Abstract Kavain is one of the main kavalactones of Piper methysticum (Piperaceae) with anxiolytic, analgesic, and antioxidant activities. Therefore, the aim of the study was to evaluate the cytotoxic, mutagenic, and antimutagenic potential of kavain in Allium cepa cells. Roots of A. cepa were transferred to the negative (2% acetone) and positive (10 µg/mL of Methylmethanesulfonate, MMS) controls and to the concentrations of kavain (32, 64 and 128 µg/mL) for 48 h. A total of 5,000 meristematic cells were analyzed under an optical microscope to determine the mitotic index, mean number of chromosomal alterations and percentage of damage reduction. Data were analyzed by Kruskal-Wallis test (p <0.05). All concentrations of kavain were not cytotoxic and did not show significant chromosomal changes when compared to 2% acetone. Kavain showed a cytoprotective effect in the pre (128 μg/mL) and in the post-treatment (32 and 64 μg/mL) and reduced damage against the mutagenic action of MMS in all concentrations of the pre and simultaneous and at the highest of post (128 μg/mL). Kavain promoted a significant reduction in micronuclei, nuclear buds and chromosomal losses in relation to MMS. The observed data indicate the importance of kavain for the inhibition of damage and chemoprevention.

Resumo: Os fungos são considerados os mais importantes dentre os patógenos devido ao maior número de espécies e aos danos causados tanto no rendimento quanto na qualidade das sementes. Assim, objetivou-se verificar o efeito do tratamento fungicida e inseticida na qualidade sanitária e no desempenho fisiológico de sementes de soja antes e após o armazenamento. Sementes das cultivares NS 7494, NS 8693 e NS 7338 IPRO foram analisadas separadamente por um delineamento inteiramente casualizado em esquema fatorial 3 x 6, sendo três aplicações de tratamento químico: 1) tratadas e avaliadas; 2) tratado, armazenado e avaliado; 3) armazenado, tratado e avaliado; e seis misturas de fungicidas e inseticidas: (Imidacloprido + Tiocarbe) + (Carbendazim + Tiram), (Imidacloprido + Tiocarbe) + (Metalaxil-M + Fludioxonil), Tiametoxam + (Carbendazim + Tiram), Tiametoxam + (Metalaxil-M + Fludioxonil) ), (Fipronil + Piraclostrobina + Metil-tiofanato) e o controle, que foi adicionado apenas água. Os testes de sanidade, germinação, frio e envelhecimento acelerado foram avaliados. Os produtos de tratamento de sementes requerem pelo menos dois meses para serem eficazes no controle de Penicillium spp e Fusarium spp. As misturas contendo Carbendazim + Tiram em sua composição são eficientes no controle de patógenos, independentemente do tempo de aplicação dos produtos.

Abstract: Fungi are considered as the most important among pathogens due to the higher number of species and to the damage caused both in yield and in seed quality. Thus, aimed to verify the effect of fungicide and insecticide treatment on sanitary quality and physiological performance of soybean seeds before and after storage. Seeds from cultivars NS 7494, NS 8693 and NS 7338 IPRO were used, which were analyzed separately by a completely randomized design in a 3 x 6 factorial design, being three chemical treatment applications: 1) treated and evaluated; 2) treated, stored and evaluated; 3) stored, treated and evaluated; and six mixtures of fungicides and insecticides: (Imidacloprido + Tiocarbe) + (Carbendazim + Tiram), (Imidacloprido + Tiocarbe) + (Metalaxil-M + Fludioxonil), Thiamethoxam + (Carbendazim + Tiram), Thiamethoxam + (Metalaxil-M + Fludioxonil), (Fipronil + Piraclostrobina + Metil-tiofanato) and the control, which was added only water. The healthy test, germination, cold and accelerated aging tests were evaluated. Seed treatment products require at least two months to be effective in controlling Penicillium spp. and Fusarium spp. The mixtures containing Carbendazim + Tiram in its composition are efficient in the control of pathogens regardless of the application time of products.

Resumo: O momento de aplicação do tratamento de sementes é importante para a manutenção da qualidade de sementes de soja. Assim, objetivou-se verificar o efeito do tratamento fungicida e inseticida na qualidade de sementes de soja antes e após o armazenamento. Utilizou-se sementes das cultivares NS 7494, NS 8693 e NS 7338 IPRO, que foram analisadas separadamente, em esquema fatorial de 3x6. Três momentos de aplicação em que as sementes foram: tratadas e avaliadas; tratadas, armazenadas e avaliadas; armazenadas, tratadas e avaliadas; e seis combinações de inseticidas e fungicidas: Cropstar(r) + Derosal Plus(r); Cropstar(r) + Maxim xl(r); Cruiser(r) + Derosal Plus(r); Cruiser(r)+ Maxim xl(r); Standak Top(r) e o controle. Realizou-se testes de germinação, emergência, envelhecimento acelerado, frio e sanidade. Concluiu-se que as misturas com Cruiser(r) não afetam a qualidade fisiológica de sementes de soja tratadas e avaliadas, e tratadas após dois meses de armazenamento. Cropstar(r)+ Derosal Plus(r)mantém a qualidade fisiológica de sementes de soja armazenadas tratadas por dois meses, enquanto que o Standak Top(r)tem efeito negativo. As misturas com Cropstar(r) prejudicam a qualidade fisiológica de sementes de soja tratadas após dois meses de armazenamento. O fungicida Derosal Plus(r) melhora a qualidade sanitária de sementes de soja independentemente do momento de tratamento.

Abstract: The timing of seed treatment application is important to keep soybean seeds quality. Therefore, the aim of this study was verify the effect of fungicides and insecticides treatment in soybean seeds quality before and after storage. Seeds of NS 7494, NS 8693 and NS 7338 IPRO were utilized and analyses separately, through a factorial scheme 3x6, with three application moments: treated and assessed; treated, stored and assessed; stored, treated and assessed; and six combination of fungicides and insecticides: Cropstar(r) + Derosal Plus(r); Cropstar(r) + Maxim xl(r); Cruiser(r) + Derosal Plus(r); Cruiser(r)+ Maxim xl(r); Standak Top(r) and the control group. Germination, seedling emergence, accelerate aging, cold and health tests were performed. It was determined that the combination with Cruiser(r) doesn't affect the physiological quality of soybean seeds treated and assessed, and treated after two month of storage. Cropstar(r) + Derosal plus(r) keeps physiological quality of soybean seeds stored and treated for two months, while Standak top(r) has negative effect. The combination with Cropstar(r) damages the physiological quality of soybean seeds treat after two months of storage. The fungicide Derosal plus(r) improves the health quality of soybean seeds regardless treatment moment.

Resumo: A análise da atividade enzimática é uma importante ferramenta no monitoramento e caracterização da qualidade fisiológica de sementes e no entendimento da deterioração. Objetivou-se com este trabalho estudar a expressão de isoenzimas aliada a qualidade de sementes de milho híbrido colhidas com diferentes teores de água e submetidas ao tratamento químico. O delineamento utilizado foi o inteiramente casualizado, com quatro repetições, em esquema fatorial 3x2, sendo três teores de água (45%, 40% e 35%), e duas formas de manejo das sementes (tratadas e não tratadas). Foram utilizadas sementes de milho dos híbridos: BM 810, semiduro e BM 3061, dentado, colhidas manualmente em espigas. O tratamento químico foi realizado com os produtos comerciais Maxin(r) + K-obiol(r) + Actellic(r). A qualidade das sementes foi avaliada pelos testes de umidade, incidência de danos mecânicos, primeira contagem de germinação, germinação, emergência, índice de velocidade de emergência, tempo médio de emergência, envelhecimento acelerado e condutividade elétrica. As expressões isoenzimáticas foram avaliadas por meio dos sistemas superóxido dismutase (SOD), catalase (CAT), esterase (EST), álcool desidrogenase (ADH), malato desidrogenase (MDH), peroxidase (PO) e α-amilase. A expressão de isoenzimas é divergente dependendo do teor de água na colheita, do híbrido e da qualidade das sementes. O tratamento de sementes não interfere na expressão das isoenzimas.

Abstract: The analysis of isoenzyme activity is an important monitoring and characterization tool of the physiological quality of seeds and to understand the deterioration. The purpose of this work was to study the isoenzyme expression allied to the quality of maize hybrid seeds harvested at different moisture levels and subjected to chemical treatment. A completely randomized experimental design was used with four replicates, in a 3x2 factorial arrangement with three moisture levels (45%, 40% and 35%), and two forms of seeds tillage (with and without treatment). Seeds from maize hybrids, semi-hard BM 810 and dented BM 3061, were used. Seeds were manually gathered on ears. Chemical treatment was performed with commercial products Maxin(r) + K-obiol(r) + Actellic(r). Seed quality was assessed by moisture test, incidence of mechanicals damage, first count of germination, germination, emergence, emergence speed index, mean emergence time, accelerated aging, and electrical conductivity. Isoenzyme expressions were assessed by means of the systems superoxide dismutase (SOD), catalase (CAT), esterase (EST), alcohol dehydrogenase (ADH), malate dehydrogenase (MDH), peroxidase (PO) and α-amilase. Isoenzyme expressions are different, depending on moisture levels at harvest, the hybrid maize and seeds quality. Seeds treatment does not interfere in their isoenzymes expression.

O Mn pode estar associado à resistência das plantas aos fungos, por meio da lignificação ou inibição direta. Objetivou-se neste trabalho avaliar a aplicação foliar de Mn sobre a incidência de fungos, teores de Mn e de lignina nas sementes de soja oriundas de cultivares convencionais e de suas derivadas transgênicas resistentes ao glifosato (RR). O ensaio foi conduzido na Universidade Federal de Lavras, em blocos casualizados, com três repetições e esquema fatorial 4 x 4 x 2, sendo quatro cultivares de soja, duas convencionais e suas derivadas RR (BRS Celeste e BRS Baliza RR; BRSGO Jataí e BRS Silvânia RR), quatro doses de Mn via foliar (0; 200; 400 e 600 g Mn ha-1) e dois estádios de aplicação (R1 ou R3). Antes e após seis meses de armazenamento, as sementes foram submetidas ao teste de sanidade (Blotter test). Foram determinados os teores de Mn nas sementes e de lignina no tegumento. Verificou-se que a aplicação foliar de Mn proporciona menores incidências de Cercospora kikuchii, Fusarium spp. e Aspergillus spp. nas sementes de soja produzidas. O Mn aplicado via foliar na soja, tanto no estádio R1 como em R3, transloca até as sementes, mas não afeta a produção de lignina no tegumento das mesmas. Os teores de lignina no tegumento das sementes de soja são inferiores nas cultivares convencionais, Celeste e Jataí, em relação às suas derivadas transgênicas resistentes ao glifosato, Baliza RR e Silvânia RR.

Manganese may be associated with plant resistance to fungi, either through lignification or direct inhibition. The aim of this study was to evaluate the foliar application of Mn on the incidence of fungi and the levels of Mn and lignin in soybean seeds from conventional cultivars and their transgenic glyphosate-resistant derivatives (RR). The test was conducted at the Federal University of Lavras, in the state of Minas Gerais, Brazil, using a randomized block design with three replications in a 4 x 4 x 2 factorial scheme, comprising four soybean cultivars, two conventional and their RR derivatives (BRS Celeste and BRS Baliza RR; BRSGO Jataí and BRS Silvânia RR), four foliar dosages of Mn (0, 200, 400 and 600 g Mn ha-1) and two application stages (R1 and R3). Prior to, and after six months of storage, the seeds were subjected to a health test (Blotter test). The levels of Mn in the seeds and lignin in the seed coat were determined. It was found that foliar application of manganese affords a lower incidence of Cercospora kikuchii, Fusarium spp. and Aspergillus spp. in the soybean seeds produced. Foliar applications of Mn in the soybean, both at the R1 and R3 stages, translocates to the seeds, but does not affect lignin production in the seed coat. Levels of lignin in the seed coat of the soybean are lower in the conventional cultivars, Celeste and Jataí, in relation to their transgenic glyphosate-resistant derivatives, Baliza RR and Silvânia RR.

A colheita de sementes de milho tem sido recomendada com elevados teores de água, no entanto, após a colheita, as espigas são submetidas à despalha, sendo necessário avaliar a compatibilidade destes processos. O objetivo foi avaliar os efeitos da despalha mecânica na qualidade de sementes de milho colhidas com elevados teores de água. Foram utilizadas sementes de milho híbrido BM 3061 dentado, peneira circular 20/64, colhidas manualmente em espigas, com teores de água de 45%, 40% e 35%, despalha mecânica (despalhadora CWA, 312 rpm), secagem em secador estacionário a 35 ºC até atingirem 22% de teor de água e posterior secagem a 42 ºC até 12%. Procedeu-se a debulha seguida do tratamento químico com Maxin® + K-obiol® + Actellic®. A qualidade das sementes foi avaliada pelos testes de incidência de danos mecânicos, primeira contagem, germinação, emergência de plântulas, índice de velocidade de emergência, tempo médio de emergência, condutividade elétrica, envelhecimento acelerado, teste frio e teste de sanidade, a cada três meses, durante nove meses. A despalha mecânica interfere na qualidade das sementes de milho, dependendo da umidade das mesmas na colheita e do material estudado. Sementes desse híbrido podem ser colhidas com teores de água de 40% com utilização da despalha mecânica.

Harvest of maize seeds with high moisture levels has been recommended; nevertheless, after harvest, the ears undergo the process of husking and there is a need to assess the compatibility of these processes. The purpose of this study was to assess the effects of mechanical husking on the quality of maize seeds collected at high moisture levels. Dent BM 3061 hybrid maize seeds, manually collected from the ear, with moisture contents of 45%, 40% and 35%, and a 20/64 round sieve were used. The ears were mechanically husked (CWA machine, 312 rpm), dried in a stationary drier at 35 ⁰C until reaching 22% moisture, followed by drying at 42 ºC until reaching 12%. Then, they underwent shelling, followed by a chemical treatment with Maxin® + K-obiol® + Actellic®. Seed quality was assessed according to the occurrence tests of mechanical damage, first count, germination, seedling emergence, emergence speed index, average emergence time, electrical conductivity, accelerated aging, cold test, and seed health test at every quarter for 9 months. The mechanical husking affects maize seed quality, depending on the degree of seed moisture at harvest and on the material studied. This hybrid's seeds can be collected with seed moisture levels at 40% with the use of mechanical husking.

Objetivou-se avaliar a aplicação de ativador de defesa (Bion®, acibenzolar-S-metil) e fungicida foliar (Priori Xtra®, azoxystrobin + ciproconazole) no controle da ferrugem asiática (Phakopsora pachyrhizi) e o efeito da doença sobre a produtividade e a qualidade das sementes de soja. O delineamento foi em blocos casualizados em esquema fatorial 4 X 10 com três repetições, envolvendo quatro linhagens de soja, duas tolerantes e duas suscetíveis à ferrugem e dez manejos de controle, com a utilização de Bion® e Priori Xtra® e a combinação dos mesmos, aplicados uma, duas e três vezes, e uma testemunha. Avaliou-se o índice de vegetação por diferença normalizada, produtividade, massa de mil sementes e qualidade das sementes por meio dos testes de germinação, emergência de plântulas, envelhecimento acelerado e sanidade. A utilização de apenas ativador de defesa no número máximo de três aplicações não é eficiente no controle da ferrugem asiática. O fungicida utilizado em conjunto ou não com o ativador de defesa é eficaz no controle da doença e na melhoria da qualidade das sementes, quando em três aplicações. A desfolha causada pela ferrugem afeta a formação das sementes de soja, a produtividade e as porcentagens de germinação e emergência de plântulas, sendo influenciada pelo genótipo.

The objective of this study was to evaluate the influence of fungicide application (PrioriXtra®, azoxystrobin + ciproconazole) and defense activator (Bion®, acibenzolar-S-methyl) on the control of Asiatic rust (Phakopsora pachyrhizi), as well as the effect of the disease on the quality of soybean seeds. The experimental design was a randomized block in factorial 4 X 10 with three replications, with four soybean breeding lines, two disease-tolerant and two susceptible and ten managements of rust control, using the products Bion®, Priori Xtra® and their combination, applied once, twice and three times, and a control. It evaluated the rate of normalized difference vegetation, productivity, mass of 1000 seeds and seed quality by means of the germination, seedling emergence, accelerated aging and seed health tests. The use of the defense activator alone in the maximum of three applications is not effective in controlling rust. The fungicide used in conjunction or not with the defense activator is effective in controlling rust and improving seed quality, when used with three foliar applications. The defoliation caused by rust affects considerably the formation of seeds, as well as productivity and also the percentage of germination and seedling emergence, and is influenced by genotype.
