Computer-aided drug planning strategies have contributed to the research and development of new anti-tuberculosis (TB) drugs to avoid resistance and reduce treatment time and the number of drugs used in therapy. The aim of work was to carry out a docking study to identify the possible mechanism of action of the hits LabMol73, 84, 86 and 93 previously tested in the Microplate Assay Blue Alamar (MABA), Low Oxygen Recovery Assay (LORA) assays in sensitive strains from M.tb. H37Rv and resistant to the standard drugs rifampicin and isoniazid, due to the promising inhibitory result against these strains, suggesting a different mechanism of action from existing drugs. The reverse virtual screening was performed on the Pharmmapper platform, which identifies targets by pharmacophoric model. The most promising targets have been validated. The DM was performed in the OpenEye Maestro program for analysis of poses and energy score. Sixteen targets M.tb. H37Rv were identified and only nine demonstrated viability for computational testing. The most promising results were observed in the mycolic acid cyclopropane synthase (PDB:1L1E) and pantothenate synthetase (PDB:1N2B) targets. Since, in the target 1L1E score results obtained were between -6.998 to -7.767 kcal/mol. In the 1N2B target the results were between -6.421 to -7.293 kcal/mol, presenting themselves as the most promising targets due to their similar score scores between the two targets suggesting that the mechanism of action may be the inhibition of one of these targets. These targets proved to be promising for elucidating the mechanism of action of the analyzed nitro heteroaryl chalcones, as they corroborate the assay against resistant strains, demonstrating that standard drugs have activity against other targets and also because mycolic acid and pantothenate are directly linked to virulence and resistance of M.tb. H37Rv.

Abstract: The adverse effects of oral pre-exposure prophylaxis (PrEP) using tenofovir disoproxil fumarate are barriers to PrEP initiation and continuation. Although serious effects are rare and predictable, evidence for this assessment among men who have sex with men (MSM) and transgender women (TGW) is still limited. This study assesses the adverse effects of daily oral PrEP in MSM and TGW. This is a systematic review and meta-analysis of clinical trials and cohort studies on the use of daily oral PrEP selected from the PubMed/MEDLINE, Embase, LILACS, and Cochrane CENTRAL databases. Data extraction included adverse effects and changes in renal and hepatic markers. Random effects models were used to summarize the risk of adverse effects throughout the study. Heterogeneity was assessed using the Cochran’s Q test and the inconsistency test (I2). The risk of bias and the certainty of the evidence were assessed using the Cochrane Collaboration recommendations. The search identified 653 references. Of these, 10 were selected. All studies assessed the eligibility of renal and hepatic markers. The use of daily oral PrEP was not associated with grade 3 or 4 adverse events (RR = 0.99; 95%CI: 0.83-1.18; I2 = 26.1%), any serious adverse event (RR = 1.04; 95%CI: 0.58-1.87; I2 = 88.4%), grade 3+4 creatinine level (RR = 0.66; 95%CI: 0.24-1.84; I2 = 79.9%), and grade 3 or 4 hypophosphatemia (RR = 0.56; 95%CI: 0.15-2.10). The certainty of the evidence ranged from high to moderate for the outcomes analyzed. Daily oral PrEP is safe and well tolerated by MSM and TGW. Adverse effects were minimal and evenly distributed between intervention and control.

ABSTRACT Purpose: to assess the evolution of educational actions on hearing health, in the remote format, for Community Health Agents from Santa Maria, RS, Brazil, and to identify if there is a relationship between the level of knowledge acquired and the variables age, length of service and education. Methods: a quantitative and interventional research, carried out with 44 Community Health Agents, who participated in three meetings, via Google Meet. In the first, data on the professionals' knowledge were collected and, in the second and third meetings, there was training on 1) hearing, 2) hearing health and disorders, 3) organization of the Hearing Health Service, 4) use/hygiene of hearing aids and 5) importance of users' follow-up at the service, and the collection of knowledge acquired on these topics. Such collection took place through Quizzes, elaborated in Google Forms. The statistical tests Chi-square and Two-sample Test of Proportions were used to statistically analyze the data, with a significance of 5%. Results: there was a difference in the rate of correct answers in the post-training Quiz and the acquisition of knowledge was more evident when compared to the performances "worse" and "mistake" in themes 1, 2, 3 and 4. No relationship was found between the level of knowledge acquired and the studied variables. Conclusion: the educational actions proved to be effective and the level of knowledge acquired on the topics is not related to the studied variables.

SUMMARY Bixa orellana L. is a native plant from Brazil, but it is also present in other tropical countries such as Peru, Colombia, Ecuador, Mexico, Indonesia, India and East Africa. It is popularly known as Urucum in Brazil. This review shows the potential of bioactive compounds derived from B. orellana to treat infectious diseases due their antimicrobial and antioxidant properties. This plant is also related as an antiinflammatory agent for treatment of pulmonary diseases, or even as eye drops for redness. Its leaves are used for treatment of snakebite, diarrhea, gonorrhea, hepatitis, gastritis, diuretic, antipyretic, and for skin disease. This popular knowledge has encouraged the identification of bioactive compounds in this plant. Compounds as β-cryptoxanthin, geranylgeraniol, lutein, procyanidin B2, procyanidin B3, ellagi tannin isomer and ellagic acid deoxyhexose have been described. These compounds inhibited pathogenic microorganisms such as bacteria, fungi, protozoan and viruses. In addition, some compounds with anti-inflammatory and antioxidant activities were also described. In this sense, B. orellana is a promising source of compounds that could be applied in antimicrobial therapy. This review work may help in the understanding and incentive of new research for antimicrobial discoveries using different B. orellana compounds.

ABSTRACT For at least 30 years researchers have called for a deeper reflection on the paths we desire for ethnobotanical research. Although the discipline of ethnobotany is growing, as measured by the number of publications in the area, there is still work to be done regarding the homogeneity of theoretical and methodological approaches and the implications of ethnobotanical research findings for society as a whole. In this article we present 10 questions/issues that we believe can guide the research and actions of ethnobotanists for the coming years.

ABSTRACT Studies involving coastal sediment resuspension have shown that trace metals could be released to the water column due to changes in physical-chemical conditions. Therefore, if environmental agencies adopt screening protocols with insufficient evaluations of contaminant mobilization risks for a given area, the prediction of environmental impacts caused by dredging may be limited. This work evaluates the influence of spatial variation on the geochemical mobility of trace metals (Cd, Cu, Ni, Pb, Zn) after artificial sediment resuspension, using as study case the Iguaçu River estuary (Guanabara Bay, Brazil). The Iguaçu River drains the industrial complex of the metropolitan area of Rio de Janeiro State, besides the presence of agriculture and the input of untreated domestic wastes. Surface estuarine sediments were submitted to resuspension experiments in an open system, during 1h and 24h of agitation on local water. A clear tendency of metals’ solubilization was observed after resuspension, especially considering the dissolved concentrations of Cu (average: 8.0 µg L-1) and Zn (average: 0.9 mg L-1), especially for the samples from the transects 2 and 3. However, evaluations of water quality changes due to sediment resuspension are not requested by the legislation regulating the dredging activities. In the sediments, the results suggested a higher geochemical mobility of Cu, indicated by the massive increase on the bioavailable fraction after resuspension, mainly on the transect number 3, on the river’s mouth. The effects of resuspension were distinct between samples, suggesting that even in a small scale, important differences on metals’ mobility are found. Thus, the combined assessment of changes in metal concentrations in water and in the metal partitioning linked to the solid-phase was demonstrated to be a promising tool for predicting the environmental risks of dredging due to changes in the bioavailability of metals.

ABSTRACT: Orthopedic diseases are common in dogs and cats, especially, those caused by traumatic injury. Overall, among the significant changes in this group, the fractures are the major cause of pain and dysfunction in dogs of all ages, sizes and breeds. Therefore, a retrospective study of dogs examined between January 2004 and December 2013 at the University Veterinary Hospital of Universidade Federal de Santa Maria, was conducted aiming to identify and determine the prevalence of appendicular fractures caused by trauma and characterizing the population/etiology. In a population of 1,200 dogs with suspected traumatic orthopedic diseases in the locomotor system, 955 (79.6%) had appendicular fractures. Of the dogs 23.5% had fractures on the femur (n=225), 23.4% had pelvic fractures (n=223), 22% had tibial and fibular fractures (n=210), 17.6% had radius and ulna fractures (n=168), 7.5% had humeral fractures (n=72) and 6% had distal limb fractures (tarsus, carpus, metacarpus, metatarsus and phalanges (n=57)). The most frequent cause was car accidents (72.2%). Most affected dogs were male (52.5%), juvenile (42%), mixed breed (51.4%) and small size (42.7%). In conclusion, the profile of dogs with fractures in the Central Region of Rio Grande do Sul State is: male dogs, mixed breed, immature and small size, presenting femoral fractures by car accident.

Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Hemicelluloses were extracted from corn husk and converted into cationic hemicelluloses using 2,3-epoxypropyltrimethylammonium chloride. The degree of substitution was determined as 0.43 from results of elemental analysis. The cationic derivative was also characterized by Fourier transform infrared spectroscopy and Carbon-13 magnetic nuclear ressonance. The produced polymer was employed as coagulant aid in a sewage treatment station (STS) of the municipal department of water and sewer (Departamento Municipal de Água e Esgoto - DMAE) in Uberlândia-Minas Gerais, Brazil, using Jar test experiments. Its performance was compared to ACRIPOL C10, a commercial cationic polyacrylamide regularly used as a coagulant at the STS. The best result of the jar-test essays was obtained when using cationic hemicelluloses (10 mg L- 1) as coagulant aid and ferric chloride as coagulante (200 mg L- 1). The resultsof color and turbidity reduction, 37 and 39%, respectively, were better than when using only ferric chloride. These results were also higher than those of commercial polyacrylamide, on the order of 32.4 and 38.7%, respectively. The results showed that the cationic hemicelluloses presented similar or even superior performance when compared to ACRIPOL C10, demonstrating that the polyelectrolytes produced from recycled corn husks can replace commercial polymers in sewage treatment stations.

Ostrich's meat has low contents of colestherol and intramuscular fat and high percentages of poliunsaturated fat acid -3 so it's known as a healthy red meat with consumer great acceptability. Considering that ostrich trim (meat resulting from boning) is considered a less noble part, three formulations of ostrich (trim) Brazilian sausage were produced. Physicochemical analyses and color evaluation (L*, a*, b*) were made to determine formulations' physicochemical characteristics and colorimetric profile. The physicochemical results of all formulations were in accord with the Brazilian legislation pattern's (BRASIL, 2000) and no differences (P>0,05) between formulations have been found except in the ash content, higher in mixed sausages. Differences (P>0,05) have been found also between the pH of the treatments, which increased with the heightening of the percentage of ostrich meat in formulation. The sausages formulated with 100% ostrich meat were darker and redder in appearance than the mixed ones. The mixed of ostrich, pork and chicken Brazilian sausage were the lightest. The Brazilian sausages prepared with ostrich meat, pure or mixed, are a viable option for production and sale, and can be placed in the market and compete with the traditional sausages with the prospect of being successful.

INTRODUCTION: Visceral leishmaniasis is a serious public health problem that requires global control strategies, especially with respect to factors that may intervene in reducing the incidence of endemicity. In this work, rainfall density and temperature were correlated with the incidence of human cases in an area endemic for leishmaniasis in São Luis do Maranhão, Northeastern Brazil. METHODS: Notification of human cases by the National Health Foundation/Regional Coordination of Maranhão (FUNASA/COREMA) from 2002 to 2010 was used. Ecological data (mean temperature and rainfall density) were provided by the Meteorological Office of State. RESULTS: A significant association was verified between the number of VL cases and rainfall rate but not in the analysis concerning mean temperatures. CONCLUSIONS: These data suggest that the control actions in visceral leishmaniasis should be performed during rainy season in the State of Maranhão, which is in the first half of the year.

The TMJ has been to the Dental community a key point in the search of knowledge, being it part of the temporomandibular joint complex and of the estomatognathic system which are in charge of the mastication, speech, swallowing, as well as participation in breathing and taste perception. For the majority of the women in serious state of depression, which do not respond psychotherapeutic treatment, pharmacological treatment it's applied. The antidepressants serotonin selective reuptake inhibitors (SSRIs) are the most recommended in these cases. The teratogenic effect of the SSRIs is considered controversial, studies done with women who used these drugs during the pregnancy showed that the respiratory and central nervous systems are the most affected, was also recorded a deficit of body growth and the decrease of the encephalon and skull measures. In the present study, our goal was to assess whether the administration of Fluoxetine during the pregnancy modified the embryology and morphology of the TMJ of rats. For that, 16 Wistar female rats from the Nutrition Department of the UFPE vivarium were selected; 8 for the control group, which received daily 0.9% of saline in subcutaneous dose of 10ml/g, with schedules previously established after daily weighing and 8 for the experimental one that were treated with fluoxetine hydrochloride with the dose of 10mg/Kg in a volume 10ml/g of weight, were injected subcutaneously with the same standards established for the control group. It was observed, with this dose that the embryological development of the TMJ, especially of the mandibular condyle, does not present any difference between the degree of maturation of the tissue that forms the TMJ, especially of the condyle between the treated and control groups.
