RESUMO: A bactéria Xylella fastidiosa, agente causal da clorose variegada dos citros, é dependente da ação de insetos vetores para sua disseminação e infecção em plantas cítricas hospedeiras. Os insetos cigarrinhas (Hemiptera: Cicadellidae) transmitem a bactéria para plantas sadias depois de se alimentarem das plantas contaminadas. O objetivo deste trabalho foi desenvolver e comparar métodos de extração de DNA genômico de cigarrinhas a fim de detectar a bactéria X. fastidiosa. A extração de DNA foi realizada de acordo com protocolo à base de fenol-clorofórmio em conjunto com dois kits comerciais: Dneasy® Plant Mini Kit and Blood & Tissue Dneasy® Handbook (Qiagen Inc., Valencia, CA, USA). Utilizou-se a cabeça das seguintes espécies de cigarrinhas: D. costalimai, A. citrina, O. facialis, B. xanthophis, M. leucomelas e H. ignorata. Com base nas diferenças numéricas entre as amostras independentes, os resultados mostraram o uso do método de extração fenol-clorofórmio (36/50 positivos) e o kit comercial DNeasy® Plant Mini Kit (33/50) resultando nas maiores detecções de X. fastidiosa nas amostras de cigarrinhas. Como esses dois métodos detectaram a presença de X. fastidiosa no maior número de amostras infectadas, eles podem ser mais eficientes para o uso na detecção em cigarrinhas.

ABSTRACT: The bacterium Xylella fastidiosa, the causal agent of citrus variegated chlorosis (CVC), is dependent on vector insects for its spread and infection of citrus hosts. The insects, leafhoppers (Hemiptera: Cicadellidae) transmit the bacteria to healthy plants after feeding on infected plants. The objective of this study was to develop and compare methods for extracting genomic DNA from sharpshooters to detect X. fastidiosa. The DNA extraction from insects was performed according to a phenol-chloroform based DNA extraction in conjunction with two commercial kits, Dneasy® Plant Mini Kit and blood & tissue Dneasy® Handbook (Qiagen Inc., Valencia, CA, USA). The heads of the following species of sharpshooter were used: Dilobopterus costalimai, Acrogonia citrina, Oncometopia facialis, Bucephalogonia xanthophis, Macugonalia leucomelas and Homalodisca ignorata. Based on the numeric differences between independent samples, the results showed the use of the phenol:chloroform extraction method (36/50 positives) and the DNeasy® Plant Mini Kit (33/50) resulting in the most detections of X. fastidiosa from leafhopper samples. As these two methods detected Xylella in the greatest number of infected samples, they may be more efficient to use for detection purposes in leafhoppers.

RESUMO. O cancro cítrico, causado pela bactéria Xanthomonas citri subsp. citri é uma das doenças mais importantes da citricultura. A utilização de genótipos resistentes à doença assume um papel importante no manejo e controle do patógeno, sendo essa uma medida viável ao produtor e sustentável ao ambiente. O conjunto de dados utilizado neste trabalho consistiu das observações obtidas de um experimento em que foram empregados como material vegetal, nove genótipos de Laranja doce, variedade Pera enxertado em quatro diferentes porta-enxertos. Este experimento teve inicio em 2010 e foram realizadas avaliações trimestrais para determinar a incidência de cancro nas folhas das plantas. Para a análise dos observações resultantes desse experimento foi utilizado a regressão Beta inflacionada de zero (RBIZ), que é a metodologia adequada para descrever proporções com grandes quantidades de zeros. A partir da análise residual, pode-se perceber que os dados se apresentaram de maneira homogênea indicando um bom ajuste do modelo. Para o componente discreto a variável explicativa, porta enxerto, foi significativa para o não aparecimento da doença, em contraste com o componente contínuo, em que a variável genótipo mostrou-se significativa para explicar a incidência de cancro cítrico.

ABSTRACT. Citrus canker, caused by the bacterium Xanthomonas citri subsp. citri, is one of the most important diseases of citrus. The use of resistant genotypes plays an important role in the management and control of the disease and is the most environmentally sustainable approach to disease control. Citrus canker incidence was recorded in an experiment on nine genotypes of the sweet orange variety 'Pera' grafted on four rootstocks. The experiment was started in 2010 and the incidence of citrus canker on the leaves was recorded on a quarterly basis. The incidence data from the experiment were analyzed using a zero-inflated Beta regression model (RBIZ), which is the appropriate method to describe data with large numbers of zeros. Based on the residual analysis, the data fit the model well. The discrete component of the explanatory variable, rootstock, was not significant as a factor affecting the onset of disease, in contrast with the continuous component, genotype, which was significant in explaining the incidence of citrus canker.

RESUMO. Em diversas áreas do conhecimento, dados com excesso de zeros são encontrados com frequência. Para a análise de tais dados, é recomendado utilizar modelos que permitam uma contagem deste excesso de zero de forma adequada. Neste artigo, o modelo binomial negativo inflacionado de zero (ZINB) foi utilizado para descrever o número de plantas doentes, acometidas por Huanglongbing, em pomares comercias de laranjeiras na região noroeste do Estado do Paraná. Entretanto, deve-se levar em consideração que cada pomar foi avaliado ao longo do tempo, sendo assim, neste contexto, o modelo ZINB com efeito aleatório em ambas as funções de ligação, logarítmica e logística, apresentou melhor ajuste aos dados, pois a introdução destes efeitos consideraram as variações entre os pomares e a dependência entre número de plantas doentes. A partir deste modelo é possível perceber que as plantas mais velhas tendem a apresentar menor probabilidade de adquirir a doença. Todavia, a aplicação de inseticidas e o manejo por calendário apresentam três vezes mais chance de apresentar a doença do que o manejo somente pela presença do vetor.

ABSTRACT. Zero-inflated data from field experiments can be problematic, as these data require the use of specific statistical models during the analysis process. This study utilized the zero-inflated negative binomial (ZINB) model with the log- and logistic-link functions to describe the incidence of plants with Huanglongbing (HLB, caused by Candidatus liberibacter spp.) in commercial citrus orchards in the Northwestern Parana State, Brazil. Each orchard was evaluated at different times. The ZINB model with random effects in both link functions provided the best fit, as the inclusion of these effects accounted for variations between orchards and the numbers of diseased plants. The results of this model show that older plants exhibit a lower probability of acquiring HLB. The application of insecticides on a calendar basis or during new foliage flushes resulted in a three times larger probability of developing HLB compared with applying insecticides only when the vector was detected.

RESUMO Genótipos de citros mostram uma gama de resistência à infecção de Xanthomonas citri subsp. citri (Xcc), agente causal do cancro cítrico asiático (ACC). O objetivo deste estudo foi avaliar um método de inoculação em folha destacada para comparar nove genótipos diferentes de citros que apresentam graus de resistência à ACC e, avaliar a infecção, colonização e desenvolvimento de sintomas das variedades utilizando Microscopia Eletrônica de Varredura (MEV). A inoculação de Xcc em folhas foi realizada através de ferimentos produzidos com agulhas. Aos 7, 10 e 13 dias após a inoculação (DAI), diâmetros de lesão foram mensurados. A variedade Pêra IAC foi a mais resistente ao patógeno e apresentou lesões com menor diâmetro e a variedade Washington foi a mais suscetível e apresentou as maiores lesões, indicando baixa resistência à Xcc. As imagens de MEV foram feitas no desenvolvimento da lesão, e secções foram cortadas a partir das lesões da doença nas variedades testadas. A variedade mais resistente, Pêra IAC, teve aberturas estomáticas menores e produziram uma maior quantidade de exsudados foliares em comparação com a variedade Washington. Nossa hipótese é que os exsudados podem conter compostos tóxicos de Xcc e, portanto, pode ser um mecanismo de defesa para a infecção por Xcc, embora novas pesquisas são necessárias para estabelecer quaisquer efeitos antibióticos de exsudato contra o cancro cítrico.

ABSTRACT Citrus genotypes show a broad range of resistance to infection with Xanthomonas citri subsp citri (Xcc), the causal agent of Asiatic citrus canker (ACC). The aim of this study was to evaluate a detached leaf inoculation method to compare nine different citrus genotypes that exhibit varied resistance levels to ACC, and to evaluate infection, colonization and symptom development of the varieties by using Scanning Electron Microscopy (SEM). Inoculation of Xcc in the leaves was carried out by producing injuries with needles. Lesion diameters were measured at 7, 10 and 13 days after inoculation (DAI). The variety Pera IAC was most resistant to the pathogen and had lesions with smaller diameters, while the variety Washington was most susceptible and had the largest lesions, indicating low resistance to Xcc. SEM images were taken from the lesion development, and sections were obtained from the disease lesions in the tested varieties. The most resistant variety, Pera IAC, had smaller stomatal openings and produced greater quantity of foliar exudates, compared with the variety Washington. We hypothesize that the exudates may contain Xcc toxic compounds and thus may be a defense mechanism against infection by Xcc, but further research is required to establish any antibiotic effects of the exudates against citrus canker.

A limpeza clonal seguida de pré-imunização com complexos protetivos do Citrus tristeza virus(CTV) possibilitaram o cultivo comercial da laranja Pêra, variedade de relevante importância para a citricultura brasileira, mas sensível ao vírus. O emprego de isolados fracos protetivos em outras variedades de citros, mesmo com maior tolerância ao CTV, pode ser também de interesse para evitar a dispersão de isolados severos. O objetivo desta pesquisa foi caracterizar, por meio da análise SSCP (Single Strand Conformational Polymorphism) do gene da proteína do capsídeo, isolados de CTV presentes em plantas das cultivares de laranjas doce Pêra, Hamlin e Valência, propagadas de quatro fontes de borbulhas: 1) clones velhos, 2) clones nucelares, 3) clones microenxertados e 4) clones microenxertados pré-imunizados com o isolado fraco protetivo 'PIAC' do CTV. Avaliou-se ainda a correlação dos padrões SSCP obtidos com intensidade de caneluras no lenho, vigor e produção de frutos. Resultados do SSCP demonstraram baixa diversidade genética entre isolados presentes em diferentes plantas da mesma variedade e mesma fonte de borbulhas, e em alguns casos, em diferentes fontes de borbulhas e variedades. Para os sintomas de tristeza, observou-se menores intensidades em plantas dos clones novos, microenxertados e, dos microenxertados pré-imunizados sobre clones velhos das três variedades. Os padrões SSCP e a sintomatologia observados sugeriram que complexos mais fortes do CTV estão infectando as plantas dos clones velhos das três variedades. A estabilidade do complexo protetivo foi observada nos padrões SSCP dos isolados de CTV de alguns dos clones microenxertados e pré-imunizados. Concluiu-se que as alterações detectadas em outros perfis eletroforéticos deste tratamento não provocaram a perda da capacidade protetiva do isolado 'PIAC' de CTV, inoculado na pré-imunização.

Clonal cleaning, followed by pre-immunization with protective complexes of Citrus tristeza virus(CTV), allowed the commercial cultivation of Pêra sweet orange, a variety that has great importance for Brazilian citriculture but is sensitive to the virus. The use of mild protective isolates in other citrus varieties, even those more tolerant to CTV, can also be of interest to prevent the spread of severe isolates. The aim of this study was to characterize, by means of SSCP (Single Strand Conformational Polymorphism) analysis of the coat protein gene, CTV isolates present in plants of the sweet orange cultivars Pêra, Hamlin and Valencia propagated from four budwood sources: 1) old lines, 2) nucellar lines, 3) shoot-tip-grafted lines, and 4) shoot-tip-grafted lines pre-immunized with the mild CTV protective isolate 'PIAC'. We also evaluated the correlation of the obtained SSCP patterns to stem pitting intensity, tree vigor and fruit yield. SSCP results showed low genetic diversity among the isolates present in different trees of the same variety and same budwood source and, in some cases, in different budwood sources and varieties. Considering tristeza symptoms, lower intensity was noted for plants of new, shoot-tip-grafted and pre-immunized shoot-tip-grafted lines, compared to old lines of the three varieties. The observed SSCP patterns and symptomatology suggested that more severe CTV complexes infect the plants of old lines of all three varieties. The protective complex stability was observed in the SSCP patterns of CTV isolates of some shoot-tip-grafted and pre-immunized clones. It was concluded that the changes detected in other electrophoretic profiles of this treatment did not cause loss of the protective capacity of CTV isolate 'PIAC' inoculated in the pre-immunization.

The citrus blackfly Aleroucanthus woglumi Ashby, (Hemiptera Aleyrodidae) is an important pest that occurs in citrus groves, native to south-east Asia. In Brazil, according to the Ministry of Agriculture, this is a quarantine pest (A2) under official control IN 52, 2007 (MAPA) and is not widespread in the country. The insect can infest more than 300 host plants, including cultivated plants, ornamentals and weeds, but mostly occurs in the plants of the genus citrus. This paper is the first report of citrus blackfly in the State of Paraná.

A gomose dos citros é considerada uma doença de grande importância para a citricultura no Brasil e em nível mundial. A etiologia desta doença compreende um complexo de espécies de Phytophthora. Embora importante, pouco se conhece sobre a gomose nas regiões produtoras de citros no Estado do Paraná. Por isso, este trabalho teve como objetivo identificar espécies de Phytophthora associadas à gomose em pomares de citros no Paraná. Nas regiões Norte, Noroeste e Vale do Ribeira foram retiradas amostras de raízes de plantas com sintomas de gomose e também de solo da rizosfera. Em laboratório, empregando pêra cv. D'anjou como isca e meio de cultivo batata-dextrose-ágar, foram obtidos 21 isolados de Phytophthora spp. Todos os isolados infectaram mudas de limão 'Cravo', reproduzindo os sintomas de gomose e também apresentaram crescimento micelial a 8º C e a 36º C, com exceção do isolado PR20 para 36º C . "In vitro", esses isolados foram heterotálicos, sendo 20 compatíveis ao tipo padrão A2 e um compatível ao tipo padrão A1. Vinte isolados formaram esporângios persistentes e papilados, com 25,5 - 62,0 µm de comprimento (C) e 27,9 - 49,6 µm de largura (L) e a relação C/L foi de 1,38:1. Um isolado (PR20) apresentou esporângios medindo 40,3 - 55,8 µm de comprimento e 27,9 - 37,2 µm de largura, formando esporângios persistentes, papilados ou bipapilados e de formas distorcidas, não formando clamidósporos. A temperatura ótima para crescimento desse isolado foi entre 20 a 28º C, enquanto para os demais foi de 24 a 32º C, tendo estes produção abundante de clamidósporos globosos de diâmetro variando entre 21,7 a 43,4 µm. De acordo com as características morfofisiológicas apresentadas, dos 21 isolados analisados, 20 pertenceram à espécie P. nicotianae e um à espécie P. citrophthora. A análise de sequências de genes da região ITS1-5.8S-ITS2 do rDNA e usando o teste de "Single-Strand Conformation Polymorphism" (SSCP), confirmou P. nicotianae e P. citrophthora como as duas espécies de Phytophthora associadas à gomose em pomares de citros no estado do Paraná.

Citrus root rot is considered a disease of great importance to citriculture in Brazil and around the world. The etiology of this disease comprises a complex of Phytophthora species. Although citrus root rot is important, little is known about it in the producing regions of Paraná State, Brazil. Therefore, this study aimed to identify Phytophthora species associated with root rot in citrus orchards in Paraná. In the North and Northwest regions of this state and in Vale do Ribeira, samples were collected from the roots of plants showing symptoms of citrus root rot, as well as from the rhizosphere soil. In the laboratory, employing pear cv. D'anjou as bait and the culture medium potato-dextrose-agar, 21 isolates of Phytophthora spp. were obtained. All isolates infected seedlings of Rangpur lime, reproducing the symptoms of root rot and presenting mycelial growth at 8 ºC and 36 ºC, except for the isolate PR20 at 36 ºC. In vitro, these isolates were heterothallic, and 20 of them were compatible with the standard type A2, while one of them was compatible with the standard type A1. Twenty isolates formed papillate persistent sporangia of 25.5 - 62.0 µm length (C) and 27.9 - 49.6 µm width (L); the C/L ratio was 1.38:1. One isolate (PR20) had sporangia of 40.3 - 55.8 µm length and 27.9 - 37.2 width, forming papillale or bipapillate persistent sporangia of distorted forms and not forming chlamydospores. The optimum temperature for the growth of this isolate was between 20 and 28ºC, while for the remaining isolates it was from 24 to 32ºC; the latter had abundant production of globose chlamydospores with diameter ranging from 21.7 to 43.4 µm. According to the presented morphophysiological characteristics, of the 21 analyzed isolates, 20 belonged to the species P. nicotianae and one to the species P. citrophthora. Sequence analysis of genes of the rDNA ITS1-5.8S-ITS2 region, using the test Single-Strand Conformation Polymorphism (SSCP), confirmed P. nicotianae and P. citrophthora as the two Phytophthora species associated with root rot in citrus orchards in the state of Paraná.

A identificação de cultivares com determinados níveis de resistência ao cancro cítrico, com características agronômicas e comerciais desejáveis, assume grande importância para o manejo desta doença. A avaliação de genótipos promissores pode ser fetuada em condições controladas ou em campo. O trabalho teve como objetivo avaliar genótipos de laranja doce (Citrus sinensis), tangerinas (Citrus reticulata) e híbridos quanto à resistência ao cancro cítrico, em diferentes épocas de avaliação. O experimento foi conduzido em pomar experimental no município de Maringá, Paraná. Para a determinação da severidade média da doença, foram utilizadas escalas diagramáticas, sendo avaliadas quatro plantas por bloco e amostrados ramos na porção mediana de cada planta. Além disso, foi calculada a Área Abaixo da Curva de Progresso da Severidade (AACPS). De acordo com os dados obtidos, ocorreram diferenças significativas na severidade e na AACPS, ao longo das avaliações, sendo que a severidade variou de 0,25% a 4,88% e a AACPS de 49,80 a 742,65. As variedades cítricas Tang. Ponkan, Rosehaugh Nartjee, Harris, De wildt, Dekopon, Lee, África do Sul, Tarocco#23, Solid Scarlet, Satsuma Okitsu SPA 29, Cristalina, Shamouti e Pêra Olimpia se destacaram e apresentaram as menores médias de severidade, sendo essas classificadas como resistentes. Os resultados sugerem que esses genótipos podem ser importantes ferramentas nos programas de melhoramento de genótipos de citros, visando resistência a Xanthomonas citri subsp. citri.

Identification of cultivars with certain levels of resistance to citrus canker and desirable agronomic and commercial features is of great importance for the management of this disease. Evaluation of promising genotypes can be performed under controlled conditions or in the field. This study aimed to evaluate genotypes of sweet orange (Citrus sinensis), mandarin (Citrus reticulata) and hybrids for resistance to citrus canker at different times. The experiment was conducted in an experimental orchard in Maringá, Paraná State. To determine the mean severity of the disease, diagrammatic scales were used to evaluate four plants per block, and branches in the middle portion of each plant were sampled. In addition, we calculated the Area Under the Disease Progress Curve (AUDPC). According to the obtained data, there were significant differences in the severity and AUDPC along the evaluations, and the severity varied from 0.25% to 4.88%, while AUDPC varied from 49.80 to 742.65. The citrus varieties Tang. Ponkan, Rosehaugh Nartjee, Harris, De wildt, Dekopon, Lee, South Africa, Tarocco#23, Solid Scarlet, Satsuma Okitsu SPA 29, "Cristalina", Shamouti and "Pêra Olimpia" had the lowest severity levels and were classified as resistant. Results suggest that these genotypes can be important tools in breeding programs of citrus genotypes for resistance to Xanthomonas citri subsp. citri.

The use of tolerant rootstocks and preimmunization has satisfactorily controlled losses associated with the Citrus tristeza virus (CTV). Several researchers have shown that CTV mild isolates that are selected in the same region where they are used are superior to isolates obtained from other areas. Thus, budwoods of 20 outstanding citrus trees were collected in north and northwestern Paraná state (Brazil) citrus-producing areas and established to be used in a preimmunization program. These budwoods were tested to evaluate the potential protection of the inherently present viral complex. Based on biological indexing and molecular characterization of the capsid protein gene by RFLP (restriction fragment length polymorphism, which indicated that the plants were infected with mild isolates of CTV), some of the selected plants could be used in a preimmunization program. These potentially mild and protective isolates were challenged with severe 'Rolândia' isolate inoculations by grafting and by the brown citrus aphid (Toxoptera citricida Kirkaldy) vector, which was faster in transmitting the virus. Some isolates had a better protective value than others, particularly when challenged with the severe CTV isolate. The SSCP (single strand conformational polymorphism) molecular analysis was an excellent complementary tool for monitoring the performance of the experiments and the stability of the viral complex present in the plants. Isolate number 1, collected in the municipality of Cruzeiro do Sul (CS-1), was the most promising for protecting commercial Pêra sweet orange (C. sinensis L.) orchards in northern and northwestern Paraná. The Rolândia severe CTV isolate was stable and had a high genetic divergence among the severe isolates used as a control (Capão Bonito and Barão B) and all of the isolates tested.

Plants of Pera sweet orange on Rangpur lime rootstocks, from orchards of the northwest and north of Paraná state, Brazil, were evaluated for severity of symptoms and genetic diversity of Citrus tristeza virus. The severity of symptoms was evaluated by the development of tree, fruit size and stem pitting symptoms. Isolates that infect these plants were compared with known mild and severe isolates by analysis of restriction fragments length polymorphism (RFLP) of the coat protein nucleotide sequences (CPNS), amplified by the polymerase chain reaction (PCR) and undergone digestion with the restriction enzymes Hinf I and Rsa I. The severity of symptoms showed that the analyzed plants from the northwest orchards presented mild to moderate tristeza symptoms, while the plants from the north orchards exhibited moderate to severe symptoms. The RFLP analysis revealed that the CTV isolates are constituted by haplotype mixtures. Rsa I was the enzyme that best discriminated the genetic diversity among the analyzed isolates of CTV. Two main groups were generated by the UPGMA analysis. The isolates from the northwest orchards grouped with most of the mild isolates used as control, and a great part of the isolates from the north orchards, was correlated with the severe isolate Capão Bonito. Correlation between the stem pitting intensity and RFLP patterns, was demonstrated with some exceptions. The failure of protection of some isolates and the contamination of the rootstocks by the severe isolates, in field nursery conditions, before grafting with scions with mild isolates, were the hypotheses considered to explain the occurrence of severe CTV isolates in the North area of Paraná State.

A Clorose variegada dos citros (CVC) é uma doença causada pela bactéria de xilema Xylella fastidiosa Wells. A disseminação ocorre por meio de insetos vetores pertencente à ordem Hemiptera, família Cicadellidae (subfamília Cicadellinae), os quais transmitem a bactéria depois de se alimentarem em plantas contaminadas. Neste trabalho, objetivou-se identificar e monitorar as espécies de cigarrinhas vetoras em um pomar comercial no município de Paranavaí, Paraná. O experimento foi realizado em um talhão comercial de laranja doce (Citrus sinensis (L.) Osbeck), variedade Pera, com 1.000 plantas de dez anos de idade. A amostragem foi realizada por meio de armadilhas adesivas amarelas, distribuídas na área periférica e central do pomar, com duas repetições por rua amostrada. As etiquetas foram distribuídas entre a 5ª e 30ª plantas em 10 ruas, totalizando 20 armadilhas que foram renovadas no pomar, a cada trinta dias, durante o período de avaliação que foi entre junho de 2005 e setembro de 2006. As principais espécies de Cicadellinae capturadas foram Acrogonia citrina Marucci & Cavichioli, Dilobopterus costalimai Young e Macugonalia cavifrons Stal. Essas espécies apresentaram ocorrência constante e frequência de 3,97%, 4,2%, 13,0% respectivamente, em relação ao total de cigarrinhas coletadas.

Citrus variegated chlorosis (CVC) is the disease caused by the bacteria Xylella fastidiosa Wells. Dissemination occurs through insect vectors belonging to the order Hemiptera, family Cicadellidae (subfamily Cicadellinae), which transmit the bacteria after feeding on infected plants. The objective of this study was to identify the species of insect vectors in an orchard in the municipality of Paranavaí, in the State of Paraná. The experiment was conducted in a commercial stand of sweet orange (Citrus sinensis (L.) Osbeck), Pêra variety with 1,000 10-year-old plants. Monitoring was performed using yellow sticky traps, distributed in the central and peripheral areas of the orchard with two traps on each one. The stickers were distributed between the 5th and 30th plant in each 10 rows, totaling 20 traps in the orchard and renewed every thirty days during the evaluation period, which was between June 2005 and September 2006. The main species caught were Cicadellinae Acrogonia citrina Marucci & Cavichioli Dilobopterus costalimai Young and Macugonalia cavifrons Stal. These species occurred constantly, with frequencies of 3.97%, 4.2%, and 13.0% respectively, compared to the total leafhoppers collected.

A citricultura é afetada por diversas doenças, dentre as quais a Clorose Variegada dos Citros (CVC) causada pela bactéria Xylella fastidiosa (Wells). Para a região noroeste do Estado do Paraná, onde foi avaliada a CVC, propõe-se encontrar modelos não-lineares de curvas de progresso de incidência da CVC que representem o percentual de plantas acometidas pela doença. Para avaliar o comprometimento dos pomares com relação à doença, foram escolhidos pomares comerciais em dois municípios, onde foi determinada a proporção de plantas doentes. Foram selecionados talhões de laranja doce (Citrus sinensis Osbeck) nas variedades "Pêra", "Valência", "Natal" e "Folha Murcha" e a avaliação de todas as plantas do talhão foi realizada visualmente em relação à presença ou à ausência de sintomas de CVC. Para estimar o modelo que melhor se ajustou aos dados de progresso da proporção da doença em cada talhão, foram considerados modelos não-lineares de crescimento sigmoidal, o logístico e o de Gompertz, conforme sugerido na literatura. A escolha do melhor modelo foi com base em medidas de curvatura intrínseca e paramétrica de Bates e Watts, viés de Box e medida de assimetria de Hougaard. O modelo que melhor ajustou aos dados de Loanda foi o modelo de Fermi-Dirac e aos dados de Nova Esperança foi outra reparametrização do Modelo Logístico, denominado de L5.

In Brazil, the production of sweet oranges has been threatened by the Citrus Variegated Chlorosis (CVC) incited by the gram-negative bacterium Xylella fastidiosa (Wells). Commercial citrus groves in two counties at the Northwestern Paraná state were evaluated to estimate the disease progression by using parameterizations of nonlinear models. Groves of Citrus sinensis Osbeck, variety "Pêra", "Valência", "Natal" and "Folha Murcha" had all the plants evaluated for the presence of disease symptoms. Thereafter, different parameterizations of the Logistic and Gompertz models were fitted to these data. The goodness of fit was evaluated by the intrinsic (IN) and parameter-effects (PE) curvatures of Bates and Watts, the bias of Box and the Hougaard measures of skewness. In Loanda, the best model was the Fermi-Dirac, and in Nova Esperança the data were best fitted to the parameterization L5, which is also a parameterization from the Logistic model.

O objetivo deste trabalho foi monitorar a manutenção da estabilidade de isolados protetores contra Citrus tristeza virus (CTV) em clones selecionados de laranja 'Pêra' (Citrus sinensis) pré-imunizados ou infectados naturalmente pelo vírus, após sucessivas propagações clonais. O trabalho foi realizado em condições de campo, no norte do Estado do Paraná. A análise do gene da capa protéica (GPC) de 33 isolados, coletados de 16 clones de laranjeira 'Pêra', foi realizada com o uso da técnica polimorfismo conformacional da fita simples (SSCP). Inicialmente, os isolados foram caracterizados por meio de sintomas de caneluras observados nos clones. Em seguida, o genoma viral foi extraído e utilizado como molde para a amplificação do GCP com uso da transcrição reversa da reação em cadeia da polimerase (RTPCR). Os perfis eletroforéticos dos produtos da RTPCR foram analisados com emprego do coeficiente de Jaccard e do método UPGMA. A maioria dos clones apresentou sintomas fracos a moderados de caneluras, bem como alterações nos perfis dos isolados de CTV. Contudo, a estabilidade dos complexos protetores foi mantida, com exceção dos isolados presentes em dois dos clones analisados. Foi observada baixa variabilidade genética nos isolados durante os anos avaliados.

The objective of this work was to monitor the maintenance of Citrus tristeza virus (CTV) protective isolates stability in selected clones of 'Pêra' sweet orange (Citrus sinensis), preimmunized or naturally infected by the virus, after successive clonal propagations. The work was carried out in field conditions in the north of Paraná State, Brazil. Coat protein gene (CPG) analysis of 33 isolates collected from 16 clones of 'Pêra' sweet orange was performed using single strand conformational polymorphism (SSCP). Initially, the isolates were characterized by symptoms of stem pitting observed in clones. Then viral genome was extracted and used as template for the amplification of CPG by reverse transcription polimerase chain reaction (RTPCR). RTPCR products electrophoretic profiles were analyzed using the Jaccard coefficient and the UPGMA method. The majority of the clones had weak to moderate stem pitting symptoms and its CTV isolates showed alterations in the SSCP profiles. However, the stability of the protective complex has been maintained, except for isolates from two analised clones. Low genetic variability was observed within the isolates during the studied years.

A Clorose variegada dos citros (CVC) é uma importante doença que ocorre nos citros, cujo agente causal é a bactéria Xylella fastidiosa, Wells. A bactéria depende, obrigatoriamente, de insetos vetores para sua disseminação, que são as cigarrinhas sugadoras do xilema (Hemiptera: Cicadellidae, Cicadellinae). No presente estudo objetivou-se avaliar a flutuação populacional de espécies de cigarrinhas nas diferentes variedades de laranja doce [Citrus sinensis (L.) Osbeck], Natal, Pêra, Valência e Folha Murcha, em um pomar comercial localizado na região Noroeste do Paraná, no período de janeiro de 2000 a dezembro de 2002. Amostragens quinzenais foram realizadas com o uso de armadilhas adesivas amarelas, num total de 24 armadilhas em cada avaliação. Após a identificação das espécies observou-se, que as mais representativas foram Dilobopterus costalimai da tribo Cicadellini e Acrogonia citrina da tribo Proconiini, sendo que a variedade de laranja Pêra apresentou o maior número de espécies vetoras durante os anos avaliados.

The aim of the present study was to assess the population flutuation of the sharpshooters species subfamily Cicadellinae belonging to the tribes Cicadellini and Proconiini, in sweet orange [Citrus sinensis( L). Osbeck] commercial orchards of the northwest region of Paraná State , Brazil. Samplings were carried out the employing every time 24 yellow sticky cards. Identification of the species showed that the most representative were Dilobopterus costalimai of the Cicadellini tribe and Acrogonia citrina of the Proconiini tribe.

O experimento teve como objetivo caracterizar biológica e molecularmente os isolados de CTV, obtidos de árvores de laranja Pêra em pomares das regiões Norte e Noroeste do Paraná, visando selecionar isolados fracos do Citrus tristeza virus com capacidade protetiva para pré-imunização. Os resultados apontaram sintomas fracos a moderados de caneluras e revelaram similaridade genética da maioria dos isolados analisados e o isolado fraco controle, sugerindo que as plantas selecionadas estão infectadas por haplótipos fracos de CTV, com potencial para serem utilizadas como plantas matrizes.

A cross-protection program citrus tristeza disease, caused by Citrus tristeza virus (CTV) has been developed in the Parana State, to obtain Pera orange mother plants. Thus, 20 plants were selected in the orange commercial fields of the North and Northwest region. Comparisons between mild and severe recognized isolates were made. The results showed mild-to-moderate stem pitting symptoms and the great majority of the isolates grouped up with the mild control isolate. These results suggest that the plants selected were infected by CTV mild haplotypes showing that they have mother plant potential.
