Real-time polymerase chain reaction for detection and enumeration of Staphylococcus aureus and Streptococccus agalactiae using different milk samplings

ABSTRACT The objective of the present study was to evaluate the qPCR for detection and enumeration of Staphylococcus aureus and Streptococcus agalactiae using different milk samplings in comparison to the conventional microbiology. Four dairy herds with a history of subclinical mastitis caused by S. aureus and S. agalactiae were selected. Sampling approach included milk samples from bulk tank (BT), cow level (composite samples, CO), and mammary quarter level (MQ) from 785 lactating cows. Three consecutive monthly milk samplings were carried out, totaling 3347 MQ milk samples, 912 CO, and 12 from BT. All collected milk samples were subjected to conventional microbiology and qPCR for detection and enumeration of S. aureus and S. agalactiae. The qPCR showed 71.5% of diagnostic sensitivity for S. aureus isolated from MQ milk samples, 71.8% for CO, and 50% for BT milk samples compared with conventional microbiology methodology. Taken together, the diagnostic sensitivity for S. agalactiae isolated from MQ milk samples was 90.2, 87.7 for CO, and 90.9% for BT milk samples. In general, the qPCR methodology enabled the detection of S. aureus and S. agalactiae, regardless of the type of milk sampling. The direct use of milk samples to estimate the counting of S. aureus by qPCR demonstrated lower sensitivity than the counting of S. agalactiae, which can be explained by the pathogen infection dynamics and differences in milk sample type.

RESUMO No cultivo do inhame a adubação mineral e o uso de tutoramento proporciona boa produção. O objetivo deste trabalho foi avaliar a produção e a qualidade do inhame submetidos a doses de nitrogênio e tipos de tutoramento. O trabalho foi realizado em condições de campo no delineamento experimental em blocos casualizados em esquema fatorial 5 × 3, com cinco doses de nitrogênio (0, 50, 100, 150 e 200 kg ha-1) e três tipos de tutoramento (espaldeiramento, vara e sem tutor), com quatro repetições. Na adubação em cobertura foram aplicadas as doses de nitrogênio parceladas em partes iguais aos 60 e 90 dias após o plantio. Foram avaliados comprimento, massa média, teores de amido e N foliar, produtividades total e comercial de túberas. O tutoramento promoveu maior rendimento total, massa média, comprimento e teor de nitrogênio foliar, entretanto não foram observadas alterações no teor de amido entre os tipos de tutoramento. A produtividade comercial é maior com a utilização do tutor em vara. Recomenda-se o plantio do inhame com tutoramento e dose de nitrogênio de 120,70 kg ha-1 para promover a máxima produtividade.
ABSTRACT Adequate mineral fertilization and staking are necessary to obtain good productivity in yam cultivation. The objective of this study was to determine the increase in yield and quality of yams subjected to nitrogen doses and staking methods. This study was carried out under field conditions using a randomized block design in a 5 × 3 factorial scheme, with five nitrogen doses (0, 50, 100, 150 and 200 kg ha-1) and three staking methods (wire trellises, single staking, and without staking) with four replicates. Nitrogen doses were applied in topdressing, and split into equal parts at 60 and 90 days after seed tuber planting. Tuber length and weight, starch and leaf nitrogen concentrations, and total and marketable yields were determined. Staking promoted higher total yield, mass, length, and leaf nitrogen concentration, but no changes were observed in starch concentration among the staking methods. Commercial productivity was higher with single staking. Planting yam with staking and a nitrogen dose of 120.70 kg ha-1 is recommended to promote maximum productivity.

ABSTRACT The Atlantic Forest is made up of different vegetation types over extensive latitude. Most of its remnants are in mountain ranges, which are responsible for the maintenance of most endemic and threatened species in this domain. Here we present the floristic composition of the Área de Proteção Ambiental Mestre Álvaro (APAMA), a coastal massif at Espírito Santo State with expressive altitudinal amplitude (100-800 a.s.l.) containing remnants never floristically studied. Knowing the composition of this protected area is important because it includes an altitudinal range not yet evaluated in floristic comparisons involving highland and lowland forests between the latitudes 18-21° S. We found 493 species (in 292 genera and 91 families): ten are regional endemic species, nine are new records of occurrence for the state, and 52 are under some degree of threat of extinction. Our results indicate the presence of flora elements' typical of both highland and lowland forests, which makes the APAMA essential in a scenario extremely fragmented. Finally, our data provide an unprecedented floristic list for regional forest restoration projects, as well as an invaluable source of data for biogeographic studies that seek to understand the effect caused by neighboring vegetation types on the composition of lowland forests.

Resumo Fundamento: O exercício físico tem sido considerado uma importante terapia não farmacológica para a prevenção e tratamento das doenças cardiovasculares. No entanto, seus efeitos na remodelação cardíaca leve não são claros. Objetivo: Avaliar a influência do exercício aeróbico sobre a capacidade funcional, estrutura cardíaca, função ventricular esquerda (VE) e expressão gênica das subunidades da NADPH oxidase em ratos com infarto do miocárdio pequeno (IM). Métodos: Três meses após a indução do IM, ratos Wistar foram divididos em três grupos: Sham; IM sedentário (IM-SED); e IM exercício aeróbico (IM-EA). Os ratos se exercitaram em uma esteira três vezes por semana durante 12 semanas. Um ecocardiograma foi realizado antes e após o treinamento. O tamanho do infarto foi avaliado por histologia e a expressão gênica por RT-PCR. O nível de significância para análise estatística foi estabelecido em 5%. Resultados: Ratos com IM menor que 30% da área total do VE foram incluídos no estudo. A capacidade funcional foi maior no IM-EA do que nos ratos Sham e IM-SED. O tamanho do infarto não diferiu entre os grupos. Ratos infartados apresentaram aumento do diâmetro diastólico e sistólico do VE, diâmetro do átrio esquerdo e massa do VE, com disfunção sistólica. A espessura relativa da parede foi menor no grupo IM-SED do que nos grupos IM-EA e Sham. A expressão gênica das subunidades NADPH oxidase NOX2, NOX4, p22phox e p47phox não diferiu entre os grupos. Conclusão: Infarto do miocárdio pequeno altera a estrutura cardíaca e a função sistólica do VE. O exercício aeróbico tardio pode melhorar a capacidade funcional e a remodelação cardíaca por meio da preservação da geometria ventricular esquerda. A expressão gênica das subunidades da NADPH oxidase não está envolvida na remodelação cardíaca, nem é modulada pelo exercício aeróbico em ratos com infarto do miocárdio pequeno.
Abstract Background: Physical exercise has been considered an important non-pharmacological therapy for the prevention and treatment of cardiovascular diseases. However, its effects on minor cardiac remodeling are not clear. Objective: To evaluate the influence of aerobic exercise on the functional capacity, cardiac structure, left ventricular (LV) function, and gene expression of NADPH oxidase subunits in rats with small-sized myocardial infarction (MI). Methods: Three months after MI induction, Wistar rats were divided into three groups: Sham; sedentary MI (MI-SED); and aerobic exercised MI (MI-AE). The rats exercised on a treadmill three times a week for 12 weeks. An echocardiogram was performed before and after training. The infarction size was evaluated by histology, and gene expression was assessed by RT-PCR. The significance level for statistical analysis was set at 5%. Results: Rats with MI lower than 30% of the LV total area were included in the study. Functional capacity was higher in MI-AE than in Sham and MI-SED rats. The infarction size did not differ between groups. Infarcted rats had increased LV diastolic and systolic diameter, left atrial diameter, and LV mass, with systolic dysfunction. Relative wall thickness was lower in MI-SED than in the MI-AE and Sham groups. Gene expression of the NADPH oxidase subunits NOX2, NOX4, p22phox, and p47phox did not differ between groups. Conclusion: Small-sized MI changes cardiac structure and LV systolic function. Late aerobic exercise is able to improve functional capacity and cardiac remodeling by preserving the left ventricular geometry. NADPH oxidase subunits gene expression is not involved in cardiac remodeling or modulated by aerobic exercise in rats with small-sized MI.

Abstract Wickerhamomyces anomalus LBCM1105 is a yeast isolated from cachaça distillery fermentation vats, notable for exceptional glycerol consumption ability. We report its draft genome with 20.5x in-depth coverage and around 90% extension and completeness. It harbors the sequences of proteins involved in glycerol transport and metabolism.

Resumo Fundamentado na Gramática Textual-Interativa, este trabalho discute o reconhecimento de uma unidade linguística essencialmente sistemática no domínio da organização textual da língua. Para tanto, investiga a Organização Tópica em editoriais paulistas do século XXI, objetivando avaliar se, em editoriais, a organização interna de Segmentos Tópicos mínimos (SegTs), constituinte da Organização Tópica, configura-se como um processo sistemático, podendo ser descrito segundo uma regra geral de organização, ou se é um processo essencialmente variável. Seguindo o método de análise tópica, que possibilita a análise textual com base na categoria do tópico discursivo, analisa 25 editoriais dos jornais Folha de S. Paulo e O Estado de S. Paulo e mostra que os SegTs mínimos nos editoriais se organizam segundo um mesmo princípio, que envolve a combinação potencialmente recursiva de duas unidades de organização intratópica - posição e suporte, o que permite defender que o texto é altamente regular, fundado em propriedades constitutivas próprias.
Abstract Based on the theoretical framework of Textual-Interactive Grammar, this work discusses the recognition of a basically systematic linguistic unit in the field of the textual organization of language. Therefore, it investigates the Topic Organization in newspaper editorials of the state of São Paulo in 21 st century, with the aim of evaluating if, in editorials, the internal structuring of minimal Topic Segments (SegTs), part of the Topic Organization, is configured as a systematic process, which can to be described according to a general rule of organization, or if is an essentially variable process. Using the method of topic analysis that enables the textual analysis based on the category of the discourse topic, the paper analyzes 25 editorials from newspapers Folha de S. Paulo and O Estado de S. Paulo, and it shows that the minimal SegTs in the editorials are organized according with a single principle, that involves the potentially recursive combination of two units of intratopic organization - position and support, which allows arguing the text is highly regular, founded in own constitutive properties.
Resumen Basado en el marco teórico de la Gramática Textual-Interactiva, este trabajo discute el reconocimiento de una unidad lingüística esencialmente sistemática en el ámbito de la organización textual de la lengua. Para ello, investiga la Organización Tópica en editoriales del Estado de São Paulo del siglo XXI, con el objetivo de evaluar si, en editoriales, la organización interna de Segmentos Tópicos mínimos (SegTs), que es parte da la Organización Tópica, se constituye como un proceso sistemático que puede ser descrito según una regla general de organización, o si es un proceso esencialmente variable. Desde el método de análisis tópica, que posibilita el análisis textual basado en la categoría del tópico discursivo, analiza 25 editoriales de los periódicos Folha de S. Paulo y O Estado de S. Paulo, y enseña que los SegTs mínimos en los editoriales se organizan conforme el mismo principio, que comprende la combinación potencialmente recursiva de dos unidades de organización intratópica - posición y sustentación, lo que permite decir que el texto es altamente regular, estructurado según propiedades propias de constitución.

Effects of grape maturity, maceration length during winemaking and interaction of both of them on physicochemical parameters, phenolic and volatile composition were investigated in tropical Syrah wines with the aim of finding the best conditions to produce wines that are rich in phenols and positive aroma active compounds. A headspace solid phase micro extraction and gas chromatography with mass spectrometry (HS-SPME-GC-MS) method was improved and validated for the analysis of 41 volatiles. Principal component analysis (PCA) and analysis of variance (ANOVA)-PCA coupled with inspection of statistically significant loadings were important to access the effects of maturation and maceration on wine composition. Wines made from riper grapes (21-23 ºBrix) stood out in relation to color intensity, total phenolic compounds, and presented high levels of anthocyanins, flavonols, procyanidin A2, and linalool (floral). Thirty days of maceration were linked to higher levels of some flavan-3-ols, gallic acid, a few esters (fruity) and alcohols, (E)-nerolidol (floral), while nonanoic acid (unpleasant aroma) reduced its concentration with prolonged maceration.

Resumo Shewanella xiamenensis G5-03 isolada de solo contaminado de aterro sanitário foi eficiente em descolorir cinco corantes têxteis azos em condições estáticas. Um deles, Reactive Red 239 (RR239), foi completamente descolorido na faixa de pH de 7 a 11 (a 35°C) dentro de 3-6 h. A bactéria também foi eficiente em descolorir RR239 em uma ampla faixa de temperatura de 25-40°C (em pH 8). A cinética de descoloração do RR239 pela G5-03 se ajustou ao modelo de Michaelis-Menten (K m = 443,3 mg L-1, V max = 166,7 mg L-1 h-1). A descoloração do RR239 foi monitorada por espectroscopia UV-Vis e FTIR, que mostrou mudanças significativas nas posições dos picos quando comparadas ao espectro do corante. No geral, a capacidade de S. xiamenensis G5-03 para descolorir os corantes têxteis azo em uma ampla faixa de temperatura e pH (neutro-alcalino) indica que esta cepa é uma candidata em potencial para o tratamento de efluente contendo corante.
Abstract Shewanella xiamenensis G5-03 isolated from contaminated landfill soil efficiently decolorized five textile azo dyes under static conditions. One of them, Reactive Red 239 (RR239), was completely decolorized at a pH range of 7 to 11 (at 35°C) within 3-6 h. The bacterium was also efficiently decolorized RR239 in a wide temperature range of 25-40°C (at pH 8). The kinetics of RR239 decolorization by G5-03 fitted to the Michaelis-Menten Model (K m = 443.3 mg L-1, V max = 166.7 mg L-1 h-1). The decolorization of RR239 was monitored by UV-Vis and FTIR spectroscopy, which showed significant changes in peak positions when compared to the dye spectrum. Overall, the ability of S. xiamenensis G5-03 to decolorize textile azo dyes in a wide range of temperatures and pH (neutral-alkaline) indicate that this strain is a potential candidate for treating dye-containing effluent.

Resumo O objetivo deste estudo foi avaliar as condições higiênico-sanitárias e o perfil da comunidade microbiana dos utensílios e das mesas de um serviço de alimentação localizado no município do Rio de Janeiro. A caracterização do processo de higienização dos utensílios (pratos, bandejas e talheres) e das mesas foi realizada por observação sistemática. Verificou-se que os utensílios eram lavados em máquina de lavar e as mesas, manualmente. Após a higienização, os utensílios apresentavam umidade e resíduos de alimentos. Pelo método dependente de cultivo, foram analisadas 126 amostras higienizadas (utensílios: n=90 e mesas: n=36). Pesquisaram-se bactérias mesófilas, coliformes, Escherichia coli, Staphylococcus aureus e fungos. Das amostras analisadas, 100% dos utensílios e 80% das mesas apresentaram contagens microbianas superiores ao recomendado na literatura, estando em condições higiênico-sanitárias inadequadas. E. coli foi isolada nos utensílios e S. aureus, nas mesas. Pelos métodos independentes de cultivo (PCR-DGGE e sequenciamento da subunidade 16S do rRNA), foram analisadas 36 amostras (utensílios: n=27 e mesas: n=9). Klebsiella sp. e Acinetobacter sp. Foram detectadas em todas as amostras, Citrobacter sp. sobre as mesas e Aeromonas hydrophila, nos talheres. Houve falha no processo de higienização, que foi confirmada pelas análises realizadas, que evidenciaram a presença de microrganismos indicadores e patogênicos, que podem causar a perda da qualidade das refeições, assim como danos à saúde dos comensais. Desta forma, é necessário adequar o processo de higienização, a fim de minimizar o risco de contaminação e o surto de doenças transmitidas por alimentos (DTA).
Abstract The objective of the study was to evaluate the hygienic-sanitary conditions and the microbial community profile of tables and tableware of a food service located in the city of Rio de Janeiro. The characterization of the sanitation process of the tableware (plates, trays and cutlery) and the tables was carried out by systematic observation. The tableware were washed in washing machine and the tables, cleaned by hand. After this process, the tableware presented moisture and food residues. For the culture-dependent method, 126 sanitized samples (tableware: n = 90 and tables: n = 36) were analyzed. Mesophilic bacteria, coliforms, fungi, Escherichia coli and Staphylococcus aureus were investigated. 100% of the sampled tableware and 80% of the sampled tables presented microbial counts higher than recommended in literature. The surfaces were in inadequate hygienic-sanitary conditions. E. coli was isolated in the tableware and S. aureus in the tables. By independent culture methods (PCR-DGGE and 16S rRNA sequencing), 36 samples (tableware: n = 27 and tables: n = 9) were analyzed. Klebsiella sp. and Acinetobacter sp. were detected in all samples, Citrobacter sp. was found on the tables and Aeromonas hydrophila in the cutlery. Results showed that the analyzed surfaces had indicative and pathogenic microorganisms, which can cause loss of quality in meals, as well as damages to the consumers’ health. Therefore, it is necessary to adapt the tableware and tables hygiene process in order to minimize the risk of contamination and food disease outbreaks.

Abstract Denture adhesives (DA) improve the retention and stability of ill-fitting dentures, especially for older adults. These materials should be biocompatible, i.e., they cannot cause undesired biological responses and be non-cytotoxic to oral tissues. However, in vitro testing of DA biocompatibility employing primary cell culture may possibly be affected by other factors, such as the donor age. Objective To compare the cytotoxicity of three different denture adhesives when assessed in primary gingival fibroblasts from a young donor or from an older donor, as well as the release of the basic fibroblast growth factor (bFGF), and the inflammatory response marker interleukin-6 (IL-6). Material and Methods Gingival fibroblasts isolated from a 30- and a 62-year-old donor were assayed for proliferation (1-7 days) and sensitivity to latex (positive control). Fibroblasts were indirectly exposed to Corega Ultra (cream), Corega powder and Fixodent Original for a 24 h period and assayed by XTT and Crystal Violet tests. The release of IL-6 and bFGF by exposed cells was determined by ELISA. Results While cells from the young donor presented higher cell growth after 7 days, the sensitivity to increasing concentrations of latex extracts was very similar between young and older cells. Both XTT and CVDE detected no difference between the DA and the control group. All materials induced higher levels of IL-6 and bFGF compared to control. Cells from the older donor exposed to Corega Ultra released lower levels of cytokine and growth factor. Conclusions All materials were considered non-cytotoxic, but affected cytokine and growth factor release. The biological differences found between fibroblasts from both donors could be due to individual or age-related factors. The authors suggest the use of cells from older donors on studies of dental products aimed at older patients, to better simulate their physiological response.

Mobilization of energetic substrates in the endangered catfish Steindachneridion parahybae (Siluriformes: Pimelodidae): changes in annual reproductive cycle in captivity

RESUMO O objetivo deste estudo foi analisar a composição do substrato energético (SE) nos principais tecidos de armazenamento de Steindachneridion parahybae, durante todo o ciclo reprodutivo em cativeiro. Diferentemente do ambiente natural, em cativeiro, a alimentação desses animais não é interrompida durante o período reprodutivo, as fêmeas não desovam espontaneamente, e são sedentárias. Fêmeas adultas foram amostradas mensalmente e baseada na histologia e no índice gonadossomático (IGS), os ovários foram classificados: estádios pré-vitelogênico (PRV), vitelogênico (VTG) e regressão (REG). Os ovários no estádio VTG apresentaram uma maior concentração de lipídeos e proteínas em relação ao estágio PRV. Esses substratos correlacionaram-se positivamente com o IGS. O músculo foi a principal fonte de proteína transferida aos ovários, como foi confirmado pela análise de correlação negativa entre esses órgãos. Os lipídeos mantiveram-se inalterados no fígado, considerado um importante órgão fornecedor de lipídeos para a vitelogênese, padrão que possivelmente ocorreu devido à contínua alimentação. A concentração do glicogênio muscular foi mais elevada durante os estágios VTG e REG em relação ao PRV. A concentração de triglicerídeos plasmática apresentou maiores valores no estádio REG enquanto a concentração de glicose no plasma foi maior durante os estádios VTG. Esses resultados sugerem que com alimentação constante, as fêmeas de S. parahybae apresentam um distinto padrão de mobilização dos substratos energéticos em relação ao que já foi descrito para outras espécies potamódromas de ambiente natural.
ABSTRACT This study aimed at analyzing the energetic substrate (ES) in the main storage tissues of Steindachneridion parahybae, throughout the reproductive cycle in captivity. Differently from wild, in captivity, feeding is not interrupted during the reproductive period, the females do not spawn spontaneously, and they are sedentary. Adult females were sampled monthly and based on their histology and gonadosomatic index (GSI), ovaries were classified into: previtellogenic (PRV), vitellogenic (VTG), and regression (REG) stages. Ovaries at the VTG stage showed higher protein and lipids levels than at the PRV stage with a positive correlation between these substrates and the GSI. Muscle was the main source of proteins transferred to the ovaries, according to the negative correlation between these organs. Lipids remained unchanged in the liver, which is an important supplier in vitellogenesis, a pattern that probably occurs due to the continuous feeding. Muscular glycogen levels were higher at the VTG and REG than at the PRV stages. Plasma triglycerides were also higher during REG, while glucose levels were more elevated during the VTG stage. These results suggest that with constant food supply, the pattern of deposition of ES in S. parahybae is different from that described for other wild potamodromous species.

Resumo Sarcocystis neurona e Neospora hughesi são protozoários coccídios que infectam equídeos e são responsáveis por doenças neurológicas nessas espécies. Neste estudo, a soroprevalência de infecção porS. neurona e Neospora spp. foi determinada em amostras de 333 soros de jumentos da Região Nordeste do Brasil. Anticorpos contra Neospora spp. foram detectados em 2% (7 jumentos) dos 333 animais examinados pela reação de imunofluorescência indireta (RIFI), com ponto de corte de 40. Anticorpos contra S. neurona foram detectados em 3% (10 jumentos) das amostras pela RIFI (ponto de corte de 50) e em 21% (69 jumentos) pela técnica de aglutinação direta (SAT - ponto de corte de 50). SAT e RIFI, para diagnóstico de S. neurona, apresentaram uma baixa concordância (Kappa = 0,051). Essa é a primeira observação de anticorpos anti-N. caninum em jumentos brasileiros e a primeira detecção de anticorpos contra S. neurona nessa espécie.
Abstract Sarcocystis neurona and Neospora hughesi are coccidian protozoa that can cause neurological illness in horses in America. In this study we report seroprevalence of Neospora spp. andS. neurona in sera of 333 donkeys from the northeastern region of Brazil. Antibodies to Neospora spp. were detected in 2% (7 donkeys) of 333 sera tested by the indirect fluorescent antibody test (IFAT) with a cut-off dilution of 1:40. Antibodies to S. neurona were found in 3% (10 donkeys) of the samples tested by IFAT (cut-off ≥50) and 21% (69 donkeys) by the direct agglutination test (SAT ≥50). The SAT and IFAT results for S. neurona showed a poor concordance (value of Kappa=0.051). This is the first report ofNeospora spp. antibodies in Brazilian donkeys and the first detection of antibodies against S. neurona in this animal species.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Short-term physiological responses to copper stress in Salvinia auriculata Aubl.

OBJETIVO: avaliar em curto tempo de exposição às respostas fisiológicas de Salvinia auriculata Aubl. sob diferentes concentrações de Cu. MÉTODOS: as plantas foram expostas a tratamentos com 0,0; 0,01; 0,1; 1 e 10 mM de Cu em um período de 2 dias. Em seguida foram avaliadas variáveis de desenvolvimento de S. auriculata (peso, pigmentos fotossintéticos e carboidratos solúveis), peroxidação lipídica (malonaldeído, aldeídos e extravasamento de eletrólitos) e produção de antioxidantes (antocianinas, carotenóides, flavonóides e prolina). RESULTADOS: Foram verificadas reduções das massas frescas nas concentrações acima de 1 mM de Cu. A clorofila a reduziu com o aumento da concentração de Cu, diferentemente da clorofila b. A razão clorofila a/clorofila b foi alterada, devido à degradação dos pigmentos fotossintéticos. As reduções dos carotenóides foram mais acentuada do que a de clorofila total. Os valores de extravasamento de eletrólitos variaram de 14 a 82 % e peroxidação lipídica de 7 a 46 nmol.g-1. Flavonóides e carboidratos solúveis mostraram reduções com o aumento da concentração de Cu. Antocianinas, compostos fenólicos e prolina quando submetidas a 0.1 mM de Cu apresentaram aumento, sugerindo adaptabilidade da planta ao estresse causado diretamente pelo metal e espécies reativas de oxigênio. Nas concentrações superiores, possivelmente, ocorreram degradação e/ou modificações diretas destas moléculas. CONCLUSÕES: Os dados sugerem que S. auriculata é provida de um mecanismo eficiente contra estresse causado por Cu na concentração de 0.1 mM. Já nas concentrações mais elevadas (1 e 10 mM), apesar de seu papel como micronutriente, Cu foi tóxico para a planta devido ao comportamento redox deste metal, que leva à formação exacerbada de espécies reativas de oxigênio, induzindo danos severos como degradação de membranas biológicas e desnaturação protéica.
AIM: To evaluate, in a short-time exposure, the physiological responses of Salvinia auriculata Aubl. under different concentrations of Cu. METHODS: The plants were exposed to treatments with 0.01, 0.1, 1 and 10 mM of Cu in a period of 2 days. Then development variables of S. auriculata (weight, photosynthetic pigments, and soluble carbohydrate), lipid peroxidation (malondialdehyde, aldehydes, and electrolyte leakage) and production of antioxidants (anthocyanins, carotenoids, flavonoids, and proline) were evaluated. RESULTS: It was observed fresh weight reductions in concentrations above 1 mM of Cu. Chlorophyll a decreased with the increase of Cu concentrations unlike chlorophyll b. The ratio chlorophyll a / chlorophyll b was changed due to the degradation of photosynthetic pigments. The reductions of carotenoids were more pronounced than that of total chlorophyll. The values of electrolyte leakage ranged from 14 to 82 % and lipid peroxidation from 7 to 46 nmol.g-1. Flavonoids and soluble carbohydrates showed reductions with the increase of Cu concentration. Anthocyanins, phenolic compounds, and proline when subjected to 0.1 mM of Cu had increased, suggesting adaptability of plant stress caused directly by metal and reactive oxygen species. In higher concentrations, degradation and/or direct modifications of these molecules possibly occurred. CONCLUSIONS: The data suggest that S. auriculata is provided with an efficient mechanism against stress caused by Cu in the concentration of 0.1 mM. As for higher concentrations (1 and 10 mM), despite its role as micronutrients, Cu was toxic to the plant due to the redox behavior of this metal, which leads to the exacerbated formation of reactive oxygen species, inducing to severe damage such as biological membrane degradation and protein denaturation.

A doença celíaca é definida como a intolerância às proteínas do glúten presente em certos cereais usados na produção de alimentos. Três formulações de biscoitos sem glúten, contendo Linum usitatissimum L. e diferentes concentrações de Chenopodium quinoa BRS Piabiru, foram desenvolvidos e avaliados em relação as caracterísiticas fisico-químicas, nutricionais e sensoriais. Não foi detectado glúten nos biscoitos formulados. O conteúdo de proteína bruta e lipídios totais variaram 85,58 a 97,55 e 121,69 a 166,19 g por kg de amostra, respectivamente. A variação da razão entre os ácidos graxos n-6:n-3 e poliinsaturados/saturados foi de 0.85:1 a 0.92:1 e 3.08:1 a 4.38:1, respectivamente. A Formulação C apresentou melhores teores de ácido alfa-linolênico, índices nutricionais da fração lipídica e conteúdo mineral por porção, com excelentes características sensoriais. A análise multivariada destacou o efeito da concentração de quinoa nas qualidades nutricionais e sensoriais do produto.
Celiac disease is defined as intolerance to the gluten proteins present in certain cereals used to prepare foodstuffs. We developed and performed physico-chemical, sensory, and nutritional assessments of three formulations of gluten-free cookies containing Linum usitatissimum L. and different levels of whole Chenopodium quinoa BRS Piabiru flour. No gluten was detected in the prepared cookie formulations. The crude protein and total lipid contents ranged from 85.58 to 97.55 and 121.69 to 166.19 g per kg of sample, respectively. The polyunsaturated/saturated and n-6:n-3 fatty acid ratios ranged from 0.85:1 to 0.92:1 and 3.08:1 to 4.38:1, respectively. Formulation C had the best alpha-linolenic acid content, lipid fraction nutritional indices and mineral content per portion, with excellent sensory characteristics. Multivariate analysis highlighted the effect of the concentration of quinoa on the nutritional and sensory qualities of the product.
