Chemical Characterization of Aspartic Protease from Artichoke Flower (Cynara cardunculus L. var scolymus) Using MIR Spectroscopy and Analytical Measurements

Vegetable proteases have been studied as milk coagulants, and artichoke flower (Cynara cardunculus L. var scolymus) has potential coagulant action as a substitute for microbial chymosin. The objective was to perform chemical characterization of aspartic proteases from artichoke flower (Cynara cardunculus L. var scolymus) by mid-infrared spectroscopy (MIR) and analytical measurements. Artichoke flower extracts were obtained and crude thistle flower extract and microbial chymosin were used as a reference. Plant extracts and microbial chymosin were analyzed for protein concentration, proteolytic activity (PA), milk clotting activity (MCA), specificity ratio (SR), effects of pH, temperature, NaCl and CaCl2 concentration on MCA and PA and characterized by electrophoretic, spectroscopic, chromatographic and storage stability. The results indicated that the crude extract of artichoke flower showed high MCA (510.08 SU mL 1), low PA (10.50 µg mg-1), SR (48.57) and storage stability for up to 90 days under frozen. The electrophoretic profile of artichoke flower resulted in a protein band with an apparent molecular weight of 32 kDa, associated with cardosin A. Artichoke flower may be a new alternative for commercial plant proteases with active enzymes for milk coagulation.

RESUMO Plantas cultivadas sob salinidade estão sujeitas a muitos distúrbios morfológicos e fisiológicos. Neste sentido, práticas que possam viabilizar o uso de águas salinas são essenciais. Os efeitos deletérios da salinidade podem ser mitigados ao utilizar microrganismos benéficos, especialmente bactérias diazotróficas. Nesse contexto, objetivou-se avaliar o efeito da inoculação e co-inoculação bacteriana utilizando Bradyrhizobium spp. e Azospirillum brasilense no crescimento, nodulação e produção de feijão-caupi sob estresse salino. O experimento foi conduzido em blocos casualizados, em arranjo fatorial 4 × 5, sendo quatro fontes de nitrogênio (F1 - sem nitrogênio e sem inoculante; F2 - adubação com nitrogênio mineral e sem inoculante; F3 - inoculação de Bradyrhizobium ssp. e F4 - coinoculação de Bradyrhizobium ssp. e Azospirillum brasilense) e cinco valores de condutividade elétrica da água de irrigação - CEa (0,4; 1,9; 3,4; 4,9 e 6,4 dS m-1), com cinco repetições. A coinoculação com Azospirillum brasilense e Bradyrhizobium favoreceu o crescimento, a produção e a nodulação de plantas de feijão caupi quando irrigadas até CEa de 0,4 dS m-1. O aumento da condutividade elétrica da água de irrigação afetou negativamente o crescimento, a produção e a nodulação de plantas de feijão Caupi da variedade Corujinha independentemente da fonte de nitrogênio utilizada.
ABSTRACT Plants grown under salinity are subject to many morphological and physiological disorders. In this context, practices that can enable the use of saline water are essential. The deleterious effects of salinity can be mitigated by using beneficial microorganisms, especially diazotrophic bacteria. In this context, this study aimed to evaluate the effect of bacterial inoculation and co-inoculation using Bradyrhizobium spp. and Azospirillum brasilense on the growth, nodulation and production of cowpea under salt stress. The experiment was conducted using a randomized block design, in a 4 × 5 factorial arrangement, corresponding to four nitrogen sources (N1 - without nitrogen and without inoculant; N2 - fertilization with mineral nitrogen and without inoculant; N3 - inoculation of Bradyrhizobium spp.; and N4 - co-inoculation of Bradyrhizobium spp. and Azospirillum brasilense) and five values of electrical conductivity of irrigation water - ECw (0.4, 1.9, 3.4, 4.9 and 6.4 dS m-1), with five replicates. Co-inoculation with Azospirillum brasilense and Bradyrhizobium spp. favors the growth, production and nodulation of cowpea plants up to ECw of 0.4 dS m-1. Increase in the electrical conductivity of irrigation water negatively affects the growth, production and nodulation of cowpea plants in Corujinha variety, regardless of the nitrogen source used.

Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development.

Amphibians of Serra das Torres Natural Monument: a reservoir of biodiversity in the Atlantic Forest of southeastern Brazil

Resumo A Mata Atlântica brasileira guarda importante porção da riqueza de anfíbios e altas taxas de endemismos. Neste estudo, nós realizamos pesquisas usando o Método de Avaliação Rápida (RA) com o objetivo de inventariar a fauna de anfíbios de um remanescente da Mata Atlântica no sudeste do Brasil, o Monumento Natural Serra das Torres (MONAST). Amostramos ativamente com uma equipe de 6 a 10 pessoas para coletar amostras padronizadas entre 09:00 e 12:00 horas durante o peíodo diurno e entre 18:00 e 22:00 duranto período crepuscular/noturno, totalizando aproximadamente 1320 horas de esforço amostral. Complementamos estes dados com 720 horas de amostragem passiva usando armadilhas de queda com cercas-guia (30 dias de balde). Registramos 54 espécies de anfíbios (dois gimnofionos e 52 anuros) e a riqueza de espécies estimada pelo Bootstrap indicou um número relativamente maior de espécies (n = 60). A família mais especiosa foi Hylidae (n = 21), seguida por Brachycephalidae (n = 8). No geral, 25% das espécies (n = 13) foram registradas apenas uma vez - singletons e 15% (n = 8) apenas duas vezes (doubletons). A maioria dos anfíbios registrados neste estudo (71%, n = 37 espécies) esteve restrita ao bioma Mata Atlântica, duas espécies (Euparkerella robusta e Luetkenotyphlus fredi) são endêmicas do estado do Espírito Santo, sendo uma delas, a espécie de serapilheira E. robusta, endêmica do MONAST. Euparkerella robusta está atualmente listada como Vulnerável pela IUCN e classificada como Criticamente Ameaçada na lista vermelha do estado do Espírito Santo, enquanto L. fredi ainda não foi avaliada devido a sua descrição ser muito recente. Thoropa lutzi está atualmente listada como Ameaçada (EN) pela IUCN e na lista estadual. Nove espécies estão listadas como Deficiente de Dados (DD) e as populações de 13 espécies são consideradas em declínio pela IUCN. Estendemos a distribuição geográfica de duas espécies de anuros (Hylodes babax e Phasmahyla lisbella) e preenchemos uma importante lacuna na distribuição de Siphonops hardyi. Os anfíbios associados ao chão da floresta representaram 42% das espécies do MONAST e 43% destas espécies habitavam exclusivamente a serapilheira. Nosso estudo revelou que a Serra das Torres preserva diversidade considerável de anfíbios da Mata Atlântica, o que reforça a necessidade de conservação desse remanescente florestal.
Abstract The Brazilian Atlantic Forest holds a major part of the country’s amphibian species richness and high rates of endemism. In this study, we conducted surveys using the Rapid Assessment (RA) method to sample the amphibian fauna of the Serra das Torres Natural Monument (MONAST), an Atlantic Forest remnant in southeastern Brazil. We sampled actively with a 6-10-person team to collect standard samples from 09:00 to 12:00 hours for the daytime period, and from 18:00 to 22:00 hours for the crepuscular/nighttime period, with a total of approximately 1,320 hours of sampling effort. We supplemented these data with 720 hours of passive sampling, using pitfall traps with drift fences (30 bucket-days). We recorded 54 amphibian species (two gymnophionans and 52 anurans), and the species richness estimated by the Bootstrap method indicates that a slightly larger number of species (n = 60) may occur in the study area. The most speciose family was Hylidae (n = 21), followed by Brachycephalidae (n = 8). Overall, 25% of the species (n = 13) were recorded only once (singletons) and 15% (n = 8) only twice (doubletons). Most amphibians recorded in this study (71%, n = 37 species) were restricted to the Atlantic Forest biome, two species (Euparkerella robusta and Luetkenotyphlus fredi) are endemic to the Espírito Santo state, and one of them, the leaf litter species E. robusta, is endemic to the MONAST. Euparkerella robusta is currently listed as Vulnerable by the IUCN and is classified as Critically Endangered in the Espírito Santo State red list, while L. fredi has yet to be evaluated due to its recent description. Thoropa lutzi is currently listed as Endangered (EN) by both the IUCN and in the State list. Nine species are listed as Data Deficient (DD) and populations of 13 species are considered to be declining by the IUCN. We extend the geographical distribution of two anuran species (Hylodes babax and Phasmahyla lisbella) and fill an important gap in the distribution of Siphonops hardyi. Amphibians associated with the forest floor represented 42% of the species richness from MONAST, and 43% of these species inhabit the leaf litter exclusively. Our study revealed that Serra das Torres preserves a considerable diversity of Atlantic Forest amphibians, which reinforces the need for the conservation of this forest remnant.

Bioactivity and molecular properties of Phenoxyacetic Acids Derived from Eugenol and Guaiacol compared to the herbicide 2,4-D

Abstract Herbicides are agrochemicals applied in the control of weeds. With the frequent and repetitive use of these substances, serious problems have been reported. Compounds of natural origin and their derivatives are attractive options to obtain new compounds with herbicidal properties. By aiming to develop compounds with potentiated herbicidal activity, phenoxyacetic acids were synthesized from eugenol and guaiacol. The synthesized compounds were characterized and the herbicidal potential of phenoxyacetic acids and precursors was evaluated through bioassays regarding the germination and initial development of Lactuca sativa and Sorghum bicolor seedlings, with the induction of DNA damage. The induction of changes in the mitotic cycle of meristematic cells of roots of L. sativa was also analyzed. At the concentration of 3 mmol L-1, phenols and their respective phenoxyacetic acids presented phytotoxic and cytotoxic activities in L. sativa and S. bicolor. Eugenol and guaiacol also presented genotoxic action in L. sativa. The toxic effect of eugenoxyacetic acid was more pronounced in L. sativa than in S. bicolor, similar to the commercial 2,4-D herbicide. Molecular properties of the phenols and their derivatives phenoxyacetic acids were compared with the ones obtained for the herbicide 2,4-D, where it was found a correlation between their molecular properties and bioactivity.

Resumo Fundamento Eventos isquêmicos recorrentes decorrem de instabilidade de placa aterosclerótica, enquanto morte após um evento isquêmico decorre da gravidade do insulto. A natureza diversa desses tipos de eventos pode fazer com que dados clínicos e anatômicos tenham diferentes capacidades prognósticas a depender do tipo de desfecho. Objetivo Identificar as predileções prognósticas de dados clínicos e dados anatômicos em relação a desfechos coronários fatais e não fatais durante hospitalização de pacientes com síndromes coronarianas agudas (SCA). Métodos Pacientes consecutivamente admitidos por SCA que realizaram coronariografia foram recrutados. O escore SYNTAX foi utilizado como modelo anatômico e o escore GRACE como modelo clínico. A capacidade preditora desses escores foi comparada quando à predição de desfechos isquêmicos não fatais (infarto ou angina refratária) e de morte cardiovascular durante hospitalização. Significância estatística foi definida por p < 0,05. Resultados Entre 365 indivíduos, 4,4% foi a incidência de óbito hospitalar e 11% de desfechos isquêmicos não fatais. Para morte cardiovascular, ambos os escores — SYNTAX e GRACE — apresentaram capacidade discriminatória, com estatísticas-C similares: 0,80 (95%IC: 0,70–0,92) e 0,89 (95%IC 0,81–0,96), respectivamente — p=0,19. Quantos aos desfechos isquêmicos não fatais, o escore SYNTAX apresentou valor preditor (estatística-C = 0,64; 95%IC 0,55–0,73), porém o escore GRACE não mostrou associação com esse tipo de desfecho (estatística-C = 0,50; 95%IC: 0,40–0,61) — p=0,027. Conclusão Os modelos clínico e anatômico predizem satisfatoriamente morte cardiovascular em SCA, enquanto a recorrência de instabilidade coronária é melhor prevista por características anatômicas do que por dados clínicos. Abstract Background Recurrent ischemic events are mediated by atherosclerotic plaque instability, whereas death after an ischemic event results from gravity of insult and ability of the organism to adapt. The distinct nature of those types of events may respond for different prediction properties of clinical and anatomical information regarding type of outcome. Objective To identify prognostic properties of clinical and anatomical data in respect of fatal and non-fatal outcomes of patients hospitalized with acute coronary syndromes (ACS). Methods Patients consecutively admitted with ACS who underwent coronary angiography were recruited. The SYNTAX score was utilized as an anatomic model and the GRACE score as a clinical model. The predictive capacity of those scores was separately evaluated for prediction of non-fatal ischemic outcomes (infarction and refractory angina) and cardiovascular death during hospitalization. It was considered as significant a p-value <0,05. Results EAmong 365 people, cardiovascular death was observed in 4,4% and incidence of non-fatal ischemic outcomes in 11%. For cardiovascular death, SYNTAX and GRACE score presented similar C-statistic of 0,80 (95% IC: 0,70 – 0,92) and 0,89 (95% IC 0,81 – 0,96), respectively – p = 0,19. As for non-fatal ischemic outcomes, the SYNTAX score presented a moderate predictive value (C-statistic = 0,64; 95%IC 0,55 – 0,73), whereas the GRACE score did not presented association with this type of outcome (C-statistic = 0,50; 95%IC 0,40-0,61) – p = 0,027. Conclusion Clinical and anatomic models similarly predict cardiovascular death in ACS. However, recurrence of coronary instability is better predicted by anatomic variables than clinical data. We reported experimental and theoretical investigation of conformers of 1,2,3-triazole derivatives, substances of exclusively synthetic origin, subject of extensive studies, because of several biological properties, such as antiviral, antimicrobial and antileishmaniasis. We reported molecular/supramolecular X-ray structures of antiophidian compounds I and II. For I and II there are two crystallographic different molecules in the unit cell (A and B). To explore the causes of the similarities in the compound’s crystal structures, intermolecular interactions were explored using the Hirshfeld surface as the fingerprint plots. In addition, density functional theory (DFT) calculations were carried out at the ωB97x-D/6-31G(d,p)-PCM-CHCl3 level aiming to contribute to the interpretation of the experimental data and complement the experimental findings. Two structures named 2A and 5B were found in good agreement with the respective X-ray solid state ones (A and B). Theoretical 1H nuclear magnetic resonance (NMR) spectra calculated for 5B rotated structure (torsion angles deviation around 40° to 90°) was in fine agreement with experimental results (in CDCl3) indicating that the solution molecular structure is considerably different from optimized equilibrium geometries and solid-state structure. Therefore, care is needed when using X-ray structures or DFT geometries to model interaction of drugs with biological targets since significant conformational changes may take place in solution.

Resumo: O conhecimento das assembleias de répteis para muitos ecossistemas no Brasil pode ser considerado ainda relativamente restrito. Neste estudo, nós realizamos amostragens no Monumento Natural Serra das Torres, localizado nos municípios de Atílio Vivacqua, Muqui e Mimoso do Sul, no estado do Espírito Santo, utilizando o método de avaliação rápida (RAP) durante 30 dias na estação chuvosa de 2018. Amostramos cerca de 1320 horas de busca ativa durante os períodos diurno e noturno, com uma equipe de 6 a 10 pessoas, suplementada por 720 horas de amostragem com armadilhas de queda com cercas guia (30 dias-balde). Registramos 34 espécies de répteis squamatas durante as amostragens (2 anfisbenídeo, 11 lagartos e 21 serpentes) e um encontro ocasional posterior que acrescentou uma espécie de quelônio à lista, Hydromedusa maximiliani, totalizando 35 espécies de répteis. Dipsadidae foi a família com a maior riqueza de serpentes, e Gymnophtalmidae foi a família com maior riqueza de lagartos. A riqueza de espécies que registramos no Monumento Natural Serra das Torres (Ntotal = 35) representa ca. 27% de todas as espécies de répteis encontradas no estado do Espírito Santo (N = 130). A espécie de lagarto mais abundante foi Leposoma scincoides seguido por Ecpleopus gaudichaudii, enquanto a espécie mais abundante de serpente foi a Bothrops jararaca, sendo marcadamente maior do aquela registradas em estudos similares. Vinte e sete por cento das espécies de registradas em nosso estudo são endêmicas da Mata Atlântica e trinta por cento das espécies (N = 10) tinham menos de cinco indivíduos registrados anteriormente no estado do Espírito Santo. Nosso estudo reforça a necessidade de conservação do Monumento Natural Serra das Torres devido à sua importância como reservatório de uma considerável parcela da biodiversidade de répteis do estado do Espírito Santo, bem como do bioma Mata Atlântica.
Abstract: Data on the composition of local reptile assemblages in several Brazilian ecosystems can still be considered relatively restricted in scope in most cases. In this study, we conducted surveys in the Serra das Torres Natural Monument, located in the municipalities of Atílio Vivacqua, Muqui, and Mimoso do Sul, using the Rapid Assessments method (RAP) during 30 days in the rainy season of 2018. We sampled actively for approximately 1320 hours with a 6-10 person crew, supplemented by 720 hours of passive sampling (30 bucket-days) using pitfall traps with drift fences. We recorded 34 reptile species during our sampling method (2 amphisbaenid, 11 lizards, and 21 snakes) and an occasional encounter, after the end of sampling, that added a chelonian species to the list, Hydromedusa maximiliani, totaling 35 reptile species. The Dipsadidae was the family with the greatest snake species richness and, the Gymnophtalmidae had the greatest lizard species richness. The species richness recorded in the Serra das Torres Natural Monument (Ntotal = 35) represents ca. 27% of all reptile species found in the state of Espírito Santo (N = 130). The most abundant lizard species was Leposoma scincoides followed by Ecpleopus gaudichaudii and, the most abundant snake species was Bothrops jararaca being markedly higher than that recorded in similar studies. Twenty-seven percent of the reptile species recorded in our study are endemic to the Atlantic Forest and 30% (N = 10) have been recorded less than five times previously in the Brazilian state of Espírito Santo. Our study reinforces the need for the conservation of the Serra das Torres Natural Monument because of its importance as a reservoir of a considerable portion of the reptile biodiversity of Espírito Santo state, and of the Atlantic Forest biome as a whole.

This work deals with preparation and characterization of H3PW12O40 (H3PW) supported on silica-alumina. Impregnation of H3PW (15, 20, 30 and 40 wt.%) on commercial silica-alumina support in acidic aqueous solution is effective for preparing this catalyst keeping its Keggin structure, according to different methods of characterization. The catalysts were tested in a model reaction of acetic acid with ethanol and 30 wt.% H3PW/SiO2-Al2O3 had the highest activity under the conditions: catalyst calcination at 300 ºC, temperature of 100 ºC, acetic acid:ethanol molar ratio of 2:1 and catalyst:acetic acid mass ratio of 10 wt.%. The reaction yield was 79 and 100% selectivity for ethyl acetate over three reutilizations, for reaction time of 2 h. The calculated total acid site distribution was 0.299 mmol g-1 (97% of the theoretical probed by pyridine), and most of these (0.236 mmol g-1) were Brønsted weak-medium strength (pyridine desorption between 300 and 500 ºC).

Individual, spatial and inter-sex variation in somatic growth: a study of Piaractus mesopotamicus (Characiformes: Serrasalmidae), a long-distance freshwater Neotropical migratory fish

RESUMO O crescimento é um processo biológico fundamental, impulsionado por múltiplos fatores endógenos (intra-individual) e exógenos (ambientais) que mantém o fitness individual e a estabilidade populacional. Este trabalho tem como objetivo avaliar se ocorre variação individual, espacial (região de cabeceira e planície de inundação) e entre sexo no crescimento de Piaractus mesopotamicus na bacia do rio Cuiabá. Amostragens foram realizadas mensalmente entre julho de 2006 a julho de 2007, em duas áreas da bacia do rio Cuiabá (região de cabeceira e planície de inundação). Três modelos de crescimento (indivíduos, indivíduos e sexo como fator, indivíduos e área como fator) foram elaborados e comparados os parâmetros de crescimentos dos peixes com base no critério de Informação de Akaike (AIC). O melhor ajuste aos dados de comprimento na idade data foi obtido pelo modelo que considerou a variação individual e o sexo. O comprimento médio máximo teórico ( L∞ ) das fêmeas foi 64,99 cm e para os machos 63,23 cm. O coeficiente de crescimento (k) médio foi de 0,230 yr-1 e 0,196 yr-1, respectivamente para fêmeas e machos. Portanto, pode-se concluir que variabilidade individual e sexo foram as principais origem de variação dos parâmetros de crescimento somático de P. mesopotamicus.
ABSTRACT Growth is a fundamental biological process, driven by multiple endogenous (intra-individual) and exogenous (environmental) factors that maintain individual fitness and population stability. The current study aims to assess whether individual, spatial (headwaters and floodplains) and inter-sex variation occurs in the growth of Piaractus mesopotamicus in the Cuiabá River basin. Samples were collected monthly from July 2006 to July 2007, at two areas in the Cuiabá River basin (headwaters and floodplain). Three growth models (individuals; individuals and sex factors; individuals and areas factors) were developed and compared the fish growth parameters using Akaike information criterion (AIC). The best fit to the length-at-age data was obtained by a model that considered individual variation and sex. The theoretical maximum average length ( L∞ ) was 64.99 cm for females, and 63.23 cm for males. Females showed a growth rate (k) of 0.230 yr-1and males of 0.196 yr-1. Thus, could be concluded that individual variability and sex were the main sources of variation in P. mesopotamicus somatic growth parameters.

ABSTRACT Laetia suaveolens (Poepp.) Benth., Salicaceae, popularly known as "casinga-cheirosa", "caferana", or "laranjinha", is native to Brazil but not endemic to this country. A crude organic extract was obtained from the leaves and stem and intraperitoneally administered in male Balb-c mice. Its behavioral effects were evaluated in the open field and elevated plus maze in a two-stage experiment that assessed ten different parameters related to behavior as locomotion, emotionality, and anxiety. In the first stage of the experiment, intraperitoneal the crude organic extract administration dose-dependently impaired locomotion and emotionality 30–120 min after administration. A significant decrease in defecation was observed, which was related to emotionality. No alterations in the elevated plus maze were found; thus, this apparatus was not used in the next stage of the experiment. In the second stage, the previously determined non-lethal dose of 0.1563 g/kg was intraperitoneally administered, which impaired locomotion and rearing frequency and increased immobility time. Necropsy revealed smooth intestine hemorrhage. Rutin, leucoside, nicotiflorin, guaijaverin, and astragalin were isolated from the crude organic extract. This is the first time that these compounds have been identified in L. suaveolens. In conclusion, the crude organic extract impaired locomotion and emotionality and caused hemorrhage in male Balb-c mice, indicating that its consumption can be harmful to humans and animals. The present results provide a basis for further studies on the pharmacology, toxicology, and natural product chemistry of the crude organic extract.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Introducción: La fibrosis quística es la enfermedad letal autosómica recesiva más frecuente en caucásicos, donde su incidencia es de 1 en 2000 nacidos vivos. Se debe a mutaciones en el gen CFTR, de las cuales la más frecuente es la F508del presente en 66% de los enfermos y en 1 de cada 25 personas sanas de origen caucásico. Objetivo: Identificar la tasa de portadores de la mutación F508del en una muestra de estudiantes de la Facultad de Medicina de la Universidad del Rosario. Materiales y métodos: Se determinó la presencia de la mutación F508del mediante PCR y análisis de heterodúplex en 110 estudiantes de IV y VII semestre de la Facultad de Medicina. Resultados: Se obtuvieron 4 heterocigotos para la mutación F508del, es decir, una frecuencia de portadores de 1 en 27 estudiantes. Conclusiones: La frecuencia de portadores de esta mutación en la población analizada es considerable, e indica que hay un alto número de personas en riesgo de heredar la enfermedad y que deben recibir asesoramiento genético. Es necesario aumentar el tamaño de la muestra para obtener datos representativos de la población colombiana.
Introduction: Cystic fibrosis (CF) is the most frequent autosomical recessive disorder in Caucasian population with an incidence of 1 in 2000 newborns. The disease is caused by mutations in the cftr gene, but the most common mutation is F508del, which accounts for 66% of CF chromosomes worldwide and a carrier frequency for Caucasian population of 1 in 25. Objective: To determine the carrier frequency of the F508del mutation in 110 unrelated, healthy students from the Facultad de Medicina, Universidad del Rosario. Methods: The presence of F508del mutation using PCR and heteroduplex analysis was determined. Results: Only four heterozygotes for F508del mutation were discovered. This represents a carrier frequency of 1 in 27 students. Conclusions: This estimated frequency of F508del carriers is higher than expected, encouraging further screening in normal control individuals from different regions of Colombia.

O objetivo deste trabalho foi selecionar plantas com atividade inseticida, para isolar, identificar e avaliar a bioatividade de compostos inseticidas presentes nessas plantas, contra as seguintes pragas de produtos armazenados da ordem Coleoptera: Sitophilus zeamais Mots. (Curculionidae), Rhyzopertha dominica F. (Bostrichidae) e Oryzaephilus surinamensis L. (Silvanidae). As espécies de plantas usadas foram: anis (Ocimum selloi Benth), arruda (Ruta graveolens L.), cordão-de-frade (Leonotis nepetifolia L.), datura (Datura stramonium L.), erva baleeira (Cordia verbenacea L.), hortelã (Mentha piperita L.), melão-de-são-caetano (Mormodica charantia L.) e mentrasto (Ageratum conyzoides L.). Avaliou-se a toxicidade dos extratos hexânico e etanólico dessas plantas sobre R. dominica. Somente o extrato hexânico de A. conyzoides apresentou atividade inseticida. O extrato hexânico desta planta foi fracionado, sucessivamente, por cromatografia de coluna de sílica gel, para isolamento e purificação dos compostos ativos. Os compostos 5,6,7,8,3',4',5'-heptametoxiflavona; 5,6,7,8,3'-pentametoxi-4',5'-metilenodioxiflavona e cumarina foram identificados. Entretanto, somente a cumarina apresentou atividade inseticida às três espécies de insetos (DL50 de 2,72 a 39,71 mg g-1 de i.a.). A ordem crescente de suscetibilidade à cumarina foi R. dominica, S. zeamais e O. surinamensis.
The objective of this work was to screen plants with insecticide activity, in order to isolate, identify and assess the bioactivity of insecticide compounds present in these plants, against Coleoptera pests of stored products: Oryzaephilus surinamensis L. (Silvanidae), Rhyzopertha dominica F. (Bostrichidae) and Sitophilus zeamais Mots. (Curculionidae). The plant species used were: basil (Ocimum selloi Benth.), rue (Ruta graveolens L.), lion's ear (Leonotis nepetifolia (L.) R.Br.), jimson weed (Datura stramonium L.), baleeira herb (Cordia verbenacea L.), mint (Mentha piperita L.), wild balsam apple (Mormodica charantia L.), and billy goat weed or mentrasto (Ageratum conyzoides L.). The insecticide activity of hexane and ethanol extracts from those plants on R. dominica was evaluated. Among them, only hexane extract of A. conyzoides showed insecticide activity; the hexane extract of this species was successively fractionated by silica gel column chromatography, for isolation and purification of the active compounds. Compounds 5,6,7,8,3',4',5'-heptamethoxyflavone; 5,6,7,8,3'-pentamethoxy-4',5'-methilenedioxyflavone and coumarin were identified. However, only coumarin showed insecticide activity against three insect pests (LD50 from 2.72 to 39.71 mg g-1 a.i.). The increasing order of insects susceptibility to coumarin was R. dominica, S. zeamais and O. surinamensis.

First gonadal maturation of Pinirampus pirinampu (Siluriformes: Pimelodidae) in the Pantanal, Mato Grosso do Sul State, Brazil

Informações sobre o ciclo reprodutivo de espécies exploradas pela pesca são imprescindíveis para o seu manejo adequado. O tamanho de início da atividade reprodutiva do Pinirampus pirinampu (Spix, 1829) foi determinado. Os exemplares foram capturados em setembro, outubro e dezembro de 1997 e fevereiro e março de 1998. O estádio de desenvolvimento gonadal foi identificado macroscopicamente. O índice gonadossomático médio (IGS), calculado mensalmente, foi utilizado como indicador da época de desova. Para determinação do comprimento da primeira maturação gonadal agruparam-se, separadamente, machos e fêmeas por classes de comprimento em imaturos e adultos. Os resultados referentes aos indivíduos adultos foram lançados em gráficos e a mediana correspondeu à estimativa do comprimento no qual 50% dos indivíduos atingem a maturidade (L50). Foi também determinado o L100, estimativa do comprimento em que todos os indivíduos estão aptos à reprodução. Machos e fêmeas em processo de maturação gonadal foram encontrados a partir de outubro, com maior freqüência em fevereiro, e, somente a partir deste mês, foram encontrados indivíduos com gônadas esvaziadas. O índice gonadossomático mostrou que a partir de setembro inicia-se o processo de desenvolvimento gonadal, com seu valor máximo em fevereiro. O L50 para fêmeas foi 574 mm e para machos foi 536 mm. O L100 para fêmeas foi 590 mm e para os machos, 580 mm.
Information regarding the reproductive cycle of commercially exploited fish species is extremely important for their appropriate management. The reproductive size of Pinirampus pirinampu (Spix, 1829) was determined for a population studied in the Pantanal wetlands of western Brazil. Samples were taken in September, October, and December 1997, and in February and March 1998. Gonadal stages were macroscopically determined and a monthly gonadossomatic index (GSI) was used to determine the breeding period. The length of the first gonadal maturation was determined by classifying male and female adults and immature fish into length classes. The data on the adult individuals were plotted on graphs and the median values estimated to find the length at which 50% of the individuals reached maturity (L50). L100 was also determined. Males and females in the gonadal maturation process were recorded from October on, and the highest frequency was found to occur in February. Individuals with empty gonads occurred from February onwards. The GSI showed that gonadal development began in September and reached its peak in February. The L50 was 574 mm for females and 536 mm for males, while the L100 was 590 mm for females and 580 mm for males.
