Resumo: A piometra é uma enfermidade comum em cadelas, caracterizada pela inflamação do útero com acúmulo de exsudato purulento. A avaliação ultrassonográfica abdominal é um dos principais exames utilizados para o diagnóstico da doença e o tratamento de eleição é a ovário-histerectomia (OSH). A proteína C reativa (PCR) é uma proteína de fase aguda que apresenta concentração sérica aumentada na ocorrência de processos inflamatórios. O objetivo deste trabalho foi avaliar a eficácia do teste rápido para detecção da PCR sérica (FASTest® CRP canino), como auxiliar no diagnóstico de piometra em cadelas com suspeita da doença ao exame ultrassonográfico. Das 25 cadelas com imagem ultrassonográfica sugestiva de piometra incluídas no estudo, apenas 12 (48,0%) tiveram o diagnóstico confirmado por exame histopatológico uterino realizado após a OSH. Em todas as pacientes com o diagnóstico de piometra confirmado pelo exame histológico a PCR foi positiva. O FASTest® CRP apresentou valor preditivo positivo de 92,3%, valor preditivo negativo e sensibilidade de 100,0% e 92,3% de especificidade. Logo, a acurácia do FASTest® CRP canino para diagnóstico de piometra em cadelas com suspeita ao exame ultrassonográfico foi de 96,0%. Conclui-se que o teste rápido para detecção da PCR sérica pode ser utilizado como exame auxiliar para o diagnóstico de piometra em cadelas.
Abstract: Pyometra is a common disease in bitches characterized by an inflamed uterus filled with pus. Abdominal ultrasonographic evaluation is one of the most requested exam used to diagnose the disease, whereas ovariohysterectomy is the most commonly chosen treatment. C-reactive protein (CRP) is an acute phase protein whose serum concentration increases in inflammatory processes. The goal of this study was to evaluate the effectiveness of the rapid test for detecting serum CRP (FASTest® canine CRP) in the diagnosis of pyometra in bitches with suspicion of the disease by ultrasonography. From 25 bitches with ultrasonographic image suggestive of pyometra included in this study, only 12 (48.0%) has the diagnosis confirmed by histopathological examination of the uterus after ovariohysterectomy. In all patients diagnosed with pyometra, confirmed by histology, the CRP was positive. The FASTest® CRP showed a positive predictive value of 92.3 %, negative predictive value and sensitivity of 100.0% and 92.3% of specificity. Therefore, the accuracy of FASTest® CRP for diagnosis of pyometra in bitches with suspicion by ultrasonography was 96.0%. It was concluded that the rapid test for detecting serum CRP can be used as an aid to diagnose pyometra in bitches.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Potential of soil fumigation with mustard essential oil to substitute biofumigation by cruciferous plant species

Soil fumigation with the synthetic essential oil of mustard (93% allyl isothiocyanate) (EOM) was evaluated as a substitute of bio-fumigation with cruciferous plant species, using Sclerotium rolfsii and Sclerotinia sclerotiorum as test pathogens, together with its non-target effect on the general population of soil microorganisms. The mortality of the sclerotia of both fungi was dependent upon the concentration of EOM and the exposure period. Exposure to the EOM vapors delayed in vitro germination of the sclerotia.Total germination of the control sclerotia of S. rolfsii, after 120 h incubation, was 94%, of which 88% germinated within 48 h. In contrast, to the total germination of 77% or 47%, less than 3% germinated in 48 h when exposed for 4 or 7 days to 50 µl EOM/L. Exposure to 100 µl/l for 4 or 7 days resulted in the mortality of 50 and 100% sclerotia, respectively. The tendency for delayed germination and mortality was similar for the sclerotia of S. sclerotiorum. No viable sclerotia of either fungus were detected to the depth of 10 cm in the air dried, moist or wet soil fumigated for 7 days with the EOM at the rate of 150 µl/L. In the field plots, no viable sclerotia of S. rolfsii were detected after 7-day exposure to the EOM applied to the upper soil layer at the rate of 9, 12, or 18 ml/m² and covered with a plastic sheet, while 73, 50 and 15%, respectively, were recovered from uncovered plots. In field plots, the fluorescein diacetate hydrolysis decreased by EOM treatment. However, the colony forming units of actinomycetes and bacteria increased, but those of fungi decreased significantly. Soil fumigation with EOM can be used with several advantages, as a substitute of bio-fumigation with cruciferous plant species.

An enzyme-linked immunosorbent assay (ELISA) for the detection of IgM antibodies against Leishmania chagasi in dogs

A leishmaniose visceral é uma zoonose emergente, com elevado número de novos casos no Brasil, onde o cão doméstico é um importante reservatório do parasito no ciclo infeccioso da Leishmania chagasi. Um ensaio de imunoadsorção enzimática (ELISA) baseado em antígeno bruto de L. chagasi foi adaptado para a detecção de anticorpos IgM em soros de cães infectados. As diluições ótimas do antígeno e dos soros controles positivo e negativo foram determinadas através de titulação em bloco. A sensibilidade e especificidade do ELISA teste foram de 100%. Um total de 110 amostras de soros foram obtidas de cães oriundos de Belo Horizonte, Minas Gerais, Brasil, e avaliadas pelo ELISA e pela reação de imunofluorescência indireta (RIFI) para anticorpos IgM anti-L. chagasi. Aproximadamente 25% (n=27) dos cães testados foram sorologicamente positivos para L. chagasi pela RIFI, enquanto 89.09% foram soropositivos pelo ELISA. Os resultados obtidos pelo ELISA e pela RIFI foram significativamente diferentes (P<0.01). A associação do ELISA e da RIFI deve ser recomendada a fim de permitir a detecção mais eficiente da leishmaniose visceral canina pelos serviços veterinários.
Visceral leishmaniasis is an emergent zoonosis with an increasing number of new cases in Brazil where the domestic dog is an important parasite reservoir in the infectious cycle of Leishmania chagasi. An enzyme-linked immunosorbent assay (ELISA), based upon the use of a total soluble antigenic preparation of L. chagasi, was adapted for the detection of IgM antibodies in the serum of infected dogs. Optimal dilutions of the antigen, using positive and negative reference sera, were determined by checkboard titrations. The specificity and sensitivity of the ELISA were 100 %. A total of 110 serum samples were taken from dogs in Belo Horizonte, Minas Gerais, Brazil, and examined for anti-L. chagasi IgM antibody by ELISA and indirect fluorescent antibody test (IFAT). About 25% (n=27) of all the dogs tested were found serologically positive for L. chagasi by IFAT, while 89.09% (n=98) were seropositive by ELISA. The results obtained by ELISA and IFAT were significantly different (P<0.01). The combined use of ELISA and IFAT is recommended in order to enable veterinary services to more efficiently detect canine visceral leishmaniasis.
