Efficient Synthesis and Antimicrobial Activities of Long Alkyl Chain Trifluoromethyl-1H-pyrazol-1-(thio)carboxamides and Trifluoromethyl-1H-pyrazol-1-yl-thiazoles

The synthesis of 3-alkyl-5-trifluoromethyl-1H- pyrazole-1-carboxamides, 3-alkyl-5-trifluoromethyl-1H-pyrazole-1-thiocarboxamides, and 2-(3-alkyl-5-trifluoromethyl-1H-pyrazol-1-yl)-thiazoles derivatives are reported. [3 + 2] cyclocondensations for a series of long alkyl chain 1,1,1-trifluoro-4-methoxyalk-3-en-2-ones and semicarbazide or thiosemicarbazide were carried out in ethanol, an eco-friendly medium. The series of trifluoromethyl-1H-pyrazol-1-yl-thiocarboxamide following a [3 + 2] cyclocondensation with 2-bromoacetophenone were converted into two series of 2-(3-alkyl-5-trifluoromethyl-1H-pyrazol-1-yl)-thiazoles. Good yields (69-96%) of the isolated products were obtained. The structures of the new long alkyl chain 1H-pyrazoles and 2-(1H-pyrazol-1-yl)thiazoles were characterized using 1H, 13C, and 19F nuclear magnetic resonance (NMR) spectroscopy and electrospray ionization tandem mass spectrometric (ESI MS/MS) data. Moreover, some of the products were evaluated for their antimicrobial activity against Gram-negative Escherichia coli American Type Culture Collection (ATCC) 35218, Salmonella enteritidis ATCC 13076, and Pseudomonas aeruginosa ATCC 15692; and Gram-positive Staphylococcus aureus ATCC 6538, methicillin resistant Staphylococcus aureus (MRSA clinical isolate), Streptococcus sp. (clinical isolate), and Candida albicans ATCC 14053 and Candida krusei ATCC 6258 fungi. All the tested 1H-pyrazoles exhibited antibacterial and antifungal activities at the tested concentrations. The compounds from series 4 were found to be powerful against MRSA.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Assessment of environmental quality through analysis of frequency of the black spot disease in an assemblage of fish, Guaíba lake, RS, Brazil

A frequência da doença dos pontos pretos, provocada por trematódeos digenéticos em peixes, foi testada como indicador da qualidade da água na bacia hidrográfica do lago Guaíba (RS). As amostragens foram padronizadas e realizadas trimestralmente com redes do tipo picaré em onze pontos desta bacia. Foram coletados e analisados 53.408 exemplares, totalizando 66 espécies pertencentes a 22 famílias e oito ordens. A maior frequência de ocorrência da doença foi verificada em Astyanax fasciatus. Um Qui-Quadrado simples aplicado para essa espécie indicou frequência estatisticamente significativa de ocorrência da doença para três locais amostrados. Dois destes locais apresentaram as maiores frequências durante dois períodos consecutivos de um ano de amostragem e os melhores níveis de qualidade de água. Os resultados deste trabalho sugerem que a associação entre a elevada frequência de infestação pelo parasito que causa a doença dos pontos pretos e os ecossistemas com menor degradação na qualidade da água, com registros de variáveis físicas, químicas e microbiológicas, pode ser utilizada como indicador de melhor qualidade ambiental.
The frequency of the black spot disease caused by digenetic trematodes in fish was tested as an indicator of the quality of water in the watershed of Lake Guaíba. Samples were standardised and quarterly made using a seine net at eleven sites in the basin. A total of 53,408 individuals of 66 specimens pertaining to 22 families and 8 orders were collected and analysed. The highest frequency of the disease was found in Astyanax fasciatus. The simple Chi-Squared test applied to the species showed statistically significant frequency of occurrence for three sampled sites. Two of these sites showed the highest frequencies for two consecutive sampling periods of one year and the best levels of water quality. Results of this study suggest that the association between high frequency of infestation by a parasite that causes the black spot disease in fish, and environments with less degradation in water quality, recorded from physical, chemicals and microbiological variables, could be used as an indicator of water quality in these environments.

Histopathologic alterations observed in fish gills as a tool in environmental monitoring

As brânquias de peixes apresentam uma grande superfície de contato com o meio externo e são particularmente sensíveis às mudanças químicas e físicas do ambiente aquático. Este trabalho descreve as alterações histopatológicas observadas nas brânquias das espécies Astyanax fasciatus e Cyanocharax alburnus, testando-se a presença de correlação entre o grau de severidade das alterações e degradação ambiental. Pode ser utilizada na avaliação da qualidade da água em programas de monitoramento. Brânquias de 107 exemplares de Astyanax fasciatus e 116 de Cyanocharax alburnus, coletados sazonalmente, foram preparadas com a técnica histológica de rotina para inclusão e impregnação em parafina e coradas com H&E. As principais alterações observadas em ambas as espécies foram alteração da estrutura do epitélio, vacuolização, hiperplasia das células do epitélio da lamela primária, elevação do epitélio, alteração da estrutura e ocorrência de aneurismas nas lamelas secundárias. Os pontos Gasômetro e F. do Celupa foram os que apresentaram as maiores frequências de intensidade de alterações moderadas e severas como as médias mais elevadas de IAH. Foi verificado que as alterações com maior grau de severidade estavam relacionadas aos ambientes mais degradados, indicando a presença de agentes estressores na água.
The gills of fish have a great external contact surface and are particularly sensitive to chemical and physical changes in the aquatic environment. The aim of this study was to examine the histopathologic alterations in the gills of Astyanax fasciatus and Cyanocharax alburnus and to determine if there is a correlation between the severity of the alterations and environmental degradation and if this biological system can be used as a tool for evaluating water quality in monitoring programmes. The gills of 107 specimens of Astyanax fasciatus and 116 of Cyanocharax alburnus were collected seasonally and processed using routine histologic techniques for fixing and embedding in paraffin and staining of sections with haematoxylin and eosin. The main alterations observed in both species were alteration of the structure of the epithelium, vacuolisation, hyperplasia of the epithelium of the primary lamella, epithelial lifting, and alteration of the structure and occurrence of aneurysms in the secondary lamella. The locations Gasometer and F. do Celupa were the ones that showed the highest frequencies of moderate and severe alterations as the highest "histopathologic alterations index" means. The most severe alterations were found to be related to the most impacted environment, indicating the presence of stressors in the water.

A manifestação de patologias infecciosas em populações de peixes costuma ser incrementada em ambientes onde são lançados despejos orgânicos. Exemplares da espécie Mugil platanus Günther, 1880 coletados em três pontos do rio Tramandaí foram utilizados para análise das alterações, e amostras de água foram tomadas durante o período de um ano para verificação da presença de bactérias. A observação macroscópica revelou ulcerações na região do pedúnculo caudal revestidas por uma massa de tecido amorfo e esbranquiçado. A análise histopatológica mostrou a presença de bactérias gram negativas, que provavelmente foram as responsáveis pela alteração da estrutura normal dos tecidos de revestimento do corpo. O resultado da análise estatística não paramétrica para a concentração de amônia mostrou variação significativa entre os três pontos amostrados bem como nas comparações múltiplas entre os pontos dois a dois. Este trabalho descreve as lesões cutâneas observadas em Mugil platanus e testa sua correlação com a concentração de amônia.
Manifestation of infectious pathologies in fishes usually increases in environments where organic wastes are disposed. Specimens of Mugil platanus Günther, 1880 and water samples collected at three points of the Tramandaí river were analyzed during a one year period. The macroscopic observation revealed ulcerations in the caudal peduncle area covered with a mass of amorphous and whitened tissues. Histopathologic analysis showed the presence of negative gram bacteria, probably responsible for alterations of the normal structure of the epidermic tissues. Non-parametric statistical analysis for ammonia concentration showed a significant variation among the three collected spots as well as in the multiple comparison between two spots. In this study, we describe cutaneous lesions observed in Mugil platanus specimens and tested their correlation with environmental ammonia concentration.

Little information is available about natural populations of fishes exposed to polluted waters in Brazil. Contaminants may be lethal or predispose fishes to sickness, being the analysis of aquatic organisms a good indicator of environmental quality. The violinha, Rineloricaria strigilata (Hensel, 1868), is a common loricariid catfish in the Guaíba lake basin, Rio Grande do Sul, Brazil, and has been selected as a bioindicator for being resident, occurring in more than 50% of the samples, and by showing externally detectable neoplasies. These are characterized by its dark pigmentation, bony plates hypertrophy, and hypertrophy and irregular arrangement of the odontodes. Histology showed hypertrophied epitelial tissues, with an increased number of secretory cells, blood cells, and melanina pigments, giving the dark appearance to the neoplasm. Observed frequencies of neoplasm were higher in lake Guaíba than in comparativo samples. Inside lake Guaíba, a pattern of distribution of neoplasm was observed, with the absence of neoplasm in fishes at the collected site that receives water from rio Jacuí, and frequencies of 3.04 to 16.81% in the collection localities in front of Porto Alegre and downstream localities, that receive water from rio Jacuí, rio dos Sinos and rio Gravataí, that drain urban and industrial area of Porto Alegre and neighborhood cities.
