Application of Euphorbia tirucalli sap in sarcoid treatment in horses - case report

RESUMO O sarcoide é a neoplasia cutânea mais comum nos equinos, não apresentando uma terapêutica estabelecida, devido à variável manifestação neoplásica e à oscilante resposta individual às terapias. A planta Euphorbia tirucalli, conhecida como Aveloz, possui em sua seiva terpenos e esteróis com atividade antitumoral, sendo amplamente empregada na medicina popular. Foram atendidos, no Hospital Veterinário, oito animais diagnosticados com sarcoide e tratados unicamente com a solução aquosa contendo a seiva de Euphorbia tirucalli. As regiões predominantemente acometidas pelos tumores eram base de orelha e peitoral; sete dos oito animais apresentaram tumores em mais de uma região. A injeção intratumoral contendo Aveloz foi aplicada até a turgência do tumor, visualizando-se, em 24 a 72 horas, o enegrecimento, o ressecamento e o desprendimento do sarcoide. O uso da seiva de Euphorbia tirucalli apresentou 87,5% de eficácia no tratamento de sarcoide, e foram observados efeitos adversos nos sete animais em que a técnica se mostrou eficaz. Os equinos tratados foram acompanhados no intervalo de três mês a sete anos e nenhum apresentou recidivas do tumor. A aplicação intralesional da seiva da Euphorbia tirucalli se apresentou eficaz, segura e de baixo custo no tratamento de sarcoide em equinos, sendo uma alternativa viável para esse tipo de neoplasia cutânea.
ABSTRACT Sarcoid is the most common cutaneous neoplasm in horses, with no established therapy, due to the variable neoplastic manifestation and the oscillating individual response to therapies. The Euphorbia tirucalli plant, known as Aveloz, has terpenes and sterols in its latex with antitumor activity, being widely used in popular medicine. Eight horses were referred to the Veterinary Hospital diagnosed with sarcoid, and they were treated only with an aqueous solution containing Euphorbia tirucalli. The most common location of tumors was base of the ear and pectoral; seven of the eight animals had tumors in more than one region. The intratumoral injection containing the sap of Aveloz was applied until the tumor became turgent, visualizing blackening, dryness, and a detachment of the sarcoid in 24 to 72 hours. The use of Euphorbia tirucalli sap showed 87.5% efficacy in the treatment of sarcoids and adverse effects were observed in the seven animals in which the technique was effective. The treated horses were evaluated for three months to seven years, and none had tumor recurrences. The intralesional application of Euphorbia tirucalli sap has been proved to be effective, safe, and low-cost in the treatment of sarcoid in horses, being a viable alternative for this type of cutaneous neoplasm.

Abstract Aim: This study aimed to evaluate the acute supplementation effects of capsaicin, caffeine, and the combined capsaicin plus caffeine on total volume (total repetitions x weight lifted), rate of perceived exertion (RPE), and side-effects on resistance training. Methods: Eleven men (21.5 ± 2.1 years, 1.75 ± 0.08 m, 79.64 ± 10.1 kg), trained in resistance training (experience of 4.5 ± 2.6 years, weekly frequency 5 ± 0.8 days) were recruited. This was a crossover, randomized, double-blind study. Each volunteer went through four experimental conditions: supplemented with capsaicin (12 mg), caffeine (400 mg), capsaicin plus caffeine (12 mg and 400 mg, respectively), or placebo. After supplementation, the volunteers completed four sets of back squats until failure, at 70% of one-repetition maximum, with 90 s of rest interval between sets. RPE was registered at the end of each set. Volunteers were asked about the occurrence of side effects, right after exercise and 24 h after supplementation. Results: No significant differences were found (p > 0.05) in total volume between placebo (5505.2 ± 810.7 kg), capsaicin (6010.0 ± 1067.0 kg), capsaicin plus caffeine (5885.1 ± 1219.3 kg), and Caffeine (5628.6 ± 894.4 kg). No significant differences were found in RPE (p > 0.05) between the experimental conditions. The effect size of the total volume was small in capsaicin and capsaicin plus caffeine (d = 0.62 e d = 0.47, respectively), and trivial in the caffeine condition (d = 0.15). Conclusion: The supplementation failed to influence resistance training performance as well as had no effects on increasing the total volume or reducing RPE.

Resumo: Objetivo As amebas testáceas são protistas de vida livre reconhecidas como bioindicadores, capazes de revelar alterações ambientais apresentando variação de riqueza, diversidade e abundância de espécies. Para avaliar a produção científica relativa ao uso de amebas testáceas como bioindicadores, foi realizada uma análise cienciométrica no período de 1960 a 2020. Métodos A seleção de artigos científicos foi feita nas plataformas Google Acadêmico, Scielo, Science Direct e Online Library. Todos os artigos selecionados foram categorizados por cronologia, periódico de publicação, país, afiliação dos autores, se o estudo era empírico ou experimental, se incluía apenas amebas testáceas e aspectos do meio ambiente. Resultados Um total de 215 artigos de noventa e dois periódicos revelou um aumento notável no número de publicações nas últimas décadas. Os dois periódicos que mais publicaram dados sobre o tema foram Microbial Ecology e Ecological Indicators. Embora o maior número de trabalhos tenha sido publicado no continente europeu, os países que mais contribuíram para o tema foram o Canadá na região temperada e o Brasil nos trópicos. Edward A.D. Mitchell publicou o maior número de estudos e em termos de instituição, a Universidade Franche-Comté. A maioria dos trabalhos que associaram amebas testáceas como bioindicadores foram empíricos, realizados tecamebas habitantes de musgos e, como tal, dissociadas de outros grupos. Conclusões Apesar do aumento verificado nas publicações com testáceas como bioindicadores, nossos resultados indicam uma concentração de estudos em regiões limitadas do globo. Sob outro aspecto, um grande número de estudos avaliou a comunidade de testáceas como um todo, mostrando uma escassez de conhecimento aprofundado sobre espécies e grupos funcionais. Com isso, nosso levantamento aponta para uma grande variedade de ecossistemas aquáticos desprovidos de estudos sobre o assunto, indicando a necessidade de fomentar a pesquisa sobre o papel das amebas testáceas como bioindicadoras, em especial nas regiões tropicais.
Abstract: Aim The free-living protists testate amoebae are recognized as being bioindicators, able to reveal environmental alteration via remodifying richness, diversity and abundance of species. To assess the scientific production on the use of testate amoebae as bioindicators, a scientometric analysis was undertaken from 1960 to 2020. Methods The sourcing of scientific articles derived from Google Academic, Scielo, Science Direct and Online Library research platforms. All selected papers were factored according to chronology, journal of publication, country, authors’ affiliation, whether the study was empirical or experimentation, if it included solely testate amoebae or other groups of organisms, and aspects of the environment. Results A total of 215 papers from ninety-two journals revealed a notable increase in publication numbers over the last decades. The two journals that most published data on this theme were Microbial Ecology and Ecological Indicators. Whilst the largest number of papers was published in the European Continent, the countries that most contributed to the subject were Canada in the temperate region and Brazil in the tropics. Edward A.D. Mitchell published the largest number of studies and regarding institutions the Franche-Comté University. The majority of papers that associated testaceans as bioindicators were empirical, conducted with Thecamoebian inhabiting moss and as such, dissociated from other groups. Conclusions Despite a suggested uptick in research of testaceans as bioindicators, our results indicate a concentration of studies to limited regions of the globe. In another aspect, a great number of studies assess testaceans’ community as-a-whole, showing a shortage of in-depth knowledge into species and functional groups. Therefore, our survey points to a wide variety of aquatic ecosystems devoid of study matter, indicating the need to foster the research on testate amoeba's role as bioindicators especially in the tropical regions.

The APOB rs693 polymorphism impacts the lipid profile of Brazilian older adults

The apolipoprotein B (APOB) gene contains several polymorphic sites described as risk modifiers for cardiovascular events. The objective of this study was to verify the association of the classic APOB Xba I polymorphism (rs693) with atherosclerotic risk factors in a segment of the Brazilian elderly population considering their usual dietary intake. Clinical and biochemical characteristics as well as total caloric and fat intake data were determined from 644 elderly individuals. Polymorphism analysis was performed by conventional polymerase chain reaction followed by enzyme restriction. Statistical analyses compared measures and proportions according to different APOB genotypic combinations. Statistically significant association was found between Xba I polymorphism and serum LDL, total cholesterol, and total lipid levels, with important elevations among T homozygotes compared to the other genotypes. There was homogeneity in all other parameters analyzed (including intake pattern), with a tendency for reduced levels of circulating apolipoprotein B among TT individuals. Our results pointed out that genetic variation in APOB affected the lipemic profile of elderly individuals in a context not biased by diet, generating a pattern suggestive of secretory disorder of lipoprotein particles, with possible implication in atherosclerotic risk.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The occurrence of Orthoreovirus, Rotavirus and chicken anemia virus in chickens of the poultry industry in Minas Gerais, Brazil

Avaliou-se a ocorrência de Orthoreovirus (ARV) e Rotavirus (AvRV) aviários na avicultura industrial de Minas Gerais. Foram colhidas cinquenta e quatro amostras de fezes de frangos de corte entre um e 45 dias e de frangas de postura de 10 a 13 semanas de idade. Para análise de ARV, o RNA foi imediatamente extraído (Trizol), transcrito em cDNA e avaliado em uma PCR com oligonucleotídeos iniciadores específicos para ARV. Para a investigação de AvRV, os extratos de RNA foram obtidos por fenol-clorofórmio e submetidos à eletroforese em gel de poliacrilamida. Todas as amostras foram também avaliadas para o DNA do vírus da anemia das galinhas (CAV) em uma nested-PCR específica. Em frangos de corte, a positividade encontrada para ARV foi de 5,55% e para AvRV de 9,25%. CAV foi detectado em coinfecção em um plantel com refugagem, claudicação e prostração. Nenhuma amostra de poedeiras foi positiva para ARV ou AvRV. Material de plantel com sinais clínicos foi purificado e inoculado em ovos SPF embrionados, sendo obtidas lesões hemorrágicas e focos brancos na membrana cório-alantóide. O sequenciamento dos produtos de PCR e de embrião agrupou os isolados de ARV com a estirpe S1133, historicamente usada como vacina viva. Os resultados sugerem a continuada circulação da infecção por estirpes assemelhadas a ARV S1133 nas regiões de avicultura industrial. Os índices de detecção de ARV, AvRV e CAV indicam que a intensificação nas regiões produtoras tem resultado em falhas de biosseguridade.
Fifty-four fecal samples taken from broiler chickens from 1 to 45 days of age, and of pullets from 10 to 13 weeks of age, original from eight different poultry regions in the state of Minas Gerais, Brazil, were collected from March 2008 to January 2010 for avian Orthoreovirus (ARV) and avian Rotavirus (AvRV) analyses. For the assay of ARV, RNA was immediately extracted (Trizolâ) and transcribed into cDNA for assaying in a nested-PCR with ARV-specific primers. For AvRV, polyacrylamide gel electrophoresis (PAGE) was performed with RNA extracts obtained by phenol-chloroform extraction. CAV was additionally investigated through a nested-PCR of thymus and spleen. Results found 5.55% positive for ARV and 9.25% for AvRV. Also, CAV and ARV genomes were detected in co-infection, in a highly prostrated and claudicating chicken flock. No ARV or AvRV infections were detected in pullets. Material of a clinically affected flock was inoculated into SPF embryos, resulting in embryonic hemorrhage, whitish foci in the chorio-allantoic membrane and death. Sequencing of ARV amplicons and isolate cDNA grouped local strains with the ARV S1133 strain, historically used in live vaccines, suggesting the continued circulation of this vaccine virus strain in intensive poultry regions. Detection rates for ARV and AvRV, as well as the presence of CAV, were additionally indicative of failing biosecurity strategies for the intensive poultry regions examined.
