RESUMO Este estudo teve como objetivo caracterizar genótipos de M. synoviae circulantes em poedeiras comerciais, na Região Sudeste do Brasil. Para estabelecer a relação evolutiva entre as cepas de MS, foi analisada a sequência genética do gene vlhA de 11 cepas de MS identificadas no Rio de Janeiro e em São Paulo, de 10 outras cepas de MS identificadas no Brasil e cujas sequências foram depositadas no banco de dados GenBank, e da cepa vacinal MSH. O método da máxima verossimilhança e o modelo de Kimura com dois parâmetros foram utilizados para comparar as cepas. As sequências obtidas foram depositadas no Genbank, sob os números de acesso OP279775 a OP279785. Foi possível verificar a presença de diferentes cepas circulantes no Brasil, com alta similaridade entre as cepas do Rio de Janeiro pela análise do gene vlhA. As duas cepas paulistas detectadas no presente estudo possuem o baixo percentual (68%) de similaridade, demonstrando a variabilidade das cepas dessa localidade.

RESUMO A doença periodontal é uma afecção comum, relacionada ao aprisionamento de alimentos em diastemas não fisiológicos, em equinos. O tratamento consiste na correção da causa primária, limpeza e desbridamento do sulco gengival, denominado tratamento convencional (TC). Frequentemente antimicrobianos são necessários, pela gravidade ou patogenicidade dos agentes. A terapia fotodinâmica adjuvante (TF) tem sido estudada pelo seu potencial combate bacteriano, sem causar resistência bacteriana. O objetivo deste estudo foi analisar o uso da TF na doença periodontal, experimentalmente induzida, em dentes incisivos de equinos, e compará-la com o TC. O TC não resultou em melhora clínica estatisticamente, tanto em graus como em profundidade, apenas numérica na profundidade aos 30 dias. A TF foi empregada em dentes com profundidade maior da bolsa periodontal que a do grupo TC e, mesmo assim, apresentou melhora clínica já com sete dias, e mais efetiva aos 30, atingindo, em média, o valor considerado normal, três milímetros. A TF apresentou potencial para ser aplicada na rotina, pelo incremento nos resultados, sem causar efeitos colaterais.

ABSTRACT Periodontal disease is a common condition, related to the entrapment of food in non-physiological diastemas in horses. Treatment consists of correction of the primary cause, cleansing and debridement of the gingival sulcus, called Conventional Therapy (CT). Often antimicrobials are requested due to the severity or pathogenicity of the agents. Adjuvant Photodynamic Therapy (PDT), has been studied for its potential bacterial combat, without causing bacterial resistance. The objective of this study was to analyze the use PDT with the experimentally induced periodontal disease in the incisor teeth of horses, and to compare with the CT. The CT did not result in clinical improvement, either in degrees or millimeters. The PDT was used in teeth with a greater depth of the periodontal pocket than the TC group, and even then, showed clinical improvement in only seven days, and more effective at 30, reaching the three millimeter value considered normal on average. The PDT presented the potential to be applied in the routine by the increase in the results without causing side effects.

RESUMO Este trabalho objetivou estabelecer valores de referência por meio da elastografia ARFI (“Acoustic Radiation Force Impulse”) do tendão flexor digital superficial, tendão flexor digital profundo, ligamento acessório inferior e ligamento suspensor do boleto em equinos hígidos. Vinte e quatro equinos foram submetidos aos exames ultrassonográfico e elastográfico (método ARFI quantitativo) dos tendões e ligamentos da porção distal do membro torácico, em nove regiões anatômicas diferentes. Os elastogramas mostraram tendões e ligamentos de coloração alaranjada a vermelha, que representam maior rigidez. Não houve diferenças em relação ao membro avaliado; as velocidades de cisalhamento foram maiores nos planos longitudinais. No plano transversal, os ligamentos suspensores foram mais rígidos em comparação aos demais. No plano longitudinal, os ligamentos mostraram-se mais rígidos que os tendões, e o tendão flexor digital profundo, mais rígido que o tendão flexor digital superficial. As estruturas mostraram-se mais rígidas nos machos, e houve uma tendência à maior rigidez em animais mais velhos. A elastografia ARFI comprovou ser um método viável e reprodutível para avaliação dos tendões e ligamentos dos membros torácicos de equinos. As características qualitativas obtidas a partir da rigidez dessas estruturas podem ser utilizadas para testes em equinos com distúrbios musculoesqueléticos e também em outras espécies animais.

ABSTRACT This study set out to determine normal values for acoustic radiation force impulse (ARFI) elastography of the superficial digital flexor tendon, deep digital flexor tendon, accessory ligament of the deep digital flexor tendon and suspensory ligament in horses. Twenty-four horses were submitted to conventional sonographic assessment and elastography imaging (quantitative ARFI method) of distal forelimb tendons and ligaments at nine anatomical regions. Elastograms depicted tendons and ligaments in orange to red tones consistent with greater stiffness. Shear wave velocity was higher in the longitudinal compared to the cross-sectional plane but did not differ significantly between the left and right forelimbs. The suspensory ligament was stiffer compared to remaining structures in the cross-sectional plane. Ligaments were stiffer than tendons and the deep digital flexor tendon was stiffer than the superficial digital flexor tendon in the longitudinal plane. Target structures were stiffer in male compared to female horses and stiffness tended to increase with age. ARFI elastography proved to be a feasible and reproducible imaging modality for assessment of distal forelimb tendons and ligaments in horses. Qualitative features derived from stiffness quantification in these structures can be applied to horses with musculoskeletal disorders, as well as to other animal species.

Currently, there is great clinical need for suitable synthetic grafts that can be used in vascular diseases. Synthetic grafts have been successfully used in medium and large arteries, however, their use in small diameter vessels is limited and presents a high failure rate. In this context, the aim of this study was to develop tissue engineering scaffolds, using poly(trimethylene carbonate-co-L-lactide) (PTMCLLA), for application as small diameter vascular grafts. For this, copolymers with varying trimethylene carbonate/lactide ratios – 20/80, 30/70, and 40/60 – were submitted to electrospinning and the resulting scaffolds were evaluated in terms of their physicochemical and biological properties. The scaffolds produced with PTMCLLA 20/80, 30/70, and 40/60 showed smooth fibers with an average diameter of 771±273, 606±242, and 697±232 nm, respectively. When the degradation ratio was evaluated, the three scaffold groups had a similar molecular weight (Mw) on the final day of analysis. PTMCLLA 30/70 and 40/60 scaffolds exhibited greater flexibility than the PTMCLLA 20/80. However, the PTMCLLA 40/60 scaffolds showed a large wrinkling and their biological properties were not evaluated. The PTMCLLA 30/70 scaffolds supported the adhesion and growth of mesenchymal stem cells (MSCs), endothelial progenitor cells, and smooth muscle cells (SMCs). In addition, they provided a spreading of MSCs and SMCs. Given the results, the electrospun scaffolds produced with PTMCLLA 30/70 copolymer can be considered promising candidates for future applications in vascular tissue engineering.

Realizou-se a detecção do gene de Staphylococcus aureus, de enterotoxinas e de resistência à meticilina com extração de DNA feita diretamente de amostras de leite. Das 200 amostras estudadas, 145 (72,5%) amplificaram o gene femA, e estas foram analisadas quanto à presença dos genes sea, seb, sec e mecA. Os genes das enterotoxinas mais prevalentes foram: sea (60%), seb (37,9%) e sec (6,9%). Foram encontradas 18 amostras de leite (11,0 %) com S. aureus portadores do gene mecA. A detecção de S. aureus diretamente do leite, sem a necessidade de isolamento bacteriano e a caracterização do potencial enterotoxigênico, demonstra que a técnica de PCR é muito útil para estudos epidemiológicos das infecções estafilocócicas da glândula mamária. O alto percentual (72,5%) de amostras de leite positivas para a presença do gene femA sugere que S. aureus constitui um dos principais agentes causadores de infecções intramamárias na microrregião de Sete Lagoas-MG e que seu potencial enterotoxigênico e presença do gene mecA, que identifica o S. aureus resistente à meticlina, representa um risco potencial à saúde pública.

This work was performed to detect the Staphylococcus aureus gene, enterotoxins resistance to methicillin with the extraction of DNA directly from milk samples. Of the 200 samples studied 145 (72.5%) amplified the femA gene, which were analyzed regarding the presence of sea, seb, sec and mecA genes. The most prevalent enterotoxins genes were: sea (60%), seb (37.9%) and sec (6.9%). 18 milk samples (11 %) had S. aureus carrying the mecA gene. The detection of S. aureus directly from the milk, with no need for bacterial isolation and the characterization of the enterotoxigenic potential demonstrate that the PCR technique is very useful for epidemiological studies of staphylococcal infections of the mammary gland. The high percentage (72.5%) of positive milk samples for the presence of the femA gene suggests that S. aureus constitutes of the main agents which cause intramammary infections in the micro region of Sete Lagoas-MG and that its enterotoxigenic potential and the presence of the mecA gene, which identifies the S. aureus resistant to methicillin, represent a potential risk to public health.

The objective of this work was to study the effects of different supplementation levels on forage intake, animal performance and nutrients digestibility in Nellore steers finished in Bahia Southwest pastures. The experiment was done in an area of 52.0 ha, divided into eight sectors of approximately 6.5 ha each, with a watering point at center of field. There were studied four levels of supplementation according to the body weight which were T00= mineral, T03= 0.3%, T06= 0.6% and T09= 0.9% of the animal live weight. During the supplementation phase it was a decreasing effect on pasture and in neutral detergent fiber intake. Also it was a linear effect, with increasing the supplementation levels, on intake of crude protein, ether extract, non fibrous carbohydrates, total nutrients digestibility, digestibility energy and me-tabolizable energy. During the rainy season none of variables was affected. The animal performance presented a linear effect during supplementation and remained unchanged in rainy season. The feed conversion and feed efficiency were positively affected by supplementation levels. There was an increasing linear effect on digestibility coefficients of dry matter, crude protein and non fibrous carbohydrates. The digestibility of neutral detergent fiber presented a quadratic effect while the ether extract was not affected. During the rainy season, none of the variables was affected.

Objetivou-se com este estudo verificar os efeitos de diferentes níveis de suplementação em pastagens sobre o consumo de forragem, o desempenho animal e a digestibilidade dos nutrientes em novilhos Nelore na fase de terminação em pastagens no Sudoeste da Bahia. A parte de campo foi implantada numa área de 52,0 ha, dividida em oito piquetes de aproximadamente 6,5 ha cada, subdivididas em setores com aguada central. Testaram-se quatro níveis de suple-mentação em função do peso vivo: T00, T03, T06 e T09. Na fase de suplementação, houve efeito decrescente sobre o consumo da pastagem e da fibra em detergente neutro. Houve efeito linear crescente em função dos níveis de suplementação para os consumos de proteína bruta, extrato etéreo, carboidrato não-fibroso, nutrientes digestíveis totais, energia digestível e energia bruta. No período chuvoso, nenhuma variável foi alterada. O desempenho animal apresentou efeito linear crescente na fase de suplementação e manteve-se inalterado no período chuvoso. A conversão alimentar e a eficiência alimentar foram alteradas positivamente pela inclusão dos níveis de suplementação. Houve efeito linear crescente sobre os coeficientes de digestibilidade da matéria seca, proteína bruta e carboidratos não-fibrosos. A digestibilidade da fibra em detergente neutro apresentou efeito quadrático enquanto a do extrato etéreo não foi alterada. No período chuvoso, nenhuma das variáveis foi alterada.

Estudou-se efeito da infecção por Goezia leporini Martins & Yoshitoshi, 2003 (Nematoda: Anisakidae) sobre as características hematológicas de Leporinus macrocephalus (Osteichthyes:Anostomidae) cultivado. Palidez das brânquias, rins, fígado e coração, pontos negros nos rins e acúmulo de líquido na cavidade visceral, estômago e intestinos foram observados. O conteúdo da vesícula biliar tinha aparência pálida e translúcida. Observaram-se alta e moderada correlações positivas entre número de nematóides e peso do peixe estimadas dentro dos grupos de peixe de 0-100g e 100-200g, respectivamente. As extensões sangüíneas revelaram variações no tamanho (anisocitose) e forma (poiquilocitose) dos eritrócitos, bem como eritrócitos em divisão. Não houve alteração (P>0,05) na contagem total de eritrócitos, de leucócitos, na taxa de hemoglobina e nos percentuais de trombócitos e monócitos. A infecção provocou redução (P<0,05) no percentual de hematócrito, no volume corpuscular médio, na concentração de hemoglobina corpuscular média e no percentual de linfócitos, e aumento (P<0,05) no percentual de neutrófilos e eosinófilos no sangue circulante de peixes infectados. Este é o primeiro relato no Brasil que relaciona hematologia e infecção por nematóides em peixes cultivados.

The effect of Goezia leporini Martins & Yoshitoshi, 2003 (Nematoda: Anisakidae) infection on the haematological characteristics of cultivated Leporinus macrocephalus (Osteichthyes:Anostomidae) was studied. Paleness of gills, kidneys, liver and heart, black spots on the kidney and accumulation of fluid in the visceral cavity, stomach and intestines were observed. Gall bladder content had pale and translucent aspect. Strong and slight positive correlations between number of nematodes and fish weight were estimated within the 0-100g and 100-200g fish weight group, respectively. Blood smears from infected fish showed variation in erythrocyte size (anisocytosis) and shape (poikilocytosis), and also dividing erythrocytes. No significant alteration (P>0.05) was shown as to erythrocyte, leukocyte count, haemoglobin concentration and thrombocyte and monocyte percentage. Parasite infection provoked significant reduction (P<0.05) in hematocrit, mean corpuscular volume, mean corpuscular haemoglobin concentration and lymphocyte percentage. On the other hand, significant increase (P<0.05) in neutrophil and eosinophil percentage in circulating blood of infected fish was observed. This is the first report regarding haematology of nematode infected freshwater cultivated fish in Brazil.
