ABSTRACT Purpose: To explore the role and mechanism of curcumin (Cur) in reducing oxidative stress damage in rats with nephrolithiasis induced by ethylene glycol (EG). Methods: Thirty male rats were divided into normal control, model, positive (10% potassium citrate), Cur-10 (10 mg/kg curcumin) and Cur-20 (20 mg/kg curcumin) groups. Results: The results of kidney tissue section stained by hematoxylin-eosin and von Kossa showed that curcumin treatment can inhibit the formation of kidney stones. The biochemical test results showed that the urea (Ur), creatinine (Cr), uric acid (UA), inorganic phosphorus and Ca2+ concentrations in urine decreased after being treated with curcumin. There were significant differences between different doses of curcumin (P < 0.05). Compared with the Cur-10 group, Cur-20 had a more significant inhibitory effect on malondialdehyde (MDA) (P < 0.05). In addition, reverse transcription polymerase chain reaction (PCR) detection and immunohistochemical results indicated that the osteopontin (OPN) in the kidney was significantly reduced after curcumin treatment. Conclusions: Curcumin could reduce the oxidative stress damage caused by EG-induced kidney stones.

Abstract Purpose To investigate whether heat shock protein 90 (HSP90) is involved in complement regulation in ischemic postconditioning (IPC). Methods The left coronary artery of rats underwent 30 min of occlusion, followed by 120 min of reperfusion and treatment with IPC via 3 cycles of 30s reperfusion and 30s occlusion. The rats were injected intraperitoneally with 1 mg/kg HSP90 inhibitor geldanamycin (GA) after anesthesia. Eighty rats were randomly divided into four groups: sham, ischemia-reperfusion (I/R), IPC and IPC + GA. Myocardial infarct size, apoptosis index and the expression of HSP90, C3, C5a, tumor necrosis factor (TNF)-alpha, interleukin (IL)-1β and c-Jun N-terminal kinase (JNK) were assessed. Results Compared with the I/R injury, the IPC treatment significantly reduced infarct size, release of troponin T, creatine kinase-MB, and lactate dehydrogenase, and cardiomyocyte apoptosis. These beneficial effects were accompanied by a decrease in TNF-α, IL-1β, C3, C5a and JNK expression levels. However, all these effects were abrogated by administration of the HSP90 inhibitor GA. Conclusion HSP90 exerts a profound effect on IPC cardioprotection, and may be linked to the inhibition of the complement system and JNK, ultimately attenuating I/R-induced myocardial injury and apoptosis.

Abstract: The great potential of crowdsourcing contest is bringing the issue of how to sustain solvers’ participation intention to be a hot topic in research and practice. This study uses the framework of Expectation-confirmation model to explain solvers’ continuance intention. Due to the uncertainties inherent in crowdsourcing contest, trust, a salient psychological belief, should be taken into account with the Expectation-confirmation model framework to predict solvers’ continuance intention. In addition, the intensive demand of intelligence and competition indicate interaction and fairness as two crucial factors for solvers to achieve expectation, thus suggesting that they may have influence on the confirmation level. Corresponding to these challenges, this study integrates platform trust, interaction, and perceived fairness into an extended Expectation-confirmation model to examine solvers’ continuance intention. Using a sample of 306 solvers, empirical results show that satisfaction, perceived benefits, and platform trust, which are positively associated with confirmation, are three significant antecedents of solvers’ continuance intention. Confirmation is further found to be significantly determined by interaction and perceived fairness. These findings provide some implications in both theory and practice for understanding the process of triggering sustained intention with an Expectation-confirmation model framework in crowdsourcing contest.
