ABSTRACT Objectives: Currently, not much is known about the interactions between voice and growth hormone (GH). We have described large kindred with isolated GH deficiency (IGHD) due to a GHRH receptor mutation, resulting in severe short stature and high-pitched voice. These IGHD individuals have little interest in GH treatment, as they consider themselves “short long-lived people”, rather than patients. Interestingly, they report normal general quality of life, but they rate their Voice-Related Quality of Life (V-RQOL) as low. Here, we assessed the social and auditory-perceptual impacts of artistic-intervention voice therapy with semioccluded vocal tract exercises (SOVTE) and choral singing, on their voices. Material and methods: Seventeen GH-naïve adult IGHD individuals were enrolled in a single-arm interventional pre-post study with 13 weekly sessions of choir singing over 90 days. Outcome measures were V-RQOL scores, self-assessment of voice, and auditory-perceptual analysis (GRBAS scale, G: grade of the severity of dysphonia; R: roughness; B: breathiness; A: asthenia; and S: strain). Results: Marked improvements in total (p = 0.0001), physical (p = 0.0002), and socioemotional (p = 0.0001) V-RQOL scores and in self-assessment of voice (p = 0.004) were found. The general grades of vocal deviation (p = 0.0001), roughness (p = 0.0001), breathiness (p = 0.0001) and strain (p = 0.0001) exhibited accentuated reductions. Conclusions: Voice therapy with semioccluded vocal tract exercises and choral training improved social impact and perceptual voice assessments in IGHD subjects and markedly improved their voice-related quality of life. This is particularly important in a setting where GH replacement therapy is not widely accepted.

Abstract Background Eugenol shows both antibacterial and antiparasitic activities, suggesting that it might be evaluated as an option for the treatment of praziquantel-resistant schistosome. Methods The in vitro activities of three eugenol derivatives (FB1, FB4 and FB9) on adult worms from Schistosoma mansoni were examined by fluorescence and scanning electron microscopy to analyze effects on the excretory system and integument damage, respectively. Biochemical tests with verapamil (a calcium channel antagonist) and ouabain (a Na+/K+-ATPase pump inhibitor) were used to characterize eugenol derivative interactions with calcium channels and the Na+/K+-ATPase, while in silico analysis identified potential Na+/K+-ATPase binding sites. Results The compounds showed effective doses (ED50) of 0.324 mM (FB1), 0.167 mM (FB4), and 0.340 mM (FB9). In addition, FB4 (0.322 mM), which showed the lowest ED50, ED90 and ED100 (p < 0.05), caused the most damage to the excretory system and integument, according to both fluorescence and scanning electron microscopy analysis. The death of adult worms was delayed by ouabain treatment plus FB1 (192 versus 72 hours) and FB9 (192 versus 168 hours), but the response to FB4 was the same in the presence or absence of ouabain. Besides, no changes were noted when all of the eugenol derivatives were combined with verapamil. Moreover, FB1 and FB9 inhibited Na+/K+-ATPase activity according to in silico analysis but FB4 did not show a time-dependent relationship and may act on targets other than the parasite Na+/K+-ATPase. Conclusion Eugenol derivatives, mainly FB4 when compared to FB1 and FB9, seem to act more effectively on the integument of adult S. mansoni worms.

OBJECTIVES: Few studies have investigated whether post-exercise hypotension (PEH) after concurrent exercise (CEX) is related to changes in cardiac output (Q) and systemic vascular resistance (SVR) in older individuals. We tested whether PEH after a single bout of CEX circuits performed in open-access facilities at the Third Age Academies (TAA) in Rio de Janeiro City (Brazil) would be concomitant with decreased Q and SVR in individuals aged ≥60 years with prehypertension. Moreover, we assessed autonomic modulation as a potential mechanism underlying PEH. METHODS: Fourteen individuals (age, 65.8±0.9 y; systolic/diastolic blood pressure [SBP/DBP], 132.4±12.1/72.8±10.8 mmHg; with half of the patients taking antihypertensive medications) had their blood pressure (BP), heart rate (HR), Q, SVR, HR variability (HRV), and spontaneous baroreflex sensitivity (BRS) recorded before and 50 min after CEX (40-min circuit, including seven stations of alternate aerobic/resistance exercises at 60-70% HR reserve) and non-exercise control (CONT) sessions. The study protocol was registered in a World Health Organization-accredited office (Trial registration RBR-7BWVPJ). RESULTS: SBP (Δ=−14.2±13.1 mmHg, p=0.0001), DBP (Δ=−5.2±8.2 mmHg, p= 0.04), Q (Δ=−2.2±1.5 L/min, p=0.0001), and BRS (Δ=−3.5±2.6 ms/mmHg; p=0.05) decreased after CEX as compared with the CONT session. By contrast, the HR increased (Δ=9.4±7.2 bpm, p<0.0001), and SVR remained stable throughout the postexercise period as compared with the CONT session (Δ=0.10±0.22 AU, p=0.14). We found no significant difference between the CEX and CONT with respect to the HRV indexes reflecting autonomic modulation. CONCLUSION: CEX induced PEH in the older individuals with prehypertension status. At least in the first 50 min, PEH occurred parallel to the decreased Q and increased HR, while SVR was not different. The changes in autonomic outflow appeared to be unrelated to the acute cardiac and hemodynamic responses.

Abstract Background: Snakebite envenoming can be a life-threatening condition, for which emergency care is essential. The Bothrops (lancehead) genus is responsible for most snakebite-related deaths and permanent loss of function in human victims in Latin America. Bothrops spp. venom is a complex mixture of different proteins that are known to cause local necrosis, coagulopathy, and acute kidney injury. However, the long-term effects of these viper envenomings have remained largely understudied. Case presentation: Here, we present a case report of a 46-years old female patient from Las Claritas, Venezuela, who was envenomed by a snake from the Bothrops genus. The patient was followed for a 10-year period, during which she presented oliguric renal failure, culminating in kidney failure 60 months after the envenoming. Conclusion: In Latin America, especially in Brazil, where there is a high prevalence of Bothrops envenoming, it may be relevant to establish long-term outpatient programs. This would reduce late adverse events, such as chronic kidney disease, and optimize public financial resources by avoiding hemodialysis and consequently kidney transplantation.

Abstract Real-time PCR (qPCR) has been used for rapid identification of Salmonella Typhimurium and Staphylococcus aureus in dairy foods, but is unable to differentiate viable and unviable pathogens. Ethidium bromide monoazide (EMA), a DNA-intercalating agent, can detect only viable cells because selectively enter cells considered unviable and bind to their DNA, inhibiting its amplification during qPCR. The objective was to establish a protocol for detection of viable Salmonella Typhimurium and S. aureus, experimentally inoculated in coalho cheese, by the use of EMA combined with qPCR. The protocol was effective for the identification of viable Salmonella Typhimurium in coalho cheese but not for the S. aureus cells. Concentrations of viable Salmonella Typhimurium cells of 1 CFU/10 g of coalho cheese could be detected. The monoplex protocol enables the rapid and specific identification of viable Salmonella Typhimurium in coalho cheese, making it an alternative method for the quality and safety control of cheeses.

Two new synthetic melanin-like oligomers inspired by the synthesis of pheomelanin (cys-DOPA) were prepared using L-DOPA as a precursor and the amino acids serine (ser-DOPA) and threonine (thr-DOPA) for modifications. The products obtained exhibited appreciable antioxidant activity. The compounds were characterized by liquid chromatography-mass spectrometry (LC-MS), elemental analysis and infrared spectroscopy. The pKa values of the groups present in each melanin suspension were determined in aqueous solution by potentiometric and spectrophotometric titration. Cytotoxicity assays showed an IC50 > 500 mg L-1 resulting in non-toxic compounds for living cells. Finally, cys-DOPA, ser-DOPA and thr-DOPA exhibited antioxidant activity determined by the DPPH scavenging assay, with EC50 = 7.69, 1.02 and 27.60 mg L-1, respectively.

Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

For the study of thermal expansion in various materials it is necessary to know the coefficient of linear expansion, but in some cases it is difficult to measure the final temperature of the sample under consideration. We present a low-cost experimental setup in order to show a teaching methodology to address the problem of measuring this final temperature. The methodology uses materials that are easy to find, and the positive result are described in this paper.
The activity of artisanal fishing for the sea-bob shrimp, using the method of motorized dragnet with rings, has been practised on the Santa Catarina coast, where it generates jobs and income, and helps keep the traditional Azorean cultural tradition alive, since 1960. With the aim of analyzing the quality and quantity of the ichthyofauna bycatch, two trawls were carried out per isobath (10-20-30 m), seasonally, between November 2009 and August 2010 (i.e. summer, spring and winter), for 20 minutes each, at an average speed of two knots, and the bottom water temperatures and salinities recorded. The captures totalled 10868 fish (208.34 kg), distributed among 31 families and 62 species, with a fish/shrimp ratio of 5.19/1 kg. The most abundant family was Sciaenidae (86.13%), followed by Batrachoididae (2.70%) and Trichiuridae (2.44%). The diversity and evenness indices presented similar patterns of variation, the highest values occurring in spring at 30 m and the lowest in winter at 20 m. Cluster analysis led to the creation of four groups; one of them small and dominated by the Sciaenidae Stellifer brasiliensis, S. rastrifer, Paralonchurus brasiliensis, Isopisthus parvipinnis and Larimus breviceps. The young of this and other species, which are of little or no commercial value, along with tiny shrimps and other macroinvertebrates that enter the dragnets, are thrown back into the sea, usually dead. Besides the negative impact on the food chain, this continued practice could exacerbate the situation of artisanal fishermen even further, with the gradual, future decline of these resources.

The purpose of this study was to compare goniometric values for forelimb and hind limbs in clinically healthy Santa Ines sheep with two ages (young and adult). Thirty female Santa Ines sheep were separated into two groups of 15 animals each: Group 1 with 6 to 12-month-old sheep (classified as young), and Group 2 with 3 to 6-year-old sheep (classified as adult). Maximum flexion, maximum extension, and range of motion (ROM) of the shoulder, elbow, carpal, hip, stifle, and tarsal joints, in the right and left limbs were measured with a standard transparent plastic goniometer. The measurements were performed in triplicate by two independent investigators with the sheep in a standing position. There was no statistically significant difference between the right and left sides, between investigators or between groups. In conclusion, in clinically healthy sheep the goniometric values were not influenced by age.

BACKGROUND: Varicose vein surgery of the lower limbs is one of the most frequent procedures carried out in the Brazilian public health system (Sistema Único de Saúde), especially during task forces focused on elective surgeries. However, detailed information on the results of such initiatives are absent in the literature, particularly with regard to the population that benefits from the task forces. OBJECTIVES: To present and discuss the profile of patients operated on during a task force for varicose vein surgery carried out in a public health institution. METHODS: The patients were selected from a database organized by the Brazilian Health Ministry for the program of elective surgeries, and they had no previous link with the hospital institution. A protocol for objective evaluation with the clinical data and demographic profile of the patients was applied. The patients were staged according to CEAP classification. RESULTS: From September 2005 to January 2006, 100 patients (106 procedures) underwent surgery, females being predominant (85% of the cases). Mean age was 35±9.8 years. Predominant schooling level among patients was primary school (53%). C2 (49.5%) and C3 (39%) were the most frequent CEAP classes in the clinical evaluation of lower limbs. A total of 106 surgeries was performed (100 patients). The preferred anesthetic technique was block anesthesia (93% of the procedures). Six patients were submitted to two different procedures due to great volume of lower limb varicose veins; 33 patients were submitted to internal saphenous vein stripping (four cases of segmental saphenous vein stripping); and eight patients were submitted to bilateral total saphenous vein stripping. CONCLUSION: Profile of the population that benefited from the task force for varicose vein surgeries was: young symptomatic female patients, with low schooling level, moderate stages of disease, especially in CEAP clinical classes C2 and C3, and who require surgeries of moderate and long duration for the treatment of their diseases.

The mechanical properties of composites with dispersed diamond particles in epoxy matrix cured with different proportions of hardener to monomer ratio, characterized by the resin/hardener ratio (phr) were studied. An investigation on the thermo-mechanical behavior of these composites was also carried out by dynamical mechanical analysis (DMA). In the present work a complete evaluation of the mechanical properties was carried in a wide interval of phr associated with possible technological applications. Composites with up to 30 wt. % of diamond particles dispersed in type DGEBA/TETA epoxy matrix were fabricated with phr ratios varying from 7 to 21. For all investigated conditions, the composite strength decreased with the amount of incorporated diamond. Matrices with phr above the stoichiometric 13 were associated with composites with better mechanical performance. The DMA results showed that the storage modulus increases with the amount of diamond particles incorporated in the composite. The values obtained for the delta tangent, allowed an evaluation of possible mechanisms that contribute to the thermal mechanical performance of these composites.

Coffee is one of the most valuable agricultural commodities and ranks second on international trade exchanges. The genus Coffea belongs to the Rubiaceae family which includes other important plants. The genus contains about 100 species but commercial production is based only on two species, Coffea arabica and Coffea canephora that represent about 70 % and 30 % of the total coffee market, respectively. The Brazilian Coffee Genome Project was designed with the objective of making modern genomics resources available to the coffee scientific community, working on different aspects of the coffee production chain. We have single-pass sequenced a total of 214,964 randomly picked clones from 37 cDNA libraries of C. arabica, C. canephora and C. racemosa, representing specific stages of cells and plant development that after trimming resulted in 130,792, 12,381 and 10,566 sequences for each species, respectively. The ESTs clustered into 17,982 clusters and 32,155 singletons. Blast analysis of these sequences revealed that 22 % had no significant matches to sequences in the National Center for Biotechnology Information database (of known or unknown function). The generated coffee EST database resulted in the identification of close to 33,000 different unigenes. Annotated sequencing results have been stored in an online database at <A HREF="http://www.lge.ibi.unicamp.br/cafe">http://www.lge.ibi.unicamp.br/cafe</A>. Resources developed in this project provide genetic and genomic tools that may hold the key to the sustainability, competitiveness and future viability of the coffee industry in local and international markets.
