Abstract UVB-irradiation increases the risk of various skin disorders, therefore leading to inflammation and oxidative stress. In this sense, antioxidant-rich herbs such as Rosmarinus officinalis may be useful in minimizing the damage promoted by reactive oxygen species. In this work, we report the efficacy of a R. officinalis hydroethanolic extract (ROe)-loaded emulgel in preventing UVB-related skin damage. Total phenols were determined using Folin-Ciocalteu assay, and the main phytocomponents in the extract were identified by UHPLC-HRMS. Moreover, in vitro sun protection factor (SPF) value of ROe was also assessed, and we investigated the in vivo protective effect of an emulgel containing ROe against UVB-induced damage in an animal model. The ROe exhibited commercially viable SPF activity (7.56 ± 0.16) and remarkable polyphenolic content (24.15 ± 0.11 mg (Eq.GA)/g). HPLC-MS and UHPLC-HRMS results showcased that the main compounds in ROe were: rosmarinic acid, carnosic acid and carnosol. The evaluation of the in vitro antioxidant activity demonstrated a dose-dependent effect of ROe against several radicals and the capacity to reduce iron. Therefore, we demonstrated that topical application of the formulation containing ROe inhibited edema formation, myeloperoxidase activity, GSH depletion and maintained ferric reducing (FRAP) and ABTS scavenging abilities of the skin after UVB exposure.

Resumo O presente estudo avalia a influência da anestesia sobre a fauna parasitária de monogeneas em peixes, sua intensidade e sua viabilidade. Dois experimentos foram realizados: Avaliação de um método anestésico por aspersão eugenol diretamente nas brânquias e avaliação da motilidade das monogeneas e sua viabilidade; e Comparação entre imersão e aspersão diretamente nas brânquias com benzocaína e eugenol, seguido de avaliação sobre a intensidade parasitária. Os resultados sugerem que a aspersão do anestésico não interferiu na motilidade, morfologia, superfície corporal e integridade do parasita, analisadas pelo método de fluorescência. A intensidade de monogenéticos nas brânquias foi menor nos peixes anestesiados pelo método de imersão em comparação com o método de aspersão e o grupo controle. O método de anestesia por aspersão nas brânquias pode ser utilizado em estudos parasitológicos.
Abstract The present study evaluates the influence of anesthesia on the parasitic fauna of monogenea fish parasites, as its intensity and viability. Two experiments were conducted: Evaluation of an anesthetic method by sprinkling eugenol directly on gills and evaluation of monogenea motility and viability; Comparison of immersion and directly sprinkling on the gills with benzocaine and eugenol followed by evaluation on parasite intensity. The results suggest that the anesthetic sprinkling didn’t interfere in the parasite motility, morphology and body surface integrity analyzed by fluorescence method. The monogenean intensity in the gills was lower in fish anesthetized by immersion method compared to the sprinkling method and the control group. This method of anesthesia can be used in parasitological studies.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Hypertensive diabetic patients: guidelines for conduct and their difficulties

OBJECTIVE: To assess the effect of blood pressure (BP) control and other cardiovascular risk factors in patients with diabetes mellitus in a referral service for the treatment of hypertension. METHODS: A retrospective study where diabetic patients (at least 2 fasting glucose levels above 126 mg/dL, use of hypoglycemic agents or insulin, or both of these) were included. They were evaluated at the first appointment (M1) and at the last appointment (M2), regarding blood pressure, body mass index (BMI), use of hypertensive drugs, glycemia, total cholesterol (TC), creatinine, and potassium. RESULTS: Of 1,032 patients studied, 146 patients with a mean age of 61.6 years had diabetes, and 27 were men (18.5%). Mean follow-up was 5.5 years. BP values were 161.6 x 99.9 mmHg in M1 and 146.3 x 89.5 mmHg in M2. In M1, 10.4% of the patients did not use medications, 50.6% used just 1 drug, 30.8% used 2 drugs, and 8.2% used 3 or more drugs. In M2, these values were 10.9%, 39%, 39.7%, and 10.4%, respectively. Diuretics were the most commonly used medication, whereas angiotensin-converting enzyme inhibitors (ACE inhibitors) were those drugs which presented greater increase when comparing M1 to M2 (24.6% and 41.7%, respectively). Only 17,1% reached the recommended goal (BP<130x85 mmhg). The other cardiovascular risk factors did not change significantly. CONCLUSION: Our data reinforce the necessity of a more aggressive approach in the treatment of these patients, despite the social and economic difficulties in adhering to treatment.

Isolation and antigenic characterization of human immunodeficiency virus (HIV) in Brazil

Um retrovírus foi isolado de um paciente brasileiro com "Síndrome de Imunodeficiência Adquirida" (SIDA/AIDS) e caracterizado em termos de sua reatividade com soros de indivíduos infectados com vírus da imunodeficiência humana dos tipos 1 e 2 (HIV-1 e HIV-2). A análise antigênica por "Western blot" revelou que o isolado brasileiro é bastante similar a uma cepa de HIV-1 bem caracterizada. A identificação do retrovírus como HIV-1 e não HIV-2 é reforçada pelo fato dos anticorpos do paciente do qual foi isolado o vírus não terem reagido com a glicoproteínas de envelope de 140 kDa, específica para HIV-2.
A retrovirus infecting a Brazilian AIDS patient was isolated and characterized in terms of its reactivity with sera from individuals infected with human immunodeficiency viruses 1 and 2 (HIV-1 and HIV-2). The Western blot analysis revealed that the Brazilian isolate is very similar to the well characterized HIV-1 strain. The serum of the patient from whom the virus was isolated did not react with the 140 kDa envelope glycoprotein specific for HIV-2.

Estudos "in vitro" dos níveis de resistência do Plasmodium falciparum a drogas, de 1983 a 1986

Foram realizados 171 testes de sensibilidade (microtécnica) com cepas de Plasmodium falciparum da Região Amazônica brasileira para cloroquina, mefloquina, amodiaquina e quinino. Os testes tiveram duração de 24 horas com as drogas preparadas na hora da realização de cada teste. Os resultados mostraram elevada resistência a cloroquina (83%) e sensibilidade em quase a totalidade das amostras testadas para mefloquina (97,7%). Para amodiaquina e quinino observou-se sensibilidade em 51,0% e 56,5% das cepas, respectivamente. Este estudo demonstra a emergência de um possível foco de resistência do Plasmodium falciparum a mefloquina, em Tucuruí.
Drug resistance tests were performed with 171 strains of P. falciparum obtained from the Amazon Region, using chloroquine, mefloquine, amodiaquine and quinine. Tests lasted about 24 hours and the drug solutions were prepared just before use. Results show high percentage of resistance to chloroquine (83%) whereas nearly all strains were shown to be sensitive to mefloquine (97.7%). For both amodiaquine and quinine sensitivities were of 51.0% and 56.5% respectively. The present study demonstrates that resistance to mefloquine may be starting in Tucuruí, Pará.

Caracterização de cepas de plasmodium falciparum do Estado de Rondônia, Brasil, utilizando microtestes de sensibilidade aos antimaláricos, tipificação enzimática e anticorpos monoclonais

Nove amostras de Plasmodium falciparum foram coletadas de migrantes infectados no Estado de Rondônia. Estas amostras foram mantidas em cultivo para caracterização por tipificação enzimática em acetato de celulose, sensibilidade á cloroquina, amodiaquina, mefloquina e quinino e análise da diversidade antigê-nica através de anticorpos monoclonais específicos. Os resultados obtidos mostraram variação entre todas as amostras estudadas: encontrou-se resistência crescente à cloroquina, resistência intermediária à amodiaquina e quinino e sensibilidade a baixos níveis de mefloquina; apenas dois isolados mostraram sensibilidade a todas as drogas. A tipificação enzimática mostrou presença de parasitas GPI 1 e 2 e ADA 1 e 2, enquanto que para PEP e LDH todas as amostras foram do tipo 1. Sorotipagem com anticorpos monoclonais PSA mostrou presença de três sorotipos diferentes (II, III e IV). Estes resultados mostraram: a) variação entre as amostras para os marcadores analisados; b) nesta região, para o pequeno número de amostras analisadas, não foram observadas diferenças significativas ou novos tipos de parasitas.
Nine samples of Plasmodium falciparum were collected from migrant workers who were infected in the State of Rondônia, Brazil. These samples were maintained in culture for characterization by enzyme typing using acetate cellulose electrophoresis, drug susceptibility to chlo-roquine, amodiaquine, mefloquine and quinine and antigenic diversity analysis using strain specific monoclonal antibodies. The results obtained have confirmed major variation among all samples studied: it was found consistent resistance to chloroquine moderated resistance to amodiaquine and quinine and susceptibility to low levels of mefloquine; only two isolates have shown susceptibility to all drugs in the in vitra assays. Enzyme typing showed presence of both GPI 1 and 2 and ADA 1 and 2 parasites, while for PEP and LDH all samples were type 1. Se-rotyping with PSA monoclonal antibodies showed presence of three different serotypes (II, III and IV). These results confirm: a) variation among samples for markers analysed; b) in this area and for the small number of samples studied, no significant differences or new types of parasites were seen.
