Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

Application of a Quantitative HPLC-ESI-MS/MS Method for Flavonoids in Different Vegetables Matrices

Plant flavonoids have been increasingly studied for its importance in plant metabolism, inter-species interactions and human health-promoting effects. Each botanical species has a distinct flavonoid profile, and as such, this work aimed to develop and apply a high-performance liquid chromatography coupled to electrospray ionization tandem mass spectrometry (HPLC-ESI-MS/MS) method to quantify specific flavonoids in different plants and parts. Six flavonoids (isoquercitrin, quercetin, naringin, naringenin, hesperidin and rutin), two isoflavonoids (daidzin and daidzein) and one coumarin (umbelliferone) were targeted. Calibration curves showed good correlations (R2 > 0.994) and limits of quantification (≤ 500 ng mL-1). The method was applied for detection of analytes in common bean seeds (Phaseolus vulgaris), soybean leaves (Glycine max), and sour orange (Citrus aurantium), sweet orange (Citrus sinensis), and Tahiti lime (Citrus latifolia) flavedo after ultrasound assisted acidic hydroalcoholic extraction. The method was tuned for quick and efficient extraction. Principal component analysis was helpful for the discrimination of matrices based on their flavonoid profiles.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

A reassessment of the role of serotonergic system in the control of feeding behavior

O papel do sistema serotonérgico no comportamento alimentar foi avaliado através de lesões eletrolíticas do núcleo dorsal da rafe (L-NDR) e da administração de para-clorofenilalanina (PCPA, 3 mg/5 mil, icv). Avaliações crônicas foram realizadas durante 120 e 360 dias em ratos injetados com PCPA e L-NDR, respectivamente. Avaliações agudas foram realizadas em ratos em jejum e injetados com PCPA e l-triptofano (LHTP, 30 mg/kg, ip). Ratos lesionados apresentaram um aumento de 22-80% na ingestão de alimento até o sexto mês enquanto a obesidade foi evidenciada e mantida por todo o período. Ratos injetados com PCPA apresentaram um aumento da ingestão alimentar seguido de uma hipofagia do 25º ao 30º dia e um aumento transitório do peso corporal do 5º ao 60º. Agudamente, o LHTP reverteu parcialmente o aumento da ingestão de alimento em ratos tratados com PCPA e jejuados, sugerindo a preservação da capacidade de descarboxilação do precursor pelos neurônios serotonérgicos. A lenta recuperação dos níveis de ingestão alimentar em ratos lesionados revela um mecanismo de neuroplasticidade dos sistemas de regulação do comportamento alimentar. Estabelecimento de platô na curva de peso corporal dos ratos lesionados representaria o estabelecimento de um novo e mais elevado ponto de calibração do balanço energético.
The role of serotonergic system in the feeding behaviorwas appraised by electrolytic lesions in the dorsal raphe nucleus (DRN) and administration of para-chlorophenylalanine (PCPA, 3 mg/5 mul, icv). Chronic evaluations were accomplished through 120 and 360 days in PCPA-injected and DRN-lesioned rats, respectively. Acute food intake was evaluated in fasted rats and submitted to injection of PCPA and hydroxytryptophan (LHTP, 30 mg/kg, ip). DRN-lesioned rats exhibited 22-80% increase in food intake up to sixth month, whereas the obesity was evident and sustained by whole period. In PCPA-injected rats was observed an initial increase in the food intake followed by hypophagy from 25th to 30th day and a transitory increase of body weight from 5th to 60th day. In the acute study, the LHTP reverted partially the PCPA-induced increase in food intake of fasted rats suggesting a sustained capacity of decarboxylation of precursor by serotonergic neurons. Slow restoration of the levels of food intake in DRN-lesioned rats reveals a neuroplasticity in the systems that regulate feeding behavior. A plateau on the body weight curve in lesioned rats possibly represents the establishment of a new and higher set point of energetic balance.

Objetivamos avaliar a influência da depleção cerebral de serotonina no apetite por sódio. Ratos depletados de serotonina através da administração sistêmica de paraclorofenilalanina (300 mg/kg, ip, por 2 dias) exibiram intensa resposta natriorexigênica induzida pela depleção de sódio (furosemida, 20 mg/kg, sc, 24 h antes da oferta de água e NaCl 1.8%). A ingestão de NaCl 1,8% atingiu valores sempre acima do grupo controle (12,9 ±1,4 e 21,4 ±3,0 mL vs 5,7 ±1,2 e 12,7 ±1,6 mL, 30 e 300 min após apresentação de água e salina hipertônica, P < 0,05). Ao final de 24 h a resposta natriorexigênica ainda mantinha-se significativamente mais elevada em relação aos controles (33,6 ±5,1 vs 21,9 ±3,6 mL, P < 0,05) e comparável aos controles no 14° dia. Ratos depletados de serotonina exibiram uma precoce resposta natriorexigênica após administração de isoproterenol, no terceiro dia após a primeira injeção de paraclorofenilalanina. O aumento da ingestão de NaCl 1,8% iniciou-se aos 120 min (1,9 ±0,3 vs 0,45 ±0,3 mL, P < 0,05) e manteve-se elevada ao final de 24 h (17,3 ±3,2 vs 1,1 ±0,5 mL, P < 0,05) após a oferta de fluidos. Decorridos sete e 14 dias a resposta natriorexigênica tornou-se comparável à dos controles. Os dados apresentados demonstram que a depleção de serotonina cerebral com paraclorofenilalanina, exagera o apetite por sódio em paradigma de depleção de sódio ou após estimulação b-adrenérgica.
We investigate the influence of brain serotonin depletion on the sodium appetite. Rats depleted of serotonin through the systemic administration of p-chlorophenylalanine (300 mg/kg, ip, for 2 days) showed an intense natriorexigenic response induced by sodium depletion (furosemide, 20 mg/kg, sc, 24 h before water and 1.8% NaCl presentation). Intake of 1.8% NaCl was always higher than that observed for the control group (12.9 ± 1.4 and 21.4 ± 3.0 mL vs 5.7 ± 1.2 and 12.7 ± 1.6 mL, 30 and 300 min after water and saline presentation). After 24 h, the natriorexigenic response continued to be significantly higher compared to control (33.6±5.1 vs 21.9±3.6 mL,P <0.05). Fourteen days after p-chlorophenylalanine administration, 1.8% NaCl intake did not differ from controls. Serotonin-depleted rats expressed an early natriorexigenic response after isoproterenol administration on the third day after the first injection of p-chlorophenylalanine. An increase in 1.8% NaCl intake was first observed at 120 min (1.9 ± 0.2 vs 0.45 ± 0.3 mL,P <0.05) and remained high up to the end of the 24-h observation period (17.3±3.2 vs 1.1±0.5 mL,P <0.05). After 7 and 14 days, the natriorexigenic response became comparable to that of control animals. Present results show that brain serotonin depletion exaggerates the sodium appetite induced by the paradigm of sodium depletion or after beta-adrenergic stimulation.

Notes on artificial substrates for black fly (Diptera: Simuliidae) larvae and microsporidian infection in Central Amazonia, Brazil

Comparou-se três tipos de substratos (filamento plástico branco; ladrilho, blocos de ardósia cinza; e fita plástica, cor amarela) na captura de larvas de simulídeo em 5 períodos de exposição (1, 5, 7, 10 e 14 dias). O estudo foi realizado a 51 km a SE de Manaus, AM, onde foram coletadas 3 espécies: Simulium quadrifidum Lutz (81%), Simulium sp. (17,3%) e Simulium perflavum Roubaud (1,7%). Através da análise de variância dos parâmetros estudados verificou-se diferenças significativas entre os substratos utilizados (P < 0,001), mas não entre os períodos de colonização. A fita plástica foi o substrato mais eficiente para capturar imaturos de simulideos (P < 0,05), não havendo diferença significativa entre os substratos ladrilho e filamento plástico. Durante o período de estudo foi observado infecção por microsporídio (Amblyospora bracteata (Strickland)) apenas em S. quadrifidum.
