RESUMO Este trabalho apresenta os resultados obtidos em um projeto de construção civil, no qual se dimensiona a isolação térmica do telhado de uma pousada na região de São Joaquim, SC, considerada uma das regiões mais frias do Brasil. Propôs-se o uso de um isolante produzido à base de asfalto reciclado, apontando sua eficiência de isolação térmica e viabilidade econômica com relação aos isolantes comumente usados pela construção civil brasileira. Os resultados obtidos demonstraram que a manta de asfalto reciclado reduz a perda de calor do telhado entre 68 e 72% e o seu custo com material é de duas a cinco vezes menor que as demais mantas estudadas. Além disso, o material do qual o produto é feito é mais ecológico do que as demais mantas térmicas. Assim, este projeto mostrou que o uso da manta de asfalto reciclado é viável economicamente, além de ser muito mais eficiente do que as mantas comercializadas atualmente no Brasil.

ABSTRACT This work shows the results obtained in a civil construction project, in which the thermal insulation of an inn’s roof is measured in são joaquim, sc, which is considered to be one of the coldest regions in brazil. it was proposed the use of an insulator produced with recycled asphalt, indicating its thermal insulation efficiency and economic viability in relation to the insulators commonly used by brazilian civil construction. the results showed that the recycled asphalt blanket reduces the roof heat loss between 68 and 72% and its material cost is two to five times smaller in relation to the other blankets studied. in addition, the material from which the product is made is more environmentally friendly than other thermal blankets. thus, this project showed that the recycled asphalt blanket use is economically viable, besides being much more efficient than the blankets currently commercialized in brazil.

Resumo Algumas classes de drogas representam maior criticidade como contaminação ambiental; os antibióticos em geral representam os contaminantes mais potentes e que causam maiores danos ao meio ambiente. O objetivo deste trabalho é quantificar a porcentagem de contaminação do solo por Azitromicina em apresentações de 500 mg, através da aplicação da Espectroscopia no Infravermelho com Transformada de Fourier. Verificou-se que o fármaco, mesmo na característica de micropoluente, exerce impactos significativos ao meio ambiente e que sua extração, quando realizada com o Acetonitrila pura, permite quantificar satisfatoriamente seus resíduos em amostras de solo.

Abstract Some classes of drugs represent greater criticality as environmental contamination; antibiotics in general represent the most potent contaminants and cause greater damage to the environment. The purpose of this work is to quantify the percentage of soil contamination by Azithromycin in presentations of 500 mg, through the application of Infrared Spectroscopy with Fourier Transform. It was verified that the drug exerts significant impacts to the environment, even in the characteristic of micropollutant, and that its extraction when performed with Pure Acetonitrile allows its residues in soil samples to be satisfactorily quantified.

Resumo A gestão da qualidade em serviços tem sido um tema frequentemente abordado como fator de diferenciação para as organizações nos dias atuais. Aliado a esse fato, os serviços de manutenção de utilidades são fundamentais para o funcionamento e desempenho adequado da infraestrutura das empresas. Nesse contexto, este artigo tem como objetivo caracterizar, mensurar e analisar a percepção da qualidade dos usuários de serviços internos de manutenção de utilidades em uma empresa de manufatura, líder em seu mercado de atuação. Por meio da elaboração de um questionário estruturado considerando o método SERVQUAL, foram coletados dados de forma não probabilística junto aos usuários. Para os dados coletados foi utilizada a análise com base no alfa de Cronbach, bem como o teste-t, buscando-se verificar a consistência e confiabilidade do instrumento. Os resultados indicaram diferenças entre expectativa e percepção dos usuários na maioria dos fatores analisados, sendo tais gaps discutidos no presente trabalho.

Abstract Service quality management is currently a subject that companies frequently consider as a differentiated factor. Maintenance services are essential to the suitable operation and performance of the internal services maintenance of a manufacturing company that is a market leader. Non-probabilistic data were collected from service users with an instrument based on the SERVQUAL scale. Cronbach alpha and t-test were used to verify the consistency and reliability of the instrument. The results highlighted differences between the performance and expectations of users regarding the most analyzed factors; discrepancies are subsequently discussed in the paper.

In this study, seven wine samples were prepared varying the amount of pulp of acerola fruits and the sugar content using the simulated annealing technique to obtain the optimal sensory qualities and cost for the wine produced. S. cerevisiae yeast was used in the fermentation process and the sensory attributes were evaluated using a hedonic scale. Acerola wines were classified as sweet, with 11°GL of alcohol concentration and with aroma, taste, and color characteristics of the acerola fruit. The simulated annealing experiments showed that the best conditions were found at mass ratio between 1/7.5-1/6 and total soluble solids between 28.6-29.0 °Brix, from which the sensory acceptance scores of 6.9, 6.8, and 8.8 were obtained for color, aroma, and flavor, respectively, with a production cost 43-45% lower than the cost of traditional wines commercialized in Brazil.

This study aimed to obtain the condition of maximum bromalein activity in different parts of pineapple plants produced in vitro, by micropropagation. The sStems and leaves of Pérola and Imperial cultivar plants were evaluated after three and eight months of in vitro cultivation in Murashige and Skoog medium without growth phytoregulator, macerated in potassium phosphate buffer at different pH values (5.7, 6.7 and 7.7). Total protein and proteolytic activity were determined in the samples after three- and eight-month cultivation periods. For both the cultivars, the best results were obtained at pH 5.7 in extraction media. Pérola cultivar, showed higher bromelain activity in the leaves cultivated in vitro for three months (0.0194U/mL) while in the Imperial cultivar, it was higher in the stem after eight months (0.0179 U/mL). Imperial cultivar showed higher bromelain activity than the Pérola's.

The aim of this work was to study the production of e bromelain from the Ananas comosus L. Merril, by determining the process conditions using flat membranes. The production system modeling generated a hyperbolical curve and the optimization by response surfaces showed an influence of the transmembrane pressure higher than the pH influence. The cost of the production of bromelain from A. comosus was estimated 9 to 13 times lower than Sigma's retail sales price and 6.5 to 8.5 times lower than when this enzyme was obtained through a liquid-liquid extraction, which showed the economical feasibility of the process.

This work aimed to study the partitioning of a lipase produced by Burkholderia cepacia in PEG/Phosphate aqueous two phase system (ATPS) and its characterization. Lipase was produced by B. cepacia strains in a fermenter. Enzyme partitioning occurred at pH 6.0 and 8.0, using PEG 1500 and 6000 on two tie lines. Metal ions, pH and temperature effects on enzyme activity were evaluated. Five milliliter of 7.5% olive oil emulsion with 2.5% gumarabic in 0.1M sodium phosphate buffer at pH 8.0 and 37ºC were used for the activity determinations. Results showed that crude stratum from B. cepacia was partitioned by PEG1500/phosphate ATPS at pH 6.0 or 8.0 for, which the partitioning coefficients were 108-and 209-folds. Lipase presented optimal activity conditions at 37ºC and pH 8.0; it showed pH-stability for 4 h of incubation at different pH values at 37ºC. Metal ions such as Mn2+ , Co2+, I-and Ca2+ sustained enzymatic activities; however, it was inhibited by the presence of Fe2+, Hg2+ and Al3+ . Km and Vmax values were 0.258 U/mg and 43.90 g/L, respectively. A molecular weight of 33 kDa and an isoelectric point at pH 5.0 were determined by SDS-PAGE and IFS electrophoresis, respectively.

In this work, the thermodynamic equilibrium and applying of PEG4000/Phosphate ATPS on the purification of bromelain extracted from pineapple was studied. A rigorous study of the equilibrium curves and tie-line length from PEG4000/phosphate ATPS were done for the pH 6-11 at 25ºC. Results showed that there was augment in the PEG and salt contents with the high pH value from PEG4000/Phosphate ATPS and two-phase formation needed only increasing the PEG content. Two tie-line length at pH 11 from PEG4000/Phosphate ATPS were optimal condition for bromelain purification, one on composition of 14% PEG and 13% salt and other at 12.6% PEG and 12.2% salt, while a 25-62 folds of enzyme was found. SDS-PAGE electrophoreses had one band only, which showed that bromelain was purified. Optimum conditions of bromelain use were found at pH 7 and between 30-40ºC.

Este trabalho objetivou otimizar o processo de hidrólise do amido de mandioca com α-amilase de A. niger e obter o álcool deste xarope. Os ensaios foram realizados a pH 4,8; em que variaram-se a concentração do amido (entre 7-22 g.L-1) e a temperatura (entre 30-59,1 ºC). Durante a fermentação, usaram-se nos mostos 2,2 e 5% de amido de mandioca. Os resultados da hidrólise mostraram que o tempo ficou entre 20-200 minutos; a análise RSM mostrou que o rendimento diminuiu nas concentrações médias; e as condições ótimas foram encontradas entre 55-59,1 ºC e com a concentração entre 7,9-10 ou 20-22 g.L-1, em que se hidrolisou 80% do amido. A melhor condição de fermentação foi obtida para o mosto contendo 5% de amido. Sua composição final foi de 0,668 g.L-1 de ART, 0,572 g.L-1 e de AR, 3,71 ºGL. O rendimento alcoólico foi de 45%, demonstrando que este processo é uma alternativa eficiente à indústria sucroalcooleira.

The objective of this study was the optimization of the manioc root starch hydrolysis by α-amylase from A. niger and alcohol production from this starch syrup. Hydrolysis assays were performed at pH 4.8, the starch concentration and the temperature varied from 7 to 22 g.L-1, and 30 to 59.1 ºC, respectively. The starch contents during the syrup fermentation were 2.2 and 5%. The results of the starch hydrolysis showed that the hydrolysis time was between 20-200 minutes, the RSM analysis showed a decrease in the starch yield at the average concentration and above of 70% at high temperatures, and the optimal conditions were found at temperature between 55-59.1 ºC and concentration between 7.9-10 or 20-22 g.L-1; conditions at which 80% of the starch was hydrolyzed. The best fermentation condition was obtained for the must containing 5% of starch. The final fermentation composition obtained was of 0.68 g.L-1 ART and 0.572 g.L-1 AR. After the fermentation process, an alcohol yield of 45% was obtained showing that this process is a very good alternative for sugar-alcohol industries.

Neste trabalho realizou-se a purificação das enzimas α e β-amilases de malte de milho combinando os processos de adsorção em leito expandido com o fracionamento por etanol. Alimentaram-se 5 mL de uma solução a 2% de malte de milho em tampão fosfato 0,05 M a pH 5 em uma coluna contendo as resinas de troca iônica Amberlite IRA 410, em leito fixo de 4 cm fluidizado para 8 cm. A eluição foi promovida com NaCl 0,25M com 14 mL/minuto de vazão descendente e o material eluído foi fracionado com álcool etílico nas concentrações de 40-70%, 50-80% e 60-90% (v/v) a 20 e 30 ºC. A recuperação da atividade e o fator de purificação foram de 95% e 23 vezes, respectivamente, na primeira etapa. A melhor condição de fracionamento das enzimas foi apresentada a 50-80% de álcool e 20 ºC, e o fator de purificação ficou acima de 100 vezes.

In the present work the purification of α and β-amylase enzymes of maize malt was carried out by expanded bed adsorption and ethanol fractionation downstream processes. 5 mL of 2% maize malt solution in 0.05 M phosphate buffer at pH was injected into a chromatographic column containing Amberlite 410 ion-exchange resin, on a 4 cm fixed bed fluidized to 8 cm. 0.25 M NaCl solution at 14 mL/ minuto of descending flux was used for promotion of the elution, the eluted matter was fractioned by 40-70, 50-80 and 60-90% ethyl alcohol concentration at 20 and 30 ºC. Activity recovery and purification factor were of 95% and 23 times, respectively, in the first step. 50-80% alcohol concentration at 20ºC was the best condition, in which a purification factor of more than 100 times was achieved.

Este trabalho teve como objetivo principal otimizar as condições para extração da bromelina do suco do abacaxi (Ananas comosus) por micelas reversas. A purificação foi feita usando o processo de extração em batelada e contínuo, este último em uma micro-coluna de campânulas pulsantes. A solução micelar catiônica foi preparada com o surfactante BDBAC, i-octano como solvente e hexanol como co-solvente. Na extração em batelada encontrou-se um fator de purificação de 3 vezes, e seus melhores valores de concentração do agente surfactante, co-solvente e sal, de pH da re-extração e extração, foram respectivamente iguais a: 100 mM, 10% v/v, 1 M, 3,5 e 8. Para a operação contínua, as variáveis independentes ótimas foram: 0,67 para a razão entre as taxas de fluxos da fase leve e a total e 1 s para o intervalo de tempo entre pulsos das campânulas. Este ponto ótimo leva a uma produtividade de 1,29 mL/min e a um fator de purificação igual a 4,96.

The main aim of this study was to optimize the conditions for bromelain extraction by reversed micelles from pineapple juice (Ananas comosus). The purification was carried out in batch extraction and a micro-column with pulsed caps for continuous extraction. The cationic micellar solution was made of BDBAC as a surfactant, isooctane as a solvent and hexanol as a co-solvent. For the batch process, a purification factor of 3 times at the best values of surfactant agent, co-solvent and salt concentrations, pH of the back and forward extractions were, 100 mM, 10% v/v, 1 M, 3.5 and 8, respectively. For the continuous operation, independent variables optimal point was determined: ratio between light phase flow rate and total flow rate equal to 0.67 and 1 second for the time interval between the pulses. This optimal point led to a productivity of 1.29 mL/min and a purification factor of 4.96.

Neste trabalho as enzimas α e β-amilases foram obtidas de malte de milho e depois foram caracterizadas bioquimicamente. Um estudo da germinação foi feito para obtenção do malte com boa atividade amilásica. A germinação ocorreu em escala laboratorial e a atividade enzimática foi medida diariamente. Um modelo exponencial do tipo A=A0eµt foi ajustado a dependência do tempo de germinação com a atividade, mostrando que o comportamento da atividade enzimática no processo de germinação é semelhante ao crescimento de microorganismos. Este modelo pode ser aplicado para descrever o mecanismo de produção da α-amilase para cada variedade de milho e de outros cereais. A caracterização do malte de milho mostrou que as α e β-amilase têm pH ótimo entre 4,0-6,5, temperatura ótima de 50 e 90ºC, e massa molar de 67,4 e 47,5 kDa, respectivamente. Este trabalho contribuiu com os avanços da biotecnologia gerando condições de emprego de uma nova e barata fonte de amilases.

In this work the α and β-amylase enzymes were obtained from maize (Zea mays) malt and were biochemistry characterized. A germination study to obtain the maize malt with good amylase activity was made. The maize seeds were germinated in laboratory and the enzymatic activity was measured daily. Activity dependence to germination time were fitted to an exponential model (A=A0eµt), which showed that the behaviour of enzymatic activity in the germination process was similar to the growth of the microorganism. Its model could be applied to describe the mechanism of α-amylase production for each maize varieties and others cereals. Maize malt characterization showed that α and β-amylase had optimal pH between 4-6.5, optimal temperature 50 and 90ºC, and molecular weight of 67.4 and 47.5kDa, respectively. This work contributed with the advances in biotechnology generating of conditions for application of a new and of low price amylases source.

A concentração do suco de abacaxi é necessária para manter a atividade da bromelina e padronizar a composição e atividade proteolítica. Assim, este trabalho objetivou a obter um extrato de bromelina pura do suco do Ananas comosus L. Merril por processos de separação por membranas. Um planejamento experimental do tipo 2² foi feito para estudar a influência do pH a da pressão transmembranar sobre a recuperação da atividade por micro-filtração usando uma membrana plana. Em uma segunda etapa, purificou-se a enzima alvo por ultra-filtração usando um "kit millipore" de 10 kDa. A melhor condição para a concentração da bromelina foi a pH 7,5 e pressão transmembranar de 0,05 bar, onde 85% da atividade da bromelina foi recuperado. A ultra-filtração manteve 100% da atividade proteolítica e concentrou em 10 vezes o extrato de bromelina. A eletroforese via SDS-PAGE mostrou que o ultra-filtrado teve alta pureza e a bromelina da polpa do Ananas comosus tem um peso molecular de 24.5 kDa.

The concentration of pineapple juice is needed to retain the bromelain activity and to standardize the composition and proteolytic activity. Thus, this work aimed to obtain a pure bromelain extract from the Ananas comosus L. Merril juice by membrane separation process. A 2² experimental planning was used to study the influence of pH and transmembrane pressure on the activity recovery by micro-filtration using a plain membrane. In second step, this enzyme was purified by the ultra-filtration using a 10 kDa millipore kit. The best operation condition to bromelain concentration using the plain membrane was at pH 7.5 and transmembrane pressure of 0.05 bar, while 85% of bromelain activity was recovered. Ultra-filtration retained 100% of proteolytic activity and concentrated in 10 fold the bromelain extract. SDS-PAGE electrophoresis showed that the ultra-filtrated had high purity and the bromelain from A. comosus pulp had a molecular weight of 24.5 kDa.

No presente trabalho foi estudado o efeito da expansão do leito sobre a adsorção de BSA na resina de troca iônica Amberlite IRA 410. O comportamento hidrodinâmico de uma coluna de adsorção em leito expandido sob efeito da adição de biomoléculas, sal e variação da temperatura também foi estudado para obter as condições ótimas de recuperação da BSA sob a resina de troca iônica. A distribuição do tempo de residência mostrou que a HEPT, a dispersão axial e o número de Pecletl aumentaram com a temperatura, altura do leito, porosidade do leito e velocidade linear. A capacidade de ligação da resina aumentou com a expansão do leito. A resina de troca iônica Amberlite IRA 410 mostrou ter afinidade pela BSA, com uma recuperação de 78,36 % da proteína total.

In the present work, the effect of bed expansion on BSA adsorption on Amberlite IRA 410 ion-exchange resin was studied. The hydrodynamic behavior of an expanded bed adsorption column on effects of the biomolecules and salt addition and temperature were studied to optimize the conditions for BSA recovery on ion-exchange resin. Residence time distribution showed that HEPT, axial dispersion and the Pecletl number increased with temperature and bed height, bed voidage and linear velocity. The binding capacity of the resin increased with bed height. The Amberlite IRA 410 ion-exchange showed an affinity for BSA with a recovery yield of 78.36 % of total protein.

Este trabalho objetivou a otimização da imobilização das enzimas amilases extraídas do malte do milho, usando alginato de sódio. A concentração do malte no extrato, a porcentagem de alginato de sódio e o pH foram usados como fatores que influenciam na imobilização das enzimas. Os resultados mostraram que as melhores condições de imobilização foram obtidas quando se usou as soluções de malte de milho em duas faixas de concentrações, uma entre 3,75 a 5 g.L-1 e outra entre 15 a 16,25 g.L-1, em pH entre 4,83 a 6,6 e 4% (m/v) de alginato de sódio, condições nas quais se conseguiu imobilizar 100% das enzimas com baixa perda de atividade. Este trabalho mostrou como se obter amilases de malte de milho imobilizadas por oclusão em alginato de sódio e que podem ser usadas em processos industriais de hidrólise de amido.

This work aimed at optimizing the immobilization of amylases obtained from corn malt by the occlusion method with sodium alginate. Corn malt concentration, sodium alginate mass percentage and pH were used as factors that influence enzyme immobilization. The results showed that the best immobilization conditions were obtained at two ranges of corn malt concentration, one between 3.75-5 g.L-1 and the other between 15-16.25 g.L-1, at pH between 4.83-6.6 and at 4% (m/m) sodium alginate. In these conditions, 100% of the enzymes were immobilized with a low loss of activity. This work showed how to obtain immobilized corn malt amylases that can be used in industrial processes of starch hydrolysis by occlusion in sodium alginate.
