Doxorubicin cardiotoxicity and Camellia sinensis cardioprotection determined by speckle-tracking echocardiography

Abstract Doxorubicin (Dox) is a medication used in the treatment of cancerous tumors and hematologic malignancies with potentially serious side effects, including the risk of cardiotoxicity. Flavonoids are plant metabolites with antioxidant properties and can be extracted from Camellia sinensis (CS). The aim of this study is to evaluate the possible cardioprotective effect of CS against injuries induced by Dox in rats. A total of 32 animals were distributed into four groups: (1) control - intraperitoneal injection (I.P.) of 0.5 mL saline weekly and 1.0 mL water by gavage daily; (2) CS - 0.5 mL saline I.P. weekly and 200 mg/kg CS by gavage daily; (3) Dox - 5.0 mg/kg Dox I.P. weekly and 1.0 mL water by gavage daily; and (4) Dox+CS -5.0 mg/kg Dox I.P. weekly and 200 mg/kg CS by gavage daily. Clinical examinations, blood profiles, electrocardiograms, echocardiograms, and histological analyses of hearts were performed over 25 days. The animals in the Dox group showed changes in body weight and in erythrogram, leukogram, electrocardiography, and echocardiography readings. However, animals from the dox+CS group had significantly less change in body weight, improved cardiac function, and showed more preserved cardiac tissue. This study demonstrated that CS prevents dox-induced cardiotoxicity, despite enhancing the cytotoxic effect on blood cells.

Resumo Niedenzuella (Tetrapterys) multiglandulosa, uma videira encontrada no Brasil, tem sido correlacionada a surtos de intoxicações em bovinos e búfalos, gerando perdas econômicas relacionadas à morte por insuficiência cardíaca, aborto, natimorto e mortalidade neonatal. O objetivo deste estudo foi examinar o potencial embriotóxico do extrato vegetal aquoso em embriões bovinos in vitro. O estudo in vitro foi realizado em cinco repetições de cultura de embriões bovinos distribuídos em dois grupos: controle, meio de cultura de embriões in vitro sem o extrato aquoso da planta; grupo tratado, com adição de 2,7mg / mL de extrato vegetal aquoso (10%) à cultura do embrião no sexto dia de cultivo. A taxa de clivagem foi avaliada no dia 2 da cultura de células. Viabilidade, eclodibilidade e subdesenvolvimento de blastocistos no sétimo, oitavo e nono dia (D7, D8 e D9, respectivamente) de cultura foram avaliados em microscópio estereoscópico. No dia 7, os blastocistos foram submetidos ao ensaio TUNEL para determinar o índice apoptótico. Observamos redução significativa da produção de blastocisto / número de embriões clivados (60,6% vs 41,5%); redução da produção de blastocistos / número total de oócitos bovinos maturados (35,1% vs 21,3%); e taxas de eclosão embrionária (38,0% vs 10,0%). No entanto, nenhum efeito foi observado na taxa de apoptose. Em conclusão, o extrato aquoso das folhas de N. multiglandulosa reduz a viabilidade do embrião bovino in vitro, sugerindo possíveis efeitos prejudiciais no desenvolvimento embrionário.
Abstract Niedenzuella (Tetrapterys) multiglandulosa, a vine plant found in Brazil, has been correlated to outbreaks of poisoning in cattle and buffaloes, generating economic losses related to the death due to heart failure, miscarriage, abortion, stillbirth, and neonatal mortality. The aim of this study was to examine the embryotoxic potential of the aqueous plant extract on in vitro bovine embryos. In vitro study was performed in five replicates of bovine embryo culture assigned in two groups: control, in vitro embryo culture medium without the aqueous plant extract; treated group, with addition of 2.7mg/mL of aqueous plant extract (10%) to the embryo culture on the sixth day of culture. Cleavage rate was evaluated at day 2 of the cell culture. Viability, hatchability and underdevelopment of blastocysts on the seventh, eighth, and ninth days (D7, D8, and D9, respectively) of culture were assessed under stereoscopic microscope. On day 7, blastocysts were submitted to TUNEL assay to determine apoptotic index. In vitro exposure of bovine embryos to of N. multiglandulosa resulted in reduced embryo development and survival, evaluated by dark cytoplasm indicating poor morphology and poor quality with marked reduction of hatchability. We observed a significant reduction of blastocyst production/number of cleaved embryos (60.6% vs 41.5%); reduction of blastocysts production/total number of matured bovine oocytes (35.1% vs 21.3%); and embryonic hatching rates (38.0% vs 10.0%). However, no effects were observed on the apoptotic rate. In conclusion, aqueous extract of N. multiglandulosa leaves reduces bovine embryo viability in vitro, suggesting possible detrimental effects on embryo development.

Resumo Este estudo teve como objetivo avaliar a eficácia das células-tronco mesenquimais (CTMs), isoladas ou associadas à dapsona (DAP), no tratamento de feridas dermonecróticas causadas pelo veneno de Loxosceles laeta. Vinte e cinco coelhos machos foram distribuídos em cinco grupos. O controle negativo recebeu água ultrapura (C-), enquanto todos os outros grupos foram injetados com 20 μg de veneno de L. laeta. Após 4 horas, cada grupo recebeu um dos seguintes tratamentos: PBS (C+), DAP, CTMs e DAP + CTMs. Os animais foram avaliados diariamente durante 12 dias, e feitos registros fotográficos para análise da ferida e no 12º dia, foram eutanasiados e, retiradas amostras de pele para análise histológica. Observou-se que a DAP apresentou o melhor percentual de contração da ferida no terceiro dia. Nos tratamentos com CTMs, observou-se uma contração negativa da ferida tanto para as CTMs isoladas, bem como a associação CTMs + DAP em relação ao PBS, possivelmente, pelo aumento da infamação inicial após a aplicação de células-tronco. Isso é devido ao fato de que as CTMs secretam um amplo espectro de moléculas bioativas como citocinas e fatores de crescimento que favorecem a regeneração. Histologicamente, observou-se que os animais de C+ apresentaram extensas áreas de necrose, úlceras, infiltrado neutrofílico, além de mineralização. Houve aumento de deposição de colágeno no tratamento CTMs + DAP, no entanto, a vascularização permaneceu inalterada. Este é o primeiro relato usando CTMs e CTMs + DAP como tratamento para loxoscelismo cutâneo e mais estudos são necessários para determinar seu uso como terapia alternativa para lesões demonecróticas causadas pela aranha Loxosceles.
Abstract This study aimed to evaluate the efficacy of mesenchymal stem cells (MSC), alone or associated with dapsone (DAP), in treating dermonecrotic wounds caused by Loxosceles laeta venom. Twenty-five male rabbits were distributed into five groups. Negative control received ultrapure water (C-), whilst all other groups were injected with 20 μg of L. laeta venom. After 4 hours, each group received one of the following treatments: PBS (C+), DAP, MSC, and DAP+MSC. Animals were evaluated daily and photographic records made for analysis of wound area. Twelve days after, animals were euthanized and skin samples removed for histological analysis. We observed that DAP showed the best percentage of wound contraction at day 3. In the treatments using MSCs, a negative value of wound contraction was observed for the isolated MSCs, as well as a lower contraction value for the association of the MSC + DAP when compared to PBS, probably, by the increase in initial infammation after the application of stem cells, due to the fact that MSCs secrete a broad spectrum of bioactive molecules such as cytokines and growth factors that favor regeneration. Histologically, it was observed that animals of C+ showed extensive areas of necrosis, ulcers, neutrophilic infiltrate, and mineralization. Collagen deposition showed increase in MSC+DAP treatment, however vascularization remained unchanged. This is the first report using MSC and MSC+DAP as a treatment for cutaneous loxoscelism and more studies are needed to determine its use as an alternative therapy for dermonecrotic lesions caused by Loxosceles spider.

Resumo A pandemia causada pelo Coronavírus da Síndrome Respiratória Aguda Grave 2 (SARS-CoV-2) e o distanciamento social causaram impactos sobre a saúde das pessoas. Dentre eles tem-se o estresse ambiental, compreendido como um conjunto de reações objetivas e subjetivas que ocorrem quando as pessoas enfrentam influências negativas presentes no ambiente. Objetivou-se investigar variáveis associadas ao estresse ambiental domiciliar durante a pandemia de coronavírus no Brasil. Para isso, realizou-se uma pesquisa online com 2000 brasileiros com idade média 38,7 anos (DP = 14,55), sendo que 76,80% eram mulheres. Os resultados mostraram que 81,90% (f = 1.639) da amostra apresentaram algum nível de estresse ambiental, que foi predito pela região geográfica (norte e nordeste brasileiro), menor renda, menor avaliação da qualidade residencial e a falta de um ambiente restaurador na casa. Conclui-se com a identificação de alguns fatores ambientais negativos e positivos associados ao estresse ambiental, que pode auxiliar na implementação de políticas para o enfrentamento da Doença por Coronavírus-19 (COVID-19) e promoção de saúde mental no contexto da pandemia.
Abstract The Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) coronavirus pandemic and social distancing caused impacts on people’s health, among them, environmental stress – a set of objective and subjective reactions that occur when people face negative environmental influences. The objective was to investigate variables associated with residential environment stress during the pandemic in Brazil. An online survey was carried out with 2000 Brazilians with a mean age of 38.7 years (SD = 14.55), 76.80% of whom were women. The results showed that 81.90% (f = 1,639) of the sample had some level of environmental stress, predicted by geographic region (Brazil’s northern and northeastern regions), lower income, lower residential quality scores, and absence of a restorative environment. It is then concluded the identification of some negative and positive environmental factors associated with environmental stress, which may assist in the implementation of policies for coping with 2019 Coronavirus Disease (COVID-19) and promoting mental health in the pandemic context.

Resumo É complexa a tarefa de definir medidas de adaptação às mudanças climáticas diante da existência de demandas sociais, ambientais e econômicas, sobretudo em cidades que apresentam infraestrutura urbana deficiente. Como resultado da análise do processo de revisão do Plano Diretor do Recife (PDDR), vê-se que a redução das vulnerabilidades socioambientais implica a realização de estudos mais consistentes, que possam acarretar a implementação de medidas estruturadoras, e que a sustentabilidade ambiental demanda uma governança multinível, com reformas políticas em escala global, regional e local, de difícil aplicação no curto prazo, mas necessárias para reorientar as políticas do clima e superar a inabilidade de prover os recursos para uma infraestrutura adequada à adaptação. O caminho para construir uma cidade resiliente, que proporcione um ambiente mais seguro para as futuras gerações, depende de um modelo de desenvolvimento inclusivo, que permita melhorar as condições urbanas para a população e minimizar os impactos ocasionados pelos eventos climáticos extremos.
Abstract Defining measures for climate change adaptation is a complex task given the existence of social, environmental, and economic demands, particularly in cities with poor urban infrastructure. As a result of analyzing the revision process of the Recife Master Plan, it is possible to observe that a reduction in the social and environmental vulnerabilities has implied carrying out more consistent studies, which may entail the implementation of structuring measures, and that environmental sustainability requires multilevel governance, with policy reforms on a global, regional and local scale, difficult to implement in the short term, although necessary for refocusing climate policies and for overcoming the inability to provide resources for a tailored adaptation infrastructure. The path to building a resilient city that provides a safer environment for the future depends on an inclusive development model, which enables the population to improve urban conditions and minimize the impacts brought about by extreme weather events.

Outbreak of organophosphorus compound-induced delayed neurotoxicity in water buffaloes

RESUMO: Quarenta búfalos foram simultaneamente tratados com clorpirifós e triclorfom na dose recomendada para bovinos. Após uma semana, 19 animais apresentaram sinais clínicos caracterizados por apatia, diarreia, agressividade, desidratação e incoordenação motora, seguidos por paralisia flácida e decúbito lateral permanente. Todos os búfalos afetados morreram após um curso clínico de 1-4 dias. Redução da atividade da colinesterase sérica em três casos foi indicativa de exposição significativa a organofosforados (OPs). O exame patológico de três búfalos não revelou lesões macroscópicas e histológicas. Por cromatografia em camada delgada, resíduos de clorpirifós e traços de resíduos de triclorfon foram detectados em amostras de tecidos frescos. Os achados epidemiológicos, clínicos, patológicos e toxicológicos foram compatíveis com neuropatia tardia induzida por OPs, uma manifestação neurológica raramente descrita em animais domésticos.
ABSTRACT: Forty 1-2-y-old water buffaloes were simultaneously treated with trichlorfon and chlorpyrifos products in the recommended dose for cattle. After a week, 19 animals started presenting clinical signs characterized by apathy, diarrhea, aggressiveness, dehydration, and motor incoordination, followed by flaccid paralysis and permanent lateral recumbency. All affected buffaloes died after a clinical course of 1-4 days. Reduction of serum cholinesterase activity in three cases was indicative of significant exposure to organophosphorus compounds (OPs). Pathological examination of three buffaloes revealed no gross and histological lesions. By thin layer chromatography, chlorpyrifos residues and trace of trichlorfon residues were detected in fresh tissue samples. The epidemiological, clinical, pathological, and toxicological findings were highly compatible with OPs-induced delayed neurotoxicity, a neurological manifestation rarely described in domestic animals.

Resumo: O objetivo deste trabalho foi avaliar o efeito da anemia infecciosa equina (AIE) no desempenho físico de equinos da região do Pantanal brasileiro. Foram avaliados 16 machos, divididos em dois grupos: 8 soronegativos (G1) e 8 soropositivos (G2) para AIE. Dois testes de esforço progressivo foram realizados, antes (T1) e após (T2) 42 dias de treinamento. Foram registrados frequência cardíaca, concentração de lactato, distância percorrida e níveis de hematócrito. Em ambos os testes, o G1 percorreu uma distância maior. No T2, o G2 apresentou menores níveis de hematócrito e menor velocidade obtida a diferentes concentrações de lactato e frequências cardíacas. O desempenho atlético dos equídeos avaliados é afetado pela anemia infecciosa equina.
Abstract: The objective of this work was to evaluate the effects of equine infectious anemia (EIA) on the physical performance of equines from the Brazilian Pantanal region. A total of 16 males were evaluated, divided into two groups: 8 seronegative (G1) and 8 seropositive (G2) for EIA. Two graded exercise tests were carried out before (T1) and after (T2) 42 days of training. Heart rate, lactate concentration, distance covered, and hematocrit level were recorded. In both tests, G1 covered a greater distance. In T2, G2 had lower hematocrit levels and lower speeds reached at different lactate concentrations and heart rates. The athletic performance of the evaluated equines is affected by equine infectious anemia.

RESUMO: O objetivo do presente trabalho foi avaliar a utilização das células tronco mesenquimais (MSC) no tratamento da mionecrose induzida pelo veneno de Bothrops alternatus em ratos. 75 ratos Wistar adultos foram distribuídos em três grupos experimentais. G1 e G2 receberam 120μg de veneno de B. alternatus, enquanto o G3 recebeu apenas 200μL de PBS. Três dias após a administração do veneno, os ratos do grupo G1 foram tratados com 5.0 x 106 MSC, enquanto G2 e G3 receberam exclusivamente PBS. A cada três dias, amostras de sangue e tecido muscular foram coletadas de cinco animais de cada grupo para avaliação bioquímica sérica e patológica, respectivamente. A análise histológica revelou lesão muscular mais intensa após a aplicação das MSC, caracterizada pela desorganização e perdas das fibras musculares, com necrose focal e infiltrado inflamatório mononuclear. É possível concluir que a utilização das MSC para o tratamento da lesão local causada pela inoculação do veneno B. alternatus deteriorou a regeneração muscular e interferiu com o processo de cicatrização.
ABSTRACT: The aim of this study was to evaluate the use of mesenchymal stem cells (MSC) in the treatment of myonecrosis induced by Bothrops alternatus venom in rats. Seventy-five male adult Wistar rats were divided into three experimental groups. G1 and G2 were injected in the gastrocnemius muscle with 120μg of B. alternatus venom, while G3 received 200μL of PBS only. Three days after the venom injection, 12 rats from G1 were treated with 5.0 x 106 MSC in PBS, whereas G2 and G3 rats received PBS. Every three days, blood and muscle samples of five animals from each group were taken for serum biochemical and pathological analyses. Histological examinations showed more intense muscle lesions following MSC treatment, characterized by disorganization and loss of muscle fibers, with focal necrosis and inflammatory infiltration by mononuclear cells. In conclusion, the use of MSC for the treatment of local damage caused by inoculation of B. alternatus venom impaired muscle regeneration and interfered in the healing process.

RESUMO: No Brasil, pelo menos 16 espécies de plantas tóxicas podem afetar o sistema nervoso central dos bovinos. Recentemente no estado do Piauí, no Nordeste do Brasil, Brunfelsia uniflora foi relatada como causadora de sinais nervosos em jumentos, bovinos e pequenos ruminantes. Para avaliar a toxicidade de B. uniflora, 20 camundongos foram distribuídos em quatro grupos de cinco animais recebendo diferentes extratos da planta. Os extratos de alcalóides, flavonóides, saponinas ou solução salina (grupo controle) foram administrados por gavagem. Após a administração dos extratos, em dose única (5g kg-1), todos os animais foram observados diariamente. Os camundongos que receberam os extratos de B. uniflora apresentaram sinais clínicos moderados a graves, incluindo piloeração, vocalização e convulsões. Todos os camundongos que receberam extrato de saponinas morreram entre 10 e 20 minutos após. A avaliação bioquímica sérica dos animais que receberam saponinas, revelou discretos aumentos nos níveis de proteína total e diminuição nos níveis de magnésio e cloro. Conclui-se que, as saponinas presentes nas folhas de B. uniflora induzem efeitos neurológicos tóxicos agudos e, morte em camundongos.
ABSTRACT: In Brazil, at least 16 poisonous plant species can affect the central nervous system of livestock. Recently in the state of Piauí, Northeastern Brazil, Brunfelsia uniflora was reported as a cause of nervous signs in donkeys, cattle, and small ruminants. In order to assess the toxicity of B. uniflora extracts, 20 Swiss mice were distributed into four groups (n=5) receiving by gavage alkaloids, flavonoids, saponins, or saline (control group). After administration of the extracts in a single dose (5g kg-1), all mice were observed daily for clinical signs. Mice that received the extracts showed moderate to severe clinical signs, including piloerection, vocalization, and seizures. All mice dosed with saponins died between 10 and 20min after administration. Serum biochemical evaluation of animals that received saponins revealed slight increases in total protein levels and decreased magnesium and chlorite levels. In conclusion, saponins of B. uniflora leaves induced acute toxic neurological effects and death in mice.

RESUMO Objetivou-se realizar uma avaliação físico-química de amostras de água, sedimento e mata ciliar em uma piscicultura localizada próxima às margens do Ribeirão da Mata, Minas Gerais. As coletas de água foram semanais em quatro pontos assim denominados: (1) nascente; (2) entrada do sistema; (3) efluente concentrado e (4) ponto de lançamento. Foram monitorados os seguintes parâmetros físico-químicos: temperatura (ºC), pH, oxigênio dissolvido, demanda bioquímica de oxigênio, fósforo total, alcalinidade, sólidos dissolvidos totais, fosfato, nitrato, nitrito, sulfato, amônia e fenol das amostras de água. Foram pesquisados metais pesados (chumbo, cádmio, mercúrio e zinco) e agrotóxicos (organoclorados, organofosforados, clorofosforados, carbamatos, piretroides e rodenticidas anticoagulantes) nas amostras de água, sedimento e mata ciliar. Alcalinidade e sólidos totais apresentaram as maiores concentrações, provavelmente devido às características geológicas,por se tratar de região cárstica. Fenol, em todos os pontos, e fosfato, nos pontos 3 e 4, apresentaram valores acima dos limites permitidos para a classe II de água doce, segundo a Resolução CONAMA nº 357. Os outros parâmetros (amônia, nitrato, nitrito e sulfato), em todos os pontos de coleta, apresentaram baixos valores. Nas amostras de água não foram detectados chumbo e mercúrio; zinco apresentou valores abaixo de 0,007 mg.L-1. As amostras de sedimento apresentaram valores de chumbo entre 11,1566 e 37,0438 mg.L-1, estando 12 dessas acima do valor de referência de qualidade. Também em todas as amostras de sedimento foi detectado mercúrio, que variou de 0,35 a 0,82 mg.L-1. Em relação ao cádmio, esse não foi detectado nas amostras de sedimento e plantas. Também não foram detectados agrotóxicos em nenhuma das amostras estudadas.
ABSTRACT The aim of this study was to carry out a physicochemical analysis of water, sediment and riparian vegetation in a fish farm located near the Ribeirão da Mata basin in Minas Gerais State (Brazil). The water samples were collected weekly at four points: (1) source, (2) system entry, (3) discharge point, (4) and point release. The monitored physicochemical parameters in the water samples were: temperature (ºC), pH, dissolved oxygen, biochemical oxygen demand, total phosphorus, alkalinity, total dissolved solids, phosphate, nitrate, nitrite, sulfate, ammonia and phenol. Heavy metals (lead, cadmium, mercury and zinc) and pesticides (organochlorine, organophosphates, clorofosforados, carbamates, pyrethroids and anticoagulant rodenticides) were searched in the samples of water, sediment and riparian vegetation. Alkalinity and total solids presented the highest concentration, probably due to geological features of a carstic area. Phenol, at all points, and phosphate, at points 3 and 4, showed values above the limits allowed for class II freshwater, according to the CONAMA Resolution n. 357. The other parameters (ammonia, nitrate, nitrite and sulfate) in all sampling sites showed low values. It was not detected lead and mercury in the water samples; and zinc values were below 0.007 mg.L-1. However, the sediment samples showed lead values from 11.1566 to 37.0438 mg.L-1, with 12 of those samples above the quality reference value. In addition, it was detected mercury in all sediment samples, which ranged from 0.35 to 0.82 mg.L-1. It was not detected cadmium in samples of sediment neither plants. No pesticides were detected in the samples.

Abstract Background Hadruroides lunatus is the most abundant scorpion species in the Peruvian central coast, where most of the accidents involving humans are registered. In spite of its prevalence, there are only very few studies on H. lunatus envenomation. The aim of the present study was to analyze the cardiorespiratory alterations caused by H. lunatus envenomation in rodents. Methods Wistar rats injected with H. lunatus scorpion venom were submitted to electrocardiography. After euthanasia, rat lungs were collected and histopathologically analyzed. Mouse cardiomyocytes were used to perform immunofluorescence and calcium transient assays. Data were analyzed by ANOVA or Student’s t-test. The significance level was set at p< 0.05. Results It was observed that H. lunatus venom increased heart rate and caused arrhythmia, thereby impairing the heart functioning. Lungs of envenomed animals showed significant alterations, such as diffuse hemorrhage. In addition, immunofluorescence showed that H. lunatus venom was capable of binding to cardiomyocytes. Furthermore, mouse ventricular cardiomyocytes incubated with H. lunatus venom showed a significant decrease in calcium transient, confirming that H. lunatus venom exerts a toxic effect on heart. Conclusion Our results showed that H. lunatus venom is capable of inducing cardiorespiratory alterations, a typical systemic effect of scorpionism, stressing the importance of medical monitoring in envenomation cases.

Objetivou-se investigar o efeito neuroprotetor da aplicação intralesional da MVIIA em ratos submetidos ao trauma medular. Foram utilizados ratos Wistar, machos, com peso entre 300g±23.4, distribuídos em cinco grupos: controle negativo (SHAM), placebo (PLA), 5µM MVIIA, 10µM MVIIA e 20µM MVIIA. Após a laminectomia da vértebra torácica 12 (SHAM), os grupos PLA, 5µM MVIIA, 10µM MVIIA e 20µM MVIIA foram submetidos ao trauma medular agudo compressivo por cinco minutos e, cinco minutos após o trauma, receberam o tratamento específico em volume total padrão de 2µL, pela via intralesional, sendo utilizado como placebo o PBS estéril. A atividade locomotora foi avaliada pela escala proposta por Basso Beattie Bresnahan (BBB), com intuito de mostrar a padronização do trauma medular. Com 48 horas do trauma, os animais foram submetidos à eutanásia, coletou-se amostra do líquor no espaço atlantooccipital e um segmento medular, incluindo o epicentro e região caudal à lesão. Foram realizados ensaios de viabilidade mitocondrial, dosagem de glutamato, produção de espécies reativas de oxigênio (ERO) e peroxidação lipídica (PL). O delineamento do estudo foi inteiramente casualizado e os dados submetidos ao ANOVA, com comparação de médias pelo teste de SNK e os dados do teste BBB foram comparados utilizando o teste Kruskal-Wallis (P<0.05). Em relação aos escores do BBB, não houve diferença entre os grupos. A MVIIA não promoveu a diminuição dos níveis do glutamato, ERO, PL e não preservou a mitocôndria na aplicação intralesional, cinco minutos após o trauma medular em ratos.
This study aimed to investigate the neuroprotective effect of ω-conotoxin MVIIA (MVIIA) intralesional application in rats submitted to spinal cord injury. Male Wistar rats, weighing 300g±23.4, were distributed in five groups: negative control (SHAM), placebo (PLA), 5μM MVIIA, 10μM MVIIA and 20μM MVIIA MVIIA. After laminectomy of the 12th thoracic vertebra (SHAM), the PLA, 5μM MVIIA, 10μM MVIIA and 20μM MVIIA groups were subjected to acute compressive spinal cord trauma for five minutes, and then five minutes later, the animals received specific treatment in a standard total volume of 2µL, by intralesional route, using sterile PBS as placebo. Locomotor activity was assayed using Basso Beattie Bresnahan (BBB) scale to show the patterning of SCI. With 48 hours of injury, the animals were euthanized, the liquor sample was collected in atlantooccipital space, and also the spinal segment, including the epicenter and caudal region to injury. Assays were performed for mitochondrial viability, serum glutamate, production of reactive oxygen species (ROS) and lipid peroxidation (LP) were performed. The study design was randomized and the data submitted to ANOVA and comparison of means by SNK test, and data from BBB scale were evaluated using Kruskal-Wallis test (P<0.05). There was no significant difference between groups in BBB scores. The MVIIA did not promote decrease in the levels of glutamate, ROS, LP, and did not preserve the mitochondria in the intralesional application five minutes after spinal cord injury in rats.

Arrancamento de penas psicogênico em maritacas: haloperidol e enriquecimento ambiental

Este trabalho avaliou a eficácia dos métodos de enriquecimento ambiental (EA) e da administração do fármaco haloperidol no controle de arrancamento de penas em maritacas mantidas em cativeiro. Foram formados três grupos: G1 tratado com haloperidol, G2 receberam enriquecimento ambiental e G3 aves sem arrancamento de penas. Utilizou-se registro scan, instantâneo dos comportamentos: antes, durante e depois dos tratamentos. Fotografias auxiliaram na avaliação da plumagem através de escore de 0 a 10. No G1, as maritacas reduziram significativamente a atividade física, aumentando o tempo em que ficavam paradas sobre o poleiro e redução também na expressão de outros comportamentos. Nenhuma maritaca do G1 apresentou melhora na qualidade da plumagem, já, no G2, apenas um indivíduo não melhorou a condição da plumagem. Sendo assim, a utilização do enriquecimento ambiental promoveu melhores condições de bem estar animal e proporcionou o crescimento de novas penas nas áreas de arrancamento, ao contrário das observações realizadas em maritacas tratadas com o haloperidol
This study evaluated the effectiveness of environmental enrichment methods and the haloperidol administration in controlling feather picking in parakeets kept in captivity. Three groups were formed: G1 haloperidol treatment, G2 subjected to environmental enrichment and G3 no feather picked parakeets. In the behavioral study was used scan, instant registration: before, during and after the treatments. The plumage evaluation was done following a score from 0 to 10. G1 parakeets reduced significantly the activity, increasing stopped time on the perch and also reduced their expression of other behaviors. No G1 birds improved the plumage quality, already only one G2 parakeet has not improved feather condition. Thus, the use of environmental enrichment promotes better animal welfare conditions and provided the growth of new feather in the picked areas, in contrast to the observations made in psychogenic feathers picked parakeet treated with haloperidol

Nerium oleander é uma planta ornamental responsável por intoxicações em animais e humanos. Todas as partes da planta contém glicosídeos cardiotóxicos, principalmente a oleandrina. Alguns autores apontam que a toxicidade da planta apresentaria variação em função da cor da flor. O objetivo deste trabalho foi determinar se existe variação na concentração de oleandrina nas folhas de N. oleander de exemplares da planta com diferentes cores de inflorescências. Foram coletadas 10 amostras de folhas para cada tipo de flor (branca, rosea e vermelha). Os níveis de oleandrina foram determinados por meio de HPLC-UV após extração com metanol, precipitação com acetato de chumbo e separação em cartucho de C18. A média da concentração obtida em todas as folhas analisadas foi de 4,89mg g-1, sendo 6,20±4,08mg g-1 na variedade de flores brancas, 4,16±3,44mg g-1 na de flores roseas e 4,31±1,99mg g-1 na de flores vermelhas. As concentrações de oleandrina não apresentaram diferença estatisticamente significante entre as variedades de flores
Nerium oleander is an ornamental plant responsible for poisoning in animals and humans. All parts of the plant contain cardiotoxic glycosides, mainly oleandrin. Some authors suggest that the toxicity of the plant would present a dependence on flower color. The objective of this study was to determine whether there is variation in the concentration of oleandrin in the leaves of N. oleander specimens of different colors of flowers. Ten samples of leaves from each variety of flower (white, pink and red) were collected. Oleandrin levels were determined by HPLC-UV after extraction with methanol, precipitation with lead acetate and separation with C18 cartridge. The average concentration obtained in all analyzed leaves was 4.89mg g-1, being 6.20±4.08mg g-1 in variety of white flowers, 4.16±3.44mg g-1 in pink flowers, and 4.31±1.99mg g-1 in red flowers. The concentrations of oleandrin did not show statistically significant difference between the varieties of flowers

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.
