OBJECTIVES: We designed a cohort study to describe characteristics and outcomes of patients with coronavirus disease (COVID-19) admitted to the intensive care unit (ICU) in the largest public hospital in Sao Paulo, Brazil, as Latin America becomes the epicenter of the pandemic. METHODS: This is the protocol for a study being conducted at an academic hospital in Brazil with 300 adult ICU beds dedicated to COVID-19 patients. We will include adult patients admitted to the ICU with suspected or confirmed COVID-19 during the study period. The main outcome is ICU survival at 28 days. Data will be collected prospectively and retrospectively by trained investigators from the hospital’s electronic medical records, using an electronic data capture tool. We will collect data on demographics, comorbidities, severity of disease, and laboratorial test results at admission. Information on the need for advanced life support and ventilator parameters will be collected during ICU stay. Patients will be followed up for 28 days in the ICU and 60 days in the hospital. We will plot Kaplan-Meier curves to estimate ICU and hospital survival and perform survival analysis using the Cox proportional hazards model to identify the main risk factors for mortality. RESULTS: We expect to include a large sample of patients with COVID-19 admitted to the ICU and to be able to provide data on admission characteristics, use of advanced life support, ICU survival at 28 days, and hospital survival at 60 days. CONCLUSIONS: This study will provide epidemiological data about critically ill patients with COVID-19 in Brazil, which could inform health policy and resource allocation in low- and middle-income countries.

Resumo Objetivo Determinar a frequência de infecções sexualmente transmissíveis (ISTs) em mulheres assintomáticas e a associação destas infecções com a neoplasia intraepitelial cervical (NIC). Métodos Foi realizado um estudo transversal recrutando mulheres atendidas em uma clínica ginecológica geral e em um centro de referência para colposcopia, de outubro de 2014 a outubro de 2015. O grupo de colposcopia consistiu de 71 mulheres, e o grupo de ginecologia geral consistiu de 55 mulheres. Amostras cervicais foram coletadas para citologia cervical e uma reação em cadeia de polimerase (RCP) multiplex em tempo real para detecção do vírus do papiloma humano (HPV) e das ISTs provocadas pelos seguintes micro-organismos: Chlamydia trachomatis, Mycoplasma hominis, Mycoplasma genitalium, Ureaplasma urealyticum e Neisseria gonorrhoeae. Foi realizada uma análise multivariada por regressão logística, considerando-se o nível de significância de 0,05. Resultados A frequência geral de ISTs foi: 46,8% (HPV); 27,8% (C. trachomatis); 28,6% (M. genitalium); 0,8% (M. hominis); 4,8% (U. urealyticum); e 4,8% (N. gonorrhoeae). Os fatores de risco significantes para NIC foram: infecção pelo HPV (razão de probabilidades [RP] = 2,53; p = 0,024); C. trachomatis (RP = 3,04; p = 0,009); M. genitalium (RP = 2,37; p = 0,04); e coinfecção por HPV e C. trachomatis (RP = 3,11; p = 0,023). Após a análise multivariada, foi encontrada uma associação significante entre HPV e NIC (RP = 2.48; intervalo de confiança de 95% [IC95%]: 1,04-5,92; p = 0,04) e entre C. trachomatis e NIC (RP = 2,69; IC95%: 1,11-6,53; p = 0,028). Conclusões A frequência de ISTs foi alta em mulheres assintomáticas. Infecções por HPV e C. trachomatis foram independentemente associadas com a presença de NIC. A alta frequência de ISTs em mulheres assintomáticas sugere a necessidade de rastreamento rotineiro dessas infecções.
Abstract Objective To determine the frequency of sexually transmitted infections (STIs) in asymptomatic women and the association of STIs with cervical intraepithelial neoplasia (CIN). Comparative morphology of the stomach ossicles of four freshwater crabs species of Dilocarcinini (Crustacea, Decapoda, Trichodactylidae)

Abstract The morphology of the foregut ossicles of representatives of four genera of Dilocarcinini were described to assess the usefulness of the ossicles as possible morphological characters in studies on the intrafamilial relationships of Trichodactylidae. Stomachs of specimens of Dilocarcinus pagei Stimpson, 1861, D. septemdentatus (Herbst, 1783), Moreirocarcinus emarginatus (H. Milne Edwards, 1853) and M. laevifrons (Moreira, 1901) were heated for 60 minutes in 10% KOH solution and then were stained with Alizarin Red 1% added into the KOH solution. The foregut ossicles of Dilocarcinini consists of 11 ossicles of the gastric mill, 10 of the lateral supporting cardiac region, 4 of the cardio-pyloric valve, 6 supporting the dorsal pyloric stomach, 9 supporting the ventral pylorus and bulb, 3 supporting the supra-ampullary region, 5 supporting the lateral pylorus region, totaling 48 ossicles and the following 34 are similar in shape, size and degree of calcification. The morphological differences between the majority of the ossicles were not very clear among the species, showing inconsistency intrageneric differences, however, species of different genera showed small similarities between them. There is a clear need for including more taxa in a comparative analysis, combined with external morphology and perhaps even molecular data to properly evaluate the phylogenetic relationships within Trichodactylidae and their systematic position within Brachyura.

Description of ten additional ossicles in the foregut of the freshwater crabs Sylviocarcinus pictus and Valdivia serrata (Decapoda: Trichodactylidae)

ABSTRACT The morphology of stomach ossicles of decapod crustaceans provides valuable information on their phylogeny and biology. We herein described ten new ossicles in the foreguts of two trichodactylid crabs, Sylviocarcinus pictus (H. Milne-Edwards, 1853) and Valdivia serrata White, 1847, in addition to previously described 38 ossicles, which are also recognized and listed. Five specimens each of S. pictus and V. serrata were selected for morphological analysis of gastric ossicles. The stomachs were obtained after removing the carapace, and they were fixed in 10% formalin for 24 hours. After this procedure, the stomachs were immersed in a solution of 10% Potassium Hydroxide (KOH) and heated to 100 °C during 60 minutes for tissue maceration. At this point, the clean skeletons were colored by adding 1% Alizarin Red to the KOH solution in order to facilitate visualization of the internal structures such as the setae and ossicles. The ten new ossicles are: dorsomedial cardiac plate; dorsolateral cardiac plate; suprapectineal lateral ossicle; inferior cardiac valve; lateral mesopyloric ossicle; ampullary roof-medium portion ossicle; process of the ampullary roof-upper portion; lateral-inferior post-ampullary plate; pleuro-pyloric valve’s ossicle; and lateral pleuro-pyloric plate. Some ossicles are thin plates that together with the main ossicles assist in the structure and support of the stomach, which are similar in the two species studied herein. The current knowledge on gastric ossicles will be useful in establishing taxonomic characters, which can evaluate phylogenetic relationships among brachyuran crabs.

Resumo: A citologia é um importante exame complementar utilizado para a detecção de alterações no perfil celular do trato respiratório e auxílio no diagnóstico de doenças. No entanto, como os primeiros meses de vida compõem o período de adaptação à vida extrauterina resultando em possíveis alterações das populações celulares, é essencial a padronização das características comuns aos animais sadios, possibilitando a identificação e análise de qualquer mudança no quadro esperado. O objetivo desta pesquisa foi caracterizar o perfil celular por citologia do lavado broncoalveolar obtido semanalmente por broncoscopia de dez bezerros durante os três primeiros meses de vida. A análise estatística foi realizada utilizando-se o software Minitab® 15.0, empregando-se o teste T pareado para as amostras paramétricas e Mann-Whitney para as amostras não paramétricas, considerando nivel de significância P≤0,05. Os resultados obtidos apontaram que o perfil celular do lavado broncoalveolar de bezerros hígidos durante os primeiros 90 dias de vida apresentou predomínio de macrófagos, com média geral de 63,17%, seguido de 33,69% de neutrófilos. Porém observou-se diminuição significativa na porcentagem de macrófagos (P=0,04) e aumento de neutrófilos (P=0,05) ao longo dos momentos, comprovada pela forte correlação negativa entre as porcentagens de macrófagos e neutrófilos ao longo dos momentos. Não houve diferença entre as porcentagens de células gigantes, células epiteliais, linfócitos, eosinófilos e basófilos durante o experimento. Apesar dos resultados serem influenciados por fatores ambientais e de manejo, os resultados dessa pesquisa fornecem subsídios para a identificação de alterações críticas que descaracterizem o perfil celular de bezerros acometidos por doenças respiratórias.
Abstract: Cytology is an important supplementary test used for the detection of changes in the mobile profile of the respiratory tract and aid in the diagnosis of diseases. However, as the first few months of life make up the period of adjustment to extrauterine life resulting in possible changes in cellular populations, it is essential to the standardization of the characteristics common to healthy animals, enabling the identification and analysis of any change in the expected frame. The objective of this research was to characterize the cellular profile by cytology of bronchoalveolar lavage obtained weekly for bronchoscopy of ten calves during the first three months of life. Statistical analysis was performed using the Minitab® 15.0 software, using the paired T test for parametric samples, and Mann-Whitney for nonparametric samples, considering significance level as P≤0,05. The results obtained showed that the profile of bronchoalveolar lavage of healthy calves during the first 90 days of life showed a predominance of macrophages, with medium average of 63.17%, followed by 33.69% neutrophils. However it was observed significant decrease in percentage of macrophages (P=0.04) and increased neutrophils (P=0,05) over the times, proven by the strong negative correlation between percentages of macrophages and neutrophils along the times. There was no difference between the percentages of giant cells, epithelial cells, lymphocytes, eosinophils and basophils during the experiment. Although the results are influenced by environmental factors and management, the results of this research provide subsidies for the identification of critical changes in cell profile of calves affected by respiratory diseases.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Molecular typing of Clostridium perfringens isolated from swine in slaughterhouses from São Paulo State, Brazil

Clostridium perfringens é uma bactéria Gram positiva anaeróbica, conhecida por infectar os seres humanos, animais domésticos e de vida selvagem. Apesar de as infecções causadas por C. perfringens tipo C e A em suínos serem bastante estudadas, poucos relatos descrevem a relação genética entre as linhagens envolvidas na cadeia epidemiológica da clostridiose suína, bem como a presença do microorganismo em abatedouros. O objetivo do presente estudo foi isolar C. perfringens a partir das fezes e carcaças de suínos no abatedouro, caracterizar os isolados quanto à presença dos genes codificadores de enterotoxina, toxina alfa, beta, épsilon, iota e beta 2 através da PCR e comparar os isolados através da eletroforese em campo pulsado (PFGE). A frequência de isolamento do agente em carcaças e em intestinos de suínos foi de 58,8% para ambos os tipos de amostras. De acordo com a reação em cadeia pela polimerase, somente a toxina alfa foi detectada, sendo todos os isolados negativos para toxina beta2 e enterotoxina. Através da técnica de PFGE, as cepas foram caracterizadas em 35 pulsotipos, sendo que, em apenas um caso, um isolado de amostras de carcaças foi agrupado no mesmo pulsotipo do isolado de fezes do mesmo animal, indicando que a possibilidade de contaminação cruzada no processamento da carcaça foi baixa. Apesar da alta prevalência de C. perfringens em carcaças de suínos provenientes dos abatedouros avaliados, o risco de intoxicação alimentar para os consumidores de carne suína brasileira é baixo, já que todas as cepas foram negativas para o gene cpe, codificador de enterotoxina.
Clostridium perfringens is an anaerobic Gram-positive bacterium known as common pathogen for humans, for domestic and wildlife animals. Although infections caused by C. perfringens type C and A in swine are well studied, just a few reports describe the genetic relationship among strains in the epidemiological chain of swine clostridioses, as well as the presence of the microorganism in the slaughterhouses. The aim of the present study was to isolate C. perfringens from feces and carcasses from swine slaughterhouses, characterize the strains in relation to the presence of enterotoxin, alpha, beta, epsilon, iota and beta-2 toxins genes, using polymerase chain reaction (PCR) and comparing strains by means of Pulsed field gel electrophoresis (PFGE). Clostridium perfringens isolation frequencies in carcasses and finishing pig intestines were of 58.8% in both types of samples. According to the polymerase chain reaction assay, only alfa toxin was detected, being all isolates also negative to enterotoxin and beta2 toxin. Through PFGE technique, the strains were characterized in 35 pulsotypes. In only one pulsotype, the isolate from carcass sample was grouped with fecal isolate of the same animal, suggesting that the risk of cross-contamination was low. Despite the high prevalence of C. perfringens in swine carcasses from the slaughterhouses assessed, the risk of food poisoning to Brazilian pork consumers is low, since all strains were negative to cpe-gene, codifying enterotoxin.
