Spatial analysis of the natural infection index for Triatomines and the risk of Chagas disease transmission in Northeastern Brazil

ABSTRACT This study aimed to analyze the spatial pattern of natural infection index (NII) for triatomines and the risk of Chagas disease transmission in an endemic area of Northeastern Brazil. An ecological study was conducted, based on 184 municipalities in five mesoregions. The NII for triatomines was evaluated in the Pernambuco State, Brazil, from 2016 to 2018. Spatial autocorrelations were evaluated using Global Moran Index (I) and Local Moran Index (II) and were considered positive when I > 0 and p < 0.05, respectively. In total, 7,302 triatomines belonging to seven different species were detected. Triatoma brasiliensis had the highest frequency (53%; n = 3,844), followed by Triatoma pseudomaculata (25%; n = 1,828) and Panstrongylus lutzi (18.5%; n=1,366). The overall NII was 12%, and the higher NII values were P. lutzi (21%) and Panstrongylus megistus (18%). In the mesoregions of Zona da Mata, Agreste, Sertao, and Sertao do Sao Francisco, 93% of triatomines were detected indoors. The global spatial autocorrelation of I to NII was positive (0.2; p = 0.01), and II values calculated using BoxMap, MoranMap, Lisa Cluster Map were statistically significant for natural infections. With regard to the risk areas for the presence of triatomines, Zone 2 (the Agreste and Sertao regions) presented a relative risk of 3.65 compared to other areas in the state. Our study shows the potential areas of vector transmission of Chagas disease. In this study, the application of different methods of spatial analysis made it possible to locate these areas, which would not have been identified by only applying epidemiological indicators.

Abstract This study evaluated the chemical profile and antimicrobial activity of essential oil (EO) ofP. dioica. The EOs were extracted by hydrodistillation and chemically characterized by gas chromatography coupled to mass spectrometry (GC/MS). The total phenolics were quantified by the FolinCiocalteu method. For the antimicrobial assay, the Disc Diffusion and Broth Dilution method were applied to obtain the minimum inhibitory concentration and minimum bactericidal concentration. The main constituent of the EO was eugenol. The EO showed bactericidal activity against E. coli,S. aureus, P. aeruginosa, Salmonella sp., B. cereus, P. mirabilis, K. pneumoniae, S. sonnei, C. albicans, Fusarium sp., Penicillium sp.andAspergilus sp.The results obtained are encouraged by the potential use of the EO studied in the control and combat of pathogenic microorganisms.
Resumen Este estudio evaluó el perfil químico y la actividad antimicrobiana del aceite esencial (AE) de P. dioica. Los AE fueron extraídos por hidrodestilación y caracterizados químicamente por cromatografía de gases acoplados a espectrometría de masas (GC/MS). Los fenólicos totales fueron cuantificados por el método Folin-Ciocalteu. Para el ensayo antimicrobiano, se aplicó el método de difusión de discos y dilución de caldo para obtener la concentración inhibitoria mínima y la concentración bactericida mínima. El componente principal de la AE fue el eugenol. La AE mostró actividad bactericida contra E. coli, S. aureus, P. aeruginosa, Salmonella sp., B. cereus, P. mirabilis, K. pneumoniae, S. sonnei, C. albicans, Fusarium sp., Penicilliumsp. yAspergilussp. Los resultados obtenidos son estimulados por el uso potencial de la AE estudiada en el control y combate de microorganismos patógenos.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Helicometrina nimia Linton, 1910 (Digenea: Opecoelidae) in dusky grouper Epinephelus marginatus (Lowe, 1834) (Teleostei: Serranidae) from southeastern Brazil

Helicometrina nimia (Opecoelidae) é um digenético com distribuição mundial. As famílias de peixes mais comumente utilizadas como hospedeiros por H. nimia são Serranidae, Pomodasydae, Scorpaenidae e Clinidae. No presente estudo, um novo hospedeiro e uma nova localidade são apresentados para a espécie. A descrição dos espécimes estudados, além de comentários sobre o seu status taxonômico e uma tabela comparativa de medidas de H. nimia são apresentados. O status taxonômico dos membros de Helicometrina tem sido questionável. A maior controvérsia dentro do gênero parece estar relacionada com a validade das características diagnósticas, especialmente no que diz respeito ao número de testículos. No presente estudo, todos os espécimes estudados apresentaram número permanente e estável de testículos (n=9) e, portanto, sua utilização como caráter diagnóstico é suportada pelos presentes autores. Epinephelus marginatus é considerada um novo hospedeiro para Helicometrina nimia e São Paulo, sudeste do Brasil, uma nova localidade para a espécie.
Helicometrina nimia (Opecoelidae) is a digenean with wide distribution. Fish families most commonly used as hosts for H. nimia are Serranidae, Pomodasydae, Scorpaenidae and Clinidae. In the present study, a new host and a new host locality are presented for the species. A description of the studied specimens, besides comments concerning its taxonomic status and biometrically compared tables of H. nimia reports are given. The taxonomic status of members of Helicometrina has been questionable. The greatest controversy for the genus seems to be related to the validity of diagnostic features, especially in regard to the number of testes. In the present study, all studied specimens presented a permanent and steady number of testes (n=9) and therefore its use as a diagnostic character is supported by the present authors. Epinephelus marginatus is considered a new host for Helicometrina nimia, and São Paulo state, southeastern Brazil, a new locality for the species.

Hydrographic and hydrogeological basin of Entre-Ribeiros: probable recharge zone delimitation and environmental impact assessment

Apresenta-se uma proposta de delimitação das prováveis zonas de recarga da Bacia Hidrográfica de Entre-Ribeiros (Noroeste de Minas Gerais/Brasil). A delimitação teve como subsídio estudos de litoestratigrafia, geomorfologia, unidades geoambientais e hidrogeologia. A partir da delimitação proposta, e conjugando-se à análise da ocupação do solo entre o período de 1975 a 2008, procura-se identificar as tendências de ocupação das zonas prováveis de recarga. Por fim, tecem-se comentários sobre os impactos ambientais decorrentes da ocupação dessas áreas.
The probable recharge zone delimitation of Entre-Ribeiros Basin (Northwest of the state of Minas Gerais / Brazil) is proposed in this study. The delimitation is based upon stratigraphy, geomorphology, geo-environmental domains and hydrogeology studies. Combining the recharge zone map with the land use variation between 1975 and 2008, the occupation trends of possible recharge zones are identified. Concluding, the environmental impacts for this basin are discussed.

Effects of nitrate and phosphate availabilities on growth, photosynthesis and pigment and protein contents in colour strains of Hypnea musciformis (Wulfen in Jacqu.) J.V. Lamour. (Gigartinales, Rhodophyta)

In Brazil, Hypnea musciformis is the main raw material for carrageenan production and the knowledge of nitrogen and phosphorus metabolism in algae is critical for the success of cultivation because these elements can limit seaweed productivity. Thus, the objective of this study was to evaluate the effects of nitrate (zero to 100 μM) and nitrate plus phosphate (zero to 25 μM) availabilities on the growth, the contents of photosynthetic pigments (phycobiliproteins and chlorophyll a) and proteins, and the photosynthesis and respiration of the brown (BR) and light green (LG) strains of H. musciformis. The results revealed metabolic differences between the colour strains of H. musciformis for nitrogen metabolism: upon nitrate addition, the LG strain stored nitrogen mainly as proteins, while the BR strain stored it as proteins and pigments. Moreover, the respiration of the LG strain and the photosynthesis of the BR strain increased with nitrate concentrations, indicating that the BR strain fixed more photosynthetic carbon than the LG strain.

Evaluation of the allergenicity of spore and mycelia extracts of Pisolithus tinctorius

Foi realizada a análise bioquímica, antigênica e alergênica de extratos de esporos e micélios de Pisolithus tinctorius. Os esporos foram coletados de basidiocarpos em plantações de Eucaliptus spp e os micélios obtidos de cultura em meio de MNM. Baseado na curva de crescimento do fungo, obteve-se massa miceliana aos 10, 20, 30 e 40 dias de incubação, que correspondem respectivamente ao início, meio, final da fase log e início da fase de declínio. A massa miceliana, juntamente com os esporos, foram submetidos a ação de três líquidos extratores (Coca, Tris-HCl e bicarbonato de amônio). Determinou-se os teores de carboidratos e proteínas. A análise eletroforética por SDS-PAGE revelou, além de frações comuns, frações distintas nestes extratos, em função do tempo de cultivo e métodos de extração. Os extratos selecionados para os testes alérgicos foram aqueles com maior número de frações. Realizou-se o "prick-test" em trabalhadores rurais, trabalhadores de áreas de plantações de Eucaliptus spp e em estudantes universitários, totalizando 374 pacientes. A positividade ao "prick-test" com extrato antigênico de P. tinctorius foi, respectivamente, 3,78%, 28,20% e 6,40%. A maioria dos pacientes positivos ao "prick-test" (82,75%) apresentava também sintomas de alergia respiratória (asma e/ou rinite). Não houve diferença de reatividade quando do emprego de extratos de esporos e de micélio do fungo. A análise dos soros dos pacientes positivos, revelou a presença de IgE específica aos antígenos do P. tinctorius. Considerando que o Pisolithus tinctorius é encontrado como micorriza de Eucaliptus spp e que o reflorestamento, na maioria dos países, é feito com esta planta, deve ser relevada a importância deste fungo como possível causador de alergias respiratórias, principalmente em trabalhadores ocupacionalmente expostos
The antigenic and allergenic chemical analysis of spore and mycelia extracts of Pisolithus tinctorius was carried out. The spores were collected from basidiocarps in plantations of Eucalyptus spp and the mycelia from culture in MNM medium. With basis on the fungus growth curve, the mycelia masses were obtained after 10, 20, 30, and 40 days of incubation, which correspond, respectively, to the beginning, middle and end of the log phase, and beginning of the decline phase. The mycelia masses, together with the spores, were submitted to the action of three extractors (Coca, Tris-HCl, and ammonium bicarbonate). The contents of carbohydrates and proteins were determined. The SDS-PAGE electrophoretical analysis revealed separate fractions in these extracts, besides common fractions, in function of cultivation time and extraction methods. The selected extracts for the allergic tests were the ones with the highest number of fractions. The prick-tests were conducted in 374 patients – rural workers, eucalyptus plantation works and college students. The positivity to the "prick test" with the antigenic extract of P. tinctorius was, respectively, 3.78%, 28.20% and 6.40%. Most prick-test positive patients (82.75%) also presented symptoms of respiratory allergy (asthma and rhinitis). There was no reactivity difference when the spore and mycelia extracts were employed. The analysis of the positive patients’ sera revealed the presence of IgE specific to the P. tinctorius antigens. Since Pisolithus tinctorius is found as mycorrhyza of Eucalyptus spp, and this plant is used in reforestation in most countries, the importance of that fungus should be regarded as a possible cause of respiratory allergies, especially in occupationally exposed workers
