RESUMO: As micoplasmoses ocasionam prejuízos econômicos nas criações de ovinos e caprinos, e estão associados com quadros de mastite, poliartrite, agalaxia, conjuntivite, pneumonia e falhas reprodutivas. Objetivou-se neste estudo determinar a ocorrência de Mycoplasma agalactiae em amostras de leite e investigar os principais fatores de risco associados à infecção em caprinos provenientes de propriedades rurais do estado da Paraíba, Brasil. Para o diagnóstico de Mycoplasma agalactiae, foram analisadas 251 amostras de leite, que foram submetidas à extração do DNA genômico usando um kit comercial, seguindo as recomendações do fabricante. Para diagnóstico da infecção utilizou-se a Reação em Cadeia da Polimerase (PCR). Além disso, foram aplicados questionários para identificar os principais fatores de risco associados infecção à agalaxia contagiosa. Das 251 amostras analisadas, 50 (19,9%; I.C. 15,1-25,4%) foram positivas na PCR para M. agalactiae. Observaram-se na análise dos fatores de risco, algumas associações para as seguintes variáveis: tamanho do rebanho (P<0,001; OR 7,1), reposição de animais da propriedade (P<0,001; OR 4,7) e participação dos animais em feiras e exposições (P= 0,029; OR 2,0). Os resultados permitiram confirmar a ocorrência do Mycoplasma agalactiae em amostras de leite de caprinos da Paraíba. Portanto, é necessário o monitoramento dos rebanhos caprinos leiteiros e a conscientização dos produtores rurais para a importância econômica da doença, visto que a mesma acarreta severos prejuízos econômicos para os produtores do estado. A identificação dos fatores de risco são imprescindíveis para a adoção de medidas de controle e para a correção dos fatores de manejo em propriedades que tenham animais com diagnóstico positivo, evitando assim, a disseminação do patógeno.

ABSTRACT: Mycoplasmosis is a disease that may cause severe economical losses in goat and sheep herds, and it is associated with mastitis, polyarthritis, agalactia, conjunctivitis, pneumonia and reproductive failure. The objective of this study was to determine the occurrence of Mycoplasma agalactiae in milk samples and investigate the main risk factors associated with infection in goats from farms of the state of Paraíba, Brazil. For Mycoplasma agalactiae diagnosis, 251 milk samples were submitted to DNA extraction using a commercially available kit, following the manufacturer’s instructions and Polymerase Chain Reaction (PCR) was performed. In addition, questionnaires were applied to identify the main risk factors associated with contagious agalactia. Out of the two hundred fifty-one samples analyzed, 50 (19.9%, I.C. 15.1-25.4%) were PCR positive for M. agalactiae. In the risk factors analysis, some associations were observed for the following variables: size of the herd (P<0.001, OR=7.1, I.C. 2.4-20.6), replacement of farm animals (P<0.001, OR=4.7, I.C. 1.8-12.2) and participation of animals in fairs and exhibitions (P=0.029, OR=2.0, I.C.1.0-3.9). The results allowed confirming the occurrence of Mycoplasma agalactiae in milk samples of goats from Paraíba. Therefore, it is strictly necessary to monitor dairy goat flocks and to raise the awareness of farmers about the economic importance of the disease, since it causes severe economic losses for producers of the state. Identification of risk factors is essential for adoption of control measures and for the correction of the management factors in farms where there are animals with positive diagnosis, avoiding, so, pathogen dissemination.

As macroalgas marinhas, por estarem sujeitas a numerosas interações biológicas e, por vezes, a condições abióticas extremas, desenvolveram, entre outros mecanismos de defesa, a capacidade de produzir substâncias biologicamente ativas. Assim, esses organismos produzem, principalmente, terpenos e fenóis. Entre outras, a atividade antifúngica, por sua importância na saúde humana e animal e na preservação de produtos agrícolas, tem sido objeto de diversos estudos. No presente trabalho, esta atividade foi investigada em dez extratos de macroalgas marinhas, por ensaios de bioautografia direta, frente ao fungo Colletotrichum lagenarium e por ensaio de difusão em disco, frente ao Aspergillus flavus. Os organismos estudados foram os seguintes: Stypopodium zonale, Laurencia dendroidea, Ascophyllum nodosum, Sargassum muticum, Pelvetia canaliculata, Fucus spiralis, Sargassum filipendula, Sargassum stenophyllum, Laminaria hyperborea e Gracilaria edulis. Os extratos de S. zonale, L. dendroidea, P. canaliculata, S. muticum, A. nodosum e F. spiralis inibiram significativamente o crescimento de C. lagenarium, porém, não significativamente, o do fungo A. flavus. A presença de terpenos em todos esses extratos foi confirmada por cromatografia em camada delgada e a de compostos fenólicos, apenas nos extratos de P. canaliculata, A. nodosum e S. muticum. No estudo químico por cromatografia em coluna, seguido por análise em cromatógrafo a gás/espectrômetro de massas, foram identificados, em L. dendroidea, os terpenos neofitadieno, cartilagineol, elatol e obtusol e o éster hexadecanoato de etila. Esse é o primeiro relato sobre a atividade de extratos de algas frente a C. lagenarium, fungo de importância agrícola.

Seaweeds are subject to numerous biological interactions and sometimes to extreme abiotic conditions, so they have developed among other defense mechanisms, the ability to produce biologically active substances. Thus, these organisms produce mainly terpenes and phenols. Among others, the antifungal activity, due to its importance in human and animal health and the production of agricultural products, has been the subject of several studies. In the present work, this activity was investigated in ten seaweeds extracts, by direct bioautography assays, compared to Colletotrichum lagenarium and disk diffusion assay, compared to Aspergillus flavus. The organisms studied were: Stypopodium zonale, Laurencia dendroidea, Ascophyllum nodosum, Sargassum muticum, Pelvetia canaliculata, Fucus spiralis, Sargassum filipendula, Sargassum stenophyllum, Laminaria hyperborea and Gracilaria edulis. S. zonale, L. dendroidea, P. canaliculata, S. muticum, A. nodosum and F. spiralis extracts significantly inhibited the C. lagenarium growth, but not inhibited significantly the A. flavus growth. The presence of terpenes in all of these extracts was confirmed by thin layer chromatography whereas the presence of phenolic compounds was confirmed only in extracts of P. canaliculata, A. nodosum and S. muticum. In chemical study by column chromatography, followed by gas chromatography/mass spectrometry analysis, the terpenes neophytadiene, cartilagineol, obtusol elatol; and the ester ethyl hexadecanoate were identified in the L. dendroidea extract. This is the first report on the activity of seaweed extracts against C. lagenarium, a fungus bearing agricultural importance.

A presença de micotoxinas como resultado do ataque fúngico pode ocorrer antes, após e durante a colheita e também no armazenamento de grãos e alimentos. Considerando as propriedades inibitórias dos óleos essenciais de plantas no desenvolvimento do micélio dos fungos e a importâncias do Aspergillus flavus, principal produtor de aflatoxinas, relatou-se neste trabalho, a atividade tóxica do óleo essencial do Pittosporum undulatum em cultura de A. flavus. Os óleos essenciais de P. undulatum foram obtidos a partir de folhas coletadas em diferentes meses e analisado por CG/EM. Os óleos se mostraram ricos em hidrocarbonetos, monoterpenos e sesquiterpenos e foi observada uma significante variação na composição química destes óleos nos diferentes meses de coleta. Os óleos essenciais mostraram diferentes espectros de inibição do crescimento de A. flavus, porém todos foram capazes de inibir a produção de aflatoxina B1.

The presence of mycotoxins as a result of fungal attack can occur before, after and during the harvest and storage operations on agricultural crops and food commodities. Considering the inhibitory property of essential plant oils on the mycelial development of fungi and the importance of Aspergillus flavus, the main producer of aflatoxins, this research was designed to evaluate the toxicity of essential oil from Pittosporum undulatum against A. flavus. The essential oils were obtained from P. undulatum leaves, collected in different months and analyzed by GC/MS. The oils were rich in hydrocarbon, monoterpenes and sesquiterpenes and it was observed a significant variation on the chemical composition of the essential oil of leaves at different months. Besides, the essential oils were tested against fungal growth and the results showed different spectrum of inhibition on A. flavus. However, the essential oils inhibited the aflatoxin B1 production.

O proteassomo é uma protease intracelular multicatalítica envolvida na regulação do ciclo e sinalização celular, apresentação antigênica e apoptose. Uma vez que inibidores do proteassomo promovem morte celular por apoptose, esses têm sido propostos como novas drogas anti-tumorais. A terreína, um metabólito secundário secretado pelo fungo Aspergillus terreus, foi primeiramente descrita em 1935. Neste trabalho demonstramos que a terreína, isolada através da bioprospecção de inibidores do proteassomo, mostrou efeito inibitório das atividades do tipo quimiotripsina e tripsina da unidade catalítica do proteassomo 20S. Apesar da alta concentração inibitória determinada in vitro, aquela verificada após incubação de células em cultura na presença de terreína (fibroblasto e tumor pulmonar humano) foi 4 vezes menor, o que sugere que o proteassomo seja um alvo intracelular específico. A terreína promoveu morte celular por apoptose nas duas linhagens ensaiadas. Embora as concentrações de terreína necessárias para desencadear apoptose nos modelos celulares aqui testados tenham sido altas (ordem de mM) quando comparadas com doses utilizadas de outros inibidores descritos recentemente (ordem de μM e nM), sua estrutura química não está relacionada a nenhum outro inibidor conhecido até o momento. Concluímos que estes resultados apontam para a possibilidade de explorar a terreína como um novo fragmento molecular para o desenvolvimento de inibidores sintéticos do proteassomo.

The proteasome is an intracellular multicatalytic protease involved in the cell cycle regulation, signaling response, antigen presentation and apoptosis. Since proteasome inhibitors promote cell death by apoptosis, they have been proposed as new anti-tumoral drugs. Terrein, a secondary metabolite secreted by the fungus Aspergillus terreus, was firstly described in 1935. In the present work we report that terrein isolated through the screening for inhibitors of the 20S proteasome showed inhibitory effect upon both chymotrypsin- and trypsin-like activities of the multicatalytic core particle, the 20S proteasome. Despite of the high inhibitory concentration determined in vitro, that verified by incubating cells (fibroblasts and a pulmonary tumor cell line) in the presence of terrein was 4-fold lower indicating the proteasome as a selective intracellular target. Moreover, terrein promoted apoptotic cell death on both fibroblasts and pulmonary tumor cell line tested. Although terrein concentrations (mM range) necessary to elicit apoptosis in the cellular models herein tried were high when compared to those (μM and nM range) of other inhibitors recently described, its chemical structure is not correlated to any other inhibitor reported thus far. Therefore, the present results point out for the possibility of exploring terrein as a new molecular fragment for the development of synthetic proteasome inhibitors.
