Abstract The Program for Biodiversity Research (PPBio) is an innovative program designed to integrate all biodiversity research stakeholders. Operating since 2004, it has installed long-term ecological research sites throughout Brazil and its logic has been applied in some other southern-hemisphere countries. The program supports all aspects of research necessary to understand biodiversity and the processes that affect it. There are presently 161 sampling sites (see some of them at Supplementary Appendix), most of which use a standardized methodology that allows comparisons across biomes and through time. To date, there are about 1200 publications associated with PPBio that cover topics ranging from natural history to genetics and species distributions. Most of the field data and metadata are available through PPBio web sites or DataONE. Metadata is available for researchers that intend to explore the different faces of Brazilian biodiversity spatio-temporal variation, as well as for managers intending to improve conservation strategies. The Program also fostered, directly and indirectly, local technical capacity building, and supported the training of hundreds of undergraduate and graduate students. The main challenge is maintaining the long-term funding necessary to understand biodiversity patterns and processes under pressure from global environmental changes.

Culture media to detect and criteria to evaluate and report the activity of extracellular enzymes produced by phytopathogenic fungi

RESUMO: As enzimas extracelulares estão envolvidas na patogênese de fungos em plantas. Atualmente, não há uma padronização de meios de cultura, formas de analisar e divulgar os dados de enzimas extracelulares produzidas em condições in vitro. Assim, o presente trabalho objetivou comparar os meios de cultura específicos relatados na literatura (normal) com o meio de batata-dextrose-ágar mais adição do substrato específico para produção de enzimas extracelulares por três diferentes fungos fitopatogênicos habitantes de solo (Fusarium solani f. sp. passiflorae, Sclerotium rolfsii e Rhizoctonia solani AG-4 HGI), bem como avaliar os dados das enzimas por cinco critérios diferentes. O delineamento experimental adotado foi o inteiramente ao acaso, em esquema de três fatores (meios, isolados e enzimas), com seis repetições. As enzimas investigadas foram amilase, carboximetilcelulase, lipase, lacase, catalase e gelatinase. Os meios normais detectaram mais enzimas, e essa detecção foi mais precisa em comparação com os meios de batata-dextrose-ágar mais o substrato específico. Os critérios que calcularam a área da coroa circular para as enzimas amilase, carboximetilcelulase, lipase e lacase e adotaram a escala de notas para medir a intensidade (0=ausência até 4=intensa) de catalase e gelatinase foram os melhores para avaliar e divulgar os resultados das enzimas. Assim, sugere-se padronizar os meios normais para estudos de produção de enzimas, bem como os critérios citados para avaliar e divulgar os dados das atividades das referidas enzimas.
ABSTRACT: Extracellular enzymes are involved in the fungal pathogenesis in plants. Currently, culture media, data analyses, and data report related to extracellular enzymes produced in vitro conditions are different and therefore, lack standardization. This work aimed to compare the culture media cited on the literature (normal) with the potato-dextrose-agar (PDA) medium combined with a specific compound to produce extracellular enzymes through three soilborne phytopathogenic fungi (F. solani f. sp. passiflorae, S. rolfsii, and R. solani AG-4 HGI), as well as to analyze and report enzyme data based on five different criteria. The assay was randomized, with three factors (culture media, isolates, and enzymes) and six repetitions. The studied enzymes were amylase (AM), carboxymethylcellulase (CMCase), lipase (LP), laccase (LC), catalase (CT), and gelatinase (GT). The normal media detected more enzymes and was more precise compared to the PDA medium plus specific compound. The criteria that calculated the area of the circular crown of AM, CMCase, LP, and LC and measured the intensity (0 = absence, up to 4 = intense) of CT and GT adopting note scale were the best to evaluate and report the results of the enzymes. We suggest the normal media culture to study enzyme production, as well as the criteria mentioned to assess and report the data related to enzyme activities.

RESUMO Objetivou-se avaliar o efeito dos resíduos da extração hidroalcoólica de própolis dos tipos verde e marrom sobre o desenvolvimento de bactérias Gram-positivas e Gram-negativas. As própolis brutas foram adquiridas da apicultura “Companhia da Abelha”, instalada em Contagem, Minas Gerais. A própolis verde foi derivada de “alecrim-do-campo” (Baccharis dracunculifolia), e a própolis marrom foi derivada de “alecrim-do-campo” (B. dracunculifolia) e “assa-peixe” (Vernonia polyanthes). Foram usados cinco níveis de inclusão (0; 0,625; 0,125; 0,25; 0,5g) dos resíduos, além do controle para o inóculo (CI), e seis amostras bacterianas, sendo duas delas Gram-positivas e quatro Gram-negativas. O método utilizado foi o de contagem de unidades formadoras de colônias em placas de petri (pour plate). Entre as bactérias Gram-positivas, o maior efeito inibidor foi detectado para amostras de Staphylococcus aureus, e maior susceptibilidade foi para a Escherichia coli dentre as Gram-negativas. Houve efeito antimicrobiano do resíduo da extração da própolis verde sobre as bactérias S. aureus e S. intermedius. O resíduo da extração da própolis marrom apresentou maior inibição para S. intermedius somente nos tratamentos com 0,5 e 0,25g de resíduo. Não houve efeito do resíduo da extração de própolis verde ou marrom sobre o crescimento de bactérias gênero Pseudomonas. Os resíduos da extração hidroalcoólica de própolis verde e marrom atuaram como inibidores de crescimento das bactérias Staphylococcus aureus, Staphylococcus intermedius e Escherichia coli, e em menor grau para Salmonella e Klebsiella. O resíduo tem maior efeito inibidor em bactérias Gram-positivas.
SUMMARY This study aimed to evaluate the effect of residues extraction hydroalcoholic of propolis, green and brown, on the development of Gram-positive and Gram-negative bacteria. The crude propolis was obtained from “Companhia da Abelha” in Contagem, Minas Gerais. The green propolis derived from “alecrim-do-campo” (Baccharis dracunculifolia), and the brown propolis derived from “alecrim-do-campo” (B. dracunculifolia) and “assa-peixe” (Vernonia polyanthes). We used five levels of inclusion (0; 0.625; 0.125; 0.25; 0.5g) of the residue, and control for the inoculum (CI) and six bacterial strains, two of which are Gram-positive and four Gram-negative. The method used was to count colony-forming units in petri dishes (pour plate). Among the Gram-positive bacteria, the greatest inhibitory effect was detected for Staphylococcus aureus strains, and increased susceptibility was observed for Escherichia coli among Gram-negative bacteria. There was antimicrobial effect of the residue of the propolis extract on S. aureus and S. intermedius. The residue of brown propolis extract showed greater inhibition for S. intermedius only in 0.5 and 0.25g of residue inclusion levels. There was no effect of residues extraction hydroalcoholic of propolis, green and brown on the growth of Pseudomonas. The residues of the extraction of propolis green and brown acted as inhibitors of growth of Staphylococcus aureus, Staphylococcus intermedius and Escherichia coli, and to a lesser extent Salmonella and Klebsiella. The residue has greater inhibitory effect on Gram-positive bacteria.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

INTRODUÇÃO: As doenças inflamatórias intestinais são enfermidades crônicas, que afetam significativamente a qualidade e expectativa de vida dos pacientes. Existe polimorfismo clínico e a abordagem terapêutica tem sido modificada nos últimos anos. OBJETIVO: Reavaliar o perfil dos pacientes em acompanhamento ambulatorial analisando o comportamento das doenças, sua prevalência e conduta terapêutica. MÉTODOS: Foi realizado um estudo transversal da última atualização da base de dados dos pacientes de ambulatório de doença inflamatória intestinal em 2010. Os itens analisados foram sexo e idade dos pacientes, tipo da doença (retocolite ulcerativa ou doença de Crohn), localização da doença, tipo de medicação em uso, e se os pacientes estavam sintomáticos ou assintomáticos na última consulta. RESULTADOS: Foram estudados 171 pacientes. O sexo feminino mostrou-se predominante (60,8%) e a média de idade dos pacientes foi de 42,3, variando de 16 a 84 anos. Em relação ao tipo de doença inflamatória, a retocolite ulcerativa mostrou-se mais prevalente (58,5%). As localizações mais frequentes na retocolite ulcerativa foram pancolite e retite, ambas com 26% (n=26). Na doença de Crohn a localização ileocolônica foi a mais prevalente, com 47,9% (n=34), sendo seguida pela colônica com 25,4% (n=18). A monoterapia foi a mais utilizada, correspondendo a 54,4% dos pacientes, sendo que os imunossupressores foram as drogas de uso mais frequente (35,5%). A associação de medicamentos foi necessária em 36,3% dos casos, sendo a combinação mais frequente salicilato local com sistêmico em 33,9%. Nos portadores de retocolite ulcerativa 82% estavam em uso de salicilatos, seja local ou sistêmico. Na doença de Crohn 57,7% faziam uso de imunossupressores. Em relação à atividade da doença na última consulta, 71,3% dos pacientes apresentavam-se assintomáticos. CONCLUSÃO: A retocolite ulcerativa foi pouco mais prevalente que a doença de Crohn, sendo pancolite e retite as localizações mais frequentes. Na doença de Crohn a localização ileocolônica foi a mais encontrada seguida pela colônica. A monoterapia com imunossupressores foi o esquema terapêutico mais utilizado. A maioria dos pacientes apresentava-se assintomático na última consulta.

There is clinical polymorphism and the therapeutic approach has been modified in recent years. AIM: To re-analyze the profile of outpatient patients studying the behavior of the disease, its prevalence and therapeutic approach. METHODS: Was conducted a cross-sectional study of the last 2010 update database of patients with inflammatory bowel disease. The items analyzed were age and sex of patients, type of disease (ulcerative colitis or Crohn's disease), disease location, type of medication used, and whether patients were symptomatic or asymptomatic at the last visit. RESULTS: In a total of 171 patients women were found to be predominant (60.8%) and the mean age was 42.3, ranging from 16 to 84 years. Regarding the type of inflammatory disease, ulcerative colitis was more prevalent (58.5%). The most common locations on ulcerative colitis were pancolitis and rectitis, both with 26% (n = 26). In Crohn's disease ileocolic location was the most prevalent, with 47.9% (n = 34), followed by colonic 25.4% (n = 18). Monotherapy was the most used, accounting for 54.4% of patients, the immunosuppressive drugs were the most frequently used (35.5%). The combination of drugs was necessary in 36.3% of cases, being the most frequent combination with systemic salicylate in 33.9%. In patients with ulcerative colitis 82% were using salicylates, whether local or systemic. In Crohn's disease 57.7% were taking immunosuppressive drugs. In relation to the disease in the last visit, 71.3% of patients were asymptomatic. CONCLUSION: The ulcerative colitis was slightly more prevalent than Crohn's disease, pancolitis and rectitis being the most common locations. In Crohn's disease ileocolic location was the most common followed by colonic. Monotherapy immunosuppressive regimen was the most popular. Most of the patients were asymptomatic at last visit.

Characterization of HBeAg-negative chronic hepatitis B in western Brazilian Amazonia

The present study was conducted with 55 patients native from western Brazilian Amazonia, who were HBV-DNA positive after seroconversion of HBeAg. It is a descriptive case study, with the patients separated into two groups: with hepatitis and without hepatitis on histological examination. The aim of the present study was to describe the clinical and molecular characteristics of patients who are chronic carriers of HBsAg. The prevalence of hepatitis was 63.64%, with a predominance of males (41.82%) and a mean age of 42.5 years, occurring mostly in natives of the southeast sub-region (32.73%). Time was a variable proportional to the course of the disease and the most frequent symptoms were: dyspepsia, asthenia and loss of libido with the majority of the patients having history of prior contact with HBV or positive family history. Splenomegalia was the most frequent sign (40%). Among the tests, platelet count, serum albumin and prothrombin activity were significant in the diagnosis of hepatitis. Alpha-fetoprotein was greater in patients with hepatitis, and hepatocellular carcinoma was detected in 3.63% of the patients with hepatic cirrhosis. Three types of HBV genotypes were diagnosed: A, D and F in the samples amplified for gene S. Genotype A (AA) was observed in 54.54% of the cases with hepatitis, in contrast to other studies showing the predominance of genotype F in this region. We observed mutations in 36.36%, with a predominance of the mutations in the core promoter region (31.81%), due to the greater prevalence of genotype A in this study.

O Staphylococcus aureus é uma bactéria do grupo dos cocos gram-positivos que faz parte da microbiota humana, mas que pode provocar doenças que vão desde uma infecção simples, como espinhas e furúnculos, até as mais graves, como pneumonia, meningite, endocardite, síndrome do choque tóxico e septicemia, entre outras. Essa bactéria foi uma das primeiras a serem controladas com a descoberta dos antibióticos, mas, devido a sua enorme capacidade de adaptação e resistência, tornou-se uma das espécies de maior importância no quadro das infecções hospitalares e comunitárias. Neste artigo faremos uma revisão sobre esse agente infeccioso e as bases dos mecanismos das patologias por ele provocadas, de forma a ressaltar a necessidade de mantê-lo como alvo para o desenho de novos antibióticos.
Staphylococcus aureus is a bacterium from Gram-positive coccus group, which is part of the human microbiota. It may cause diseases that may vary from simple infections (i.e., pimples and furuncles) to severe infections, such as pneumonia, meningitis, endocarditis, toxic shock syndrome and septicemia, among others. This bacterium was one of the first bacteria affected by antibacterial agents, however, its ability of adaptation and resistance turns it into an important hospital and communitarian pathogenic species of great concern. In this article we will discuss some important points related to the S. aureus and the pathologies related to it to reinforce it as a target for the design of new antibiotics.

This review deals with the homo- and copolymerization of styrene with nickel catalysts. The catalytic activity, polymer stereoregularity, polymer molecular weight and polydispersity are dependent upon nickel ligands and reaction parameters. Catalysts supported on silica, treated with methylaluminoxane (MAO), have shown higher stereospecificity and activity compared to homogeneous ones. The influence of these parameters is discussed focusing on the elucidation of some aspects of the polymerization mechanism.

Production of recombinant proteins in Escherichia coli

Attempts to obtain a recombinant protein using prokaryotic expression systems can go from a rewarding and rather fast procedure to a frustrating time-consuming experience. In most cases production of heterologous proteins in Escherichia coli K12 strains has remained an empirical exercise in which different systems are tested without a careful insight into the various factors affecting adequate expression of the encoded protein. The present review will deal with E. coli as protein factory and will cover some of the aspects related to transcriptional and translational expression signals, factors affecting protein stability and solubility and targeting of proteins to different cell compartments. Based on the knowledge accumulated over the last decade, we believe that the rate of success for those dedicated to expression of recombinant proteins based on the use E. coli strains can still be significantly improved.

Mutantes de Escherichia coli resistentes aos aminoglicosídios por seleção in vitro em meio de cultivo de alta osmolaridade contendo canamicina podem apresentar ausência da proteína periplásmica oligopeptídio permease (OppA). O comportamento desse mutante em presença de canamicina ou do peptídio tóxico triornitina sugere que a proteína OppA e um outro componente do envoltório bacteriano contribuem para a resistência osmodependente em E. coli.
The oligopeptide permease (OppA) protein was found to be missing in the periplasmic fractions of Escherichia coli kanamycin-resistant mutants selected under high osmotic conditions. The growth behavior of one mutant in media containing kanamycin or the toxic peptide triornithine suggests that OppA and another cell envelope component contribute to the osmolarity-dependent aminoglycoside resistance of E. Coli.
