Abstract. Technological development for Gymnotus sp. farming is relevant because this species has a socio- economic impact in South American regions where sport fishing is an important activity. Although Gymnotus sp. breeds in captivity, fry survival in ponds can be unacceptable for culture systems. The objective of this work was to test the response to hormonal induction of Gymnotus sp. with carp pituitary extract (CPE) and gonadotrophin-releasing hormone (GnRH) in relation to different male and female reproductive parameters. CPE treatment specimens were stripped 12 and 20 hours after the triggering dosage, and GnRH treatment specimens were stripped 10 and 12 hours after hormonal application. We could obtain positive results using CPE with a short capture-induction interval (C-II) with broodstock in mature stages of gonadal development. Percentage of oocytes with peripheral germinal vesicle (%OPGV) and oocyte diameter showed significant differences between groups treated with CPE. Oocytes could be successfully extracted from two females treated with CPE and stripped 12 hours after the last dosage. Semen was only extracted from males treated with CPE. GnRH stimulation did not give positive results because gametes could not be extracted with different doses and C-II used. This work is the first description of semen quality parameters in Gymnotus sp. Future works should focus on combining hormonal induction strategies with CPE and the management of environmental variables.
Resumen El desarrollo tecnológico para el cultivo de Gymnotus sp. es relevante porque este género tiene un impacto socioeconómico en regiones sudamericanas donde la pesca deportiva es una actividad importante. Aunque Gymnotus sp. se reproduce en cautiverio, la supervivencia de los alevines en los estanques puede ser insuficiente para un sistema de cultivo. El objetivo de este trabajo fue evaluar la respuesta de diferentes parámetros reproductivos a la inducción hormonal en Gymnotus sp. con extracto bruto de hipófisis de carpa (EBH) y hormona liberadora de gonadotrofinas (GnRH). Las muestras de gametos del tratamiento con EBH se extrajeron 12 y 20 horas después de la dosis desencadenante, las muestras del tratamiento con GnRH se extrajeron 10 y 12 horas después de la aplicación hormonal. El porcentaje de ovocitos con vesícula germinal periférica (%OVGP) y el diámetro de los ovocitos mostraron diferencias significativas entre grupos tratados con EBH con un intervalo captura-inducción (IC-I) de corta duración, con reproductores en etapas avanzadas de desarrollo gonadal. Se pudieron extraer ovocitos con éxito a dos hembras inducidas con EBH, 12 horas después de la dosis desencadenante y, además, se extrajo semen sólo de los machos inducidos con EBH. La estimulación con GnRH no dio resultados positivos en las diferentes dosis e IC-I utilizados, dado que no se pudieron extraer gametos. Este trabajo es la primera descripción de los parámetros de calidad del semen en Gymnotus sp. Futuras líneas de trabajo deberán centrarse en el manejo de variables ambientales para establecer una estrategia de reproducción que se pueda utilizar a nivel productivo.

RESUMO: Na micropropagação, o nitrato de potássio (KNO3), reagente puro para análise (P.A.), utilizado no preparo dos meios de cultura, possui custo elevado e a sua aquisição depende de trâmites burocráticos, por se tratar de substância controlada pelo Exército Brasileiro. O objetivo deste trabalho foi avaliar o efeito da substituição do KNO3 P.A. por fertilizante comercial (com fonte de KNO3), encontrado livremente no comércio, na micropropagação de palma (Opuntia stricta (Haw.) Haw. cv Orelha de Elefante. Os tratamentos foram de seis concentrações do fertilizante (0; 0,5; 1; 1,5; 2 e 2,5 g L-1) e um controle constituído de 1,9 g L-1 de reagente KNO3, conforme mostrado nos sais MS. Avaliou-se a sobrevivência, tamanho e número de brotações do explante, e o valor da biomassa fresca. Após a aclimatização das mudas avaliou-se a sobrevivência, número de brotações, comprimento da parte aérea, número de raízes, formação da raquete, massa média da biomassa fresca, absorção de macronutrientes e alterações morfológicas das mudas. Os explantes inoculados em meio com fertilizantes nas concentrações de 0,0; 2,0 e 2,5 g L-¹ não se desenvolveram. A resposta dos explantes nas concentrações de 0,5 e 1,5 g L-1 do fertilizante foram iguais aos desenvolvidos em meio contendo KNO3, e na concentração de 1,0 g L-1, em todas as variáveis, as médias foram superiores em relação as do controle. Dessa forma, constatou-se a viabilidade do uso do fertilizante no cultivo in vitro da palma, o que propiciou a eliminação dos entraves burocráticos e redução no custo de 99,12% na compra do produto.
ABSTRACT: In micropropagation, potassium nitrate (KNO3), an ACS reagent grade chemical, used in the preparation of growing mediums is expensive and its procurement depends on bureaucratic procedures, as it is controlled by the Brazilian Army. This research to assessed the effect of replacing the ACS KNO3 for a commercially available fertilizer (KNO3- based) on the micropropagation of the prickly pear cactus (Opuntia stricta (Haw.) Haw. cv. Elephant Ear. Treatments used six different fertilizer concentrations (0, 0.5, 1, 1.5, 2 and 2.5 g L-1) and a control consisting of 1.9 g L-1 KNO3, as shown in the MS salts. The survival, size and number of sprouts and the value of fresh biomass were evaluated. After seedling acclimation, we assessed the survival, number of sprouts, length, and number of roots, racket formation, average fresh biomass mass, macronutrient absorption and morphological changes of the seedlings. Explants inoculated with fertilizers at concentrations of 0.0; 2.0 and 2.5 g L-¹ did not grow. The response of explants at concentrations of 0.5 and 1.5 g L-1 of the fertilizer were the same as those developed in a KNO3 medium, and at a concentration of 1.0 g L-1, in all variables, the means were higher than those of the control medium. Therefore, it showed the feasibility of using fertilizers in the in vitro cultivation of the prickly pear cactus, which may remove bureaucratic barriers and reduce product costs by 99.12%.

Abstract The 2019 oil spill was the biggest in Brazilian history. Oil was found along more than 3,000 km of the Brazilian coastline, mainly in the Northeast, in more than 1,000 localities. This article analyzes the disaster’s damage using a sample of interviewers who were impacted - fishers, tourism and beach hawkers - distributed along 40 of the affected municipalities in the Northeast Region of Brazil. The socio-economic indicators obtained by the research show that the impacts were not homogeneous between the segments and cities researched. Localities specialized in tourism and with a workforce relatively more specialized in fishing were the most affected. Accordingly, the populations of fishers and beach hawkers suffered the most severe impacts in terms of income reduction and the sale of products. These agents report a negative impact of the disaster on their work activities of 73% (fishers) and 65% (beach vendors), while the lodging and food sectors reported losses in about 38% of the cases. The interviewees’ health indicators demonstrated that the volunteers at the oil spill clean- up suffered damage due to the exposure experienced, evidencing the public health emergency dimension of the disaster.

SiOx Top Layer on DLC Films for Atomic Oxygen and Ozone Corrosion Protection in Aerospace Applications

Every year, billions of dollars are invested in research and development for space applications, including new systems, new technologies, and new materials. DLC (Diamond-Like Carbon) is a promising material for use in these applications, but its use faces a technological barrier, since it is severely etched by atomic oxygen and ozone. In this study, SiOx-DLC thin films were deposited as a top layer of diamond-like carbon (DLC) films on Ti-6Al-4V substrates to increase resistance against corrosion by atomic oxygen and ozone as well as meet the requirements for use in Low Earth Orbit (LEO) satellites. The corrosion resistance of the films was evaluated using oxygen plasma, and the tribological and mechanical properties were investigated. The SiOx-DLC top layer reduced the corrosion rate two orders of magnitude and increased the critical load from 16.2 ± 1.5 N to 18.4 ± 0.4 N.

RESUMO A fauna de anfíbios do estado de Mato Grosso do Sul foi compilada e elaborada pela primeira vez, totalizando 97 espécies, o que representa pouco mais de 10% das espécies que ocorrem no Brasil. Nove famílias e 25 gêneros de Anura e apenas uma espécie de Gymnophiona foram registradas. As áreas de Cerrado apresentaram a maior riqueza, seguidas pelas florestas estacionais semideciduais, Chaco e veredas. Em relação às macrorregiões consideradas, foram registradas 56 espécies para a planície de inundação (majoritariamente representada pelo Pantanal) e 92 para o planalto. As regiões mais amostradas em Mato Grosso do Sul são o Parque Nacional da Serra da Bodoquena e algumas áreas do Pantanal, o que resulta em lacunas de amostragens por todo o estado, principalmente nas regiões norte, leste/sudeste e parte do oeste/sudoeste. Foram identificadas áreas consideradas importantes para amostragens em decorrência da complexidade de fitofisionomias e relevos encontrados no estado: Maciço do rio Apa, Serra de Maracaju, grande parte do Chaco brasileiro, leque do rio Taquari, planalto do Taquari-Itiquira, escarpas do planalto Central, áreas úmidas/florestas estacionais da planície do rio Paraná e áreas interioranas do Pantanal, onde o acesso é difícil.
ABSTRACT Amphibian species from Mato Grosso do Sul state were compiled and checked for the first time. A total of 97 species were recorded, representing more than 10% of the known Brazilian amphibians. Twenty five anuran genera in nine families and only one caecilian were recorded. Cerrado was the richest area followed by semideciduous forests, Chaco, and veredas (palm swamps). It was recorded 56 species in floodplain areas and 92 in the surrounding cerrados. The Serra da Bodoquena National Park and some Pantanal areas were better sampled than other regions in Mato Grosso do Sul, resulting in sampling gaps across the state. Important areas for future inventories were identified: mountainous regions in Serra de Maracaju and those bordering the Apa river, Brazilian chacoan areas, the Taquari river alluvial fan, the broad Taquari-Itiquira flat area, the Central Plateau scarps, wet areas from Parana river bordering São Paulo state, and inland areas from Pantanal.

Emergence and Control of Rottboelia spp. in Different Months in Sugarcane Cultivation1

RESUMO Objetivou-se neste estudo avaliar o controle de Rottboelia spp. com a mistura formulada diuron + hexazinone + sulfometuron-methyl e a emergência dessa planta daninha nos meses de abril, maio, junho, julho, agosto, setembro, outubro e novembro de 2012 na cultura da cana-de-açúcar, variedade RB867515. Utilizou-se o delineamento em blocos casualizados, com os tratamentos dispostos em esquema fatorial 4 x 4, com quatro repetições para os meses de abril a novembro. Nos ensaios de emergência utilizou-se o delineamento de blocos casualizados, em esquema de parcelas subdivididas 8 x 5, com quatro repetições. Os fatores analisados foram as doses da mistura formulada (diuron + hexazinone + sulfometuron-methyl), que consistiram em: 1.809,00 + 510,00 + 43,50 g ha-1 (T2); 1.507,50 + 425,00 + 36,25 g ha-1 (T3) e 1.386,90 + 391,00 + 33,35 g ha-1 (T4), além da testemunha (T1) sem aplicação. Controles superiores a 80,00% foram verificados para a aplicação realizada no mês de novembro nas avaliações aos 30 e 60 DAA, independentemente da dose utilizada. Todos os tratamentos foram seletivos para a variedade RB867515 de cana-de-açúcar. Os ensaios instalados nos meses de agosto, setembro, outubro e novembro registraram a maior densidade populacional.
ABSTRACT This study aimed to evaluate the control of Rottboelia spp. with the formulated mixture diuron + hexazinone + sulfometuron-methyl and the emergence of this weed in the months of April, May, June, July, August, September, October and November 2012 in sugarcane cultivation, cultivar RB867515. The experimental design consisted of randomized blocks, with treatments in a 4 x 4 factorial arrangement with four replicates for the months of April to November. The emergence tests used a randomized block design, in a 8 x 5 split plot arrangement with four replicates. The analyzed factors were the rates of the formulated mixture (diuron + hexazinone + sulfometuron-methyl), which consisted of: 1,809.00 + 510.00 + 43.50 g ha-1 (T2); 1,507.50 + 425.00 + 36.25 g ha-1 (T3) and 1,386.90 + 391.00 + 33.35 g ha-1 (T4), in addition to the control (T1) without application. Controls over 80.00% were found for the application performed in the month of November in evaluations at 30 and 60 DAA, regardless of the rate in use. All treatments were selective for the sugarcane cultivar RB867515. The tests conducted in the months of August, September, October and November showed the highest population density.

The aim of this study was to evaluate the effects of sodium hyaluronate (HY), single-walled carbon nanotubes (SWCNTs) and HY-functionalized SWCNTs (HY-SWCNTs) on the behavior of primary osteoblasts, as well as to investigate the deposition of inorganic crystals on titanium surfaces coated with these biocomposites. Primary osteoblasts were obtained from the calvarial bones of male newborn Wistar rats (5 rats for each cell extraction). We assessed cell viability using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide assay and by double-staining with propidium iodide and Hoechst. We also assessed the formation of mineralized bone nodules by von Kossa staining, the mRNA expression of bone repair proteins, and the deposition of inorganic crystals on titanium surfaces coated with HY, SWCNTs, or HY-SWCNTs. The results showed that treatment with these biocomposites did not alter the viability of primary osteoblasts. Furthermore, deposition of mineralized bone nodules was significantly increased by cells treated with HY and HY-SWCNTs. This can be partly explained by an increase in the mRNA expression of type I and III collagen, osteocalcin, and bone morphogenetic proteins 2 and 4. Additionally, the titanium surface treated with HY-SWCNTs showed a significant increase in the deposition of inorganic crystals. Thus, our data indicate that HY, SWCNTs, and HY-SWCNTs are potentially useful for the development of new strategies for bone tissue engineering.

Although long regarded as the gold standard for liver fibrosis staging in chronic hepatitis C (CHC), liver biopsy (LB) implies both the risk of an invasive procedure and significant variability. The aim of this study was to evaluate the diagnostic performance for transient elastography (TE) and aspartate aminotransferase to platelet index (APRI) used alone and in combination compared to liver biopsy and to analyze false positive/negative results. Patients with CHC, and no previous clinical diagnosis of cirrhosis were enrolled to undergo liver biopsy, TE and APRI. A total of 182 adult patients with a median age of 55 years and median body mass index of 26.71 kg/m2 were analyzed. On LB, 56% of patients had significant levels of fibrosis (METAVIR F≥2) and 28% had advanced fibrosis (F3/F4). The strongest performance for both tests was observed for exclusion of advanced fibrosis with good negative predictive values (89 and 86%, respectively). Low necroinflammatory activity on LB was associated with false negative TE. False positives were associated with NASH and smaller LB fragments. Correlation between APRI and Fibroscan for F≥2 was 100% and 84% for F≥3 and remained high in both false negative and false positive instances, correctly identifying F<2 in 71% of cases and F<3 in 78% (and potentially foregoing up to 84% of LB). We concluded that low individual performance indicators could be attributable to limitations of LB. Poorer differentiation of lower levels of fibrosis is a known issue for LB and remains so for noninvasive tests. Good predictability is possible, however, for advanced fibrosis.

Endodontics pastes formulated with copaiba oil: action on oral microbiota and dentin bridge formation in dogs

Foi avaliada a ação de pastas formuladas com hidróxido de cálcio P.A. e diferentes veículos (solução fisiológica - pasta A e óleo de Copaifera langsdorffii Desfon - pasta B) sobre microrganismos bucais e formação de ponte dentinária em cães. A ação antimicrobiana das pastas e de seus componentes individuais foi avaliada pela técnica da concentração inibitória mínima pela diluição em ágar. Os materiais foram diluídos e avaliados contra quinze cepas padrão de microrganismos relacionados a doenças endodônticas. Os microrganismos foram cultivados e, após a incubação, os dados foram analisados (Anova One-Way, Tukey, P≤0,05). Para a avaliação da formação de ponte dentinária, quatro incisivos superiores de dez animais foram tratados, sendo dois capeados com pasta A (GA) e dois com a pasta B (GB). Após 90 dias, os dentes foram extraídos para obtenção de cortes histológicos, com o objetivo de se avaliar o grau de formação de ponte dentinária. Os dados obtidos foram submetidos à análise estatística (Kruskal-Wallis, P<0,05). Segundo a ação antimicrobiana, os materiais foram classificados em ordem decrescente: hidróxido de cálcio P.A., pasta A, pasta B e óleo de Copaifera langsdorffii Desfon, com diferenças estatisticamente significantes entre o hidróxido de cálcio P.A., as pastas e os veículos. Ao final do estudo, observou-se que as duas pastas avaliadas foram semelhantes quanto à formação de ponte dentinária. Considerando-se os microrganismos selecionados, conclui-se que as pastas avaliadas foram semelhantes entre si em termos de potencial antimicrobiano, diferindo dos componentes individuais. Em relação à formação de ponte dentinária, não foram observadas diferenças significantes entre os dentes tratados com as diferentes pastas.
This study analysed the effect of pastes formulated with calcium hydroxide P.A. and different vehicles (saline solution - paste A and Copaifera langsdorffii Desfon oil - paste B) on oral microorganisms and dentin bridge formation in dogs. The antimicrobial action of the pastes and their components was analysed by the minimum inhibitory concentration in agar gel technique. The components were diluted and tested on fifteen standard strains of microorganisms associated with endodontic diseases. The microorganisms were cultivated and after incubation data was analysed using One-Way ANOVA and Turkey's test (P≤0.05). Four superior incisors of ten animals were used to evaluate dentin bridge formation. Two incisors were capped with paste A (GA) and two with paste B (GB). After 90 days, the teeth were extracted for histological analysis and the degree of dentin bridge formation evaluated. Data was analysed by the Kruskal-Wallis test (P<0.05). The pastes and their components were classified in the following decreasing order of antimicrobial action: calcium hydroxide P.A., paste A, paste B and Copaifera langsdorffii Desfon oil. Calcium hydroxide P.A. showed significantly higher antimicrobial action than the pastes or their vehicles. No significant difference was observed between the two pastes in dentin bridge formation. Based on the microorganisms studied, it can be concluded that the pastes analysed showed similar antimicrobial potential but differed significantly from their individual components. No significant difference was observed in dentin bridge formation between the different pastes tested.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Subcutaneous administration of ketoprofen delays Ehrlich solid tumor growth in mice

O cetoprofeno é uma droga anti-inflamatória não esteroidal (NSAID), que exerce efeito anti-inflamatório, antiproliferativo e antitumoral em condições neoplásicas e não neoplásicas. Foram investigados os efeitos desse composto sobre o desenvolvimento tumoral de camundongos Swiss previamente inoculados com células do tumor de Ehrlich. Para realização deste estudo, o tumor sólido foi obtido a partir de células do fluido ascítico do tumor de Ehrlich suspendido em solução salina fisiológica para se obter 2,5x106 de células em 0,05mL. Após a inoculação do tumor, os animais foram separados em dois grupos (n = 10). Os animais do grupo tratado receberam cetoprofeno 0,1μg/100μL/animal por via intraperitoneal, em intervalos de 24 horas durante 10 dias consecutivos, e o controle 100µL de solução salina. No final do experimento, os animais foram sacrificados, e o tumor removido. Foram analisados o crescimento do tumor e as características histomorfológicas e imunoistoquímicas para CDC47 (marcador de proliferação celular) e para CD31 (marcador de vasos sanguíneos). Os animais tratados com o cetoprofeno 0,1μg/100μL/animal apresentaram menor crescimento do tumor. O tratamento não influenciou significativamente o tamanho das áreas de neoplasia, inflamação, necrose e hemorragia. Em contraste, uma menor taxa de proliferação das células neoplásicas foi observada nos animais tratados com cetoprofeno em comparação com o grupo controle. A participação do cetoprofeno em controlar a proliferação de células neoplásicas abre perspectivas para sua utilização em terapia clínica antineoplásica.
Ketoprofen, a nonsteroidal anti-inflammatory drug (NSAID) has proven to exert anti-inflammatory, anti-proliferative and anti-angiogenic activities in both neoplastic and non-neoplastic conditions. We investigated the effects of this compound on tumor development in Swiss mice previously inoculated with Ehrlich tumor cells. To carry out this study the solid tumor was obtained from cells of the ascites fluid of Ehrlich tumor re-suspended in physiological saline to give 2.5x106cells in 0.05mL. After tumor inoculation, the animals were separated into two groups (n = 10). The animals treated with ketoprofen 0.1µg/100µL/animal were injected intraperitoneally at intervals of 24h for 10 consecutive days. Animals from the control group received saline. At the end of the experiment the mice were killed and the tumor removed. We analyzed tumor growth, histomorphological and immunohistochemical characteristics for CDC47 (cellular proliferation marker) and for CD31 (blood vessel marker). Animals treated with the ketoprofen 0.1µg/100µL/animal showed lower tumor growth. The treatment did not significantly influence the size of the areas of cancer, inflammation, necrosis and hemorrhage. Moreover, lower rates of tumor cell proliferation were observed in animals treated with ketoprofen compared with the untreated control group. The participation of ketoprofen in controlling tumor malignant cell proliferation would open prospects for its use in clinical and antineoplasic therapy.

INTRODUÇÃO: Medicamentos endodônticos apresentam componentes tóxicos que provocam algum grau de reação inflamatória.OBJETIVO: Este estudo avaliou o efeito da laserterapia na resposta inflamatória causada por medicações intracanais, em tecido subcutâneo de ratos, por meio da análise quantitativa de mastócitos.MATERIAL E MÉTODO: Tubos de polietileno contendo as medicações foram implantados no dorso de 60 ratos, distribuídos em seis grupos: HS (pasta de hidróxido de cálcio P.A.); HL (pasta de hidróxido de cálcio P.A. e laserterapia); HPS (pasta de hidróxido de cálcio P.A. com paramonoclorofenol canforado); HPL (pasta de hidróxido de cálcio P.A. com paramonoclorofenol canforado e laserterapia); IS (iodofórmio e soro fisiológico) e IL (iodofórmio, soro fisiológico e laserterapia). Os animais foram eutanasiados oito e quinze dias após a cirurgia, as peças cirúrgicas foram removidas, processadas para inclusão em parafina e os cortes histológicos corados em Azul de Toluidina 0.2%, para quantificação dos mastócitos. A análise de variância (ANOVA) e o teste de Tukey post hoc foram aplicados para determinar diferenças significativas entre os grupos quanto ao número de mastócitos (p<0.05).RESULTADO: Nos grupos HL, HPL e IL houve uma diminuição de mastócitos em ambos os períodos experimentais em relação aos grupos HS, HPS e IS, porém não se observou diferença estatística significativa entre o grupo HPS e o HPL aos oito dias.CONCLUSÃO: A laserterapia foi eficaz em modular a intensidade da resposta inflamatória induzida pelos medicamentos endodônticos a partir da redução significativa na quantidade de mastócitos.
INTRODUCTION: Endodontic medications contain toxic components that cause varying degrees of inflammation.OBJECTIVE: This study evaluated the effect of laser therapy on the inflammatory response induced by intracanal medications implanted into the subcutaneous tissue of rats using a quantitative analysis of mast cells.MATERIAL AND METHOD: Polyethylene tubes containing the medications were implanted in the dorsum of 60 rats divided into six groups, including HS (P.A. calcium hydroxide paste), HL (P.A. calcium hydroxide paste and laser therapy), HPS (P.A. calcium hydroxide paste with camphorated paramonochlorophenol), HPL (P.A. calcium hydroxide paste with camphorated paramonochlorophenol and laser therapy), IS (iodoform with saline) and IL (iodoform with saline and laser therapy). The animals were euthanized eight or fifteen days after surgery, and samples were removed and embedded in paraffin. Histological sections were stained with 0.2% toluidine blue for the quantification of mast cells. Analysis of variance (ANOVA) and Tukey's post-hoc test were applied to determine significant differences in the number of mast cells between groups (p<0.05).RESULT: There was a decrease in mast cells for the HL, HPL and IL groups when compared with the HS, HPS and IS groups at both time points. There was no statistically significant difference between the HPS and HPL groups at the eight-day time point.CONCLUSION: Laser therapy was effective at modulating the inflammatory response induced by endodontic medications by significantly reducing the number of mast cells.

Evolution of the biochemical profile of children treated or undergoing treatment for moderate or severe stunting: consequences of metabolic programming?

OBJETIVO: avaliar as mudanças no perfil bioquímico de crianças tratadas ou em tratamento para déficit estatural moderado ou grave em um centro de recuperação e educação nutricional. MÉTODOS: estudo longitudinal retrospectivo com 263 crianças semi-internas no Centro no período de agosto/2008 a agosto/2011, com idade entre 1 e 6 anos e diagnóstico de déficit estatural moderado, escore-Z da altura-para-idade (AIZ) < -2 e grave (AIZ < -3). Foram coletados dados socioeconômicos, dietéticos e bioquímicos e a evolução estatural segundo a idade. RESULTADOS: com a intervenção nutricional observou-se incremento no escore-z das crianças com déficit estatural moderado (0,51 ± 0,4; p = 0,001) e grave (0,91 ± 0,7; p = 0,001) durante o período de acompanhamento. Observou-se, ainda, aumento nos níveis de fator de crescimento insulina-símile (IGF-1) (inicial: 71,7 ng/dL; final: 90,4 ng/dL; p = 0,01), redução nos triglicérides (TG) tanto nas crianças graves (inicial: 91,8 mg/dL; final: 79,1 mg/dL; p = 0,01) como nas moderadamente desnutridas (109,2 mg/dL para 88,7 mg/dL; p = 0,01) e aumento significativo do lipoproteína de alta densidade (HDL-C) apenas no terceiro ano de intervenção (inicial: 31,4 mg/dL; final: 42,2 mg/dL). Os valores de colesterol total (CT) e lipoproteína de baixa densidade (LDL-C) continuaram elevados durante todo o tratamento (inicial: 165,1 mg/dL; final: 163,5 mg/dL e inicial: 109,0 mg/dL; final: 107,3 mg/dL, respectivamente). CONCLUSÃO: o tratamento nutricional para as crianças de baixa estatura mostrou-se eficaz na redução do déficit estatural e dos TG e melhora dos níveis de HDL-C após três anos de intervenção. Porém, os níveis de LDL-C e CT mostraram-se sempre elevados mesmo nas crianças tratadas. Questiona-se, pois, se essas alterações seriam resultantes da programação metabólica a desnutrição.
OBJECTIVE: to evaluate changes in the biochemical profile of children treated or being treated for moderate or severe stunting in a nutrition recovery and education center. METHODS: this was a retrospective longitudinal study of 263 children treated at this center between August of 2008 to August of 2011, aged 1 to 6 years, diagnosed with moderate (z-score of height-for-age [HAZ] < -2) or severe stunting (HAZ < -3). Data were collected on socioeconomic conditions, dietary habits, and biochemical changes, as well as height according to age. RESULTS: the nutritional intervention showed an increase in HAZ of children with moderate (0.51 ± 0.4, p = 0.001) and severe (0.91 ± 0.7, p = 0.001) stunting during the monitoring. Increased levels of insulin-like growth factor 1 (IGF-1) (initial: 71.7 ng/dL; final: 90.4 ng/dL; p = 0.01) were also observed, as well as a reduction in triglycerides (TG) in both severely (initial: 91.8 mg/dL; final: 79.1 mg/dL; p = 0.01) and in moderately malnourished children (initial: 109.2 mg/dL; final 88.7 mg/dL; p = 0.01), and a significant increase in high-density lipoprotein cholesterol HDL-C only in the third year of intervention (initial: 31.4 mg/dL; final: 42.2 mg/dL). The values of total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) levels remained high throughout the treatment (initial: 165.1 mg/dL; final: 163.5 mg/dL and initial: 109.0 mg/dL; final: 107.3 mg/dL, respectively). CONCLUSION: the nutritional treatment for children with short stature was effective in reducing stunting and improving TG and HDL-C after three years of intervention. However, the levels of LDL-C and TC remained high even in treated children. It is therefore speculated that these changes may result from metabolic programming due to malnutrition.

Objetivou-se avaliar a substituição do farelo de soja por torta de amendoim no concentrado da dieta de novilhos, por meio do comportamento ingestivo e respostas fisiológicas. Foram utilizados cinco novilhos, dispostos em um quadrado latino 5 x 5. As dietas foram compostas de feno de tifton, milho moído, farelo de soja e torta de amendoim, constituídas de zero (100% de farelo de soja), 25, 50, 75 e 100% de substituição por torta de amendoim no concentrado. Os níveis de torta de amendoim não influenciaram (P>0,05) as eficiências de ingestão e ruminação da matéria seca e da fibra em detergente neutro. Em relação às atividades comportamentais, o único parâmetro em que se observou efeito da substituição foi o número de períodos de ócio no turno da manhã (P<0,05), no qual se constatou efeito linear crescente. A taxa de secreção salivar e a quebra de partículas foram semelhantes, pois não houve efeito (P>0,05) das dietas sobre as características de mastigação. Como não houve efeito (P>0,05) das dietas sobre as frequências respiratória, cardíaca e a temperatura retal, o calor endógeno produzido pela fermentação ruminal foi equivalente. A torta de amendoim, por conter aflatoxinas, quando fornecida em alta relação volumoso x concentrado, pode substituir o farelo de soja na dieta de novilhos, pois ocasiona comportamento ingestivo e respostas fisiológicas semelhantes.
This study aimed to evaluate the replacement of soybean meal with peanut cake in the concentrate of steer diets through intake behavior and physiological responses. Five steers, arranged in a 5 x 5 latin square design were used. The diets were composed of Tifton hay, ground corn, soybean meal and peanut cake, made from zero (100% soybean meal), 25, 50, 75 and 100% replacement for peanut cake in the concentrate. The levels of peanut cake did not influence (P>0.05) the intake and rumination efficiencies of dry matter and fiber in neutral detergent. Regarding the behavioral activities, the only parameter that observed a substitution effect was the number of idle periods in the morning shift (P<0.05) which had an increasing linear effect. The rate of salivary secretion and the particle breakdown were similar, because there was no effect (P>0.05) of the diets on the chewing characteristics. As there was no effect (P>0.05) of the diets on the respiratory rate, heart rate and rectal temperature, the endogenous heat produced by rumen fermentation was equivalent. The peanut cake, by containing aflatoxin, when provided in high roughage x concentrate rate, can replace soybean meal in the steers' diet because it causes similar intake behavior and physiological responses.

High-frequency electroacupuncture versus carprofen in an incisional pain model in rats

The objective of the present study was to compare the effect of electroacupuncture (EA) and carprofen (CP) on postoperative incisional pain using the plantar incision (PI) model in rats. A 1-cm longitudinal incision was made through skin, fascia and muscles of a hind paw of male Wistar rats and the development of mechanical and thermal hypersensitivity was determined over 4 days using the von Frey and Hargreaves methods, respectively. Based on the experimental treatments received on the third postoperative day, the animals were divided into the following groups: PI+CP (CP, 2 mg/kg, po); PI+EAST36 (100-Hz EA applied bilaterally at the Zusanli point (ST36)); PI+EANP (EA applied to a non-acupoint region); PI+IMMO (immobilization only); PI (vehicle). In the von Frey test, the PI+EAST36 group had higher withdrawal force thresholds in response to mechanical stimuli than the PI, PI+IMMO and PI+EANP groups at several times studied. Furthermore, the PI+EAST36 group showed paw withdrawal thresholds in response to mechanical stimuli that were similar to those of the PI+CP group. In the Hargreaves test, all groups had latencies higher than those observed with PI. The PI+EAST36 group was similar to the PI+IMMO, PI+EANP and PI+CP groups. We conclude that 100-Hz EA at the ST36 point, but not at non-acupoints, can reduce mechanical nociception in the rat model of incisional pain, and its effectiveness is comparable to that of carprofen.
