A espécie pinhão manso (Jatropha curcas L.), vem se destacando como a principal matéria-prima para produção de biodiesel. O objetivo deste estudo foi caracterizar dois isolados de Colletotrichum spp., obtidos de sementes dessa espécie, através de análise morfológica, cultural e molecular, além de avaliar a patogenicidade desses isolados em folhas e frutos da planta. Para análise morfológica foram medidos o comprimento e a largura de 30 esporos de cada isolado, produzidos em meio de cultura Extrato de Malte-Agar, a 25±1 ºC, sob luz contínua. A análise cultural foi realizada pelo crescimento do fungo em meio de BDA, com avaliações diárias do crescimento do micélio e pela coloração da colônia, após sete dias de incubação. O DNA dos isolados foi extraído e oligonucleotídeos específicos (região ITS4) foram identificados pela técnica de PCR e utilizados para identificar C. capsici (CcInt) e C. gloeosporioides (CgInt). A patogenicidade dos isolados foi avaliada em plantas com diferentes idades (10, 20, 30, 40, 50 e 60 dias após semeadura) e em frutos em seis estádios de maturação. Com base nestas análises os isolados foram identificados como C. capsici e C. gloeosporioides. Plantas jovens e frutos desenvolvidos foram mais suscetíveis à infecção pelas duas espécies do fungo.

The species known as physic nut (Jatropha curcas L.) has become important as one of main sources of feedstock for biodiesel production. The aims of this study were characterizing two different isolates of Colletotrichum spp. obtained from seeds of this species, through morphological, cultural, and molecular analyses; as well as assessing pathogenicity of both isolates on leaves and fruit of this plant species. For morphological analysis, length and width of 30 spores of each isolate, produced on medium Malt Extract-Agar (MEA), at 25 ± 1 ºC, and under constant light, were measured. Cultural analysis was performed by the fungus growth on PDA medium, through daily measurements of the mycelial growth, and the color of colonies after seven days incubation. DNA of the isolates was extracted and specific oligonucleotides primers (region ITS4) were identified by PCR and used to identify C. capsici (CcInt) and C. gloeosporioides (CgInt). Pathogenicity of isolates was assessed on plants aged 10, 20, 30, 40, 50, and 60 days after sowing and fruits at six maturation stages. Based on these analyses, isolates were identified as C. gloeosporioides and C. capsici. Young plants and the older fruits were more susceptible to infection by the two fungus species.

Objetivou-se determinar a taxa de transporte de população de fungos associados às sementes de pinhão manso, a patogenicidade desses microrganismos a plântulas e frutos e a transmissibilidade fruto-semente e semente-plântula. Avaliaram-se a taxa de transporte, por meio de blotter test, de sementes produzidas nos estados de Minas Gerais, São Paulo, Bahia e Tocantins. As sementes foram submetidas aos tratamentos: sem desinfestação com tegumento (SDCT), sem desinfestação sem tegumento (SDST), com desinfestação com tegumento (CDCT) e com desinfestação sem tegumento (CDST). A incidência (%) dos fungos foi avaliada sob microscópio estereoscópico binocular. Para o teste de patogenicidade em plântulas e frutos inocularam-se suspensões de 10(6) esporos e discos de BDA com micélio, respectivamente. Para os fungos fitopatogênicos avaliaram-se a transmissibilidade fruto-semente e semente-plântula. O tratamento SDCT permitiu a detecção de maior número de fungos. Os fungos identificados foram Colletotrichum gloeosporioides, C. capsici, Curvularia sp., Verticillium sp., Fusarium sp., Penicillium sp., Aspergillus sp., A. niger e Rhizopus sp. Apenas as espécies de Colletotrichum são patogênicas às plântulas e frutos. Para ambas espécies há transmissibilidade fruto-semente, entretanto não é observada transmissão semente-plântula.

The objective of the present study was to determine the transport rate of a fungus population associated with Jatropha curcas seeds and the pathogenicity of these microrganisms to seedlings and fruit and also the fruit-seed and seed-seedling transmissibility. The transport rate of seeds produced in Minas Gerais, São Paulo, Bahia and Tocantins states was evaluated using the blotter test. The seeds were submitted to the following treatments: without disinfestation, with tegument (SDCT), without disinfestation, without tegument (SDST), with disinfestation, with tegument (CDCT) and with disinfestations, without tegument (CDST). The incidence (%) of the fungi was assessed using a binocular stereomicroscope. Suspensions of 10(6) spores and PDA discs with mycelium were inoculated for the pathogenicity tests on seedlings and fruits, respectively. For the phytopathogenic fungi, the fruit-seed and seed-seedling transmissibility were evaluated. The SDCT treatment allowed the detection of a greater number of fungi. The identified fungi were Colletotrichum gloeosporioides, C. capsici, Curvularia sp., Verticillium sp., Fusarium sp., Penicillium sp., Aspergillus sp., A. Niger, Rhizopus sp. Only the Colletotrichum species is pathogenic to seedlings and fruit. For both species there is fruit-seed transmissibility but no seed-seedling transmission.

Objetivou-se detectar a influência do estádio fenológico e da idade da folha de soja na infecção por Phakopsora pachyrhizi, agente causal da ferrugemasiática (FA). Plantas das cultivares BRS 154 e BRS 258 foram inoculadas, com suspensão de 10(5) urediniósporos/mL, nos estádios fenológicos V3, R1 e R5. Após 24 horas de câmara úmida, as plantas foram acondicionadas em condições de casa de vegetação por 20 dias. Avaliou-se o período latente médio (PLM) e a severidade. Para a avaliação da suscetibilidade de trifólios à FA utilizou-se a cultivar BRS 154 (V5). A inoculação foi realizada nos quatro primeiros trifólios. Aos 15 dias após a inoculação, os folíolos foram avaliados quanto à severidade, tamanho médio de lesão e freqüência de infecção. O estádio das plantas de soja não influenciou no PLM. As cultivares BRS 154 e BRS 258 tiveram PLM de 8 e 9 dias, respectivamente. As cultivares não se diferenciaram quanto à severidade da doença. Não houve diferença de severidade nos estádios V3 e R1, porém, os valores de severidade nesses estádios foram superiores ao valor de severidade no estádio R5, na avaliação realizada 8 dias após a inoculação. Em relação à suscetibilidade de folhas, o trifólio mais velho apresentou maiores valores de doença.

This work was conducted to study the influence of soybean growth stage and leaf age on the infection of Phakopsora pachyrhizi, the soybean rust pathogen. Soybean plants (cv. BRS 154 and BRS 258) at the V3, R1 and R5 growth stages were inoculated with a 1 x 10(5) urediniospores per mL suspension. After a period of 24 hours in dew chambers, all plants were removed from the chambers and placed under greenhouse conditions for 20 days. Mean latent period (PLM) and disease severity were estimated. The susceptibility of trifoliate leaves to soybean rust was estimated on cv. BRS 154 at the growth stage R5. Pathogen inoculation was done at the first four trifoliate leaves. Fifteen days after inoculation, leaflets of each trefoil were evaluated for disease severity, lesion mean size and infection frequency. Plants' growth stage did not influence the PLM. Cultivars BRS 154 and BRS 258 presented PLM of 8 and 9 days, respectively. There was no difference in disease severity at the growth stages V3 and R1, but those values were higher than at the R5 growth stage, 8 days after inoculation. The oldest trefoil showed the highest disease values.
