Methodology for seed germination and vigor tests of Bowdichia virgilioides

ABSTRACT Bowdichia virgilioides Kunth., a member of the Fabaceae family, is known as sucupira or paricarana. In addition to its medicinal properties, it has heavy, fibrous wood with ornamental features and high strength. Consequently, the present study aimed to evaluate the effect of the amount of water of the substrate, substrate temperature and the effect of different light regimes on germination and vigor of B. virgilioides seeds. In the first experiment, the volumes of water used for wetting the substrate were equivalent to 1.5, 2.0, 2.5, 3.0 and 3.5 times the weight of the dry substrate, at constant temperatures of 25, 30 and 35 °C, and alternating between 20-30 °C. In the second experiment, the tested light regimes were white, green, red, far-red and dark light at constant tempera tures of 25, 30 and 35 °C and alternated between 20-30 °C. A completely randomized design was used in both experiments, in a 5 x 4 factorial model, with four replicates of 25 seeds each. The variables analyzed included percentage, first count and index of germination speed, length and dry mass of roots and shoot length. The seeds of B. virgilioides germinated under all light regimes, and classified as neutral photoblastics. The combination of a 30 °C temperature and volume of water 2.5 times larger than the weight of the dry substrate were shown to be the most appropriate conditions to perform the seed germination test of this species.
RESUMEN Bowdichia virgilioides Kunth. perteneciente a la familia Fabaceae, es conocida como sucupira o pariarana. Además de sus propiedades medicinales posee madera pesada, fibrosa, con características decorativas y de gran resistencia. En este contexto. el presente trabajo tuvo como objetivo evaluar el efecto de la cantidad de agua en el sustrato, temperaturas y diferentes regímenes de luz en la germinación y vigor de las semillas de B. virgilioides. En el primer experimento, los volúmenes de agua utilizados para humedecimiento del sustrato fueron equivalentes a 1,5, 2,0, 2,5, 3,0 y 3,5 veces el peso del sustrato seco. en las temperaturas constantes de 25, 30 y 35 °C, y alternada de 20-30 °C. En el segundo experimento. los regímenes de luz probados fueron el de luz blanca, verde, roja, rojo-distante y oscuro continuo, a las temperaturas constantes de 25, 30 y 35 °C y alternada de 20-30 °C. Se adoptó en ambos experimentos el delineamiento enteramente al azar. en factorial 5 x 4, con cuatro repeticiones de 25 semillas cada una. Las variables analizadas fueron porcentaje, primer conteo e índice de velocidad de germinación, longitud y masa seca de raíces y parte aérea. Las semillas de B. virgilioides germinan en todos los regímenes de luz, pudiendo ser clasificadas como fotoblásticas neutras y la combinación entre la temperatura de 30 °C y el volumen de agua de 2,5 veces el peso del sustrato seco es la condición más apropiada para realizar la prueba de germinación de semillas de esta especie.

AgAu Nanotubes: Investigating the Effect of Surface Morphologies and Optical Properties over Applications in Catalysis and Photocatalysis

We herein report the preparation of AgAu nanotubes displaying controlled surface morphologies and optical properties by varying the reaction temperature during the galvanic reaction between Ag nanowires and AuCl4-(aq). As the AgAu nanotubes presented similar sizes and compositions, they enabled us to isolate the influence of surface morphology and optical properties over their catalytic and plasmonic photocatalytic activities towards methylene blue oxidation. At 25 °C, AgAu nanotubes (AgAu 25) presented branched walls and surface plasmon resonance (SPR) band with low intensities in the visible were obtained. However, at 100 °C, the AgAu nanotubes (AgAu 100) presented smooth surfaces and SPR bands that closely matched the emission spectra of a commercial halogen-tungsten lamp. The AgAu 25 nanotubes displayed better catalytic performances in classical heterogeneous catalysis as a result of its branched walls that lead to increased surface areas relative to the smooth nanotubes. Conversely, AgAu 100 nanotubes showed better activities in plasmonic photocatalysis due to its broader and more intense SPR bands. Thus, our results demonstrate the potential of the control over morphological and optical features towards the optimization of distinct catalytic phenomena.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Ocorrência de anticorpos e fatores de risco para infecção por Toxoplasma gondii em cães, nas cidades de Lages e Balneário Camboriú, Santa Catarina, Brasil

Com os objetivos de conhecer a ocorrência de anticorpos e identificar fatores de risco para a infecção por Toxoplasmagondii em cães, nas cidades de Lages e Balneário Camboriú, SC, amostras de sangue de 400 cães domiciliados foram processadas para a detecção de anticorpos contra T. gondii pela técnica da imunofluorescência indireta (RIFI). Dados referentes à raça, idade, sexo, tipo de dieta, ambiente, presença de gatos e acesso à rua foram obtidos por meio de questionário. Análise estatística foi realizada (teste de χ²). Dos 400 cães, 89 (22,3%) apresentaram anticorpos contra T. gondii. Dos cães de Lages, 52 (26%) foram positivos para T. gondii, enquanto os de Balneário Camboriú, 37 (18,5%)foram sororreagentes. Não foram observadas diferenças estatísticas entre as prevalências de T. gondii nos dois municípios. Maior ocorrência de sororreagentes para toxoplasmose foi verificada entre os cães sem raça definida (P = 0,002), que tinham acesso à rua (P = 0,003) e que recebiam dieta caseira (P = 0,028). Forte tendência para reagentes ao T. gondii foi observada entre os animais adultos, indicando exposição pós-natal ao agente, e entre os cães em contato com felinos.
The aim of this study was to identify the occurrence of antibodies and risk factors for infection for Toxoplasma gondii in dogs in the cities of Lages and Balneário Camboriú, Santa Catarina State, Brazil. Blood samples from 400 domiciliated dogs were processed for antibody detection against T. gondii by indirect immunofluorescence antibody test (IFAT). The following variables were obtained by questionnaire: breed, age, sex, diet, environment, presence of cats and street access. A Chi-Square test was performed to verify the association between the seroprevalence with the studied variables. From all evaluated dogs, 89 (22.3%) had antibodies against T. gondii. A total of 52 (26%) and 37 (18.5%) dogs were positive for T. gondii in Lages and Balneário Camboriú, respectively. No statistical differences were observed on the prevalences of T. gondii between both cities. Greater occurrences of seropositives for toxoplasmosis were observed in mixed-breed dogs (P = 0.002), who had access to the street (P = 0.003) and received homemade diet (P = 0.028). A strong trend for seropositivity against T. gondii was observed among adult dogs, indicating post-natal exposure, and among those that have previous contact with cats.

We investigate the nature of one-electron eigenstates in power-law diluted chains for which the probability of occurrence of a bond between sites separated by a distance r decays as p(r) = 1/r1+sigma. Using an exact diagonalization scheme on finite chains, we compute the spreading of an initially localized wave-packet, the time dependent participation number as well as the return probability. Our results show the existence of a phase of extended states. By considering the scale invariance of the fluctuations of the participation number at the Anderson transition, we obtained that extended states emerges for sigma < 0.68. This limiting value is larger than the one reported in the literature for the emergence of extended states in one-dimensional Anderson models with power-law decaying couplings.

Oito reprodutores suínos foram divididos em três grupos e inoculados com Toxoplasma gondii [GI (n=3) 1.5x10(4) oocistos cepa P; GII (n=3) 1.0x10(6) taquizoítos cepa RH, e GIII (n=2) controle, não inoculados]. Exames clínicos, hematológicos, de parasitemia e sorológicos foram realizados para avaliar a infecção toxoplásmica. Pesquisa do parasito no sêmen, por meio do bioensaio e pela técnica da PCR, e em órgãos do sistema reprodutor (bioensaio e imunohistoquímica) foi realizada. Sangue e sêmen foram colhidos nos dias -2, -1, 1, 3, 5, 7, 9, 11, 14, e semanalmente até o 84º dia pós-infecção (DPI). Nenhuma alteração clínica ou hematimétrica foi observadda nos animais. Parasitemia foi detectada em um animal inoculado com oocistos no 7º DPI e em outro inoculado com taquizoítos (GII) nos 3º e 49º DPI. A sorologia revelou a presença de anticorpos contra T. gondii nos animais inoculados com oocistos ou taquizoítos no 7º DPI com títulos de 1:256 e 1:64, que atingiram picos de 1:4096 nos dias 11 e 9, respectivamente. O bioensaio revelou a presença do parasita em amostras seminais de um animal inoculado com oocistos (GI) nos 3º, 49º e 56º DPI, e de dois animais infectados com taquizoítos (GII), um deles no 5º DPI e os dois ao 49º DPI. Pela PCR, o DNA de T. gondii foi detectado no sêmen dos Suínos 1 e 3 inoculados com taquizoítos e oocistos, respectivamente. A imunohistoquímica revelou T. gondii em órgão do aparelho reprodutor dos Suínos 1 e 2, inoculados com taquizoítos e oocistos, respectivamente. Esses achados sugerem a possibilidade da ocorrência da transmissão venérea do T. gondii em suínos.
Eight reproductive boars were divided into three groups and inoculated with Toxoplasma gondii [GI (n=3) 1.5x10(4) oocysts strain P; GII (n=3) 1.0x10(6) tachyzoites strain RH; and GIII (n=2) non-inoculated control]. Clinical, hematological, parasitemia and serological tests and studies of the parasite in the semen through bioassay and PCR, and in reproductive organs (Bioassay and immunohistochemical analyses) were conducted to evaluate the toxoplasmic infection. Blood and semen were collected on day -2, -1, 1, 3, 5, 7, 9, 11, 14 and weekly up to 84 days post-inoculation (DPI). No clinical or hematimetric alteration was observed in the boars. Parasitemia was detected in one boar inoculated with oocysts at the 7th DPI and in another boar infected with tachyzoites (GII) at the 3rd and 49th DPI. Serological tests revealed antibodies against T. gondii in animals inoculated with oocysts or tachyzoites at the 7th DPI with dilutions of 1:256 and 1:64, which reached peaks of 1:4096 at day 11 and 9, respectively. The bioassays revealed the presence of the parasite in semen samples of a boar inoculated with oocysts (GI) at 3, 49 and 56 DPI and from two boars infected with tachyzoites (GII), one animal at 5 and two animals at 49 days DPI. Mice inoculated with semen from the control group (GIII) remained serologically negative. PCR analysis showed T. gondii DNA in the semen of Boar 1 and Boar 3 inoculated with tachyzoites and oocysts, respectively. The immuno-histochemical tests showed T. gondii in the reproductive organs of Boar 1 and Boar 2, inoculated with tachyzoites and oocysts, respectively. These findings suggest the possible occurrence of venereal transmission of T. gondii in swine.
