Spontaneous and experimental poisoning by Froelichia humboldtiana in cattle

RESUMO: Os objetivos deste trabalho foram descrever os aspectos epidemiológicos, clínicos e patológicos de dois surtos de intoxicação por Froelichia humboldtiana em bovinos em Pernambuco e reproduzir experimentalmente essa intoxicação em bovinos. Intoxicações espontâneas foram observadas após o início do período chuvoso nos municípios de Cachoerinha e São Caetano. Vinte e dois bovinos apresentaram sinais clínicos e lesões cutâneas compatíveis com fotossensibilização primária, dentre os quais, dois bezerros lactentes. Todos os bovinos se recuperaram totalmente cerca de 21 dias após serem retirados da pastagem. Para reproduzir experimentalmente a intoxicação, três vacas, uma delas com bezerro ao pé, foram mantidas em um piquete de 1ha composto por F. humboldtiana por 14 dias consecutivos. O quadro clínico e as lesões tegumentares, tanto nos bovinos intoxicados nos surtos espontâneos, quanto nos bovinos do experimento consistiram em prurido e hiperemia em áreas despigmentadas de pele, que evoluíam para edema, dermatite exsudativa e necrose de áreas extensas de pele. Em todos os bovinos examinados, os níveis séricos de aspartato aminotransferase (AST), gamaglutamiltransferase (GGT), bilirrubina total, direta e indireta estavam normais. Histologicamente, as lesões consistiram em necrose da epiderme, hiperqueratose com grande quantidade de neutrófilos degenerados e acantose. Na derme havia edema e infiltrado inflamatório composto por eosinófilos, linfócitos e plasmócitos principalmente ao redor dos vasos sanguíneos. Nos bovinos do experimento, sinais clínicos de fotossensibilização foram observados após o terceiro dia de consumo de F. humboldtiana. O bezerro lactente apresentou sinais clínicos leves de fotodermatite no 8º dia do experimento. Estimou-se que o consumo médio de matéria seca de F. humboldtiana necessário para iniciar os sinais clínicos em cada bovino adulto foi de 78kg.
ABSTRACT: The aim of this work was to describe the epidemiological, clinical and pathological aspects of two outbreaks of spontaneous poisoning caused by Froelichia humboldtiana in cattle in Pernambuco, northeastern Brazil and reproduce experimentally this poisoning in cattle. Spontaneous poisonings of primary photosensitization occurred in two farms at the municipalities of Cachoeirinha and São Caetano and affected twenty-two adult bovines and two suckling calves after the rainy season. All bovines have recovered 21 days after they were removed from the pasture. To reproduce experimental poisoning, three cows and a calf were maintained in a pasture with 1ha composed by F. humboldtiana during 14 days. Clinical signs and skin lesions were similar in both spontaneous and experimental poisoning and consisted of cutaneous itching and hyperemia of non-pigmented areas of skin that evolved into edema, exudative dermatitis and extensive areas of skin necrosis. Serum levels of aspartate aminotransferase (AST), gamma glutamyltransferase (GGT), total, direct and indirect bilirubin were normal in all cattle examined. Histologically, lesions consisted of epidermal necrosis, hyperkeratosis with large amounts of degenerate neutrophils and acanthosis. In the dermis, edema and inflammatory infiltrate composed of eosinophils, lymphocytes and plasma cells mainly around the blood vessels were observed. In the experimental group, clinical signs of photosensitization were observed after the third day of F. humboldtiana consumption. The suckling calf displayed mild clinical signs of photodermatitis on the 8th day of the experiment. It was estimated that the average consumption of F. humboldtiana necessary to initiate clinical signs in each adult bovine was 78kg.

Drying kinetics and mass transfer properties in the drying of thistle flower

Resumo A flor de cardo e, em particular, os seus estigmas, são utilizados para coagular o leite na produção de vários queijos tradicionais portugueses, devido à sua elevada atividade de coagulação, proporcionada pelo elevado teor de proteases aspárticas. O objetivo do presente trabalho foi determinar as propriedades de transferência de massa da secagem de flor do cardo em diferentes condições: secagem natural e convectiva. A secagem convectiva decorreu em câmara de convecção a diferentes temperaturas (35 a 65 °C) e o processo foi finalizado quando a amostra apresentava um teor de umidade de cerca de 5% ou inferior. Além disso, adotou-se também o método tradicional de secagem, colocando a flor de cardo em local seco e protegido do sol, deixando-a desidratar à temperatura ambiente variável. O presente trabalho permitiu concluir que a secagem convectiva foi muito mais rápida que a secagem natural e ainda que a velocidade de secagem aumentou com a temperatura. A curva de secagem revelou um período inicial de velocidade constante, seguido de fase de velocidade decrescente. Todos os cinco modelos de camada fina testados para se ajustarem aos dados experimentais revelaram descrever adequadamente a secagem da flor de cardo, no entanto, o modelo que se revelou melhor foi o de Page. A constante de secagem aumentou com a temperatura, assim como a difusidade efetiva e o coeficiente de transferência de massa. Os resultados permitiram estimar a energia de ativação para difusão de umidade (57 kJ/mol) e para a transferência de massa por convecção (78 kJ/mol). Desta forma, este estudo mostrou as possibilidades de projetar processos de secagem eficientes para a flor de cardo utilizada como coagulante do leite na fabricação de queijos tradicionais.
Abstract Thistle flowers, and particularly their stigmas, are used to coagulate milk in the production of a number of traditional Portuguese cheeses due to their high milk-clotting activity provided by the high content of aspartic proteases. The aim of the present work was to determine the mass transfer properties of thistle flower under different drying conditions: natural drying and convective drying. Convective drying took place in a convection chamber set at different temperatures (35 to 65 °C) and the process was terminated when the sample presented a moisture content of about 5% or less. The traditional drying method was also used, placing the thistle flowers in a dry place sheltered from the sun, and leaving them to dehydrate at the variable room temperature. The present work allowed for the conclusion that convective drying was much faster than natural drying, and that the drying rate increased with temperature. The drying curve revealed an initial constant rate period followed by a falling rate. All the five thin layer models tested to fit the experimental data were shown to adequately describe the drying of the thistle flowers, but the best one was the Page model. The drying constant increased with temperature as did the effective diffusivity and the mass transfer coefficient. The results allowed one to estimate the activation energy for moisture diffusion (57 kJ/mol) and for convective mass transfer (78 kJ/mol). Thus this study showed the possibilities for designing efficient drying processes for the thistle flower used for milk-clotting in the manufacture of traditional cheeses.

ABSTRACT The organic extracts from stems, roots and leaves of Tephrosia egregia Sandwith, Fabaceae, provided a new flavone, 5-hydroxy-8-(1",2"-epoxy-3"-hydroxy-3"-methylbutyl)-7-methoxyflavone (1), in addition to eleven known compounds: pongaflavone (2), praecansone B (3), 12a-hydroxyrotenone (4), praecansone A, 2',6'-dimethoxy-4',5'-(2",2"-dimethyl)-pyranochalcone, pongachalcone, maackiain, β-sistosterol and its glucoside, p-cumaric acid and cinnamic acid. The structures of all compounds were established on the basis of spectroscopic methods, mainly 1D and 2D NMR and HRESIMS, involving comparison with literature data. Cytotoxicity of compounds 1–4 was evaluated against AGP-01 (cancerous ascitic fluid), HCT-116 (colon adenocarcinoma), HL-60 (leukemia), PC-3 (prostate carcinoma), SF-295 (glioblastoma) and SKMEL 28 (melanoma) cell lines.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

A alimentação de peixes e os padrões ecomorfológicos estão relacionados com vários fatores como ontogenia. O presente trabalho foi realizado a fim de testar a hipótese de que a dieta e os padrões ecomorfológicos apresentam correlações positivas entre os diferentes estágios de ontogenia de Plagioscion squamosissimus (Heckel, 1840), pois à medida que a espécie passa da fase jovem para adulta altera a sua alimentação e morfologia. A pescada foi escolhida por ser a espécie mais abundante no reservatório da Estação Ecológica de Tapacurá, Estado de Pernambuco. Os padrões ecomorfológicos foram avaliados e relacionados com a alimentação dos indivíduos capturados no reservatório no mês de abril de 2013. As medidas morfométricas foram retiradas de todos os indivíduos capturados e posteriormente recolhidos seus estômagos. Os peixes foram separados de acordo com a fase de desenvolvimento, juvenis e adultos. Também foram tomados seu peso total, comprimento padrão e aferidas onze medidas lineares, utilizadas para obter os índices que representam os atributos ecomorfológicos. Os itens alimentares foram identificados e separados até menor nível taxonômico possível, sendo o mais frequente o camarão Macrobrachium amazonicum (Heller, 1862). Foi possível observar que nos juvenis os itens ingeridos apresentavam maior amplitude no tamanho em comparação com os adultos. Dos dados morfométricos observa-se que os valores de largura da cabeça são maiores nos juvenis e os valores da altura do corpo são mais elevados nos adultos, sendo estes fatores importantes para determinar o tamanho da presa ingerida pelo peixe. Conclui-se que a espécie Plagioscion squamosissimus mostrou relação positiva entre a morfologia e alimentação ao longo do desenvolvimento ontogenético evidenciado pela variação de comprimento dos itens ingeridos entre juvenis e adultos, porém não houve distinção em relação ao alimento ingerido entre eles.
The fish feeding and the ecomorphological patterns are related to several factors such as ontogeny. The present report was realized to test the hypothesis that the diet and the ecomorphological patterns present positive correlations between the different ontogeny stages of Plagioscion squamosissimus (Heckel, 1840) because as the species pass the young to adult phase to alter their diet and morphology. The croaker was chosen because is the more abundant specie in reservoir of Tapacurá Ecologic Station, state of Pernambuco. The ecomorphological patterns were valued and related with the feeding of individuals captured in a reservoir in April 2013. The morphometric measures were taken of all captured individuals and after were collected their stomachs. Fish were separated according to development stage, in juveniles and adults. Also total weight and their standard length were measured and eleven linear measurements were taken, used to obtain the indices that represent ecomorphological attributes. The food items were identified and separated until the smaller taxonomic level, been the most abundant the shrimp Macrobrachium amazonicum (Heller, 1862). It was observed that in the juveniles the items ingest presented wide amplitude size than adults. Through morphometric data it was observed that the head width values are bigger in juveniles and the body weight values are bigger in adults, being this important factors to determine the prey size ingested by the fish. It is concluded that the specie Plagioscion squamosissimus showed the relationship positive between morphology and feeding along the ontogenetic development evidenced by the variation in length of the ingest items between juveniles and adults, but there was no distinction in relation to ingest food between them.

O objetivo deste trabalho foi reproduzir a intoxicação por Ipomoea verbascoidea em caprinos e descrever os aspectos epidemiológicos, clínicos e histopatológicos da intoxicação espontânea por essa planta no Estado de Pernambuco. Para isso, realizou-se o acompanhamento da epidemiologia da doença em sete municípios do semiárido pernambucano. Três caprinos espontaneamente intoxicados foram examinados e, em seguida eutanasiados e necropsiados (Grupo I). Para reproduzir experimentalmente a doença, as folhas secas de I. verbascoidea contendo 0,02% de swainsonina, foram fornecidas na dose de 4g/kg (0,8mg de swainsonina/kg) a dois grupos de três animais. Os caprinos do Grupo II receberam a planta diariamente por 40 dias e foram eutanasiados no 41º dia de experimento. Os caprinos do Grupo III receberam a planta diariamente por 55 dias e foram eutanasiados no 120º dia de experimento. Outros três caprinos constituíram o grupo controle (Grupo IV). Nos grupos experimentais, as lesões encefálicas foram avaliadas por histopatologia e adicionalmente avaliaram-se as lesões cerebelares por morfometria, mediante mensuração da espessura da camada molecular, do número de neurônios de Purkinje e da área dos corpos celulares dessas células. Os principais sinais clínicos e lesões microscópicas foram semelhantes aos previamente reportados em animais intoxicados por plantas que contem swainsonina. Nos caprinos do GII e GIII, os primeiros sinais clínicos foram observados entre o 22º e 29º dia de experimento; clinicamente a doença desenvolvida por esses animais foi semelhante aos casos espontâneos. Nenhum dos caprinos do GIII se recuperou dos sinais neurológicos. Esse resultado evidencia que o consumo da planta por 26-28 dias após a observação dos primeiros sinais clínicos é suficiente para provocar lesões irreversíveis. Pela análise morfométrica, a camada molecular do cerebelo dos caprinos do Grupo I e III eram mais delgadas que às dos caprinos do grupo controle, e os neurônios de Purkinje estavam atróficos. Sugere-se que essas alterações sejam responsáveis pelo quadro clínico neurológico observado nos caprinos que deixam de ingerir a planta e apresentam seqüelas da intoxicação.
The aim of this paper was to reproduce the poisoning of Ipomoea verbascoidea in goats and describe the epidemiological, clinical and pathological aspects of spontaneous poisoning by this plant in Pernambuco. For this, we studied the epidemiology of the disease in seven municipalities in the semiarid region of the State. Three spontaneously poisoned goats were examined and then euthanized and necropsied (Group I). To reproduce the disease, the dried leaves of I. verbascoidea containing 0.02% swainsonine were supplied at doses of 4g/kg (0.8mg swainsonine/kg) to two groups of three animals. The goats in Group II received daily doses of the plant during 40 days and were euthanized on the 41st day of the experiment. Goats from Group III received daily doses of the plant during 55 days and were euthanized on the 120th day of the experiment. Other three goats constituted the control group (Group IV). In experimental groups, the brain lesions were evaluated by histopathology; additionally the cerebellar lesions were evaluated by morphometry, by measuring the molecular layer thickness, the number of Purkinje cells and the area of the cell bodies of these cells. The main clinical signs and microscopic lesions in goats poisoned were similar to those reported by swainsonine containing plants. In goats of GII and GIII, the first nervous signs were observed between 22th and 29th days; clinically, the disease developed by these animals was similar to the spontaneous cases. The goats of GIII did not recover from the neurologic signs. These results show that the consumption of the plant by 26-28 days after observation of the first clinical signs is enough to cause irreversible damage. By morphometric analysis, the molecular layer of the cerebellum of the goats of Group I and III were thinner than those of goats in the control group, and Purkinje neurons were atrophic. It is suggested that these changes are responsible for the neurological picture observed in goats that stop eating the plant and have sequelae of poisoning.

Os microrganismos patogênicos multirresistentes apresentam-se como grandes responsáveis por milhões de mortes em todo o mundo, principalmente Pseudomonas aeruginosa e Staphylococcus aureus, responsáveis por grande parte das infecções hospitalares. A preocupação com estas espécies faz com que novas pesquisas busquem alternativas para controlar estes microrganismos de uma forma mais eficiente e também mais econômica. Os extratos fitoterápicos são alternativas promissoras para este fim, visto que são uma imensa fonte de compostos de ação biológica. O objetivo do trabalho foi a elucidação da atividade antimicrobiana do extrato de Lafoensia pacari A. St.-Hil., Lythraceae, frente a linhagens de bactérias multirresistentes (P. aeruginosa, S. aureus) isoladas de pacientes com múltiplas infecções internados na Unidade de Emergência de Maceió. Os testes de atividade antibacteriana foram avaliados pelo método de difusão em meio sólido (Kirby-Bauer) modificado. De acordo com os ensaios in vitro, foi constatado que 96,4% das linhagens de bactérias utilizadas na pesquisa apresentaram-se sensíveis ao extrato da folha da planta, demonstrando atividade antibacteriana. Halos de inibição de crescimento de até 26 mm foram encontrados. Dessa forma, conclui-se que o extrato de Lafoensia pacari apresenta possibilidades de se encontrar substâncias úteis no combate a bactérias multirresistentes.
Multiresistant pathogenic microorganisms are responsible for million of death all the world, mainly Pseudomonas aeruginosa and Staphylococcus aureus that are responsible for great part of hospital infections. The concern with this species does new researches to find out alternatives to control these microorganisms in the way more efficient and more economic. The phytoterapic extracts are promissory alternatives for that purpose because they are an immense source of biological action. The objective of this study was to evaluate the antimicrobial activity of Lafoensia pacari A. St.-Hil., Lythraceae, extract on multiresistant bacterial strains (P. aeruginosa, S. aureus) that have been isolated from patients with multiple infections occupying an Emergency Unit from Maceió in Brazil. The antibacterial activity experiments were evaluated by agar diffusion tests. Agreeable us experiments in vitro, ascertain that 96,4% of bacterial strains utilized in this research have showed susceptible to the plant's leaf extract, it means an excellent antibacterial activity. Halos of bacterial inhibition until 26 mm were observed. Thus, it can be concluded that the Lafoensia pacari extract has showed as an excellent product to combat multiresistant bacterial.

OBJETIVO: estimar a validade da inspeção visual após aplicação de ácido acético (IVA) no rastreamento das neoplasias intra-epiteliais cervicais (NIC) e lesões induzidas por HPV, comparando seu desempenho com o da colpocitologia e da colposcopia. MÉTODOS: estudo de validação de teste diagnóstico realizado em 893 mulheres de 18 a 65 anos, rastreadas simultaneamente com colpocitologia, IVA e colposcopia, em unidade de saúde pública de Recife, PE. A IVA foi realizada por embrocação do colo com ácido acético a 5% e observação a olho nu, com auxílio de foco clínico comum. Considerou-se como positividade o achado de qualquer lesão aceto-branca no colo. O padrão-ouro foi o histopatológico de biópsia cervical, realizado sempre que qualquer um dos três testes resultasse anormal. Foram estimados e comparados os indicadores de validade de cada teste, com os respectivos intervalos de confiança a 95%. A concordância entre os resultados dos testes foi avaliada pelo coeficiente kappa (k). RESULTADOS: das 303 mulheres biopsiadas, o estudo histopatológico foi anormal em 24. Deste total, a IVA foi positiva em 22, conferindo-lhe sensibilidade estimada de 91,7%, especificidade de 68,9%, valor preditivo positivo de 7,5% e valor preditivo negativo de 99,7%. Comparando-se os intervalos de confiança a 95%, a IVA mostrou maior sensibilidade que a colpocitologia, mas com menores especificidade e valor preditivo positivo. Houve fraca concordância entre os resultados da IVA e da colpocitologia (k=0,02) e excelente concordância com os da colposcopia (k=0,93). CONCLUSÃO: a IVA foi muito mais sensível que a colpocitologia no rastreamento das NIC e lesões HPV-induzidas e teve o mesmo desempenho da colposcopia. Sua baixa especificidade foi responsável por um elevado número de resultados falso-positivos.
PURPOSE: to estimate the validity of visual inspection of cervical intraepithelial neoplasia (CIN) and HPV-induced lesion screening, after acetic acid application (VIA), and to compare its performance with that of colpocytology and colposcopy. METHODS: a diagnostic test validation study involving 893 women aged 18 to 65 years, simultaneously screened with colpocytology, VIA and colposcopy was carried out at a public health unit in Recife, PE. VIA was performed by applying 5% acetic acid onto the cervix and observing it with the help of a clinical spotlight. The finding of any aceto-white lesion on the cervix was considered positive. The gold standard was the histopathology of cervical biopsy, carried out whenever any of the three test results was abnormal. Validity indicators were estimated for each test, within 95% confidence intervals. The analysis of agreement between test results was done by the kappa coefficient. RESULTS: of 303 women submitted to biopsy, the histopathological study was abnormal in 24. Among this total, VIA was positive in 22, yielding an estimated 91.7% sensibility, 68.9% specificity, and 7.5% positive predictive value and 99.7% negative predictive value. Comparing 95% confidence intervals, VIA was more sensitive than colpocytology, despite a lower specificity and positive predictive value. There was poor agreement between VIA and colpocytology (k=0.02) and excellent agreement with colposcopy (k=0.93). CONCLUSION: VIA was much more sensitive than colpocytology in the screening of CIN and HPV-induced lesions and presented a performance similar to colposcopy. Its low specificity determined a high number of false-positive results.
