Abstract Introduction: Currently, wherever you are in the world, the COVID-19 pandemic has had a global impact on every human being. This pandemic has had major consequences on the majority of the population, increasing feelings of instability, distrust and tension in access to health care. This study aimed to assess the level of satisfaction and the feeling of safety of clients who underwent endoscopic exams at a Gastroenterology unit during the second wave of the COVID-19 pandemic, as well as the impact of the measures taken to mitigate the influence of this circumstance on the experience of patients. Methods: A closed clinical interview made up of 10 questions focused on the feeling of safety and user satisfaction. Sixty individuals were interviewed, selected randomly, after using the Gastroenterology unit to perform endoscopic exams. Inclusion criteria were: age over 18, minimum education level, without cognitive or cerebral impairment. Results: The results have shown that, in general, users are satisfied with having done the exams (90%). However, some of the patients showed some concern before doing them (45%). Due to the equipment used in the context of the pandemic, some patients experienced difficulties in communicating with health professionals (63.3%). In spite of this, they felt confident and safe in carrying out the exams (66.9%), as well as safe in the hygienic conditions of the unit (58.3%). Conclusion: Despite the context of insecurity, tension and instability, the measures taken and the approach of the unit's professionals managed to transmit safety and tranquillity in the performance of the exams, enhancing a good level of patient satisfaction.

Resumo Introdução: Atualmente, em qualquer ponto do mundo, a pandemia pela COVID-19 não é indiferente a nenhum indivíduo. Esta pandemia teve um grande impacto na maioria da população, potenciando um sentimento de instabilidade, desconfiança e tensão no acesso aos cuidados de saúde. Este trabalho teve como objetivo avaliar o nível de satisfação e o sentimento de segurança de utentes que realizaram exames endoscópicos num serviço de Gastrenterologia durante a segunda vaga da pandemia por COVID-19, bem como o impacto das medidas instituídas para mitigar a influência dessa circunstância na experiência dos utentes. Métodos: Foi construída uma entrevista clínica fechada composta por 10 perguntas focadas no sentimento de segurança e satisfação dos utentes. Foram entrevistados 60 indivíduos, selecionados de forma aleatória, após o seu recurso ao serviço de Gastrenterologia para realizar exames endoscópicos. Os critérios de inclusão foram: maiores de 18 anos, nível de escolaridade mínima, sem comprometimento cognitivo ou cerebral. Resultados: Os resultados mostraram que, de uma forma geral, os utentes se encontram satisfeitos por terem feito os exames (90%), no entanto algumas destas pessoas demonstraram alguma inquietação antes de o fazerem (45%). Apesar de, devido aos equipamentos usados em contexto de pandemia, terem sentido algumas dificuldades na comunicação com os profissionais de saúde (63,3%), os utentes sentiram confiança e segurança na realização do exame (66,9 %), bem como nas condições de higiene da unidade (58,3%). Conclusão: Não obstante o contexto propício a sentimentos de insegurança, tensão e instabilidade, as medidas instituídas e a atitude dos profissionais da unidade conseguiram transmitir segurança e tranquilidade no desempenho dos exames, potenciando um bom nível de satisfação dos utentes.

Abstract Chagas disease is a neglected parasitic disease caused by Trypanosoma cruzi, whose treatment has remained unsatisfactory for over 50 years, given that it is limited to two drugs. Benznidazole (BZN) is an efficient antichagasic drug used as the first choice, although its poor water-solubility, irregular oral absorption, low efficacy in the chronic phase, and various associated adverse effects are limiting factors for treatment. Incorporating drugs with such characteristics into nanostructured lipid carriers (NLC) is a promising alternative to overcome these limiting obstacles, enhancing drug efficacy and bioavailability while reducing toxicity. Therefore, this study proposed NLC-BZN formulations in different compositions prepared by hot-melt homogenization followed by ultrasound, and the optimized formulation was characterized by FTIR, DRX, DSC, and thermogravimetry. Biological activities included in vitro membrane toxicity (red blood cells), fibroblast cell cytotoxicity, and trypanocidal activity against epimastigotes of the Colombian strain of T. cruzi. The optimized NLC-BZN had a small size (110 nm), negative zeta potential (-18.0 mV), and high encapsulation (1.64% of drug loading), as shown by infrared spectroscopy, X-ray diffraction, and thermal analysis. The NLC-BZN also promoted lower in vitro membrane toxicity (<3% hemolysis), and 50% cytotoxic concentration (CC50) for NLC-BZN in L929 fibroblast cells (110.7 µg/mL) was twice the value as the free BZN (51.3 µg/mL). Our findings showed that the NLC-BZN had higher trypanocidal activity than free BZN against the epimastigotes of the resistant Colombian strain, and this novel NLC-BZN formulation proved to be a promising tool in treating Chagas disease and considered suitable for oral and parenteral administration.

Abstract Scientific diving (SD) is defined as any diving activity that applies scientific procedures to produce subsidies for studies and technical works in underwater environments. The first report of an underwater scientific study in Brazil dates to the 19th century, in the Abrolhos reefs. Currently, in Brazil, scientific diving has been performed in various areas, from shallow coastal regions to remote and sometimes hard-to-reach places, such as oceanic islands, flooded caves, and icy areas like Antarctica. However, the regulation of SD in Brazil still lacks more concrete actions towards an effective and efficient self-regulation that offers physical safety to practitioners and institutional safeguards for organizations that use it in their research projects. Thus, this article aims to contribute to a better understanding of this critical issue in Brazil and to serve as a reference and incentive for the training of professionals and the development of these activities in the country. It includes: 1) a historical review of SD; 2) a diagnosis of the training and application of SD in Brazil; 3) the evolution of marine sciences in Brazil from the perspective of SD; 4) a review of the use of environmental assessment and underwater conservation techniques in oceans and internal waters; 5) an analysis of the evolution of scientific diver training in Brazil, including a diagnosis on training; 6) the history and updates of the rules, regulations, and safety of SD. Given all the potential of diving combined with specific techniques for research, monitoring, and marine and limnic science in Brazil, we aim to understand the evolution of scientific diving teaching and to outline perspectives in the country, as it is crucial for the training of qualified scientists capable of performing these underwater tasks. Finally, we present future plans for the development of this activity in Brazil from the point of view of research and the labor market.

Abstract The quality of the raw material for chocolate manufacture depends on the control of cacao seed fermentation. Practices affect the diversity and metabolic activities of spontaneous yeasts in fermentations. The control of the initial inoculum is important to standardize fermentations and to improve the final seed quality. We conducted fermentations using non-cleaned or cleaned fermentation boxes to evaluate the dynamics of the microorganism populations and their effect in seed composition in two consecutive years. The isolated yeasts were identified by sequencing the D1-domain of the 26S ribosomal genes. Pulp and cotyledon parameters were monitored during fermentations. Wickerhamomyces anomalus was the dominant yeast in the fermentations, but the species was less represented in cleaned boxes. In the cleaned-box fermentation, there was a delay in temperature rise of the fermentation, which retarded the suitable moment for seed-mass turning. The reduction in seed-pulp quality caused by restricted rainfall in one year limited the seed-mass heating and voided the seed turning. Thus, under substrate restriction, the initial reduced inoculum caused by cleaning the boxes impaired fermentation, mainly under environmental conditions that compromise the quality of the pulp. RESUMO Tumor triquilemal proliferante (TTP) é uma rara neoplasia que se desenvolve a partir de lesões císticas foliculares, cuja característica histológica é a presença de queratinização triquilemal. Manifestação mais comum é um nódulo solitário no couro cabeludo de mulheres idosas. Descreve-se caso de tumor triquilemal proliferante, que se apresenta como lesão tumoral cística em couro cabeludo de mulher idosa, com recidiva após remoção sem investigação diagnóstica.

ABSTRACT Proliferating trichilemmal tumor (PTT) is a rare neoplasm that develops from cystic follicular lesions, whose histological characteristic is the presence of trichilemmal keratinization. The most common manifestation is a solitary lump on the scalp of older women. It describes a case of proliferating trichilemmal tumor, which presents as a cystic tumor lesion on the scalp of an older woman, with recurrence after removal without diagnostic investigation.

RESUMO Introdução: pacientes submetidos a ressecção pulmonar podem apresentar complicações locais ou remotas no pós-operatório decorrente da resposta inflamatória, que aumenta o tempo de internação e de custos hospitalares. O objetivo deste trabalho foi estabelecer perfil ampliado do comportamento das interleucinas, identificar as principais interleucinas que atuam na resposta inflamatória no período pós-operatório e associá-las com dados clínicos e laboratoriais dos pacientes submetidos a ressecção pulmonar. Métodos: Estudo prospectivo com 27 casos de ressecção pulmonar realizados no HC-UNICAMP. Foram analisados níveis séricos de IL-1 α, IL-1 β, IL-1 ra, IL-2, IL-13, IL-6, IL-8, IL-10, IL-12 (p40), IL-12 (p70), IL-17a, TNF- α, TNF- β, IFN-γ, TGF- β, MIP-1 α, MIP-1 β, MCP-1, MCP-3, VEGF e dados clínicos antes, durante e após a operação. Resultados: indivíduos apresentaram mediana de idade de 63 anos, sendo 16 (59%) do sexo masculino e 11 (41%) do sexo feminino. Os fatores clínicos que influenciam na resposta inflamatória são: índice de massa corporal, tabagismo e uso prévio de corticóide, enquanto que os dados laboratoriais se expressam nos números de leucócitos e plaquetas. Discussão: dentro deste ampliado perfil das interleucinas na resposta inflamatória das ressecções pulmonares, este estudo mostrou que devem ser valorizadas para avaliar inflamação humoral as interleucinas: IL-6, IL-8, IL-10, IL-1 β e TNF- α. Conclusão: este estudo pode colaborar na identificação de intervenções clínicas ou farmacológicas que modulem a resposta inflamatória no período peri-operatório das ressecções pulmonares, mitigando as complicações locais e sistêmicas.

ABSTRACT Introduction: patients undergoing pulmonary resection may experience local or remote complications in the postoperative period due to the inflammatory response, which increases the length of hospital stay and costs. This study objective was to establish an expanded interleukins profile, identifying the main actors in the postoperative inflammatory response, and to correlate them with clinical and laboratory data of patients submitted to pulmonary resection. Methods: this was a prospective, interventional, longitudinal study of 27 cases of pulmonary resection performed at HC-UNICAMP, in which we analyzed serum levels of IL 1 α, IL 1 β, IL 1 ra, IL 2, IL 13, IL 6, IL 8, IL 10, IL 12 (p40), IL 12 (p70), IL 17a, TNF α, TNF β, IFN γ, TGF β, MIP 1α, MIP 1β, MCP 1, MCP 3, VEGF, and clinical data before, during, and after surgery. Results: Individuals had a median age of 63 years, 16 (59%) being male and 11 (41%), female. The clinical factors that influenced inflammatory response were body mass index, smoking, and previous use of corticosteroids, while the influencing laboratory data were the numbers of leukocytes and platelets. Discussion: within this expanded interleukin profile in the inflammatory response of lung resections, our study showed that interleukins IL 6, IL 8, IL 10, IL 1 β, and TNF α should be considered for assessing humoral inflammation. Conclusion: this study can aid in the identification of clinical or pharmacological interventions that modulate the inflammatory response in the perioperative period of pulmonary resections, mitigating local and systemic complications.

RESUMO: O presente estudo foi conduzido para investigar a soroprevalência em 20 fazendas de criação extensiva de ovinos quanto à presença de anticorpos para Corynebacterium pseudotuberculosis (n=402) e Toxoplasma gondii (n=228). Ensaio de imunoabsorção enzimático (ELISA) foi utilizado para a detecção de anticorpos contra C. pseudotuberculosis/T. gondii. Observou-se que C. pseudotuberculosis apresentou a maior prevalência na região (34,07%), com presença estatisticamente significante (p <0,05) nas categorias de ovelha. Anticorpos contra T. gondii foram encontrados em 14,91% dos animais estudados. Sobre a coinfecção de C. pseudotuberculosis/T. gondii, as categorias carneiro apresentaram diferenças significativas (p <0,05), sugerindo que esse gênero poderia perpetuar as doenças nos rebanhos. Concluiu-se que o conhecimento sobre as doenças na região em estudo facilitaria a execução de medidas profiláticas, principalmente contra as doenças que apresentam riscos à saúde pública e causam danos ao produtor.

ABSTRACT: The present study was conducted to investigate in 20 extensive sheep farms for the seroprevalence of Corynebacterium pseudotuberculosis (n=402) and Toxoplasma gondii (n=228). Enzyme-linked immunosorbent assay (ELISA) was used for the detection of antibodies to C. pseudotuberculosis/T. gondii. It was observed that C. pseudotuberculosis showed the highest prevalence in the region (34.07%) with statistically significant presence (p<0.05) in ewes. Antibodies to T. gondii was reported in 14.91% of the animals studied. About C. pseudotuberculosis/T. gondii coinfection the categories of rams showed significant (p<0.05) differences, suggesting that this gender could perpetuate the diseases in the flocks. It was concluded that the knowledge about the diseases in the region under study would facilitate the execution of prophylactic measures, especially against the diseases that pose risks to the public health and cause damages to the producer.

Abstract OBJECTIVE To describe the thermoregulation protocol implementation for newborns (NB). METHODS An experimental report, conducted at a neonatal unit in Salvador, Bahia, from January 2016 to January 2017. The Plan, Do, Check, Action cycle guided the construction, implementation and applicability of the protocol. RESULTS Implementation of the protocol that allowed the reduction of adverse events due to thermal instability during surgical procedures and introduction of new technologies. CONCLUSION The protocol could improve and strengthen the care practices related to safe surgery in newborns.

Resumen OBJETIVO Describir la implementación del protocolo de termorregulación para procedimientos quirúrgicos en recién nacido (RN). MÉTODOS Relato de experiencia, realizado en una unidad neonatal en Salvador-BA, en el período de enero de 2016 a enero de 2017. El ciclo Plan, Do, Check, Action orientó la construcción, la implementación y la aplicabilidad del protocolo. RESULTADOS Se pudo implementar el protocolo que permitió reducir eventos adversos por inestabilidad térmica durante procedimientos quirúrgicos y se introdujo nuevas tecnologías. CONCLUSIÓN El protocolo permitió una mejora y un fortalecimiento de las prácticas asistenciales, relacionadas con la cirugía segura en RN.

Resumo OBJETIVO Descrever a implantação do protocolo de termorregulação para procedimentos cirúrgicos em recém-nascido (RN). MÉTODOS Relato de experiência, realizado em uma unidade neonatal em Salvador-BA, no período de janeiro de 2016 a janeiro 2017. O ciclo Plan, Do, Check, Action norteou a construção, a implantação e a aplicabilidade do protocolo. RESULTADOS Implantação do protocolo que possibilitou a redução de eventos adversos por instabilidade térmica durante procedimentos cirúrgicos e introdução de novas tecnologias. CONCLUSÃO O protocolo possibilitou a melhoria e o fortalecimento das práticas assistenciais relacionadas com a cirurgia segura em RN.

RESUMO A teca é uma espécie florestal que assumiu grande importância no Brasil, onde encontrou excelentes condições de desenvolvimento desde a sua introdução na década de 1960. Entretanto, problemas fitossanitários começam a ameaçar a exploração desta espécie madeireira. Um exemplo é o cancro da teca, causado pelo fungo Lasiodiplodia theobromae (Lt), cuja etiologia foi recentemente elucidada no Brasil e, portanto, ainda não existem métodos de controle recomendados. Sendo assim, o objetivo deste trabalho foi avaliar o controle deste patógeno, investigando o potencial dos agentes de controle biológico (ACB) Trichoderma spp., Bacillus sp. e Enterobacter sp., inicialmente por meio de ensaios in vitro e, posteriormente, com realização de testes in vivo. De acordo com os resultados dos testes in vitro, os isolados de Trichoderma CEN162 e CEN1153 e a estirpe UnB1366 de Bacillus sp., foram os tratamentos que se destacaram, sendo capazes de inibir completamente o crescimento micelial de alguns isolados de Lt. Quando esses isolados foram testados de forma preventiva, observou-se variação nos níveis de controle, dependendo do isolado de Lt e da interação antagonista-clone, onde CEN162 (T. asperellum) e UnB166 (Bacillus sp.) apresentaram 100% de controle. Dessa forma, houve uma correlação positiva entre os testes in vitro e in vivo, uma vez que os mesmos ACB se destacaram. Apesar de ter-se obtido bons níveis de controle com os ACB utilizados, pode-se concluir que existe uma variação no antagonismo a diferentes isolados ou mesmo na interação antagonista-clone, corroborando as informações disponíveis na literatura científica sobre este fungo fitopatogênico.

ABSTRACT Teak is a forest species that has assumed great importance in Brazil, where it has found excellent conditions for development since its introduction into the country in the 1960s. However, phytosanitary problems are beginning to threaten the production of this timber species. An example is teak canker, caused by the fungus Lasiodiplodia theobromae (Lt), which has only recently been reported in Brazil, and for which, therefore, there are no recommended control methods. Thus, this study evaluated the control of this pathogen, investigating the potential of the biocontrol agents (BCAs) Trichoderma spp., Bacillus sp. and Enterobacter sp., initially through in vitro assays and, subsequently, with in vivo tests. According to the in vitro assay results, the Trichoderma isolates CEN162 and CEN1153 and the strain of Bacillus sp. (UnB1366) were the treatments that stood out, as they were able to completely inhibit mycelial growth of some isolates of Lt. When these isolates were tested in a preventive way, the control levels varied depending on the Lt isolate and the antagonist-clone interaction, where CEN162 (T. asperellum) and UnB166 (Bacillus sp.) showed 100% control. Thus, there is a positive correlation between the in vitro and in vivo tests, since the same BCAs stood out. Although good levels of control have been obtained with the BCAs used, it can be concluded that there is a variation in the antagonism to different Lt isolates or even in the antagonist-clone interaction, corroborating the information available in the scientific literature on this plant-pathogenic fungus.

Resumo Antibiose é um mecanismo pelo qual certos microrganismos respondem à presença de outros, secretando compostos ou metabólitos capazes de inibir ou impedir o seu desenvolvimento. O extrato bruto de Trichoderma contém uma mistura de compostos secundários e tem sido utilizado na prospecção deste fungo para o controle biológico e demais fins industriais. Diante da crescente demanda da agricultura por alternativas ecologicamente compatíveis para o manejo de doenças, este trabalho teve como objetivo investigar o espectro de ação de Metabólitos Não Voláteis (MNVs), produzidos por isolados de Trichoderma, contra diferentes fungos fitopatogênicos. O potencial antagônico dos MNVs foi avaliado através do método de incorporação do extrato líquido filtrado em meio BDA. Os ensaios mostraram que todos os MNVs produzidos inibiram de forma semelhante o fungo Sclerotinia sclerotiorum. Por outro lado, os isolados CEN1245 e CEN1274, ambos Trichoderma brevicompactum, mostraram um amplo espectro de ação, atuando contra Sclerotium rolfsii, Colletotrichum gloesporioides, Verticillium dahliae, Fusarium oxysporum e Cylindrocladium sp. O presente estudo descreve isolados que produziram metabólitos não voláteis com amplo espectro de ação antifúngico, assim como patógeno-específico. Os MNVs de Trichoderma spp. obtidos de diferentes amostras de solo cultivadas com vegetais, mandioca e milho, foram eficientes na inibição de fungos fitopatogênicos pertencentes a outros patossistemas, como os de espécies florestais e frutíferas, o que poderia aumentar sua potencial aplicação no controle de doenças de plantas. Adicionalmente, estes fungos antagonistas devem ser mais bem estudados para identificação de moléculas bioativas de interesse industrial ou formulação de produtos para o controle biológico de patógenos de plantas.

Abstract Antibiosis is the mechanism by which certain microorganisms respond to the presence of others, secreting compounds or metabolites capable of inhibiting or impeding their development. The crude extract of Trichoderma contains a mixture of secondary compounds, which may show antibiotic effect, and has been used for the prospect of this fungus for biological control and other industrial purposes. Faced with the increasing demand of agriculture for ecologically compatible alternatives for the management of diseases, this work aimed to investigate the spectrum of action of Non-Volatile Metabolites (NVMs) of Trichoderma isolates against different plant pathogenic fungi. The antagonistic potential of NVMs was evaluated through the incorporation method of the filtered liquid extract in PDA medium. The assays showed that all the NVMs produced inhibited the fungus Sclerotinia sclerotiorum similarly. On the other hand, strains CEN1245 and CEN1274, both belonging to the species Trichoderma brevicompactum, showed broad spectrum against Sclerotium rolfsii, Colletotrichum gloesporioides, Verticillium dahliae, Fusarium oxysporum and Cylindrocladium sp. The present study describes isolates producing non-volatile metabolites with broad spectrum of antifungal action, as well as pathogen-specific. The Trichoderma spp. NVMs obtained from different soil samples cultivated with vegetables, cassava and maize were efficient in inhibiting plant pathogenic fungi belonging to other patossystems, such as forest or fruit, which could increase their potential application in biological control of plant diseases. In addition, these antagonistic fungi should be studied in greater depth for the identification of bioactive molecules of industrial interest or in commercial formulations of products for biological control of plant pathogens.

ABSTRACT To report a case of tuberous breast with significant breast asymmetry, describe the technique used and evaluate the outcome of the case. Different techniques were used on each breast, although studies in literature recommend the use of similar strategy in both breasts. The patient progressed without complications and had, 6 months after the surgery, extremely satisfactory result without tuberous breast stigmas, and significant improvement of breast asymmetry. The literature establishes that not only one surgical technique is adequate to correct different types of malformations. Tuberous breast constitutes a challenge in breast plastic surgery and it becomes more complex when the asymmetry is more severe. However, surgeons who is trained in a variety of aesthetic and breast reconstructive techniques can achieve a satisfactory result.

RESUMO Descrever um caso de mama tuberosa com importante assimetria mamária, descrever a técnica e avaliar o resultado de caso. Diferentes técnicas foram usadas em cada mama, apesar de na literatura dizer que é importante empregar uma estratégia semelhante em ambas as mamas. A paciente evoluiu sem complicações e apresenta, com 6 meses de pós-operatório, resultado extremamente satisfatório, sem estigmas de mama tuberosa e com melhora importante da assimetria mamária. Na literatura é estabelecido que não há uma única técnica cirúrgica adequada para a correção dos diferentes tipos de malformações. A mama tuberosa representa um desafio nas cirurgias plásticas da mama e se tornam mais complexas quando a assimetria é mais exacerbada. Porém, o cirurgião que dominar as mais diversas técnicas mamárias estéticas e reconstrutoras poderá alcançar um resultado satisfatório.

RESUMO Objetivo: avaliar a intensidade de sudorese reflexa com o grau de ansiedade e sua interferência na qualidade de vida de indivíduos submetidos à simpatectomia por videotoracoscopia nos períodos pré e pós-operatório. Métodos: foram avaliados 54 pacientes com média de idade de 26 anos (16 a 49 anos), submetidos à simpatectomia em nível R3-R4. Dois questionários foram aplicados em três momentos diferentes: "Qualidade de vida em pacientes com hiperidrose primária e "Escala para ansiedade e depressão". Resultados: dos pacientes estudados, 93% mostrou melhora significativa na qualidade de vida após 30 dias da cirurgia, com os efeitos remanescentes após seis meses. Não houve complicações pós-operatórias. A análise mostrou que o nível de ansiedade do paciente é altamente correlacionado com a intensidade da sudorese reflexa após 30 e 180 dias. Conclusão: a simpatectomia torácica por videotoracoscopia melhora a qualidade de vida de pacientes com hiperidrose primária, mesmo com o surgimento de sudorese reflexa. A ansiedade está diretamente relacionada com a intensidade da sudorese reflexa, sem comprometer o grau de satisfação do paciente.

ABSTRACT Objective: to compare the intensity of reflex sweating with the degree of anxiety and its interference in the quality of life of patients undergoing Thoracoscopic (VATS) sympathectomy in the pre- and postoperative period. Methods: we evaluated 54 patients with a mean age of 26 years (16-49 years) undergoing sympathectomy in the R3-R4 level. We applied two questionnaires at three different times: "Quality of life in patients with primary hyperhidrosis" and "Scale for anxiety and depression". Results: of the patients studied, 93% showed significant improvement in quality of life 30 days after surgery, the effects remaining after six months. There were no postoperative complications. The patient's level of anxiety is highly correlated with the intensity of reflex sweating after 30 and 180 days. Conclusion: Thoracoscopic sympathectomy improves quality of life of patients with primary hyperhidrosis, even with the emergence of reflex sweating. Anxiety directly relates to the intensity of reflex sweating, without compromising the degree of patient satisfaction.

Resumo Quarenta e nove isolados de Trichoderma obtidos no centro-oeste do Brasil foram avaliados quanto a sua atividade antagônica in vitro contra Sclerotinia sclerotiorum (agente causal do mofo branco) e identificados com base nas sequências ITS do DNA ribossômico nuclear. Os testes de cultivo pareado mostram que todos os isolados exibiram algum antagonismo, com um máximo de 77% de inibiação micelial e inibição total da produção de escleródios. Dois isolados se destacaram como os mais promissores, considerando ambos os parâmetros avaliados (CEN1253 - T. koningiopsis e CEN1265 - T. brevicompactum). Cinco espécies diferentes foram identificadas: T. harzianum (23), T. spirale (9), T. koningiopsis (8), T. brevicompactum (7) and T. asperellum (2). Estes isolados estão armazenados na Coleção de Fungos para Controle Biológico da Embrapa e as informações obtidas nos experimentos serão incorporadas na base de dados de ativos biológicos, no sistema de informações de recursos genéticos, e disponibilizados para estudos futuros.

Abstract Forty-nine isolates of Trichoderma from the Brazilian Midwest were evaluated for their antagonistic activity in vitro against Sclerotinia sclerotiorum (causal agent of white mold), which were then identified based on their nuclear ribosomal ITS sequences. Paired culture tests showed that all isolates exhibited some antagonism, with a maximum of 77% mycelial inhibition and complete inhibition of sclerotia production. Two isolates were found to be the most promising biocontrol agents, considering both antagonistic parameters (CEN1253 - T. koningiopsis and CEN1265 - T. brevicompactum). Five different species were identified: T. harzianum (23), T. spirale (9), T. koningiopsis (8), T. brevicompactum (7) and T. asperellum (2). These isolates are stored in the Embrapa Fungi Collection for Biological Control and the information obtained in the experiments will be incorporated into the database of biological assets within the genetic resources information system (Allele) and be made available for further studies.

OBJECTIVE: To evaluate hematological, cytogenetic and molecular responses as well as the overall, progression-free and event-free survivals of chronic myeloid leukemia patients treated with a third tyrosine kinase inhibitor after failing to respond to imatinib and nilotinib/dasatinib. METHODS: Bone marrow karyotyping and real-time quantitative polymerase chain reaction were performed at baseline and at 3, 6, 12 and 18 months after the initiation of treatment with a third tyrosine kinase inhibitor. Hematologic, cytogenetic and molecular responses were defined according to the European LeukemiaNet recommendations. BCR-ABL1 mutations were analyzed by Sanger sequencing. RESULTS: We evaluated 25 chronic myeloid leukemia patients who had been previously treated with imatinib and a second tyrosine kinase inhibitor. Nine patients were switched to dasatinib, and 16 patients were switched to nilotinib as a third-line therapy. Of the chronic phase patients (n=18), 89% achieved a complete hematologic response, 13% achieved a complete cytogenetic response and 24% achieved a major molecular response. The following BCR-ABL1 mutations were detected in 6/14 (43%) chronic phase patients: E255V, Y253H, M244V, F317L (2) and F359V. M351T mutation was found in one patient in the accelerated phase of the disease. The five-year overall, progression-free and event-free survivals were 86, 54 and 22% (p<0.0001), respectively, for chronic phase patients and 66%, 66% and 0% (p<0.0001), respectively, for accelerated phase patients. All blast crisis patients died within 6 months of treatment. Fifty-six percent of the chronic phase patients lost their hematologic response within a median of 23 months. CONCLUSIONS: Although the responses achieved by the third tyrosine kinase inhibitor were not sustainable, a third tyrosine kinase inhibitor may be an option for improving patient status until a donor becomes available for transplant. Because the long-term outcome for these patients is poor, the development of new therapies for resistant chronic myeloid leukemia patients is necessary.
