RESUMO A fusariose do trigo, ou giberela, causada por um complexo de Fusarium spp., é uma importante doença no Paraguai. Dentre as estratégias utilizadas para o seu controle, a resistência genética é considerada altamente eficiente e de baixo custo. Objetivou-se avaliar e comparar o efeito de F. graminearum em seis genótipos de trigo, incluindo duas variedades testemunha, em duas datas de semeadura. Os genótipos foram inoculados artificialmente na espiga, por meio da injeção de um pool de quatro isolados patogênicos de F. graminearum, na fase de floração. O desenvolvimento da fusariose foi avaliado pela pontuação da incidência e severidade da doença, porcentagem de espiguetas doentes e grãos danificados, bem como pela área sob a curva de progresso da doença. Além da infecção de grãos, foi determinado o seu impacto no desenvolvimento de micotoxinas (deoxinivalenol) e sua interação com os genótipos. Os resultados identificaram uma linhagem avançada (Lin 84), com nível de resistência à fusariose comparável ao das fontes universalmente conhecidas de resistência (Sumai 3 e Frontana). Os outros três genótipos (Caninde 11, Caninde 12 e Caninde 21), apesar de apresentarem maior potencial de rendimento de grãos, foram avaliados como moderadamente suscetíveis a suscetíveis. Esses resultados sugerem que, embora seja possível transferir a resistência da fusariose do trigo para um tipo agronômico melhorado, combinar a resistência com um maior potencial de rendimento de grãos continua sendo um desafio contínuo.
ABSTRACT Fusarium head blight (FHB), or scab, caused by a Fusarium spp. complex, is an important wheat disease in Paraguay. Among the strategies used to control it, the genetic resistance is considered highly efficient and cost effective. This study aimed to evaluate and compare the effects of F. graminearum on six wheat genotypes, including two comparison varieties, in two seeding dates. The genotypes were artificially inoculated in the spike, at the flowering stage, by injecting a pool of four pathogenic F. graminearum isolates. The FHB development was evaluated by scoring the disease incidence and severity, percentage of diseased spikelets and damaged kernels, as well as using the area under the disease progress curve. Besides the kernel infection, its impact on the development of mycotoxins (deoxynivalenol) and interactions with the genotypes were also evaluated. The results identified an advanced breeding line (Lin 84) with a resistance level to FHB comparable to that of the universally known resistance sources (Sumai 3 and Frontana). The other three genotypes (Caninde 11, Caninde 12 and Caninde 21), in spite of presenting a higher grain yield potential, were evaluated as moderately susceptible to susceptible. These results suggest that, although it is possible to transfer the FHB resistance to a higher agronomic type, combining such resistance with a higher grain yield potential remains an ongoing challenge.

Effects of bleaching using 10% carbamide peroxide with calcium or amorphous calcium phosphate on enamel mineral content and hardness

RESUMO Este estudo avaliou o conteúdo mineral do esmalte e a microdureza superficial antes e após o tratamento clareador, utilizando peróxido de carbamida 10% (PC) contendo cálcio (Ca) ou fosfato de cálcio amorfo (ACP) em sua composigao. Trinta e seis espécimes de esmalte bovino foram alocados aleatoriamente em 3 grupos (n = 12) de acordo com os tratamentos clareadores: G1 - Opalescence PF 10% (CP), G2 -NiteWhite (CP+ACP); e G3 - Opalescence PF (10%) com cálcio (CP + CA). O agente clareador foi aplicado na superficie do esmalte por 6 h/dia por um periodo de 21 dias. A superficie do esmalte foi avaliada por microdureza Knoop (KNH) e espectrometria de fluorescencia de raios X micro-dispersiva (p-EDXRF) no inicio e após o tratamento clareador. Os dados foram analisados estatisticamente pelo teste ANOVA de medidas repetidas e Tukey (a = 0,05). Houve uma diminuigao significativa da microdureza após os tratamentos clareadores para todos os grupos estudados, mas nao houve diferenga entre os diferentes géis. Nao houve diferenga da relagao Ca/P mensurada por p-EDXRF para todos os grupos nos tempos estudados; no entanto, o grupo CP+CA apresentou menor valor comparado ao grupo CP+ACP. O grupo CP foi similar aos grupos CP+ACP e CP+CA. Portanto, pode-se concluir que houve redugao significativa da microdureza do esmalte após o clareamento, independente da presenga de cálcio ou APC na composigao dos géis, embora nao tenha havido alteragao significando na relagao Ca/P do esmalte após o clareamento. Isto indica um potencial erosivo dos géis clareadores, causando o amolecimento sem perda da estrutura do esmalte, independente da presenga dos íons cálcio e ACP.
ABSTRACT This study evaluated enamel mineral content and surface microhardness before and after bleaching treatment using 10% carbamide peroxide (CP) containing calcium (Ca) or amorphous calcium phosphate (ACP). Thirty-six bovine slabs were randomly allocated into 3 groups (n = 12) according to bleaching treatment: G1 - Opalescence PF 10% (CP), G2 -NiteWhite ACP (CP+ACP), and G3 - Opalescence PF (10%) with calcium (CP+CA). The bleaching agent was applied on enamel surface for 6 h/day over a period of 21 days. Enamel surface was evaluated by Knoop microhardness (KNH) and micro energy-dispersive X-ray fluorescence spectrometry (p-EDXRF) at baseline and at after bleaching treatment. Data were statistically analyzed by repeated measures ANOVA and Tukey's test (a = 0.05). There was a significant decrease in microhardness after bleaching treatments for all study groups, but no difference between bleaching gels. There was no difference in the Ca/P ratio measured by p-EDXRF for all groups at the study times, but the mean value was lower in group CP+CA than in group CP+ACP. Group CP was similar to both CP+ACP and CP+CA. It can be concluded that enamel microhardness decreased after the bleaching process, regardless of the presence of calcium or ACP, but there was no significant change in the Ca/P ratio of enamel after bleaching for each tested gel. This indicates that the bleaching gels have erosive potential, causing softening of enamel without promoting surface loss, regardless of the presence of calcium of ACP ions.

Chalcone-quinolinone hybrid compounds, as well as the synthesis of such compounds, have few reports in the literature. Such compounds may be quite useful in therapeutics, since various biological activities are reported for both chalcones and quinolinones. In the present work, several novel chalcone-quinolinone compounds have been synthesized. The reaction conditions developed allowed to obtain the compounds in a single step of synthesis from the chalcones. The products precipitated pure and were isolated by filtration. The yields of such reactions, from 45 to 94%, were promising. The product structures were confirmed by nuclear magnetic resonance (NMR) and electrospray ionization mass spectrometry (ESI-MS) techniques. Their antitumor activities were evaluated in HCT-116 (colon) cell lines by the 3-(4,5-dimethyl-2-thiazole)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) test. The half maximal inhibitory concentration (IC50) values obtained for the most active chalcones were between 2.9 and 7.5 and for active quinolinone was 19.3 mg L-1. The antitumor activities results suggest that the class of compounds studied has potential for use in cancer research.

From the natural product 4-(4'-O-acetyl-α-L-rhamnosyloxy)benzylisothiocyanate (1), isolated from the flowers of Moringa oleifera Lam (Moringaceae), four new semisynthetic derivatives, N-[4-(4'-O-acetyl-α-L-rhamnosyloxy)benzyl]-2-(pyridinil-4-carbonil)hydrazine-1-carbothioamide (3), 4-(4'-O-acetyl-2',3'-dimesyloxy-α-L-rhamnosyloxy)benzylisothiocyanate (4), N-[(4'-O-acetyl-α-L-rhamnosyloxy)benzyl]hydrazinecarbothioamide (5), 4-[4'-O-acetyl-2',3'-O-bis(decanoiloxy)-α-L-rhamnosyloxy]benzylisothiocianate (6), and the known compound 4-(2',3',4'-O-triacetyl-α-L-rhamnosyloxy)benzylisothiocyanate (2), were obtained. All compounds were tested for their cytotoxicity against the tumor cell lines SF-295, HL-60, HCT-116 e PC-3. The natural product 1 and the semisynthetic derivatives 2 and 4 were the most active compounds (IC50 from16.0 to 3.7 µmol L-1) against all tumor cell lines.

ABSTRACT Natural steroids and triterpenes such as b-sitosterol, stigmasterol, lupeol, ursolic and betulinic acids were transformed into its hexanoic and oleic esters, to evaluate the influence of chemical modification towards the cytotoxic activities against tumor cells. The derivatives were evaluated against five tumor cell lines [OVCAR-8 (ovarian carcinoma); SF-295 (glioblastoma); HCT-116 (colon adenocarcinoma); HL-60 (leukemia); and PC-3 (prostate carcinoma)] and the results showed only betulinic acid hexyl ester exhibits cytotoxic potential activity.

Resumo Apresenta-se uma síntese atualizada de informações sobre algas no Brasil objetivando refinar os dados reunidos até o presente, bem como avaliar os avanços sobre o conhecimento da diversidade de algas no país desde a publicação do Catálogo de Plantas e Fungos do Brasil. Os resultados de 2015 mostraram um acréscimo de 1.250 espécies (35.7%) a um total de 4.747 em relação a 2010. As classes mais diversas em número de espécies foram Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae e Euglenophyceae. Bacillariophyceae e Cyanophyceae tiveram o maior acréscimo de espécies no intervalo de cinco anos. A região Sudeste e Sul foram as mais diversas, porém, as regiões Nordeste com os estados do Piauí e Sergipe e Centro-Oeste com os estados de Mato Grosso, Goiás e Distrito Federal destacaram-se no cenário da biodiversidade nacional. Apesar da escassez de taxonomistas e da infraestrutura limitada, os resultados obtidos evidenciaram um avanço significativo no conhecimento da diversidade de algas no país nesse período de cinco anos, iniciando uma mudança quanto as diferenças geográficas regionais.
Abstract An updated synthesis of cyanobacteria and algae information is presented for Brazil aiming to refine the data gathered to date and evaluate the progress of the biodiversity knowledge about these organisms in the country since the publication of the Catálogo de Plantas e Fungos do Brasil. The results of 2015 showed an increase of 1,250 species (35.7%) when compared to 2010, reaching a total of 4,747 species. The most diverse classes in species number were the Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae and Euglenophyceae. Bacillariophyceae and Cyanophyceae had the highest increase in species number in the five-year interval. The Southeast and South regions were the most diverse, however, the Northeast, with the states of Piauí and Sergipe, and the Central-west region, with Mato Grosso, Goiás and Distrito Federal, also stood out in the national algal biodiversity scenario. Despite the shortage of taxonomists and limited infrastructure, the results showed a significant improvement in the knowledge regarding the diversity of cyanobacteria and algae in the country during the study period, starting to even out regional geographical differences caused by subsampling.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Controle biológico da murcha do tomateiro causada por Ralstonia solanacearum e Fusarium oxysporum f. sp. lycopersici por rizobactérias

A busca por alternativas ao uso intensivo de agrotóxicos no controle de doenças tem recebido grande atenção da pesquisa agrícola. Rizobactérias têm reconhecida capacidade de reduzir doenças em diversas culturas e promover de crescimento de plantas. O objetivo deste trabalho foi avaliar a eficácia de seis isolados de rizobactérias, pré-selecionadas, no controle de Ralstonia solanacearum e Fusarium oxysporum f. sp. lycopersici (FOL), em casa de vegetação e relacionar este comportamento a produção de compostos "in vitro". Foi avaliada a capacidade de estas rizobactérias produzirem quitinases, amilases, lipases, compostos antibióticos e de solubilizar fosfato. A microbiolização das sementes com a rizobactéria DFs1421 (Pseudomonas sp.) reduziu os valores de AACPD de murcha bacteriana em ambos os ensaios (36,6 e 91,7% no primeiro e segundo ensaios respectivamente). Este controle pode ser associado à produção de compostos responsáveis pela antibiose observada "in vitro". Isolados de Streptomyces (DFs1296 e DFs1315), bem como de Bacillus (DFs1414) e o indutor químico ASM reduziram significativamente a murcha de fusário, variando de 22,5 a 76%. O controle observado por parte das rizobactérias pode ser atribuído à atividade quitinolítica e/ou antibiótica por compostos voláteis.
Replacing the intensive use of pesticides for alternative methods in disease control has been an important aim for agricultural research. Rhizobacteria are known to be capable of reducing diseases levels in many crops and also of promoting plant growth. The objective of this work was to evaluate the efficacy of six pre-selected rhizobacteria isolates in controlling Ralstonia solanacearum and Fusarium oxysporum f. sp. lycopersici (FOL) under controlled conditions in a greenhouse, and to link such capacity to the "in vitro" production of selected biologically active compounds. The ability of these rhizobacteria isolates to produce chitinases, amylases, lipases, antibiotic compounds was investigated. Additionally, their ability in solubilizing phosphate was also checked. Microbiolization of seeds with one rhizobacterium DFs1421 (Pseudomonas sp.) reduced the tomato wilt AUDPC in both assays (36.6 and 91.7% in the first and second assays respectively). Such efficacy in wilt control is likely to be linked with the production of antibiotics as observed "in vitro". Streptomyces (DFs1296 and DFs1315) and Bacillus (DFs1414), and the chemical inducer (ASM) reduced significantly fusarium wilt ranging 22.5 to 76%. This may be owing to the observed chitinolytic activity and / or antibiosis in the presence of volatile compounds.

Consumo médio e digestibilidade aparente dos nutrientes do feno de aveia (Avena sativa L.) e ervilha (Pisum sativum L.) em ovinos submetidos a dois regimes alimentares

O experimento foi realizado com os objetivos de determinar o consumo médio diário (CMD), o coeficiente de digestibilidade aparente (CDA) dos nutrientes do feno de aveia e ervilha e avaliar o balanço de nitrogênio em ovinos. Foram utilizados dez ovinos machos, castrados, com peso médio de 36,64 kg, alojados em gaiolas para metabolismo, em um delineamento experimental inteiramente casualizado com dois tratamentos (T1 = 80% de feno de aveia + 20% de ervilha e T2 = 60% de feno de aveia + 40% de ervilha) e cinco repetições por tratamento. Os tratamentos foram avaliados sob dois regimes alimentares (consumo voluntário e restrito). O CMD de matéria seca (MS) e fibra em detergente neutro (FDN) não diferiram entre tratamentos, e o CMD de fibra em detergente ácido (FDA) e fibra bruta (FB), foram maiores para o tratamento 1. Para animais sob regime de consumo voluntário, as médias de consumo desses nutrientes foram maiores para aqueles recebendo ração com 60% de feno + 40% de ervilha. Não houve diferença significativa entre os regimes alimentares para o coeficiente de digestibilidade aparente (CDA) da matéria seca (MS), extrato não nitrogenado (ENN) e proteína bruta (PB), entretanto, o CDA da matéria orgânica (MO), fibra em detergente ácido (FDA), fibra em detergente neutro (FDN) e fibra bruta (FB) foram maiores no consumo restrito. Observou-se que houve maiores CDA da MS, ENN, MO e PB no tratamento 2 e não houve diferenças entre os tratamentos para o CDA da FDN e FB.
This experiment was carried out with the objectives of determining the daily average intake (DAI), apparent digestibility coefficient (ADC) of oat hay and pea nutrients and to evaluate the nitrogen balance in sheep. Ten wethers with average weight of 36.64 kg, alloted to suitable cages, were used. A completely randomized design with two treatments (T1= 80% oat hay + 20% pea and T2= 60% oat hay + 40% pea) and five replicates, was used. The treatments were evaluated under two feeding systems (voluntary and restrict). Daily average intake of dry matter (DM) and neutral detergent fiber (NDF) did not differ between treatments.Daily average intake of acid detergent fiber (ADF) and crude fiber (CF) were higher for treatment one. Animals submmitted to voluntary intake, showed higher nutrients average intake than the ones fed diet with 60% oat hay + 40% pea. There was no significant difference between both feeding systems to ADC of DM, nitrogen free extract (NFE) and crude protein (CP). However, ADC of organic matter (OM), ADF, NDF and CF were higher in restrict intake. There was higher ADC of DM, NFE, OM and CP in treatment two and there was no differences between treatments to ADC and NDF and CF.

Naftoquinonas isoladas de madeiras de árvores das famílias Bignoniaceae e Verbanaceae vêem sendo estudadas de modo interdisciplinar desde a década de 70, quando Dr. Benjamin Gilbert, na UFRJ, iniciou um programa inédito sobre a química de produtos naturais voltada ao combate de doenças endêmicas. Neste artigo apresentamos a síntese de cinco naftoimidazóis decorrentes deste programa e sua ação sobre T. cruzi, agente da doença de Chagas. Revisamos também a influência da estrutura química sobre a ação tripanocida de naftoquinonas e de hetero-derivados dos tipos imidazólico, oxazólico, fenoxazínico, indólico, dipirânico e ciclopentênico. A análise dos resultados corrobora a tendência de uma maior atividade tripanocida de compostos com anel imidazólico ou oxazólico ligado à estrutura naftopirânica. Dois naftoimidazóis apresentaram atividades superiores (14,5x e 34,8x) ao padrão cristal violeta. Ênfase foi dada à biodiversidade da flora brasileira como ponto de partida para o desenvolvimento de uma química medicinal autônoma e criativa.
Naphthoquinones isolated from the wood of trees of the families Bignoniaceae and Verbenaceae have been subjected to an interdisciplinary study since the seventies, when Dr. Benjamin Gilbert, at the Federal University of Rio de Janeiro, launched a program on the chemistry of natural products active against endemic diseases. In this paper we describe the synthesis of five naphthoimidazoles, derived from this program and their activity towards T. cruzi, the etiologic agent of Chagas disease. We also review the influence of chemical structure on trypanocidal action of naphthoquinones and of derived heterocycles with imidazole, oxazole, phenoxazine, indole, dipyrane and cyclopentene rings. The overall analysis corroborates the tendency of trypanocidal activity in compounds with an imidazole or oxazole ring linked to a naphthopyrane structure. Two naphthoimidazoles presented higher activities (14.5x and 34.8x) than the standard crystal violet. Emphasis is given to the biodiversity of the Brazilian flora as a starting point for the development of an autonomous and creative medicinal chemistry.
