ABSTRACT The objectives were to evaluate the effects of monensin and virginiamycin, alone or combined, on supplemented Nellore cattle grazing tropical grass during the rainy season. Two experiments were conducted simultaneously to evaluate intake, digestibility, CH4 emissions, blood parameters, performance, and carcass characteristics (Exp. 1), and ruminal fermentation and relative abundance of ruminal microorganisms (Exp. 2). Animals (n = 92 Exp. 1 and n = 12 Exp. 2) were distributed in a completely randomized design and allocated in twelve paddocks composed of Urochloa brizantha (A. Rich.) Stapf. cv. Xaraés. A protein-energetic supplementation of 3 g/kg of BW per day was provided to all animals. Supplements were: without additives (WA), monensin alone at 80 mg/kg of product (MN), virginiamycin alone at 150 mg/kg of product (VM), and monensin (80 mg/kg of product) combined with virginiamycin (150 mg/kg of product; MNVM). Treatments did not affect intakes of total dry matter (DM), supplement DM, and nutrients. However, the intakes of forage DM and crude protein decreased in cattle fed MNVM compared with animals fed WA, MN, and VM. Total volatile fatty acids increased in animals fed VM. Ruminal NH3-N decreased, and pH increased in animals fed MN, VM, and MNVM. Relative abundance of total F. succinogenes and S. ruminantium decreased and R. flavefaciens increased in animals fed MN and VM at d 118. Treatments had no effect on enteric CH4 emissions. The average daily gain (ADG) and total gain were greater in cattle fed MNVM than in cattle fed MN. Combination of monensin and virginiamycin altered the rumen microbial populations but did not decrease enteric CH4 emissions. However, it decreased forage dry matter intake without altering the ADG and total weight gain, leading to an increase in feed efficiency. Results from this study indicate an advantage in including feed additives combined in the diet of supplemented Nellore cattle grazing tropical grass during the rainy season.

Resumo As sempre-vivas são plantas que mantêm o aspecto vivo de suas estruturas mesmo após serem coletadas, e por esta característica são comercializadas como ornamentais de alto valor econômico; contudo, devido à coleta extrativista de suas inflorescências algumas espécies se encontram em perigo de extinção. O gênero Comanthera compreende as sempre-vivas de maior importância econômica para o Brasil. Estudos apontam a cultura de tecidos como estratégia para a propagação e conservação in vitro de espécies deste gênero; entretanto as pesquisas publicadas ainda são incipientes. Diante disso, o objetivo desta revisão foi reunir publicações sobre o cultivo in vitro do gênero Comanthera com o intuito de fundamentar estudos futuros com suas espécies. Para tanto, esta revisão está estruturada em dois tópicos principais: micropropagação e conservação in vitro.

Abstract The term “sempre-viva” denotes plants whose structures retain their natural form and color after being cut and dried. For these reasons, they are commercially valuable for ornamental purposes. However, due to extractive overexploitation of their inflorescences, some of these species are considered endangered. The genus Comanthera includes the sempre-vivas species with greatest economic importance in Brazil. Previous studies have shown that tissue culture is a workable strategy for in vitro propagation and conservation of species of this genus. However, these studies are still incipient. Therefore, the objective of this review is to summarize the findings on the in vitro cultivation of species of the Comanthera genus, to serve as the basis for future research. The text is structured in two main topics: micropropagation and in vitro conservation.

Resumo Este trabalho teve o objetivo de avaliar a resposta morfogênica da micropropagação de Aechmea miniata e Aechmea blanchetiana após diferentes tratamentos com citocinina e auxinas. O estabelecimento in vitro se deu a partir de sementes de plantas adultas, com aproximadamente 2 anos de idade. Após descontaminação as sementes foram inoculadas em meio MS. Após 30 dias da germinação, foram isolados segmentos caulinares com aproximadamente 5 mm, e em seguida, foram transferidos para meio MS suplementado com BAP nas concentrações 4,44; 8,88 ou 13,32 µM. Ao final de 225 dias de cultivo, foi avaliado o número de brotos / explante, altura de brotações, porcentagem de enraizamento, número de raízes e comprimento de raízes. Verificou-se que, para as duas espécies, a concentração 4,44 µM de BAP mostrou maior eficiência para a multiplicação. Para a etapa de enraizamento, os brotos obtidos in vitro foram inoculados em meio MS com metade da concentração salina de macronutrientes, acrescido com 1, 2 e 3 µM de AIB ou ANA. Aos 60 dias, verificou-se que as duas espécies apresentaram 100% de enraizamento em meio de cultura sem a adição de reguladores de crescimento. Após enraizamento as plantas foram transferidas para substrato, e aos 180 dias de cultivo foi verificado sobrevivência superior a 80%, independente da espécie.

Abstract The objective of this study was to evaluate the morphogenic response of Aechmea miniata and Aechmea blanchetiana after different treatments with cytokinins and auxins during micropropagation. In vitro cultures were established from seeds of adult plants, approximately 2 years old. After sterilization, the seeds were inoculated in MS médium. 30 days after germination, caulin segments of approximately 5 mm were isolated, then, transferred to MS medium supplemented with BAP at concentrations of 4.44, 8.88, and 13.32 µM. At the end of 225 days of culture, the number of buds/explants, height of shoots, percentage of rooting, number of roots, and length of roots were evaluated. It was found that 4.44 µM BAP induced relatively higher frequency of multiplication in both species. For rooting, the shoots obtained in vitro were inoculated in MS medium with half of the concentration of macronutrient salts, and 1, 2, and 3 µM AIB or ANA. At 60 days, the two species exhibited 100% rooting in the médium without the addition of growth regulators. After rooting the plants were transferred to substrate, and at 180 days of culture, the survival rate was more than 80%, independent of the species.

RESUMO: Sincoraea mucugensis (Wand. & A.A. Conc.) LOUZADA & WAND é uma bromélia vulnerável de ocorrência restrita ao município de Mucugê, Chapada Diamantina. Estudos indicam que a cultura de tecidos é uma alternativa viável para a propagação in vitro desta espécie. Contudo, em função das baixas taxas de multiplicação obtidas em estudos anteriores, objetivou-se estabelecer um protocolo de micropropagação via organogênese direta para S. mucugensis. Explantes caulinares foram inoculados em meio de cultura MS ½ contendo diferentes concentrações de BAP (0,00; 6,66; 8,88; 11,10; 13,20 µM) e ANA (0,00; 2,60; 5,20 µM). Os brotos obtidos foram submetidos a diferentes períodos de enraizamento (30 e 60 dias) com carvão ativado; posteriormente as microplantas foram aclimatizadas em casa de vegetação, testando-se o efeito da cobertura com copos plásticos transparentes como estratégias contra perda de água. Os dados foram submetidos à análise de variância (ANOVA) e as médias analisadas por regressão ou comparadas pelo Teste de Tukey. Os resultados demonstram que altas taxas de multiplicação de S. mucugensis são geradas a partir de explantes caulinares cultivados em meio contendo ANA (6,42 a 7,43 brotos/explantes); o enraizamento dos brotos é realizado por 30 dias em meio com carvão ativado e a aclimatização é feita com exposição direta das microplantas ao ambiente ex vitro com 95% de sobrevivência.

ABSTRACT: Sincoraea mucugensis (Wand. & A.A. Conc.) LOUZADA & WAND, an endangered bromeliad, is confined to the central region of the Chapada Diamantina, in the municipality of Mucugê, Brazil. From various researches, it is evident that for the propagation of this species, the in vitro technique is a feasible option. However, due to the low multiplication rates reported in various papers, this study aimed to establish a micropropagation protocol of direct organogenesis for S. mucugensis. First, the inoculation of the stem explants was done in MS ½ culture medium which contained different levels of BAP (0.00; 6.66; 8.88; 11.10; 13.20 µM) and NAA (0.00; 2.60; 5.20 µM). These shoots were then subjected to a couple of distinct rooting periods (of 30- and 60-day duration) using activated charcoal; finally, these microplants were transferred to a greenhouse for acclimatization, and covered with transparent plastic cups, as a water loss prevention test method. All the data were submitted to the analysis of variance (ANOVA), and the means were subjected to regression analysis or compared using the Tukey test. The findings revealed that the S. mucugensis stem explants raised in the NAA-rich medium (6.42 to 7.43 shoots/explants) showed high multiplication rates; the shoot rooting was done for 30 days using activated charcoal with the medium. Acclimatization, which was performed by directly exposing the microplants to the ex vitro environment, showed 95% survival rate.

RESUMO Physalis peruviana L. (Solanaceae) é uma frutífera herbácea, que vem se consolidando no mercado em virtude de suas potencialidades nutricionais e medicinais. As limitações à sua produção são o ciclo reprodutivo curto, o acometimento dos frutos por pragas e a carência de estudos, o que justifica a busca de estratégias para a sua propagação. Objetivou-se avaliar os efeitos de 6-benzilaminopurina (BAP) e explantes no potencial morfogênico de P. peruviana, bem como estabelecer um protocolo para a micropropagação da espécie via organogênese direta. Para avaliar a morfogênese, explantes (nó cotiledonar, cotilédone, folha, epicótilo, hipocótilo e raiz) foram inoculados em meio de cultura Murashige & Skoog com metade das concentrações salinas e suplementado com a citocinina BAP (0,00 µM; 2,22 µM; 4,44 µM; 6,66 µM; ou 8,88 µM), acrescido de 30 g L-1 de sacarose e 7 g L-1 de ágar. Visando à produção de brotos via direta, o explante nó cotiledonar foi submetido a 0,00 µM; 2,22 µM; 4,44 µM; 6,66 µM; 8,88 µM; 13,32 µM; 17,76 µM; ou 22,20 µM de BAP. Os brotos obtidos foram submetidos ao enraizamento em meios com e sem adição de carvão ativado e, posteriormente, transferidos para aclimatização. Verificou-se que o nó cotiledonar e a folha foram as fontes de explantes mais eficientes para a regeneração dos brotos via organogênese direta e indireta, respectivamente. Os resultados mais significativos para a produção de brotos via direta foram obtidos com 12,50 µM de BAP. Estes brotos foram enraizados in vitro em meio isento de carvão, e as microplantas aclimatizadas em terra vegetal alcançaram 100 % de sobrevivência aos 90 dias da aclimatização.

ABSTRACT Physalis peruviana L. (Solanaceae) is an herbaceous fruit-bearing species that has been gaining market acceptance due to its nutritional and medicinal potential. The main limitations to its cultivation are the short reproductive cycle, the susceptibility of the fruits to pests and the lack of information about the crop management. Hence, studies are necessary to develop strategies for its propagation. This study aimed to evaluate the effects of 6-benzylaminopurine (BAP) and explants on the morphogenetic potential of P. peruviana, as well as to establish a protocol for the micropropagation of the species via direct organogenesis. To evaluate the morphogenesis, cotyledonary node, cotyledon, leaf, epicotyl, hypocotyl and root explants were inoculated in Murashige & Skoog culture medium with half the normal concentration of salts and supplemented with cytokinin BAP (0.00 µM, 2.22 µM, 4.44 µM, 6.66 µM or 8.88 µM), plus 30 g L-1 of sucrose and 7 g L-1 of agar. Aiming at a direct production of shoots, the cotyledonary node explant was submitted to 0.00 µM, 2.22 µM, 4.44 µM, 6.66 µM, 8.88 µM, 13.32 µM, 17.76 µM or 22.20 µM of BAP. The obtained shoots were tested regarding their rooting potential in media with and without the addition of activated charcoal and then were transferred for acclimatation. The cotyledonary node and leaf explants were the most efficient sources for the regeneration of shoots via direct and indirect organogenesis, respectively. The most significant results for direct shoot production were obtained with 12.50 µM of BAP. These shoots were successfully rooted in vitro in medium without activated charcoal, and the microplants acclimated in vegetable earth attained 100 % of survival after 90 days of acclimatation.

RESUMO: Este trabalho teve como objetivo avaliar a germinação, o crescimento inicial e o co-cultivo in vitro de Comanthera curralensis Moldenke, uma "sempre viva" nativa da Chapada Diamantina-BA. Para germinação e crescimento inicial, foram testados os meios de cultura MS completo e MS1/2 suplementados com duas concentrações de sacarose (15 e 30gL-1); no cultivo in vitro, foram testadas três quantidades de plantas por recipiente (8,10 e 12). A utilização do meio MS1/2 com 15gL-1 de sacarose proporcionou maiores porcentagem de germinação crescimento das plantas no estabelecimento in vitro de C. curralensis , e o uso de 12 plantas por recipiente é indicado para a redução de custos na produção in vitro da espécie.

ABSTRACT: The goal of the present study was to evaluate the germination, initial growth, and in vitro co-cultivation of Comanthera curralensis Moldenke, a "sempre viva" native of the Chapada Diamantina state of Bahia. Full strength (MS) and half-strength MS (MS1/2) growth media supplemented with two different sucrose concentrations (15 and 30g L-1) were tested for germination and initial plant growth. Three different plant densities were tested by in vitro culture (8, 10 and 12 plants per container). MS1/2 medium with 15g L-1 sucrose resulted in a higher percentage of germination and plant growth for the in vitro establishment of C. curralensis. The use of 12 plants per container is indicated for cost reduction in C. curralensis in vitro production.

O objetivo do estudo foi determinar se o anel de tensão capsular (ATC), isoladamente ou associado à implantação de uma lente intraocular, poderia diminuir a ocorrência de opacificação da cápsula posterior (OCP) após a cirurgia de catarata por facoemulsificação em cães. Para tanto, cães portadores de catarata imatura ou madura foram avaliados, perfazendo um total de vinte olhos. Em todos os animais, foram excluídas quaisquer doenças oftálmicas e sistêmicas concomitantes. Os olhos foram selecionados aleatoriamente para receber o anel (ATC) isolado ou em associação com a lente. Flaremetria a laser e avaliações clínicas foram realizadas aos 2, 7, 14, 21, e 28 dias de pós-operatório. Fotografias foram tomadas aos 28 e a seguir a cada 30 dias, até os 180 dias. Os resultados mostraram inflamação mais importante nos olhos com IOL, embora, entre os parâmetros avaliados, apenas nas sinéquias houve diferença estatisticamente significativa. No entanto, em ambas as técnicas, quase nenhuma OCP ocorreu até o momento final das avaliações. Tais resultados sugerem que os anéis de tensão capsular (ATC) podem ser úteis na prevenção da opacidade capsular pós-operatória, sem complicações decorrentes

The purpose of the study was to determine whether capsular tension ring (CTR) alone or associated with the implantation of an intraocular lens (IOL) could decrease the occurrence of posterior capsular opacification (PCO) after phacoemulsification cataract surgery in dogs. Twenty eyes of client-owned dogs with immature or mature cataracts were included in this study. In all the animals, any ophthalmic or systemic concurrent disease was excluded. The eyes were randomly selected to receive CTR alone or in association with the IOL. Laser flaremetry and clinical evaluations were conducted on post-operative days 2, 7, 14, 21 and 28. Fundus photographs were taken at the 28th and then every 30 days, up to 180 days. The results showed more inflammatory response on eyes with IOLs, although among the parameters evaluated, solely the synechiae showed significant statistical differences. Nevertheless, in both techniques almost no PCOs occurred at the end of the evaluations. These results suggest that the CTRs can be useful in the prevention of the post-operative capsular opacities, with minor complications

O experimento foi realizado em 8ha de pasto de Panicum maximum cv. Tanzania, com pastejo rotacionado de 7 dias de ocupação e 21 dias de descanso. Quatro tratamentos foram avaliados: bovinos pastejando isoladamente (BOV), ovinos pastejando isoladamente (OVI), bovinos e ovinos pastejando simultaneamente (SIM), e bovinos pastejando previamente aos ovinos (alternado - ALT). Vinte novilhas e 30 cordeiros Santa Inês foram utilizados. Contagem de ovos nas fezes (FEC) e coproculturas foram realizados. Sangue também foi colhido para examinar a série vermelha e branca, proteínas plasmáticas totais (TPP), albumina e hemoglobina. FEC e índice de patogenicidade estimada de nematoides nos ovinos foram menores no tratamento SIM. A proporção de Haemonchus spp. foi maior nos sistemas isolados de pastejo. Para os ovinos, os sistemas consorciados apresentaram redução na infecção endoparasitária, sendo SIM melhor que ALT. Para os bovinos, nenhuma diferença entre os sistemas de pastejo foi verificado. Entretanto, o pastejo simultâneo (ovinos e bovinos) pode ser uma ferramenta para reduzir a necessidade de tratamentos anti-helmínticos em ovinos.

This experiment was carried out on 8 ha of Panicum maximum cv. Tanzania pastures, with rotational grazing consisting of 7 days of occupation and 21 days of rest. Four treatments were evaluated: cattle grazing alone (BOV), sheep grazing alone (OVI), cattle and sheep grazing simultaneously (SIM) and cattle grazing followed by sheep (alternate - ALT). Twenty heifers and 30 male Santa Inês lambs were used. Fecal egg count (FEC) and fecal cultures were carried out. Blood was also collected to examine red and white cell series, total plasma protein (TPP), albumin and hemoglobin. FEC and estimated nematode pathogenicity index in sheep were lower in the SIM treatment. The Haemonchus spp. proportion was higher in isolated grazing systems. For sheep, mixed grazing was shown to reduce endoparasite infection, and SIM was better than ALT. For cattle, no difference between grazing systems was seen. Therefore, simultaneous grazing (sheep and cattle) may be a tool for reducing the need for anthelmintic treatments in sheep.

A conservação in vitro é uma estratégia de conservação ex situ que garante a manutenção da integridade genética e biológica das espécies. O objetivo deste estudo foi avaliar o efeito de agentes osmóticos e temperatura na conservação in vitro de Syngonanthus mucugensis Giul. subsp. mucugensis. Os brotos foram inoculados em meio de cultura MS ½ contendo 7g L-1 de ágar e suplementado com 60g L-1 de sacarose, e com as concentrações de sacarose 15, 30 e 45g L-1 combinados com 0 e 15g L-1 de sorbitol ou manitol. As culturas foram mantidas em duas temperaturas (18 e 25°C). A porcentagem de sobrevivência das plantas foi avaliada mensalmente e ao final de 180 dias foram analisados o comprimento da parte aérea e da raiz, a porcentagem de folhas verdes, a porcentagem de explantes com brotos, o número de brotos por explante e o comprimento dos brotos. Os agentes osmóticos promoveram um decréscimo no crescimento das plantas, no entanto reduziram a sua viabilidade. Os resultados observados nos experimentos mantidos a 18°C foram significativamente superiores aos encontrados a 25°, para todas as variáveis analisadas. A conservação de S. mucugensis subsp. mucugensis pode ser feita à 18°C em meio de cultura MS ½ contendo 15g L-1 de sacarose, por até 180 dias, sem subcultivo.

The in vitro conservation is an ex situ conservation strategy that ensures the maintenance of genetic and biological integrity of species. The present study evaluated the effects of, osmotic agents and different temperature regimes on the in vitro conservation of Syngonanthus mucugensis Giul. subsp. mucugensis. The shoots were inoculated into half salt strength Murashige and Skoog culture medium (MS ½) containing 7g L-1 of agar. The culture medium was supplemented with 60gL-1 sucrose and with the sucrose concentrations 15, 30 and 45g L-1 combined with 0 and 15g L-1 of sorbytol or mannitol. Two different temperatures were used in these experiments (18 and 25°C). The percentage of plant survival was evaluated monthly and at 180 days were analyzed length of shoot and root, the percentage of green leaves, the percentage of explants with shoots and number of shoots per explants and shoot length. The addition of osmotic agents resulted in decreased growth of the plants and therefore reduced their viability. The averages observed in the experiments undertaken at 18°C were significantly superior to those observed at 25°C for all of the variables analyzed. S. mucugensis subsp. mucugensis can be cultured at 18°C in MS½ culture medium containing 15g L-1 of sucrose, for up to 180 days, without subculturing.

O gênero Calliandra Benth. (Leguminosae) possui 134 espécies exclusivamente neotropicais, das quais 42 ocorrem na Chapada Diamantina, sendo 32 endêmicas da área. O objetivo deste trabalho foi avaliar o efeito da luz e substrato na germinação e desenvolvimento inicial de C. viscidula e C. hygrophila. As sementes foram submetidas a três tratamentos em laboratório: luz, escuro e luz/escuro (12 h/12 h). Em viveiro foram utilizados três substratos: (I) areia; (II) terra vegetal + areia; (III) vermiculita + terra vegetal + areia, mantidos a 100%; e 70% de luminosidade. As avaliações da germinação foram diárias durante 21 dias, utilizando-se os parâmetros: germinabilidade, índice de velocidade de germinação (índice de velocidade de emergência para viveiro), tempo médio de germinação e frequência relativa. Os parâmetros analisados no desenvolvimento inicial foram: comprimentos da parte aérea e raiz, número de folhas, massa seca, relação parte aérea/sistema radicular e razão do peso da folha. C. viscidula e C. hygrophila foram classificadas como fotoblásticas neutras e apresentaram melhor desenvolvimento a 70% de luminosidade nos substratos III e I e II , respectivamente.

The genus Calliandra Benth. (Leguminosae) has 134 exclusively Neotropical species; 42 species are distributed in the Chapada Diamantina and 32 are endemics to this area. The aim of this work was to evaluate the effect of light and growing media on germination and initial seedling development of the Calliandra viscidula and C. hygrophila. Seeds were placed under three different light conditions in laboratory conditions: light, dark, and light/dark (12h/12h). In nursery conditions, seeds were germinated in three different growing medias: (I) sand; (II) soil + sand; (III) vermiculite + soil + sand and they were submitted to 100% and 70% of natural light conditions. Evaluations were daily for 21 days for germinability, speed germination index (speed emergency index to nursery), average germination time and relative frequency. The variables evaluated during initial seedling development were: shoot and root lengths, leaf numbers, dry weight, shoot/root system, and leaf weight ratio. C. viscidula and C. hygrophila are photoblastically neutral and have better results to the 130 days under 70% light condition. The best development was achieved when seeds were grown in growing media III for C. viscidula and I and II for C. hygrophila.
