RESUMO Os fungicidas químicos fornecem meios primários para o controle da infecção fúngica de frutas e legumes no período pós-colheita. A exploração de produtos naturais para controlar a deterioração e prolongar o tempo de armazenamento de alimentos perecíveis tem recebido mais atenção. Neste trabalho, extratos hidroetanólicos de folhas de saboeiro (Sapindus saponaria L.) foram analisados quanto a sua atividade inibitória contra o fungo Colletotrichum gloeosporioides, agente causador da antracnose em frutos de mamoeiro. Para avaliar a atividade antifúngica do extrato vegetal, este foi incorporado em meio de batata-dextrose-agar (BDA) à diferentes concentrações (10, 50 e 100 mg mL-1), e a inibição do crescimento micelial, inibição de produção de esporos e mudanças morfológicas foram avaliadas à temperatura ambiente após sete dias. A eficácia do extrato foi observada quando foram utilizados 50 mg mL-1 e 100 mg mL-1 do extrato, resultando em redução de crescimento micelial (. <0.05). À 50 mg mL-1, o extrato causa alterações morfológicas com a redução do tamanho das hifas que se tornam mais curtas e mais ramificadas. À 100 mg mL-1 de extrato, os danos na estrutura da hifa foram mais severos, causando ruptura micelial. O tratamento in vivo com 100 mg mL-1 de extrato impediu o desenvolvimento de sintomas de antracnose em frutos de mamão frescos. O extrato de folha de saboeiro tem uma forte atividade inibitória contra C. gloeosporioides, impedindo o crescimento micelial e reduzindo o aparecimento de sintomas de antracnose. Em conjunto, estes resultados demonstraram o seu potencial do extrato como fungicida natural constituindo uma medida alternativa para o controle da doença em frutos de mamoeiro.

ABSTRACT Chemical fungicides provide the primary means to control fungal infection in fruits and vegetables in the postharvest. Exploitation of natural products to control decay and prolong the storage life of perishables has received more attention. In this study, hydroethanolic extracts from the leaves of soapberry (Sapindus saponaria L.) were investigated for their inhibitory activity against the fungus Colletotrichum gloeosporioides, the causal agent of anthracnose in papaya fruits. To evaluate the antifungal activity of the plant extract, the latter was incorporated into potato-dextrose-agar (PDA) medium at different concentrations (10, 50 and 100 mg mL-1), and mycelial growth inhibition, spore production inhibition and morphological changes were evaluated at room temperature after 7 days. Efficacy of the extract was noted when it was used at 50 mg mL-1 and 100 mg mL-1, resulting in a reduction in the mycelial growth (P< 0.05). At 50 mg mL-1, the extract causes morphological changes by reducing the size of hyphae, which become shorter and more branched. At 100 mg mL-1 extract, damages in the hyphal structure were more severe, causing mycelial rupture. In vivo treatment with 100 mg mL-1 extract prevented the development of anthracnose symptoms in fresh papaya fruits. Soapberry leaf extract has strong inhibitory activity against C. gloeosporioides, preventing the mycelial growth and reducing the appearance of anthracnose symptoms. Taken together, these results demonstrated the potential of this extract as a natural fungicide, constituting an alternative measure for disease control in papaya fruits.

The success of sexual plant reproduction is directly influenced by specific interactions between the pollen and pistil. Light, fluorescence and scanning electron microscopy techniques were used to evaluate the steps of pollination in sour passion fruit plants (Passiflora edulis Sims). In the compatible interaction, pollen tubes grow through stigma projections towards the ovary. The pollen grain surface was found to be spheroidal and to consist of heteroreticulate exine with six colpi. Furthermore, analysis in vivo of pollen-pistil interactions indicated that stigmas of flowers 24 hours before anthesis are unable to discriminate compatible (genetically unrelated) and incompatible (genetically related) pollen grains. Taken together, these results provide insight into the cellular mechanisms underlying pollination in passion fruit plants.

As características anatômicas de folhas de sol e sombra de Alseis pickelii foram investigadas a fim de se avaliar as consequências do corte seletivo de madeira sobre a estrutura foliar em uma floresta atlântica estacional semidecidual. Folhas expandidas de sol e de sombra foram coletadas em dois distintos setores da mata, setor explorado (área com recente atividade de corte seletivo), e setor preservado. Somente folhas do setor preservado revelaram significantes diferenças de resposta aos regimes de luz, produzindo folhas com características estruturais nitidamente associadas com diferentes níveis de irradiação. Folhas de sol deste setor apresentaram maiores espessuras da superfície adaxial, mesofilo, parênquima paliçádico e lacunoso, parede secundária das fibras e menor área foliar comparada às folhas de sombra. Entretanto, no setor explorado, as cutículas das folhas de sol e sombra não apresentaram diferenças significativas e as áreas foliares das folhas de sol foram maiores que a das folhas de sombra. De acordo com estes resultados, conclui-se que o setor preservado produziu árvores com típicas folhas de "sol" e "sombra". Em contraste, no setor explorado as árvores apresentaram menor variação nos tipos de folhas, onde nem típicas folhas de "sol" ou de "sombra" foram produzidas, sugerindo menor plasticidade desta espécie secundária tardia nesta área.

The anatomical characteristics of both sun and shade leaves of Alseis pickelii were investigated in order to evaluate the consequences of selective logging (in seasonally dry Atlantic Forest) on the leaf structure of this species. Fully expanded sun and shade leaves were collected in two distinct stands of tabuleiro forest; a stand of recently logged forest and an unlogged stand. Only leaves from the unlogged stand revealed significantly different magnitudes of response to light regimes, producing leaves with structural characteristics associated with different levels of irradiance. The sun leaves from this stand had a thicker adaxial surface, mesophyll, palisade and spongy parenchyma, a secondary cell wall of fibers and a lower leaf area compared with the shade leaves. However, in the logged stand, the leaf cuticles of sun and shade leaves showed no significant differences, although the leaf area of the sun leaves was higher than the shade leaves. According to these data, we concluded that the unlogged stand produced typical "sun" and "shade" leaves. In contrast, leaves from the logged stand showed a lower variation of types, where neither typical "sun" nor typical "shade" leaves were produced, suggesting lower leaf plasticity of this late successional tree in this area.

(Ultraestrutura da fase secretora e da senescente dos coléteres de Bathysa gymnocarpa e B. stipulata (Rubiaceae)). Coléteres são estruturas secretoras formadas por um eixo parenquimático que inclui feixes vasculares, circundado por uma camada de células epidérmicas secretoras em paliçada. Em estudos sobre a estrutura dos coléteres tem sido observada a ultraestrutura das células secretoras, mas não discriminam as fases secretora e senescente. Geralmente, em células secretores de mucilagem como os coléteres, o retículo endoplasmático e o complexo de Golgi estão envolvidos na produção e no transporte da secreção. Neste estudo, foram determinadas duas fases baseadas na estrutura dos coléteres de Bathysa gymnocarpa K. Schum. and B. stipulata (Vell.) C. Presl. (Rubiaceae): a fase secretora e a fase senescente. Amostras foram coletadas e processadas utilizando técnicas usuais de microscopia óptica e eletrônica. Os coléteres estudados são constituídos por uma camada epidérmica em paliçada e um eixo central parenquimático com traços vasculares raros. Durante a fase secretora, as células epidérmicas se apresentam com o citoplasma denso, pequenos vacúolos, retículo endoplasmático liso e rugoso evidente e complexo de Golgi próximo a grandes vesículas. Durante a fase senescente, as células epidérmicas apresentaram o sistema endomembranar desorganizado. Nenhuma organela intacta ou vesícula foi observada. A parede celular mais externa exibiu camadas similares às observadas durante a fase secretora. A fase senescente é facilmente definida pela morfologia do coléter, mas não é bem definida em nível subcelular nas células secretoras. Nossa investigação sugere que a morte celular programada se inicia na fase secretora. Contudo, mais evidências são necessárias para avaliar esse fenômeno.

(Ultrastructure of secretory and senescence phase in colleters of Bathysa gymnocarpa and B. stipulata (Rubiaceae)). Colleters are secretory structures formed by a parenchymatic axis with vascular bundles, bound by a layer of secretory palisade-like epidermis. Some studies regarding the structure of colleters have focused on secretory cells structure, but not distinguished the secretory and senescent phases. Generally, in mucilage-secreting cells such as colleters, the endoplasmic reticulum and Golgi apparatus are involved in secretion production and transport. In these study, colleters structure of Bathysa gymnocarpa K. Schum. and B. stipulata (Vell.) C. Presl. (Rubiaceae) were determined in two phases: a secretory phase and a senescence one. Samples were collected and processed by usual light and electron microscopy techniques. Studied colleters are constituted by an epidermal palisade layer and a central axis formed by parenchymatic cells with rare vascular traces. During the secretory phase, epidermal cells presented a dense cytoplasm, small vacuoles, enhanced rough and smooth endoplasmic reticulum, and a Golgi apparatus close to large vesicles. During the senescence phase epidermal cells presented a disorganized membrane system. No intact organelles or vesicles were observed. The outer cell wall exhibited similar layers to that observed during the secretory phase. The senescent phase is easily defined by the morphology of the colleters, but not well defined at subcellular level. Our research suggests that programmed cell death starts on secretory phase. However, more evidences are needed to evaluate the phenomena.

Uma beta-1,3-glucanase foi purificada a partir da secreção de Simira glaziovii, através de vários processos cromatográficos, tendo a análise do perfil protéico acompanhado de SDS-PAGE. A enzima purificada apresentou uma massa molecular estimada de 35 kDa. O pH ótimo obtido para a enzima foi de 5,2.

In this study, beta-1,3-glucanase was isolated from Simira glaziovii secretion. The purification process was achieved by a combination of chromatographic methods and was analyzed by SDS-PAGE. The purified enzyme presented an estimated molecular mass of 35 kDa. The optimum pH of enzyme was 5.2
