Abstract The present study investigated the motherhood experience of mothers of extremely preterm infants during their hospitalizations in the NICU. In particular, we sought to investigate the maternal feelings related to this context. Participants included 18 mothers whose babies were between 25 and 28 gestational weeks and weighed between 625g and 1000g. Mothers were interviewed 15 days after delivery and the responses were examined using qualitative content analysis. Three axes were developed, derived from the interviews: feelings about the NICU; feelings about the baby’s manifestations; and feelings about motherhood. The findings revealed the complexity of maternal feelings in the context of the NICU. However, the mothers’ narcissistic investment in their babies, which led them to communicate with them using small gestures and sensory expressions, stood out in the maternal verbalizations, which simultaneously made it possible to build a symbolic place for the infant.

Resumen El presente estudio investigó la experiencia de maternidad de madres de bebés nacidos extremadamente prematuros durante sus hospitalizaciones en la UCI Neonatal. En particular, se buscó conocer los sentimientos maternos relacionados con este contexto. Participaron 18 madres cuyos bebés tenían entre 25 y 28 semanas de gestación y pesaban entre 625 g y 1000 g. Las madres fueron entrevistadas 15 días después del parto y las respuestas se examinaron mediante análisis de contenido cualitativo. Se desarrollaron tres ejes, derivados de las entrevistas: sentimientos sobre la UCI Neonatal; sentimientos sobre las manifestaciones del bebé; y sentimientos sobre la maternidad. Los hallazgos revelaron una complejidad de los sentimientos maternos en el contexto de la UCI Neonatal. Sin embargo, la inversión narcisista de las madres en sus bebés se evidenció en sus verbalizaciones, que las llevaba a comunicarse con ellos a través de pequeños gestos y expresiones sensoriales que, simultáneamente, permitían la construcción de un lugar simbólico para el bebé.

Resumo O presente estudo investigou a experiência da maternidade de mães de bebês nascidos extremamente prematuros durante suas hospitalizações na UTI Neo. Em particular, buscou-se conhecer os sentimentos maternos relacionados a esse contexto. Participaram 18 mães cujos bebês tinham entre 25 e 28 semanas gestacionais e pesavam entre 625g e 1000g. As mães foram entrevistas 15 dias após o parto e as respostas foram examinadas por meio de análise de conteúdo qualitativa. Foram desenvolvidos três eixos, derivados das entrevistas: sentimentos sobre a UTI Neo; sentimentos sobre as manifestações do bebê; e sentimentos sobre a maternidade. Os achados revelaram uma complexidade de sentimentos maternos no contexto da UTI Neo. Porém, destacaram-se nas verbalizações maternas o investimento narcísico das mães nos seus bebês, que as levavam a se comunicar com eles a partir de pequenos gestos e expressões sensoriais, o que oportunizava, simultaneamente, a construção de um lugar simbólico para o bebê.

Abstract Haemonchus contortus is a constraint to sheep production. Seeking to reduce the use of hosts and produce parasitic stages in large-scale, a 42-day in vitro culture protocol of H. contortus third-stage larvae was optimized using Dulbecco’s modified Eagle’s medium (DMEM). In cell-free culture, larvae were maintained at 39.6°C, in acidic media (pH 6.1) for 3 or 6 days with Δ4-dafachronic acid followed by DMEM pH 7.4 supplemented or not with Fildes’ reagent. In DMEM pH 7.4 at 37°C, supplementation with Caco-2 cells was compared to Fildes. On Day 14, fourth-stage larvae (L4) development rates in acidic media supplemented (86.8-88.4%) or not (74.4-77.8%) with Fildes and in Caco-2 cell co-culture (92.6%) were similar, and superior to DMEM pH 7.4 with Fildes (0.0%). On Day 21, Caco-2 cell co-culture resulted in higher larvae differentiation (25.0%) and lower degeneration (13.9%) compared to acidic media (1.5-8.1% and 48.6-69.9%, respectively). This is the first report of prolonged in vitro culture of H. contortus larvae using commercial media in co-culture with Caco-2 cells. Although no progression to the adult stage, Caco-2 cell co-culture resulted in morphological differentiation of H. contortus L4 and larval viability for up to 28 days.

Resumo Haemonchus contortus provoca grandes prejuízos à ovinocultura. Visando reduzir a utilização de ovinos hospedeiros e produzir estágios parasitários em larga escala, um protocolo para o cultivo in vitro por 42 dias de larvas de terceiro estádio de H. contortus foi realizado em meio Eagle, modificado por Dulbecco (DMEM). No cultivo sem células, as larvas foram mantidas a 39,6°C e incubadas em DMEM ácido (pH 6,1) por 3 ou 6 dias com 4Δ-ácido dafacrônico seguido por DMEM pH 7,4 suplementado ou não com reagente de Fildes. Em DMEM pH 7,4 a 37°C, a suplementação com células Caco-2 foi comparada à suplementação com Fildes. No Dia 14, as taxas de desenvolvimento até o quarto estádio larvar (L4) foram similares em meio ácido sem células suplementado (86,8-88,4%) ou não (74,4-77,8%) com Fildes e em co-cultura com células Caco-2 (92,6%), e superiores ao desenvolvimento em DMEM pH 7,4 com Fildes (0,0%). No Dia 21, o co-cultivo com células Caco-2 resultou em maior diferenciação (25,0%) e menor degeneração (13,9%) das larvas em comparação ao meio ácido (1,5-8,1% e 48,6-69,9%, respectivamente). Este é o primeiro relato de cultivo in vitro prolongado de H. contortus em meio comercial em co-cultivo com células Caco-2. Apesar da ausência de progressão até o estágio adulto, o co-cultivo com células Caco-2 resultou em diferenciação de L4 e manutenção da viabilidade das larvas de H. contortus por até 28 dias in vitro.

RESUMO O uso de zeólita na suplementação alimentar pode melhorar a resistência e resiliência aos parasitas gastrintestinal. Os efeitos da suplementação nutricional com zeólita de cordeiros Santa Inês no desempenho, crescimento, infecção por nematoides gastrintestinais e análise econômica foram investigados. Animais fêmeas e machos de 110 dias foram suplementados com quatro níveis de zeólita: 0, 25, 50, e 75 gramas por dia em um delineamento experimental em blocos ao acaso. Os ovos por grama de fezes (OPG), proteína sérica total (PST) e volume globular (VG) foram obtidos de cada animal semanalmente. A cada 14 dias os animais foram pesados e as coproculturas preparadas. Foram abatidos os machos que atingiram peso vivo médio de 34 kg. A análise de viabilidade econômica das dietas foi baseada nos períodos de confinamento de 56 e 91 dias. A zeólita não mitigou significativamente (P > 0,05) a infecção por nematoides, não afetou a PST, o PCV e o desempenho dos cordeiros confinados. A zeólita não foi um suplemento dietético eficiente para evitar a infecção por nematoides gastrintestinais ou melhorar o desempenho dos cordeiros. Os machos abatidos aos 56 dias de confinamento apresentaram maior viabilidade econômica (P < 0,05) comparados àqueles abatidos aos 91 dias. Os machos confinados tiveram desempenho superior (P < 0,05) e melhores resultados econômicos (P < 0,05) comparados às fêmeas. O aumento do período de confinamento reduziu a lucratividade.

ABSTRACT Zeolite as a dietary supplement can improve animal resistance and resilience against gastrointestinal parasites. The effects of different levels of zeolite supplementation on gastrointestinal nematode infection, growth performance, and economic viability of male and female Santa Ines lambs were investigated. Four zeolite levels (0, 25, 50, and 75 g per day) were fed to 110 day-old animals in a randomized block experimental design. Eggs per gram of feces (EPG), total serum protein (TSP), and packed cell volume (PCV) were measured individually every week. The animals were weighed every 14 days and coprocultures were prepared. Male animals that reached an average live weight of 34 kg were harvested. The economic viability analysis of the experimental diets was based on feedlot periods of 56 and 91 days. Dietary supplementation with zeolite did not significantly mitigate infections by nematodes and did not affect TSP, PCV, or feedlot lamb growth performance. Therefore, zeolite was not an effective (P > 0.05) dietary supplement for avoiding gastrointestinal nematode infection or improving growth performance. Male animals slaughtered after 56 days of feeding were more economically viable (P < 0.05) than those slaughtered after 91 days. Feedlotting demonstrated that male animals had superior performance (P < 0.05) and better economic (P < 0.05) results than female animals.

RESUMO As leucemias são caracterizadas pela proliferação de células imaturas da linhagem branca, denominadas blastos, na medula óssea e/ou sangue. Dentre elas destacamos a Leucemia Mielóide Aguda Monocítica, que representa uma neoplasia maligna, cuja proliferação anormal de monoblastos resulta na supressão das células da série mielóide. Manifestações orais são comuns, surgindo no início da doença e também associadas à quimioterapia durante o tratamento gerando complicações que comprometem a oncoterapia. O presente trabalho trata-se de um paciente portador deste tipo agressivo de leucemia, com complicações bucais da doença e posteriormente, do tratamento oncológico. Destaca ainda a atuação do cirurgião-dentista em fases distintas do tratamento realçando a importância da manutenção da saúde bucal em pacientes oncológicos. A presença do Cirurgião-Dentista na equipe multiprofissional contribui para diminuição das manifestações que ocorrem tanto devido à doença quanto ao tratamento, garantindo melhor qualidade de vida do paciente oncológico e evitando maiores gastos ao sistema de saúde.

ABSTRACT Leukemia’s are characterized by the proliferation of immature white blood cells, called blasts, in the bone marrow and / or blood. Among them, we highlight the Monocytic Acute Myeloid Leukemia, which represents a malignant neoplasm whose unnatural monoblast proliferation results in suppression of myeloid series cells. Oral manifestations are common, arising at the onset of the disease and are also associated with chemotherapy during treatment leading to complications that compromise oncotherapy. The present study deals with a patient with this type of aggressive leukemia, with oral complications of the disease and later, oncological treatment. It also give prominence the performance of the dental surgeon in distinct phases of the treatment emphasizing the importance of maintaining oral health in cancer patients. The presence of the Dentist in the multi-professional team contributes to the reduction of manifestations that occur both due to the disease and the treatment, ensuring a better quality of life for cancer patients and avoiding higher expenses to the health system.

Resumo A implantação dos Núcleos Ampliados de Saúde da Família e Atenção Básica (NASF) fomentou estratégias de cuidados mais dialógicas e o acompanhamento de grupos populacionais que, historicamente, tinham pouco acesso à Atenção Primária à Saúde (APS). No município do Rio de Janeiro, oscilações na gestão federal e municipal implicaram redução ou eliminação dessas equipes. Para apreender os impactos desse cenário no cotidiano das equipes do NASF, esta pesquisa analisou documentos normativos e a percepção dos profissionais a respeito da organização do trabalho. Para tanto, realizou-se pesquisa qualitativa que utilizou coleta de dados, análise documental e entrevistas semiestruturadas com oito profissionais do NASF de uma região do Rio de Janeiro, entre agosto de 2019 e fevereiro de 2020. O tratamento dos dados baseou-se na análise de conteúdo. Os resultados expõem os impactos das mudanças nas diretrizes de organização e financiamento do NASF e abordam a percepção dos trabalhadores sobre o processo de trabalho. Assim, evidenciou-se que o NASF apresenta grande potencial de qualificação das ações e intervenções no contexto da APS, porém, a crescente precarização do trabalho sobrecarrega os trabalhadores e dificulta o alcance de seus objetivos originais.

Abstract The implementation of Extended Family and Primary Health Care Centers (EFPHCC) has fostered more dialogic care strategies and the monitoring of population groups that, historically, had little access to Primary Health Care (PHC). However, in the municipality of Rio de Janeiro, oscillations in federal and municipal management implied the reduction or elimination of these teams. To understand the impacts of this scenario on the daily routine of EFPHCC teams, this research analyzed regulatory documents and the professionals' perception of the work organization. Accordingly, a qualitative study was conducted using data collection, document analysis, and semi-structured interviews with eight EFPHCC professionals from a region of Rio de Janeiro between August 2019 and February 2020. Data treatment was based on content analysis. The results expose the impacts of the changes in the organization and financing guidelines of EFPHCC and address the workers' perception of the work process. Thus, it was evident that EFPHCC has the potential to qualify actions and interventions in the context of PHC; however, the growing job insecurity overloads the workers and hinders the achievement of its original goals.

Resumo Este estudo avaliou o impacto econômico da infecção por nematoides gastrintestinais (NGI), em cordeiros Morada Nova, sob diferentes condições de controle químico dos parasitas. Para isso, 246 cordeiros, na estação chuvosa e seca, foram randomizados em grupos de acordo com o tratamento com levamisol: controle (TC: sem tratamento); tratamento rotineiro (TR: tratado a cada 42 dias); e tratamento seletivo direcionado (TST: tratado de acordo com o ganho de peso médio diário, GMD). Dos 63 dias de idade (D63) ao D210, os cordeiros foram pesados e monitorados quanto aos parâmetros de infecção por NGI. O gasto com anti-helmínticos no sistema produtivo foi de 1,3% do resultado econômico total. O resultado econômico por animal (R$ 5,00 = US$ 1,00) foi maior no grupo RT, totalizando US$ 6,60 na estação chuvosa e US$ 5,69 na seca, devido ao maior GMD. Assim, o RT apresentou resultados econômicos 14,4% e 10,9% superiores ao TC, e 7,2% e 1,9% superiores ao TST, no período chuvoso e seco, respectivamente. Entretanto o rápido desenvolvimento de resistência inviabiliza o TR. O impacto econômico da infecção por NGI em escala nacional são aqui discutidos, demonstrando sua importância e a impossibilidade de uma ovinocultura lucrativa e sustentável sem o controle adequado.

Abstract This study evaluated the economic impact of gastrointestinal nematode (GIN) infection in Morada Nova lambs under different parasite chemical control conditions. For this, 246 lambs, in the rainy and dry season, were randomized into groups according to their anthelmintic treatment with levamisole: control (CT: no treatment); routine treatment (RT: treated every 42 days); and targeted selective treatment (TST: treated according to the average daily weight gain, DWG). From 63 days of age (D63) to D210, the lambs were weighed and monitored for GIN infection parameters. Spending on anthelmintics in the production system was 1.3% of the total economic result. The economic result per animal (R$ 5.00 = US$ 1.00) was higher in the RT group, amounting to US$ 6.60 in the rainy and US$ 5.69 in the dry season, due to higher DWG. Thus, RT presented economic results 14.4% and 10.9% higher than CT, and 7.2% and 1.9% higher than TST, in the rainy and dry season, respectively. However, fast development of resistance made RT unfeasible. Here, the economic impact of GIN infection on a national scale is discussed, demonstrating its importance and the impossibility of profitable and sustainable sheep production without adequate control.

Resumo Objetivos: Avaliar os conhecimentos e atitudes relativos à saúde oral na gravidez e na infância de uma amostra de grávidas de Coimbra, Portugal. Métodos: Um questionário auto-aplicável foi administrado a uma amostra aleatória de grávidas em seguimento pré-natal em duas instituições de saúde públicas de Coimbra. Colheram-se dados sociodemográficos e referentes aos conhecimentos e práticas de saúde oral na gravidez e relativos ao bebé. Todos os requisitos éticos foram cumpridos. Procedeu-se a análise estatística descritiva e inferencial dos resultados. Verificou-se a independência das variáveis através do teste de qui-quadrado para um intervalo de confiança de 95%. Resultados: A amostra incluiu 120 grávidas. Apesar de 68.9% das participantes ter revelado realizar escovagem bi-diária, 36.4% não usava fio dentário, verificando-se uma associação estatisticamente significativa deste hábito com a idade (p=0.004). Metade das inquiridas não visitou o médico dentista antes da gravidez e 59.2% considerava que a gravidez era potencialmente danosa para a saúde oral. Foram evidenciadas algumas lacunas no conhecimento relativamente às possíveis consequências de gengivite ou periodontite no desfecho da gravidez e acerca da importância da dieta para a saúde oral. Destacou-se também falta de informação no que respeita à cárie precoce de infância e à sua prevenção. Conclusões: Em geral, as grávidas apresentaram conhecimentos e comportamentos de saúde oral insuficientes. Atendendo a que a gravidez constitui uma fase de particular interesse para a aquisição de conhecimentos e modificações comportamentais decisivos para a gestante e bebé, os presentes resultados suportam a necessidade de implementar programas de saúde oral pré-natal nesta população.

Abstract Objectives: To assess the level of knowledge and attitudes regarding prenatal and infant oral health in a sample of pregnant women from Coimbra, Portugal. Methods: A self-applied questionnaire was administered to randomly selected pregnant women who attended prenatal check-ups at two public health institutions in Coimbra. Collected data included sociodemographic information, oral health knowledge and practices in pregnancy, and knowledge on oral health in children. All ethical requirements were met. Statistical analysis was conducted using descriptive and inferential methods. Variables were tested for independence using a chi-square test with a 95% confidence interval. Results: A total of 120 women enrolled in the study. Although 68.9% of participants brushed their teeth twice daily, 36.4% reported not using floss, with a statistically significant association with age (p=0.004). Half of the respondents had not attended a dental appointment before pregnancy, and 59.2% believed pregnancy could be harmful to oral health. Findings showed limited knowledge of the possible consequences of gingivitis and periodontitis to the course of pregnancy and the importance of diet in oral health. Moreover, a low level of understanding was noted regarding the existence and prevention of early childhood caries. Conclusions: Oral health-related knowledge and practices of surveyed women were in general deficient. Considering pregnancy is a period of particular interest for acquiring knowledge and good oral health practices, which are decisive for both the expectant mother na the child, our results highlight an urgent need to implement prenatal oral health care programs in this study population.

Abstract Objective To describe the clinical, laboratory, and radiological characteristics, as well as the outcomes of children with MIS-C. Method Multicenter, prospective cohort study, conducted in 17 pediatric intensive care units in five states in Brazil, from March to July 2020. Patients from 1 month to 19 years who met the MIS-C diagnostic criteria were included consecutively. Results Fifty-six patients were included, with the following conditions: Kawasaki-like disease (n = 26), incomplete Kawasaki disease (n = 16), acute cardiac dysfunction (n = 10), toxic shock syndrome (n = 3), and macrophage activation syndrome (n = 1). Median age was 6.2 years (IQR 2.4−10.3), 70% were boys, 59% were non-whites, 20% had comorbidities, 48% reported a contact with COVID-19 cases, and 55% had a recent SARS-CoV-2 infection confirmed by RT-PCR and/or serology. Gastrointestinal symptoms were present in 71%, shock symptoms in 59%, and severe respiratory symptoms in less than 20%. -Dimer was increased in 80% and cardiac dysfunction markers in more than 75%. Treatment included immunoglobulin (89%); corticosteroids, antibiotics, and enoxaparin in about 50%; and oseltamivir and antifungal therapy in less than 10%. Only 11% needed invasive mechanical ventilation, with a median duration of five days (IQR 5–6.5). The median length of PICU stay was six days (IQR 5–11), and one death occurred (1.8%). Conclusions Most characteristics of the present MIS-C patients were similar to that of other cohorts. The present results may contribute to a broader understanding of SARS-CoV-2 infection in children and its short-term consequences. Long-term multidisciplinary follow-up is needed, since it is not known whether these patients will have chronic cardiac impairment or other sequelae.

This work presents some modifications in the post-synthetic processing for a classical arsenic reagent: AsI3. In comparison with the widely used analog, the trichloride, arsenic triiodide presents several advantages such as low toxicity, air stability, and low volatility. It was used as a synthetic precursor in the preparation of a variety of arsenic(III) derivatives like arsines, arsenites, and thioarsenites. Besides that, AsI3 was submitted to a diversity-oriented Grignard reaction in the preparation of a homologous series of trialkyl arsines ranging from AsC3H9 to AsC12H27. The series was analyzed by comprehensive two-dimensional gas chromatography coupled with time-of-flight mass spectrometry to provide a trialkyl arsines library that can be used for the direct analysis of natural samples.

OBJECTIVES: This observational, cross-sectional study based aimed to test whether heart failure (HF)-disease management program (DMP) components are influencing care and clinical decision-making in Brazil. METHODS: The survey respondents were cardiologists recommended by experts in the field and invited to participate in the survey via printed form or email. The survey consisted of 29 questions addressing site demographics, public versus private infrastructure, HF baseline data of patients, clinical management of HF, performance indicators, and perceptions about HF treatment. RESULTS: Data were obtained from 98 centers (58% public and 42% private practice) distributed across Brazil. Public HF-DMPs compared to private HF-DMP were associated with a higher percentage of HF-DMP-dedicated services (79% vs 24%; OR: 12, 95% CI: 94-34), multidisciplinary HF (MHF)-DMP [84% vs 65%; OR: 3; 95% CI: 1-8), HF educational programs (49% vs 18%; OR: 4; 95% CI: 1-2), written instructions before hospital discharge (83% vs 76%; OR: 1; 95% CI: 0-5), rehabilitation (69% vs 39%; OR: 3; 95% CI: 1-9), monitoring (44% vs 29%; OR: 2; 95% CI: 1-5), guideline-directed medical therapy-HF use (94% vs 85%; OR: 3; 95% CI: 0-15), and less B-type natriuretic peptide (BNP) dosage (73% vs 88%; OR: 3; 95% CI: 1-9), and key performance indicators (37% vs 60%; OR: 3; 95% CI: 1-7). In comparison to non- MHF-DMP, MHF-DMP was associated with more educational initiatives (42% vs 6%; OR: 12; 95% CI: 1-97), written instructions (83% vs 68%; OR: 2: 95% CI: 1-7), rehabilitation (69% vs 17%; OR: 11; 95% CI: 3-44), monitoring (47% vs 6%; OR: 14; 95% CI: 2-115), GDMT-HF (92% vs 83%; OR: 3; 95% CI: 0-15). In addition, there were less use of BNP as a biomarker (70% vs 84%; OR: 2; 95% CI: 1-8) and key performance indicators (35% vs 51%; OR: 2; 95% CI: 91,6) in the non-MHF group. Physicians considered changing or introducing new medications mostly when patients were hospitalized or when observing worsening disease and/or symptoms. Adherence to drug treatment and non-drug treatment factors were the greatest medical problems associated with HF treatment. CONCLUSION: HF-DMPs are highly heterogeneous. New strategies for HF care should consider the present study highlights and clinical decision-making processes to improve HF patient care.

Resumo Este estudo avaliou o status de resistência de Haemonchus contortus em rebanhos ovinos do estado de São Paulo, Brasil, através de um estudo comparativo entre o teste de redução da contagem de ovos nas fezes (TRCOF) e o teste de desenvolvimento larvar (TDL). Para o TRCOF, 35 ovinos foram selecionados em cada um dos cinco rebanhos e divididos em grupos tratados com: benzimidazol, levamisol, ivermectina, monepantel e controle. Fezes foram coletadas para OPG e coproculturas. O TDL foi realizado com tiabendazol-TBZ, levamisol-LEV, ivermectina aglicona-IVM-A e Zolvix-ZLV. Resistência a todos os fármacos foi detectada por meio do TRCOF em 100% dos rebanhos, exceto para ZLV (40% resistentes e 20% suspeitos de resistência). O LDT indicou resistência ao TBZ e IVM-A em todos os rebanhos; em 80% dos rebanhos ao LEV e 10% ao ZLV. Foi obtida concordância total entre os dois testes para TBZ e IVM (k = 1,0), enquanto para LEV (k = 0,8) e ZLV (k = 0,9) foram obtidas concordância substancial e quase perfeita, respectivamente. A concordância entre os testes foi significante, sendo possível usar o resultado do LDT para prever o do TRCOF, validando-se o primeiro como um teste diagnóstico rápido para criadores de ovinos no Brasil.

Abstract This study evaluated the resistance status of Haemonchus contortus from sheep flocks in the state of São Paulo, Brazil, through comparison between the fecal egg count reduction test (FECRT) and the larval development test (LDT). For the FECRT, 35 sheep were selected in each of five flocks and divided into groups treated with: benzimidazole, levamisole, ivermectin, monepantel and control. Feces were collected for EPG and fecal cultures. The LDT was performed using thiabendazole (TBZ), levamisole (LEV), ivermectin aglycone (IVM-A) and Zolvix (ZLV). Resistance to all drugs was detected using FECRT in 100% of the flocks, except in relation to ZLV (40% resistant and 20% suspected of resistance). LDT indicated resistance to TBZ and IVM-A in all flocks, to LEV in 80% of flocks and to ZLV in 10%. Total agreement was obtained between the two tests for TBZ and IVM (k = 1.0), while for LEV (k = 0.8) and ZLV (k = 0.9), substantial and almost perfect agreement were obtained, respectively. The concordance between the tests was significant, thus showing that it is possible to use the outcome of the LDT to predict the FECRT, and hence validating the former as a fast diagnostic test for use by sheep farmers in Brazil.

Resumo Objetivos Avaliar e comparar as características histológicas de meniscos frescos e meniscos congelados armazenados em banco de tecidos por 1 mês e por 5 anos. Métodos Foi feito um estudo histológico com enxertos meniscais. Avaliamos 10 meniscos, sendo 2 que ficaram armazenados sob congelamento por 5 anos, 4 armazenados congelados por 1 mês, e 4 frescos, recém captados. Foram feitos cortes histológicos corados com hematoxilina e eosina e Tricrômico de Masson, para avaliação das propriedades histológicas. Resultados Os meniscos congelados por 1 mês apresentaram preservação parcial da estrutura das fibras colágenas, sem degeneração hidrópica significativa do tecido. Nos meniscos congelados por 5 anos, observamos dissociação evidente das fibras colágenas, com presença de múltiplos focos de degeneração hidrópica. Discussão Encontramos degeneração bem mais significativa nos meniscos armazenados por 5 anos, o que indica que o longo período de congelamento leva à progressão significativa da degeneração do tecido. Isto pode sugerir que o período de 5 anos, considerado período máximo que o enxerto pode permanecer armazenado antes de ser transplantado, é um período muito longo. Conclusão Nos enxertos armazenados por 1 mês, existiu apenas discreta alteração degenerativa das fibras colágenas, enquanto que nos meniscos com 5 anos de congelamento foi observada degeneração significativa do tecido. Abstract Objectives The present paper aims to evaluate and compare the histological features of fresh and frozen menisci stored in a tissue bank for 1 month and for 5 years. Methods The meniscal grafts were subjected to a histological study. A total of 10 menisci were evaluated; 2 were frozen for 5 years, 4 were frozen for 1 month, and 4 were fresh, recently harvested specimens. Histological properties were evaluated in sections stained with hematoxylin and eosin and Masson trichrome methods. Results The menisci frozen for 1 month showed partially preserved collagen fiber structure and no significant hydropic tissue degeneration. The menisci frozen for 5 years presented an evident dissociation of collagen fibers and multiple foci of hydropic degeneration. Discussion Degeneration was much more significant in menisci stored for 5 years, indicating that a long freezing period results in substantial progression of tissue deterioration. This may suggest that the 5-year period, considered the maximum time for graft storage before transplant, is too long. Conclusion Grafts stored for 1 month showed a slight degenerative change in collagen fibers, whereas menisci frozen for 5 years presented significant tissue degeneration.

Abstract Willing to increase the shelf life of the food matrix, studies regarding new preservations methods are developed to preserve fish deterioration. The application of natural additives as preservatives is increasingly common, with a current emphasis on the growing use of protein hydrolysates, in substitution to artificial ones. These compounds have been the subject of recent studies, focusing on determining their functional properties and the best form of applying peptide chains. The present study aimed to evaluate the effects of the protein hydrolysate obtained by the enzymatic hydrolysis of commercial fish gelatin added to rainbow trout fillets (Oncorhynchus mykiss) packed under vacuum and maintained under refrigeration. The fillets were distributed into three sample groups (controls and hydrolysate addition at 1:10 and 1:1 ratios). Microbiological and physico-chemical assessments were carried out. The results were compared and correlations observed regarding fish preservation assessments. A relationship between pH, redox potential and N-TVB was verified when compared to microbial development, especially with regard to psychrotrophic aerobic heterotrophic bacteria count. It was concluded that the application of protein hydrolysate as an additive is promising and can be effective in the control of food quality and maintenance of freshness for a longer period.
