Mari-mari (Cassia leiandra Benth.) is an Amazonian native fruit that is highly appreciated in the northern region of Brazil, but there are few studies regarding its chemical composition and/or biological properties. A phytochemical study was conducted on the extracts of the pulp, peel and seeds; and their antioxidant, cytotoxic and antimicrobial potential were also determined. Methanol and acetone extracts from different fruit parts were subjected to total phenolic, 2,2-diphenyl-1 picrylhydrazyl (DPPH) radical scavenging and FeIII reducing capacity (FRAP) assays. From the peel extracts, the engeletin and astilbin could also be isolated and characterized based on nuclear magnetic resonance spectroscopy and high-resolution mass spectrometry. A quantification using ultra-fast liquid chromatography of acetone dry crude extracts (DCE) of the peel showed that engeletin was the main compound (532.77 mg g-1 DCE). Astilbin content was also determined (131.53 mg g-1 DCE). The antibacterial and antiproliferative activities were evaluated and the methanol extract of the peel showed higher growth inhibition activity against all tested bacteria. Engeletin showed a relevant antimicrobial effect against Staphylococcus aureus, Staphylococcus epidermidis and Escherichia coli. Acetone and methanol peel extracts exhibited a relevant inhibitory effect on the proliferation of breast carcinoma cells (MCF-7), with no toxicity to the normal cell line (NIH/3T3).

In this work, plasma nitriding (PN) and cathode cage plasma deposition (CCPN) treatments were carried out with temperatures of 400 and 450 °C to evaluate the modifications caused on the surface of pure niobium samples. XRD, SEM, Vickers microhardness, and sphere-disk analyses were used to characterize the coatings’ composition, morphology, hardness, and wear resistance. The results showed that the treated samples increased hardness and wear resistance, with the sample submitted to CCPN at 450 °C presenting the best tribological behavior.

Abstract Ortho-eugenol is a synthetic derivative from eugenol, the major compound of clove essential oil, which has demonstrated antidepressant and antinociceptive effects in pioneering studies. Additionally, its effects appear to be dependent on the noradrenergic and dopaminergic systems. Depression and anxiety disorders are known to share a great overlap in their pathophysiology, and many drugs are effective in the treatment of both diseases. Furthermore, high levels of anxiety are related to working memory deficits and increased oxidative stress. Thus, in this study we investigated the effects of acute treatment of ortho-eugenol, at 50, 75 and 100 mg/kg, on anxiety, working memory and oxidative stress in male Swiss mice. Our results show that the 100 mg/kg dose increased the number of head-dips and reduced the latency in the hole-board test. The 50 mg/kg dose reduced malondialdehyde levels in the prefrontal cortex and the number of Y-maze entries compared to the MK-801-induced hyperlocomotion group. All doses reduced nitrite levels in the hippocampus. It was also possible to assess a statistical correlation between the reduction of oxidative stress and hyperlocomotion after the administration of ortho-eugenol. However, acute treatment was not able to prevent working memory deficits. Therefore, the present study shows that ortho-eugenol has an anxiolytic and antioxidant effect, and was able to prevent substance-induced hyperlocomotion. Our results contribute to the elucidation of the pharmacological profile of ortho-eugenol, as well as to direct further studies that seek to investigate its possible clinical applications.

Resumo Ortho-eugenol é um derivado sintético do eugenol, composto principal do óleo essencial de cravo, que demonstrou efeitos antidepressivos e antinociceptivos em estudos pioneiros. Além disso, seus efeitos parecem ser dependentes dos sistemas noradrenérgico e dopaminérgico. Sabe-se que os transtornos de depressão e ansiedade compartilham uma grande sobreposição em sua fisiopatologia, e muitas drogas são eficazes no tratamento de ambas as doenças. Além disso, altos níveis de ansiedade estão relacionados a déficits de memória de trabalho e aumento do estresse oxidativo. Assim, o presente estudo investigou os efeitos do tratamento agudo de orto-eugenol, nas doses de 50, 75 e 100 mg/kg, na ansiedade, memória de trabalho e estresse oxidativo em camundongos Swiss machos. Nossos resultados mostram que a dose de 100 mg/kg aumentou o número de mergulhos e reduziu a latência no teste da placa perfurada. A dose de 50 mg/kg reduziu os níveis de malondialdeído no córtex pré-frontal e o número de entradas no labirinto em Y em comparação com o grupo de hiperlocomoção induzida por MK-801. Todas as doses reduziram os níveis de nitrito no hipocampo. Também foi possível avaliar uma correlação estatística entre a redução do estresse oxidativo e a hiperlocomoção após a administração do orto-eugenol. No entanto, o tratamento agudo não foi capaz de prevenir os déficits de memória de trabalho. Portanto, o presente estudo mostra que o orto-eugenol tem efeito ansiolítico e antioxidante, sendo capaz de prevenir a hiperlocomoção induzida pela substância. Nossos resultados contribuem para a elucidação do perfil farmacológico do orto-eugenol, bem como para direcionar novos estudos que busquem investigar suas possíveis aplicações clínicas.

The formation of solid waste is a global challenge regarding environmental preservation. Due to the lifestyle of modern society, which aims to improve practicality, new products are constantly being released in the market and increasing the formation of waste. The decomposition of waste from various origins produces a dark liquid named leachate, which presents high rates of organic and inorganic compounds. The objective of this study was to assess the concentration of potentially toxic metals in the leachate collected from the Aurá dump. The concentrations of Cd, Cu, Cr, Mn, Ni, Pb, and Zn were evaluated in leachate samples collected in a period of a year, covering two typical seasonal periods of the Amazon region with more and less rainfall. The leachate samples collected in the Aurá dump were determined in the Toxicology Laboratory of Instituto Evandro Chagas (IEC), by means of Plasma/Inductively Coupled Optical Emission Spectrometry (ICP-OES). The Cr, Mn, and Pb metals analyzed in the slurry samples are in a concentration above the maximum limits specified by CONAMA Resolution No. 430/2011, causing significant environmental concerns.

Resumo As infecções agudas por Giardia geralmente causam diarreia e dores de estômago. As infecções crônicas podem levar à desnutrição, deficiências de micronutrientes, má absorção e perda de peso. Este estudo avaliou a prevalência da infecção por G. lambia e os fatores de risco associados em pacientes imunocomprometidos em tratamento quimioterápico no sul do Brasil. Um total de 110 pacientes imunocomprometidos de Pelotas, RS, Brasil, consentiram em participar deste estudo e foram recrutados. O perfil socioeconômico e epidemiológico dos pacientes foi coletado por meio de questionário. A prevalência de Giardia foi determinada através de microscopia pela técnica de centrifugação-flutuação utilizando amostras de fezes de cada paciente. Além disso, a caracterização genética do parasita foi realizada pela amplificação e sequenciamento do gene da glutamato desidrogenase (gdh). À microscopia, a prevalência de giardíase foi de 17,3% (19/110). Além disso, as sequências de DNA revelaram que 7 (36,8%) dos 19 isolados pertenciam ao agrupamento B, enquanto 6 deles (31,6%) pertenciam ao agrupamento C, 5 (26,3%) ao agrupamento A e 1 (5,3%) ao agrupamento D. Os fatores de risco (p ≤ 0,05) para giardíase foram, escolaridade (OR=8,0 (1,02 – 62,91), dividir casa com mais de três pessoas (OR=14,1 (3,77 – 52,51), fontes de água (OR=38,9 (10,4 – 145,7), tratamento de esgoto (OR=14,2 (3,1 – 65,5), destinação do lixo (OR=7,44 (2,0 – 27,3), possuir animais de estimação (OR=4,6 (1,0 – 21,2) e cultivar horta (OR=4,2 (1,3 – 13.6). A prevalência de G. lamblia em pacientes imunocomprometidos foi considerada elevada com a identificação de quatro conjuntos do parasito (A, B, C e D).

Abstract Acute Giardia infections often cause diarrhea and stomach upset. Chronic infections can lead to malnutrition, micronutrient deficiencies, malabsorption and weight loss. This study assessed the prevalence of G. lambia infection and assessed associated risk factors among immunocompomised patients undergoing chemotherapeutic treatment in southern Brazil. A total of 110 immunocompromised patients in Pelotas, RS, Brazil, consented to participate in this study and were recruited. Socioeconomic and epidemiological profile of patients was collected by questionnaire. The prevalence for Giardia were determined through microscopy by the centrifugation-flotation technique using stool samples of every patient. In addition, the genetic characterization of the parasite was carried out by amplifying and sequencing the glutamate dehydrogenase (gdh) gene. By microscopy, the prevalence of giardiasis was 17.3% (19/110). Furthermore, the DNA sequences revealed that 7 (36.8%) out of 19 isolates belonged to assemblage B, while 6 of them (31.6%) belonged to assemblage C, 5 (26.3%) to assemblage A and 1 (5.3%) to assemblage D. Risk factors (p ≤ 0.05) for giardiasis were schooling level (OR=8.0 (1.02 – 62.91) sharing a house with more than three people (OR=14.1 (3.77 – 52.51), water sources (OR=38.9 (10.4 – 145.7), sewage treatment (OR=14.2 (3.1 – 65.5), waste destination (OR=7.44 (2.0 – 27.3), owning pets (OR=4.6 (1.0 – 21.2) and cultivating a vegetable garden (OR=4.2 (1.3 – 13.6). The prevalence of G. lamblia in immunocompromised patients was considered elevate with the identification of four assemblage of the parasite (A, B, C and D).

Resumo Hymenaea martiana é uma espécie popularmente conhecida no Nordeste do Brasil como “jatobá” e usada na medicina popular para tratar a dor e a inflamação. O objetivo deste trabalho foi avaliar a atividade antinociceptiva e anti-inflamatória de H. martiana. No presente estudo, foram avaliados os efeitos do extrato etanólico bruto (Hm-EtOH) e da fração acetato de etila (Hm-AcOEt) em modelos de nocicepção e inflamação em camundongos. Foram utilizados estímulos químicos (contorções abdominais induzidas por ácido acético e teste da formalina) e estímulo térmico (teste da placa quente) para avaliação da atividade antinociceptiva, enquanto no perfil anti-inflamatório foi utilizado o teste do edema de pata induzido por carragenina e migração de leucócitos para a cavidade peritoneal. A presença do flavonoide astilbina nas amostras foi caracterizada através de análise por CLAE-DAD-EM. Hm-EtOH e o Hm-AcOEt (100, 200 e 400 mg.kg-1, i.p.) reduziram significativamente o número de contorções abdominais e diminuíram o tempo de lambida da pata no teste da formalina. No teste da placa quente, houve aumento do tempo de latência dos animais. Hm-EtOH e Hm-AcOEt inibiram significativamente o aumento do edema após a administração de carragenina, bem como inibiram a migração de leucócitos no teste de peritonite. A análise por CLAE-DAD-EM de Hm-EtOH e Hm-AcOEt revelou a presença do flavonoide astilbina nas amostras. De acordo com os resultados deste estudo, tanto Hm-EtOH quanto o Hm-AcOEt possuem atividades antinociceptiva e anti-inflamatória, o que pode estar relacionado à presença do flavonoide. Os resultados reforçam o uso popular desta planta.

Abstract Hymenaea martiana is a species popularly known in Northeastern Brazil as “jatobá” and used in folk medicine to treat pain and inflammation. The aim of this work was to evaluate the antinociceptive and anti-inflammatory activity of H. martiana. In the present study, we carried out an investigation about the effects of the crude ethanolic extract (Hm-EtOH) and the ethyl acetate fraction (Hm-AcOEt) in models of nociception and inflammation in mice. Chemical (acetic acid-induced writhing and formalin) and thermal stimuli (hot plate) were used for the evaluation of antinociceptive activity, while for the anti-inflammatory profile paw edema induced by carrageenan was used, along with leukocyte migration to the peritoneal cavity. The presence of the flavonoid astilbin in the samples was characterized through HPLC-DAD-MS analysis. Hm-EtOH and Hm-AcOEt (100, 200 and 400 mg.kg-1, i.p.) significantly reduced the number of abdominal contortions and decreased the paw licking time in the formalin test. In the hot plate, the extract increased the latency time of animals. Hm-EtOH and Hm-AcOEt inhibited significantly the increase in the edema after the administration of carrageenan. Hm-EtOH and Hm-AcOEt inhibited leukocyte migration in the peritonitis test. HPLC-DAD-MS analysis of Hm-EtOH and Hm-AcOEt revealed the presence of the flavonoid astilbin in the samples. According to the results of this study, both Hm-EtOH and Hm-AcOEt have antinociceptive and anti-inflammatory activities, which could be related with the presence of flavonoid in the extracts. The results reinforce the popular use of this plant.

Abstract Background: The evaluation of selective herbicides for weed control in winter cereals is extremely important. Simple methods to evaluate alterations caused by herbicides in the growth and development of winter cereals can be performed with vegetation indices. Objective: Evaluate the potential of different vegetation indices by optical sensors to detect phytotoxicity caused by herbicides in winter cereals. Methods: The experiment was conducted under field conditions, in a randomized block design with four replications. Herbicides were sprayed when the plants were at Z24. The phytotoxicity was evaluated at 7, 14 and 21 days after treatment (DAT) and the normalized difference vegetation index (NDVI) and the normalized difference red edge index (NDRE) at 16, 22 and 37 DAT. Results: The herbicide 2,4-D was less phytotoxic to winter cereals, while metribuzin and saflufenacil caused higher percentages (up to 50%) of visible injuries. The NDRE discriminated more precisely than NDVI the variations in winter cereals, showing alteration in the chlorophyll content due to the phytotoxicity caused by the herbicides. The canonical correlation analysis found an inverse relationship between phytotoxicity and vegetation indices. Conclusions: The winter cereals studied were tolerant to herbicides, enabling the recovery of growth and development. The NDRE index was the most sensitive and showed greater potential in identifying injuries caused by herbicides. The canonical correlation analysis confirmed the inverse relationship between phytotoxicity and the reduction of vegetation indices.

Abstract The extensive marine biodiversity has proved to be a promising source of substances with biomedical potential. In this study, the cytotoxicity of the Brazilian octocoral Phyllogorgia dilatata (Gorgoniidae) was evaluated against two tumor cell lines and three bacterial strains. The methanol/dichloromethane crude extract presented no antibacterial activity up to the highest concentration tested (512 µg/mL), however it revealed a noteworthy antiproliferative effect against HCT-116 (80%) and MCF-7 (54%) cell lines at 50 μg/mL. Therefore, guided by the cytotoxic activity, a multistep chemical fractionation of the extract provided the subfraction 5 (PDPH2-5) with IC50 values of 3.18 and 17.80 μg/mL against HCT-116 and MCF-7, respectively. The LC-HRMS/MS analysis of PDPH2-5 showed ions of m/z 219.1742 and 219.1743, characterized as (E,E) and (Z,E) germacrone, after a LC-DAD-SPE/NMR analysis of the hexanic fraction and comparisons of NMR data with the literature. Previously reported assessments to the cytotoxic activity of the (E,E)-diastereoisomer disclosed higher IC50 values than that obtained for the PDPH2-5 fraction, suggesting, herein, a potentiated effect of the diastereoisomeric mixture. Such remark encourage further bioactivity studies with stereoisomer mixtures and reduce the urge for compound isolation.

Resumo A reciclagem tem sido apontada como alternativa ao descarte de resíduos em aterros industriais. No presente trabalho é mostrada a transformação de resíduos (areia descartada de fundição - ADF, grits e lama de cal) em materiais vitrocerâmicos. Os vidros foram obtidos pelo método de fusão/resfriamento. Os materiais precursores, compostos vítreos e vitrocerâmicos foram caracterizados pelas técnicas de difração de raios X (DRX), fluorescência de raios X (FRX) e calorimetria exploratória diferencial/análise termogravimétrica (CED/ATG). Os materiais vítreos foram moídos, pastilhados e tratados termicamente nas temperaturas de cristalização obtidas por CED para preparar os materiais vitrocerâmicos (885, 961 e 1090 ºC). As fases obtidas foram cristobalita, α-wolastonita (parawolastonita) e β-wolastonita (pseudowolastonita). As vitrocerâmicas apresentaram baixa absorção de água e porosidade aparente (0,26 até 0,88 wt% e 0,66 até 1,77 vol%, respectivamente). Os resultados confirmaram que os resíduos estudados podem ser utilizados como matéria-prima na fabricação de materiais vítreos e vitrocerâmicos.

Abstract Recycling has been pointed out as an alternative to the disposal of waste materials in industrial landfills. In the present study, the transformation of residues (discarded foundry sand - DFS, grits, and lime mud) in glass-ceramic materials is shown. The glasses were obtained by the melting/cooling method. The precursor materials, glasses, and glass-ceramics were characterized by X-ray diffraction (XRD), X-ray fluorescence (XRF), and differential scanning calorimetry/thermal gravimetric analysis (DSC/TGA). The glassy materials were milled, pelleted, and thermally treated at the crystallization temperatures given by DSC data to obtain the glass-ceramics (885, 961, and 1090 ºC). The main formed phases were cristobalite, α-wollastonite (parawollastonite), and β-wollastonite (pseudowollastonite). The glass-ceramics showed very low water absorption and apparent porosity (0.26 to 0.88 wt% and 0.66 to 1.77 vol%, respectively). The results confirmed that the studied residues can be used as raw materials for the manufacture of vitreous and glass-ceramic materials.

Resumo Este estudo descreve a formação de novas ligações C-C através da reação de alilação de diferentes aldeídos promovida pela zeólita beta. Este método ecologicamente amigável foi caracterizado por sua eficiência, versatilidade e quimiosseletividade, na formação de diferentes álcoois homoalílicos. Os álcoois homoalílicos foram obtidos em bons rendimentos (80% a 96%), tempos reacionais curtos (30 a 75 min) em temperatura ambiente e sem a necessidade de purificações adicionais. Além disso, foi utilizada água como cossolvente garantindo ao método um desenvolvimento no âmbito da química verde. Adicionalmente, constatou-se que a zeólita beta pode ser reutilizada por até um ciclo reacional sem perda de sua eficiência.

Abstract This study describes the formation of new C-C bonds through the allylation reaction of different aldehydes promoted by beta zeolite. This environmentally friendly method was characterized by its efficiency, versatility, and chemoselectivity in the formation of different homoallylic alcohols. Homoallylic alcohols were obtained in good yields (80% to 96%), short reaction times (30 to 75 min) at room temperature, and without the need for additional purifications. In addition, water was used as a co-solvent, ensuring the method a development in the field of green chemistry. Moreover, it was found that the zeolite beta can be reused for up to one reaction cycle without loss of its efficiency.

Resumo Os concretos apresentam alto potencial de uso nas mais diversas áreas da construção civil e suas propriedades podem ser melhoradas através da utilização de materiais alternativos por substituição ou adição ao cimento Portland. A diatomita é um material sedimentar composto principalmente por sílica e impurezas amorfas e possui granulometria muito fina, alta porosidade e baixa massa específica. Foram produzidos concretos em proporção de 1:1:2 (cimento: areia: brita) com adição de diatomita em níveis de 5% e 10% em massa de cimento e relação água/cimento de 0,54. As propriedades físico-mecânicas dos concretos foram avaliadas por meio dos ensaios de resistência à compressão e absorção de água por imersão. Também foram realizados ensaios de difratometria de raios X e microscopia eletrônica de varredura para análises das principais fases na pasta de cimento. Os resultados indicaram que a adição de diatomita no concreto aumentou a resistência à compressão devido ao teor de sílica no material e reduziu levemente a porosidade.

Abstract Concretes present great potential to be used in many areas of civil construction. Their properties can be improved by substitution or addition of alternative materials to the Portland cement. The diatomite is a sedimentary material composed mainly by silica and amorphous impurities, presenting very fine particle size, high porosity, and low specific mass. Concrete was produced in a ratio of 1:1:2 (cement: sand: gravel) with the addition of diatomite at contents of 5% and 10% by cement weight and a water/cement ratio of 0.54. The physico-mechanical properties of the concretes were evaluated by tests of compressive strength and water absorption by immersion. Also, X-ray diffraction and scanning electron microscopy were carried out to analyze the main phases in the cement paste. The results indicated that the addition of diatomite in the concrete increased the compressive strength due to the silica content in the material and reduced slightly the porosity.

Abstract Rabies is a fatal zoonotic infection of the central nervous system of mammals and has been known to humans for millennia. The etiological agent, is a neurotropic RNA virus in the order Mononegavirales, family Rhabdoviridae, genus Lyssavirus. There are currently accepted to be two cycles for rabies transmission: the urban cycle and the sylvatic cycle. The fact that both cycles originated from a common RABV or lyssavirus ancestor and the adaptive divergence that occurred since then as this ancestor virus adapted to a wide range of fitness landscapes represented by reservoir species in the orders Carnivora and Chiroptera led to the emergence of the diverse RABV lineages currently found in the sylvatic and urban cycles. Here we study full genome phylogenies and the time to the most recent common ancestor (TMRCA) of the RABVs in the sylvatic and urban cycles. Results show that there were differences between the nucleotide substitution rates per site per year for the same RABV genes maintained independently in the urban and sylvatic cycles. The results identify the most suitable gene for phylogenetic analysis, heterotachy among RABV genes and the TMRCA for the two cycles.

Resumo Membranas cerâmicas são de grande interesse industrial em processos de separação. Elas são caracterizadas por elevada estabilidade química/térmica e capacidade de filtração em temperaturas acima de 500 °C em uma larga faixa de valores de pH. Entretanto, a grande desvantagem das membranas cerâmicas é o elevado custo de produção envolvendo matérias-primas sintéticas, geralmente alumina e zircônia. Nesse contexto, membranas cerâmicas de baixo custo foram preparadas por sinterização reativa usando uma mistura de argila caulinítica e cinza de bagaço de cana de açúcar. Análises de distribuição de tamanho de partícula, térmica, mineralógica e de composição química foram realizadas para estudar as matérias-primas. As propriedades tecnológicas e de permeabilidade à água foram determinadas em amostras queimadas entre 800 e 1000 °C. A eficiência de filtração foi medida através de análises comparativas entre a água bruta e o filtrado. O tamanho médio dos poros entre 2,5 e 6,0 µm viabiliza o uso das membranas derivadas de cinza de bagaço de cana de açúcar em processos de microfiltração.

Abstract Ceramic membranes are of great industrial interest in separation processes. They are characterized by high chemical and thermal stabilities and filtration capacity at high temperatures (>500 °C) in a wide range of pH values. However, the major disadvantage of ceramic membranes is the high-cost of production involving synthetic raw materials, usually alumina and zirconia. In this work, low-cost ceramic membranes were prepared by solid-state reactive firing using a mixture of kaolinite clay and sugarcane bagasse ash. Particle size distribution, thermal, mineralogical, and chemical composition analyses were carried out to study the raw materials. Technological properties and water permeability were investigated in samples fired between 800 and 1000 °C. The filtration efficiency was measured by comparative analyses between the raw water and the filtrate. The mean pore size ranging from 2.5 to 6.0 μm makes the sugarcane bagasse ash derived ceramic membranes suitable for microfiltration processes.
