Abstract Soil salinization is an environmental factor that frequently occurs in semi-arid regions around the world and seed priming technique is one of the alternatives to obtain the greatest establishment of seedlings in the field, through the induction of tolerance to environmental stresses. This research aimed to evaluate the effect of seed priming with NaCl on the induction of salinity tolerance in Myracrodruon urundeuva Allemão in vitro. The research was carried out at the Center for Strategic Technologies of the Northeast, in Recife / PE-Brazil M. urundeuva seeds were disinfected and submitted to two treatments: water (control) or NaCl (2mM), and inoculated in WPM medium at three saline concentrations (0.0; 25.0 and 50.0 mM NaCl) for 45 days. Plant height, number of leaves, fresh biomass (total, shoot, and root), shoot/root ratio, sodium and potassium and Na/K ratio, antioxidant enzyme activity, and peroxide content were evaluated malondialdehyde hydrogen. The data were subjected to variance analysis and the results were compared using the Student-Newman-Keuls test at 5% probability, using the R software. Seed priming with NaCl promoted a beneficial effect on the height of seedlings exposed to salinity. On the other hand, height was inversely proportional to saline concentrations, regardless of seed priming. The shoot/root ratio was also lower in seedlings cultivated under 25 and 50 mM NaCl. Despite being favorable for most growth variables, seed priming with NaCl induced an increase in lipid peroxidation in seedlings that were not exposed to salinity, which is linked to a decrease in the activity of antioxidant enzymes. The antagonistic response to NaCl stimulation between growth parameters and plant defense observed in the present research raises the need for further complementary studies that make it possible to delineate the metabolic alterations of M. urundeuva against a chemical stimulus to induce tolerance to a given stress.

Resumo A salinização do solo é um fator ambiental que ocorre frequentemente em regiões semi-áridas ao redor do mundo e a técnica de seed priming é uma das alternativas para se obter o maior estabelecimento de mudas no campo, através da indução de tolerância aos estresses ambientais. Este trabalho objetivou avaliar o efeito do seed priming com NaCl na indução da tolerância à salinidade em Myracrodruon urundeuva Allemão in vitro. A pesquisa foi realizada no Centro de Tecnologias Estratégicas do Nordeste, em Recife / PE-Brasil. Sementes de M. urundeuva foram desinfestadas e submetidas a dois tratamentos: água (controle) ou NaCl (2mM), e inoculadas em meio WPM em três concentrações salinas (0.0; 25.0 and 50.0 mM NaCl) por 45 dias. Foi avaliada a altura, número de folhas, biomassa fresca (total, parte aérea e raiz), razão parte aérea / raiz, sódio, potássio, razão Na / K, atividade de enzimas antioxidantes, teores de peróxido de hidrogênio e malondialdeído. Os dados foram submetidos à análise de variância e os resultados comparados pelo teste Student-Newman-Keuls a 5% de probabilidade, utilizando o software R. O priming com NaCl promoveu efeito benéfico na altura de mudas expostas à salinidade. Todavia, a altura foi inversamente proporcional às concentrações salinas, independentemente do priming. A razão parte aérea / raiz foi menor nas mudas cultivadas sob NaCl 25 e 50 mM. Apesar de ser favorável para a maioria das variáveis de crescimento, o seed priming com NaCl induziu aumento na peroxidação lipídica em mudas não expostas à salinidade, o que está relacionado à diminuição da atividade de enzimas antioxidantes. A resposta antagônica à estimulação do NaCl entre parâmetros de crescimento e defesa das plantas na presente pesquisa levanta a necessidade de novos estudos complementares que possibilitem delinear as alterações metabólicas de M. urundeuva frente a um estímulo químico para induzir tolerância a um determinado estresse.

ABSTRACT Polyhydroxyalkanoates (PHA) are a viable alternative to replacing traditional plastics, harmful to nature. The objective of this study was to evaluate the production of PHA by Cupriviadus necator using agro-industrial residues, its physicochemical characterization, and of the membranes produced. Cell growth and PHA production were compared in three carbon sources: residual glycerol, whey cheese with and without a mineral solution. After cultivation, the biopolymer was extracted and characterized by FTIR and 1D and 2D NMR experiments. Membranes containing 5.0% of PHA had their physicochemical properties measured for mechanical properties, color, and water vapor permeability. The results demonstrated that the yield of PHA.g-1 of biomass was higher for cultivation using residual. Yet the PHA.L-1 was higher in cultures with whey cheese. The NMR and FTIR result allowed to characterize the molecular structure of PHA produced. It was calculated 8 carbon atoms in the methylene main chain and a maximum of 13 carbons (Poly-3-hydroxytridecanoato) in the polymer side chain. The membranes showed a strong whiteness tendency as well as a high opacity, expected low water vapor permeability, and rigidity. In conclusion, mechanical properties indicated that the formed PHA membranes could be applicable to plastic packaging in substitution for petroleum derivatives.

RESUMO: Um dos maiores entraves para o crescimento da produção de cana-de-açúcar é a infecção por fitopatógenos, principalmente pela bactéria Leifsonia xyli subsp. xyli (Lxx), agente causal do raquitismo-da-soqueira, doença que mais causa perdas a cultura. Assim, este trabalho teve como objetivo, avaliar os efeitos do antimicrobiano casugamicina sobre o crescimento in vitro, bem como seu efeito sobre a infecção endógena por Lxx em cana-de-açúcar. Explantes da variedade SP791011 de cana-de-açúcar foram inoculados em meio de cultura MS suplementados com o antimicrobiano nas concentrações de 0,00; 0,87; 1,08; 1,74 e 3,48 mL.L-1, em que permaneceram por 30 dias. Após este período, foi avaliada a taxa de sobrevivência, número de brotações por explante, altura dos explantes, fitomassa, fitomassa seca e fitossanidade em relação à presença de DNA genômico de Lxx. Foi verificado que o cultivo em casugamicina influenciou de forma negativa nas variáveis morfológicas, apesar disso, o antimicrobiano não mostrou fitotoxidade às plantas. Todos os tratamentos testados neste experimento foram diagnosticados como positivos, com amplificação de DNA para Lxx, apesar de ter sido observada uma redução na carga bacteriana, sugerindo que a casugamicina em doses superiores pode ser avaliada como alternativa na tentativa de eliminar completamente a bactéria no cultivo in vitro de cana-de-açúcar.

ABSTRACT: One of the significant obstacles to the growth of sugarcane production is the infection by phytopathogens, mainly by the bacterium Leifsonia xyli subsp. xyli (Lxx) causal agent of Ratoon stunting disease. Thus, this research aimed to evaluate the effects of kasugamycin on the in vitro growth of sugarcane, as well as its effect on the bacterium Lxx. Explants of strain SP791011 from sugarcane were inoculated in MS culture medium supplemented with the antimicrobial kasugamycin at concentrations of 0.00; 0.87; 1.08; 1.74 and 3.48 mL.L-1, where they remained for 30 days. After this period, the survival rate, shoot number per explant, height of the explants, phytomass, dry phytomass and phytosanitary were evaluated based on the presence of genomic DNA of Lxx. It was verified that the culture in kasugamycin influenced the morphological variables negatively; nevertheless, the antimicrobial did not demonstrate phytotoxicity to the plants. All treatments tested in this experiment were diagnosed as positive, with DNA amplification for Lxx, despite it was observed a reduction in bacterial load, suggesting that kasugamycin at higher doses can be evaluated as an attempt to eliminate the bacterium in the in vitro cultivation of sugarcane.

RESUMO: O objetivo do presente trabalho foi relatar, pela primeira vez, população de Pratylenchus brachyurus associada a raízes de mangueira cv. Tommy Atkins no estádio de muda em solo naturalmente infestado no município de Assú, Rio Grande do Norte. A caracterização da referida população do nematóide foi fundamentada em estudo morfométrico realizado em 20 fêmeas coletadas das raízes. Os dados morfométricos foram comparados aos dados publicados por diferentes autores em trabalhos sobre a ocorrência da referida espécie de nematóide das lesões e permitiram a identificação da população como pertencente à P. brachyurus.

ABSTRACT: The aim of this study was to report for the first time Pratylenchus brachyurus population associated to mango cv. Tommy Atkins seedling roots, on naturally infested soil, on Assú municipality, Rio Grande do Norte State. The population of nematode was characterized based on a morphometric study that was done using 20 females collected from the roots. The morphometric data were compared to other data published by different authors in studies concerning to reporters of this species of root-lesion nematode, and it was possible to identify the population as P. brachyurus.

RESUMOO estudo foi realizado com explantes de ápices caulinares de nim (Azadirachta indica A. Juss) para identificar um processo viável de regeneração. Brotos foram obtidos a partir de embriões de nim cultivados alternadamente em meio DKW suplementado com BAP e meio sem hormônios. O desenvolvimento inicial dos brotos foi influenciada pela presença dos cotilédones. Calos basais, isolados in vitro a partir da base dos caules, também apresentam potencial de organogênese. Em alguns casos, as linhagens de plantas, obtidas a partir de cada semente, apresentaram características diferentes. A característica mais comum observada in vitro foi a formação de calos na base do caule. Entretanto, as características mais raras foram a formação de calo ao longo de todo o caule e a ocorrência de senescência foliar. Os apices caulinares regenerados foram posteriormente micropropagadados e enraizados em um meio suplementado com IBA, permitindo a obtenção de plantas completas. As plantas enraizadas foram transferidas para casa de vegatação e aclimatizadas com sucesso. Nenhuma diferença no desenvolvimento in vivo foi observada entre plantas obtidas de calos e da propagação de ápices caulinares de nim.

ABSTRACTThe study was conducted with shoot tip explants of neem (Azadirachta indica A. Juss) to identify a viable regenerative process. Shoot tips were obtained from neem embryos cultured alternatingly in DKW medium supplemented with BAP and medium without hormones. Initial shoot development was influenced by cotyledon presence. Basal callus, excised from in vitro stem base, also presented organogenic potential. In some cases, plant lines, obtained from each seed, presented different characteristics. The most common characteristic observed in vitro was callus formation at the stem base. However, the rarest characteristics were stem callus formation and leaf senescence. The regenerated shoot tips were further subculture and rooted on a medium supplemented with IBA so that complete plants could be obtained. The rooted plants were transplanted to a greenhouse and successfully acclimatized. No significant differences in in vivo development were observed between neem plants from callus and from shoot tip propagation.

Neste trabalho é descrito um método de regeneração de plantas, a partir de células de bananeira, em suspensão, e o efeito da biobalística no potencial regenerativo. Embriões somáticos foram obtidos de inflorescências masculinas de bananeira cv. Maçã (grupo AAB) por meio de embriogênese indireta. Parte dos calos (40%) apresentou características embriogênicas (não-friáveis, compactas e amareladas). Suspensões celulares obtidas desses calos continham pequenas massas celulares, com citoplasmas ricos em grânulos de amido, núcleos grandes e nucléolos densos. Após quatro meses, embriões somáticos começaram a se desenvolver. O número máximo de plantas regeneradas ocorreu 45-60 dias após a formação dos embriões. No primeiro experimento foram regeneradas 401 plantas. No segundo, 399 plantas foram obtidas de uma suspensão celular 6 meses mais velha do que a do primeiro experimento. Transformações celulares com uma das três construções plasmidiais utilizadas, que continham o gene uid-A, resultaram em fortes sinais de expressão cinco dias após as transformações; todavia, o número de plantas regeneradas foi muito menor do que o observado no material não bombardeado.

A plant regeneration method with cell suspension cultures of banana, and the effect of biobalistic on regeneration potential are described in this report. Somatic embryos of banana were obtained from indirect embryogenesis of male inflorescence of banana cultivar Maçã (AAB group). Part of the calluses formed (40%) showed embryogenic characteristics (nonfriable, compact and yellow color). The cell suspension, originated from embryogenic calluses, contained clusters of small tightly packed cells with dense cytoplasms, relatively large nuclei and very dense nucleoli. After four months of culture, somatic embryos started to regenerate. The maximum number of regenerated plants was observed between 45 and 60 days after embryo formation.In the first experiment, 401 plants were regenerated from approximately 10 mL of packed cells. In the second experiment, 399 plants were regenerated from a cell suspension six months older than that of the first experiment. Cell transformation using particle bombardment with three different plasmid constructions, containing the uid-A gene, resulted in a strong GUS expression five days after bombardment; however, plant regeneration from bombarded cells was much lower than nonbombarded ones.
