Concrete slurry waste is a material removed from sedimentation tanks that receive water used to wash concrete mixer trucks and patios of concrete batching plants (CBP). The objectives of this study were to determine the chemical composition of this residue from three different producers, in three consecutive months and at different collection points using wavelength dispersive X-ray fluorescence, investigate the inconstancy of content through analysis of variance, compare the qualitative and quantitative results with the literature and discuss the possibility of inserting the material in the proposal of different authors. Twenty-two oxides and elements were identified; among them, five showed their incidence in a punctual way. The analysis of variance showed that there are significant differences in the concentration of components between batching plants, months and collection points. The comparison with the literature showed that CaO, Al2O3 and SiO2 are the majority, both in the samples under study and in the samples from different authors. However, the average amounts of CaO are higher in the waste sludge from the concrete batching plants in question and, consequently, if this material were reused in the same proposals as the authors, the products would possibly present different characteristics and performance.

On-Flow Ligand Screening Assay Based on Immobilized Nucleoside Diphosphate Kinase B from Homo sapiens

We describe an on-flow zonal affinity-based chromatography assay to screen ligands for the nucleoside diphosphate kinase B enzyme (NME2) from Homo sapiens. For the first time, we have covalently immobilized NME2 on the surface of an open fused silica capillary reactor (NME2-ICER) and placed the reactor before the analytical column, which resulted in a two-dimensional liquid chromatography-based system. We evaluated the pH effect on immobilized NME2 activity and carried out steady-state kinetic studies to compare free and immobilized NME2. Steady-state kinetic studies with the substrates adenosine 5’-triphosphate di(tris) salt dihydrate (ATP) and guanosine 5’-diphosphate sodium salt (GDP) resulted in apparent Michaelis-Menten constant values of 1136 and 713 mmol L-1, respectively. The ping-pong catalysis mechanism and substrate specificity were preserved after NME2 immobilization. By employing a reference inhibitor, (-)-epicatechin gallate (ECG), we verified the potential application of this method in NME2 ligand screening and NME2 inhibitor identification. The half maximum inhibitory concentration (IC50) for ECG was 161.3 ± 1.0 μmol L-1.

Termites are insects with a complex social organization on castes among which soldiers are the only responsible for the colony defense. The soldiers of the Nasutitermes genus use chemical defense, which comprises a mixture of mono, sesqui and mainly diterpenes. The new diterpene 2b,3a-dihydroxy-trinervita-1(15),11-diene, found in Nasutitermes macrocephalus, was isolated using preparative high performance liquid chromatography (HPLC) and identified by gas chromatography-mass spectrometry (GC-MS) and nuclear magnetic resonance (NMR) techniques. The absolute stereochemistry determined was supported by ab initio calculations and by comparison of the experimental circular dichroism data with the theoretical results. The isolated diterpene was evaluated against the bacteria Staphylococcus aureus (Gram-positive) and Escherichia coli (Gram-negative), and the fungi Candida albicans and Aspergillus brasiliensis. The isolated diterpene presented minimal inhibitory concentrations of 62.5 and 62.5 µg mL-1 against S. aureus and C. albicans, respectively.

Resumo Fundamento: O número de síndromes genéticas descritas que apresentam alguma forma de cardiopatia e manifestações oculares associadas é grande. Contudo, estas síndromes ainda não foram reunidas e sintetizadas para melhor consulta e comparação. Objetivo: O objetivo deste trabalho é sistematizar a literatura, avaliando evidências disponíveis sobre síndromes que cursam com cardiopatia congênita associada a alterações oculares, salientando os tipos de alterações anatômicas e funcionais descritas. Métodos: Dois pesquisadores independentes fizeram uma busca sistemática utilizando as bases eletrônicas Medline (PubMed, Embase, Cochrane, Lilacs), de trabalhos publicados até o mês de janeiro de 2016. Os critérios de elegibilidade utilizados pelos autores incluíram somente artigos publicados sob a forma de relatos de caso ou revisão, que abordassem a associação de alterações oftalmológicas e cardiológicas em pacientes menores de 18 anos e que apresentassem alguma síndrome genética. Resultados: As síndromes genéticas mais frequentes foram: Síndrome de Down, Síndrome Velo-cardio-facial / DiGeorge, Síndrome de Charge e Síndrome de Noonan. Entre as malformações cardíacas, a comunicação interatrial (77,4%), a comunicação interventricular (51.6%), a persistência do canal arterial (35,4%), estenose da artéria pulmonar (25,8%) e a tetralogia de Fallot (22,5%) foram as mais associadas com achados oculares. Conclusão: Devido à sua variedade clínica, as malformações cardíacas congênitas revelam defeitos que evoluem de maneira assintomática até aqueles que provocam grande morbimortalidade. Dessa forma, encontrar características extra-cardíacas que, de alguma maneira, possam auxiliar no diagnóstico da doença ou revelar a gravidade dessa enfermidade tornam-se de grande relevância.
Abstract Background: Numerous genetic syndromes associated with heart disease and ocular manifestations have been described. However, a compilation and a summarization of these syndromes for better consultation and comparison have not been performed yet. Objective: The objective of this work is to systematize available evidence in the literature on different syndromes that may cause congenital heart diseases associated with ocular changes, focusing on the types of anatomical and functional changes. Method: A systematic search was performed on Medline electronic databases (PubMed, Embase, Cochrane, Lilacs) of articles published until January 2016. Eligibility criteria were case reports or review articles that evaluated the association of ophthalmic and cardiac abnormalities in genetic syndrome patients younger than 18 years. Results: The most frequent genetic syndromes were: Down Syndrome, Velo-cardio-facial / DiGeorge Syndrome, Charge Syndrome and Noonan Syndrome. The most associated cardiac malformations with ocular findings were interatrial communication (77.4%), interventricular communication (51.6%), patent ductus arteriosus (35.4%), pulmonary artery stenosis (25.8%) and tetralogy of Fallot (22.5%). Conclusion: Due to their clinical variability, congenital cardiac malformations may progress asymptomatically to heart defects associated with high morbidity and mortality. For this reason, the identification of extra-cardiac characteristics that may somehow contribute to the diagnosis of the disease or reveal its severity is of great relevance.

SUMMARY Prader-Willi syndrome (PWS) is a genetic disorder frequently characterized by obesity, growth hormone deficiency, genital abnormalities, and hypogonadotropic hypogonadism. Incomplete or delayed pubertal development as well as premature adrenarche are usually found in PWS, whereas central precocious puberty (CPP) is very rare. This study aimed to report the clinical and biochemical follow-up of a PWS boy with CPP and to discuss the management of pubertal growth. By the age of 6, he had obesity, short stature, and many clinical criteria of PWS diagnosis, which was confirmed by DNA methylation test. Therapy with recombinant human growth hormone (rhGH) replacement (0.15 IU/kg/day) was started. Later, he presented psychomotor agitation, aggressive behavior, and increased testicular volume. Laboratory analyses were consistent with the diagnosis of CPP (gonadorelin-stimulated LH peak 15.8 IU/L, testosterone 54.7 ng/dL). The patient was then treated with gonadotropin-releasing hormone analog (GnRHa). Hypothalamic dysfunctions have been implicated in hormonal disturbances related to pubertal development, but no morphologic abnormalities were detected in the present case. Additional methylation analysis (MS-MLPA) of the chromosome 15q11 locus confirmed PWS diagnosis. We presented the fifth case of CPP in a genetically-confirmed PWS male. Combined therapy with GnRHa and rhGH may be beneficial in this rare condition of precocious pubertal development in PWS.

RESUMO: Rhyzopertha dominica F. (Coleoptera: Bostrichidae) é uma praga primária de grãos armazenados em muitas regiões do mundo. Este trabalho teve como objetivo avaliar a atividade fumigante dos óleos essenciais de Ocimum basilicum L., Citrus aurantium L., Mentha spicata L. e Croton pulegiodorus Baill. sobre adultos de R. dominica em grãos de milho armazenados. Para isso, testes para determinação das concentrações letais (CL50 e CL100) e mortalidade (fumigação) foram realizados em câmaras constituídas por recipientes de vidro, contendo 10 indivíduos de R. dominica, com óleos essenciais em diferentes concentrações: O. basilicum e M. spicata (5, 10, 15, 20, 30 e 40 µL/L de ar), C. aurantium (10, 20, 30, 40, 50 e 60 µL/L de ar) e C. pulegiodorus (0, 20, 30, 50, 70 e 90 µL/L de ar). Após 48 h de exposição aos óleos, avaliou-se a percentagem de mortalidade dos insetos. Com base nas CL50 e CL100, a toxicidade dos óleos essenciais decresceu na seguinte ordem: O. basilicum > M. spicata > C. pulegiodorus > C. aurantium. O óleo essencial de O. basilicum exibiu forte toxicidade por fumigação sobre adultos de R. dominica, com valores de CL50 de 17,67 µL/L de ar e de CL100 de 27,15 µL/L de ar. O óleo essencial de C. aurantium requereu concentrações mais altas que os óleos de O. basilicum, M. spicata e C. pulegiodorus para matar os insetos. Contudo, todos os óleos avaliados apresentaram propriedades fumigantes para controlar a R. dominica e demonstraram potencial de utilização no manejo deste coleóptero.
ABSTRACT: The Rhyzopertha dominica F. (Coleoptera: Bostrichidae) is a primary pest of stored grains in many regions of the world. In this work we evaluated the fumigant activity of essential oils of Ocimum basilicum L., Citrus aurantium L., Mentha spicata L. and Croton pulegiodorus Baill on adult R. dominica in stored maize. Tests were conducted to determine lethals concentrations (CL50 and CL100) and mortality (fumigation).The fumigation test was done in containers made of glass containing 10 individuals of R. dominica, where essential oils were applied at different concentrations: O. basilicum and M. spicata (5, 10, 15, 20, 30 and 40µL/L of air), C. aurantium (10, 20, 30, 40, 50 and 60µL/L of air) and C. pulegiodorus (0, 20, 30, 50, 70 and 90µL/L of air). After 48 hours of exposure to the oils the percentage of insect mortality was evaluated. According to LC50 and LC100 the toxicity of essential oils decreased in the following order: O. basilicum> M. spicata> C. pulegiodorus> C. aurantium. The essential oil of O. basilicum exhibited strong fumigant toxicity against R. dominica adults, with a LC50 value of 17.67 µL/L air and LC100 value of 27.15 µL/L air. The C. aurantium essential oil required higher concentrations than O. basilicum, M. spicata and C. pulegiodorus to kill insects. However, all oils evaluated presented fumigating property to promote the control of R. dominica and demonstrated potential use in the management of this coleoptera.

<p>This paper describes the correlation between the phenolic composition and the molecular phylogenetic reconstruction of five <italic>Spondias</italic> species (Anacardiaceae). Two of these species (<italic>S. venulosa</italic> and <italic>Spondias</italic> sp.) occur in rainforest areas and the other three are widely distributed in Brazil (<italic>S. dulcis</italic>, <italic>S.mombin</italic>, and <italic>S. purpurea</italic>). The flavonoid enriched fraction of the <italic>S. venulosa</italic> leaf extract also underwent a chemical study. The results indicate that the presence of flavonol 3-<italic>O</italic>-glycosides are a synapomorphic character of the studied American <italic>Spondias</italic> and the production of rhamnetin 3-<italic>O</italic>-rutinoside is a synapomorphy of the Atlantic forest species. This is the first report of flavonoids in <italic>S. venulosa</italic>, an endemic species from the Brazilian Atlantic rainforest.</p>

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Biodegradable nanoparticles (NPs) have received considerable attention because of their possible use in the development of strategies for the topical delivery of oils and therapeutic drugs, particularly when drug penetration in dermis is desired. Zein is a prolamine and is a promising material for the design of drug delivery systems. In this study, NPs were prepared with zein and were used to encapsulate and release terpinen-4-ol, which is a therapeutic agent for the treatment of melanoma. The results show that the zein NPs are promising nanostructured systems for the prolonged delivery of T4OL with potential applications in anti-melanoma therapy.

Avaliação da sensibilidade e especificidade dos exames não invasivos no diagnóstico da estenose de artéria renal

FUNDAMENTO: O envelhecimento e a aterosclerose estão relacionados à hipertensão renovascular em indivíduos idosos. Independentemente das comorbidades, a estenose de artéria renal é, por si só, importante causa de morbidade e mortalidade cardiovascular. OBJETIVO: Definir a sensibilidade, a especificidade, o valor preditivo positivo e o valor preditivo negativo dos exames não invasivos utilizados no diagnóstico de estenose da artéria renal. MÉTODOS: Um grupo de 61 pacientes recrutados permitiram a análise de 122 artérias e a definição de sensibilidade, especificidade e da contribuição relativa de cada exame realizado (Doppler, cintilografia e angiotomografia, comparados a arteriografia renal). RESULTADOS: A média das idades foi de 65,43 (desvio padrão: 8,7) anos. Das variáveis relacionadas à população do estudo e comparadas à arteriografia, duas estiveram correlacionadas à estenose da artéria renal, à disfunção renal e aos triglicerídeos. A mediana do ritmo de filtração glomerular foi de 52,8 mL/min/m². O Doppler identificou sensibilidade de 82,90%, especificidade de 70%, valor preditivo positivo de 85% e valor preditivo negativo de 66,70%. Para a tomografia, encontraram-se sensibilidade de 66,70%, especificidade de 80%, valor preditivo positivo de 87,50% e valor preditivo negativo de 55,20%. Esses achados permitiram identificar os exames que melhor detectavam a estenose. CONCLUSÃO: A tomografia e o Doppler mostraram qualidade e grande possibilidade no diagnóstico de estenose da artéria renal, com vantagem para o segundo, pois não há necessidade do uso de meio de contraste na avaliação de uma doença que, frequentemente, ocorre em diabéticos e associa-se à disfunção renal e à disfunção ventricular esquerda grave.
BACKGROUND: Aging and atherosclerosis are related to renovascular hypertension in elderly individuals. Regardless of comorbidities, renal artery stenosis is itself an important cause of cardiovascular morbidity and mortality. OBJECTIVE: To define the sensitivity, specificity, positive predictive value, and negative predictive value of noninvasive imaging tests used in the diagnosis of renal artery stenosis. METHODS: In a group of 61 patients recruited, 122 arteries were analized, thus permitting the definition of sensitivity, specificity, and the relative contribution of each imaging study performed (Doppler, scintigraphy and computed tomographic angiography in comparison to renal arteriography). RESULTS: The mean age was 65.43 years (standard deviation: 8.7). Of the variables related to the study population that were compared to arteriography, two correlated with renal artery stenosis, renal dysfunction and triglycerides. The median glomerular filtration rate was 52.8 mL/min/m². Doppler showed sensitivity of 82.90%, specificity of 70%, a positive predictive value of 85% and negative predictive value of 66.70%. For tomography, sensitivity was 66.70%, specificity 80%, positive predictive value 87.50% and negative predictive value 55.20%. With these findings, we could identify the imaging tests that best detected stenosis. CONCLUSION: Tomography and Doppler showed good quality and efficacy in the diagnosis of renal artery stenosis, with Doppler having the advantage of not requiring the use of contrast medium for the assessment of a disease that is common in diabetics and is associated with renal dysfunction and severe left ventricular dysfunction.

The phytochemical investigation of the ethanol extract from leaves of Lecythis pisonis Cambess., Lecythidaceae, resulted in the isolation of seven triterpenes: α- and β-amyrin, uvaol and erythrodiol, ursolic and oleanolic acids and 3β-friedelinol, as well as a mixture of sitosterol and stigmasterol steroids and a diterpene (E)-phytol. The structures of these compounds were identified by¹H and 13C NMR spectral analysis and compared with literature data. The mixture of triterpenes ursolic and oleanolic acids isolated from the active ethereal fraction showed moderate cytotoxic activity. This paper describes for the first time the phytochemical and cytotoxic study of Lecythis pisonis' leaves.

As indústrias galvânicas utilizam, durante o seu processo produtivo, vários tipos de soluções químicas, além de água, que, após o seu ciclo de vida, devem ser descartadas. Entre esses insumos, está o cianeto cúprico, que é tóxico ao ser humano e ao meio ambiente e que está presente em vários tipos de revestimentos nos quais uma camada de cobre é necessária. Esse estudo tem a finalidade de comparar revestimentos eletrodepositados a partir de soluções com cianetos e livres de cianetos, avaliando a qualidade do tratamento superficial de peças. Dessa forma, buscou-se produzir um revestimento com as mesmas qualidades de peças produzidas pelo processo tradicional. Nesse trabalho, foram utilizadas soluções tradicionais com 25 g.L-1 de cianeto e uma solução alternativa isenta de cianeto. As peças revestidas eram de zamak (liga de zinco com alumínio). Para avaliar a qualidade do revestimento obtido foram realizados ensaios acelerados de corrosão, dureza, comparação de espessura e morfologia da camada eletrodepositada. Os primeiros vestígios de corrosão foram verificados após 432 horas nos corpos de prova tradicionais e 1032 horas nos corpos-de-prova produzidos sem cianeto. Nos ensaios de dureza, obteve-se um pequeno ganho de dureza nas peças sem cianeto. A espessura da camada eletrodepositada, assim como a morfologia, foi similar para os dois tipos de processos. Os resultados obtidos demonstram a viabilidade técnica da utilização de soluções livres de cianeto na galvanoplastia.
The electroplating industries use in their productive process several types of chemical solutions, besides water, which after their life cycle must be discarded. Among them, there is cupric cyanide, which is highly toxic for humans and the environment, and it is present in many coatings where the copper layer is necessary. This study aims to compare coatings electrodeposited from solutions with cyanide and free cyanide, with those produced by the traditional process. This comparison involved parts and components of the same quality. In this study, traditional solutions with 25 g.L-1 of cyanide and an alternative solution with 0,015 mg.L-1 of cyanide were used. The parts were produced with Zamak (alloy of Zinc and Aluminum). In order to evaluate the quality of the coating, tests of accelerated corrosion, hardness, comparison of thickness and morphology of the electroplated layer were carried out. The first corrosion traces were detected after 432 hours in the traditional samples and 1032 hours in the free cyanide samples. In hardness tests, there was a better result for the free cyanide samples. The electroplated layer thickness and morphology was similar in both processes. The study results showed that there is technical feasibility in using free cyanide solutions in the electroplating sector.

O objetivo deste trabalho foi avaliar o efeito do óleo essencial de Eucalyptus globulus (OEEG) sobre a eclosão e desenvolvimento de larvas de Haemonchus contortus. A determinação da composição química do OEEG foi feita por meio de cromatografia gasosa acoplada à espectrometria de massa. No teste de eclosão de ovos (TEO), utilizaram-se as concentrações 21,75; 17,4; 8,7; 5,43 e 2,71 mg.mL-1; e no de desenvolvimento larvar (TDL) as concentrações foram 43,5; 21,75; 10,87; 5,43 e 2,71 mg.mL-1. Cada ensaio foi acompanhado por um controle positivo, 0,02 mg.mL-1 tiabendazol, no TEO e 0,008 mg.mL-1 ivermectina no TDL, e por um controle negativo com o diluente Tween 80 (3%). A eficácia máxima obtida pelo OEEG sobre ovos foi de 99,3% na concentração de 21,75 mg.mL-1 e, sobre larvas, 98,7% na concentração de 43,5 mg.mL-1. A concentração do OEEG, que inviabilizou 50% dos ovos e das larvas do parasito, foi de 8,3 e 6,92 mg.mL-1, respectivamente. A análise química do óleo identificou como principal componente o monoterpeno 1,8-cineol. O OEEG apresentou atividade ovicida e larvicida in vitro sobre H. contortus, determinando um bom potencial para utilização no controle de nematóides gastrintestinais de ovinos e caprinos.
The objective of this work was to evaluate ovicidal and larvicidal effects of Eucalyptus globulus essential oil (EGEO) on Haemonchus contortus. The chemical composition determination of EGEO was through gas chromatography and mass spectrometry. Egg hatch test (EHT) was performed in concentrations 21.75; 17.4; 8.7; 5.43 e 2.71 mg.mL-1. In larval development test (LDT) were used the concentrations 43.5; 21.75; 10.87; 5.43 e 2.71 mg.mL-1. Each trial was conducted by negative control with Tween 80 (3%) and positive control, 0.02 mg.mL -1 of thiabendazole in EHT and 0.008 mg.mL-1 of ivermectin in LDT. The maximum effectiveness of EGEO on eggs was 99.3% in concentration of 21.75 mg.mL-1 and on larvae was 98.7% in concentration 43.5 mg.mL-1. The concentration of EGEO that inhibits 50% of the eggs and larvae was 8.3 and 6.92 mg.mL-1, respectively. The oil chemical analysis identified as main component the monoterpen 1,8-cineol. EGEO presented ovicidal and larvicidal activities in vitro, revealing a good potential for use in the control of sheep and goat gastrointestinal nematodes.
