Abstract The Neotropical zone stands out for having the greatest richness of amphibians in the world. About 625 species of anurans are known on Atlantic Forest. The southeastern mountainous regions of Brazil called Serra do Mar and Serra da Mantiqueira, are very important areas for the diversity of anurans in the country because they shelter remaining strata of the Atlantic Forest that have several endemic species. The present study aims to expand the knowledge about the Serra da Mantiqueira anurans, specifically from the Rio Preto and Rio Grande upper basins. The study area comprised 10 sample centers. We realized 38 campaigns, lasting two days each. During this period, we used active search as visual and auditory cues. The passive search was comprised of pitfall traps. Bioacoustic analyses were used to determine the cryptic species. The sufficiency of the sampling effort was estimated through the accumulation curve using Bootstrap and Jackknife-1 as estimators. During the study 55 species were found, 40 of them in the basin of the Rio Preto, and 44 in the basin of the Rio Grande. Twenty-nine species were found in both areas. The Hylidae family was predominant, followed by Leptodactylidae. In addition to species of the Atlantic Forest, the place houses typical components of the Cerrado biome and altitudinal swamps with unexpected sympatries. This inventory expands the area of occurrence of Scinax duartei, a species considered threatened. Although adjacent to the oldest national park in Brazil, the composition of anuran species of this area was unknown until then.

Abstract Guarana [Paullinia cupana var. sorbilis (Mart.) Ducke] is a species of great economic and social important in Brazil, as it is the only commercial guarana producer in the world. The vegetative propagation method indicated for the culture is stem cuttings, which aims at productivity, tolerance, and uniformity of clonal cultivars, because reproduction by seeds has slow germination and high genetic variability, which in traditional varieties is an undesirable factor. Genetic factors can interfere with the rooting capacity of the crop. Studies seek alternatives that can improve this condition and enhance the production system. Use of growth regulators, microorganisms that promote plant growth, variation of substrates and fertilization, have been strategies used. Preliminary tests on the rate of stem rooting and seed germination with the use of exogenous phytohormone did not demonstrate in relation to the non-application of these inducers. The use of rhizobacteria, which presents itself as a promising activity in many cultures, has not yet been demonstrated in the culture of guarana. On the other hand, the influence of different substrates on rooting has already shown consistent results as a function of rooting rate. Fertilizing the mother plants as recommended by the production system for the crop has proven to be an efficient procedure. There are still few studies aimed at improving the spread of guarana, demonstrating that new protocols need to be explored, or that the protocols already used are reviewed from another perspective.
ABSTRACT For studies of thermal performance, computational modeling has been presented as an important decision-making tool to solve complex and everyday problems in poultry production and research. Thus, the present study aimed to validate computational models developed using the EnergyPlus software package and, based on these, evaluate the thermal performance of a poultry house with different types of tiles (thermoacoustic, ceramic, fiber-cement, and metal). The research consisted of two stages: an experimental phase and another phase in which the climatic conditions in four aviary models are numerically simulated, considering different types of tiles. The experimental phase was conducted in two climate seasons, winter (2019) and summer (2020), where air temperature and relative air humidity data were measured and recorded using six HOBO model H12 data loggers distributed inside an aviary located in the semiarid region of Pernambuco state, Brazil. For the computer simulation, a virtual model of the house was developed using the OpenStudio graphical user interface in Sketchup, and the thermal performance was calculated using the EnergyPlus software package. The models were validated, showing a strong correlation between the experimental and simulated data, with Pearson’s correlation coefficient (r) values greater than 0.95. The simulations demonstrated the influence of the roof tiles on the thermal performance of the evaluated building. For the climatic conditions of the semiarid region of the Pernambuco state, thermoacoustic roof tiles presented the best thermal performance, followed by ceramic, fiber-cement, and metal roof tiles.

Abstract Currently, available fish anesthetics can produce important side effects, including respiratory arrest and distress. Easy-to-implement alternatives with low toxicity are needed to ensure fish health as well as to help artisanal fisheries and fish sellers in handling and transporting fishes, and native plants seems to be the best alternative. We aimed to implement an anesthetic protocol using crude ethanolic extracts from flowers and leaves of two Amazonian plants, the Acmella oleracea and Piper alatabaccum. We first tested the extracts for anesthesia, using the zebrafish as model. Even though in some treatments the animals apparently entered deep anesthesia, many of them presented aberrant behaviors and even died. Thus, we performed new experiments testing the extracts effects on seizure-like behaviors of the fish. Only the leaf extract of A. oleracea has potential effects for fish anesthesia. Both the flower extract from this plant and the leaf extract from P. alatabaccum induced seizure-like behavior in the animals. In conclusion, besides bringing a possible new anesthetic protocol for fish, our work draws attention for the neurotoxic effects the anesthetic solutions may cause, since several studies defend other Piper species as anesthetic for fish and A. oleracea flowers’ extract was already pointed as fish anesthetic.

ABSTRACT Background Triatoma infestans (Hemiptera: Reduviidae) is a hematophagous insect and the main vector of Trypanosoma cruzi (Kinetoplastida: Trypanosomatidae). In the present study, the authors investigated whether a serine protease activity from the saliva of T. infestans has a role in vasomotor modulation, and in the insect-blood feeding by cleaving and activating protease-activated receptors (PARs). Methods T. infestans saliva was chromatographed as previously reported for purification of triapsin, a serine protease. The cleavage activity of triapsin on PAR peptides was investigated based on FRET technology. Mass spectrometry was used to analyze the sites of PAR-2 peptide cleaved by triapsin. NO measurements were performed using the DAN assay (2,3-diaminonapthalene). The vasorelaxant activity of triapsin was measured in vessels with or without functional endothelium pre-contracted with phenylephrine (3 µM). Intravital microscopy was used to assess the effect of triapsin on mouse skin microcirculation. Results Triapsin was able to induce hydrolysis of PAR peptides and showed a higher preference for cleavage of the PAR-2 peptide. Analysis by mass spectrometry confirmed a single cleavage site, which corresponds to the activation site of the PAR-2 receptor. Triapsin induced dose-dependent NO release in cultured human umbilical vein endothelial cells (HUVECs), reaching a maximum effect at 17.58 nM. Triapsin purified by gel-filtration chromatography (10-16 to 10-9 M) was applied cumulatively to mouse mesenteric artery rings and showed a potent endothelium-dependent vasodilator effect (EC30 = 10-12 M). Nitric oxide seems to be partially responsible for this vasodilator effect because L-NAME (L-NG-nitroarginine methyl ester 300 µM), a nitric oxide synthetase inhibitor, did not abrogate the vasodilation activated by triapsin. Anti-PAR-2 antibody completely inhibited vasodilation observed in the presence of triapsin activity. Triapsin activity also induced an increase in the mouse ear venular diameter. Conclusion Data from this study suggest a plausible association between triapsin activity mediated PAR-2 activation and vasodilation caused by T. infestans saliva.

ABSTRACT Negative pressure ventilation in poultry houses has been used to enable the correction of their internal microclimates, and studies point to the heterogeneous distribution of air along the aviaries and the inadequacy of the environmental variables to the recommended ranges for thermal comfort of adult birds, especially in the hottest hours of the day. This study aimed to diagnose the facilities of a poultry house in the state of Paraíba, Brazil, regarding the distribution of environmental variables and thermal comfort; develop a computational model and validate it for Computational Fluid Dynamic - CFD simulations. Air temperature (Tair), air relative humidity (RH) and air velocity (Vair) data allowed characterizing the internal environment by comparison with the recommended ranges for each variable and by the temperature-humidity-velocity index (THVI). The poultry house does not provide comfort for the housed adult birds, between 12 and 14 h, with THVI indicating alert and Tair, RH and Vair values outside the recommended ranges; the CFD model for the poultry house was validated with Tair averages collected in the field of 27.75 ± 1.35 ºC and simulated of 27.85 ± 0.55 ºC, mean values of RH collected of 83 ± 12% and simulated of 78 ± 3%, and means of Vair collected of 2.35 ± 1.35 m s-1 and simulated of 2.50 ± 1.50 m s-1.

Abstract The present work aims to study the photocatalytic degradation of three organophosphate esters considered environmental emerging contaminants by the UV/H2O2 system with the use of 31P NMR spectroscopy to investigative their removal after the treatment. Results demonstrated the efficiency of the oxidation system in removing the esters tris(2-butoxyethyl) phosphate (TBEP), tris(2-chloroethyl) phosphate (TCEP) and tri-butyl phosphate (TBP) from aqueous solutions when they were individually present and mixed. High levels of degradation of these chemicals were achieved, in addition to the good performance of the analytical technique applied in the study, which represents some advantages in comparison with other techniques reported in the literature. An increase in the 31P NMR signal removal could also be observed when the oxidizing agent concentration increases. Decreases in solution acute toxicity were also verified for both TBP and TBEP treated samples when compared with the samples before the treatment.

ABSTRACT Although endophytic bacteria impact the health, and ultimately the fitness, of their hosts, our understanding of the diversity of endophytic species remains limited. Here we report on the endophytic microbiota inhabiting the roots, healthy leaves and leaves attacked by a gall-inducing insect of Baccharis dracunculifolia, a species of major economic relevance in South America, using 16S rRNA gene new generation sequencing. Rhodoplanes and Nitrospira were well represented in the communities of roots and leaves; known to be important for nitrogen cycling. The difference in bacterial diversity between healthy and galled leaves was not pronounced. The leaves seem to harbor specialized bacteria with high tolerances to abiotic stresses such as wide variation in temperature, low humidity, shallow and nutrient-poor soils and high solar irradiation. These findings suggest taxon-specific ecological niches in the leaves and roots, which may be the result of different physicochemical characteristics between these structures. This study provides a basis for further investigations and adds significant new information to the current knowledge of the endophytic bacterial composition in B. dracunculifolia.

The production of chicken eggs, following the evolution of research and development of innovative projects, is gaining significance every year in national and international market. The objective of this study was to evaluate the influence of air temperature and relative humidity on the quality of chicken eggs for human consumption during transport between the farm and the final warehouse. The research was conducted on a poultry farm (egg-laying) in Esperança, in the State of Paraíba, Brazil, and six loads were conducted. A F-4000 truck model was used without climate control. Red eggs of Lohmann (Brown) lineage were used and the following quality parameters were evaluated: egg mass, specific gravity, Haugh unit, shell thickness, yolk index and pH. For statistical analysis, the Dunnett’s test at 0.05 probability level was performed, thus comparing the final samples with the control from the farm. It was found that under the loading conditions only Haugh unit showed loss of quality during long trips.

Two new alkaloids containing Nb-methyl-triptamine units linked to bis-quinoline moieties, N-8”-formyl-calycosidine and N-8”-methyl-N-1’-demethyl-iso-calycosidine, were isolated from the aerial part of Margaritopsis carrascoana, along with known calycosidine and hodgkinsine. The cytotoxicity of the isolated alkaloids has been evaluated against cancer cell lines. The first two compounds showed only weak activity against HL-60 cell line, while hodgkinsine was the most active compound against HCT-116, OVCAR-8, HL-60 and SF-295 cancer cell lines. The structures of the isolated compounds were established by spectroscopic and molecular mechanical methods.

Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The objective of this study was to evaluate the performance and egg quality of laying hens raised in furnished cages and controlled environment, housed in a climate chamber under three temperatures: 20, 26 and 32 ºC and relative humidity of 60%. The cages had drinkers, feeders, nests and an area with sand and sandpaper. Thirty six Dekalb White lineage hens were used with 27 weeks of age with six animals per cage. The consumption of food and water, production and egg quality were analysed. The experimental design was in completely randomized design with three treatments and six replications. In ambient conditions of 20 and 26 ºC, birds were within the ZTC (Thermal Confort Zone), with no significant difference, with positive effect on egg quality with best values in mean quality parameters. In ET (Environmental Temperature) of 32 ºC, considered out of ZTC, the birds showed evidence of heat stress, providing increased water intake, reduced feed intake, values of percentage of production and egg quality parameters. Enriched cages were adequate to laying hens, showing egg production with good shell strength.

Henhouse thermal comfort and bird age influence broiler breeder hen productivity; this way, the present research aimed to assess thermal comfort indexes and broiler breeder hen performance during winter in North East semi-arid region from Brazil, using a Cobb Avian 48 broiler strain at the age of 38 and 50 weeks. We measured the following bioclimatic indexes: air temperature and relative humidity, black globe temperature and humidity, CO2 concentration and sound pressure levels at henhouse facility. Data analysis design was complete randomized in a 3 x 3 factorial scheme with three treatments (months) and three replications (week number of collection by month). A number of 25 climatic data were observed. Bioclimatic indexes at the most warm hours throughout the day promoted bird discomfort yet sound pressure and carbon dioxide levels were under the Brazilian standards. Even in thermal comfort zone at some hours, the productivity indexes were in compliance with those recommended by the strain creation guidebook.
