Abstract Healthcare workers present an increased risk of contagion for the SARS-CoV-2 virus due to their labor exposure. Here, we describe the clinical, laboratory, and immunological characteristics of healthcare workers, before vaccine application, exposed to SARS-CoV-2-infected patients. We collected sociodemographic, clinical, and laboratory information from 50 professionals who worked during the COVID-19 pandemic at the Clinical Hospital of the Northwest in Brazil. The results showed that most workers are women, over 50 years old, and worked as nursing technicians. Approximately 56% of workers were positive for a previous infection by RT-PCR and/or anti-SARS-CoV-2-immunoglobulin tests. Increased levels of hematocrit, neutrophils, NK lymphocytes, and fibrinogen, were found in positive healthcare workers, suggesting a light inflammatory status. The immunological findings showed an increase in IL-17 production and a Th2/Th17/Th22 profile followed by high serology for anti-SARS-CoV-2 IgM and IgG. Those data reveal the importance of studies with healthcare workers to investigate if the continuous exposition to the virus may result in chronic activation of the immune system and/or pulmonary inflammation in this target group.

Benzophenone Derivatives Showed Dual Anti-Inflammatory and Antiproliferative Activities by Inhibiting COX Enzymes and Promote Cyclin E Downregulation

Considering the promising antitumor effects of compounds with dual anti-inflammatory and antiproliferative activities, thus benzophenones analogs (2-7) were evaluated on in vivo anti-inflammatory assay and molecular docking analysis. Those with the best molecular docking results were in vitro evaluated on cyclooxygenase (COX) enzymes and tested regarding antiproliferative activity. All derivatives displayed in vivo anti-inflammatory activity. Among them, the substances 2’-hydroxy-4’-benzoylphenyl-β-D-glucopyranoside (4), 4-hydroxy-4’-methoxybenzophenone (5) and 4’-(4’’-methoxybenzoyl)phenyl-β-D-glucopyranoside (7) showed the best values of Glide Score in COX-2 docking evaluation and 4 and 5 selectively inhibited COX-2 and COX-1 in vitro enzymatic assay, respectively. Thus, 4 and 5 were tested against breast cancer (MCF-7, MDA MB-231, Hs578T) and non-small-cell-lung cancer (A549) cell lines. The estrogen-positive MCF-7 cell line was more responsive compared to other tested cell lines. They induced cell cycle arrest at G1/S transition in MCF-7 cell line once there was an increase in G0/G1 population with concomitant reduction of S population. The antiproliferative activity of these substances on MCF-7 was associated with their ability to inhibit cyclin E expression, a critical regulator of G1/S transition. Taken together, the data indicate that 4 and 5 have dual anti-inflammatory and antiproliferative activities and support further studies to evaluate their antitumor potential.

Abstract Health professionals working to mitigate the COVID-19 pandemic are one of the main risk groups for the disease, being prioritized for vaccination. Considering this, the aim of this study was to analyze the immune response of these professionals immunized with CoronaVac in the first and second doses. Blood samples were collected after the first and second doses of the vaccine (CoronaVac) and used to investigate hematological and biochemical parameters, analysis of immunoglobulin production, cytokines, and gene expression profile, as well as the identification of subsets of immune cells. Post-first dose immunological phenotypic memory (CD27+) profiles (T CD4+, TCD8+ and CD19+) showed a significant increase, as did Monocyte APCs (CD80+HLA-DR+) in relation to the second dose. The cytokines IL-2, IL-6 and IFN-° showed increased values in relation to the other analyzed cytokines. The Th2/Th17 profile in the second dose was characterized by gene expression analysis. The production of IgM and IgG after vaccination showed statistically significant values in the comparison between doses. CoronaVac showed activation of APCs monocytes, memory response of T and B lymphocytes, with immunoglobulins production. This set of responses is characterized by the Th2/Th17 immunological profile.

Resumo Devido à importância ecológica de Lophiosilurus alexandri, o presente trabalho avaliou sua representatividade genética, comparando estoques selvagens com plantéis de reprodutores de três larviculturas ao longo do Rio São Francisco. Noventa e sete amostras foram genotipadas com marcadores microssatélites recém-desenvolvidos. Baixos níveis de diversidade genética (número médio de alelos de 4,2) foram detectados em todos os casos, sendo mais severo no cativeiro. Os valores de FST e DEST par a par, as análises do Structure e DAPC mostraram a estruturação dos animais selvagens em dois grupos, e um terceiro formado pelas larviculturas, evidenciando a necessidade de adoção de critérios genéticos para retenção da diversidade genética no cativeiro. Por essa razão, três famílias de irmãos completos foram construídas para selecionar o melhor estimador de parentesco para a espécie e estabelecer os valores mínimos de corte para evitar cruzamentos indesejados. Dois estimadores, Wang (RW) e Lynch & Li (RLL), foram eficientes em refletir as relações de parentesco para irmãos completos nessa espécie. Segundo eles, menos de 50% dos potenciais cruzamentos são recomendáveis nas três larviculturas. A baixa diversidade genética inerente ao L. alexandri destaca a importância de minimizar a consanguinidade e evitar perda de diversidade genética, visando a recuperação da espécie.
Abstract Due to the ecological importance of Lophiosilurus alexandri, the present work evaluated its genetic representativeness by comparing wild stocks to broodstocks that were kept at three restocking hatcheries along the São Francisco River. A total of 97 samples were genotyped for newly developed microsatellite markers. Low levels of genetic diversity (average alleles number of 4.2 alleles) were detected in all cases, being more severe in captive groups. Significant pairwise FST and DEST values, Structure, and DAPC analyses showed that wild animals were structured in two groups, and a third group was formed by captive animals, evidencing the need to adopt genetic criteria to retain genetic diversity in the hatcheries. For this reason, three full-sib families were constructed to select the best relatedness estimator for L. alexandri and establish a cut-off value aimed to avoid full-sibling matings in the hatcheries. Two estimators, Wang (RW) and Lynch & Li (RLL), were accurate in reflecting the relatedness level for full-sibs in this species. According to them, less than 50% of the potential breeding matings in the three hatcheries are advisable. The innate low diversity of L. alexandri highlights the importance of minimizing inbreeding and retaining genetic diversity towards the species recovery.

The secondary metabolites of the aerial parts of Phyllanthus acuminatus Vahl (Phyllanthaceae) and the biological activity of one of them were characterized in this study. This chemical investigation led to isolation of two lignans, luclaricin (1), a new arylnaphtalene-typelignan, and justicidin B (2). The structures of isolated compounds were determined based on 1D and 2D-NMR, HRESIMS and IR. The cytotoxic activity of justicidin B was assessed against tumor cell lines (MCF-7, U251, NCI-ADR/RES, NCI-H460, OVCAR-3, HT29 and K562) and a non-tumor cell line (HaCat) using sulforhodamine B assay. Justicidin B showed strong cytotoxic activity against the tumor cell lines HT29, K562 and U251 with GI50 values of 2.28, 3.29 and 5.76 mol L-1, respectively.

Synthesis and Application of Bimetallic Zinc(II) Phenoxy-Imine Complexes as Initiators for Production of Lactide Polymers

This article reports the synthesis of two bimetallic zinc(II) complexes containing phenoxy-imine ligands and their application as initiators of ring opening polymerization (ROP) of L-lactide (LA). The phenoxy-imine ligands were obtained from salicylaldehyde derivatives and 2,3,5,6-tetramethylphenylenediamine, yielding two bidentate regions per ligand. The reactions of both phenoxy-imine ligands with ZnEt2 in the presence of n-BuOH afforded [Zn2(L1)(OBu)2] and [Zn2(L2)(OBu)2], which were characterized by elemental analysis, Fourier transform infrared (FTIR) and 1H nuclear magnetic resonance (NMR) spectroscopy. In addition, the geometries of both complexes were investigated by DFT (B3LYP/LACVP**) calculations. [Zn2(L1)(OBu)2] and [Zn2(L2)(OBu)2] were tested as initiators of ROP of LA at 180 °C using different LA/Zn molar ratios, namely 500, 1000 and 2500. Both complexes showed good activity, resulting in conversions up to 96% in 2 h. The poly-LLA exhibited average molecular weight (Mw) ranging from 45,000-92,000, relatively low polydispersity (Mw/Mn = 1.6-2.0) and high stereoregularity with melting temperature Tm = 164 °C.

Infection by Streptococcus pneumoniae in children with or without radiologically confirmed pneumonia

Resumo Objetivo: Avaliar o papel do raios X de tórax na identificação de casos de pneumonia adquirida na comunidade (PAC) causada por agentes bacterianos. Métodos: A frequência de infecção por Streptococcus pneumoniae, Haemophilus influenzae e Moraxella catarrhalis em crianças com PAC não hospitalizadas foi comparada com a presença de confirmação radiológica da pneumonia (n = 249 crianças com pneumonia radiologicamente confirmada e 366 crianças com raios X de tórax normal). Infecção por S. pneumoniae foi diagnosticada com base na resposta sorológica a pelo menos uma dentre oito proteínas pneumocócicas investigadas (aumento ≥ 2 vezes nos níveis de IgG em relação a Ply, CbpA, PspA1 e 2, PhtD, StkP-C e PcsB-N ou aumento≥ 1,5 vez em relação aPcpA). Infecção por H. influenzae e M. catarrhalis foi definida por aumento ≥ 2 vezes nos níveis de IgG específica a antígenos de cada agente. Resultados: Crianças com pneumonia radiologicamente confirmada apresentaram maior taxa de infecção pelo pneumococo. Além disso, a presença de infecção pneumocócica foi um fator preditor de pneumonia radiologicamente confirmada, o que aumenta sua chance de detecção em 2,8 vezes (IC 95%: 1,8-4,3). O valor preditivo negativo do raios X normal para a infecção por S. pneumoniae foi 86,3% (IC95%: 82,4%-89,7%). Não houve diferença nas frequências de infecção por H. influenzae e M. catarrhalis entre crianças com PAC com ou sem confirmação radiológica. Conclusão: Crianças com diagnóstico clínico de PAC submetidas a um raios X de tórax que apresentam confirmação radiológica têm maior taxa de infecção por S. pneumoniae comparadas com as crianças com raios X normal.
Abstract Objective: Community-acquired pneumonia is an important cause of morbidity in childhood, but the detection of its causative agent remains a diagnostic challenge. The authors aimed to evaluate the role of the chest radiograph to identify cases of community-aquired pneumonia caused by typical bacteria. Methods: The frequency of infection by Streptococcus pneumoniae, Haemophilus influenzae, and Moraxella catarrhalis was compared in non-hospitalized children with clinical diagnosis of community acquired pneumonia aged 2-59 months with or without radiological confirmation (n = 249 and 366, respectively). Infection by S. pneumoniae was diagnosed by the detection of a serological response against at least one of eight pneumococcal proteins (defined as an increase ≥2-fold in the IgG levels against Ply, CbpA, PspA1 and PspA2, PhtD, StkP-C, and PcsB-N, or an increase ≥1.5-fold against PcpA). Infection by H. influenzae and M. catarrhalis was defined as an increase ≥2-fold on the levels of microbe-specific IgG. Results: Children with radiologically confirmed pneumonia had higher rates of infection by S. pneumoniae. The presence of pneumococcal infection increased the odds of having radiologically confirmed pneumonia by 2.8 times (95% CI: 1.8-4.3). The negative predictive value of the normal chest radiograph for infection by S. pneumoniae was 86.3% (95% CI: 82.4-89.7%). There was no difference on the rates of infection by H. influenzae and M. catarrhalis between children with community-acquired pneumonia with and without radiological confirmation. Conclusions: Among children with clinical diagnosis of community-acquired pneumonia submitted to chest radiograph, those with radiologically confirmed pneumonia present a higher rate of infection by S. pneumoniae when compared with those with a normal chest radiograph.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

A phytochemical study on the aerial parts of Wissadula periplocifolia using chromatographic techniques has led to the isolation of sitosterol (1a), stigmasterol (1b), sitosterol 3-O-β-D-glucopyranoside (2a), stigmasterol 3-O-β-D-glucopyranoside (2b), phaeophytin A (3), 13²-hydroxy-(13²-S)-phaeophytin A (4), phaeophytin B (5), 17³-ethoxyphaeophorbide (6), 3,4-seco-urs-4(23),20(30)-dien-3-oic acid (7), 3-oxo-21β-H-hop-22(29)-ene (8), dammaradienone (9a), and taraxastenone (9b). The isolated compounds were characterised by spectroscopic analysis. A preliminary assay to evaluate the antibacterial activity of W. periplocifolia extracts and fractions showed that the dichloromethane, ethyl acetate, and n-butanol fractions were active against Enterococcus faecalis.

Lentivírus em caprinos leiteiros do semiárido paraibano: prevalência de anticorpos, fatores de risco e detecção molecular

Os objetivos do presente trabalho foram determinar a prevalência de caprinos leiteiros soropositivos para a infecção por Lentivirus de pequenos ruminantes no semiárido do Estado da Paraíba, Nordeste do Brasil, identificar fatores de risco associados à prevalência de rebanhos positivos, e realizar a detecção molecular do agente. Foram utilizadas 1047 cabras leiteiras de 110 propriedades selecionadas aleatoriamente no Município de Monteiro, Estado da Paraíba, no período de março de 2009 a dezembro de 2011. Para o diagnóstico da infecção por Lentivirus, foi utilizado o teste de imunodifusão em gel de ágar (AGID). Um ano após foi realizada nova sorologia, e PCR em tempo real foi aplicada em amostras de sangue e leite de 48 cabras procedentes de quatro propriedades com animais soropositivos. As prevalências de propriedades positivas e de animais soropositivos na AGID foram 44,6% (IC 95% = 35,1% - 54,3%) e 8,1% (IC 95% = 5,6% - 16,8%), respectivamente. Realizar corte e desinfecção de umbigo (odds ratio = 2,44; p = 0,048) e condições de aglomeração de animais (odds ratio = 3,45; p = 0,048) foram associadas com a prevalência de propriedades positivas. Um ano após a realização do inquérito sorológico, foi verificada a permanência de animais infectados, detectados por PCR em tempo real a partir de amostras de sangue e leite. A PCR em tempo real das amostras de leucócitos circulantes apresentou boa performance, com sensibilidade de 100%, especificidade de 92,86%, concordância de 93,75% e indicador Kappa de 0,765. Sugere-se que seja realizado um trabalho de educação sanitária junto aos produtores sobre medidas de prevenção com o objetivo de reduzir a disseminação da infecção nos rebanhos.
The aims of this study were to determine the seroprevalence of infection by ruminants Lentivirus in dairy goats in the semiarid of the Paraiba State, Northeastern Brazil, to identify risk factors associated with the herd-level prevalence and to perform molecular detection of the agent. A total of 1,047 dairy goats from 110 herds were randomly selected from the county of Monteiro, Paraiba State, and serum samples were collected from March 2009 to December 2011. For the diagnosis of Lentivirus infection, the agar gel immunodiffusion test (AGID) was used. One year after that a new serology was performed and the real-time PCR assay was applied in blood and milk samples from 48 goats from four herds with seropositive animals. Prevalence of positive herds and seropositive animals at AGID were 44.6% (95% CI=35.1-54.3%) and 8.1% (95% CI =5.6-16.8%), respectively. Umbilical cord cutting and disinfection (odds ratio = 2.44; p = 0.048) and conditions of animal agglomeration (odds ratio=3.45; p=0.048) were associated with herd-level prevalence. One year after the serological profile, the permanence of infected animals detected by real-time PCR in blood and milk samples was verified. Real-time PCR using white blood cells had a good performance, with sensitivity of 100%, specificity of 92.86%, concordance of 93.75% and Kappa index of 0.765. It was suggested to teach sanitary measures to the herd owners in order to encourage them to adopt prevention measures aiming to reduce the spread of the infection in the herds.

OBJETIVO: Estimar a frequência de história pregressa de chiado, descrevendo as características clínicas e formas de tratamento utilizadas. MÉTODOS: Coorte retrospectiva incluindo lactentes (6-23 meses de vida) com sintomas de infecção de vias aéreas superiores e relato de episódio prévio de chiado. Os dados foram registrados em questionário anteriormente validado. RESULTADOS: Dos 451 avaliados, 164 (36,4%; IC95% = 31,9-41,0) apresentaram história pregressa de chiado, 148 (32,8%; IC95% = 28,5-37,4) no primeiro ano de vida. A média de idade no primeiro episódio de chiado foi de 5,3±3,9 meses. Para quem começou a chiar no primeiro ano de vida, 38,5% referiram entre três e seis episódios e 14,2% > seis episódios. A média da idade no primeiro episódio foi menor para os que apresentaram > três episódios em comparação aos que apresentaram até dois episódios (3,2±2,7 versus 5,7±2,5 meses, p < 0,001). CONCLUSÃO: Um terço dos lactentes apresentou chiado no primeiro ano de vida. Quanto mais cedo ocorre o primeiro episódio, mais frequente é a recorrência do chiado.

METHODS: Infants aged 6-23 months with upper respiratory tract complaints and reporting previous wheezing were followed-up retrospectively. Data were registered on a validated standardized form. RESULTS: Out of 451 infants, 164 (36.4%; 95%CI: 31.9-41.0) had a report of prior history of wheezing, 148 (32.8%; 95%CI: 28.5-37.4) during the first year of life. The mean age at the first episode of wheezing was 5.3±3.9 months. Among those who had had their first episode before 12 months of age, 38.5% reported 3 to 6 episodes and 14.2% > 6 episodes. Mean age at first episode was lower for those with > 3 episodes in comparison with those with < 2 episodes (3.2±2.7 vs. 5.7±2.5 months, p < 0.001). CONCLUSION: One third of the infants reported wheezing during the first year of life. The earlier the first episode occurs, the more frequently wheezing recurs.

Acetic acid-induced writhing, hot-plate, carrageenan-induced pleurisy, formalin-induced pain, croton oil-induced ear edema, vascular permeability tests and phospholipase A2 activity assay were used to study the analgesic and/or anti-inflammatory activity of the hydromethanolic fraction of ethanolic extract from Spiranthera odoratissima A. St.-Hil., Rutaceae, leaves (HMF) and its subfraction (sub-Fr10-28). HMF and sub-Fr10-28 reduced the leukocyte migration on the carrageenan-induced pleurisy test; sub-Fr10-28 reduced the pain reaction time in the second phase of formalin-induced pain, as well as the ear edema and vascular permeability. Both HMF and sub-Fr10-28 inhibited the phospholipase A2 activity. These results suggest that the analgesic effect of this plant could be, in part, due to an anti-inflammatory action produced by the inhibition of phospholipase A2 activity.

O aumento da incidência das infecções fúngicas, bem como o registro crescente de resistência e falha terapêutica, têm impulsionado a realização de estudos de prospecção de fitoquímicos com propriedades antifúngicas. Diante do exposto, o presente estudo investigou o potencial antifúngico de extratos de Baccharis ligustrina, B. schultzii, Croton jacobinensis, Licania rigida, Moringa oleifera, Vernonia sp. e V. brasiliana, e de óleos essenciais de Lippia alba (Quimiotipos 1, 2, 3 e 4) e Ocimum gratissimum. Inicialmente, foi realizada uma avaliação qualitativa da atividade antifúngica de cada amostra por meio do método de difusão em ágar, frente a cepas de Candida albicans e Microsporum canis, mostrando que apenas os extratos de M. oleifera (MLF-C) e Vernonia sp. (TVS-H) apresentaram atividade frente a C. albicans e M. canis, com halos de inibição =10mm. Também foram determinadas a concentração inibitória mínima (CIM), frente a 12 cepas de C. albicans e M. canis, e a toxicidade aguda de MLF-C e TVS-H, através de protocolos do Clinical and Laboratory Standards Institute (CLSI) e ensaio com Artemia sp., respectivamente. A CIM (80%) de MLF-C e TVS-H variou de 0,156 a 2,5mg mL-1 frente C. albicans e de 0,039 a 1,25 e 0,039 a 0,625mg mL-1 para M. canis, respectivamente. A CIM (100%) de MLF-C e TVS-H variou de 0,625 a 2,5mg mL-1 frente C. albicans é de 0,039 a 2,5 e 0,078 a 1,25mg mL-1 para M. canis, respectivamente. As doses letais (DL50) para o MLF-C e TVS-H foram de 201,09 e 204,17mg mL-1, respectivamente, sendo, portanto, demonstrada a baixa toxicidade desses extratos. Os extratos de M. oileifera e Vernonia sp. apresentaram atividade antifúngica frente cepas de C. albicans e M. canis, abrindo a perspectiva de estudos para caracterização dos seus componentes bioativos.
The increase in the incidence of fungal infections and the frequent report of resistance and therapeutic failure has promoted the performance of phytochemical screening for compounds with antifungal properties. Based on this, the present study investigated the antifungal potential of extracts of Baccharis ligustrina, B. schultzii, Croton jacobinensis, Licania rigida, Moringa oleifera, Vernonia sp. and V. brasiliana and of essential oils of Lippia alba (Chemotypes 1, 2, 3 and 4) and Ocimum gratissimum. Initially, a qualitative evaluation of the antifungal activity of each vegetal sample against strains of Candida albicans and Microsporum canis, through the agar diffusion method, was performed. Extracts of M. oleifera (MLF-C) and Vernonia sp. (TVS-H) presented activity against C. albicans and M. canis with inhibition halos =10mm. Then, minimum inhibitory concentrations (MICs) for MLF-C and TVS-H against 12 strains of C. albicans and M. canis were determined through the methodology established by the Clinical and Laboratory Standards Institute (CLSI), and acute toxicity tests against Artemia sp. were performed for both extracts. MICs (80%) for MLF-C and TVS-H varied from 0.156 to 2.5mg mL-1 against C. albicans and from 0.039 to 1.25mg mL-1 and 0.039 to 0.625mg mL-1 against M. canis, respectively. MICs (100%) for MLF-C and TVS-H varied from 0.625 to 2.5mg mL-1 for C. albicans and from 0.039 to 2.5mg mL-1 and 0.078 to 1.25mg mL-1 against M. canis, respectively. Lethal doses (DL50) of MLF-C and TVS-H were 201.09 and 204.17mg mL-1, respectively, being, therefore, demonstrated the low toxicity of these extracts. M. oleifera and Vernonia sp. extracts presented in vitro antifungal activity against C. albicans and M. canis, creating perspectives for the development of studies on the characterizations of their bioactive components.
