ABSTRACT Objective: To assess the impact of different vertical positions on lung aeration in patients receiving invasive mechanical ventilation. Methods: An open-label randomized crossover clinical trial was conducted between January and July 2020. Adults receiving invasive mechanical ventilation for > 24 hours and < 7 days with hemodynamic, respiratory and neurological stability were randomly assigned at a 1:1 ratio to the sitting position followed by passive orthostasis condition or the passive orthostasis followed by the sitting position condition. The primary outcome was lung aeration assessed using the lung ultrasound score (score ranges from 0 [better] to 36 [worse]). Results: A total of 186 subjects were screened; of these subjects, 19 were enrolled (57.8% male; mean age, 73.2 years). All participants were assigned to receive at least one verticalization protocol. Passive orthostasis resulted in mean lung ultrasound scores that did not differ significantly from the sitting position (11.0 versus 13.7; mean difference, -2.7; [95%CI -6.1 to 0.71; p = 0.11). Adverse events occurred in three subjects in the passive orthostasis group and in one in the sitting position group (p = 0.99). Conclusion: This analysis did not find significant differences in lung aeration between the sitting and passive orthostasis groups. A randomized crossover clinical trial assessing the impact of vertical positioning on lung aeration in patients receiving invasive mechanical ventilation is feasible. RESUMO Objetivo: Avaliar o impacto de diferentes posicionamentos verticais na aeração pulmonar em pacientes em ventilação mecânica invasiva. Métodos: Trata-se de ensaio clínico aberto, randomizado e transversal, realizado entre janeiro e julho de 2020. Adultos em ventilação mecânica invasiva por mais de 24 horas e menos de 7 dias com estabilidade hemodinâmica, respiratória e neurológica foram distribuídos aleatoriamente em uma proporção de 1:1 à postura sentada seguida da condição de ortostatismo passivo ou o ortostatismo passivo seguido de postura sentada. O desfecho primário foi a aeração pulmonar avaliada pelo lung ultrasound score. O escore varia de zero (melhor) a 36 (pior). Resultados: Foram selecionados 186 indivíduos; destes, 19 foram incluídos (57,8% do sexo masculino; média idade de 73,2 anos). Todos os participantes foram selecionados para receber pelo menos um protocolo de verticalização. O ortostatismo passivo resultou em escores médios de aeração pulmonar por ultrassonografia que não diferiram significativamente da postura sentada (11,0 versus 13,7; diferença média, -2,7; IC95% -6,1 a 0,71; p = 0,11). Ocorreram eventos adversos em três indivíduos no grupo ortostatismo passivo e em um no grupo postura sentada (p = 0,99). Conclusão: Esta análise não encontrou diferenças significativas na aeração pulmonar entre os grupos ortostatismo passivo e postura sentada. É factível conduzir um estudo clínico transversal randomizado para avaliar o impacto do posicionamento vertical na aeração pulmonar em pacientes em ventilação mecânica invasiva. Infelizmente, o estudo foi interrompido devido à necessidade de tratar pacientes com COVID-19. Registro ClinicalTrials.gov: NCT04176445

RESUMO: Anticarsia gemmatalis (Hübner: 1818) (Lepidoptera: Erebidae) é uma das principais pragas que acometem a cultura da soja, causando desfolha. Nos estágios vegetativos a desfolha ocorre juntamente com ervas daninhas, e no reprodutivo com patógenos. Nesse sentido, para manter a fitossanidade, é necessário realizar a utilização combinada de pesticidas. Assim, o objetivo do presente trabalho foi determinar a compatibilidade do vírus entomopatogênico AgMNPV com os principais herbicidas e fungicidas utilizados na soja em diferentes tempos de mistura. A dieta artificial foi imersa nas soluções dos pesticidas e suas misturas e fornecida às lagartas de A. gemmatalis, imediatamente e após uma e duas horas de mistura. A avaliação foi realizada quantificando o número de lagartas mortas. A mistura do vírus AgMNPV com herbicidas e fungicidas, mesmo após duas horas de mistura se mostrou compatível. Os cenários observados mostram a compatibilidade do vírus com os herbicidas e fungicidas, com percentuais de mortalidade entre 70 a 99% para A. gemmatalis.

ABSTRACT: Anticarsia gemmatalis (Hübner: 1818) (Lepidoptera: Erebidae) is one of the main pests that affect soybean crops, causing defoliation. In the vegetative stages, defoliation occurs together with weeds, and in the reproductive stages with pathogens. In this sense, to maintain plant health, it is necessary to carry out the combined use of pesticides. Thus, this research determined the compatibility of the entomopathogenic virus AgMNPV with the main herbicides and fungicides used in soy at different times of the mixture. The artificial diet was immersed in the solutions of the pesticides and their mixtures and supplied to A. gemmatalis caterpillars, immediately and after one and two hours of mixing. The evaluation was performed by quantifying the number of dead caterpillars by mixing the AgMNPV virus with herbicides and fungicides, even after two hours of mixing if compatible. The observed scenarios showed a compatibility of the virus with the herbicides and fungicides, with mortality rates between 70 to 99% for A. gemmatalis.

RESUMO Objetivo: Descrever as características e os desfechos de pacientes submetidos à retirada da ventilação mecânica e comparar a pacientes com ventilação mecânica e limitações de terapias de suporte à vida (limitar ou retirar), porém sem remoção da ventilação mecânica. Métodos: Este foi um estudo de coorte retrospectiva realizado entre janeiro de 2014 e dezembro de 2018 com pacientes em ventilação mecânica com alguma limitação de suporte artificial de vida admitidos a uma única unidade de terapia intensiva. Foram comparados os pacientes submetidos à retirada da ventilação mecânica e os que não passaram por esse procedimento com relação à mortalidade na unidade de terapia intensiva e ao tempo de permanência no hospital, em uma análise não ajustada e em uma amostra pareada por escore de propensão. Analisou-se também o tempo desde a retirada da ventilação mecânica até o óbito. Resultados: Dentre 282 pacientes com limitações a terapias de suporte à vida, 31 (11%) foram submetidos à retirada da ventilação mecânica. Não houve diferenças iniciais entre os grupos. As taxas de mortalidade na unidade de terapia intensiva e no hospital foram, respectivamente, de 71% versus 57% e 93% versus 80%, entre os pacientes submetidos à retirada da ventilação mecânica e os que não o foram. O tempo mediano de permanência na unidade de terapia intensiva foi de 7 versus 8 dias (p = 0,6), e o tempo de permanência no hospital foi de 9 versus 15 dias (p = 0,015). A mortalidade hospitalar não foi significantemente diferente (25/31; 81% versus 29/31; 93%; p = 0,26) após o pareamento. O tempo mediano desde a retirada da ventilação mecânica até o óbito foi de 2 dias [0 - 5] e 10/31 (32%) dos pacientes morreram dentro de 24 horas após a retirada dessa ventilação. Conclusão: Neste relato brasileiro, a retirada da ventilação mecânica representou 11% de todos os pacientes com limitação do tratamento e não se associou com aumento da mortalidade hospitalar após pareamento por escore de propensão das covariáveis relevantes.

Abstract Objective: To describe the characteristics and outcomes of patients undergoing mechanical ventilation withdrawal and to compare them to mechanically ventilated patients with limitations (withhold or withdrawal) of life-sustaining therapies but who did not undergo mechanical ventilation withdrawal. Methods: This was a retrospective cohort study from January 2014 to December 2018 of mechanically ventilated patients with any organ support limitation admitted to a single intensive care unit. We compared patients who underwent mechanical ventilation withdrawal and those who did not regarding intensive care unit and hospital mortality and length of stay in both an unadjusted analysis and a propensity score matched subsample. We also analyzed the time from mechanical ventilation withdrawal to death. Results: Out of 282 patients with life-sustaining therapy limitations, 31 (11%) underwent mechanical ventilation withdrawal. There was no baseline difference between groups. Intensive care unit and hospital mortality rates were 71% versus 57% and 93% versus 80%, respectively, among patients who underwent mechanical ventilation withdrawal and those who did not. The median intensive care unit length of stay was 7 versus 8 days (p = 0.6), and the hospital length of stay was 9 versus 15 days (p = 0.015). Hospital mortality was not significantly different (25/31; 81% versus 29/31; 93%; p = 0.26) after matching. The median time from mechanical ventilation withdrawal until death was 2 days [0 - 5], and 10/31 (32%) patients died within 24 hours after mechanical ventilation withdrawal. Conclusion: In this Brazilian report, mechanical ventilation withdrawal represented 11% of all patients with treatment limitations and was not associated with increased hospital mortality after propensity score matching on relevant covariates.

RESUMO: No presente trabalho objetivou-se avaliar a longevidade de operárias de A. mellifera alimentadas com dieta incorporada com os entomopatógenos comerciais Beauveria bassiana e Bacillus thuringiensis, e verificar possíveis alterações morfológicas em seu mesêntero. Para isso, os entomopatógenos utilizados foram B. bassiana (Produto A) (5,0 × 1011 conídios viáveis.kg-1), B. thuringiensis (Produto B) (2,5 × 109 esporos viáveis.g-1), B. thuringiensis (Produto C) (1,0 × 109 esporos viáveis.g-1); e dois controles: T1: água destilada esterilizada e T2: água destilada esterilizada + Tween 80® (0,01%). Para os bioensaios, 2 mL de cada tratamento foram incorporados à pasta Cândi. Para cada tratamento, 80 abelhas foram acondicionadas, individualmente, em tubos de vidro de fundo chato (2,5 cm Ø), cobertos com voile, contendo um pedaço de algodão embebido em água e pasta Cândi. Os tubos contendo as abelhas foram acondicionados em B.O.D (30 ± 2°C, U.R. 70% ± 10%, 12 h), e a mortalidade foi avaliada a cada seis horas, durante 10 dias. Logo após a verificação da mortalidade, foram separadas duas abelhas por tratamento para a retirada do mesêntero. Essas amostras foram processadas em metodologia padrão para Microscopia Eletrônica de Varredura (MEV). Verificou-se que os produtos A, B e C reduziram a longevidade das abelhas quando comparados aos controles T1 e T2. Nas análises qualitativas realizadas com MEV, não foi possível observar alterações morfológicas externas ou internas nos tecidos do mesêntero. Apesar dos produtos A, B e C causarem redução na longevidade, sua presença não foi verificada quando os tecidos foram analisados por MEV.

ABSTRACT: The present study had the objective of evaluating the longevity of A. mellifera workers fed on a diet incorporating commercial entomopathogens, Beauveria bassiana, and Bacillus thuringiensis. It also aimed at verifying possible morphological alterations in the midgut. To this purpose, the entomopathogens used were B. bassiana (Product A) (5.0 × 1011 viable conidia.kg-1), B. thuringiensis (Product B) (2.5 × 109 viable spores.g-1), and B. thuringiensis (Product C) (1.0 × 109 viable spores.g-1); and two controls: T1: sterilized distilled water, and T2: sterilized distilled water + Tween 80® (0.01%). For the bioassays, 2 mL of each treatment were incorporated into Candy paste. For each treatment, 80 bees were individually in flat bottom glass tubes (2.5 cm Ø) covered with voile, containing a piece of cotton soaked in water and Candy paste. These tubes were stored in a B.O.D (30 ± 2°C, R.H 70% ± 10%, 12 h), and mortality was evaluated every six hours, for 10 days. Soon after verifying mortality, two bees per treatment were selected for the removal of their midgut. Midgut samples were processed using standard methodology for Scanning Electron Microscopy (SEM). It was verified that products A, B, and C reduced the longevity of bees when compared to T1 and T2 controls. In the qualitative analyses carried out using SEM, it was not possible to observe external or internal morphological alterations to midgut tissues. Although products A, B, and C cause a reduction in longevity, their presence was not verified when tissues were analyzed using SEM.

ABSTRACT The coffee berry borer Hypothenemus hampei Ferrari, 1876 (Coleoptera: Curculionidae: Scolytinae) is considered the most serious pest of the coffee crop and is controlled primarily with the use of chemical insecticides. An alternative to this control method is the use of the entomopathogenic bacterium, Bacillus thuringiensis Berliner, 1911. Therefore, the objective of this work was to select strains of B. thuringiensis virulent against H. hampei and characterize them by morphological and molecular methods to identify possible genes for the production of genetically modified plants. To achieve this objective, 34 strains of B. thuringiensis underwent a selective bioassay to evaluate their toxicity to H. hampei first-instar larvae. Among the strains tested, 11 and the standard B. thuringiensis subspecies israelensis (IPS-82) caused mortality above 90%. Then, the median lethal concentration (LC50) was estimated for these strains followed by characterization using morphological, biochemical and molecular methods. The lowest LC50 was obtained for strain BR58, although this concentration did not differ significantly from that of the standard strain IPS-82 or from that of strains BR137, BR80 and BR67. The molecular characterization detected cry4A, cry4B, cry10, cry11 and cyt1 genes in 10 of the most virulent strains (BR58, BR137, BR80, BR81, BR147, BR135, BR146, BR138, BR139, BR140). Strain BR67 differed completely from the others and amplified only the cry3 gene. This strain was more virulent than BR135, BR146, BR138, BR139 and BR140, but it did not differ from BR58, BR137, BR80, BR81 and BR147. The protein profile revealed proteins of 28, 65, 70 and 130 kDa, and the morphological analysis identified spherical crystalline inclusions in all strains. The results showed that the 11 strains studied have potential for use as a gene source for insertion into coffee plants for the control H. hampei, especially the cry3, cry4A, cry4B, cry10, cry11 and cyt1 genes, that were repeated in the most virulent isolates.

ABSTRACT The selectivity of five entomopathogens and a chemical insecticide (positive control) to pupae and adults of the egg parasitoid Telenomus remus (Nixon) was evaluated in the laboratory under controlled environmental conditions according to protocols established by the International Organization for Biological Control. Baculovirus anticarsia (Baculovirus AEE®), Bacillus thuringiensis var. kurstaki (Thuricide®), Bacillus thuringiensis var. aizawai (Agree®), Beauveria bassiana (Boveril®), Metarhizium anisopliae (Metarril®) and Trichoderma harzianum (Trichodermil®) are harmless to T. remus pupae, and adults. Thus, our results suggest that the insect control strategies applied here are compatible since entomopathogens were classified as harmless to T. remus in most examined cases and therefore facilitate a joint application to control different pests. Bacillus thuringiensis var. kurstaki (Dipel®), despite being classified as slightly harmful in some of the evaluations, can still be considered compatible for use together with T. remus, especially when compared with chemical insecticides such as chlorpyrifos that might be considered harmful to the parasitoid survival.

RESUMO Novos genótipos híbridos de bananeira têm sido inseridos no mercado brasileiro, embora ainda não existam informações consistentes sobre seus possíveis impactos em relação às pragas da cultura. O moleque-da-bananeira (Coleoptera: Curculionidae) é fortemente atraído pelos voláteis do rizoma da bananeira e acredita-se que sua atração pelos genótipos híbridos seja diferente da observada para variedades. Este estudo objetivou avaliar a resposta olfativa do moleque-da-bananeira, Cosmopolites sordidus (Coleoptera: Curculionidae) a 20 genótipos de bananeira, em laboratório. De acordo com a semelhança genética e comercial dos genótipos, foram estabelecidos quatro agrupamentos: 1) Cavendish; 2) Prata; 3) Maçã/Conquista e 4) Ouro/Terra/Caipira/Figo. No bioensaio 1, a atração do moleque-da-bananeira foi avaliada entre os genótipos do mesmo agrupamento e, no bioensaio 2, os genótipos previamente selecionados foram comparados entre si. Em ambos os bioensaios, a testemunha foi representada pela ausência do rizoma. O estudo foi repetido com insetos e rizomas coletados em abril (primeira época) e julho (segunda época) do ano agrícola de 2011. Na primeira época, os genótipos selecionados foram: Nanicão (AAA); FHIA 18 (AAAB); Princesa (AAAB); Ouro (AA) e Prata anã (AAB) (bioensaio 1). Na segunda época, os genótipos selecionados foram FHIA 17 (AAAA), PA 9401 (AAAB), Thap Maeo (AAB), além de Nanicão e Ouro (bioensaio 1). No geral, apenas as variedades Nanicão (AAA) e Ouro (AA) foram mais atrativas que a testemunha (primeira e segunda épocas, respectivamente) (bioensaio 2). No entanto, não foi observada clara influência do genótipo sobre a resposta olfativa de C. sordidus, indicando que os híbridos de bananeira apresentam atratividade semelhante à das variedades.

ABSTRACT New banana plant hybrids have been cultivated in the Brazilian fields, although there is no consistent information about the possible impacts on banana pests. The banana root weevil (Coleoptera: Curculionidae) is strongly drawn to the volatile rhizome of banana, and it is believed that its attractiveness for hybrid genotypes is different than that observed on banana plant varieties. This study aimed to evaluate the olfactory response of banana root weevil, Cosmopolites sordidus (Coleoptera: Curculionidae), to 20 banana genotypes in laboratory. According to the genetic and commercial similarity of plants, the genotypes were divided into four groups: 1) Cavendish; 2) Prata; 3) Maçã/Conquista, e 4) Ouro/Terra/Caipira/Figo. In the bioassay 1, the attractiveness of the banana root weevil was evaluated for genotypes of the same group. In bioassay 2, the pre-selected genotypes were compared among themselves. The study was replicated with insects and rhizomes in April (first season) and July (second season) of 2011 agricultural year. In the first season, the selected genotypes were Nanicão (AAA), FHIA 18 (AAAB), Princesa (AAAB), Ouro (AA), and Prata anã (AAB) (bioassay 1). In the second season, the selected genotypes were FHIA 17 (AAAA), PA 9401 (AAAB), and Thap Maeo (AAB), besides Nanicão and Ouro (bioassay 1). In both bioassays, the control was represented by absence of rizome. Overall, only Nanicão (AAA) and Ouro (AA) genotypes were more attractive than the control (in the first and second seasons, respectively) (bioassay 2). Nevertheless, we observed no clear influence of genotypes on olfactory response of C. sordidus, indicating that banana plants hybrids and varieties have similar attractiveness to banana root weevil.

RESUMO. Elasmopalpus lignosellus (Zeller, 1848) (Lepidoptera, Pyralidae) é considerada praga para mais de 60 espécies de plantas cultivadas como o milho, cana-de-açúcar, trigo, soja, arroz, feijão, sorgo, amendoim e algodão. O objetivo deste trabalho foi selecionar e caracterizar isolados de Bacillus thuringiensis virulentos a E. lignosellus atuando como método alternativo de controle. Assim, 47 isolados foram avaliados em bioensaios seletivos contra lagartas de 1º ínstar de E. lignosellus. Para os isolados mais tóxicos, a concentração letal (CL50) foi estimada e a sua caracterização foi realizada por métodos morfológicos, bioquímicos e moleculares. Dos 47 isolados testados, 12 causaram mortalidade acima 85% e apresentaram CL50 entre 0.038E+8 a 0.855E+8 esporos mL-1. Os menores valores de CL50 foram obtidos pelos isolados BR83, BR145, BR09, BR78, S1534 e S1302, os quais não diferiram da linhagem padrão HD-1, com exceção do BR83. O perfil proteico revelou proteínas Cry entre 60 e 130 kDa, e a caracterização molecular mostrou a presença dos genes cry1, cry2, cry3 e cry11. A análise morfológica identificou três diferentes inclusões cristalinas: bipiramidais, esféricas e cuboides. Entre os isolados avaliados, 12 apresentam potencial biotecnológico para controle de E. lignosellus via formulação de novos bioinseticida ou produção de plantas transgênicas.

ABSTRACT. Elasmopalpus lignosellus (Zeller, 1848) (Lepidoptera, Pyralidae) is an insect pest of 60 economically important crops, including sugarcane, wheat, soybean, rice, beans, sorghum, peanuts, and cotton. The aim of this work was to select and characterize Bacillus thuringiensis isolates with insecticidal activity against E. Lignosellus that could be used as an alternative method of control. Selective bioassays were done to evaluate the toxicity of 47 isolates against first instar larvae of E. lignosellus. For the most toxic bacterial strains, the lethal concentration (LC50) was estimated and morphological, biochemical and molecular methods were used to characterize the isolates. Among the 47 isolates tested, 12 caused mortality above 85% and showed LC50 values from 0.038E+8 to 0.855E+8 spores mL-1. Isolates BR83, BR145, BR09, BR78, S1534, and S1302 had the lowest LC50 values and did not differ from the standard HD-1 strain; the exception was BR83.The protein profiles produced bands with molecular masses of 60-130 kDa. The genes cry1, cry2, cry3, and cry11 were identified in the molecular characterization. The morphological analysis identified three different crystal inclusions: bipyramidal, spherical and cuboidal. Among the tested isolates, 12 isolates have potential for biotechnological control of E. Lignosellus by development of new biopesticides or genetically modified plants.

ABSTRACT: We studied Spodoptera frugiperda development using different food sources in the laboratory and field. Newly hatched larvae were fed soybean, cotton, maize, wheat, and oat leaves. An artificial diet was used as the control. Duration of pre-pupal, pupal, and larva-adult period, pupal weight, sex ratio, survival, larva feeding preferences, oviposition preferences, and nutritional quality of different hosts were evaluated. Insects fed on wheat showed the shortest larva-adult period. The insects fed on cotton and soybean had longer larval development cycles and pupae of lower weight. Feeding preference was evident for third instar larvae and did not differ between wheat, oat, maize, and soybean, which were the preferred hosts. Moths oviposited to a greater extent on the upper canopy of wheat than that of other plants in both the no-choice and free-choice tests. Treatments influenced insect growth, food consumption, and digestion when nutritional variables were analyzed. Thus, grasses were better hosts for S. frugiperda development. Cotton was the least preferred food, followed by soybean. The present study can improve our understanding of S. frugiperda in these different crops and help in developing management strategies. Even though S. frugiperda is considered to be polyphagous, this pest is closely associated with grasses (maize, wheat, oat) and has lower potential as a soybean or cotton feeder. Howerver, S. frugiperda food intake regulation appears to be triggered by a complex of different mechanisms. Thus, S. frugiperda can also damage soybean and cotton and adapt to them in the absence of preferred hosts.

RESUMO: Neste estudo, objetivamos investigar a infestação de Cosmopolites sordidus ( Coleoptera: Dryophthoridae ) em diferentes genótipos de bananeira, em um pomar comercial de bananeira, com idade de 30 meses. Para isso, comparou-se a infestação da broca-da-rizoma em 20 genótipos de bananeira, sendo cinco variedades e quinze híbridos. No geral, foi observado que 94,17% da infestação da praga ocorreu na região do córtex do rizoma, e apenas 5,83% das plantas apresentaram infestação no cilindro central. Os materiais menos sensíveis à infestação foram as variedades Prata Anã (AAB) e Pacovan (AAB), nas quais não foi registrada ocorrência da praga. Entre os genótipos híbridos, de forma geral, os materiais PV 9401 e BRS FHIA 18 foram os que apresentaram sensibilidade intermediária, enquanto os híbridos BRS Tropical (AAAB), PA 9401 (AAAB), BRS Vitoria (AAAB), YB-4203 (AAAB) e Bucaneiro (AAAA) foram os mais sensíveis ao ataque da broca-do-rizoma. Este estudo demonstra que a infestação da broca-do-rizoma varia de acordo com o genótipo, e a seleção de materiais menos suscetíveis auxilia na redução da infestação de C. sordidus.

ABSTRACT: In this study, we aimed to investigate Cosmopolites sordidus (Coleoptera: Dryophthoridae ) infestation among different banana genotypes in a commercial banana orchard over the course of 30 months. Banana root borer infestation was compared in 20 banana genotypes, including five varieties and 15 hybrids. Overall, we observed that 94.17% of pest infestation cases occurred in the cortex region, and only 5.83% occurred in the central cylinder. Genotypes least sensitive to infestation were the Prata Anã (AAB) and Pacovan (AAB) varieties, where no damage was recorded. Among the hybrid genotypes, PV 9401 and BRS Fhia 18 showed intermediate levels of sensitivity, while BRS Tropical hybrids (AAAB), PA 9401 (AAAB), BRS Vitoria (AAAB), YB 4203 (AAAB), and Bucaneiro (AAAA) were the most sensitive to attack by banana root borer. This study demonstrated that the infestation of the banana root borer varies according banana plant genotype, and the utilization of less susceptible genotypes could reduce infestation rates of C. sordidus.

RESUMO: Um dos fatores limitantes à utilização em maior quantidade de fungos entomopatogênicos produzidos é a dificuldade na manutenção da viabilidade dos conídios por longos períodos de armazenamento, o que torna importante a necessidade de desenvolvimento de formulações e embalagens que aumentem a vida de prateleira desses micro-organismos. Diante disso, o objetivo deste trabalho foi identificar embalagens que pudessem manter a viabilidade dos conídios por longos períodos de armazenamento em diferentes temperaturas. O trabalho foi desenvolvido no Laboratório de Controle Microbiano de Insetos da Universidade Estadual de Londrina (UEL). Foi utilizada uma mistura granulada (conídios e arroz parboilizado) de B. bassiana (CG 432) acondicionada em 3 tipos de embalagens: Coex, poliéster metalizado e polietileno, que foram armazenadas durante 180 dias em refrigerador (6 ± 2°C) e câmara climatizada (25 ± 1°C). Os parâmetros utilizados para avaliar a influência das embalagens sobre a viabilidade dos conídios foram a porcentagem de germinação e o conteúdo de água. Os conídios acondicionados na embalagem tipo Coex foram os que apresentaram maior porcentagem de germinação em refrigerador e à temperatura ambiente durante todo o período de armazenamento, sendo também a embalagem que proporcionou menor ganho de água pelos conídios durante o armazenamento. Independentemente da embalagem, os conídios armazenados em refrigerador apresentaram maior porcentagem de germinação em relação aos armazenados à temperatura ambiente (25 ± 1°C).

ABSTRACT: One of the limiting factors in the use of more substantial amounts of entomopathogenic fungi is the difficulty in maintaining conidial viability for long periods in storage. The need for developing formulations and packaging that would increase the shelf life of these microorganisms is therefore important. Considering this, this study aimed to identify packaging types that would maintain conidial viability for long storage periods at different temperatures. A granular B. bassiana (CG 432) mixture (conidia and parboiled rice) was used, stored in three different packaging types: Coex, metallized polyester, and polyethylene, which were stored in refrigerator (6 ± 2°C) and in an incubator (25 ± 1°C) for 180 days. Germination percentage and water content were the parameters used to evaluate packaging influence on conidial viability. The conidia stored in Coex packaging showed the highest germination percentage in refrigerator and at room temperature during the entire storage period. This packaging also provided the lowest water acquisition by the conidia during storage. Regardless of packaging, the conidia stored in refrigerator showed the highest germination percentage in relation to those stored at room temperature (25 ± 1°C).

Resumo:O objetivo deste trabalho foi selecionar e caracterizar molecularmente isolados de Bacillus thuringiensistóxicos a Spodoptera eridaniaeS. frugiperda. Trinta e quatro isolados foram submetidos ao bioensaio, dos quais três foram selecionados e usados para a estimativa da CL50. Os isolados selecionados não diferiram da linhagem padrão HD-1. Na caracterização molecular, identificou-se a presença dos genes cry1 e cry2, nos isolados BR37 e BR94, e dos genes cry4A, cry4B, cry10, cry11 e cyt1 no isolado BR58, o que confirmou o perfil proteico obtido de 130, 70 e 65 kDa. Foram identificados cristais bipiramidais e esféricos. O isolado BR58, apesar de não conter os genes relacionados à toxicidade a Lepidoptera, causa mortalidade em ambas as espécies

Abstract:The objective of this work was to select and molecularly characterize Bacillus thuringiensis isolates toxic to Spodoptera eridaniaand S. frugiperda. Thirty four isolates were subjected to the bioassay, of which three were selected and used to estimate the LC50. The selected isolates did not differ from the HD-1 standard line. In the molecular characterization, the presence of the cry1 and cry2 genes was identified in the BR37 and BR94 isolates, and of the cry4A, cry4B, cry10, cry11, and cyt1 genes in the BR58 isolate, which confirmed the obtained protein profile of 130, 70, and 65 kDa. Bipyramidal and spherical crystals were identified. The BR58 isolate, although not containing the genes related to toxicity to Lepidoptera, causes mortality in both species.

The green stink bug Nezara viridula is an important pest in many crops and regions of the world. In this study the effect of pyriproxyfen on fecundity and fertility of N. viridula and the effect of diflubenzuron in the reproductive system was evaluated. Two bioassays were conducted in laboratory. In the first, low concentration of pyriproxyfen was tested on adult pairs of N. viridula. After copulation, we assessed the number of eggs/female (fecundity) and the number of hatched nymphs (fertility). In the second bioassay, we evaluated the effect of diflubenzuron in the morphology of female and male reproductive system of N. viridula. The results show that pyriproxyfen, at the tested concentration did not reduce significantly the number of eggs/female (23.61%). A decrease of 52.39% in eggs fertility, was observed preventing nymphs hatching probably due to embryo malformation. Diflubenzuron affected female and male development and reproductive organs. Thus, both pyriproxyfen and diflubenzuron have potential to decrease N. viridula progeny and can be included in integrated pest management. However, field studies are needed.

O percevejo Nezara viridula é uma praga importante em varias culturas e regiões do mundo. Neste estudo, avaliou-se o efeito do inseticida do piriproxifem sobre a fecundidade e fertilidade de N. viridula e o efeito do diflubenzurom na formação do aparelho reprodutor. Foram realizados dois bioensaios em laboratório. No primeiro bioensaio, experimentou-se uma baixa concentração do piriproxifem sobre casais adultos de N. viridula. Posterior à cópula, avaliou-se o número de ovos/fêmea (fecundidade) e o número de ninfas eclosionadas (fertilidade). No segundo bioensaio, avaliou-se o efeito do diflubenzurom na morfologia do aparelho reprodutor feminino e masculino de N. viridula. Os resultados indicam uma redução de 23,61% no número de ovos por fêmea provocada pelo piriproxifem, na concentração testada, que não foi significativa em relação à testemunha. Entretanto, 52,39% da fertilidade de ovos diminuiu, impedindo a eclosão de ninfas, provavelmente por afetar a formação do embrião. Entretanto, o diflubenzuron afetou o desenvolvimento dos órgãos femininos e masculinos. Tanto o piriproxifem e o diflubenzurom tem potencial de diminuir a progênie de N. viridula, e podem ser incluídos em programas de manejo integrado pragas, uma vez que sejam realizados estudos de eficácia dos produtos a campo.

A hipertensão arterial resistente e uma condição clinica que carece de investigação etiológica, cujo objectivo e identificar causas secundarias passiveis de serem corrigidas. Os autores descrevem uma apresentação rara de hipertensão secundaria. O caso consiste num doente que e seguido em consulta externa desde Janeiro de 2012 por insuficiência renal e hipertensão diagnosticadas ha um ano. Foi realizado um estudo etiológico no qual constou um estudo endocrinológico que demonstrou um hiperaldosteronismo secundário e uma ecografia renal que revelou um rim esquerdo de dimensões reduzidas mas com uma ecografia com Doppler sem alterações do fluxo sanguíneo ao nivel das artérias renais. Posteriormente o doente foi internado no serviço de nefrologia por hipertensão maligna com retinopatia hipertensiva com sinais de hemorragia ao nível da retina. O doente apresentou vários episódios de crises convulsivas tendo sido admitido na Unidade de Cuidados Intensivos onde foi entubado e ventilado. Para exclusão de causas de hipertensão renovascular foi realizado uma tomografia axial computorizada que revelou extenso trombo envolvendo a aorta distal e a artéria renal esquerda. O estudo imunológico realizado demonstrou positividade para os anticorpos anti-fosfolipido, anti-cardiolipina.

Resistant hypertension is a clinical condition that needs an aetiological investigation with the purpose of establishing if there is a secondary cause that can be treated. The authors describe a rare cause of secondary hypertension. We report a case of a 40-year-old Caucasian man followed in an outpatient nephrology clinic, since January 2012, with renal insufficiency and hypertension known for a year. An initial aetiological study was performed, including an endocrinological study, which revealed a secondary hyperaldosteronism, a renal ultrasound that further revealed a diminished left kidney and a renal artery Doppler ultrasound that described a normal arterial blood flow. The patient was admitted in the nephrology department presenting malignant hypertension that included hypertensive retinopathy with retinal haemorrhage. At the time, the patient initiated several convulsive crises and had to be admitted in the intensive care unit, needing invasive mechanical ventilation. To exclude renovascular disease causing the clinical scenario, a computed tomography angiography was performed, showing extensive thrombosis of the distal aorta involving at least the renal left artery. The immunologic study showed positive antiphospholipid antibodies, anticardiolipin antibodies and anti-beta2-glycoprotein 1 antibodies.

Selectivity of entomopathogenic fungus M. anisopliae to egg parasitoid T. podisi was evaluated. Three bioassays were realized: in the first, a suspension of M. anisopliae conidia was sprayed over sterilized eggs of Euschistus heros and exposed to parasitism, in the second, conidial suspensions were sprayed over parasitized eggs and in the third, the suspensions were sprayed over glass plates (IOBC/WPRS) and dried at ambient temperature, forming a layer of conidia for adult parasitoids contact. The control group was sprayed with sterile distillated water + Tween 80 (0.01%). To evaluate the percentage of parasitism, emergence and viability of parasitoids, the cards containing parasite eggs were introduced in plastic bags and observed using a stereoscopic microscope. The results indicate that M. anisopliae do not affect T. podisi viability and parasitism, but its emergence can be reduced 25.81%, if the fungus is sprayed on parasitized eggs. Possibly, the fungus penetrates the egg and infects the parasitoid at some stage. Nevertheless, emergence reduction compared to control was low. Therefore, M. anisopliae can be considered selective to T. podisi, and can be used jointly, separating at maximum the spraying of fungus and the releases of parasitoid.

A seletividade do fungo entomopatogênico M. anisopliae ao parasitóide de ovos T. podisi foi avaliada com a realização de tres bioensaios: no primeiro, foram pulverizados suspensões de conídios de M. anisopliae sobre ovos esterilizados de Euschistus heros e expostos ao parasitismo; no segundo bioensaio, as suspensões do fungo foram pulverizadas sobre ovos parasitados, e no terceiro bioensaio, as suspensões do fungo foram pulverizadas sobre placas de vidro das jaulas de exposição (IOBC/WPRS) e secas a temperatura ambiente, formando uma película de conídios para permitir o contato com os parasitóides adultos. As testemunhas foram pulverizadas com água destilada estéril + Tween 80 a 0,01%. Para avaliar a porcentagem de parasitismo, emergência e viabilidade dos parasitóides, as cartelas de ovos contendo os parasitóides foram acondicionadas em sacos plásticos e observadas a traves de um microscópio estereoscópico. Os resultados obtidos neste estudo indicam que o M. anisopliae não afeta o parasitismo e a viabilidade de T. podisi, porêm, a emergência do parasitóide pode reduzir 25,81% se o fungo é pulverizado sobre ovos parasitados. Possivelmente, o fungo penetra o ovo do hospedeiro e infeta ao parasitóide em alguma fase. Entretanto, a reducção da emergência em comparação á testemunha é baixa. Por tanto, M. anisopliae pode ser considerado seletivo a T. podisi, e ambos podem ser utilizados conjuntamente, separando ao máximo as pulverizações do fungo e as liberações do parasitóide.
