Phagocytosis by neutrophils and monocytes constitutes the main defense mechanism against bacterial challenges in periodontitis. Phagocytosis by neutrophils has already been evaluated, whereas phagocytic function of monocytes has hardly been addressed so far. Objectives: The aim of this study was to assess phagocytosis by neutrophils and monocytes in periodontitis. Material and Methods: The sample included 30 subjects with severe periodontitis and 27 control subjects without periodontal disease. The phagocytic index (PhI) was calculated as the mean number of adhered/ingested Saccharomyces cerevisiae per phagocytozing monocyte or neutrophil multiplied by the percentage of phagocytes involved in phagocytosis. Results: A significant reduction in phagocyte functions was observed in individuals with periodontitis. The median of PhI of neutrophils using nonsensitized S. cerevisiae was 3 for the control group, and 1.5 for the periodontitis group (p=0.01, Mann-Whitney test). The median of PhI of monocytes with non-sensitized S. cerevisiae was 26.13 for the control group, and 13.23 for the periodontitis group (p=0.03, Mann Whitney test). The median of PhI of monocytes assessed with sensitized S. cerevisiae was 97.92 for the control group and 60.1 for the periodontitis group (p=0.005, t-test). Conclusion: The data demonstrated a reduction in the function of phagocytes, suggesting a decrease in immune defenses in periodontitis.

Este trabalho teve como objetivo avaliar a atividade antimicrobiana da cúrcuma em pó, da curcumina disponível no comércio, dos pigmentos curcuminóides purificados e dos óleos essenciais da cúrcuma. As amostras foram analisadas quanto aos teores de pigmentos curcuminóides por cromatografia líquida de alta eficiência. O óleo essencial foi também analisado quanto à densidade, índice de refração e perfil das substâncias voláteis por cromatografia gasosa e detector de ionização de chamas. A atividade antimicrobiana in vitro foi determinada pelo método de difusão em ágar com discos de papel estéreis, impregnados com os extratos, sendo os diâmetros dos halos de inibição medidos com paquímetro. Os extratos etanólicos da cúrcuma, da curcumina comercial, e dos pigmentos curcumina e desmetoxicurcumina não inibiram o crescimento de Staphylococcus aureus, Bacillus subtilis, Salmonella choleraesuis, Escherichia coli, Aspergillus niger, Saccharomyces cerevisiae, e Candida albicans. Apenas o extrato etanólico da bisdesmetoxicurcumina apresentou atividade antimicrobiana em relação ao B. subtilis. O óleo essencial da cúrcuma apresentou atividade antimicrobiana para o B. subtilis, S. choleraesuis, E. coli, A. niger e S. cerevisiae, sendo que essa atividade aumentou em função do aumento da concentração. Os halos de inibição, obtidos com o óleo essencial, foram significativos, quando comparados aos respectivos antibióticos tradicionais, cloranfenicol e anfotericina, indicando ser o óleo essencial da cúrcuma um agente antimicrobiano em potencial.

The objective of this work was to investigate the antimicrobial activity of ground turmeric, commercially available curcumin, purified curcuminoid pigments and turmeric essential oils. The samples were analyzed for the levels of curcuminoid pigments by high performance liquid chromatography. The essential oil was also analyzed for density, refractive index and profile of volatile compounds by gas chromatography-flame ionization detection. The in vitro antimicrobial activity was determined by the agar diffusion method using sterile disks impregnated with the extracts and the diameters of the inhibition zones were measured using a caliper. The ethanolic extracts of turmeric, commercial curcumin, and the purified pigments curcumin and demethoxycurcumin did not inhibit the growth of Staphylococcus aureus, Bacillus subtilis, Salmonella choleraesuis, Escherichia coli, Aspergillus niger, Saccharomyces cerevisiae, and Candida albicans. Only the ethanolic extract of bisdemethoxycurcumin showed in vitro antimicrobial activity on B. subtilis. The essential oil of turmeric inhibited the growth of B. subtilis, S. choleraesuis, E. coli, A. niger and S. cerevisiae. The antimicrobial activity increased with an increase in the concentration of the essential oil. The inhibitory effects on the microorganisms were significant when compared to the respective traditional antibiotics chloranphenicol and anphotericin. This result indicates that the essential oil from turmeric is a potential antimicrobial agent.

Foram realizados dois experimentos com o objetivo de avaliar o efeito das concentrações de BAP e de AG3 no desenvolvimento in vitro de mandioquinha-salsa. Foram avaliadas as concentrações de 0,0; 0,2 e 0,4 mg L-1 de BAP e de 0,0; 0,125 e 0,250 mg L-1 de AG3 no desenvolvimento de ápices caulinares com cerca de 2 mm das cultivares Amarela de Senador Amaral e Amarela Comum. A interação cultivares x concentração de BAP não foi significativa para nenhuma das características avaliadas (P>0,0617). O aumento na concentração de BAP reduziu a formação de calos; entretanto, promoveu proporcionalmente redução no desenvolvimento da parte aérea. A concentração de BAP que apresentou os melhores resultados foi em torno de 0,3 mg L-1 de BAP, conciliando diâmetro de calo relativamente reduzido e bom desenvolvimento da parte aérea, cerca de 4,5 brotos com altura média de 43 mm. A interação cultivares x concentração de AG3 não foi significativa para nenhuma das características avaliadas (P>0,4310). O aumento na concentração de AG3 promoveu elevação significativa na altura média e máxima das brotações e aumento no tamanho de calos. Nos dois experimentos, observou-se maior número de brotações na cultivar Amarela Comum.

Two experiments were carried out to evaluate the effect of BAP and GA3 concentrations on the in vitro arracacha development. The concentrations tested were 0.0; 0.2 and 0.4 mg L-1 of BAP and 0.0; 0.125 and 0.250 mg L-1 of GA3 in the shoot tips with about 2 mm of arracacha, cultivars Amarela de Senador Amaral and Amarela Comum. The interaction of cultivars x BAP concentration was not significant for any one of the evaluated variables (P>0,0617). The increase in the BAP concentration reduced the callus formation; however, promoted larger reduction in the shoot development. The best BAP concentration was around 0.3 mg L-1, with relatively reduced callus diameter and good development of shoot, with a mean of 4.5 shoots and a height of 43 mm. The interaction of cultivars x GA3 concentration was not significant for any one of the studied variables (P>0,4310). The increase in the GA3 concentration promoted the improvement in the mean and maximum height of the shoots and in callus size. More shoots were observed for Amarela Comum cultivar in both trials.
