Abstract South American opossums (Didelphis spp.) are definitive hosts of Sarcocystis neurona, Sarcocystis speeri, Sarcocystis lindsayi and Sarcocystis falcatula. In Brazil, diverse studies have demonstrated a high frequency of Sarcocystis falcatula-like in sporocysts derived from opossums, and high genetic diversity has been observed in surface antigen-encoding genes (SAGs). In this study, genetic diversity of Sarcocystis spp. derived from Didelphis albiventris and Didelphis aurita from the cities of Campo Grande and São Paulo, was accessed by sequencing SAG2, SAG3, SAG4, the first internal transcribed spacer (ITS-1) and cytochrome c oxidase subunit I (cox1). Molecular identification was performed for 16 DNA samples obtained from sporocyst or culture-derived merozoites. The ITS-1, cox1, and SAG3 fragments were cloned, whereas SAG2 and SAG4 were sequenced directly from PCR products. Four alleles variants were found for SAG2, 13 for SAG3 and seven for SAG4, from which four, 13 and four, respectively, were novel. Twenty-seven allele variants were found for ITS-1, all phylogenetically related to S. falcatula-like previously described in Brazil. Sarcocystis sp. phylogenetically related to Sarcocystis rileyi was evidenced by cox1 in three opossums. Further studies are needed to clarify the role of Didelphis spp. as definitive hosts of Sarcocystis spp. other than that previous described.

Resumo Gambás sul-americanos (Didelphis spp.) são hospedeiros definitivos de Sarcocystis neurona, Sarcocystis speeri, Sarcocystis lindsayi e Sarcocystis falcatula. No Brasil, diversos estudos têm demonstrado alta frequência de Sarcocystis falcatula-like em esporocistos derivados de gambás, com grande diversidade nos genes que codificam antígenos de superfície (SAGs). Neste estudo, a diversidade genética de Sarcocystis spp., oriundos de Didelphis albiventris e Didelphis aurita, dos municípios de Campo Grande e São Paulo, foi acessada por meio do sequenciamento de SAG2, SAG3 e SAG4, da primeira região espaçadora interna transcrita (ITS-1) e citocromo c oxidase subunidade I (cox1). Identificação molecular foi realizada em 16 amostras de DNA, obtidas de esporocistos ou merozoítos derivados de cultivo. Os fragmentos de ITS-1, cox1 e SAG3 foram clonados, enquanto SAG2 e SAG4 foram sequenciados diretamente dos produtos de PCR. Quatro alelos foram observados em SAG2, 13 em SAG3 e sete em SAG4, sendo novos quatro, 13 e quatro, respectivamente. Em ITS-1, 27 alelos foram observados, todos filogeneticamente relacionados à S. falcatula-like, previamente detectados no Brasil. Sarcocystis sp. filogeneticamente relacionado à Sarcocystis rileyi foi evidenciado por cox1 em três gambás. Mais estudos são necessários para entender o papel de Didelphis spp. como hospedeiro definitivo de Sarcocystis spp. diferentes daqueles previamente descritos.

Abstract Cholesterol is a lipid molecule of great biological importance to animal cells. Dysregulation of cholesterol metabolism leads to raised blood total cholesterol levels, a clinical condition called hypercholesterolemia. Evidence has shown that hypercholesterolemia is associated with the development of liver and heart disease. One of the mechanisms underlying heart and liver alterations induced by hypercholesterolemia is oxidative stress. In this regard, in several experimental studies, gold nanoparticles (AuNP) displayed antioxidant properties. We hypothesized that hypercholesterolemia causes redox system imbalance in the liver and cardiac tissues, and AuNP treatment could ameliorate it. Young adult male Swiss mice fed a regular rodent diet or a high cholesterol diet for eight weeks and concomitantly treated with AuNP (2.5 μg/kg) or vehicle by oral gavage. Hypercholesterolemia increased the nitrite concentration and glutathione (GSH) levels and decreased the liver’s superoxide dismutase (SOD) activity. Also, hypercholesterolemia significantly enhanced the reactive oxygen species (ROS) and GSH levels in cardiac tissue. Notably, AuNP promoted the redox system homeostasis, increasing the SOD activity in hepatic tissue and reducing ROS levels in cardiac tissue. Overall, our data showed that hypercholesterolemia triggered oxidative stress in mice’s liver and heart, which was partially prevented by AuNP treatment.

RESUMO Uma nova espécie de Paralithoxus é descrita do rio Ajarani, um pequeno tributário da bacia do rio Branco, Estado de Roraima, Amazônia Brasileira. O gênero Paralithoxus é composto por espécies descritas para as drenagens do Essequibo na Guiana, Approuague e Maroni na Guiana Francesa, rio Suriname no Suriname, e mais recentemente, para os rios Jari e Amapá, no Brasil. Apesar de ocorrer em pequenos riachos com fundo rochoso e correnteza forte como o restante dos representantes de Paralithoxus, esta é a primeira espécie do gênero coletada a 900 m de altitude, na Serra da Mocidade, uma pequena cadeia de montanhas localizada em uma área isolada e de difícil acesso na porção sul do Escudo das Guianas. A nova espécie distingue-se de suas congêneres pela presença de dentes truncados, pelo padrão de coloração com manchas verde claras sobre fundo oliva-marrom escuro, alternando manchas claras e escuras nas nadadeiras e pelo comprimento relativo da nadadeira pélvica igual ou maior que o comprimento da nadadeira peitoral. O dimorfismo sexual da espécie é descrito. Comentários sobre a morfologia e osteologia da espécie são feitos e comparados com suas congêneres.

ABSTRACT A new species of Paralithoxus is described from the Ajarani River, a small tributary of the Branco River basin, Roraima State, Brazilian Amazon. The genus Paralithoxus comprises species described from the Essequibo drainage in Guyana, Approuague and Maroni in French Guiana, Suriname River in Surinam, and more recently, from Jari and Amapá rivers, in Brazil. Despite occurring in a rock-bottomed fast-flowing stream as the other species of Paralithoxus, this is the first species of the genus collected at 900 m altitude, in the Serra da Mocidade highlands, an isolated and poorly accessible small mountain chain at the southern border of the Guiana Shield. The new species is distinguished from its congeners by having truncate teeth, color pattern with green spots on dark olive-brown background, alternating dark and light blotches on fins and by the pelvic fin being as long as or longer than the pectoral fin. Sex dimorphism of the species is described. Comments on morphology and osteology are provided and compared with congeners.

RESUMO: A doença de armazenamento de glicogênio tipo II (DAG-II) e a síndrome miastênica congênita (SMC) são importantes doenças autossômicas recessivas no gado Brahman. O objetivo deste estudo foi investigar a presença das mutações responsáveis pela DAG-II (E7, c.1057_1058delTA; e E13, c.1783C>T) e pela SMC (c.470del20) em bovinos da raça Brahman e em touros Brahman que são rotineiramente utilizados em programas de reprodução no Brasil. Um total de 276 amostras de bulbo piloso de bovinos Brahman (167 fêmeas e 109 machos, com idade variando de 12 a 24 meses) e 35 amostras de sêmen congeladas de touros Brahman (22 touros americanos, 11 touros brasileiros, um touro argentino e um touro australiano) foram usados neste estudo. O DNA genômico foi purificado, das amostras de bulbo piloso e de sêmen, e utilizado na genotipagem por PCR das mutações c.1057_1058delTA (E7) e c.1783C>T (E13) no gene GAA e c.470del20 no gene CHRNE. Os produtos de PCR foram purificados e sequenciados. A frequência genotípica para cada polimorfismo foi estimada separadamente. Dos 276 Brahman testados, 7,3% foram identificados como heterozigotos para E7. Todos os Brahman foram homozigotos wild-type para o alelo E13. A mutação E7 foi identificada em homozigose em 8,6% (3/35) das amostras de sêmen comerciais, enquanto que a mutação E13 não foi identificada. A mutação c.470del20 foi identificada em heterozigose em 0,73% das amostras de bulbo piloso, mas esta mutação não estava presente nas amostras de sêmen avaliadas. Nenhum estudo prévio avaliou a prevalência das mutações responsáveis pela DAG-II ou SMC em bovinos Brahman brasileiro. Em suma, as mutações E7 e c.470del20 estão presentes no rebanho Brahman brasileiro, e medidas de controle devem ser adotadas para prevenir o aumento da incidência da DAG-II e SMC em bovinos da raça Brahman no Brasil.

ABSTRACT: Glycogen storage disease type II (GSD-II) and congenital myasthenic syndrome (CMS) are important autosomal recessive disorders in Brahman cattle. The objective of this study was to investigate the presence of mutations responsible for GSD II (E7, c.1057_1058delTA; and E13, c.1783C>T) and CMS (c.470del20) in purebred Brazilian Brahman cattle and in purebred Brahman bulls that were routinely used in breeding programs in Brazil. A total of 276 purebred Brahman cattle (167 females and 109 males, with ages ranging from 12-24 months) and 35 frozen semen samples taken from purebred Brahman bulls (22 bulls from the USA, 11 Brazilian bulls, one Argentine bull and one Australian bull) were used in this study. Genomic DNA was purified from hair root samples and from semen samples. Purified DNA was used in PCR genotyping to mutations c.1057_1058delTA (E7) and c.1783C>T (E13) in the GAA gene and c.470del20 in the CHRNE gene. The PCR products were purified and sequenced. The genotypic frequencies per polymorphism were estimated separately. Of the 276 Brahman cattle tested, 7.3% were identified as heterozygous for E7. All Brahman cattle studied were homozygous for the wild-type E13 allele. The E7 mutations was identified as heterozygous in 8.6% (3/35) of the commercial semen samples, whereas the E13 mutations was not identified. The c.470del20 mutation was identified as heterozygous in 0.73% of the hair root samples, but this mutation was not present in any semen sample assessed. No study had previously evaluated the prevalence of mutations responsible for GSD II or CMS in Brazilian Brahman cattle. In summary, the E7 and c.470del20 mutations are present in the Brazilian Brahman herd, and control measures should be adopted to prevent an increase in the incidence of GSD-II and CMS in Brahman cattle in Brazil.

RESUMO A farinha do mesocarpo do babaçu tem sido utilizada pelas indústrias farmacêuticas e de alimentação humana e animal. Contudo, há falta de padronização na produção, bem como ausência de informações do manejo sobre a qualidade do produto. Deste modo, objetivou-se, no trabalho, secar o mesocarpo do babaçu em estufa de ventilação forçada de ar e secador solar, ajustar diferentes modelos matemáticos, bem como quantificar os teores de proteínas e fibras brutas da farinha produzida. Foram usados, como critério do ajuste, o coeficiente de determinação, a magnitude do erro médio relativo, o desvio-padrão da estimativa e a tendência de distribuição dos resíduos. A secagem em menor tempo ocorreu em estufa a 60 ºC (370 min) obtendo-se teor de água de 4,62%, enquanto no secador solar foi em 6 dias com teor de água final de 8,07%. O modelo Dois Termos foi o que melhor se ajustou aos dados experimentais para secagem em estufa e o modelo Midilli em secador solar. Houve um aumento do teor de proteína com a secagem em secador solar e estufa a 40, 50 e 60 ºC (1,36; 1,33; 1,15 e 1,37%, respectivamente) em relação ao mesocarpo in natura (0,88%). A secagem em estufa e em secador solar proporcionou incremento de proteína na farinha.

ABSTRACT ‘Babassu’ mesocarp flour has been used by the pharmaceutical, human food and animal feed industries. However, there is lack of standardization in the production, as well as absence of information on the management of the product’s quality. Thus, the objective of this study was to dry the ‘babassu’ mesocarp in forced-air oven and solar dryer, adjust different mathematical models to the experimental data, as well as to quantify the levels of proteins and crude fiber of the produced flour. The criteria for the adjustment were the coefficient of determination, magnitude of the mean relative error, standard deviation of estimate and the residual distribution trend. Drying in the shortest time occurred in oven at 60 °C (370 min), leading to water content of 4.62%, while in the solar dryer the final water content was 8.07% in 6 days. The mathematical model Two Terms showed the best fit to the experimental data for oven drying and the Midilli model showed the best fit in solar dryer. There was an increase in protein content with the drying in solar dryer and oven at 40, 50 and 60 °C (1.36, 1.33, 1.15 and 1.37%, respectively) in relation to fresh mesocarp (0.88%). Drying in both oven and solar dryer promoted increase of protein in the flour.

RESUMO O gênero cisandino Aphanotorulus foi recentemente revisado e consiste de seis espécies válidas: A. ammophilus, A. emarginatus, A. gomesi, A. horridus, A. phrixosoma e A. unicolor. No presente trabalho, uma nova espécie é descrita dos tributários da bacia do rio Aripuanã, nos estados do Amazonas e Mato Grosso, Brasil. A nova espécie é facilmente distinguida de suas congêneres pelo padrão de coloração: nadadeira caudal com o lobo superior quase totalmente hialino e com pintas escuras ao longo dos raios e membranas, e o lobo inferior vermelho e desprovido de pintas; ausência de manchas escuras na série lateral de placas medianas ventrais.

ABSTRACT The cis-Andean genus Aphanotorulus was recently revised and comprises six valid species: A. ammophilus, A. emarginatus, A. gomesi, A. horridus, A. phrixosoma and A. unicolor. Herein, a new species is described from tributaries of the rio Aripuanã basin, in Amazonas and Mato Grosso states, Brazil. The new species is easily distinguished from congeners by its color pattern: caudal fin with upper lobe mostly hyaline with dark spots along rays and membranes, and lower lobe red and without dark spots; and absence of dark spots in the lateral series of mid-ventral plates.

Reduction of five substituted octalones employing lithium tri-sec-butylborohydride (L-selectride®) in THF and ethyl ether led to allylic alcohols with moderate diastereoselectivity. The stereoselectivity of addition of bulky hydrides showed to be different from most examples in the literature and was strongly influenced by substitution on the octalone ring.
