RESUMO Objetivo Investigar os efeitos do treinamento físico nos parâmetros morfológicos e metabólicos de ratos diabéticos. Métodos Ratos Wistar foram randomizados para quatro grupos: controle sedentário, controle treinado, diabético sedentário e diabético treinado. Diabetes mellitus foi induzido por administração de Aloxana (35mg/kg) nos Grupos Diabético Sedentário e diabético treinado. O protocolo de treinamento físico incluiu natação com carga de 2,5% do peso corporal, por 60 minutos por dia (5 dias por semana) para os Grupos Controle Treinado e diabético treinado, durante 6 semanas. Ao final do experimento, os ratos foram sacrificados, e o sangue foi coletado para determinação das concentrações séricas de glicose, insulina, albumina e proteínas totais. Amostras do fígado foram coletadas para determinação do glicogênio, proteínas, DNA e diâmetro mitocondrial. Resultados O Grupo Sedentário Diabético apresentou redução no peso corporal, insulinemia e glicogênio hepático, além de maior glicemia e diâmetro mitocondrial hepático. O protocolo de treinamento físico em animais diabéticos foi eficiente para restaurar o peso corporal e o glicogênio hepático, além de reduzir o diâmetro mitocondrial hepático. Conclusão O treinamento físico melhorou o metabolismo hepático e promoveu importantes adaptações morfológicas, como no diâmetro mitocondrial no fígado de animais diabéticos.

ABSTRACT Objective To investigate the effects of physical training on metabolic and morphological parameters of diabetic rats. Methods Wistar rats were randomized into four groups: sedentary control, trained control, sedentary diabetic and trained diabetic. Diabetes mellitus was induced by Alloxan (35mg/kg) administration for sedentary diabetic and Trained Diabetic Groups. The exercise protocol consisted of swimming with a load of 2.5% of body weight for 60 minutes per day (5 days per week) for the trained control and Trained Diabetic Groups, during 6 weeks. At the end of the experiment, the rats were sacrificed and blood was collected for determinations of serum glucose, insulin, albumin and total protein. Liver samples were extracted for measurements of glycogen, protein, DNA and mitochondrial diameter determination. Results The sedentary diabetic animals presented decreased body weight, blood insulin, and hepatic glycogen, as well as increased glycemia and mitochondrial diameter. The physical training protocol in diabetic animals was efficient to recovery body weight and liver glycogen, and to decrease the hepatic mitochondrial diameter. Conclusion Physical training ameliorated hepatic metabolism and promoted important morphologic adaptations as mitochondrial diameter in liver of the diabetic rats.

Abstract Epidemiological studies indicate continuous increases in the prevalence of Alzheimer’s Disease (AD) in the next few decades. The key feature of this disease is hippocampal neurodegeneration. This structure has an important role in learning and memory. Intense research efforts have sought to elucidate neuroprotective mechanisms responsible for hippocampal integrity. Insulin signaling seems to be a very promising pathway for the prevention and treatment of AD. This hormone has been described as a powerful activator of neuronal survival. Recent research showed that reduced insulin sensitivity leads to low-grade inflammation, and both phenomena are closely related to AD genesis. Concomitantly, exercise has been shown to exert anti-inflammatory effects and to promote improvement in insulin signaling in the hippocampus, which supports neuronal survival and constitutes an interesting non-pharmacological alternative for the prevention and treatment of AD. This review examines recent advances in understanding the molecular mechanisms involved in hippocampal neuroprotection mediated by exercise.

OBJETIVO: Comparar a técnica neurocirúrgica clássica a uma nova técnica simplificada, para correção de mielomeningocele, em fetos de ovelhas. MÉTODOS: Em 9 fetos, foi criado um defeito semelhante à mielomeningocele (laminectomia e excisão de dura-máter) no 90º dia de gestação. O tipo de correção foi randomizado. No Grupo 1, o defeito foi corrigido usando a técnica neurocirúrgica clássica, com a sutura de três camadas (dura-máter, músculo e pele), realizada por um neurocirurgião. No Grupo 2, um especialista em Medicina Fetal utilizou a técnica simplificada, colocando um fragmento de celulose biossintética sobre a medula e suturando apenas da pele sobre a celulose. Próximo ao termo da gestação (132 dias), os fetos foram sacrificados para análise anatomopatológica. RESULTADOS: Ocorreram dois casos de aborto e uma morte materna, restando seis casos para avaliação - três em cada grupo. No Grupo 1, todos os casos mostraram aderência da medula à cicatriz (meningoadesão) e perda da arquitetura medular, com destruição do funículo posterior e perda da visualização da substância cinzenta. No Grupo 2, observou-se, em todos os casos, a formação de uma neodura-máter, separando o tecido nervoso do músculo adjacente, sendo que o funículo posterior e a substância cinzenta estavam preservados. CONCLUSÃO: A técnica simplificada foi superior à neurocirúrgica, com maior preservação da medula e evitando as aderências do tecido nervoso. Os presentes achados sugerem que a técnica utilizada atualmente na correção de mielomeningocele em fetos humanos deva ser reavaliada.

OBJECTIVE: To compare the classical neurosurgical technique with a new simplified technique for prenatal repair of a myelomeningocelelike defect in sheep. METHODS: A myelomeningocele-like defect (laminectomy and dural excision) was created in the lumbar region on day 90 of gestation in 9 pregnant sheep. Correction technique was randomized. In Group 1 the defect was corrected using the classic neurosurgical technique of three-layer suture (dura mater, muscle and skin closure) performed by a neurosurgeon. In Group 2, a fetal medicine specialist used a biosynthetic cellulose patch to protect the spinal cord and only the skin was sutured above it. Near term (day 132 of gestation) fetuses were sacrificed for pathological analysis. RESULTS: There were two miscarriages and one maternal death. In total, six cases were available for pathological analysis, three in each group. In Group 1, there were adherence of the spinal cord to the scar (meningo-neural adhesion) and spinal cord architecture loss with posterior funiculus destruction and no visualization of grey matter. In Group 2, we observed in all cases formation of a neo-dura mater, separating the nervous tissue from adjacent muscles, and preserving the posterior funiculus and grey matter. CONCLUSION: The new simplified technique was better than the classic neurosurgical technique. It preserved the nervous tissue and prevented the adherence of the spinal cord to the scar. This suggests the current technique used for the correction of spina bifida in humans may need to be reassessed.

O futebol apresenta uma séria de situações geradoras de estresse. O momento pré-competitivo é altamente gerador de estresse. O alongamento pode contribuir para diminuir estes efeitos negativos do estresse. O objetivo deste estudo foi verificar os efeitos do alongamento no estresse pré-competitivo de jogadores de futebol. Participaram deste estudo 17 atletas juvenis, distribuídos em dois grupos: Grupo Controle (GC) e Grupo Alongamento (GA). Todos responderam ao TEF e, em seguida, o GA praticou 30 minutos de alongamento enquanto o GC vivenciou as situações habituais sem realização de atividade física. Logo após, os dois grupos responderam novamente ao TEF, sendo invertidos os grupos na partida seguinte. As situações “Dormir mal a noite anterior ao jogo” e “Ser prejudicado pelos juizes” foram avaliadas com respostas mais negativas enquanto “Cobrança de si mesmo para ganhar” e “O time adversário é o favorito” foram avaliadas como respostas mais positivas. O GA obteve aumento no número de avaliações positivas comparado ao GC, diminuição no nulo e diminuição na resposta -1 e -3, enquanto o GC obteve diminuição no valor -2. Conclui-se que a situação pré-competitiva modifica negativamente a avaliação dos atletas enquanto o alongamento interfere positivamente frente à avaliação das situações estressoras em jogadores juvenis.

Soccer show a series of stress-generated situations. The streatching may contribut to decrease this stress negative effects. The pre-competitive moment is stress generator. Yhe objetive of this study was to verify the effects of strectch on pre-competitive stress in soccer players. To this, 17 juvenile soccer players were allocated into two groups: control group (CG) and strecth group (SG). All participants responsed to TEF (Stress soccer test) and GA stretching during 30 minutes while GC not realized any physical activity. After this, both groups responded to TEF and was inverted in the next game. The situations “sleep badly on previous night” and “Be damaged for arbitrator” was avaliated with more negative reponses while “ Himself charged to win” and “ The adversary is the favourite” was was avaliated with more positive reponses. SG showed positive avaliations increased copared to CG, decreased on nule response and decreased on -1 and -3 responses. GC showed decrease in -2 responses. Concluded that pre-competitive situation modify negatively the athletes avaliation while the strecth interfere positively in front the avaliation of stressor situations in juvenile players.

Este trabalho analisou se a resposta glial após a lesão da medula espinhal é restrita à formação da cicatriz no local do trauma ou pode também estar relacionada a efeitos tróficos parácrinos em toda a medula espinhal. Hemitransecção da medula espinhal de ratos adultos foi realizada em nível torácico baixo. Após 7 dias e 3 meses as medulas espinhais dos animais foram removidas e submetidas à imuno-histoquímica para a visualização da proteína fibrilar glial ácida GFAP (marcador do astrócito), do OX42 (marcador da microglia), e do fator de crescimento básico derivado do fibroblasto (bFGF), produzido pelo astrócito. Para a quantificação das alterações dos perfis imunoreativos utilizou-se análise de imagem computadorizada. No local da lesão observou-se aumento no número de astrócitos GFAP positivos bem como de células fagocíticas OX42 positivas, caracterizando uma formação cicatricial densa que aumentou aos 3 meses da lesão. A análise morfométrica da área e microdensitométrica da intensidade das imunoreatividades do OX42 e da GFAP mostrou microglia e astrócitos reativos nas substâncias cinzenta e branca em toda a medula espinal 7 dias e 3 meses após cirurgia. Marcação imunofluorescente com 2 cromógenos demonstrou o aumento da expressão do bFGF nos astrócitos reativos. Estes resultados mostram que a ativação glial no local da lesão da medula espinhal é necessária ao processo de cicatrização e reparo do tecido nervoso. Embora a gliose constitua uma barreira à regeneração axonal, a ativação glial em níveis distantes da lesão pode contribuir para o trofismo e plasticidade neuronal favorecendo a manutenção neuronal e o crescimento de fibras nervosas.

This paper analysed whether glial responses following a spinal cord lesion is restricted to a scar formation close to the wound or they might be also related to widespread paracrine trophic events in the entire cord. Spinal cord hemitransection was performed in adult rats at the thoracic level. Seven days and three months later the spinal cords were removed and submitted to immunohistochemistry of glial fibrillary acidic protein (GFAP) and OX42, markers for astrocytes and microglia, as well as of basic fibroblast growth factor (bFGF), an astroglial neurotrophic factor. Computer assisted image analysis was employed in the quantification of the immunoreactivity changes. At the lesion site an increased number of GFAP positive astrocytes and OX42 positive phagocytic cells characterized a dense scar formation by seven days, which was further augmented after three months. Morphometric analysis of the area and microdensitometric analysis of the intensity of the GFAP and OX42 immunoreactivities showed reactive astrocytes and microglia in the entire spinal cord white and gray matters 7 days and 3 months after surgery. Double immunofluorescence demonstrated increased bFGF immunostaining in reactive astrocytes. The results indicated that glial reaction close to an injury site of the spinal cord is related to wounding and repair events. Although gliosis constitutes a barrier to axonal regeneration, glial activation far from the lesion may contribute to neuronal trophism and plasticity in the lesioned spinal cord favoring neuronal maintenance and fiber outgrowth.
