Abstract We describe the toxicity of multiwalled carbon nanotubes (MWCNTs) in two aquatic species: Danio rerio and Artemia salina. Fish were exposed to 25 mg L-1, 50 mg L-1, and 100 mg L-1 MWCNTs for 7 days. After exposure, the gills from D. rerio showed lamellar fusion and epithelial lifting in the secondary lamellae, and vasodilation in the primary lamellae. In the gills, heat shock protein 70 and Bax (pro apoptotic member) were stained more intensely in the treated groups than in the control group. The nauplii of A. salina were exposed to 25 µg L-1, 50 µg L-1, and 100 µg L-1 MWCNTs for 96 h. After exposure, the nauplii from the control group presented 20 % mortality, whereas the 25 µg mL-1, 50 µg mL-1, and 100 µg L-1 groups had 46.7 %, 70 %, and 100 % mortality, respectively. The nauplii exposed to 25 µg mL1 and 50 µg mL1 MWCNTs had black aggregates in the intestine because of the uptake of nanomaterials. In addition, the 25 µg mL-1 and 50 µg mL-1 MWCNT groups had black aggregates attached onto the body surface, appendages of the second pair, and mandibles.The results revealed the toxic effects of MWCNTs on the two aquatic organisms, and both species showed great potential for toxicological evaluation of MWCNTs in aquatic environments.

Abstract Would the general rules of Financial Law and Tax Law be the essential condition for the normative-institutional existence of Brazilian federalism from the perspective of the Constitution of the Federative Republic of Brazil of 1988? Based on the historical and dogmatic reconstruction of the function exercised by these normative species in our federalism, the present article aims to go beyond the discussion of classical theories around this phenomenon, pointing out the potentiating role of democracy at the federative level, especially in the with regard to the formation of provisional consensus. For the research, we adopted the methodological type inserted in the legal-comprehensive or legal-interpretative approach.

Resumo Seriam as normas gerais de Direito Financeiro e de Direito Tributário a condição essencial para a existência normativo-institucional do federalismo brasileiro sob a óptica da Constituição da República Federativa do Brasil de 1988? Tomando por base a reconstrução histórica e dogmática da função exercida por estas espécies normativas em nosso federalismo, o presente artigo tem como finalidade ir para além da discussão das teorias clássicas em torno desse fenômeno, apontando o papel potencializador da democracia no nível federativo, especialmente no que diz respeito à formação de consensos provisórios. Para a pesquisa, adotou-se o tipo metodológico inserido na abordagem jurídico-compreensiva ou jurídico-interpretativa.

The automotive industry is one of the largest industrial segments in the world market. The exhaust system of motor vehicles is responsible for the emission and treatment of toxic gasses released by the engine. In this sense, the application of titanium on an AISI 1020 steel substrate was carried out by plasma deposition using a cathode cage. The aim of this research was to evaluate the application of this material in the exhaust system of motor vehicles. The samples were characterized by scanning electron microscopy (SEM), confocal microscopy (CM), microhardness tests and corrosion resistance tests. The samples exhibited a thin film with higher titanium content and hardness than the uncoated sample. The corrosion potential also increased and the current density was lower than the uncoated sample. The conclusion is that the deposition of thin titanium films on AISI 1020 steel with CCPD has the potential to produce thin films.

BACKGROUND Leishmania RNA virus 1 (LRV1) is commonly found in South American Leishmania parasites belonging to the subgenus Viannia, whereas Leishmania RNA virus 2 (LRV2) was previously thought to be restricted to the Old-World pathogens of the subgenus Leishmania. OBJECTIVES In this study, we investigated the presence of LRV2 in strains of Leishmania (L.) infantum, the causative agent of visceral leishmaniasis (VL), originating from different hosts, clinical forms, and geographical regions. METHODS A total of seventy-one isolates were screened for LRV2 using semi-nested reverse transcription-polymerase chain reaction (RT-PCR) targeting the RNA-dependent RNA polymerase (RdRp) gene. FINDINGS We detected LRV2 in two L. infantum isolates (CUR268 and HP-EMO) from canine and human cases, respectively. MAIN CONCLUSIONS To the best of our knowledge, this is the first detection of LRV2 in the New World.

RESUMO Antecedentes: A Pandemia por COVID-19 tem trazido desafios subtanciais para a prática clínica no tratamento das doenças neuromusculares hereditárias (DNMh). A infecção não tem sido a única preocupação para os pacientes. O distanciamento social tem comprometido a assistência multidisciplinar, atividade física e tem trazido problemas mentais em decorrência do próprio isolamento. Nós apresentamos aqui um seguimento de 363 pacientes com DNMh de um centro terciário Brasileiro durante o pico da Pandemia de Covid-19. Objetivos: Mostrar a frequência e gravidade da infecção por Sars-Cov-2 em pacientes com DNMh e demonstrar os efeitos da pandemia nos hábitos de vida, na progressão da doença e no cuidado multidisciplinary. Métodos Trezentos e sessenta e três pacientes (58% homens and 42% mulheres) foram acompanhados por 3 meses através de 3 teleconsultas durante o pico da Pandemia de Covid-19 no Brasil. Resultados Houve um decréscimo no número de pacientes que faziam terapia física, respiratória e fonoaudiológica. Em muitos pacientes, o apetite (33%) e hábitos do sono (25%) se alteraram. Exercícios físicos e terapias foram interrompidas pela maioria dos pacientes. Physical exercises and therapies were interrupted for most of the patients. Eles relataram piora ou aparecimento de fadiga (17%), dor (17%), retrações (14%), e escoliose (7%). Irritabilidade, mudanças no sono, peso e apetite, sendo principalmente diminuição do apetite e peso foram mais frequentemente encontrados em pacientes que apresentaram piora clinica da doença. Houve uma baixa taxa de contaminação por Covid-19 (0.8%), e todos os pacientes infectado apresentaram quadro clinico leve. Conclusão O isolamento por si só se mostrou protetor na perspectiva de infecção por Covid-19, mas pode desencadear um cenário complexo com mudanças nos hábitos de vida e curso desfavorável da doença de base.

ABSTRACT Background The COVID-19 pandemic has brought substantial challenges for current practices in treating hereditary neuromuscular disorders (hNMDs). However, this infection has not been the only concern for these patients. Social distancing has compromised multidisciplinary assistance and physical activity, and has brought about several mental health issues. We presented a follow-up on 363 patients with hNMDs at a Brazilian tertiary center during the peak of the COVID-19 pandemic. Objective We aimed to show the frequency and severity of SARS-CoV-2 infection among hNMD patients and to demonstrate the effects of the pandemic on life habits, disease progression and multidisciplinary supportive care status. Methods Three hundred and sixty-three patients (58% male and 42% female) were followed for three months through three teleconsultations during the peak of the COVID-19 pandemic in Brazil. Results There were decreases in the numbers of patients who underwent physical, respiratory and speech therapies. For several patients, their appetite (33%) and sleep habits (25%) changed. Physical exercises and therapies were interrupted for most of the patients. They reported new onset/worsening of fatigue (17%), pain (17%), contractions (14%) and scoliosis (7%). Irritability and sleep, weight and appetite changes, and especially diminished appetite and weight loss, were more frequent in the group that reported disease worsening. There was a low COVID-19 contamination rate (0.8%), and all infected patients had a mild presentation. Conclusion The isolation by itself was protective from a COVID-19 infection perspective. However, this isolation might also trigger a complex scenario with life habit changes that are associated with an unfavorable course for the NMD.

BACKGROUND Leishmania (Mundinia) enriettii is a species commonly found in the guinea pig, Cavia porcellus. Although it is a dermotropic species, there is still an uncertainty regarding its ability to visceralise during Leishmania life cycle. OBJECTIVE Here, we investigated the ability of L. enriettii (strain L88) to visceralise in lungs, trachea, spleen, and liver of C. porcellus, its natural vertebrate host. METHODS Animals were infected sub-cutaneously in the nose and followed for 12 weeks using histological (hematoxilin-eosin) and molecular tools (polymerase chain reaction-restriction fragment length polymorphism - PCR-RFLP). To isolate parasite from C. porcellus, animals were experimentally infected for viscera removal and PCR typing targeting hsp70 gene. FINDINGS Histological analysis revealed intense and diffuse inflammation with the presence of amastigotes in the trachea, lung, and spleen up to 12 weeks post-infection (PI). Molecular analysis of paraffin-embedded tissues detected parasite DNA in the trachea and spleen between the 4th and 8th weeks PI. At the 12th PI, no parasite DNA was detected in any of the organs. To confirm that the spleen could serve as a temporary site for L. enriettii, we performed additional in vivo experiments. During 6th week PI, the parasite was isolated from the spleen confirming previous histopathological and PCR observations. MAIN CONCLUSION Leishmania enriettii (strain L88) was able to visceralise in the trachea, lung, and spleen of C. porcellus.

Resumo A partir da análise do sistema do Direito como sistema comunicativo, percebe-se a complexidade própria do fenômeno da Constituição Financeira. Esta é tomada como o acoplamento estrutural existente entre Política e Direito e que, em seu funcionamento orgânico, irrita diretamente o sistema da Economia. É a partir dessa visão de que a comunicação normativa da Constituição Financeira ocupa o papel central da estrutura estatal que o presente trabalho se apresenta no sentido de avaliar o valor da Constituição a partir de sua interrelação com a sociedade, com a normatividade e com os clássicos. Para construir o raciocínio, lançou-se mão de metodologia teorético-explicativa, especificamente com a contraposição de teorias e a extração dos principais significados de cada uma delas para revelar o que seria a Constituição Financeira.

Abstract From the analysis of the Law system as a communicative system, one can perceive the complexity of the phenomenon of the Financial Constitution. This is taken as the structural coupling between Politics and Law and, in its organic functioning, directly irritates the system of Economics. It is from this view that the normative communication of the Financial Constitution occupies the central role of the state structure that the present work presents itself in the sense of evaluating the value of the Constitution from its interrelationship with society, with normativity and with the classics. To construct the reasoning, theoretical-explanatory methodology was used, specifically with the counterposition of theories and the extraction of the main meanings of each of them to reveal what would be the Financial Constitution.

Resumo A atividade legislativa do Poder Executivo tem ganhado cada vez mais destaque nos sistemas jurídicos ocidentais. Sua contraposição aos princípios do Estado de Direito e da legalidade são a chave básica para avaliar o alcance de dada competência normativa. Para a presente análise, avaliar-se-á os desafios da crise do princípio da legalidade nos dias de hoje e, logo após, como se estrutura a atividade normativa do Poder Executivo no ordenamento jurídico brasileiro. Ao final, serão propostos os limites da atividade regulamentar em matéria de atos de intervenção de controle sobre o âmbito das relações desenvolvidas por particulares dentro do arcabouço de Direito Privado. Para tanto, lançou-se mão de método indutivo-hipotético, com viés explicativo-qualitativo.

Abstract Legislative activity of the Executive Branch has gained increasing prominence in Western legal systems. Its contrast with the principles of the rule of law and legality are the basic key to assess the scope of given normative competence. For the present analysis, we will evaluate the challenges of the crisis of the principle of legality nowadays and, soon after, how to structure the normative activity of the Executive Branch in the Brazilian legal system. In the end, the limits of the regulatory activity regarding acts of control intervention will be proposed on the scope of the relations developed by private individuals within the framework of Private Law. For that, an inductive-hypothetical method was used, with explanatory-qualitative bias.

OBJECTIVES: The objectives of this study were to determine the sensitivity of ovarian cancer (OC) cell lines (TOV-21G and SKOV-3) to cisplatin and to the recombinant human TRAIL (rhTRAIL), and to evaluate the expression profile of TNFRSF10B, TNFRSF10C, TP53TG5, MDM2, BAX, BCL-2 and CASPASE-8 genes and their participation in the resistance/susceptibility mechanism of these tumor cell lines. METHODS: To determine the IC50 values associated with Cisplatin and rhTRAIL, inhibition of cell growth was observed using MTT assays in two human OC cell lines (SKOV-3 and TOV-21G). The analysis of gene expression was performed using quantitative real-time polymerase chain reaction (qRT-PCR). RESULTS: Both cell lines had different susceptibility profiles to the tested drugs. In the SKOV-3 cell line, the IC50 values for cisplatin and for rhTRAIL were 270.83 ug/mL and 196.5 ng/mL, respectively. The same concentrations were used for TOV-21G. Different gene expression profiles were observed in each tested cell line. CASPASE-8 and TNFRSF10B expression levels could predict the response of both the cell lines to rhTRAIL alone or the response to a combination of rhTRAIL and cisplatin. In addition, we observed a relationship between BCL-2 and BAX expression that may be helpful in estimating the proliferation rate of the OC cell lines. CONCLUSION: SKOV-3 and TOV-21G respond differently to cisplatin and rhTRAIL exposure, and expression of CASPASE-8 and TNFRSF10B are good predictors of responses to these treatments.

Resumo O presente ensaio1 buscar relatar a experiência de traduzir dois cantos xamânicos Marubo para performance vocal a partir do trabalho de Pedro Cesarino, tanto na etnografia como na tradução poética. Para tanto, o texto se desdobra em um relato de percurso e em algumas ponderações teóricas sobre os problemas de traduzir uma poética xamânica ameríndia em português, com seus dilemas éticos e poéticos.

Abstract This essay seeks to report the experience of translating two Marubo shamanic chants for vocal performance starting from the work of Pedro Cesarino, both in ethnography and poetic translation. To this end, the text unfolds into an account of the course and into some theoretical considerations about the problems of translating an Amerindian shamanic poetics into Portuguese, with its ethical and poetic dilemmas.

BACKGROUND In addition to the limited therapeutic arsenal and the side effects of antileishmanial agents, drug resistance hinders disease control. In Brazil, Leishmania braziliensis causes atypical (AT) tegumentary leishmaniasis lesions, frequently refractory to treatment. OBJECTIVES The main goal of this study was to characterise antimony (Sb)-resistant (SbR) L. braziliensis strains obtained from patients living in Xakriabá indigenous community, Minas Gerais, Brazil. METHODS The aquaglyceroporin 1-encoding gene (AQP1) from L. braziliensis clinical isolates was sequenced, and its function was evaluated by hypo-osmotic shock. mRNA levels of genes associated with Sb resistance were measured by quantitative reverse transcription polymerase chain reaction (qRT-PCR). Atomic absorption was used to measure Sb uptake. FINDINGS Although clinical isolates presented delayed recovery time in hypo-osmotic shock, AQP1 function was maintained. Isolate 340 accumulated less Sb than all other isolates, supporting the 65-fold downregulation of AQP1 mRNA levels. Both 330 and 340 isolates upregulated antimony resistance marker (ARM) 56/ARM58 and multidrug resistant protein A (MRPA); however, only ARM58 upregulation was an exclusive feature of SbR field isolates. CA7AE seemed to increase drug uptake in L. braziliensis and represented a tool to study the role of glycoconjugates in Sb transport. MAIN CONCLUSIONS There is a clear correlation between ARM56/58 upregulation and Sb resistance in AT-harbouring patients, suggesting the use of these markers as potential indicators to help the treatment choice and outcome, preventing therapeutic failure.

RESUMO: Introdução: A Estratégia de Saúde da Família (ESF) deve ser o primeiro contato do Sistema Único de Saúde (SUS). Contudo dois modelos de atenção operam concomitantemente no âmbito da Atenção Primária à Saúde (APS): a ESF e o modelo tradicional. A expansão da ESF tem sido lenta e heterogênea em muitos municípios, tornando fundamental a condução de avaliações comparativas de atributos relacionados à qualidade dos modelos da APS. Objetivo: Comparar o desempenho dos modelos de atenção da APS de acordo com a percepção dos profissionais de saúde. Métodos: Estudo transversal com gestores e profissionais de saúde da APS do município de Divinópolis, Minas Gerais. Dados foram coletados por meio do Primary Care Assessment Tool. O desempenho dos modelos foi estimado por meio do Índice de Atenção Primária à Saúde (IAPS geral e específico). Regressão logística politômica univariada foi conduzida para comparação do desempenho dos modelos da APS de acordo com os atributos. A força da associação foi estimada por meio do odds ratio com intervalo de confiança de 95%. Resultados: Três gestores e 81 profissionais de saúde participaram do estudo. A ESF obteve melhor avaliação do que o modelo tradicional com relação ao IAPS geral e aos atributos vínculo, elenco de serviços, enfoque familiar e formação profissional. Conclusão: A ESF obteve escores superiores aos do modelo tradicional, entretanto ainda não atingiu o seu desempenho esperado. Esse cenário aponta para a necessidade de ampliação da cobertura da ESF e para a melhoria da qualidade das unidades de ESF existentes no Brasil.

ABSTRACT: Introduction: The Family Health Strategy (FHS) should be first-contact care in the Brazilian Health System. However, Primary Health Care (PHC) still encompasses two models: the FHS and the traditional health care facilities. The expansion of the FHS has been slow and heterogeneous in many cities, rendering a comparative evaluation of key quality-related elements of PHC models crucial. Objective: To compare the performance of PHC models as perceived by health professionals. Methods: A cross-sectional study involving managers and health professionals from PHC of a medium-size city in South-eastern Brazil. Data were collected by applying the Primary Care Assessment Tool. The performance was estimated through primary health care indexes (general and partial PHCI by attributes). Univariate polytomous logistic regression was performed to compare care model performances according to their attributes. Strength of association was estimated by odds ratio with 95% confidence interval. Results: Three managers and 81 health professionals participated in the study. The FHS had a better index rating than the traditional care model for general PHCI and for the attributes longitudinality, comprehensiveness, family focus and professional level. Conclusion: Although the FHS attained higher scores compared to the traditional model, it has not yet achieved the performance expected. This scenario points to the need for increased FHS cover and quality improvements at the existing units.

A avaliação científica do bem-estar tem sido importante para detectar animais não adaptados às condições de criação que lhes são impostas, situação que afeta o potencial produtivo e os coloca em maior risco de enfermidades. Com esse objetivo, foram avaliados 30 equinos da Cavalaria da Polícia Militar do Paraná, considerando-se indicadores clínicos, etológicos e hormonais. Na avaliação clínica, além da determinação do escore corporal e exame físico, verificou-se o histórico de ocorrência de cólica nos últimos doze meses. Comportamentos anormais foram pesquisados por observação etológica e a taxa do ritmo circadiano do cortisol (RCC) foi determinada por meio de duas colheitas de sangue, às 08 e às 17 horas. Apesar da significativa ocorrência de comportamentos anormais (26,7%), os achados clínicos foram normais e houve ausência de relato de cólica nos últimos 12 meses precedentes ao estudo. As médias dos resultados das análises de cortisol estiveram dentro da normalidade, nos dados absolutos e na variação do RCC, entretanto, 42% dos animais analisados apresentaram RCC alterado. Apesar da expressiva incidência de comportamentos anormais e de animais com alteração no ritmo circadiano de cortisol, sinais indicadores de estresse crônico, considerou-se que os equinos deste estudo apresentaram boas condições de bem-estar em relação àquelas relatadas por estudos nacionais prévios.

The scientific assessment of welfare has been important to detect animals non adapted to their management conditions, which can interfere with their productive potential and put them at increased risk of illness. With this objective, 30 horses from the Military Police of Paraná were evaluated, considering clinical, ethological and hormonal indicators of equine welfare. In the clinical evaluation, horses were assessed by physical examination and their body score conditions and the history of colic incidence in the last twelve months were determined. Abnormal behavior incidence were studied by ethological observation and the individual rate of circadian rhythm of cortisol (RCC) was calculated based on cortisol concentration changes in two blood samples taken at 08:00 am and 5:00pm. Despite significant incidence of abnormal behaviors (26.7%), clinical findings were normal and there were no reports of colic episodes in the last 12 months preceding the study. The mean blood cortisol concentrations and the mean RCC rate were within the normal range. However, 42% of the studied horses had abnormal RCC rates. Despite the significant incidences of abnormal behavior and abnormal RCC rates (indicators of chronic stress), these horses were considered in good welfare when compared to ones previously reported in other national studies.
