ABSTRACT Feed additives, such as β-glucans, nucleotides, ascorbic acid (vitamin C), and α-tocopherol (vitamin E) can improve fish immunity and contribute to enhanced zootechnical performance of the fish. This study aimed to evaluate the effect of different concentrations of vitamin- mineral premix with or without the inclusion of an immunostimulant boost (β-glucans, nucleotides and vitamins C and E) on the zootechnical performance, hemato-immunological parameters, histological changes, and survival of Nile tilapia juveniles. To this end, isocaloric and isoproteic diets were prepared with three different concentrations of vitamin-mineral premix (1.0, 1.5, and 2.0 kg ton-1); additionally, the effects of 4% immunostimulant boost in the diet were examined considering six diets (without the immunostimulant boost: 0.1%, 0.15%, and 0.2%, and with the immunostimulant boost: 0.1% I, 0.15% I, and 0.2% I). We used 24 experimental units, each including 15 fish. Nile tilapia juveniles (1.88 g±0.25) were fed for 50 d with supplemented diets. Further, zootechnical indexes; histological changes in the liver, spleen, and intestine; intestinal morphology; and hemato-immunological parameters were evaluated. The fish that received supplementation showed higher zootechnical values, compared to those that did not receive the supplementation of immunostimulant reinforcement. Weight gain (30.45g±3.06), daily weight gain (0.60g±0.06), final weight (32.41g±3.15) and feed conversion (0.95±0.06) were higher in tilapia fed with an inclusion concentration of 0.2%. Hematological parameters were not affected by either the pre-mix concentrations or the booster of the immunostimulant. There was an increase in the number of intestinal folds, length of the fold, total area and number of goblet cells in the group supplemented with immunostimulant reinforcement. Supplementation with the immunostimulant promoted growth and improved intestinal morphology and immunological parameters of Nile tilapia juveniles.
RESUMO Os aditivos alimentares, tais como β-glucanos, nucleotídeos, ácido ascórbico (vitamina C) e α-tocoferol (vitamina E), podem melhorar a imunidade dos peixes e contribuir para um melhor desempenho zootécnico. Este estudo teve como objetivo avaliar o efeito de diferentes concentrações de premix vitamínico mineral com ou sem a inclusão de um imunoestimulante (β-glucanos, nucleotídeos e vitaminas C e E) no desempenho zootécnico, nos parâmetros hematoimunológicos, nas alterações histológicas e na sobrevivência dos juvenis de tilápia-do-Nilo. Para esse fim, foram preparadas dietas isocalóricas e isoproteicas com três concentrações diferentes de premix vitamínico e mineral (1,0, 1,5, e 2,0kg ton-1); além disso, foram examinados os efeitos da inclusão de 4% de imunoestimulante na dieta, considerando-se seis dietas (sem o imunoestimulante: 0,1%, 0,15% e 0,2%, e com a inclusão de imunoestimulante: 0,1% I, 0,15% I e 0,2% I). Foram utilizadas 24 unidades experimentais, cada uma com 15 peixes. Os juvenis de tilápia-do-Nilo (1,88g±0,25) foram alimentados, durante 50 dias, com as dietas suplementadas. Além disso, foram avaliados índices zootécnicos; alterações histológicas no fígado, baço e intestino; morfologia intestinal; e parâmetros hematoimunológicos. Os peixes que receberam suplementação apresentaram valores zootécnicos mais elevados, em comparação aos que não receberam a suplementação de reforço imunoestimulante. O ganho de peso (30,45g±3,06), o ganho de peso diário (0,60g ± 0,06), o peso final (32,41g±3,15) e a conversão alimentar (0,95±0,06) foram maiores nas tilápias alimentadas com uma concentração de inclusão de 0,2%. Os parâmetros hematológicos não foram afetados nem pelas concentrações de premix nem pelo reforço do imunoestimulante. Foi observado o aumento no número de pregas intestinais, no comprimento da prega, na área total e no número de células caliciformes no grupo suplementado com o boost imunoestimulante. A suplementação com o imunoestimulante promoveu o crescimento e melhorou a morfologia intestinal e os parâmetros imunológicos de juvenis de tilápia-do-Nilo.

ABSTRACT Researchers from different areas of knowledge have used the Cox proportional-hazards model, due to its simplicity and easy interpretation when studying situations in which the response variable is the time until the occurrence of an event of interest. However, the traditional Cox proportional-hazards model is not suitable for modeling data sets that violate the assumption of proportionality of the risks (or failure rates) and the effects of covariates over time are not detected. The generalized time-dependent logistic (GTDL) model has been used as an alternative in the modeling of survival data, taking into account the assumption of non-proportionality of the risks. In the literature, we found a wide and relevant production in inferential procedures, but no contribution in diagnostic methods or techniques. In this paper, Cox-Snell, modified Cox-Snell, martingale, deviance, randomized quantiles, NMSP (normally-transformed modified survival probabilities) and NRSP (normally-transformed randomized survival probabilities) residuals are proposed to assess the suitability of the GTDL model to the data. A Monte Carlo simulation study is conducted in order to investigate the empirical distribution of these residuals. In summary, the obtained simulation results indicate the adequacy, for the GTDL model, of the randomized quantile and NRSP residuals, regardless of the proportion of censorship in the data. The GTDL library is built and made available in the R programming language. Finally, the methodology studied is applied to a set of real data, available in the literature, involving patients diagnosed with advanced-stage lung cancer. RESUMO Pesquisadores de diferentes áreas do conhecimento têm utilizado o modelo de riscos proporcionais de Cox devido à sua simplicidade e fácil interpretação ao estudar situações em que a variável resposta é o tempo até a ocorrência de um evento de interesse. No entanto, o modelo tradicional de Cox não é adequado para descrever conjuntos de dados que violam a suposição de proporcionalidade dos riscos (ou taxas de falha) e os efeitos das covariáveis ao longo do tempo não são detectados. O modelo logístico generalizado dependente do tempo (GTDL, do inglês generalized time-dependent logistic) tem sido utilizado como alternativa na modelagem de dados de sobrevivência, levando em consideração a suposição de não proporcionalidade dos riscos. Na literatura é encontrada uma ampla e relevante produção em procedimentos inferenciais, mas nenhuma contribuição em métodos ou técnicas de diagnóstico. Neste artigo, os resíduos de Cox-Snell, modificados de Cox-Snell, martingale, deviance, quantílicos aleatorizados, NMSP (do inglês normally-transformed modified survival probabilities) e NRSP (do inglês normally-transformed randomized survival probabilities) são propostos para avaliar a adequabilidade do modelo GTDL aos dados. Um estudo de simulação via Monte Carlo é conduzido com o proposito de investigar a distribuição empírica desses resíduos. Em suma, os resultados de simulação obtidos indicam a adequação, para o modelo GTDL, dos resíduos quantílicos aleatorizados e NRSP, independentemente da proporção de censura nos dados. A biblioteca GTDL é construída e disponibilizada na linguagem de programação R. Finalmente, a metodologia estudada é aplicada a um conjunto de dados reais, disponível na literatura, envolvendo pacientes diagnosticados com câncer de pulmão em estágio avançado. Biofilm production by Staphylococcus spp. isolated from bovine mastitis in dairy herds in state of Acre, Brazil and its implications

RESUMO O presente estudo teve como objetivos identificar as espécies de estafilococos envolvidas com mastite em rebanhos leiteiros no norte do Brasil, investigar a capacidade de produção de biofilme e analisar a associação da produção de biofilme com multirresistência e intensidade de reações de CMT que podem dificultar o tratamento e causar erros de diagnósticos, respectivamente. Amostras de leite foram coletadas em 23 propriedades leiteiras, situadas em cinco municípios do estado do Acre. Um total de 339 vacas mestiças foram testadas pelo CMT, sendo detectados 109 animais (229 tetos) reagentes ao teste. Após isolamento em ágar sangue, os cocos Gram positivos produtores de catalase foram submetidos à identificação por MALDI TOF MS. Um total de 103 estirpes foram identificadas como estafilococos, sendo mais prevalentes as espécies Staphylococcus chromogenes (58,3%) e S. aureus (19,4%). A produção de biofilme foi avaliada quantitativamente pelo teste de aderência em microplaca. Dentre todas as amostras de estafilococos, 71,8% foram produtoras de biofilme. A maioria das amostras de S. chromogenes (68,3%) apresentou capacidade de produzir biofilme, variando de fraco (43,3%), moderado (13,3%) a forte (11,7%) produtor. Entre as amostras de S. aureus, 50% não foram produtoras e nenhuma foi produtora forte. Essa capacidade de produção de biofilme mostrou-se associada à multirresistência a antimicrobianos, além de diminuir a intensidade no teste CMT, podendo mascarar o diagnóstico.
ABSTRACT This study aimed to identify the Staphylococcus species responsible for bovine mastitis in dairy herds in northern Brazil, to investigate the capacity of biofilm production, and to analyze the association of biofilm production with multiresistance and intensity of California Mastitis Tests (CMT) reactions that can make treatment more difficult and cause misdiagnoses, respectively. Milk samples were collected from 23 dairy farms located in five municipalities in the state of Acre. A total of 339 crossbred cows were tested by CMT, with 109 animals (229 udder ceilings) reacting to the test. After bacterial isolation in blood agar, the catalase-positive and gram-positive cocci were submitted for identification by MALDI-TOF MS. Of 103 strains identified as staphylococci, Staphylococcus chromogenes (58.3%) and Staphylococcus aureus (19.4%) were the most prevalent species. Biofilm production was quantitatively evaluated using a microplate adherence test. Among the Staphylococcus strains, 71.8% were biofilm producers. Most strains of S. chromogenes (68.3%) had the capacity to produce biofilms, ranging from weak (43.3%), moderate (13.3%), and strong (11.7%) producers. Among S. aureus strains, 50% were non-biofilm producers, and none were strong producers. Our data showed an association between biofilm production capacity and multidrug resistance. In addition, there was a reduction in the response to the CMT test, which can mask the diagnosis.

Abstract The search for new therapeutic strategies for leishmaniasis treatment is essential due to the side effects of available drugs and the increasing incidence of resistance to them. Marine sponges use chemical compounds as a defense mechanism, and several of them present interesting pharmacological properties. The aim of this study was to evaluate the in vitro activity of the aqueous extract of the marine sponge Dercitus (Stoeba) latex against Leishmania amazonensis. MIC and toxicity against mammal cells were evaluated through broth microdilution assays. Transmission electron microscopy analysis was performed to assess possible effects on L. amazonensis ultrastructure. Arginase and proteolytic activities were measured by spectrometric methodologies. The extract of Dercitus (Stoeba) latex displayed antileishmanial activity and moderate toxicity against peritonial macrophages. Ultrastructural changes were observed after the growth of L. amazonensis promastigotes in the presence of the extract at 150 µg.ml-1 (IC50), mainly on acidocalcysomes. The extract was able to inhibit the activity of arginase and serine proteases. This study shows that Dercitus (Stoeba) latex aqueous extract may be a novel potential source of protozoa protease inhibitors and drugs that are less toxic to be used in the treatment of L. amazonensis infections.

Kahweol is a compound derived from coffee with reported antinociceptive effects. Based on the few reports that exist in the literature regarding the mechanisms involved in kahweol-induced peripheral antinociceptive action, this study proposed to investigate the contribution of the endocannabinoid system to the peripheral antinociception induced in rats by kahweol. Hyperalgesia was induced by intraplantar injection of prostaglandin E2(PGE2) and was measured with the paw pressure test. Kahweol and the drugs to test the cannabinoid system were administered locally into the right hind paw. The endocannabinoids were purified by open-bed chromatography on silica and measured by LC-MS. Kahweol (80 µg/paw) induced peripheral antinociception against PGE2-induced hyperalgesia. This effect was reversed by the intraplantar injection of the CB1 cannabinoid receptor antagonist AM251 (20, 40, and 80 μg/paw), but not by the CB2 cannabinoid receptor antagonist AM630 (100 μg/paw). Treatment with the endocannabinoid reuptake inhibitor VDM11 (2.5 μg/paw) intensified the peripheral antinociceptive effect induced by low-dose kahweol (40 μg/paw). The monoacylglycerol lipase (MAGL) inhibitor, JZL184 (4 μg/paw), and the dual MAGL/fatty acid amide hydrolase (FAAH) inhibitor, MAFP (0.5 μg/paw), potentiated the peripheral antinociceptive effect of low-dose kahweol. Furthermore, kahweol increased the levels of the endocannabinoid anandamide, but not of the other endocannabinoid 2-arachidonoylglycerol nor of anandamide-related N-acylethanolamines, in the plantar surface of the rat paw. Our results suggested that kahweol induced peripheral antinociception via anandamide release and activation of CB1 cannabinoid receptors and this compound could be used to develop new drugs for pain relief.

Isolation and characterization of the aerobic bacterial microbiota of the esophagus and its probable association with obstructive caseous lesions in green turtles (Chelonia mydas)

RESUMO: A ocorrência de lesão caseosa no esôfago de tartarugas-verdes (Chelonia mydas) da costa do Brasil tem sido descrita como de caráter obstrutivo e pode causar a morte dos animais. No entanto, sua etiologia permanece pouco esclarecida. Objetivou-se isolar e caracterizar a microbiota aeróbica esofágica das tartarugas-verdes (C. mydas) da costa brasileira e verificar sua possível participação na etiologia das lesões caseosas. Foram utilizados 42 animais, 33 vivos e hígidos e nove mortos naturalmente que apresentavam lesão esofágica confirmada pela necropsia, provenientes de Anchieta e Piúma, Espírito Santo, nos quais foram feitos testes microbiológicos e avaliação morfológica do esôfago. Foram isolados 14 agentes bacterianos diferentes nas amostras de animais saudáveis, com prevalência de Pseudomonas aeruginosa (36,36%), Staphylococcus aureus (33,33%), Aeromonas hydrophila (27,27%) e Vibrio alginolyticus (24,24%). Nos animais mortos, foram isolados apenas três agentes distintos: S. aureus (50,00%), A. hydrophila (25,00%) e V. alginolyticus (25,00%). A avaliação morfológica revelou predominância da lesão em junção gastroesofágica, com distribuição multifocal a coalescente, intensidade discreta e ausência de obstrução. Observou-se ainda ulceração e exsudato caseoso, infiltrado inflamatório, ovos de parasitos e células gigantes do tipo corpo estranho, além de grumos bacterianos e de alterações glandulares, como necrose, adenite e fragmentos de parasitos adultos. Houve correlação positiva dos grumos bacterianos com cultivo microbiológico e negativa dos grumos bacterianos e cultivo microbiológico com parasitos. Assim, nota-se que a microbiota esofágica aeróbica de C. mydas é constituída predominantemente por bactérias Gram-negativas como P. aeruginosa, A. hydrophila e V. alginolyticus, além de diversas enterobatérias e por Gram-positivas, como S. aureus. Esses agentes são oportunistas e podem estar envolvidos na etiologia da esofagite caseosa em associação a outros patógenos como co-fatores agindo em associação, ou mesmo, por via de infecção secundária.
ABSTRACT: Caseous lesions in the esophagus of green turtles (Chelonia mydas) from the coast of Brazil have been described as obstructive lesions and can lead to the death of these animals. However, their etiology remains unclear. The aim of this study was to isolate and characterize the aerobic bacterial microbiota of the esophagus of green turtles (C. mydas) from the Brazilian coast and to verify its possible participation in the etiology of caseous lesions. For this, 42 animals were used, 33 alive and healthy and 9 naturally dead that had esophageal lesions confirmed by necropsy, from Anchieta and Piúma beaches, Espírito Santo. Microbiological tests and morphological evaluation of the esophagus were performed. We isolated 14 different bacterial agents from healthy animal samples, with the prevalence of Pseudomonas aeruginosa being (36.36%), Staphylococcus aureus (33.33%), Aeromonas hydrophila (27.27%), and Vibrio alginolyticus (24.24%). In dead animals, only three distinct agents were isolated: S. aureus (50.00%), A. hydrophila (25.00%), and V. alginolyticus (25.00%). Morphological evaluation revealed a predominance of the lesions at the gastroesophageal junction, with multifocal-to-coalescent distribution, discrete intensity, and absence of obstruction. Ulcerations and caseous exudates, inflammatory infiltrates, parasitic eggs, and giant foreign body cells were also observed as well as bacterial lumps and glandular alterations, such as necrosis, adenitis, and fragments of adult parasites. There was a positive correlation between bacterial lumps and microbiological culture and a negative correlation between bacterial lumps and microbiological culture with parasites. Thus, it was noted that the esophageal aerobic microbiota of C. mydas was predominantly composed of Gram-negative bacteria such as P. aeruginosa, A. hydrophila, and V. alginolyticus, in addition to several enterobacteria and Gram-positive bacteria, such as S. aureus. These agents are opportunists and may be involved in the etiology of caseous esophagitis in association with other pathogens as co-factors working in association or, even in a secondary way.

RESUMO: A conservação do haylage (alimento pré-seco) pode ser desafiadora, considerando o aumento do risco de crescimento de fungos, com consequente produção de micotoxinas. Entretanto, quando a qualidade da higiene e armazenamento é assegurada, o haylage aumenta a palatabilidade da forragem e fornece suplemento de matéria seca suficiente ao longo do ano. Devido à falta de informação relativa aos efeitos dessa alimentação nos parâmetros sanguíneos de equinos alimentados exclusivamente com essa dieta, o objetivo do presente estudo é avaliar o perfil bioquímico sanguíneo dos equinos após administração da haylage em comparação com feno. Doze matrizes Quarto de Milha foram distribuídas em dois grupos, cada um recebendo feno ou haylage de Tifton 85 (Cynodon spp.) por um período de 28 dias. O perfil bioquímico foi realizado em cinco tempos (T) diferentes (T0, antes do início do experimento e cronologicamente, a cada sete dias após o fornecimento das dietas - T1, T2, T3 e T4) para análise de proteína total (PT) e seu perfil fracionado, das enzimas alanina aminotransferase, aspartato aminotransferase, γ-glutamil-transferase, dos produtos de catabolismo creatinina e ureia e, dos íons cálcio e fósforo. Micotoxinas no haylage foram investigadas e mantiveram-se abaixo dos limites determinados pela legislação brasileira. O perfil bioquímico revelou, somente, elevação da PT em T4 no grupo que recebeu haylage, o que pode estar relacionado à sua maior digestibilidade proteica. Nenhuma diferença foi observada nos outros parâmetros estudados em ambos os grupos experimentais. Conclui-se que Haylage é comprovadamente seguro, quando bem preparado para equinos, sem causar efeitos na saúde geral, conforme demonstrado pelos exames bioquímicos no presente estudo.
ABSTRACT: The conservation of haylage (a pre-dried feed) can be challenging, since there is an increased risk of mould growth, which can contaminate this foodstuff with mycotoxins. However, when the hygienic quality is secured, haylage enhances grass palatability and provide enough supply of dry matter throughout the year. Due to the lack of information regarding its effect on blood parameters in horses fed exclusively with this foodstuff, the aim of this study is to provide information regarding its use in comparison to hay and ensure that it does not affect horses’ biochemical profile. Twelve Quarter Horse broodmares were distributed into two groups, each fed with Tifton-85 (Cynodon spp.) hay or haylage for a period of 28 days, and the biochemical profile was done in five different times (T0 before the experiment started and, chronologically, seven days apart - T1, T2, T3 and T4), It was analyzed total protein (TP) and its fractioning; enzymes alanine aminotransferase, aspartate aminotransferase and γ-glutamyl-transferase; endogenous catabolism products urea and creatinine; and ions calcium and phosphorus. Mycotoxins in haylage were also investigated and remained below the legislation thresholds. Only TP was higher in the last sampling (T4) of the haylage group, which may be related to the foodstuff’s higher protein digestibility. No differences were observed between serum enzymes, urea, creatinine and Ca/P from both experimental groups. Haylage has proven to be safe, when well prepared for horses, without causing impairing side effects, as shown by the normal serum biochemistry parameters presented in this study.

Cervical cancer is a public health problem and the molecular mechanisms underlying radioresistance are still poorly understood. Here, we evaluated the modulation of key molecules involved in cell proliferation, cell cycle and DNA repair in cervical cancer cell lines (CASKI and C33A) and in malignant tissues biopsied from 10 patients before and after radiotherapy. The expression patterns of epidermal growth factor receptor (EGFR), excision repair cross-complementation group 1 (ERCC1) and p53 were evaluated in cancer cell lines by quantitative PCR and western blotting, and in human malignant tissues by immunohistochemistry. The mutation status of TP53 gene was evaluated by direct sequencing. Among cell lines, absent or weak modulations of EGFR, ERCC1 and p53 were observed after exposure to 1.8 Gy. Conversely, increased expressions of p53 (5/10 patients; P=0.0239), ERCC1 (5/10 patients; P=0.0294) and EGFR (4/10 patients; P=0.1773) were observed in malignant tissues after radiotherapy with the same radiation dose. TP53 mutations were found only in one patient. Here we show that a single dose of radiotherapy induced EGFR, ERCC1 and p53 expression in malignant tissues from cervical cancer patients but not in cancer cell lines, highlighting the gap between in vitro and in vivo experimental models. Studies on larger patient cohorts are needed to allow an interpretation that an upregulation of p53, EGFR and ERCC1 may be part of a radioresistance mechanism.

RESUMO Doenças infecciosas são as maiores responsáveis por falhas reprodutivas (FR) em cadelas, causando aborto, morte fetal e natimortalidade. Este estudo teve como objetivo investigar a associação entre agentes infecciosos, FR inexplicáveis e anemia em cadelas. Todas as amostras maternas e fetais foram negativas para a presença dos principais agentes infecciosos causadores de FR: herpes vírus canino 1, Neospora caninum, Brucella spp. e B. canis, enquanto agentes como o de Leishmania spp., parvovírus canino, Ehrlichia canis e Anaplasma platys foram encontrados em sangue materno. Coinfecções de A. platys/E. canis e A. platys/Leishmania spp. foram diagnosticadas. Os resultados indicam que os animais com anemia causadas por doenças transmitidas por vetores podem ser mais suscetíveis a sofrerem FR do que animais com valores hematológicos normais.

RESUMO O presente estudo avaliou o perfil hormonal e a expressão gênica de receptores de prostaglandina F2α (PGF2α), ocitocina e estrógeno no endométrio de vacas pós-parto tratadas com cloprostenol. Vinte vacas mestiças Holandês-Zebu foram tratadas com solução salina (tratamento CONT, n = 10) ou cloprostenol (tratamento CLO, n = 10), ambos administrados dois e cinco dias após o parto. Amostras de sangue foram coletadas nos dias dois, sete, 14, 21 e 28 pós-parto para mensuração de progesterona, de metabólito de PGF2α (PGFM) e de estradiol, e foram obtidas biópsias endometriais para quantificar a expressão de PTGFR, OXTR e ESR1. No tratamento CLO, a expressão gênica de receptores de ocitocina foi menor (P<0,05). As concentrações de estrógeno aumentaram progressivamente até o dia 14 (P<0,05). A maior expressão de OXTR foi observada no dia 14 (P<0,05). A expressão de ESR1 foi semelhante entre os tratamentos (P>0,05). Os níveis de PGFM foram altos durante todo o estudo. Conclui-se que a administração de cloprostenol nos dias dois e cinco pós-parto parece diminuir a expressão de OXTR no endométrio em vacas mestiças.
ABSTRACT The present study investigated the hormonal profile and expression of prostaglandin F2α (PGF2α), oxytocin and estrogen receptors in uterine tissues of postpartum cows treated with cloprostenol. Twenty Holstein-Zebu crossbred cows were treated with saline solution (treatment CONT) or cloprostenol (treatment CLO), both administered two and five days postpartum. Blood samples were collected on days two, seven, 14, 21 and 28 postpartum for progesterone, PGF2α metabolite (PGFM) and estradiol determination, and endometrial biopsy was performed in order to quantify the expression of oxytocin receptor (OXTR), prostaglandin F receptor (PTGFR) and estrogen receptor 1 (ERS1) genes. In the CLO treatment, expression of OXTR was reduced (P<0.05) but no difference (P>0.05) between treatments was found for PTGFR and ERS1 expression. Estrogen concentrations increased progressively until day 14 (P<0.05) and the highest OXTR expression and lowest PTGFR expression were observed on day 14 (P<0.05) in both treatments. Serum PGFM concentrations were high throughout the experiment. In conclusion, cloprostenol administration at days two and five of postpartum seems to reduce OXTR expression in the endometrium in crossbred cows.

RESUMO A placentite é a principal causa de partos prematuros, aborto e nascimento de potros comprometidos, podendo causar hipóxia e septicemia. A hematologia e a gasometria venosa fornecem informações importantes para o monitoramento da saúde de potros nascidos de éguas com placentite. O objetivo deste trabalho foi descrever os valores hematológicos e hemogasométricos durante as primeiras 24h de vida em potros nascidos de éguas mestiças Crioulas submetidas à indução experimental de placentite, com diferentes graus de maturidade. Foram utilizados 16 potros nascidos de éguas submetidas à indução experimental de placentite, divididos em três grupos de acordo com o grau de maturidade: prematuro (n=8), dismaturo (n=4) e a termo (n=4). Foram realizadas coletas sanguíneas nos momentos 0h, 12h e 24h para realização de hemograma completo e gasometria venosa. No eritrograma, foi observada anemia normocítica normocrômica no grupo prematuro em relação ao grupo dismaturo nas 12h e 24h. O grupo prematuro apresentou menor contagem de leucócitos totais nas 24h em relação ao grupo a termo (P=0,01). Os valores de pH, cHCO3 e SO2 não diferiram entre os grupos, porém os animais prematuros apresentaram acidose respiratória (pH=7,28). A PCO2 nos prematuros foi maior na 0h (P=0,02). Nos três grupos, a PCO2 apresentou uma curva adaptativa com redução dos valores durante as 12h e 24h. Os potros prematuros mostraram menores valores de excesso de base (BE) no nascimento (P=0,02), confirmando o quadro de acidose respiratória. Concluiu-se que as respostas hematológicas e hemogasométricas diferem entre potros com diferentes graus de maturidade. A acidose observada no grupo prematuro ao nascimento, com estabilização e resposta compensatória durante as 12h e 24h, demonstra a necessidade de avaliação hemogasométrica sequencial em potros de risco, o que permite a identificação da resposta clínica ao processo e, assim, auxilia no estabelecimento do tratamento e prognóstico para esses potros.
ABSTRACT The placentitis is a major cause of premature birth, abortion and compromised foal delivery, and may result in hypoxia and sepsis. The blood gas analysis and hematology can provide important information for monitoring the foals born from mares with placentitis, with different degrees of maturity. The aim of this study was to describe the hematological and blood gas values during the first 24 hours of life in foals born from crossbreed mares with experimentally induced placentitis, presenting different degrees of maturity. Sixteen foals, born from mares with experimentally induced ascending placentitis were assigned to three groups according to degree of maturity: premature (n=8), dysmature (n=4), and full-term foals (n=4). Blood samples were collected at birth (0), at 12h and 24h, and hematological evaluation and blood gas variables were measured. In the premature group normocytic normochromic anemia was observed compared to dysmature group at 12h and 24h. The premature group showed lower count of white blood cells at 24h relative to the full-term group (p=0.01). The pH, cHCO3 and SO2 values do not differ among the groups; however the premature group showed respiratory acidosis (pH=7,28). The PCO2 was higher at 0h in the premature foals (p=0.02). In all groups, the PCO2 presented an adaptive curve with reduction between 12h and 24h. The premature foals showed lower base excess (BE) values at birth (p=0.02), confirming the respiratory acidosis in this group. We conclude that the hematological and blood gas response differs between foals with different degrees of maturity. Acidosis in the premature foals at birth, with stabilization and compensation of pH value during the first 12-24h demonstrate the necessity of sequential blood gas analysis in risk foals. This may help identify the clinical response to the process and assist in the establishment of adequate treatment and prognosis for these foals.

Effect of wind in the design of reinforced concrete buildings

RESUMO Este artigo apresenta os resultados de um estudo paramétrico realizado com o objetivo de quantificar o tanto que erros na etapa de projeto relacionados com a consideração da ação do vento podem comprometer a resposta em serviço e a segurança de edifícios de concreto armado. Usando-se um modelo arquitetônico como referência e variando-se o número de pavimentos do edifício, a segurança estrutural foi avaliada como uma função da intensidade da ação do vento. Os resultados mostraram que mesmo para edifícios baixos, com 10 pavimentos, a desconsideração da ação do vento pode comprometer significativamente o comportamento e a segurança e que no caso de edifícios mais esbeltos, com até 30 pavimentos, pode levar a resultados catastróficos, como a ruína da estrutura através de colapso progressivo.
ABSTRACT This paper presents the results from a parametric study carried in order to quantify how far errors in the design stage related to the consideration of the wind action may put at risk the response and safety of reinforced concrete buildings. Using an architectural model as reference and varying the number of floors of the building, the structural safety was evaluated as a function of the wind action intensity. Results showed that even for low-rise buildings, with 10 floors, ignoring the wind action can significantly jeopardize their behaviour and safety. Yet, for slenderer buildings, up to 30 floors, it can lead to catastrophic results, as the ruin of the structure by progressive collapse.

Morphology of the testes and epididymal ducts in the pampas cat Leopardus colocolo (Molina, 1782)

RESUMO: O gato-palheiro Leopardus colocolo (Molina, 1782) é uma espécie da família Felidae, com ampla distribuição na América do Sul. Está incluido no Appendix II da CITES e classificada como Próxima da Extinção na Lista Vermelha da IUCN, apresentando população em decrescimo. Com base nessas informações o objetivo deste estudo é caracterizar morfologicamente os testículos e ductos epididimários de L. colocolo. O animal, oriundo do Zoológico da Universidade Federal de Mato Grosso, Brasil, veio a óbito após procedimento anestésico e o sistema reprodutor masculino foi dissecado para coleta dos testículos. As amostras retiradas foram fragmentadas e histologicamente examinadas. A partir das análises microscópicas dos testículos foram identificados: a túnica vaginal e albugínea, formada por tecido conjuntivo denso modelado, com grande quantidade de fibras de colágeno. A túnica albugínea emite septos fibrosos para o interior do órgão. Os túbulos seminíferos são enovelados e revestidos internamente por epitélio estratificado constituído por células espermatogênicas e células somáticas de Sertoli, envolvidos por uma membrana basal com presença de células mioides. O tecido intersticial, entre os túbulos seminíferos, é constituído de tecido conjuntivo do tipo frouxo com vasos sanguíneos e linfáticos, e células de Leydig em formato poliédrico. Os ductos epididimários apresentaram epitélio cilíndrico pseudoestratificado com células secretoras dos quais projetam estereocílios, situados sobre uma membrana basal repleta por células mióides. Este epitélio apresenta células principais e basais, cujas células principais projetam estereocílios em direção ao lúmen do ducto epididimário.
ABSTRACT: The pampas cat Leopardus colocolo (Molina, 1782) is a species of the Felidae family, widely distributed in South America, included on CITES Appendix II and classified as Near Threatened on the IUCN Red List, with population trend decreasing. Based on this information, the objective of this study is to describe morphologically the testes and epididymal ducts of pampas cat. The animal, coming from the Federal University of Mato Grosso Zoo, Brazil, had died after anesthesia procedure and the male reproductive system was dissected to collect the testicles. The samples taken were fragmented and histologically examined. From the microscopic analysis of the testes were identified: vaginal and tunica albuginea, formed by dense connective tissue modeled with large amount of collagen fibers. The tunica albuginea fibrous septa emits into the body. The seminiferous tubules are coiled and coated internally by spermatogenic epithelium consisting of Sertoli cells, surrounded by a basement membrane in the presence of myoid cells. The interstitial tissue between the seminiferous tubules, is composed of loose connective tissue, blood and lymph vessels, and Leydig cells in polyhedral shape. The epididymal ducts showed pseudostratified columnar epithelium with secretory cells of which stereocilia design, situated on a basement membrane filled by myoid cells. This epithelium has principal and basal cells, the main cell design stereocilia toward the lumen of the epididymal duct.

O presente trabalho teve por objetivo avaliar a conversão alimentar (CA) por meio da inferência bayesiana considerando-se análises bivariadas. Foram utilizadas diferentes espécies animais de experimentos conduzidos na Universidade Federal de Viçosa, no estado de Minas Gerais, Brasil. O modelo proposto mostrou ser apropriado, uma vez que possibilitou a detecção de diferenças significativas entre níveis de fatores não detectados por procedimentos frequentistas em ANOVA tradicional, principalmente em pequenas amostras. No experimento com codornas, evidenciou-se que aves cujos níveis de proteína bruta eram de 23% e 29%, respectivamente, para machos e fêmeas, apresentaram uma melhor CA, de 2,83±0,03 e 2,66±0,03, respectivamente. No experimento com frangos, no grupo sem o aditivo antibiótico, a inclusão de 0,02% de extrato de ésteres naturais foi o que promoveu a melhor CA (1,72±0,01), e, de modo geral, o uso de antibiótico e a ausência de ésteres naturais promoveram CA de 1,63±0,02. Em caprinos, verificou-se que o aleitamento, seja com leite de cabra ou de vaca, promove igualmente uma melhor CA, respectivamente, no grupo de 60 e 90 dias, de 1,29±0,14 e 1,79±0,11, sugerindo que o aleitamento seja feito até os 60 dias. Em suínos, a dieta com maior nível de energia metabolizável e aminoácidos foi a que promoveu a melhor CA (2,86±0,07), quando comparada a uma dieta com nível nutricional mais baixo. Já o uso de enzimas na dieta com menor nível energético e de aminoácidos proporcionou resultado intermediário (2,90±007). Em bovinos, observou-se que o uso de 1% de concentrado na dieta promoveria uma melhor CA estimada de 7,33±0,35 entre os Nelores e que essa promoção seria de 7,40±0,58 entre os cruzados com o uso de 2% de concentrado na dieta.
The aim of this study was to evaluate the feed conversion (CA) by Bayesian inference in bivariate considering analyzes in real and simulated data. Different animal species experiments conducted at the Universidade Federal de Viçosa, state of Minas Gerais, Brazil are used. The proposed model proved to be appropriate once it enabled the detection of significant differences between levels of factors not detected by frequentist procedures with traditional ANOVA, especially in small samples. In the experiment with quails, it became clear that the birds' brute protein levels were 23% and 29%, respectively, for males and females, which presented better CA, 2.83±0.03 and 2.66±0.03, respectively. In the experiment with chickens, the group without additive antibiotic, including 0.02% extract natural esters promoted the best CA (1.72±0.01) and in general antibiotic absence of esters natural promoted 1.63±0.02 of the CA. In goats, it has been found that feeding milk from cows or goats also promotes better CA, respectively, in groups milked up to 60 and 90 days, being 1.29±0.14 and 1.79±0.11, suggesting that suckling done until 60 days. Pigs fed the highest level of metabolizable energy and aminoacids promoted the best CA (2.86±0.07) compared to a diet with lower nutritional level. But the use of enzymes in the diet with lower energy level and amino acid provided intermediate result (2.90±007). In cattle, it was observed that the use of 1% concentrate diet, CA, promotes a better estimate of 7.33±0.35 between Nellore and this promotion would be 7.40±0.58 between the cross breeds using 2% concentrate diet.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.
