Abstract Healthcare workers present an increased risk of contagion for the SARS-CoV-2 virus due to their labor exposure. Here, we describe the clinical, laboratory, and immunological characteristics of healthcare workers, before vaccine application, exposed to SARS-CoV-2-infected patients. We collected sociodemographic, clinical, and laboratory information from 50 professionals who worked during the COVID-19 pandemic at the Clinical Hospital of the Northwest in Brazil. The results showed that most workers are women, over 50 years old, and worked as nursing technicians. Approximately 56% of workers were positive for a previous infection by RT-PCR and/or anti-SARS-CoV-2-immunoglobulin tests. Increased levels of hematocrit, neutrophils, NK lymphocytes, and fibrinogen, were found in positive healthcare workers, suggesting a light inflammatory status. The immunological findings showed an increase in IL-17 production and a Th2/Th17/Th22 profile followed by high serology for anti-SARS-CoV-2 IgM and IgG. Those data reveal the importance of studies with healthcare workers to investigate if the continuous exposition to the virus may result in chronic activation of the immune system and/or pulmonary inflammation in this target group.

Accidental monensin poisoning in buffaloes in Bahia, Brazil

RESUMO: Monensina é um antibiótico ionóforo (AI) amplamente empregado na produção de ruminantes para promoção de crescimento e ganho de peso, mas que tem causado intoxicação em diversas espécies, incluindo os búfalos, principalmente, pelo desconhecimento ou desrespeito das recomendações de uso e às particularidades de cada espécie animal. Objetivou-se descrever, pela primeira vez na Bahia, dados clínico-epidemiológicos e anatomopatológicos de um surto de intoxicação acidental por monensina em búfalos e rediscutir a recomendação do uso de AI na produção de bubalinos. O surto acometeu um lote de 21 búfalos adultos após consumo de sobras de uma ração para bovinos formulada na fazenda e cujos constituintes eram misturados à mão. Os sinais clínicos e primeiros óbitos foram observados 24 horas após a ingestão dessa ração. O quadro clínico, em geral, se caracterizou por fraqueza muscular, tremores, dificuldade de locomoção e decúbito. Quinze búfalos apresentaram sinais clínicos de intoxicação (morbidade 71,5%), seguido de morte (letalidade 100%), após evolução aguda a subaguda (<24h até 96h). Exames laboratoriais indicaram acentuada elevação na atividade sérica das enzimas CPK e AST. Três búfalos foram necropsiados, sendo coletadas amostras de diversos órgãos para exame histopatológico. A principal lesão encontrada foi degeneração hialina e necrose segmentar multifocal nos músculos estriados esqueléticos e cardíacos (miopatia e cardiopatia degenerativo-necrótica tóxica multifocal polifásica). O diagnóstico foi confirmado pela avaliação toxicológica das sobras da ração suspeita, que detectou 461,67mg/kg de monensina. A morte de 71,5% dos búfalos deste lote ocorreu devido a uma sucessão de erros, que incluíram falhas na formulação da ração e, sobretudo, devido ao uso da monensina em uma espécie altamente sensível. Enfatizamos que, apesar dos possíveis efeitos benéficos do uso AIs como suplemento dietético para bubalinos, somos da opinião que os AIs nunca devem ser empregados na bubalinocultura, uma vez que os eventuais incrementos na produção não compensam o risco iminente de morte, devido a pequena margem de segurança para essa espécie e a inexistência de antídotos.
ABSTRACT: Monensin is an ionophore antibiotic (IA) widely used for growth promotion and weight gain in the production of ruminants. However, it has caused intoxication in several species, including buffaloes, mainly because of the ignorance or disrespect of the recommendations for use in each animal species. The objective of this study was to describe, for the first time, clinical-epidemiological and anatomopathological data of an outbreak of accidental poisoning by monensin in buffalos and rediscuss the recommendation of the use of IA in the production of this species. The outbreak affected 21 adult buffaloes after consumption of remains from a feed formulated on the farm and whose constituents were mixed by hand. Clinical and first death signs were observed 24 hours after ingestion of this food. In general, the clinical picture was characterized by muscle weakness, tremors, difficulty in locomotion, and decubitus. Fifteen buffaloes presented clinical signs of poisoning (71.5% morbidity), followed by death (100% lethality), after acute to subacute evolution (<24h to 96h). Laboratory tests indicated elevated serum activity of creatine phosphokinase and aspartate aminotransferase enzymes. Three buffaloes underwent necropsy, and samples from several organs were collected for histopathological examination. The main injuries found were hyaline degeneration and multifocal segmental necrosis in the skeletal and cardiac striated muscles (myopathy and degenerative-necrotic multifocal multifocal-necrotic cardiopathy). The diagnosis was confirmed by the toxicological evaluation of suspected ration remains, which detected 461.67mg/kg of monensin. The death of 71.5% buffaloes in this lot occurred due to a succession of errors, which included faults in the formulation of the ration and, above all, due to the use of monensin in a highly sensitive species. Despite the possible beneficial effects of IA use as a dietary supplement for buffaloes, we are of the opinion that IAs should never be used in bubalinoculture since any increment in production does not compensate for the imminent risk of death due to a small safety margin for this species and the absence of antidotes.

Abstract Health professionals working to mitigate the COVID-19 pandemic are one of the main risk groups for the disease, being prioritized for vaccination. Considering this, the aim of this study was to analyze the immune response of these professionals immunized with CoronaVac in the first and second doses. Blood samples were collected after the first and second doses of the vaccine (CoronaVac) and used to investigate hematological and biochemical parameters, analysis of immunoglobulin production, cytokines, and gene expression profile, as well as the identification of subsets of immune cells. Post-first dose immunological phenotypic memory (CD27+) profiles (T CD4+, TCD8+ and CD19+) showed a significant increase, as did Monocyte APCs (CD80+HLA-DR+) in relation to the second dose. The cytokines IL-2, IL-6 and IFN-° showed increased values in relation to the other analyzed cytokines. The Th2/Th17 profile in the second dose was characterized by gene expression analysis. The production of IgM and IgG after vaccination showed statistically significant values in the comparison between doses. CoronaVac showed activation of APCs monocytes, memory response of T and B lymphocytes, with immunoglobulins production. This set of responses is characterized by the Th2/Th17 immunological profile.

Effects of Zn Substitution on the Dispersion Spectrum of Permeability and Permittivity of Cu-Mn Ferrites in 1 MHz - 1 GHz Frequency Range

Abstract This work presents a study of Mn-Zn-Cu triphasic spinel ferrite according to Stoichiometry Cu(0.5-β) Znβ Mn0.5 Fe2O4 with β = 0.10, 0.20, 0.30 and 0.40 . The samples were produced by the conventional ceramic method of sintering, irrigated with partial oxygen pressure, following a determined thermal curve. The morphological, microstructural, dielectric and magnetic properties were characterized by X-ray Diffraction (XRD), Scanning Electron Microscopy (SEM), Energy Dispersive Spectroscopy (EDS) analysis and impedance measurements. The data obtained demonstrated the influence of stoichiometry on the dimensional morphological variation, density, grain size, and electromagnetic behavior in the proposed frequency range. Electromagnetic analysis revealed electrical (ε*) and magnetic (μ*) behaviors very similar to an almost linear decrease of the real and imaginary parts with the increase of frequency, with some oscillations in several parts of the curve, according to the different β. Thus, for the ferrites produced by the conventional ceramic method of powders oxides sintering, irrigated with partial oxygen pressure, within a determined thermal curve, we observed an important relationship between the different triphasic stoichiometric combinations of Mn-Zn-Cu, according to the different β, with their volume, density, grain size, and electromagnetic response in the frequency range from 1.0 MHz to 1GHz.

Myracrodruon urundeuva (Anacardiaceae) is the scientific designation of “aroeira-do-sertão”, a tree well dispersed at the northeastern Brazil. The medicinal use of this plant is widespread throughout the rural population and the inner trunk bark decoction is used to treat cutaneous affections, urinary and respiratory diseases, and gynecological problems. “Aroeira-do-sertão” has been included in the list of native species under extinction threat. In order to find a sustainable source of the plant, seedlings have been cultivated. The purpose of this work was to investigate the chemical composition of the decoction from leaves of cultivated plants (aqueous extract from Myracrodruon urundeuva (AEMU)) and its anti-inflammatory activity on human neutrophils. The addition of AEMU on cells suspension inhibited significantly its activation/degranulation induced by phorbol myristate acetate. AEMU since at 50 µg mL-1 showed better pattern of inhibitions when related to indomethacin, non-selective cyclooxygenase inhibitor used as reference anti-inflammatory. The decoction fractionation was performed by high-speed counter-current chromatography (HSCCC) and semi-preparative high-performance liquid chromatography (HPLC) using a phenyl column as the stationary phase, allowing the isolation of 24 compounds. These results showed how the AEMU is rich in bioactive metabolites, which probably contribute for the anti-inflammatory effects of this plant extract, modulating the degranulation of human neutrophils, one of the pro-inflammatory mechanism of these cells.

Currently, the pharmaceutical industry devotes great attention to drug degradation products because these compounds can offer risks to patients. A previous degradation study of betahistine (N-α-methyl-2-pyridylethylamine) conducted under different stress conditions detected three main impurities named A, B and C. Degradation products were analyzed by high-resolution mass spectrometry in electrospray source and time of flight analyzer (ESI-TOF) and nuclear magnetic resonance (NMR). Impurity mutagenicity was evaluated by Derek Nexus and Sarah Nexus softwares. Liquid chromatography hyphenate with tandem mass spectrometry (LC-MS/MS) analysis of the betahistine forced degradation sample indicated the presence of a new impurity, which was named impurity C1. 2D NMR experiments allowed the complete structural characterization of the new entity. The active pharmaceutical ingredient and degradation impurities were classified as inactive in the in silico mutagenic studies. Systematic investigation of a forced degradation sample led to the characterization of a new betahistine impurity. The in silico mutagenicity study of the betahistine degradation impurities may be useful in the risk assessment of the drug products.

RESUMO Alternaria solani, agente etiológico da pinta preta, é considerado um fitopatógeno de grande impacto econômico na cultura do tomate, gerando grandes prejuízos aos agricultores. O objetivo deste trabalho foi verificar o potencial antagonista por confronto direto, produção de voláteis e atividade antimicrobiana in vitro de filtrado de fungos sapróbios contra A. solani. Assim, confrontou-se o fitopatógeno A. solani com os fungos sapróbios Volutella minima, Memnoniella echinata, M. levispora, Curvularia eragrostidis, C. inaequalis, Gonytrichum chlamydosporium, G. macrocladum, Pseudobotrytis terrestris, Pithomyces chartarum, Lappodochium lageniforme, Dictyochaeta simplex, Stachybotrys nephrosfora e S. globosa, observando a formação de halo de inibição e agressividade dos fungos por meio de uma escala de nota. Foi avaliado também o crescimento do fitopatógeno na concentração 20% para selecionar filtrado de sapróbios com atividade antimicrobiana. Posteriormente os filtrados dos sapróbios selecionados foram estudados nas concentrações 5; 10; 15 e 20%, avaliando o crescimento micelial de A. solani. Os sapróbios L. lageniforme e G. macrocladum, apresentaram maior crescimento micelial em relação a A. solani sete dias após a repicagem do fitopatógeno. Aos 15 dias somente o L. lageniforme conteve o patógeno. No teste de confrontação direta os fungos L. lageniforme e G. macrocladum ocuparam uma maior área na placa de Petri que o patógeno. Os filtrados dos fungos G. macrocladum, C. inaequalis, P. terrestris, S. globosa e C. eragrostidis na concentração de 20% promoveram redução no crescimento micelial de A. solani de 34, 21, 19, 10 e 10%, respectivamente. Ao analisar o efeito das concentrações dos filtrados no crescimento micelial de A. solani, observou dose-dependência em todos os tratamentos, com máxima inibição do crescimento para os filtrados de G. macrocladum, C. inaequalis, P. terrestris, S. globosa e C. eragrostidis, respectivamente, na concentração 20%.
ABSTRACT Alternaria solani, the etiological agent of early blight, is considered a phytopathogen of great economic impact on tomato crop, causing great losses to farmers. The aim of this study was to assess the antagonistic potential for direct confrontation, the production of volatile compounds and the in vitro antimicrobial activity of saprobic fungal filtrate against A. solani. Thus, the phytopathogen A. solani was confronted with the saprobic fungi Volutella minima, Memnoniella echinata, M. levispora, Curvularia eragrostidis, C. inaequalis, Gonytrichum chlamydosporium, G. macrocladum, Pseudobotrytis terrestris, Pithomyces chartarum, Lappodochium lageniforme, Dictyochaeta simplex, Stachybotrys nephrosfora and S. globosa, to observe inhibition zone formation and aggressiveness of the fungi according to a score scale. The phytopathogen growth was also evaluated at the concentration of 20% to select the saprobe filtrate with antimicrobial activity. Subsequently, the selected saprobe filtrates were studied at the concentrations of 5; 10; 15 and 20%, evaluating the mycelial growth of A. solani. The saprobes L. lageniforme and G. macrocladum showed greater mycelial growth, compared to A. solani, at seven days after transferring the phytopathogen. At 15 days, only L. lageniforme contained the pathogen. In the direct confrontation test, the fungi L. lageniforme and G. macrocladum occupied a larger area on the Petri dish, in contrast to the pathogen. The fungal filtrates G. macrocladum, C. inaequalis, P. terretris, S. globosa and C. eragrostidis, at a concentration of 20%, promoted a reduction in the mycelial growth of A. solani by 34, 21, 19, 10 and 10%, respectively. Analyzing the effect of the filtrate concentrations on the mycelial growth of A. solani, dose-dependence was observed in all treatments, with maximum growth inhibition for filtrates of G. macrocladum, C. inaequalis, P. terretris, S. globosa and C. eragrostidis, respectively, at the concentration of 20%.

Electroencephalographic findings in patients with major depressive disorder during cognitive or emotional tasks: a systematic review

Objective: Major depressive disorder (MDD) is a prevalent psychiatric condition characterized by multiple symptoms that cause great distress. Uncovering the brain areas involved in MDD is essential for improving therapeutic strategies and predicting response to interventions. This systematic review discusses recent findings regarding cortical alterations in depressed patients during emotional or cognitive tasks, as measured by electroencephalography (EEG). Methods: A search of the MEDLINE/PubMed and Cochrane databases was carried out using the keywords EEG and depression, confined to article title. Results: The studies identified reveal the frontal cortex as an important brain structure involved in the complex neural processes associated with MDD. Findings point to disorganization of right-hemisphere activity and deficient cognitive processing in MDD. Depressed individuals tend to ruminate on negative information and respond with a pattern of relatively higher right frontal activity to emotional stimuli associated with withdrawal and isolation. Conclusion: Patients with MDD may have altered dynamic patterns of activity in several neuroanatomical structures, especially in prefrontal and limbic areas involved in affective regulation. Identification of these alterations might help predict the response of patients to different interventions more effectively and thus maximize the effects both of pharmacotherapeutic and of psychotherapeutic strategies.

Due to the economic potential of camu-camu, considering its high concentration of vitamin C, the aim of this work was to evaluate the quality and genetic variability seven accessions of camu-camu conserved in genebank (BAG) of Embrapa Amazônia Oriental, in the state of Pará, Amazon region, Brazil. The fruits of camucamuzeiro were analyzed for physicochemical characterization (standard methodologies) and mineral composition. The data were subjected to multivariate statistical analysis, using the techniques of cluster analysis and principal component analysis. The formation of different groups for each genotype, which shows the genetic variability and the dissimilarity of the genotypes in the species data, may be used to guide the selection of promising genotypes to enrich the programs of genetic improvement of the camucamuzeiro. Ascorbic acid showed levels above 1000 mg 100 g-1, however, genotypes 4, 1 and 2 show the highest potential and the most promising nutritional capacity, but genotype 4 showed good characteristics for the moisture, acidity, carbohydrates, Cu and Zn and differs totally from others about the total soluble solids (TSS) and flavon-3-ol, features that make it the most promising genotype. It was possible to separate the seven different genotypes using multivariate analysis (hierarchical cluster analysis-HCA and principal component analysis-PCA).

Abstract Prostate cancer is the second most common cancer among men in western populations, and despite its high mortality, its etiology remains unknown. Inflammatory processes are related to the etiology of various types of tumors, and prostate inflammation, in particular, has been associated with prostate cancer carcinogenesis and progression. Human papillomavirus (HPV) is associated with benign and malignant lesions in the anogenital tract of both females and males. The possible role of HPV in prostate carcinogenesis is a subject of great controversy. In this study, we aimed to examine the prevalence of HPV infections in prostate carcinomas of patients from northeastern Brazil. This study included 104 tissue samples from primary prostate carcinoma cases. HPV DNA was purified and then amplified using MY09/11 and GP5+/GP6+ degenerate primer sets that detect a wide range of HPV types, and with specific PCR primers sets for E6 and E7 HPV regions to detect HPV 16. None of the samples showed amplification products of HPV DNA for primer sets MY09/11 and GP5+/GP6+, or the specific primer set for the E6 and E7 HPV regions. HPV infection, thus, does not seem to be one of the causes of prostate cancer in the population studied.

RESUMO O objetivo deste trabalho foi avaliar, em ambiente protegido, a resposta de alface cultivar Vera, quanto à produção de massa seca comercial da parte aérea e ao acúmulo de macronutrientes em função da aplicação de diferentes doses de nitrogênio (N) e de fertilizante foliar à base de silicato de potássio. O cultivo de alface ocorreu entre os dias 10/08/2012 (semeadura) e 16/10/2012 (colheita). O delineamento foi inteiramente casualizado com três repetições referentes a nove tratamentos resultando de combinação entre doses em cobertura de N (9; 54; 90; 126 e 171 kg ha-1) e de Si e K2O (1,15; 6,90; 11,50; 16,10 e 21,85 kg ha-1) empregando a matriz Plan Puebla III e um tratamento controle sem fertirrigação. Utilizou-se fertirrigação por gotejamento para aplicação das doses. As maiores doses de N, Si e K2O resultaram na maior produção de massa seca comercial da parte aérea e na maior acumulação foliar de magnésio, cálcio, potássio e fósforo.
ABSTRACT The objective of this study was to evaluate, in greenhouse, the response of the lettuce cultivar 'Vera', with respect to the marketable shoot dry matter production and macronutrient accumulation, as a function of the application of different doses of nitrogen (N) and a potassium silicate-based leaf fertilizer. The lettuce was cultivated from August 10, 2012 (sowing) to October 16, 2012 (harvest). The experimental design was completely randomized with three replicates, referring to nine treatments, resulting from the combination between top-dressing doses of N (9, 54, 90, 126 and 171 kg ha-1) and Si/K2O (1.15, 6.90, 11.50, 16.10 and 21.85 kg ha-1), using the Plan Puebla III experimental matrix, and a control treatment without fertigation. Drip fertigation was used for the application of the doses. The highest doses of N and Si/K2O resulted in the highest marketable shoot dry matter production and highest leaf accumulation of magnesium, calcium, potassium and phosphorus.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Refrigeração do epidídimo canino a 4ºC e recuperação dos espermatozoides epididimários utilizando ACP-106c

Objetivou-se avaliar a qualidade dos espermatozoides recuperados da cauda do epidídimo após a refrigeração do complexo testículo-epidídimo (CTE) de cães usando o diluidor ACP-106c. Foram utilizados 60 cães machos adultos, com peso de 10-20 kg. Após a eutanásia, removeu-se o CTE que foi imerso em solução fisiológica 0,9% e transportado em caixa térmica ao laboratório a 30ºC. Para a refrigeração e recuperação dos espermatozoides epididimários, os 60 pares do CTE foram divididos em 4 grupos, de acordo com o tempo de refrigeração do CTE e posterior recuperação espermática: G0h, G6h, G12h e G18h, em que cada par do CTE permaneceu por zero, seis, doze ou dezoito horas a 4ºC, respectivamente. A recuperação dos espermatozoides da cauda do epidídimo foi realizada pela técnica de flutuação utilizando-se o diluidor ACP-106c ou Tris. Para cada epidídimo foi adicionado 1,0 mL de um dos dois diluidores, pré-aquecidos a 37ºC por 5 minutos. Em seguida foram centrifugados a 800g/5 minutos para remoção dos resíduos celulares. Avaliou-se a morfologia, funcionalidade e motilidade espermática total e progressiva, além de parâmetros obtidos pelo CASA. Os dados foram submetidos à ANOVA seguido do teste de Turkey (P < 0,05). Em todos os parâmetros avaliados, não houve diferença entre os diluidores testados (P>0,05). Os valores de motilidade total nos grupos G0h, G6h, G12h, e G18h para o ACP-106c foram 84,4±7,7; 81,6±11,6; 88,3±6,5 e 69,5±16,9, respectivamente, e para o Tris 85,2±8,7; 77,4±14,3; 79,0±17,8 e 65,4±17,9, respectivamente. Um decréscimo na qualidade espermática foi observado após 18 horas de refrigeração em ambos os diluidores. Dessa forma pode-se concluir que o ACP-106c pode ser utilizado para recuperar os espermatozoides epididimários refrigerados e podem ser viáveis por até 12h de refrigeração.
The study aimed to assess the quality of sperm recovered from the epididymal cauda after cooling the complex testis-epididymis (CTE) of dogs using ACP-106c extender. Sixty adult male dogs, weighing 10-20 kg were used. After euthanasia, CTE was removed and it was immersed in 0.9% saline and transported to the laboratory in cooler at 30ºC. For cooling and recovery of epididymal spermatozoa, the 60 pairs of CTE were divided into four groups, according to the refrigeration time of the CTE and subsequent sperm recovery: G0h, G6h, G12h and G18h, wherein each pair of CTE remained zero, six, 12 or 18 hours at 4ºC respectively. The recovery of sperm epididymal tail was conducted by flotation technique using ACP-106c or Tris extender. For each epididymis, it was added 1.0 mL of either extenders, preheated to 37ºC for 5 minutes. They were then centrifuged at 800g/5 minutes to remove the cell debris. Morphology, functionality and total and progressive motility, and parameters obtained by CASA were evaluated. Data were analyzed by ANOVA followed by Turkey test (P <0.05). In all parameters assessed, there was no difference between the extenders used (P>0.05). The values of total motility groups G0h, G6H, G12H, and G18h for ACP-106c were 84.4±7.7, 81.6±11.6, 88.3±6.5 and 69.5±16.9 respectively, and for Tris 85.2±8.7, 77.4±14.3, 79.0±17.8 and 65.4±17.9 respectively. A decrease in sperm quality was observed after 18 hours of cooling in both extenders. Thus it can be concluded that the ACP-106c may be used to recover the epididymal spermatozoa chilled and may be viable for up to 12h cooling.

Polyclonal antibodies to the putative coat protein of Citrus leprosis virus C expressed in Escherichia coli: production and use in immunodiagnosis

This work reports the in vitro expression of Citrus leprosis virus C (CiLV-C) putative coat protein (p29) and the production of a polyclonal antibody to be used as a tool for serological diagnosis of citrus leprosis. The ORF2/RNA1, corresponding to p29, was cloned in pET28a and transformed into Escherichia coli cells (BL21). Expression of p29 was induced in vitro and the protein was purified and used for immunization of rabbits to produce the polyclonal antibody. The anti p29 serum was shown to be highly specific to CiLV-C detection by immunological methods (Western blot, PTA-ELISA, tissue blot and in situ immunolocalization), without cross reaction with healthy citrus plants or other cytoplasmic and nuclear viruses transmitted by Brevipalpus mites. These results demonstrate that the antibody against CiLV-C p29 protein is highly specific for CiLV-C detection. In situ immunogold labeling assays on thin sections of sweet orange leaf cells infected by CiLV-C demonstrated that short, bacilliform particles present within cisternae of the endoplasmic reticulum were specifically labeled, confirming their viral nature. The dense cytoplasmic viroplasm was also heavily labeled indicating that it represents a site of p29 accumulation.

A Leishmaniose Visceral (LV) é causada por protozoários do gênero Leishmania e transmitida por flebotomíneos do gênero Lutzomyia, os quais vêm adaptando-se ao ambiente peridomiciliar, onde o cão é sua principal fonte de alimento, aumentando assim o risco de casos em humanos. Neste trabalho, foram utilizadas técnicas de geoprocessamento e de estastística espacial como contribuição à compreensão da dinâmica epidemiológica da LV na área urbana de Ilha Solteira-SP.
Visceral Leishmaniasis (VL) is caused by protozoan of genus Leishmania and transmitted by sand flies of genus Lutzomyia, which has been adapted to the peridomicile environment where dogs are their mainly food source, increasing the risk for human cases. In this study, techniques of geoprocessing and spatial statistics were utilized as a contribution to understanding the epidemiological dynamics of VL in the urban area of Ilha Solteira, SP.
