Abstract There are no records of autochthonous cases of canine visceral leishmaniasis in the city of Curitiba, Paraná state, Brazil. In 2020, a male French bulldog (CW01), approximately 2 years old was taken by its owners to a private veterinarian clinic. The suspicion of CVL was confirmed by means of a serology test (ELISA/IFAT reagent), rapid chromatographic immunoassay (DPP®) (ELISA - Biomanguinhos®), parasitological culture and quantitative polymerase chain reaction (qPCR). The animal routinely frequented parks in Curitiba and was taken on several trips to the municipalities of Bombinhas and Balneário Camboriú (Santa Catarina) and to Matinhos (Paraná) where CVL had not previously been reported. Treatment was initiated orally with Milteforan™ which resulted in a significant reduction in the parasitic load. The suspicion of autochthony was investigated through entomological research. A total of 10 traps were installed, one at the animal’s home, seven in adjacent city blocks and two in a forest edge. No sandflies were trapped in the dog’s home and adjacent houses. The traps in the forest edge caught one Migonemyia migonei female and five Brumptomyia spp. females. This case serves as a warning of the possible introduction of CVL in the city of Curitiba.

Resumo Não há registros de casos autóctones de leishmaniose visceral canina (LVC) no município de Curitiba, Paraná, Brasil. Em 2020, um "bulldog" francês macho (CW01), com aproximadamente 2 anos de idade, foi levado por seus donos a uma clínica veterinária particular. A suspeita de LVC foi confirmada por meio de teste sorológico (reagente ELISA/IFAT), imunoensaio cromatográfico rápido (DPP®) (ELISA - Biomanguinhos®), cultura parasitológica e reação quantitativa em cadeia da polimerase (qPCR). O animal frequentava, rotineiramente, parques de Curitiba e foi levado em diversas viagens para Bombinhas e Balneário Camboriú (Santa Catarina) e para Matinhos (Paraná), onde a LVC não havia sido relatada anteriormente. O tratamento foi iniciado por via oral com Milteforan™ (Virbac) o que resultou em redução significativa da carga parasitária. A suspeita de autóctone foi investigada por meio de pesquisa entomológica. Foram instaladas 10 armadilhas, uma na casa do animal, sete em quarteirões adjacentes e duas na borda da mata. Nenhum inseto foi capturado na casa do proprietário do animal e nas casas adjacentes. As armadilhas na borda da mata capturaram uma fêmea de Migonemyia migonei e cinco Brumptomyia spp. fêmeas. Este caso serve de alerta para a possível introdução da LVC na cidade de Curitiba.

Este estudo transversal, realizado em um hospital oncológico em Goiânia, objetivou caracterizar a prevalência da colonização oral e suscetibilidade antimicrobiana de Pseudomonas spp. isoladas da saliva de profissionais da saúde. Micro-organismos foram submetidos a testes bioquímicos, perfil de sensibilidade e detecção fenotípica. Dos 76 participantes colonizados por bastonetes Gram negativos, 12 (15,8%) albergavam Pseudomonas spp. Todos os isolados, P. aeruginosa (75,0%), P. stutzeri (16,7%) e P. fluorescens (8,3%), foram resistentes à cefoxitina e, portanto, possíveis produtores de AmpC. Os resultados são clinicamente relevantes e enfatizam a importância da vigilância para minimizar a disseminação e a multirresistência bacteriana.

This cross-sectional study, performed in an oncology hospital in Goiania, aimed to characterize the prevalence of oral colonization and antimicrobial susceptibility of Pseudomonas spp. isolated from the saliva of healthcare workers. Microorganisms were subjected to biochemical tests, susceptibility profile, and phenotypic detection. Of 76 participants colonized with Gram negative bacilli, 12 (15.8%) harbored Pseudomonas spp. Of all isolates, P. aeruginosa (75.0%), P. stutzeri (16.7%), and P. fluorescens (8.3%), were resistant to cefoxitin, and therefore likely to be AmpC producers. The results are clinically relevant and emphasize the importance of surveillance to minimize bacterial dissemination and multiresistance.

The aim of the study was to analyze epidemiological and microbiological aspects of oral colonization by methicillin-resistant Staphylococcus of health care workers in a cancer hospital. Interview and saliva sampling were performed with 149 health care workers. Antimicrobial resistance was determined by disk diffusion and minimum inhibitory concentration. Polymerase Chain Reaction, Internal Transcribed Spacer-Polymerase Chain Reaction and Pulsed Field Gel Electrophoresis were performed for genotypic characterization of methicillin-resistant Staphylococcus. Risk factors were determined by logistic regression. Methicillin-resistant Staphylococcus colonization prevalence was 19.5%, denture wearing (p = 0.03), habit of nail biting (p = 0.04) and preparation and administration of antimicrobial (p = 0.04) were risk factors identified. All methicillin-resistant Staphylococcus were S. epidermidis, 94.4% of them had mecA gene. Closely related and indistinguishable methicillin-resistant S. epidermidis were detected. These results highlight that HCWs which have contact with patient at high risk for developing infections were identified as colonized by MRSE in the oral cavity, reinforcing this cavity as a reservoir of these bacteria and the risk to themselves and patients safety, because these microorganisms may be spread by coughing and talking.

O presente trabalho teve como objetivo utilizar marcadores RAPD para conhecer a variabilidade genética de populações de Tibouchina papyrus (Pohl) Toledo, provenientes da região de Serra Dourada e Serra dos Pirineus, no Estado de Goiás. Os seis iniciadores RAPD produziram um total de 147 locos, variando entre 23 e 26 por iniciador. A avaliação hierárquica da estruturação da variabilidade genética, realizada pela a AMOVA, considerando a existência de duas regiões (Serra dos Pirineus e Serra Dourada) apresentou uma estimativa de F ST = 0,3439. O valor do componente entre regiões (F CT) foi igual a 18,96% e a variação entre populações dentro de regiões igual a 15,43%. As estimativas de fluxo gênico sugerem a existência de uma baixa proporção de migrantes entre populações. As análises multivariadas (UPGMA e NMDS) indicam que existe uma relação entre distância genética e espaço geográfico, hipótese esta que foi confirmada por uma análise de padrão espacial utilizando o teste de Mantel (r = 0,71; P = 0,015 com 1000 permutações aleatórias). Os resultados indicam assim que esta estrutura tenha se originado seguindo um modelo de diferenciação estocástica (neutro), ou seja, por um balanço entre fluxo gênico a curtas distâncias e deriva genética nas populações. Os valores de diversidade genética obtidos apóiam a hipótese de que a espécie T. papyrus é uma espécie xenógama facultativa e o fato de não serem encontrados altos níveis de homozigose, indica que devem existir mecanismos relacionados à biologia reprodutiva da espécie, que previnem, de alguma maneira, a ocorrência de elevadas taxas de endogamia, que poderia ter um efeito deletério em médio e longo prazo.

This paper has the aim of using RAPD markers to describe the genetic of variability of Tibouchina papyrus (Pohl) Toledo local populations, from the regions of Serra Dourada and Serra de Pirineus, in Goiás State, Central Brazil. The six RAPD primers generated a total of 147 loci, varying from 23 to 26 per primer. The hierarchical evaluation of genetic variability, performed using an Analysis of Molecular Variance (AMOVA) considering the local populations within the two regions (i.e., Serra Dourada and Serra de Pirineus), showed an estimate of overall diversity among populations equal to F ST = 0.3439. The value of divergence between regions (F CT) was equal to 18.96% so that the variation among local populations within regions was equal to 15.43%. Estimates of gene flow suggest a small number of migrants among local populations per generation. Multivariate analyses (UPGMA and NMDS) indicated that a relationship between genetic and geographical distances exists, which was confirmed by a spatial pattern analysis using Mantel test (r = 0.71; P < 0.015 with 1000 random permutations). Thus, this structure was originated from a stochastic neutral model of population differentiation, in which drift within populations is counteracted by short distance gene flow. Despite strong levels of population structure (i.e., high divergence among populations within regions), the diversity observed support the hypothesis that T. papyrus is a facultative xenogamous species. The lack of high levels of homozigosity indicates that ecological mechanisms related to species' reproductive biology prevent, someway, a high level of endogamy within local structured populations, which could cause deleterious effects in a long run.
