The current study aimed to assess the effects of the addition of dietary fermented feed (FF) on the growth performance, blood parameters, intestinal morphology, and number of cecal microorganisms of Sichuan white geese from 1 to 28 d of age. A total of 240, 1-day-old female geese were randomly divided into five groups with six replicates (eight birds per replicate). Geese in the control group were fed the basal diet (0.0% FF) and the experimental groups were fed the basal diet supplemented with 2.5%, 5.0%, 7.5%, and 10.0% FF, respectively. The 5.0% FF and 7.5% FF groups had greater body weights (BW), average daily feed intakes (ADFI), and average daily gains (ADG) than the control group. The feed/gain ratio (F/G) in the 7.5% FF group was higher than the control group. The concentrations of immunoglobulin A (IGA), superoxide dismutase (SOD), catalase, total antioxidant capacity (T-AOC), and glutathione peroxidase (GPx) in the 5.0% FF group were higher than those in the control group, whereas malondialdehyde (MDA) content in the 5.0% FF group was lower. Moreover, the FF did not affect the plasma biochemical indicators, number of cecal microorganisms, and intestinal morphology of geese in the groups. These findings indicate that 5.0% FF may improve the growth performance (BW, ADG, and ADFI), immunological function, and antioxidant capacity of Sichuan white geese from 1 to 28 days of age.

Abstract Cobalt metal is considered as an essential trace element for the animals. Present investigation was undertaken in the peri-urban area to analyze the cobalt availability in animal food chain by using different indices. Cow, buffalo and sheep samples along with forage and soil samples were collected from the three different sites of District Jhang and analyzed through atomic absorption spectrophotometer. Cobalt values differed in soil samples as 0.315-0.535 mg/kg, forages as 0.127-0.333 mg/kg and animal samples as 0.364-0.504 mg/kg. Analyzed cobalt concentration in soil, forage and animal samples was found to be deficient in concentration with respect to standard limits. Soil showed the minimum cobalt level in Z. mays while maximum concentration was examined in the forage C. decidua samples. All indices examined in this study has values lesser than 1, representing the safer limits of the cobalt concentration in these samples. Enrichment factor (0.071-0.161 mg/kg) showed the highly deficient amount of cobalt enrichment in this area. Bio-concentration factor (0.392-0.883) and pollution load index (0.035-0.059 mg/kg) values were also lesser than 1 explains that plant and soil samples are not contaminated with cobalt metal. The daily intake and health risk index ranged from 0.00019-0.00064 mg/kg/day and 0.0044-0.0150 mg/kg/day respectively. Among the animals, cobalt availability was maximum (0.0150 mg/kg/day) in the buffaloes that grazed on the C. decidua fodder. Results of this study concluded that cobalt containing fertilizers must be applied on the soil and forages. Animal feed derived from the cobalt containing supplements are supplied to the animals, to fulfill the nutritional requirements of livestock.

Resumo O metal cobalto é considerado um oligoelemento essencial para os animais. A presente investigação foi realizada na área periurbana para analisar a disponibilidade de cobalto na cadeia alimentar animal usando diferentes índices. Amostras de vacas, búfalos e ovelhas, juntamente com amostras de forragem e solo foram coletadas em três locais diferentes do Distrito Jhang e analisadas por meio de espectrofotômetro de absorção atômica. Os valores de cobalto diferiram em amostras de solo como 0,315-0,535mg/kg, forragens como 0,127-0,333 mg/kg e amostras de animais como 0,364-0,504 mg/kg. A concentração de cobalto analisada no solo, forragem e amostras de animais foi considerada deficiente em relação aos limites padrão. O solo apresentou o teor mínimo de cobalto em Z. mays enquanto a concentração máxima foi examinada nas amostras de forragem C. decidua. Todos os índices examinados neste estudo possuem valores menores que 1, representando os limites mais seguros da concentração de cobalto nestas amostras. O fator de enriquecimento (0,071-0,161 mg/kg) mostrou a quantidade altamente deficiente de enriquecimento de cobalto nesta área. Os valores do fator de bioconcentração (0,392-0,883) e do índice de carga de poluição (0,035-0,059 mg/kg) também foram menores que 1, o que explica que as amostras de plantas e solo não estão contaminadas com cobalto metálico. A ingestão diária e o índice de risco à saúde variaram de 0,00019-0,00064 mg/kg/dia e 0,0044-0,0150 mg/kg/dia, respectivamente. Entre os animais, a disponibilidade de cobalto foi máxima (0,0150 mg/kg/dia) nos búfalos que pastaram na forragem de C. decidua. Os resultados deste estudo concluíram que fertilizantes contendo cobalto devem ser aplicados no solo e nas forragens. A ração animal derivada dos suplementos contendo cobalto é fornecida aos animais, para atender às necessidades nutricionais do gado.

RESUMO Porcos tibetanos são caracterizados por diferenças fenotípicas significativas em relação aos porcos de planície. Nosso estudo anterior demonstrou que os genes CRYAB e CTGF eram expressos diferentemente nos tecidos do coração entre os porcos tibetanos (raça das terras altas) e Yorkshire (raça das terras baixas), indicando que eles poderiam participar da adaptação à hipoxia. CRYAB (ɑB-crystallin) e CTGF (fator de crescimento do tecido conjuntivo) também foram relatados como estando associados ao desenvolvimento pulmonar. Entretanto, os padrões de expressão do CRYAB e CTGF nos tecidos pulmonares em diferentes altitudes e sua caracterização genética não são bem compreendidos. Neste estudo, o qRT-PCR e a mancha ocidental de tecido pulmonar revelou níveis de expressão de CRYAB mais elevados em porcos tibetanos e Yorkshire de altitude e média altitude do que em seus pares de planície. Com um aumento na altitude, o nível de expressão do CTGF aumentou nos porcos tibetanos, enquanto diminuiu nos porcos Yorkshire. Além disso, foram identificados dois novos polimorfismos de um único nucleotídeo na região flanqueadora de CRYAB (g.39644482C>T e g.39644132T>C) e CTGF (g.31671748A>G e g.31671773T>G). O polimorfismo pode contribuir parcialmente com as diferenças nos níveis de expressão entre grupos a uma mesma altitude. Estas descobertas proporcionam novos conhecimentos sobre as adaptações de hipoxia a alta altitude dos porcos tibetanos.

ABSTRACT Tibetan pigs are characterized by significant phenotypic differences relative to lowland pigs. Our previous study demonstrated that the genes CRYAB and CTGF were differentially expressed in heart tissues between Tibetan (highland breed) and Yorkshire (lowland breed) pigs, indicating that they might participate in hypoxia adaptation. CRYAB (ɑB-crystallin) and CTGF (connective tissue growth factor) have also been reported to be associated with lung development. However, the expression patterns of CRYAB and CTGF in lung tissues at different altitudes and their genetic characterization are not well understood. In this study, qRT-PCR and western blot of lung tissue revealed higher CRYAB expression levels in highland and middle-highland Tibetan and Yorkshire pigs than in their lowland counterparts. With an increase in altitude, the expression level of CTGF increased in Tibetan pigs, whereas it decreased in Yorkshire pigs. Furthermore, two novel single-nucleotide polymorphism were identified in the 5′ flanking region of CRYAB (g.39644482C>T and g.39644132T>C) and CTGF (g.31671748A>G and g.31671773T>G). The polymorphism may partially contribute to the differences in expression levels between groups at the same altitude. These findings provide novel insights into the high-altitude hypoxia adaptations of Tibetan pigs.

Resumo Dendrobium nobile Lindl. é uma orquídea com importantes valores medicinais. Esta é uma colorida planta doméstica e também uma erva popular na Medicina Tradicional Chinesa (MTC). As variantes desta planta de diferentes regiões geográficas podem ser altas, e neste estudo, nosso objetivo foi desenvolver marcadores de região amplificada de sequência caracterizada (in English, Sequence Characterized Amplified Region (SCAR)) para a identificação de variante específica desta planta. Diferentes cultivares de D. nobile foram coletadas de nove locais diferentes da China e uma cultivar de Mianmar. Materiais de DNA foram extraídos das amostras de plantas, em que a Amplificação Aleatória de DNA Polimórfico (in English, Random Amplified Polymorphism DNA (RAPD)) foi desenvolvida, clonada e sequenciada para o desenvolvimento de marcadores SCAR. Desenvolvemos quatro marcadores SCAR, que são específicos para a cultivar de Luzhou na China e claramente distinguíveis (geneticamente) de outras cultivares. Esses marcadores SCAR estão depositados no GenBank (números de acesso MZ417502, MZ484089, MZ417504 e MZ417505). Quatro marcadores SCAR para D. nobile compreendem técnicas moleculares eficazes para identificar geneticamente as diferentes cultivares ou espécies, e este método é aplicável para caracterização genética e identificação de outras espécies de plantas também.

Abstract Dendrobium nobile Lindl. is an orcid plant with important medicinal values. This is a colourful houseplant, and also a popular herb in traditional Chinese medicine (TCM). The variants of this plant from different geographic regions might be high, and in this study, we aimed to develop specific sequence characterized amplified region (SCAR) markers for the identification of specific variant of this plant. Different cultivars of D. nobile were collected from nine different places of China, and one cultivar from Myanmar. DNA materials were extracted from the plant samples, random amplified polymorphic DNA (RAPD) were developed, cloned and sequenced for the development of SCAR markers. We have developed four SCAR markers, which are specific to the cultivar from Luzhou China, and clearly distinguishable (genetically) from other cultivars. These SCAR markers are deposited in GenBank (accession number MZ417502, MZ484089, MZ417504 and MZ417505). Four SCAR markers for D. nobile are effective molecular technique to genetically identify the different cultivars or species, and this method is applicable for genetic characterization and identification of other plant species too.

To explore the effects of supplementing a basal diet with green sweet sorghum stalks on the gut microbiota in geese, one hundred and twenty 28-day-old geese were divided into two groups. Group 1 was fed a basal diet and group 2 was fed 92% of the same basal diet with green sweet sorghum stalks ad libitum for 42 days. Three male geese from each group were killed at 70 days old, four gastrointestinal tract segments (duodenum, jejunum, ileum, and caecum) were collected, and the hypervariable V4 region of the bacterial 16S rRNA gene was sequenced. There were obvious changes of microbiome in the caecum compared with the other three intestinal segments after green sweet sorghum stalks were added to the diet. Group 2 had significantly higher alpha diversity in the caecum compared with group 1 (P <0.05). The phyla of bacterial communities in the caecum differed. Group 2 had more abundant Bacteroidetes and Firmicutes, but Proteobacteria were more abundant in group 1. At genus level, Bacteroides was more prevalent in group 2, as were Prevotella and YRC22 (P <0.05). Functional analysis revealed that the carbohydrate metabolism, membrane transport, endocrine system and digestive system metabolic pathways were overrepresented in group 2. In conclusion, caecal microbes might play an important role in digesting green sweet sorghum stalks in geese.

Abstract Shallow lowland lakes are critical components of the water cycle, providing an essential service function. However, the impacts of microcystin from phytoplankton communities on benthic macroinvertebrate community diversity and structure have seldom been investigated. During 2008-2012, the impacts of water environmental variables on the diversity of macrobenthic communities, including water temperature, pH, dissolved oxygen, transparency, conductivity, the permanganate index, Chlorophyll a, total phosphorus, total nitrogen, and microcystin-LR (L for leucine and R for arginine), were measured in a typical shallow lowland lake, Ge Lake. The results of the present study demonstrated that there were 31 benthic macroinvertebrate taxa in Ge Lake, including 7 oligochaetes, 7 Mollusca, 14 chironomids, and 3 other taxa. Among the macrobenthic taxa in the benthic community, opportunistic taxa such as Limnodrilus hoffmeisteri are present and can occur at greater densities in disturbed habitats. However, a significant reduction/disappearance of sensitive and clean taxa was observed among the benthic macroinvertebrates. Water temperature, dissolved oxygen, conductivity, ammonia nitrogen, transparency and total phosphorus were the main environmental variables influencing macrobenthic community structure, while water temperature, conductivity, ammonia nitrogen, Chlorophyll a, the permanganate index, total phosphorus and total nitrogen were the main factors that influenced macrobenthic community diversity indices (the numbers of taxa, Shannon’s diversity index, Margalef’s richness index, and Pielou’s evenness index). Our results underscore the severity of the effects of human activity on Ge Lake and strongly suggest that restoring the benthic invertebrate community to previous conditions would require the control and reduction of environmental pollutants and nutrients in Ge Lake.

The objective of the study was to determine the effects of antibiotics, yeast culture (YC), and Lactobacillus culture (LC) on the gut microbiome and metabolites in the serum and caecum of weaning piglets. Twenty-four weaning piglets were divided into four treatment groups: control, antibiotic (1% chlortetracycline), 1.8% yeast culture (YC), and 1.6% Lactobacillus culture groups (LC). Each group had six replicated pens with one pig per pen. Feed and water were available ad libitum. Dietary supplementation with antibiotics, YC and LC increased the abundance of phylum, Firmicutes, and decreased the abundance of phylum, Proteobacteria. Beneficial bacteria such as Lactobacillus and Megasphaera in YC and LC groups increased, whereas the proportion of Shigella was decreased. Genera Alloprevotella and Lachnospira were biomarkers in the control and antibiotic groups, respectively. Phylum, Bacteroidetes, and genus, Collinsella, were biomarkers in the YC group, and Mitsuokella, Anaerotruncus, Syntrophococcus and Sharpea were biomarkers in the LC group. Dietary supplementation with different probiotics changed the serum and caecum metabolite profiles too. Antibiotic supplementation increased the levels of D-mannose, D-glucose, and hexadecanoic acid in the serum, and the levels of myo-inositol, D-mannose and benzenepropanoic acid in the caecum. LC increased the concentrations of D-mannose, cis-9-hexadecenoic acid and heptadecanoic acid in caecum compared with the control group. YC and LC supplementation in the weaning diet could improve the abundance of beneficial bacteria by changing the concentrations of some metabolites in the serum and caecum. Therefore, dietary supplementation with YC or LC could be used as additives instead of antibiotics in weaning piglets.

Resumo Kiwis são uma fruta popular em todo o mundo; No entanto, o crescimento das plantas é ameaçado por estresses abióticos como a seca e as altas temperaturas. O tratamento com niacina em plantas mostrou aumentar os níveis de NADPH, aumentando assim a tolerância a stress abiótico. Aqui, avaliamos o efeito do tratamento com spray de solução de niacina sobre os níveis de NADPH nos cultivares de kiwis Hayward e Xuxiang. Descobrimos que o tratamento por spray com solução de niacina promoveu níveis de NADPH e NADP + e diminuiu a produção de O2·- e os teores de H2O2 nas folhas durante um curto período. Nos frutos, os teores de NADPH aumentaram durante o desenvolvimento precoce, mas diminuíram mais tarde. No entanto, não se observou qualquer efeito nos níveis de NADP + ao longo do desenvolvimento do fruto. Em resumo, este relatório sugere que a niacina pode ser utilizada para aumentar NADPH oxidases, aumentando assim a tolerância ao estresse em kiwis durante o encontro de condições estressantes de curto prazo.

Abstract Kiwifruit are a popular fruit worldwide; however, plant growth is threatened by abiotic stresses such as drought and high temperatures. Niacin treatment in plants has been shown to increase NADPH levels, thus enhancing abiotic stresses tolerance. Here, we evaluate the effect of niacin solution spray treatment on NADPH levels in the kiwifruit cultivars Hayward and Xuxiang. We found that spray treatment with niacin solution promoted NADPH and NADP+ levels and decreased both O2·- production and H2O2 contents in leaves during a short period. In fruit, NADPH contents increased during early development, but decreased later. However, no effect on NADP+ levels has been observed throughout fruit development. In summary, this report suggests that niacin may be used to increase NADPH oxidases, thus increasing stress-tolerance in kiwifruit during encounter of short-term stressful conditions.

To reduce production costs and increase efficiency in ageing coal mines, a system that utilizes two shearers in a fully mechanized longwall working face is proposed. Using theoretical and engineering experience, three issues pertaining to the system were determined: (a) the two-shearer mining technique; (b) matching and modifying the equipment; (c) and coal-cutting task allocation for each shearer. Two mining processes are proposed, with the two shearers travelling in either the same or opposing directions. To avoid breakage of chains and pan extrusion of the armoured face conveyor (AFC), the length of the AFC 'snake' was controlled. To ensure continuous cutting and transporting of coal, the cross-sectional dimensions of the AFC were checked and the shearer closer to the head drive was modified. Based on the assumption of equal cutting times for each shearer, the meeting position of the two shearers was obtained by using a theoretical model of mining task allocation. Application of this technique at No. 2 Jining Mine, China, has shown remarkable benefits: daily production capacity was increased by 54% and personnel efficiency was improved by 33 t/d per person.

The mining of steeply dipping coal seams involves significant safety risks because of instability caused by the weak nature of surrounding rocks and their movement after excavation. Physical and numerical models were developed to determine an optimum backfill approach to ground control in steeply dipping coal seams. Physical modelling showed that for mining without backfill, the thickness of the largest roof collapse was approximately twice that of the mined seam, the movement of roof strata tended to be asymmetrical, and there was a relatively large empty zone in the upper gob area. For mining with backfill, roof conditions were significantly improved; only slight roof separation appeared 1.5 m above the mined seam. The largest cumulative displacements occurred 5-15 m from the first cut of mining. With backfill, the levels of mining-induced stress release and concentration were significantly reduced, and the vertical range of mining disturbance was shortened by 18%. Numerical modelling showed that floor strata mainly undergo nearly horizontal displacements, while roof strata mainly undergo vertical subsidence, both with and without backfill. The integrity of roof strata improved as the extent of backfill increased, and the range of displacement direction increased. The conclusions are confirmed by results from a field trial.

Teosinte branched 1/cycloidea/proliferating cell factor 1 (TCP) proteins are plant-specific transcription factors playing crucial roles in various biological processes, such as leaf development, flower symmetry, shoot branching and senescence. However, no comprehensive analysis of the TCP gene family has been reported in grapevine (Vitis vinifera L). Herein, a total of 15 TCP family members were identified in the genome of grapevine, located on eight of the 19 chromosomes. Phylogenetic and structural analyses showed that the VvTCPs were classified into two groups, designated as Class I and Class II. The Class II genes were further divided into two subclasses, the CIN subclass and the CYC/TB1 subclass. Genes belonging to the same subclass shared similar gene structures, conserved domains and motifs. Real-time PCR showed that almost all members of Class II exhibited organ-specific expression patterns, while members of Class I and the CIN Class were ubiquitously expressed in all the tissues examined, indicating multiple roles in the development of different grapevine organs. In addition, many members were strongly modulated by abiotic (cold, heat, drought) and biotic (downy mildew and powdery mildew infection) stresses, suggesting important and diverse regulatory roles in adverse conditions and plant immunity. The comprehensive in silico analysis of the grapevine TCP transcription factor family gives us some references to potential functions in grapevine development and stress responses.

Kidney stone disease is a major cause of chronic renal insufficiency. The role of long non-coding RNAs (lncRNAs) in calcium oxalate-induced kidney damage is unclear. Therefore, we aimed to explore the roles of lncRNAs in glyoxylate-exposed and healthy mouse kidneys using microarray technology and bioinformatics analyses. A total 376 mouse lncRNAs were differentially expressed between the two groups. Using BLAST, 15 lncRNA homologs, including AU015836 and CHCHD4P4, were identified in mice and humans. The AU015836 expression in mice exposed to glyoxylate and the CHCHD4P4 expression in human proximal tubular epithelial (HK-2) cells exposed to calcium oxalate monohydrate were analyzed, and both lncRNAs were found to be upregulated in response to calcium oxalate. To further evaluate the effects of CHCHD4P4 on the cell behavior, we constructed stable CHCHD4P4-overexpressing and CHCHD4P4-knockdown HK-2 cells. The results showed that CHCHD4P4 inhibited cell proliferation and promoted the epithelial-mesenchymal transition in kidney damage and fibrosis caused by calcium oxalate crystallization and deposition. The silencing of CHCHD4P4 reduced the kidney damage and fibrosis and may thus be a potential molecular target for the treatment of kidney stones.

The receptor activator of nuclear factor κB ligand (RANKL)/RANK pathway plays an important role in the prognosis of several solid tumor types, but its role in gastric cancer prognosis has been poorly characterized. A total of 116 gastric cancer patients who underwent surgical resection were enrolled in this study. Expressions of RANKL and RANK in gastric cancer tissues were detected using immunohistochemical staining. Thirty-eight patients (33%) showed a high level of RANKL expression and 61 patients (53%) showed a high level of RANK expression. There was a positive correlation between expressions of RANKL and RANK (P=0.014, r=0.221). A high level of RANKL expression indicated shorter overall survival (OS) (P=0.008), and was associated with a higher pathological tumor/lymph node/metastasis (pTNM) stage (P=0.035), while no significant correlation was detected between RANK expression and clinicopathological parameters. RANKL also predicted poor prognosis in patients with high RANK expression (P=0.008) and Bormann's type III/IV (P=0.002). Furthermore, RANKL expression correlated with pTNM stage according to high RANK expression (P=0.009), while no significance was found in patients with low RANK expression (P=1.000). Together, our results revealed that high expression of RANKL could predict worse outcomes in gastric cancer especially combined with RANK detection, and thereby this pathway could be a useful prognostic indicator of gastric cancer.

Abstract This paper investigates the dynamic response of rectangular prestressed membrane subjected to concentrated impact load based on multiple scale perturbation method. The governing equations of motion of nonlinear vibration are derived based on the Föppl large deflection theory and Galerkin method. By introducing different time scales to consider the process of vibration, the results of dynamic response are obtained by applying the multiple scale perturbation method. Furthermore, the effects of pretension force, velocity of load and dimension of membrane on the dynamic response of membrane are discussed. The present work studies the problem of the dynamic response of prestressed membrane subjected to concentrated impact load in different time scales, and provides a more accurate theoretical model for design of membrane structure.

The aim of this study was to evaluate the feasibility of endoscopy to remove keratocystic odontogenic tumors (KCOTs) with virtual 3D mandibular images. Fifteen patients (mean age, 40.27±14.58 years) who underwent endoscopic mandibular KCOT enucleation between May 2009 and October 2009 were included. Virtual 3D mandibular reconstructions derived from computed tomography (CT) imaging were generated for all patients. Recurrence and pathological fracture were evaluated as the primary outcome variables at 1 and 12 months after operation. Secondary infection and inferior alveolar nerve injury were evaluated as the secondary outcome variables at 1 and 6 months after operation. None of the 15 patients exhibited signs of recurrence or pathological fracture after operation. During long-term follow-up, no symptoms of inferior alveolar nerve injury or secondary infection were observed and no signs of recurrence were found in any of the patients. Endoscopy helps surgeons to remove mandibular KCOTs with small incisions. Moreover, endoscopy can provide clear and magnified views and help to avoid damage to the inferior alveolar neurovascular bundle. Therefore, under the support of preoperative virtual 3D mandibular images, the application of endoscopy to remove the tumors should be considered to be a treatment option for KCOTs.
