RESUMO A detecção e identificação molecular de vírus são fundamentais para se definir estratégias de controle de viroses, tendo uma importancia crucial para vírus transmitidos por moscas-brancas. Plantas de algodoeiro (Gossypium) com sintomas de mosaico e redução de produtividade foram observadas em cultivares comerciais (G. hirsutum) e em acessos do banco de germoplasma de algodão da Embrapa mantidos em campo (G. hirsutum, G. barbadense, G. mustelinum). A extração de DNA a partir de plantas com sintomas de mosaico e clorose internerval e amplificação do genoma de begomovírus com primers degenerados e universais mostrou a associação com um begomovírus. Este vírus foi identificado como um isolado de sida micrantha mosaic virus (SiMMV) por sequenciamento do amplicon. O vírus não pode ser transmitido para algodão por mosca-branca (Bemisia tabaci MEAM1) quando plantas de algodão foram usadas como fonte de inóculo, em casa de cultivo protegido, sugerindo uma complexa interação entre vírus, plantas e vetores.

ABSTRACT Detection and molecular identification of viruses are fundamental to define control strategies against viral diseases, particularly for whitefly-transmitted viruses. Cotton (Gossypium) plants showing leaf mosaic symptoms and yield reduction were observed in commercial cultivars (G. hirsutum) and in plants of the cotton germplasm collection of Embrapa maintained in the field (G. hirsutum, G. barbadense, G. mustelinum). DNA was extracted from cotton plants with symptoms of mosaic and interveinal chlorosis, and a begomovirus-specific genome was amplified with degenerated universal primers, which indicated their association with a begomovirus. This virus was identified as an isolate of sida micrantha mosaic virus (SiMMV) after the amplicon sequencing. The virus could not be transmitted by whiteflies (Bemisia tabaci MEAM1) to cotton plants when the latter were used as inoculum source under protected cultivation house, suggesting a complex interaction among viruses, plants and vectors.

O objetivo deste trabalho foi avaliar o efeito do feijoeiro geneticamente modificado quanto à resistência ao Bean Golden Mosaic Vírus, BGMV (Olathe M1-4), sobre organismos não alvo. De um experimento implantado no campo, em delineamento inteiramente casualizado, com dois tratamentos (Olathe Pinto e evento elite Olathe M1-4), dois períodos amostrais (estádio V4 e R6) e dez repetições, obtiveram-se células bacterianas cultivadas e não cultivadas da rizosfera e do solo não rizosférico, para as quais se procedeu à extração de DNA total. A região V6-V8 do 16S rDNA foi amplificada para a comunidade bacteriana total, e também realizou-se amplificação com iniciadores específicos para o subgrupo alfa (α) do filo Proteobacteria a partir de células não cultivadas. Foram obtidos dendrogramas comparativos entre a variedade Olathe Pinto (convencional) e o evento elite Olathe M1-4 (geneticamente modificado) utilizando-se o coeficiente de Jaccard e o método UPGMA (Unweighted pair-group method with arithmetic mean). Os agrupamentos obtidos dos perfis de 16S rDNA PCR-DGGE indicam alterações na comunidade bacteriana da rizosfera em função da transformação das plantas são mais notáveis nos perfis obtidos para alfa-proteobacteria. A origem das amostras e o estágio de desenvolvimento das plantas afetam a comunidade bacteriana.

The objective of this work was to evaluate the effect of genetically modified common bean for Bean Golden Mosaic Virus, BGMV, resistance (Olathe M1-4) on nontarget organisms. In a field experiment established in a completely randomized design with two treatments (Olathe Pinto cultivar and M1-4 Olathe elite event), two sampling periods (V4 and R6 stages) and ten replicates, cultivated and non-cultivated bacterial cells from rhizosphere soil and bulk soil were obtained, and their total DNA was extracted. The V6-V8 region of 16S rDNA was amplified for the whole bacterial community, and primers specific for the alpha (α) subgroup of the Proteobacteria phylum were obtained from uncultured cells and used for amplification. Using the Jaccard coefficient and UPGMA (Unweighted pair-group method with arithmetic mean), dendrograms comparing the conventional Olathe Pinto and the elite event Olathe M1-4 transgenic varieties were obtained. The clusters obtained from the 16S rDNA PCR-DGGE profiles indicate changes in the rhizosphere bacterial community in genetically modified plants, being more notable in the profiles obtained for alphaproteobacteria. Sample origin and plant development stages affect bacterial community profiles.

O objetivo do presente trabalho foi determinar a herança e a estabilidade de transgenes de uma linhagem de feijoeiro-comum com expressão dos genes rep-trap-ren, do Bean golden mosaic virus, e do gene bar. Foram realizados cruzamentos entre a linhagem transgênica e quatro cultivares comerciais de feijão, seguidos de quatro retrocruzamentos. As progênies de cada cruzamento foram avaliadas quanto à presença dos transgenes, com aplicação do glifosinato de amônia nas folhas e por meio da reação da polimerase em cadeia com uso de oligonucleotídeos específicos. O vírus do mosaico comum necrótico do feijoeiro, Bean common mosaic necrosis virus (BCMNV), foi inoculado mecanicamente nas gerações avançadas. Os transgenes foram herdados em padrão mendeliano nos quatro cruzamentos estudados. As linhagens analisadas apresentaram cerca de 80% das características do parental recorrente, conforme determinado por análises com uso de marcadores de DNA, além de manter caracteres importantes, tais como resistência ao BCMNV. A presença do transgene não causou efeitos indesejáveis que pudessem ser detectados nas progênies avaliadas.

The objective of the present work was to determine the inheritance and stability of transgenes of a transgenic bean line expressing the genes rep-trap-ren from Bean golden mosaic virus and the bar gene. Crosses were done between the transgenic line and four commercial bean cultivars, followed by four backcrosses to the commercial cultivars. Progenies from each cross were evaluated for the presence of the transgenes by brushing the leaves with glufosinate ammonium and by polymerase chain reaction using specific oligonucleotides. Advanced generations were rub-inoculated with an isolate of Bean common mosaic necrosis virus (BCMNV). The transgenes were inherited consistently in a Mendelian pattern in the four crosses studied. The analyzed lines recovered close to 80% of the characteristics of the recurrent parent, as determined by the random amplified DNA markers used, besides maintaining important traits such as resistance to BCMNV. The presence of the transgene did not cause any detectable undesirable effect in the evaluated progenies.

A brusone é um dos fatores limitantes para a produção de arroz de várzea no Estado do Tocantins. A diversidade fenotípica e genética em população de Pyricularia grisea foi estudada, em amostras coletadas em quatro lavouras da cultivar Metica-1, sob condições de epidemias de brusone nas folhas. Foram testados 87 isolados em 32 genótipos, incluindo oito diferenciadoras internacionais. Considerando 80% de similaridade em virulência, dois grupos compostos por 81 isolados foram identificados, independentemente das lavouras onde foram coletados. Oitenta por cento dos isolados pertencem ao patótipo ID-14, indicando alta especificidade da cultivar e a diversidade estreita em virulência na população estudada. A virulência da população do patógeno nas cultivares comerciais BR-IRGA 409 e Rio Formoso foi baixa. A análise de isolados de P. grisea utilizando rep-PCR com dois primers Pot2 gerou de uma a nove bandas. A análise de agrupamento revelou a ocorrência de seis grupos ou linhagens com similaridade variando de 0,09 a 1. Não houve uma relação clara entre a virulência dos isolados dos 32 genótipos e o agrupamento baseado na análise usando rep-PCR de Pot2 de isolados de P. grisea coletados na cultivar Metica-1.

Rice blast is a major yield constraint of the irrigated rice in the State of Tocantins, Brazil. The objective of this investigation was to study the phenotypic and genetic diversity within the pathogen population of Pyricularia grisea in samples collected from four individual farms of rice cultivar Metica-1, under epidemic conditions of leaf blast. A set of 87 isolates was tested on 32 rice genotypes including eight international differentials. Considering 80% similarity in virulence, two groups comprising a total of 81 isolates were recognized, independently of the farms from which they were collected. Eighty percent of the isolates pertained to pathotype ID-14, indicating high cultivar specificity and narrow diversity of virulence in the sample population. The virulence in pathogen population on rice cultivars BR-IRGA 409 and Rio Formoso was low. Analysis of P. grisea isolates using rep-PCR with two primer sequences from Pot2 generated fingerprint profiles of one to nine bands. Cluster analysis revealed the occurrence of six fingerprint groups with similarities ranging from 0.09 to 1. There was no straight relationship between virulence of the isolates based on reaction pattern on 32 genotypes and grouping based on Pot2 rep-PCR analysis of P. grisea isolates collected from 'Metica-1'.

O mosaico dourado é a principal doença virótica da cultura do feijoeiro (Phaseolus vulgaris L.) na América Latina. A genética da resistência ao vírus do mosaico dourado do feijoeiro (BGMV), isolado do Brasil, foi conduzida em um dialelo 4 x 4 sem os recíprocos, entre os parentais DOR 303, EMGOPA 201 Ouro, Carnaval e Redlands Greenleaf C. Plântulas com oito dias de idade, dos pais, híbridos F1, retrocruzamentos e geração F2, foram inoculadas através da exposição a uma população virulífera de moscas brancas (Bemisia tabaci Genn.) por 24 h, antes do transplantio para condição de campo. O grupo completo de dois parentais, F1, F2 e respectivos retrocruzamentos foi considerado uma família. Foram coletados e analisados os dados de amarelecimento foliar, nanismo e de deformação de vagens, usando o delineamento experimental de blocos ao acaso com três repetições. Análise ponderada de médias de gerações foi executada para cada uma das seis famílias. Foram detectados efeitos gênicos aditivos significantes controlando todos os caracteres avaliados. Por outro lado, na maioria dos casos, houve predominância da ação gênica não aditiva. A resistência ao amarelecimento foliar, conferida por genes de DOR 303, foi altamente herdável e se expressou igualmente bem nos diferentes 'backgrounds' genéticos avaliados. Ela pode ser oligogênica. Estimativas das herdabilidades no sentido amplo e restrito foram relativamente altas para as três características. O maior coeficiente de correlação foi obtido entre nanismo e deformação de vagens. As três características estudadas foram todas positivamente correlacionadas, indicando que podem ser selecionadas simultaneamente.

Bean golden mosaic is the most important viral disease of the bean crop (Phaseolus vulgaris L.) in Latin America. The genetics of resistance to a Brazilian strain of bean golden mosaic virus (BGMV), was studied in a 4 x 4 diallel cross without reciprocals, among the parental genotypes DOR 303, EMGOPA 201 Ouro, Carnaval, and Redlands Greenleaf C. Seedlings of the four parents, six F1 hybrids, 12 backcrosses, and F2 generations for each combination were inoculated on the eighth day after sowing by exposure to a viruliferous whitefly (Bemisia tabaci Genn.) population for 24 h, in a glasshouse, prior to transplantation to field conditions. The full set of two parents, F1, F2 and respective backcrosses for each combination was considered to be a family. Data were recorded and analyzed for foliar yellowing, plant dwarfing, and pod malformation, using a randomized block design, with two replications. Weighted generation mean analysis was performed for each of the six families. An additive gene action model was significant for the three characteristics evaluated. On the other hand, non-additive gene action had greater absolute value in most cases. Resistance to foliar yellowing conferred by genes from DRO 303 was highly heritable and was expressed equally well in the different genetic backgrounds evaluated. Such resistance may be oligogenic. Broad- and narrow-sense heritabilities were relatively high for all response traits. The three traits studied were all positively correlated, indicating that they can be simultaneously selected for enhancement. The highest correlation coefficient was obtained for dwarfing x pod malformation.
