RESUMO: Os objetivos deste estudo foram: 1) determinar IgG total específica e seus subisotipos no soro e colostro de éguas vacinadas contra Theileria equi e Streptococcus equi subspécie equi; 2) determinar IgG total específica e seus subisotipos em potros provenientes dessas éguas. Em éguas vacinadas contra T. equi, o maior nível sérico de IgG total específico ocorreu aos 335 dias de gestação, decaindo 30 dias pós parto, enquanto nas vacinadas contra S. equi, ocorreu aos 328 dias de gestação e manteve-se pelo mesmo período. A avaliação de IgG total específica no colostro de éguas vacinadas demonstrou níveis de IgG proporcionais aos títulos do soro materno. Os isotipos de IgG mais detectados foram IgG3/5 e IgG4/7 para ambos antígenos avaliados. Potros provenientes de éguas vacinadas com antígeno de T. equi mantiveram os níveis de IgG específica até dois meses de idade, enquanto potros provenientes de éguas vacinadas com antígeno de S. equi mantiveram por três a quatro meses, sugerindo que esse seja o período ideal para início do esquema vacinal em potros.

ABSTRACT: The aims of this study were: 1) determine total specific IgG and subisotypes in serum and colostrum of pregnant mares vaccinated against Theileria equi and Streptococcus equi subspecies equi; 2) determine total specific IgG and subisotypes in foals born from these mares. In mares, the highest total serum IgG value for T. equi was at 335 days of gestation declining 30 days postpartum, while for S. equi was at 328 days of gestation remaining high up to 30 days postpartum. Transfer of passive immunity against both antigens was observed with specific IgG values in colostrum and foals’ serum proportional to mares’ values. The most detected specific IgG subisotypes were IgG3/5 and IgG4/7 for both antigens. Foals born from mares immunized with T. equi kept maternal IgG values until 2 months of age, while those born from mares immunized with S. equi kept maternal IgG values until 3-4 months of age. These results suggest that foals should be vaccinated after this period.

Resumo O objetivo deste trabalho foi avaliar a modulação imunológica precoce e tardia da infecção experimental de larvas de T. canis em camundongos. Estes foram infectados com 100 larvas infectantes e eutanasiados em diferentes períodos: 24 e 48 horas pós infecção (HPI), 15 e 30 dias após a infecção (DPI). A resposta humoral foi avaliada por ELISA indireto. RT-PCR quantitativo (qPCR) foi usado para quantificar a transcrição de mRNA das citocinas IL4, IL10, IL12 e Ym1 nos períodos de infecção precoce e tardia. A infecção por T. canis foi capaz de gerar IgG total específico aos 15 e 30 DPI. A análise do isótipo IgG revelou uma diferenciação significativa para IgG1 em comparação com IgG2a, IgG2b e IgG3, caracterizando uma resposta Th-2. Avaliando-se a transcrição gênica na fase inicial da infecção, foram observados níveis mais elevados de transcrição de IL10, IL4 e Ym1 e a regulação negativa de IL12. Na fase tardia, foram observados níveis aumentados de transcrição de IL4, Ym1 e IL12, e foi observada regulação negativa da transcrição de IL-10. Os dados obtidos sugerem, que durante a infecção experimental com T. canis, a participação das citocinas IL4, IL10, IL12 e Ym1 podem desempenhar um papel importante na imunomodulação de T. canis.

Abstract The objective of this work was to evaluate the early and late immunological modulation of an experimental infection of T. canis larvae in mice. Mice were infected with 100 infective larvae and euthanized at different period: 24, 48 hours post infection (HPI), 15- and 30 days post infection (DPI). The humoral response was evaluated by indirect ELISA. Quantitative RT–PCR (qPCR) was used to quantify the mRNA transcription of cytokines IL4, IL10, IL12 and Ym1 in the early and late infection periods. Infection with T. canis was able to generate specific total IgG at 15- and 30- DPI. Analyzing the IgG isotype revealed a significant differentiation for IgG1 compared with IgG2a, IgG2b and IgG3, characterizing a Th-2 response. Evaluating the gene transcription at the early phase of infection, higher transcription levels of IL10, IL4 and Ym1 and a downregulation of IL12 were observed. By the late phase, increased transcription levels of IL4, Ym1 and IL12 were observed, and downregulation of IL-10 transcription was observed. The data obtained suggest that during experimental infection with T. canis, the participation of the IL4, IL10, IL12 cytokines and Ym1 can play an important role in T. canis immunomodulation.

Abstract Coccidiosis, a disease caused by the parasitic Eimeria spp., affects birds of all ages, particularly young birds more intensely. Infected poultry presents significant economic losses. Bacillus thuringiensis var israelensis (Bti) is a Gram-positive, spore-forming bacterium that produces proteins with high specific parasiticidal activity against various orders of parasites. Thus, the aim of the present study was to evaluate the parasiticidal potential of Bti in quails that were naturally infected with Eimeria bateri. Twenty 12-week-old male quails (Coturnix coturnix coturnix), naturally infected with Eimeria bateri, were randomly divided into two groups of 10 birds: Bti treated and control. The treated group was supplemented with Bti (1×108 spores∙g-1) in the feed, while; the control group received the same feed without Bti. To evaluate the occurrence of oocysts, samples of feces were collected every week for four weeks. Significant (P < 0.05) oocysts reductions of 56.64% and 94.51% were noted in the Bti treated group at 2nd and 4th week of study, respectively. The Bti supplementation may contribute to the reduction of oocysts in quails and environmental contamination. Bacillus thuringiensis var israelensis appeared to be a promising complementary alternative in E. bateri control.

Abstract The Equine Strangles, caused by Streptococcus equi subs. equi, is a contagious disease, causing high rates of morbidity been responsible for important economic losses. The M protein synthesized by S. equi plays an important role in the pathogenesis and is a promising candidate for a vaccine antigen. The innate immune system is responsible for the first immune response against microorganisms, this response is mediated by receptors that detect PAMPs and their activation trigger crucial modulation of the adaptative immune response. This work describes the immune response of S. equi subs. equi. recombinant SeM protein, using Escherichia coli BL21 (DE3) as an expression and delivery vaccine system. To characterize and to determine the vaccine efficacy, mice were vaccinated as followed: 1. Recombinant E. coli expressing rSeM protein; 2. The same recombinant E. coli, inactivated adsorbed in Alumen; 3. Purified rSeM protein adsorbed in Alumen; 4. Inactivated S. equi whole cells adsorbed in Alumen; 5. Control group. All vaccinated mice developed protective response against S. equi infection, however the groups that received the E. coli expressing rSeM presented significant higher IgG level than other vaccinated groups. The recombinant E. coli delivery vaccine system also induced a highest IgG response than inactivated S. equi or purified rSeM vaccines in horses. This study evidence that the recombinant E. coli, live or inactivated, enhanced the humoral response, reaching significant higher antibodies levels than those obtained in the vaccination with the bacterin or purified antigen, showing the feasibility of producing low-cost vaccines against strangles.

Abstract Gonadotropin-releasing hormone (GnRH) is one of the main targets for the development of immunocontraceptives vaccines. The aim of this study was to clone and express the recombinant GnRH fused to the B subunit of Escherichia coli heat-labile enterotoxin (LTB) molecule in Pichia pastoris and Escherichia coli platforms and evaluate their immunogenicity in mice. P. pastoris (pGnRH/LTB) and E. coli (eGnRH/LTB) platforms were able to express GnRH/LTB expected band with ~ 21 kDa. Both constructions were immunogenic in mice. Similar IgG kinetics was observed for both construction when it was used as ELISA antigen respectively, showing significant (p<0.05) IgG levels 5-fold higher than a commercial vaccine and 14-fold higher than the controls. The histological effects of pGnRH/LTB as well as eGnRH/LTB proteins demonstrated a significant effect on the gonads, characterized by atrophy of seminiferous tubules, absence of spermatogenesis and reduction of Leydig cells. Both constructions were able to induce antibodies that block the hormone effect, suggesting the potential of GnRH/LTB, independently of the P. pastoris or E. coli platform used, as a vaccine candidate for immunocontraception.

Resumo As infecções por nematódeos gastrintestinais (ING) constituem a maior limitação à produção de pequenos ruminantes. Na busca do controle desses parasitos, estudos com plantas medicinais têm sido uma importante alternativa. Visto isto, o estudo desenvolvido teve como objetivo avaliar a ação ovicida e larvicida in vitro do óleo essencial de Rosmarinus officinalis. O óleo foi extraído, analisado por cromatografia gasosa e testado sobre ovos e larvas de ING em seis concentrações, 227,5mg/mL; 113,7mg/mL; 56,8mg/mL; 28,4mg/mL; 14,2mg/mL; 7,1mg/mL. Para determinar a ação ovicida, ovos de ING foram recuperados de fezes de ovinos e incubados por 48h com as diferentes concentrações do óleo. Na avaliação da migração das larvas, as larvas de terceiro estágio (L3) foram obtidas por coprocultura, e associadas ao óleo essencial por 24h nas mesmas concentrações, permanecendo por mais 24h em microtamises, seguindo-se a contagem de larvas que migraram e que não migraram. Os testes in vitro com o óleo de R. officinalis mostraram o nível de significância (p<0.05) 97,4% a 100% na inibição da eclodibilidade e 20% a 74% na inibição da migração das larvas. Na análise por cromatografia gasosa o constituinte majoritário foi o eucaliptol. Os resultados apresentados mostram que o óleo essencial de R. officinalis possui ação ovicida e larvicida sobre ING de ovinos.

Abstract Gastrointestinal Nematode Infection (GIN) are the main constraint to the production of small ruminants. Studies of medicinal plants have been an important alternative in the effort to control these parasites. Therefore, the purpose of this study was to evaluate the in vitro ovicidal and larvicidal activity of essential oil of Rosmarinus officinalis. The oil was extracted, analyzed by gas chromatography and tested on GIN eggs and larvae in six concentrations, 227.5mg/mL, 113.7mg/mL, 56.8mg/mL, 28.4mg/mL, 14.2mg/mL and 7.1mg/mL. To determine the ovicidal activity, GIN eggs were recovered from sheep feces and incubated for 48h with different concentrations of the oil. For the evaluation of larval migration, third-stage larvae (L3) were obtained by fecal culture, and associated with the essential oil for 24h at the same concentrations, after which they were left for another 24 hours on microsieves, followed by the count of migrating and non-migrating larvae. The assays of R. officinalis oil showed a significant (p<0.05) 97.4% to 100% inhibition of egg hatching and a significant (p<0.05) 20% to 74% inhibition of larval migration. The main constituent revealed by gas chromatography was Eucalyptol. The results indicate that R. officinalis essential oil has ovicidal and larvicidal activity on sheep GINs.

RESUMO: O gênero Staphylococcus abrange algumas das bactérias patogênicas mais importantes tanto para humanos como para animais. Ele é responsável pela mastite bovina e otite canina, além de estar presente na microbiota de animais e como contaminante em alimentos. Sua patogênese está relacionada à formação de cápsula e biofilme, que contribuem para sua infectividade. O objetivo deste estudo foi observar a produção de slime layer e a formação de biofilme, que estão relacionados à resistência a antibicrobianos e à presença dos genes icaA e icaD, em 41 isolados de Staphylococcus spp. de diferentes origens fornecidos pelo Laboratório Regional de Diagnóstico (LRD) da Universidade Federal de Pelotas (UFPEL). Os isolados de Staphylococcus spp. foram cultivados em ágar vermelho do Congo para detecção de cápsulas. A formação de biofilme foi detectada usando o teste de microplaca com 96 poços. O teste de susceptibilidade antimicrobiana foi realizado usando o método de difusão em placa. Parte das amostras analisadas produziram slime layer (36,6%) e formaram biofilme (17,1%). Entretanto, seis daquelas que formaram biofilmes foram sensíveis aos oito antibióticos testados no antibiograma. Em testes para determinar as concentrações bactericidas e inibitórias mínimas, a resistência à gentamicina de cepas formadoras de biofilme foi maior que aquela das cepas não formadoras. O antimicrobiano menos eficaz foi a ampicilina (51%), seguida por tetraciclina (71%), neomicina (73%) e eritromicina (73%). Alguns isolados apresentaram os genes icaA (6) e icaD (11). Portanto, sugerimos que a origem de um isolado pode determinar sua expressão de fator de virulência e resistência a certos antibióticos.

ABSTRACT: The genus Staphylococcus comprises some of the most important pathogenic bacteria for both humans and animals. It is responsible for bovine mastitis and canine otitis, besides being present in the microbiota of animals and as a contaminant in food. Its pathogenesis is related to the formation of capsule and biofilm, which contribute to its infectivity. The objective of this study was to observe the production of slime layer and formation of biofilm, which are related to the resistance to antimicrobial agents and presence of icaA and icaD genes, in 41 isolates of Staphylococcus spp. from different origins, provided by the Universidade Federal de Pelotas (UFPEL), Laboratório Regional de Diagnóstico (LRD). Strains of Staphylococcus spp. were cultivated in Congo red agar for capsule detection. Biofilm formation was detected using the 96-well microplate testing. Antimicrobial susceptibility testing was performed using the plate diffusion method. Part of the analyzed samples produced slime layer (36.6%) and formed biofilm (17.1%). However, six of those that formed biofilms were susceptible to the eight antibiotics tested in the antibiogram. In tests to determine the minimum bactericidal and inhibitory concentrations, gentamicin resistance of biofilm-forming strains was greater than that of non-forming strains. Ampicillin was the least effective antimicrobial drug (51%), followed by tetracycline (71%), neomycin (73%), and erythromycin (73%). Some isolates presented the icaA (6) and icaD (11) genes. Therefore, we suggested that the origin of an isolate can determine its expression of virulence factor and resistance to certain antibiotics.

Resumo Este estudo avaliou a ação in vitro e in vivo de extratos de Eugenia uniflora em nematódeos gastrintestinais de ovinos. No teste de inibição da eclodibilidade dos ovos, extratos aquosos e hidroalcoólicos foram testados nas concentrações de 40 a 1,25 mg/mL. O efeito citotóxico foi mensurado através do ensaio de MTT em células VERO nas concentrações de 4000 a 1,95 µg/mL. A composição química foi analisada através da marcha fitoquímica qualitativa. No teste in vivo, foram administrados 100 mg/kg, por via oral, do extrato hidroalcoólico por três dias em ovinos naturalmente infectados e realizada coleta de fezes para estimar a redução de ovos por grama de fezes (OPG). Diferentes extratos de E. uniflora inibiram a eclodibilidade dos ovos, com percentual de inibição variando de 14,56 a 99,75%, sendo os hidroalcoólicos mais promissores que os aquosos. Na marcha fitoquímica, compostos com ação anti-helmíntica comprovada foram observados, como flavonoides, saponinas, taninos e triterpenos. Nas condições testadas, os extratos apresentaram citotoxicidade nula a moderada. In vivo, a redução do OPG foi observada apenas no grupo controle (anti-helmíntico). Os extratos de E. uniflora mostraram-se promissores com ação in vitro, necessitando estudos que avaliem outras concentrações e formas de administração in vivo.

Abstract: This study evaluated the in vitro and in vivo action of Eugenia uniflora extracts on sheep gastrointestinal nematode. In the egg hatchability inhibition assay, aqueous and hydroalcoholic extracts were tested in the concentrations ranging from 40 to 1.25 mg/mL. The cytotoxic effect was measured using the MTT assay on VERO cells at concentrations ranging from 4000 to 1.95 μg/mL. The chemical composition was analyzed through the qualitative phytochemical screening. In the in vivo test, 100 mg/kg of the hydroalcoholic extract was administered orally for three days in sheep naturally infected and fecal collection was performed to estimate the egg count per gram of faeces (EPG). Different extracts of E. uniflora inhibited hatchability of eggs with a percentage of inhibition ranging from 14.56 to 99.75%, being the hydroalcoholic most effective than the aqueous. In phytochemical analysis, compounds with anthelmintic action were observed, such as flavonoids, saponins, tannins and triterpenes. Extracts showed moderate to null cytotoxicity under the conditions tested. Reduction of EPG was observed only in control group (anthelmintic). The extracts of E. uniflora proved promising with in vitro action, requiring studies that assess other concentrations and forms of in vivo administration.

RESUMO: O objetivo do presente estudo foi avaliar a eficácia da PCR multiplex na detecção da adulteração por adição e/ou substituição de carne bubalina em carne moída bovina, comercialmente disponível. A fim de determinar o limite de detecção da técnica, carnes moídas bovinas fraudadas intencionalmente foram fabricadas em triplicata e, adicionalmente, concentrações conhecidas de DNA das espécies Bostaurus e Bubalusbubalis foram diluídos. Ao mesmo tempo, 91 amostras de carne moída comercializadas como sendo de origem bovina foram coletadas em diferentes comércios na região Norte do Brasil e a PCR multiplex proposta foi realizada. Os resultados demonstraram que o DNA bubalino foi detectado em 17,5% das amostras de carne moída coletadas. Concluiu-se que a PCR multiplex é uma técnica eficaz, capaz de detectar a incorporação de carne moída bubalina em percentagens que variaram de 10 a 100%, e a incorporação de carne bovina em percentagens que variaram de 0,1 a 100% em amostras de carne moída.

ABSTRACT: The aim of the present study was to assess the efficacy ofmultiplex PCR in detecting the adulterationof commercially available ground beefvia addition and/orsubstitution ofground buffalo meat. Experimentally adulterated ground beefsamples were prepared in triplicate, and dilutions of DNA from Bos taurus and Bubalusbubalis were prepared to determine the detection limit of the method. Concurrently, 91 ground meatsamples sold as “ground beef” were collected from differentstores in northern Brazil andanalyzed bymultiplex PCR. Buffalo DNA was detected in 17.5% of the collected ground meat samples.Our results showed that multiplex PCR is an efficient method for detectingthe incorporation of groundbuffalo meatatpercentages ranging from 10 to 100% and the incorporation of beef at percentages ranging from0.1 to 100% intoground meat samples.

RESUMO: O estudo foi realizado em uma fazenda de criação de gado de corte na região do extremo sul do Brasil. O nascimento de um bezerro com opacidade unilateral da córnea foi imediatamente relatado ao Laboratório de Parasitologia da Universidade Federal de Pelotas (RS) e coletou-se sangue da vaca e do bezerro antes da ingestão do colostro. Foram coletados sangue e cordão umbilical do bezerro com opacidade na córnea bem como de outros seis bezerros aparentemente saudáveis antes da ingestão de colostro. Foram realizados exames sorológicos utilizando o teste indireto de anticorpos fluorescentes (IFAT) com ponto de corte de 1: 100 e reação em cadeia da polimerase (PCR). O resultado dos exames sorológicos mostraram que o bezerro foi positivo para N. caninum ao nascer com titulação de 1:1600 e o de sua mãe foi de 1:3200 na IFAT. A PCR do cordão umbilical foi positiva para o agente no animal com opacidade córnea unilateral e a identidade molecular do produto amplificado foi confirmada por sequenciamento. A detecção de DNA de N. caninum no cordão umbilical é uma nova alternativa para o diagnóstico deste agente em bezerros recém-nascidos com suspeita clínica de neosporose.

ABSTRACT: This study was conducted at a beef cattle breeding farm in the far southern region of Brazil. The birth of a calf with unilateral corneal opacity was immediately reported to the Laboratory of Parasitology, in the Universidade Federal de Pelotas, Rio Grande do Sul, Brazil; blood was collected from the cow and calf before colostrum intake. The umbilical cords from this calf and from six other healthy animals were collected. Serological examination, utilizing an indirect fluorescent antibody test, was done using a cut-off point of 1:100. Polymerase chain reaction (PCR) was also performed using the umbilical cord samples. Serological tests showed that the calf was positive for Neospora caninum at birth, with a titer of 1:1600; a titer of 1:3200 was reported in the dam. PCR, using umbilical cord tissue from the affected animal, was positive for the presence of this parasite, and the molecular identity of the amplified product was confirmed by sequencing. Therefore, the detection of N. caninum DNA in the umbilical cord represents a novel alternative test for the diagnosis of this parasitic infection in newborn calves that are clinically suspected to have neosporosis.

RESUMO: Eficientes programas de manejo que permitem o fornecimento de colostro de alta qualidade e em quantidades adequadas para bezerros são necessários para garantir a sua saúde e sobrevivência. Substitutos do leite tornam-se frequentemente necessários quando o colostro apresenta qualidade inadequada ou a fim de quebrar o ciclo de transmissão de doenças infecciosas. Este estudo teve como objetivo avaliar o efeito da fermentação anaeróbica do colostro bovino (silagem de colostro) sobre bactérias de interesse para a saúde animal. Alíquotas de colostro foram inoculadas com culturas de Brucella abortus , Escherichia coli , Leptospira interrogans serovar Copenhageni , Mycobacterium bovis , Salmonella Enteritidis , Salmonella Typhimurium , Staphylococcus aureus e Bacillus cereus e submetidas ao processo de fermentação anaeróbia. No primeiro dia e a cada sete dias até o 30o dia de fermentação, as alíquotas foram cultivadas e analisadas pela contagem de unidades formadoras de colônias. Aos sete dias de fermentação, unidades formadoras de colônias de B. abortus, L. interrogans e M. bovis não foram detectadas e unidades formadoras de colônia de E. coli , S. aureus , S. Enteritidis e S. Typhimurium não foram observadas no 14o dia de fermentação. No entanto, B. cereus e bactérias ácido lácticas foram detectadas até o 30o dia de fermentação. Este estudo sugere que o processo de fermentação anaeróbia do colostro bovino proporciona efeitos inibitórios sobre as bactérias avaliadas, constituindo-se num promissor método de conservação do colostro bovino.

ABSTRACT: Efficient handling programs that provide high quality colostrum in adequate amounts to dairy farm calves are needed to assure their health and survival. Replacers (or milk substitutes) often become necessary when colostrum presents inadequate quality, or in order to break the cycle of infectious disease transmission. In this study we aimed to assess the effect of anaerobic fermentation processing (colostrum silage) on bacterial that represent interest to animal health. Colostrum samples were inoculated with cultures of Brucella abortus , Escherichia coli , Leptospira interrogans serovar Copenhageni , Mycobacterium bovis , Salmonella Enteritidis , Salmonella Typhimurium , Staphylococcus aureus , and Bacillus cereus and then subjected to anaerobic fermentation. On the first day, and every seven days until 30th days after fermentation, the samples were cultured and colony forming units counted. At seven days of fermentation, B. abortus , L. interrogans , and M. bovis were not detected. At 14th days of fermentation, E. coli , S. aureus , S. Enteritidis and S. Typhimurium were no longer detected. However, we were able to detect both lactic acid bacteria and B. cereus until 30th days of fermentation. From this study we suggested that anaerobic fermentation processing can inhibit important bacteria that cause economical losses for the cattle industry. The observations suggested that colostrum silage is a promising form to conserve bovine colostrum.

El objetivo de este estudio fue evaluar diferentes concentraciones de aceite de oliva para la criopreservación de semen de verraco. Se utilizaron 21 eyaculados de 18 machos con motilidad igual o superior al 70%. Para la congelación, el semen se diluyó en los tratamientos: control (Cont) (lactosa yema), 0,25% (A025); 0,50% (A050); 0,75% (A075) y 1,0% (A10) de aceite de la variedad "Koroneiki" en lactosa yema. Pajas con 5.107 espermatozoides / ml, fueron congeladas y almacenadas en nitrógeno líquido. La descongelación se produjo a 37°C durante 30 segundos y se evaluó: la motilidad, la funcionalidad de la mitocondria, la integridad del ADN, la tasa de penetración de ovocitos y el número de espermatozoides por ovocito (en el ensayo de penetración in vitro). Las variables se compararon mediante la prueba de Kruskal-Wallis. Aunque no hubo diferencia estadística entre el control y los tratamientos que contienen aceite de oliva (p> 0,05), la concentración de 0,25% de este aceite mostró tendencias de protección de esperma, superando el control en la funcionalidad de las mitocondrias y en la preservación de la capacidad fertilizante en el ensayo de penetración in vitro. Por lo tanto, el aceite de oliva puede representar una alternativa para ayudar en la criopreservación de semen porcino

The aim of this study was to evaluate different concentrations of olive oil for cryopreservation of boar semen. A total of 21 eyaculates of 18 males with motility ≥70% were used. For freezing, the semen was diluted in treatments: control (Cont-egg yolk), "Koroneiki" oil in egg yolk at 0.25% (A025), 0.50% (A050), 0.75% (A075) and 1.0% (A10). Straws with 5.107 sperm/ml were frozen and stored in liquid nitrogen. Thawing was done at 37ºC for 30 seconds and motility, mitochondrial functionality, DNA integrity, oocite penetration rate and the number of sperm per oocite (in the in vitro penetration trial) were evaluated. The variables were compared using the Kruskal-Wallis test. Even though no statistical difference was found between control and treatments containingolive oil (p>0.05), the treatment with 0.25% oil concentration showed tendences towards sperm protection, outperforming the controls in mitochondrial functionality and preservation of the fertilizing capabilities in the in vitro penetration trial. This is why, olive oil could represent an alternative to help cryopreservation of boar semen.

O objetivo de este estudo foi avaliar diferentes concentrações de azeite de oliva para a criopreservação de sêmen suíno. Utilizaram-se 21 ejaculados de 18 machos com motilidade igual ou superior ao 70%. Para o congelamento, o sêmen se dilui-o nos tratamentos: controle (Cont) (lactose-gema), 0,25% (A025); 0,50% (A050); 0,75% (A075) e 1,0% (A10) de azeite da variedade "Koroneiki" em lactose-gema. Palhetas com uma dose de 5.107 espermatozoides/ ml foram congeladas e armazenadas em nitrogênio liquido. O descongelamento se produz a 37°C durante 30 segundos e se avaliaram: motilidade, funcionalidade da mitocôndria, integridade do DNA, taxa de penetração de ovócitos e o número de espermatozoides por ovócito (no teste de penetração in vitro). As variáveis se compararam mediante o teste de Kruskal-Wallis. Ainda que não houve diferença estatística entre o controle e os tratamentos que continham azeite de oliva (p> 0,05), a concentração de 0,25% de azeite de oliva teve tendências de proteção do esperma, superando o controle na funcionalidade das mitocôndrias e na preservação da capacidade fertilizante no teste de penetração in vitro. Assim sendo, o azeite de oliva pode representar uma alternativa para ajudar na criopreservação do sêmen suíno.

Salmonella está entre os principais micro-organismos patogênicos veiculados por alimentos. A patogenicidade dessa bactéria está relacionada a uma série de fatores de virulência e, dentre estes, podemos citar os flagelos. A expressão do flagelo está entre os fatores de virulência modulados por Quorum Sensing. A adrenalina produzida pelos mamíferos utiliza a mesma via de sinalização do autoindutor 3 das bactérias. Nesse sentido, este trabalho teve como objetivo avaliar o efeito de moléculas indutora (adrenalina) e autoindutoras (auto-indutor 2 e auto-indutor 3) e a associação destas na motilidade, no crescimento celular e na expressão dos genes flhC, fliA, fliY, motA, motB e fliC de Salmonella Typhimurium (ST). Inicialmente, ST foi semeada em caldo BHI. Após, ensaios de motilidade, curvas de crescimento e expressão gênica foram feitos, testando diferentes concentrações de adrenalina (50, 125, 250, 500µM), meio condicionado (10 e 50%) e a associação destes. A partir dos resultados obtidos, observou-se que o tratamento que utilizou 50% de meio condicionado + 500µM de adrenalina aumentou a motilidade de ST, em decorrência do aumento de genes envolvidos com montagem do flagelo.

Genus Salmonella bacteria are among the major pathogenic microorganisms in food. This bacterium pathogenicity is related to a number of virulence factors, among which its flagella. Flagellum expression is one of the virulence factors modulated by Quorum Sensing. Epinephrine produced by mammals uses the same signaling pathway of the 3 bacteria autoinducer. This study evaluated the effect of molecules inducer (epinephrine) and autoinducers (autoinducer 2 and autoinducer 3) and their association with the motility, growth and expression genes flhC, fliA, fliY, motA, motB e fliC of Salmonella Typhimurium (ST). Initially, ST was inoculated in BHI. Then, motility assays, growth curves and gene expression were performed by testing different concentrations of epinephrine (50, 125, 250, 500µM), conditioned medium (10 and 50%) and a combination of these. ST was exposed to different concentrations of epinephrine, conditioned medium and an association of both. Following, motility assays, bacterial growth and gene expression were performed. The results obtained showed that the combination of 500uM epinephrine with 50% conditioned medium increased ST bacterial motility by increasing the expression of genes involved in flagellum assembly.

A erliquiose monocítica canina, causada por Ehrlichia canis, é uma doença endêmica em diversas regiões do Brasil. Algumas técnicas de diagnóstico sorológico, utilizando proteínas imunodominantes de E. canis como antígenos, estão disponíveis, porém suas especificidades e sensibilidades são questionáveis. Com base nesse fato, o objetivo deste trabalho foi testar o potencial antigênico da proteína GP19 recombinante (rGP19) para posterior utilização em testes diagnósticos. A rGP19, expressa em E. coli cepa BL21 (DE3) C41, foi reconhecida por soros de cães experimentalmente infectados pelas técnicas de ELISA e Western blotting. Dessa maneira, conseguiu-se obter um antígeno promissor com a capacidade de diferenciar animais positivos de negativos, até mesmo uma semana após a infecção.

The canine monocytic ehrlichiosis, caused by Ehrlichia canis, is endemic in several regions of Brazil. Some serological diagnostic techniques using immunodominant proteins of E. canis as antigens are available, but their specificities and sensitivities are questionable. Based on this, the objective of this study was to test the antigenic potential of the recombinant gp19 protein (rGP19) for subsequent use in diagnostic tests. The rGP19 expressed in the Escherichia coli strain BL21 (DE3) C41 was recognized in the sera from experimentally infected dogs using ELISA and Western blotting. Thus, it was possible to obtain a promising antigen with the ability to differentiate between E. canis-positive and -negative animals, even 1 week after infection.

A neosporose é uma doença causada pelo protozoário Neospora caninum que leva a perdas econômicas importantes em muitos países. No presente estudo, é descrita a utilização da proteína recombinante NcSRS2 de N. caninum expressa em Pichia pastoris em um ensaio imunoenzimático indireto (ELISA) para o diagnóstico de infecção por Neospora em ovelhas e cães. Observou-se, que utilizando-se um ELISA, o teste produziu uma especificidade de 94,5% e uma sensibilidade de 100% para ovinos; e uma especificidade de 93,3% e sensibilidade de 100% para cães. Uma maior sensibilidade foi observada em relação à IFI que foi confirmada por Western blot. Os resultados deste estudo sugerem que a proteína recombinante expressa em P. pastoris é bom antígeno para ser utilizado no diagnóstico imunológico para detectar N. caninum em duas espécies importantes expostas a esta parasitose

Neosporosis is a disease caused by the protozoon Neospora caninum that leads to significant economic losses in many countries. In the present study, we report on use of the recombinant protein NcSRS2 of N. caninum expressed in Pichia pastoris in an indirect immunoenzymatic assay (ELISA) for diagnosing neosporosis infection in sheep and dogs. We observed that the ELISA test yielded specificity of 94.5% and sensitivity of 100% for sheep and specificity of 93.3% and sensitivity of 100% for dogs. We observed that the sensitivity was higher than shown by the indirect fluorescent antibody test, and this was confirmed by means of Western blot. The results from this study suggest that the recombinant protein expressed in P. pastoris is a suitable antigen for use in immunodiagnosis to detect N. caninum in two important species exposed to this parasitosis.
