Antifungal effect of (R) and (S)-citronellal enantiomers and their predictive mechanism of action on Candida albicans from voriconazole-resistant onychomycoses

Abstract Onychomycosis is the most common disease affecting the nail unit and accounts for at least 50% of all nail diseases. In addition, Candida albicans is responsible for approximately 70% of onychomycoses caused by yeasts. This study investigated the antifungal effect of (R) and (S)-citronellal enantiomers, as well as its predictive mechanism of action on C. albicans from voriconazole-resistant onychomycoses. For this purpose, in vitro broth microdilution and molecular docking techniques were applied in a predictive and complementary manner to the mechanisms of action. The main results of this study indicate that C. albicans was resistant to voriconazole and sensitive to the enantiomers (R) and (S)-citronellal at a dose of 256 and 32 µg/mL respectively. In addition, there was an increase in the minimum inhibitory concentration (MIC) of the enantiomers in the presence of sorbitol and ergosterol, indicating that these molecules possibly affect the integrity of the cell wall and cell membrane of C. albicans. Molecular docking with key biosynthesis proteins and maintenance of the fungal cell wall and plasma membrane demonstrated the possibility of (R) and (S)-citronellal interacting with two important enzymes: 1,3-β-glucan synthase and lanosterol 14α-demethylase. Therefore, the findings of this study indicate that the (R) and (S)-citronellal enantiomers are fungicidal on C. albicans from onychomycoses and probably these substances cause damage to the cell wall and cell membrane of these micro-organisms possibly by interacting with enzymes in the biosynthesis of these fungal structures.
Resumo A onicomicose é a doença mais comum que afeta a unidade ungueal e representa pelo menos 50% de todas as doenças ungueais. Além disso, a Candida albicans é responsável por aproximadamente 70% das onicomicoses causadas por leveduras. Nesse estudo, foi investigado o efeito antifúngico dos enantiômeros (R) e (S)-citronelal, bem como seu mecanismo de ação preditivo sobre C. albicans de onicomicoses resistentes ao voriconazol. Para este propósito, foram aplicadas técnicas in vitro de microdiluição em caldo e docking molecular de forma preditiva e complementar para os mecanismos de ação. Os principais resultados deste estudo indicam que C. albicans foi resistente ao voriconazol e sensível aos enantiômeros (R) e (S)-citronelal na dose de 256 e 32 µg/mL respectivamente. Além disso, houve aumento da concentração inibitória mínima (CIM) dos enantiômeros na presença do sorbitol e do ergosterol, indicando que estas moléculas possivelmente afetem a integridade da parede e da membrana celular de C. albicans. O docking molecular com proteínas chave da biossíntese e manutenção da parede celular e da membrana plasmática fúngica, demonstraram a possibilidade do (R) e (S)-citronelal interagir com duas importantes enzimas: 1,3-β-glucan sintase e lanosterol 14α-demetilase. Portanto, os achados desse estudo indicam que os enantiômeros (R) e (S)-citronelal são fungicidas sobre C. albicans de onicomicoses e provavelmente essas substâncias causem danos a parede e a membrana celular desses microrganismos possivelmente por interagir com as enzimas da biossíntese destas estruturas fúngicas.

The present work describes the chemical constituents of Odontadenia lutea (Vell.) Markgr. leaves and the toxicities of its extract and fractions against Atta sexdens rubropilosa Forel (Hymenoptera: Formicidae). Chromatographic procedures of the ethanolic extract resulted in the identification of the triterpene β-amyrin, the flavonoid rutin, two fatty acids palmitic and linolenic, and one glyceroglycolipid 3-O-(9,12,15-octadecatrienoyl)-glyceryl-β-D-galactopyranoside, which are known compounds, but they are described for the first time in the Odontadenia genus. The known triterpenes lupeol and α-amyrin were also identified. Structural identification of the compounds was performed by analysis of IR, ESI-MS, and 1D and 2D NMR spectra. The toxicity of its extract and fractions from O. lutea leaves was tested against leaf-cutting ants Atta sexdens rubropilosa Forel by employing ingestion bioassay procedures. The hexane fraction (2 mg mL-1) decreases the average survival of ants from sixteen to six days, causing 98% mortality on the 14th day and 100% at the end of the experiment.

OBJECTIVES: To prospectively evaluate demographic, anthropometric and health-related quality of life (HRQoL) in pediatric patients with laboratory-confirmed coronavirus disease 2019 (COVID-19) METHODS: This was a longitudinal observational study of surviving pediatric post-COVID-19 patients (n=53) and pediatric subjects without laboratory-confirmed COVID-19 included as controls (n=52) was performed. RESULTS: The median duration between COVID-19 diagnosis (n=53) and follow-up was 4.4 months (0.8-10.7). Twenty-three of 53 (43%) patients reported at least one persistent symptom at the longitudinal follow-up visit and 12/53 (23%) had long COVID-19, with at least one symptom lasting for >12 weeks. The most frequently reported symptoms at the longitudinal follow-up visit were headache (19%), severe recurrent headache (9%), tiredness (9%), dyspnea (8%), and concentration difficulty (4%). At the longitudinal follow-up visit, the frequencies of anemia (11% versus 0%, p=0.030), lymphopenia (42% versus 18%, p=0.020), C-reactive protein level of >30 mg/L (35% versus 0%, p=0.0001), and D-dimer level of >1000 ng/mL (43% versus 6%, p=0.0004) significantly reduced compared with baseline values. Chest X-ray abnormalities (11% versus 2%, p=0.178) and cardiac alterations on echocardiogram (33% versus 22%, p=0.462) were similar at both visits. Comparison of characteristic data between patients with COVID-19 at the longitudinal follow-up visit and controls showed similar age (p=0.962), proportion of male sex (p=0.907), ethnicity (p=0.566), family minimum monthly wage (p=0.664), body mass index (p=0.601), and pediatric pre-existing chronic conditions (p=1.000). The Pediatric Quality of Live Inventory 4.0 scores, median physical score (69 [0-100] versus 81 [34-100], p=0.012), and school score (60 [15-100] versus 70 [15-95], p=0.028) were significantly lower in pediatric patients with COVID-19 at the longitudinal follow-up visit than in controls. CONCLUSIONS: Pediatric patients with COVID-19 showed a longitudinal impact on HRQoL parameters, particularly in physical/school domains, reinforcing the need for a prospective multidisciplinary approach for these patients. These data highlight the importance of closer monitoring of children and adolescents by the clinical team after COVID-19.

Resumo A Bacia Hidrográfica do rio Bacanga (BHRB) apresenta problemas ambientais relacionados a urbanização e lançamentos de esgoto in natura que comprometem a qualidade desse ecossistema. Devido ao reduzido número de estudos associados à avaliação da qualidade da água no local, este estudo teve como objetivo avaliar a situação atual desse ecossistema por meio do Índice de Proteção da Vida Aquática. Seis amostragens bimestrais foram realizadas em seis pontos ao longo da bacia, coletando parâmetros físico-químicos para a aplicação do Índice de Estado Trófico e Índice de Parâmetros Mínimos para a Proteção da Vida Aquática, relacionando-os com a resolução Conselho Nacional do Meio Ambiente - CONAMA/357. Os resultados revelaram alterações significativas na qualidade da água da BHRB, o aumento de nutrientes e das concentrações de clorofila-a conduziram ao um estado geral de eutrofização. Os valores de surfactantes foram altos colocando em risco a biota aquática e as taxas de oxigênio dissolvido estiveram abaixo do permitido pela resolução na maioria dos pontos amostrados. A situação atual da qualidade da água para proteção da vida aquática é péssima em 61,92% dos pontos amostrados.
Abstract Bacanga River Basin faces environmental problems related to urbanization and discharge of untreated domestic sewage, which compromise its ecosystem health. Due to the small number of studies that assessed its water quality, the present study aimed to assess the current status of this ecosystem based on the aquatic life protection index. Samples were carried out every two months, in a total of six events, in six sites along the basin, where the water samples were collected to assess physicochemical parameters and calculate the trophic state index and the index of minimum parameters for the protection of aquatic communities. The data were also compared with values determined by the resolution National Environment Council - CONAMA 357/05. Our results reveal significant changes in the water quality of Bacanga River Basin. An increase in nutrients and chlorophyll-a concentration led it to eutrophication. The surfactant values were high and put in danger the aquatic biota. Dissolved oxygen rates were below the values allowed by the resolution in most sites sampled. The current water quality is terrible for the protection of aquatic life in 61.92% of the sites sampled.

Influência de agentes clareadores nas propriedades superficiais (rugosidade e microdureza) de uma cerâmica odontológica

Resumo O objetivo desse estudo in vitro foi avaliar a influência de agentes clareadores na rugosidade superficial e na microdureza de uma cerâmica odontológica (porcelana). Foram confeccionadas 30 amostras de cerâmica odontológica de 2 mm de espessura e 6 mm de diâmetro de acordo com as instruções do fabricante. Posteriormente, as amostras foram aleatoriamente divididas em 3 grupos (n=10) e avaliadas quanto à rugosidade e à microdureza para obtenção dos valores iniciais. Dois grupos foram submetidos ao tratamento superficial com agentes clareadores (GII: peróxido de hidrogênio a 9,5% e GIII: peróxido de hidrogênio a 37,5%). O GI, que constituiu o grupo controle, ficou imerso em água destilada. O peróxido de hidrogênio a 9,5% foi aplicado 3 h/dia por 3 semanas. O agente mais concentrado foi utilizado 3 vezes ao dia, por 8 min durante 3 semanas (uma vez por semana). Ao fim de cada exposição ao agente clareador, os espécimes foram lavados e armazenados em água destilada. Os dados obtidos relativos à rugosidade e à microdureza dos espécimes cerâmicos foram registrados nos intervalos de tempo: inicial (baseline) e 3 semanas após a aplicação dos agentes clareadores. A rugosidade e a microdureza de superfície não foram alteradas de maneira estatisticamente significativa, considerando o tempo de clareamento, independente do grupo analisado (p> 0,05). Os valores médios da rugosidade (Ra em μm) foram: GI (0,084, 0,086, 0,078, 0,083), GII (0,085, 0,085, 0,086, 0,080) e GIII (0,079, 0,088, 0,083, 0,086). Os valores médios de microdureza (VHN em kgf/mm2) foram: GI (618, 589), GII (573, 578) e GIII (584, 600). Concluiu-se que as propriedades superficiais (rugosidade e microdureza) da cerâmica odontológica não foram alteradas pelo tratamento clareador. Novas pesquisas são necessárias para explicar o efeito do clareamento em cerâmicas odontológicas.
Abstract The objective of this in vitro study was to evaluate the effect of bleaching agents on the surface roughness and microhardness of a dental ceramic (porcelain). Thirty samples of dental ceramic, 2 mm thick and 6 mm in diameter, were prepared according to manufacturer's instructions, randomly divided into three groups of 10, and their initial baseline roughness and microhardness values recorded. Two groups were submitted to surface treatment with bleaching agents (GII, hydrogen peroxide 9.5%; and GIII, hydrogen peroxide 37.5%). The third group, GI, remained immersed in distilled water constituting the control group. GII group was exposed to the bleaching agent three hours a day for three weeks. As for the GIII group, the bleaching agent was applied three times a day, for eight minutes, during three weeks (once a week). After each exposure to the bleaching agent, the samples were washed and stored in distilled water. The roughness and microhardness data for the different ceramic specimens were collected at time intervals: initial (baseline) and every 3 weeks after bleaching agent applications. None of the bleaching treatments applied altered the roughness or the micro-hardness of the ceramic surfaces in any statistically significant way (p>0.05). Average roughness (Ra in μm) values registered were: for GI (0.084, 0.086, 0.078, 0,083); for GII (0.085, 0.085, 0.086, 0.080); and for GIII (0.079, 0.088, 0.083, 0.086). Average microhardness (VHN in kgf/mm2) values registered were: for GI (618, 589); for GII (573, 578); and for GIII (584, 600). It is concluded that the dental ceramic' surface properties (roughness and microhardness) were not altered by the bleaching treatments applied. Additional studies are necessary to understand and explain the effect of bleaching on dental ceramics.

Este artigo apresenta uma melhoria para a formulação de Sherlock e Monro para a parametrização wavelet para a obtenção das restrições que garantem três momentos nulos. A fim de testar a formulação apresentada, um exemplo de detecção de transitórios em sinais é apresentada.
This article presents an improvement to the formulation of Sherlock and Monro for the wavelet parameterization for the obtainment of the restrictions which ensure three vanishing moments. In order to test the formulation presented, a transient signal detection is presented.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Las degradation in a fluidized bed reactor and phylogenetic characterization of the biofilm

A fluidized bed reactor was used to study the degradation of the surfactant linear alkylbenzene sulfonate (LAS). The reactor was inoculated with anaerobic sludge and was fed with a synthetic substrate supplemented with LAS in increasing concentrations (8.2 to 45.8 mg l-1). The removal efficiency of 93% was obtained after 270 days of operation. Subsequently, 16S rRNA gene sequencing and phylogenetic analysis of the sample at the last stage of the reactor operation recovered 105 clones belonging to the domain Bacteria. These clones represented a variety of phyla with significant homology to Bacteroidetes (40%), Proteobacteria (42%), Verrucomicrobia (4%), Acidobacteria (3%), Firmicutes (2%), and Gemmatimonadetes (1%). A small fraction of the clones (8%) was not related to any phylum. Such phyla variety indicated the role of microbial consortia in degrading the surfactant LAS.

Biocompatible polymers and copolymers are frequently being used as part of controlled delivery systems. These systems can be prepared using a "clean and environment friendly" technology like supercritical fluids. One great advantage of this process is that compressed carbon dioxide has excellent plasticizing properties and can swell most biocompatible polymeric matrixes, thus promoting drug impregnation processes. Mass sorption of two acrylate biocompatible copolymers contact with supercritical carbon dioxide is reported. Equilibrium solubility of dense carbon dioxide in poly(methylmethacrylate-co-ethylhexylacrylate) and poly(methylmethacrylate-co-ethylhexylacrylate-co-ethyleneglycoldimethacrylate) was studied by a static method at 10.0 MPa and 313 K. The reticulated copolymer had Fickean behavior and its diffusion coefficient was calculated, under operating conditions.

Evaluation of biochemical and serological methods to identify and clustering yeast cells of oral Candida species by CHROMagar test, SDS-PAGE and ELISA

Este trabalho teve o propósito de avaliar métodos bioquímicos e sorológicos para serem aplicados na caracterização e identificação de linhagens do gênero Candida isoladas da cavidade bucal. As cepas empregadas representam cinco espécies de Candida previamente identificadas: C. albicans, C. guilliermondii, C. parapsilosis, C. krusei e C. tropicalis, utilizando como controle negativo Kluyveromyces marxianus. Foram empregadas as técnicas de gel de poliacrilamida (SDS-PAGE) associado à análise estatística em software, CHROMagar Candida (CA), meio cromogênico diferencial descrito para o isolamento e identificação presuntiva de leveduras de importância clínica e um ensaio de imunoabsorção ligado a enzima (ELISA), utilizando antissoro produzido contra extratos protéicos de uma linhagem-padrão de Candida e um isolado de cavidade oral de C. albicans. O método mostrou-se adequado para a identificação presuntiva de C. albicans, C. tropicalis e C. krusei, com 100% de sensibilidade e especificidade, com base na coloração e textura das colônias. O método de ELISA utilizando imunoglobulinas G purificadas apresentou alto teor de reação cruzada com as outras espécies de Candida estudadas. A análise do perfil protéico por SDS-PAGE permitiu agrupar os isolados da cavidade oral por intermédio do coeficiente "Simple Matching", S SM = 1,0. Os perfis protéicos analisados por SDS-PAGE ampliam os conhecimentos sobre as relações taxonômicas de leveduras isoladas da cavidade oral. Esta metodologia demonstra boa reprodutibilidade e origina informações úteis para aplicação clínica e estudos que envolvem a sistemática e a epidemiologia.
The purpose of this work was to evaluate biochemical and serological methods to characterize and identify Candida species from the oral cavity. The strains used were five Candida species previously identified: C. albicans, C. guilliermondii, C. parapsilosis, C. krusei, C. tropicalis, and Kluyveromyces marxianus, as a negative control. The analyses were conducted through the SDS-PAGE associated with statistical analysis using software, chromogenic medium, and CHROMagar Candida (CA), as a differential medium for the isolation and presumptive identification of clinically important yeasts and an enzyme-linked immunoabsorbent assay (ELISA), using antisera produced against antigens from two C. albicans strains. This method enabled the screening of the three Candida species: C. albicans, C. tropicalis, and C. Krusei, with 100% of specificity. The ELISA using purified immunoglobulin G showed a high level of cross-reaction against protein extracts of Candida species. The SDS-PAGE method allowed the clustering of species-specific isolates using the Simple Matching coefficient, S SM = 1.0. The protein profile analysis by SDS-PAGE increases what is known about the taxonomic relationships among oral yeasts. This methodology showed good reproducibility and allows collection of useful information for numerical analysis on information relevant to clinical application, and epidemiological and systematical studies.
