Objetivo: avaliar o efeito cicatrizante do extrato aquoso do babaçu e do óleo de andiroba em feridas abertas no ceco de ratos. Métodos: cinquenta e quatro ratos Wistar foram divididos em três grupos de 18: 1) grupo babaçu, com aplicação do extrato aquoso de babaçu; 2) grupo andiroba, com aplicação do óleo; e 3) grupo controle, com aplicação de solução salina. Todos os procedimentos foram feitos por gavagem. Cada grupo foi dividido em três subgrupos de seis animais conforme o período de observação, aos 7, 14 ou 21 dias. De cada animal foi retirado fragmento do ceco com 1,5cm2 de diâmetro. As áreas das lesões foram analisadas por macroscopia e os segmentos ressecados das feridas por microscopia ótica em colorações de hematoxilina-eosina e tricrômico de Masson. Resultados: foram verificados abscesso e infecção em dois animais do grupo andiroba, e um com hematoma. Quanto ao grau de aderências, o grupo babaçu teve maior incidência de aderências grau II enquanto que no grupo controle e andiroba predominaram aderências grau I. Na análise microscópica no sétimo dia a proliferação fibroblástica foi maior no grupo andiroba e menor no grupo babaçu (p=0,028). No 14º dia os polimorfonucleares foram menos acentuados no grupo babaçu (p=0,007). Quanto ao teste de resistência à insuflação de ar atmosférico observou-se que o grupo andiroba em qualquer dos dias avaliados apresentou maior tensão. Quanto à colagenização, no sétimo dia, ela esteve presente em 100% dos animais do grupo andiroba. No 14º dia foi mais acentuada no grupo controle e no 21º dia resultados semelhantes para o grupo controle e andiroba. Conclusão: os animais dos grupos babaçu e andiroba apresentaram melhor cicatrização do ceco em comparação ao grupo controle.

Objective: to evaluate the healing effect of the babassu aqueous extract and andiroba oil on open wounds in the cecum of rats. Methods: fifty-four Wistar rats were divided into three groups of 18: 1) babassu group with application of aqueous extract of babassu; 2) andiroba group with application of the oil; and 3) control group, with application of saline solution. All procedures were done by gavage. Each group was divided into three subgroups of six animals according to the observation period of 7, 14 or 21 days. From each animal was removed caecum fragment of 1.5cm² diameter. The areas of the lesions were analyzed macroscopically and resected specimens by light microscopy using hematoxylin-eosin and Masson's trichrome. Results: abscess and infection were observed in two aroeira group animals, and in one only hematoma. In relationship to adhesions degree, babassu group had higher incidence of grade II while in the control and aroeira groups predominated adhesions grade I. On microscopic examination on day 7 fibroblast proliferation was greater in aroeira and lower in babassu group (p=0.028). On the 14th day polymorphonuclear were less pronounced in babassu (p=0.007). As for the resistance test of air insufflation, it was observed that in all andiroba group in all tested days showed be higher. As for collagen, on the 7th day it was present in 100% of animals of aroeira group. On the 14th day was more pronounced in the control group and at day 21 similar results were found in the control and aroeira groups. Conclusion: animals in babassu and andiroba groups showed better cecum healing compared to the control group.

PURPOSE:To compare controlled liver regeneration in rats submitted to 60% hepatic resection having L-arginine supplemented diet, based on weight changes of the regenerated liver, laboratory parameters of liver function and pathological findings.METHODS:Thirty-six rats were divided into two groups, control and L- arginine. The first received standard chow and saline solution by gavage. The second had supplementation with L- arginine. Animals were killed on postoperative period at 24h, 72h and seven days. For analysis of liver regeneration was used Kwon formula for weight, laboratory tests and mitosis.RESULTS: Weight, showed no benefit with L- arginine supplementation; however, intergroup comparison in the first 24h observed positive effect on supplementation (p=0.008). Alkaline phosphatase was increased in arginine group (p<0.04). The number of mitoses showed no difference between the two groups; however, in the first 24 hours, the supplemented group had higher number of mitoses within the groups (p=0.03).CONCLUSION:Supplementation with L-arginine did not show benefits in liver regeneration; however, supplemented group in the first 24 hours showed benefits over 72 hours and seven days of the evaluation by weight gain and number of mitosis.

OBJETIVO: Analisar a resistência (rigidez) do sistema de fixação externa tubular uniplanar, com hastes de conexão única e dupla, com traços de fraturas estáveis e instáveis. MÉTODOS: Foram utilizados 48 modelos semelhantes à tíbia. Em todos foi deixado um intervalo de 0,5 cm entre os fragmentos e realizados cortes com angulações de 15º e 45º para simular fraturas estáveis e instáveis, respectivamente. Os modelos foram divididos em quatro grupos de acordo com o traço fraturário (15º e 45º) e o número de barras metálicas na montagem (1 e 2 barras). Os modelos de prova foram adaptados à uma máquina de testes Instron®, pelas suas extremidades, e submetidos à compressão axial até que os fragmentos tiveram contato total. Avaliou-se a força necessária para efetuar o completo contato dos fragmentos do modelo. RESULTADOS: As forças instabilizadoras na montagem do fixador com barra dupla foram bastante superiores às com barra única. Observou-se ainda que o grupo com barra única instável apresentou variabilidade muito menor que os demais grupos, ou seja, apresenta resultados mais homogêneos, além de ter apresentado a menor média. CONCLUSÃO: A montagem do fixador externo com uma haste longitudinal dupla nos modelos estudados é mais estável que as demais quando submetidas à uma força de compressão axial.

OBJECTIVE: This is an experimental study with the objective of analyses the increase of rigidity in synthetic tibia bones with external fixators, with single and double connecting rods and with stable and unstable fracture patterns. METHODS: The external fixators were used in the monoplanar, half pin configuration submitted to an axial compression load using the connecting bar in different patterns. Forty-eight similar models to the human tibia had been used. In all the models were left an interval of 0,5 cm between the fragments and were made cuts of 15º and 45º to simulate stable and unstable fracture patterns, respectively. The models had been divided in four groups in accordance with the cut (15º and 45º) and the number of metallic bars in assembly (1 and 2 bars). The study used the Instron® testing machine which the models were submitted to the axial load until the fragments that were separated by a gap made full contact. RESULTS: The instability forces in the assembly with a double bar had been sufficiently superior to the forces with only one bar. It was still observed that the group with unstable fracture pattern and with only one connecting rod had a lesser variability compared with the other groups and presented more homogeneous results, with a minor average. CONCLUSION: The assembly with a double longitudinal connecting rod in the studied models is more stable than the others to stand compression load.

OBJETIVO: Avaliar a diferença do comportamento mecânico de tendões flexores solidarizados e não solidarizados para verificar se a solidarização tem função efetiva. MÉTODOS: Vinte e quatro tendões digitais bovinos frescos foram usados. Para determinar a área da secção transversal utilizou-se alginato. Dez tendões foram solidarizados seguindo as orientações do fabricante; outros 10 pares não. Foram desenvolvidas garras para fixação dos enxertos à máquina universal para simulação da fixação, sendo a superior bipartida e de passagem controlada dos pinos e a inferior com dentes alternados. RESULTADOS: A carga máxima dos corpos de prova não solidarizados foi de 849,4 N ± 386,8 a área 30,4 mm²± 7,7, tensão de 29 ± 17 Mpa. Os solidarizados obtiveram carga máxima de 871,8 N ± 484,9 área 35 mm²± 5,8, tensão de 24 ± 10 Mpa. Não houve diferença estatística entre os dois grupos (p>0,05). CONCLUSÃO: A distribuição de probabilidade mostra que para 400 N os tendões não solidarizados apresentam confiabilidade de 83,8% e os solidarizados de 78,5%%.

OBJECTIVE:To evaluate the difference of the mechanical behavior of bended tied (sewed) and not tied hamstring tendons in order to evaluate if it has any effective function. METHODS: Twenty-four fresh bovine digital tendons had been used. Alginate to determine the area of the tendon transversal section was used. Ten bovine tendons had been bended and tied following the manufacturer orientation, ten others pairs had not been tied. Claws had been developed for the grafts fixation to the universal machine, for the setting simulation. The superior one was characterized to be bipartite and to have controlled passage for the pins and the inferior claw is characterized to have alternating teeth. RESULTS: The maximum load of the not tied samples of the test was 849,4 N ± 386,8 the area was 30,4 mm² ± 7,7, and 29 ± 17 Mpa of tension. The tied ones had gotten maximum load of 871,8 N ± 484,9, area of 35 mm² ± 5,8, and 24 ± 10 Mpa of tension. It did not have statistical difference between the two groups (p>0,05). CONCLUSION: The probability distribution showed that with 400 N the not tied tendons represents 83,8% of trustworthiness and the tied ones represents 78,5%.

RACIONAL: Segundo a hipótese do intestino longo, o comprimento do intestino delgado estaria relacionado à obesidade. Existem dados evolutivos, anatômicos e neuroendócrinos em favor desta afirmação. OBJETIVOS: Verificar a medida do comprimento do intestino delgado em pacientes obesos submetidos à cirurgia bariátrica e analisar a correlação entre o seu tamanho e o IMC. MÉTODO: A casuística foi composta de 30 pacientes do Serviço de Obesidade do Hospital Universitário Evangélico de Curitiba, submetidos à cirurgia bariátrica no período de março a junho de 2009, e previamente avaliados, verificando peso, altura e IMC. A técnica cirúrgica foi a de Fobi-Capella. A mensuração do intestino delgado foi realizada desde a flexura duodenojejunal à junção ileocecal, durante o trans-operatório, utilizando uma pinça de manipulação intestinal, a qual foi marcada com a medida de 10 cm. As alças intestinais foram medidas na margem anti-mesentérica, aplicando-se tensão mínima necessária para retificar as mesmas. Os dados foram correlacionados e tabulados estatisticamente, utilizando-se o método de correlação de Pearson. RESULTADOS: A média do comprimento intestinal para o sexo masculino foi de 582,5 cm e para o sexo feminino de 509,1. A partir do cálculo do coeficiente de correlação de Pearson, confirmou-se a ausência de correlação entre IMC e comprimento do intestino delgado. CONCLUSÃO: Obteve-se uma média de tamanho intestinal de 528,7 cm para toda a amostra. O comprimento do intestino delgado não possuiu correspondência com o IMC, não sendo um fator de significância neste estudo.

BACKGROUND: According to the hypothesis of long bowel, the length of small intestine is related to obesity. There are evolutionary, anatomics, neuroendocrines evidences in favor of this assertion. AIM: To check the measure of the small intestine length in overweight patients submitted to bariatric surgery and to analyze the correlation between the intestine size and Body Mass Index (BMI). METHODS: The sample was composed of 30 patients of Hospital Universitário Evangélico de Curitiba, Obesity Service, which had been submitted to bariatric surgery in the period between March and June, 2009. These patients previously had been evaluated, according to their weight, height and BMI. The surgical technique was Fobi-Capella. The measurement of small bowel was carried through the Treitz angle towards the ileocecal junction, during the operation. The instrument used was an intestinal manipulation clamp, which was marked with the measure of 10 cm. The intestinal handles had been measured in the anti-mesenteric edge, applying minimum tension necessary to rectify them. The data were statistically correlated and automatically tabulated, using the Pearson correlation method. RESULTS: The average intestinal length for males was 582,5 cm and for 509,1 cm, medium length 528,7 cm. Pearson correlation coefficient, confirmed the absence of correlation between BMI and the intestinal length. CONCLUSION: The medium length of small intestine was 528,7 cm and it did not have correspondence to BMI.

RACIONAL: A despeito de grandes avanços na elucidação dos fatores locais e gerais que interferem na cicatrização, o estado atual ainda não permite dispensar o uso do fio de sutura em intervenções cirúrgicas e a busca pelo fio ideal ainda continua. OBJETIVO: Avaliar suturas em ceco de ratos realizadas com fio monofilamentar absorvível de glicomer 60 (Monosyn®), em relação aos fios polipropilene (Prolene®) e poliglecaprone 25 (Monocryl®). MÉTODO: Foram utilizados 72 ratos Wistar, divididos em três grupos de 24 denominados grupo polipropilene, poliglecaprone e glicomer. Os animais de cada grupo foram distribuídos em três subgrupos de oito e avaliados no 3º, 7º e 14º dias de pós-operatório. Os parâmetros estudados foram: teste de resistência à insuflação de ar atmosférico; aspectos macroscópicos; as características histológicas e taxa de hidroxiprolina tecidual. RESULTADOS: O teste de resistência à insuflação de ar atmosférico apresentou baixos valores no 3º dia, com elevação no 7º, mantendo-se nos mesmos níveis no 14º dia, sendo semelhantes em todos os fios estudados. Na análise estatística, não se observou diferença significativa entre os grupos no 3º, 7º e 14º dias de pós-operatório, porém houve aumento significativo da pressão auferida, comparando-se os dias 3 e 7, em todos os grupos. Na análise histológica, avaliada quanto aos aspectos de reação inflamatória, proliferação fibroblástica, colagenização, coaptação das bordas e reepitelização, houve no 7º dia significativamente mais animais do grupo polipropilene com colagenização moderada em relação aos grupos poliglecaprone e glicomer. Os demais resultados não diferiram entre os fios estudados. Na determinação da taxa de hidroxiprolina tecidual, obteve-se a menor taxa nos subgrupos de 3º dia de pós-operatório, de todos os três grupos estudados, observando-se assim paralelismo com o ganho de resistência tênsil à insuflação de ar. CONCLUSÃO: O fio de Monosyn® equiparou-se, nos parâmetros estudados, aos fios de Prolene® e Monocryl® na realização de sutura em ceco de ratos.

BACKGROUND: Surgeons are still looking for the ideal suture material, although the progress during the last decades. AIM: To compare cecorrhaphy in rats with three different sutures: polypropylene (Prolene®), polyglecaprone 25 (Monocryl®) and glycomer 60 (Monosyn®). METHOD: Seventy-two Wistar rats were divided into three groups of 24 each (polypropylene, polyglecaprone and glycomer groups). Within each group, the animals were assigned to three subgroups and assessed on the 3rd, 7th and 14th postoperative day. The parameters studied were the bursting strength, macroscopic mucosal healing, histology and tissue hydroxyproline rate. RESULTS: The bursting strength had its lower values on the 3rd postoperative day, with significant higher values on the 7th and 14th; there was no difference among groups. In the overall, the histological analysis of the inflammatory reaction, fibroblast proliferation, collagen formation, border attachment and re-epithelization showed the same pattern among the three groups and sub-groups; only on day 7, there was seen a significant higher collagen formation in polypropylene group than polyglecaprone and glycomer groups. The tissue hydroxyproline rate was significant lower in the sub-groups of the 3rd postoperative day, in all of the three studies groups, with increase on days 7 and 14. There was a parallel gain in bursting strength and neo-collagen formation. CONCLUSION: There was no difference among glycomer 60, polypropylene and polyglecaprone sutures in performing cecorrhaphy in rats between days 3 and 14, according to the evaluated parameters.

RACIONAL: Muitas dúvidas ainda permanecem no que se refere às ações dos fatores de crescimento e do plasma rico em plaquetas sobre o mecanismo de reparação tissular. Há necessidade de serem esclarecidos pontos controversos ainda existentes. OBJETIVO: Obter o plasma rico em plaquetas em coelhos através de um método simplificado e ao mesmo tempo adequado, introduzindo um modelo experimental que possa ser utilizado em estudos posteriores. MÉTODOS: Foram utilizados 25 coelhas da raça Nova Zelândia e sem doenças prévias. Quinze mL de sangue de cada animal foi coletado, sendo 10 mL submetidos à dupla centrifugação. Para comprovar a efetividade do método proposto realizou-se contagem mecânica do sangue, bem como do produto final. RESULTADO: Obteve-se uma concentração média de plaquetas no plasma rico em plaquetas 687% maior que a contagem inicial observada no sangue venoso periférico. Para as variáveis: contagem inicial de plaquetas, contagem de plaquetas no plasma rico em plaquetas e enriquecimento, foram obtidos os limites de 95% de confiança para suas médias, sendo que, no que se refere ao percentual de enriquecimento, existe 95% de chance de que o intervalo de (530-844) contenha a média real de enriquecimento de plaquetas. CONCLUSÃO: O método simplificado utilizado permite a obtenção de plasma rico em plaquetas adequado permitindo seu uso em estudos dos fatores de crescimento nos mecanismos de reparação tecidual.

BACKGROUND: Multiple uncertainties still exist about the action of the growth factors and the platelet-rich plasma on the mechanism of repair. AIM: To obtain the platelet-rich plasma in rabbits through a simplified and suitable method, creating an experimental model. METHODS: Twenty-five female New Zealand rabbits without previous diseases were used. Fifteen mL of blood of each rabbit was collected and 10 mL of the collected blood were twice centrifugated. To check the effectiveness of the proposed method mechanical counting of the blood and of the final product were performed. RESULTS: The mean platelet concentration was 687% higher than the initial peripheral blood counting. Mean initial platelet counting, platelet-rich plasma platelet counting and enriched were obtained with 95% CI and in terms of enrichment percentage there is a chance that the interval 530 to 844 be the real mean platelet enrichment. CONCLUSION: This simplified method permits to get an effective platelet-rich plasma to be used in trials about growth factors in mechanisms of tissue repair.
