Introduction: Evaluating the scarring differences between areas previously anesthetized with different local anesthetics is interesting to verify whether the choice of anesthetic helps in surgical healing in relation to inflammation, neovascularization and collagen deposition. Objectives: Review the existing literature with a focus on analyzing the role of local anesthetics in aiding the healing of surgical wounds. Method: Narrative review of literature selected from the SciELO – Scientific Electronic Library Online, Google Scholar, Pubmed and Scopus platforms. Initially, the review searched for keywords in the focus of the research, based on MESH/DeCS with the following terms: “local anesthetics, healing, neovascularization, collagen” with AND or OR search, considering the title and/or abstract. Next, only those that were most related to the topic were included, and the texts were read for inclusion. Results: 73 articles were included, read and interpreted focusing on the role of anesthetics in healing, pain, side effects and the best that currently emerge in analgesia and scar potential. Conclusions: No differences were observed between the anesthetics studied in terms of healing, inflammation and neovascularization. However, there was a significant difference in the production of collagens with an intensity of 6 to 12 times greater than collagen types I, II and III with the use of levobupivacaine.

Introdução: Avaliar as diferenças cicatriciais entre áreas previamente anestesiadas com diferentes anestésicos locais é interessante para verificar se a escolha do anestésico auxilia na cicatrização cirúrgica em relação à inflamação, neovascularização e deposição de colágeno. Objetivos: Revisar a literatura existente com foco de análise sobre o papel dos anestésicos locais no auxílio da cicatrização de feridas cirúrgicas. Método: Revisão narrativa da literatura selecionada das plataformas SciELO – Scientific Electronic Library Online, Google Scholar, Pubmed e Scopus.  Inicialmente a revisão realizou a busca por palavras-chave no foco da pesquisa, com base no MESH/DeCS com os seguintes termos: “anestésicos locais, cicatrização, neovascularização, colágeno” com busca AND ou OR, considerando o título e/ou resumo. A seguir, foram incluídos somente os que tinham maior relação ao tema, e realizada a leitura dos textos para inclusão. Resultados: Foram incluídos, lidos e interpretados 73 artigos enfocando o papel dos anestésicos na cicatrização, dor, efeitos colaterais e os melhores que despontam na atualidade na analgesia e potencial cicatricial. Conclusões: Não se observou diferenças entre anestésicos pesquisados quanto à cicatrização, inflamação e neovascularização. Contudo, verificou-se diferença significativa na produção de colágenos com intensidade de 6 a 12 vezes maior de colágenos tipos I, II e III com o uso da levobupivacaína.

Introduction: Evaluating the scarring differences between areas previously anesthetized with different local anesthetics is interesting to verify whether the choice of anesthetic helps in surgical healing in relation to inflammation, neovascularization and collagen deposition. Objectives: To analyze the influence of ropivacaine and levobupivacaine on the inflammatory phase of surgical incision healing. Method: Thirty male Wistar rats were divided into 3 groups to evaluate the effects of these local anesthetics in comparison with each other and with a control group. The animals were subjected to general anesthesia and subsequently scheduled for administration of the solutions and their samples were excised after 5 days. The scar area, neovascularization and collagen deposition were studied. Results: Regarding inflammation and neovascularization, there were no significant differences between the groups, with all of them presenting pronounced levels. As for collagen deposition, this group exhibited significantly higher levels of type I, II, and III collagens compared to groups C and R. Regarding collagen deposition, this group demonstrated significantly higher levels of type I, II and III collagen when compared to groups C and R. Conclusions: No differences were observed between the groups in relation to the variation in scar measurements, inflammation and neovascularization. However, there was a significant difference in the production of collagens with an intensity of 6 to 12 times greater in collagen types I, II and III with the use of levobupivacaine.

Introdução: Avaliar as diferenças cicatriciais entre áreas previamente anestesiadas com diferentes anestésicos locais é interessante para verificar se a escolha do anestésico auxilia na cicatrização cirúrgica em relação à inflamação, neovascularização e deposição de colágeno. Objetivos: Analisar a influência da ropivacaína e levobupivacaína na fase inflamatória da cicatrização de incisões cirúrgicas. Método: Trinta ratos Wistar machos foram divididos em 3 grupos para avaliar os efeitos desses anestésicos locais em comparação entre si e com grupo controle. Os animais foram submetidos à anestesia geral e, posteriormente, marcados para a administração das soluções e terem excisadas suas amostras após 5 dias. Estudou-se a área cicatricial, neovascularização e deposição de colágenos. Resultados: Em relação à inflamação e neovascularização, não houve diferenças significativas entre os grupos., todos apresentando níveis acentuados. Quanto à deposição de colágenos, este grupo demonstrou níveis significativamente mais altos de colágenos tipo I, II e III quando comparados aos grupos C e R. Conclusões: Não se observou diferenças entre os grupos em relação à variação das medidas cicatriciais, inflamação e à neovascularização. Entretanto, verificou-se sim diferença significativa na produção de colágenos com intensidade de 6 a 12 vezes maior de colágenos tipos I, II e III com o uso da levobupivacaína.

Abstract Objectives: To review and provide evidence-based recommendations for the diagnosis and treatment of otosclerosis. Methods: Task force members were educated on knowledge synthesis methods, including electronic database search, review and selection of relevant citations, and critical appraisal of selected studies. Articles written in English or Portuguese on otosclerosis were eligible for inclusion. The American College of Physicians’ guideline grading system and the American Thyroid Association’s guideline criteria were used for critical appraisal of evidence and recommendations for therapeutic interventions. Results: The topics were divided into 2 parts: 1) Diagnosis – audiologic and radiologic; 2) Treatment – hearing AIDS, pharmacological therapy, stapes surgery, and implantable devices - bone-anchored devices, active middle ear implants, and Cochlear Implants (CI). Conclusions: The pathophysiology of otosclerosis has not yet been fully elucidated, but environmental factors and unidentified genes are likely to play a significant role in it. Women with otosclerosis are not at increased risk of worsening clinical condition due to the use of contraceptives or during pregnancy. Drug treatment has shown little benefit. If the patient does not want to undergo stapedotomy, the use of hearing aids is well indicated. Implantable systems should be indicated only in rare cases, and the CI should be indicated in cases of profound deafness.

RESUMO Estabelecer os procedimentos adequados para condução do teste de germinação em espécies florestais é importante para viabilizar a comercialização de lotes de sementes, sobretudo para espécies que ainda não constam nas Instruções Normativas. Objetivou-se com este estudo determinar as condições ideais de temperatura e substrato para a realização do teste de germinação de sementes de Senna siamea, bem como: (i) caracterizar a morfo-fisiologia do estabelecimento da plântula, (ii) o período para a avaliação das plântulas, e (iii) validar a eficiência da metodologia em 10 lotes de sementes. O delineamento utilizado foi inteiramente ao acaso em arranjo fatorial 2 x 2 (temperaturas x substratos) com 12 repetições de 50 sementes para cada tratamento. Para a avaliação dos diferentes lotes, os dados foram submetidos à análise de variância e as médias comparadas pelo teste de Scott-Knott ao nível de 5% de probabilidade. Avaliou-se o percentual e a velocidade de germinação, primeira contagem, comprimento de plântulas, germinação acumulada, e morfologia de plântulas. Sementes de S. siamea possuem maior potencial germinativo em substrato rolo de papel sob temperaturas de 25 e 30 °C (86% e 87%). A primeira contagem de germinação e avaliação final de plântulas deve ser feita aos 15 e 21 dias após a semeadura, respectivamente. A espécie possui germinação do tipo epígea e suas plântulas são fanerocotiledonares. Observou-se que as condições de execução e avaliação do teste de germinação recomendadas neste estudo são adequadas na avaliação de lotes de sementes de S. siamea.

ABSTRACT Establishing the appropriate procedures for conducting germination tests in forest species is important for marketing the seeds, especially species that are not yet included in the relevant Normative Instructions. The aim of this study was to determine the ideal conditions of temperature and substrate to carry out the germination test on seeds of Senna siamea, as well as: (i) to characterise the morpho-physiology of seedling establishment, (ii) the period for seedling evaluation, and (iii) validate the efficiency of the methodology in 10 batches of seeds. The experimental design was completely randomised in a 2 x 2 factorial scheme (temperatures x substrates), with 12 replications of 50 seeds in each treatment. To evaluate the different batches, the data were submitted to analysis of variance, and the mean values compared by the Scott-Knott test at 5% probability. The percentage and speed of germination, first count, seedling length, accumulated germination, and seedling morphology were evaluated. The germination potential of S. siamea seeds is greater on paper-roll substrate at temperatures of 25 °C and 30 °C (86% and 87%). The first germination count and final seedling evaluation should be carried out 15 and 21 days after sowing, respectively. The species has epigeal germination and the seedlings are phanerocotyledonous. It was found that the method for carrying out and evaluating the germination test recommended in this study is suitable for evaluating batches of S. siamea seeds.

Abstract Background Exercise plays a positive role in the course of the ischemic heart disease, enhancing functional capacity and preventing ventricular remodeling. Objective To investigate the impact of exercise on left ventricular (LV) contraction mechanics after an uncomplicated acute myocardial infarction (AMI). Methods A total of 53 patients was included, 27 of whom were randomized to a supervised training program (TRAINING group), and 26 to a CONTROL group, who received usual recommendations on physical exercise after AMI. All patients underwent cardiopulmonary stress testing and a speckle tracking echocardiography to measure several parameters of LV contraction mechanics at one month and five months after AMI. A p value < 0.05 was considered statistically significant for the comparisons of the variables. Results No significant difference were found in the analysis of LV longitudinal, radial and circumferential strain parameters between groups after the training period. After the training program, analysis of torsional mechanics demonstrated a reduction in the LV basal rotation in the TRAINING group in comparison to the CONTROL group (5.9±2.3 vs. 7.5±2.9o; p=0.03), and in the basal rotational velocity (53.6±18.4 vs.68.8±22.1 º/s; p=0.01), twist velocity (127.4±32.2 vs. 149.9±35.9 º/s; p=0.02) and torsion (2.4±0.4 vs. 2.8±0.8 º/cm; p=0.02). Conclusions Physical activity did not cause a significant improvement in LV longitudinal, radial and circumferential deformation parameters. However, the exercise had a significant impact on the LV torsional mechanics, consisting of a reduction in basal rotation, twist velocity, torsion and torsional velocity which can be interpreted as a ventricular “torsion reserve” in this population.

Resumo Fundamento O exercício exerce um papel positivo na evolução da doença cardíaca isquêmica, melhorando a capacidade funcional e prevenindo o remodelamento ventricular. Objetivo Investigar o impacto do exercício sobre a mecânica de contração do ventrículo esquerdo (VE) após um infarto agudo do miocárdio (IAM) não complicado. Métodos Um total de 53 pacientes foram incluídos e alocados aleatoriamente em um programa de treinamento supervisionado (grupo TREINO, n=27) ou em um grupo CONTROLE (n=26) que recebeu recomendações usuais sobre a prática de exercício físico após um IAM. Todos os pacientes realizaram um teste cardiopulmonar e um ecocardiograma com speckle tracking para medir vários parâmetros da mecânica de contração do VE em um mês e cinco meses após o IAM. Um valor de p <0,05 foi considerado para significância estatística nas comparações das variáveis. Resultados Não foram encontradas diferenças nas análises dos parâmetros de strain circunferencial, radial ou longitudinal do VE entre os grupos após o período de treinamento. Após o programa, a análise da mecânica de torção revelou uma redução na rotação basal do VE no grupo TREINO em comparação ao grupo CONTROLE (5,9±2,3 vs. 7,5±2.9o; p=0,03), bem como na velocidade rotacional basal (53,6±18,4 vs. 68,8± 22,1 º/s; p=0,01), velocidade de twist (127,4±32,2 vs. 149,9±35,9 º/s; p=0,02) e na torção (2,4±0,4 vs. 2,8±0, º/cm; p=0,02). Conclusões A atividade física não causou melhora significativa nos parâmetros de deformação longitudinal, radial ou circunferencial do VE. No entanto, o exercício teve um impacto significativo sobre a mecânica de torção do VE, que consistiu em uma redução na rotação basal, na velocidade de twist, na torção, e na velocidade de torção, que pode ser interpretada como uma “reserva” de torção ventricular nessa população.

Resumo: A tecnologia de aplicação do plasma atmosférico consiste em um método rápido e prático, que apresenta potencial de modificação da superfície tegumentar de sementes com dureza tegumentar. O objetivo desta pesquisa foi avaliar o efeito da aplicação do plasma frio atmosférico em sementes de Pityrocarpa moniliformis, verificando suas implicações sobre a impermeabilidade do tegumento, germinação e vigor. As sementes foram submetidas ao plasma por 1,5; 2,0; 3,0; 4,0 e 5,0 minutos, utilizando-se como controle, sementes sem nenhum tratamento. Após a aplicação, determinou-se a molhabilidade do tegumento, curva de embebição e condutividade elétrica da solução de embebição. As sementes também foram submetidas ao teste de germinação, tendo-se, a partir dos seus resultados determinado a viabilidade, mediana, uniformidade e assimetria da germinação. As sementes submetidas ao plasma por 4,0 e 5,0 minutos apresentaram os menores ângulos de contato aparente, 64 e 61°, respectivamente, caracterizando maior molhabilidade do tegumento, entre os tratamentos testados. Maior germinação foi observada nos tratamentos com aplicação do plasma quando comparado com o controle. A tecnologia de aplicação do plasma atmosférico tem potencial para ser utilizada como acelerador da germinação de sementes de P. moniliformis.

Abstract: Atmospheric plasma application technology consists of a fast and practical method, which has the potential to modify the surface of seeds with integumentary hardness. The aim of this research was to evaluate the effect of applying atmospheric cold plasma on Pityrocarpa moniliformis seeds, assessing its implications on the seed coat impermeability, germination and vigor. Cells were subjected to plasma for 1.5, 2.0, 3.0, 4.0 and 5.0 minutes, using seeds without any treatment as a control. After application, the seed coat wettability, imbibition curve and electrical conductivity of the soaking solution were determined. Seeds were also subjected to the germination test, and its results were used to determine viability, median, uniformity and asymmetry of germination. Seeds subjected to plasma for 5.0 and 4.0 minutes showed the lowest apparent contact angles, 64 and 61°, respectively, characterizing greater wettability of the seed coat among the treatments tested. Higher germination was observed in the treatments with plasma application when compared to the control. Atmospheric plasma application technology has the potential to be used as an accelerator of P. moniliformis seed germination.

RESUMO A utilização de ciclos de hidratação descontínua pode atuar na melhoria da germinação de sementes e crescimento inicial de plântulas sob estresse hídrico. Nesse sentido, objetivou-se avaliar o efeito da hidratação descontínua na germinação de sementes de Mimosa caesalpiniifolia e Pityrocarpa moniliformis sob estresse hídrico. Para isso, sementes dessas espécies foram submetidas a 0 (sem hidratação), 2 e 4 ciclos de hidratação (18 h) e desidratação (12 e 18 h, respectivamente), com posterior germinação sob estresse hídrico simulado para os potenciais osmóticos 0,0 (água destilada); -0,2; -0,4; -0,6 e -0,8 MPa. As plântulas foram avaliadas por meio dos testes de germinação, índice de velocidade de germinação, tempo médio de germinação e comprimento de plântulas. A hidratação descontínua em sementes de M. caesalpiniifolia e P. moniliformis prejudica a germinação e o vigor de plântulas sob estresse hídrico até -0,6 MPa. No entanto, a utilização de dois ciclos em P. moniliformis favorece a germinação sob estresse hídrico mais severo (-0,8 MPa).

ABSTRACT The use of discontinuous hydration cycles can act in improving seed germination and seedling initial growth under water stress. In this context, the objective of this study was to evaluate the effect of discontinuous hydration on the germination of Mimosa caesalpiniifolia and Pityrocarpa moniliformis seeds under water stress. For this, seeds of these species were subjected to 0 (without hydration), 2 and 4 hydration cycles (18 h) and dehydration (12 and 18 h, respectively), with subsequent germination under water stress simulated for osmotic potentials of 0.0 (distilled water), -0.2, -0.4, -0.6 and -0.8 MPa. Seedlings were evaluated using tests of germination, germination speed index, mean germination time and seedling length. Discontinuous hydration in M. caesalpiniifolia and P. moniliformis seeds hampers germination and vigor of seedlings under water stress up to -0.6 MPa. However, the use of two cycles in P. moniliformis favors germination under more severe water stress (-0.8 MPa).

The purpose of this study was to analyze the spatial variability of soil properties and productivity and the chemical and physical properties of oranges (Monte Parnaso variety) in their geographical quadrants. This experiment was conducted in a commercial orange orchard in the municipality of Nova Laranjeiras in the State of Paraná, Brazil. A GPS receiver and a total station were used to measure the spatial location of the fruit and to determine which quadrant they were located in. In this experiment, 13 trees were studied. Twelve fruits were evaluated in each geographical quadrant, and 156 fruits were considered. After harvest, the productivity was expressed as kg per quadrant (kg quadrant-1). Next, physical tests (equatorial diameter (EqD), total fruit mass (TFM), juice yield (JY)) and chemical tests (total soluble solids (TSS), total titrable acidity (TTA), the TSS/TTA ratio, and the ascorbic acid (AA) content) were conducted in the laboratory. In addition, the data were subjected to analysis by descriptive statistics and geostatistics to determine the spatial variability of the physical and chemical properties of the oranges, the variations of the soil properties and the correlations between the following parameters: the pH properties of the fruit and Zn; TFM and K; EqD and K, and C and TSS. The orchard varies spatially, especially regarding the EqD, TFM, JcP, TSS and AA for the quadrants with the highest values (the Northwest, Southeast, Northeast, Southwest and Southeast, respectively). The soil potassium and zinc contents were strongly correlated with the physical and chemical properties of the orange fruits.

El objetivo de este estudio ha sido analizar la variabilidad espacial de los atributos del suelo, de la productividad y de las propiedades físicas y químicas de los frutos de la variedad de naranja "Monte Parnaso", en relación a los cuadrantes geográficos en que se ubican los frutos analizados. El experimento se realizó en un huerto comercial de naranjos en Nova Laranjeiras, Estado de Paraná, Brasil. En el procedimiento de georreferenciación de la área y de cada árbol de la muestra, se utilizó un GPS y una estación total para medir la localización espacial de la fruta y luego la definición de qué cuadrante se ubicaban. En el experimento, se utilizaron 13 árboles en las cuales se analizaron tres frutas por cuadrante geográfico de cada árbol, por un total de 156 frutas. Después de la cosecha se estimó la productividad en kg cuadrante-1 y en el laboratorio fueran realizados análisis física (diámetro ecuatorial (ED), masa total de la fruta (MT), rendimiento de jugo (RS)) y químicos (sólidos solubles totales (SST), acidez titulable (AT), relación SST/AT y ácido ascórbico (AA)) de la fruta. Los datos fueron sometidos a análisis de la estadística descriptiva y de la geoestadística señalando la variabilidad espacial de las propiedades físico-químicas de las naranjas y los atributos del suelo y también correlación entre los atributos de la fruta pH y Zn, MT y K, ED y K y finalmente SST y C. El huerto tiene variaciones espaciales principalmente para los atributos diámetro ecuatorial, masa total de la fruta, rendimiento del jugo, SST y ácido ascórbico, siendo los cuadrantes con mayores valores fueron, respectivamente, noroeste, sureste, noreste, suroeste y sureste. Especialmente potasio y zinc, las propiedades del suelo, fuertemente correlacionados con las propiedades físicas y químicas de las frutas de color naranja.

Abstract: Atomic vapor laser isotope separation has been studied at the Institute for Advanced Studies for nuclear purposes since 1982, and recently it has been questioned about its potentialities for the aerospace area. Many applications from nuclear propulsion to electricity generation and space navigation have been found, which justify the study of isotope separation for aerospace applications. One of the key process, and the first step for atomic vapor laser isotope separation, is the production of a neutral vapor jet. This paper discussed the potentiality of using laser ablation as a tool to generate neutral metal vapor jet for isotope separation purposes. The basis for the discussion is a set of experimental results obtained at the Institute for Advanced Studies. The experiments were described, the results were analyzed using basic theoretical treatment found in the literature, and it was concluded that laser ablation is a potential tool for the generation of a neutral vapor jet for atomic vapor laser isotope separation purposes.

A ferrugem da folha de trigo, causada por Puccinia triticina é uma doença de importância econômica na cultura do trigo (Triticum aestivum). Os sintomas são mais evidentes na superfície superior da folha, mas também podem ocorrer na bainha. Neste trabalho, determinou-se a taxa de progresso e a distribuição das lesões para a construção de modelos de simulação. Amostras de folhas com sintomas da doença foram coletadas em lavouras comerciais de trigo em Tamandaré do Sul, RS, em 2006 e 2007. Uma imagem digital foi obtida de cada amostra. Um software, foi usado para auxiliar na contagem e localização das lesões diretamente da imagem de cada uma das folhas amostradas. O conjunto de dados representando o progresso da epidemia foi ajustado ao modelo logístico com três parâmetros. Pela análise descritiva, as lesões de ferrugem da folha seguem um padrão aleatório no início da epidemia e passam a agregado à medida que a epidemia avança no tempo. Foi possível com o modelo log-polinomial cúbico delimitar a região da folha onde seria mais provável de formação de futuras lesões. A continuidade na exploração destas técnicas de estatística poderá contribuir na modelagem das relações patógeno-hospedeiro. Além disso, o modelo poderá ser usado útil para gerar escalas diagramáticas com gradientes de severidade.

Wheat leaf rust caused by Puccina triticina is an important disease affecting wheat (Triticum aestivum) worldwide. Disease symptoms are normally shown on the upper leaf surface, but they can be observed in the leaf sheaths. In this work, epidemic rate and lesion distribution were characterized for the development of a simulation model. Diseased leaves were collected at regular intervals during the 2006 and 2007 seasons, in Tamandaré do Sul, RS. Two commercial wheat fields were used for sampling. A digital image was obtained for each sampled leaf. Software was used to assist counting and determining the exact location of leaf rust lesions. The quantification of the number of lesions was made on individual leaves. The data set representing the epidemic progress was adjusted to a logistic model with three parameters. A descriptive analysis indicated that the position of leaf rust lesions followed a random pattern at the beginning of the epidemic, shifting to an aggregate pattern as the epidemic advanced in time. The cubic log- polynomial model successfully predicted portions of leaf area more likely to bear lesions. Further exploration of these techniques could help the statistical modeling of host-pathogen relationships. Moreover, the model can be used to generate a useful diagrammatic scale with gradients of severity.

Plântulas de feijão das cultivares Fepagro 26, Guapo Brilhante, Iraí, Macotaço e US pinto 111 foram submetidas aos tratamentos com e sem adição de ozônio ao ar ambiente, com o objetivo de avaliar os efeitos da exposição sobre a fotossíntese, permeabilidade relativa a eletrólitos, abscisão foliar e biomassa das plântulas. A exposição ao ozônio causou decréscimos significativos na assimilação líquida de todas as cultivares, exceto na Iraí. Causou também um aumento significativo na permeabilidade relativa a eletrólitos da cultivar Pinto, mas apenas quando a AOT40 foi a mais elevada. Além disso, produziu uma antecipação significativa no tempo de abscisão foliar nas cultivares Pinto, Fepagro e Guapo. A variabilidade observada nas medidas de biomassa total, refletiu as limitações para se executar estudos de longo prazo em condições ambientais controladas, um dos principais desafios ainda por ser superado antes que se possa obter dados mais conclusivos sobre os danos em espécies cultivadas resultantes do enriquecimento do ozônio toposférico.

Bean seedlings of Fepagro 26, Guapo Brilhante, Iraí, Macotaço and US Pinto 111 cultivars were submitted to treatments with or without addition of ozone to the ambient air, in order to evaluate the effects of exposure on photosynthesis, relative electrolyte leakage, foliar abscission and biomass of the seedlings. Exposure to ozone caused significant decreases in the net assimilation of all cultivars except Iraí. It also caused a significant increase in electrolyte leakage from the Pinto cultivar, but only when AOT40 was the highest. It also produced significant anticipation in the time of foliar abscission in the Pinto, Fepagro and Guapo cultivars. The variability observed in the biomass measurements reflected the limitations to perform long-term controlled-environment studies, one of the major challengers yet to be overcome in order to obtain more conclusive data on damages induced on crop species resulting from tropospheric ozone enrichment.

RACIONAL: Muitas dúvidas ainda permanecem no que se refere às ações dos fatores de crescimento e do plasma rico em plaquetas sobre o mecanismo de reparação tissular. Há necessidade de serem esclarecidos pontos controversos ainda existentes. OBJETIVO: Obter o plasma rico em plaquetas em coelhos através de um método simplificado e ao mesmo tempo adequado, introduzindo um modelo experimental que possa ser utilizado em estudos posteriores. MÉTODOS: Foram utilizados 25 coelhas da raça Nova Zelândia e sem doenças prévias. Quinze mL de sangue de cada animal foi coletado, sendo 10 mL submetidos à dupla centrifugação. Para comprovar a efetividade do método proposto realizou-se contagem mecânica do sangue, bem como do produto final. RESULTADO: Obteve-se uma concentração média de plaquetas no plasma rico em plaquetas 687% maior que a contagem inicial observada no sangue venoso periférico. Para as variáveis: contagem inicial de plaquetas, contagem de plaquetas no plasma rico em plaquetas e enriquecimento, foram obtidos os limites de 95% de confiança para suas médias, sendo que, no que se refere ao percentual de enriquecimento, existe 95% de chance de que o intervalo de (530-844) contenha a média real de enriquecimento de plaquetas. CONCLUSÃO: O método simplificado utilizado permite a obtenção de plasma rico em plaquetas adequado permitindo seu uso em estudos dos fatores de crescimento nos mecanismos de reparação tecidual.

BACKGROUND: Multiple uncertainties still exist about the action of the growth factors and the platelet-rich plasma on the mechanism of repair. AIM: To obtain the platelet-rich plasma in rabbits through a simplified and suitable method, creating an experimental model. METHODS: Twenty-five female New Zealand rabbits without previous diseases were used. Fifteen mL of blood of each rabbit was collected and 10 mL of the collected blood were twice centrifugated. To check the effectiveness of the proposed method mechanical counting of the blood and of the final product were performed. RESULTS: The mean platelet concentration was 687% higher than the initial peripheral blood counting. Mean initial platelet counting, platelet-rich plasma platelet counting and enriched were obtained with 95% CI and in terms of enrichment percentage there is a chance that the interval 530 to 844 be the real mean platelet enrichment. CONCLUSION: This simplified method permits to get an effective platelet-rich plasma to be used in trials about growth factors in mechanisms of tissue repair.

O objetivo deste trabalho foi analisar a performance agronômica e a estabilidade cromossômica de progênies transgênicas homozigotas de soja [Glycine max (L.) Merrill.], e confirmar a resistência dessas plantas a Anticarsia gemmatalis. Onze progênies com expressão dos genes cry1Ac, hpt e gusA foram avaliadas quanto às características agronômicas, em relação à cultivar parental IAS 5 não transformada. Análises citogenéticas foram realizadas em plantas transgênicas e não transgênicas. Duas das 11 progênies transgênicas também foram avaliadas quanto à resistência a A. gemmatalis, em bioensaios in vitro e in vivo. Dois controles negativos foram utilizados nos bioensaios: uma linha transgênica homozigota, com apenas o gene repórter gusA, e plantas de 'IAS 5' não transgênicas. A presença do transgene cry1Ac não afetou nem o desenvolvimento nem a produtividade das plantas. As plantas transgênicas apresentaram cariótipo normal. No bioensaio com folhas destacadas, as plantas cry1Ac exibiram completa eficácia contra A. gemmatalis, enquanto os controles negativos foram significativamente danificados. Ensaios com plantas inteiras confirmaram a elevada proteção do cry1Ac contra a lagarta-da-soja, enquanto as plantas 'IAS 5' não transgênicas e a linha homozigota gusA exibiram 56.5 e 71.5% de desfolhação, respectivamente. A presença do transgene cry1Ac não afeta a performance agronômica da soja (incluindo a produtividade) e confere elevada proteção a A. gemmatalis.

The objective of this work was to analyze the agronomic performance and chromosomal stability of transgenic homozygous progenies of soybean [Glycine max (L.) Merrill.], and to confirm the resistance of these plants against Anticarsia gemmatalis. Eleven progenies expressing cry1Ac, hpt and gusA genes were evaluated for agronomic characteristics in relation to the nontransformed parent IAS 5 cultivar. Cytogenetical analysis was carried out on transgenic and nontransgenic plants. Two out of the 11 transgenic progenies were also evaluated, in vitro and in vivo, for resistance to A. gemmatalis. Two negative controls were used in resistance bioassays: a transgenic homozygous line, containing only the gusA reporter gene, and nontransgenic 'IAS 5' plants. The presence of cry1Ac transgene affected neither the development nor the yield of plants. Cytogenetical analysis showed that transgenic plants presented normal karyotype. In detached-leaf bioassay, cry1Ac plants exhibited complete efficacy against A. gemmatalis, whereas negative controls were significantly damaged. Whole-plant feeding assay confirmed a very high protection of cry1Ac against velvetbean caterpillar, while nontransgenic 'IAS 5' plants and homozygous gusA line exhibited 56.5 and 71.5% defoliation, respectively. The presence of cry1Ac transgene doesn't affect the majority of agronomic traits (including yield) of soybean and grants high protection against A. gemmatalis.

O objetivo deste trabalho foi estimar a adaptabilidade e estabilidade de genótipos de trigo irrigado, nas condições do Cerrado do Brasil Central. O rendimento de grãos de cinco cultivares e nove linhagens de trigo irrigado foram aferidos no ensaio de valor de cultivo e uso. Os ensaios foram conduzidos nos anos de 2005 e 2006, em seis locais em Minas Gerais, três em Goiás e um no Distrito Federal. Os dados de rendimento de grãos foram submetidos à análise de variância individual e à análise de variância conjunta, para a estimativa dos parâmetros de adaptabilidade e estabilidade. Verificaram-se diferenças significativas, entre os genótipos, em 14 dos 16 ambientes avaliados. Nenhum dos genótipos avaliados apresentou o comportamento ideal preconizado pelo método de Cruz e colaboradores. Destacaram-se a cultivar BRS 264 e a linhagem CPAC 02167, pela superioridade em todas as condições do Cerrado do Brasil Central (adaptabilidade geral). Entre os demais genótipos avaliados, destacaram-se as cultivares BRS 207 e EMBRAPA 22, responsivas à melhoria das condições ambientais, e as cultivares BRS 254 e EMBRAPA 42, pela adaptabilidade específica a ambientes desfavoráveis ao cultivo de trigo irrigado.

The aim of this work was to estimate the adaptability and stability of irrigated wheat genotypes under conditions of Central Brazil savannah area. Grain yield of five cultivars and nine lines of irrigated wheat was measured in assays of cultivation value and use. The assays were conducted during the years 2005 and 2006, in six locations in Minas Gerais State, three in Goiás State and one in Federal District. The data of grain yield were submitted to individual and joint ANOVA, in order to estimate the adaptability and stability parameters. Significant differences were detected between genotypes in 14 of the 16 locations evaluated. None of the evaluated genotypes have presented the behavior predicted by the method of Cruz and collaborators. Cultivar BRS 264 and the CPAC 02167 lineage were superior in all the tested conditions in the Central Brazil savannah (general adaptability). Among the other evaluated genotypes, it can be highlighted the responsiveness of the cultivars BRS 207 and EMBRAPA 22 to the improvement of environmental conditions, and the specific fitness of BRS 254 and EMBRAPA 42 cultivars to environments unfavorable to irrigated wheat.
