Resumen Los compuestos volátiles activos olfativamente en el aroma de la fruta del pepino dulce (Solanun muricatum) se extrajeron mediante la técnica de extracción de aroma asistido por disolventes (solvent-assisted flavor evaporation, SAFE), se analizaron por cromatografía de gases acoplada a olfatometría (CGO), y cromatografía de gases acoplada a espectrometría de masas (CG/EM) y se compararon con estándares. Los compuestos con más influencia en el aroma de la fruta se diferenciaron mediante el análisis por dilución del extracto de aroma (aroma extract dilution analysis, AEDA). Con base en el cálculo del valor de la actividad del olor (odor activity value, OAV) se identificaron como compuestos de impacto de la fruta el E-6-nonenal (nota acuosa), el E-2-nonenal (nota verde-pepino), el hexanal (nota verde), el nonanal (nota verde-pepino) y la 1-penten-3-ona (nota etérea). También se estudiaron in vitro algunas de las propiedades biofuncionales de la fruta: la actividad antihipertensiva mediante la inhibición de la enzima convertidora de angiotensina-I (ECA-I) y la actividad antihipercolesterolémica con base en la inhibición de la 3-hidroxi-3-metilglutaril-coenzima A (HMG-CoA) reductasa. Se encontró que la fruta presenta una actividad inhibitoria de la ECA-I promisoria (IC50=3,72 ± 0,19 ppm), ya que se obtuvo un valor mucho menor que el obtenido para el control positivo (lisinopril). También se evidenció con el tiempo que el extracto de la fruta inhibió la actividad de la HMG-CoA reductasa con una cinética comparable a la del control positivo (pravastatina). Con estos resultados, la fruta de S. muricatum se convierte en una candidata potencial para el desarrollo de alimentos funcionales.

Abstract Odor-active volatile compounds of sweet cucumber (Solanum muricatum) fruit were extracted by using the solvent-assisted flavor evaporation (SAFE) technique and analyzed, by gas chromatography coupled to olfactometry (GC-O) and gas chromatography coupled to mass spectrometry (GC-MS), and compared with standards. The most relevant compounds in the aroma of this fruit were determined by the aroma extract dilution analysis (AEDA) technique. Based on the odor activity value (OAV) the following key-aroma compounds were identified: (ii)-6-nonenal (aqueous odor note), (E)-2-nonenal (green, cucumber-like odor note), hexanal (green odor note), nonanal (green, vegetable odor note), and 1-penten-3-one (ethereal note). Some biofunctional properties of the fruit were studied in vitro. Antihypertensive and anti-hypercholesterolemic activities were measured by using the angiotensin-converting enzyme inhibition (ACE-I) and the 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase inhibition assays, respectively. The fruit exhibited a promising ACE-I inhibitory activity (IC50=3.72 ± 0.19 ppm) but lower than that of the positive control (lisinopril). The fruit extract inhibited the activity of the HMG-CoA reductase over time with similar kinetics to the positive control (pravastatin). Based on these results, S. muricatum fruit could be considered a potential candidate for the development of functional foods.

Pouteria sapota is a tropical fruit that is commonly found in central and north part of South America where is widely consumed by its biofunctional properties. The aroma active volatile compounds of Pouteria sapota fruit were studied by using the molecular sensory approach, which includes a SAFE (Solvent Assisted Flavor Evaporation) distillation to get a volatile extract that resembles the fruit aroma, and the combination of GC-O (Gas Chromatography coupled to Olfactometry) with GC-MS analyses to identify odour-active compounds. By AEDA (Aroma Extract Dilution Analysis), β-damascenone, furaneol, linalool, (Z)-3-hexenal, and benzaldehyde, were identified as key aroma compounds in this fruit.

Abstract: With the aim of using the biofunctional ingredients of goldenberry (Physalis peruviana), oil/water emulsions (O/W) were prepared by using as the oily phase sunflower oil fruit pulp extract (ultrasound assisted). Emulsions were spontaneously obtained, with different amount of oil phase/aqueous phase. Also,Tween 20 was employed as surfactant, propylene glycol as cosurfactant, and xanthan gum as viscosity increasing agent. Physicochemical characterization of the emulsions was carried out by measuring pH, conductivity, droplet size distribution and Z potential. Also, carotenoid content and color (tristimulus colorimetry) were determined. The emulsions obtained with addition of xanthan gum showed a droplet size lower than 3.43 µm, increasing their stability. The emulsions containing lower amount of oily phase, were the most stable based on their Z potential values. However, the oily phase showed a high anti hypercholesterolemic activity under the assay of HMG-CoA reductase inhibition, exhibiting higher values than the control pravastatin. This confirms the potential of Goldenberry emulsions as biofunctional food ingredient.

Resumen: Con el objeto de utilizar los ingredientes biofuncionales de la uchuva (Physalis peruviana), se prepararon emulsiones aceite/agua (O/W) usando como fase oleosa, el extracto de uchuva obtenido con aceite de girasol y asistido con ultrasonido. Las emulsiones se obtuvieron espontáneamente utilizando diferentes proporciones de fase oleosa/fase acuosa. Se usó Tween 20 como surfactante, propilenglicol como co-surfactante y goma xantana como viscosante. Las emulsiones se caracterizaron fisicoquímicamente, midiendo pH, conductividad, distribución de tamaño de gota, y potencial Z. También se realizó la medición de contenido de carotenoides y color mediante colorimetría triestímulo. Las emulsiones obtenidas con adición de goma xantana presentaron un tamaño de gota menor a 3.43 µm, incrementando así su estabilidad. Las emulsiones con bajo contenido de fase oleosa se identificaron como las más estables por la medición de potencial Z. Sin embargo, en la fase oleosa se comprobó la actividad antihipercolerestolémica frente a la inhibición de la HMG-CoA reductasa, con valores mayores que el control pravastatina. Esto confirma el potencial de las emulsiones de uchuva como ingrediente biofuncional alimentario.

RESUMEN Antecedentes: Los frutos del roble se tuestan y muelen para producir un polvo que es empleado en la preparación de una bebida sustituta del café. Objetivos: Identificar los compuestos volátiles activos olfativamente presentes en el polvo proveniente de los frutos tostados de Quercus humboldtii; así como llevar a cabo un análisis cuantitativo descriptivo (QDA) de la apariencia, olor y sabor de este polvo. Métodos: Los compuestos volátiles activos olfativamente del polvo de los frutos tostados del roble se extrajeron por DES y se analizaron por CG-EM y análisis olfativo (CG-O). La caracterización sensorial de esta bebida sustituta de café fue realizada mediante el empleo de QDA. Resultados: Se identificaron diecinueve compuestos y se evidenció la presencia de notas de olor como manteca/caramelo, ahumado/tostado y frutal, similares a las encontradas en el café tostado. Los componentes mayoritarios identificados para este sustituto de café fueron cetonas, aldehídos, ácidos alifáticos, alcoholes furánicos y pirazinas. Los compuestos mayoritarios detectados fueron furfural, 5-metil furfural, alcohol furfurílico y 3-hidroxi-2-butanona, algunos de ellos relacionados con la descomposición térmica de azúcares. Los atributos de olor caracterizados por QDA fueron similares a los detectados por CG-O, aunque se detectaron algunas notas de sabor desagradable. Conclusiones: Los resultados del molecular sensory approach confirmaron que este producto se parece al café; sin embargo el QDA mostró la presencia de un sabor desagradable. Estudios adicionales son necesarios para mejorar la calidad del sabor de este sustituto de café.

ABSTRACT Background: Oak acorns are roasted and grounded to produce a powder that is used to prepare a coffee substitute beverage. Objective: To identify the odour-active volatiles of the powder derived from roasted acorns of Quercus humboldtii and to perform a quantitative descriptive analysis (QDA) of its appearance, odour, and taste. Methods: The odour-active volatiles of roasted oak acorn powder were extracted by SDE and analyzed by GC/MS and olfactory analyses (GC/O). Sensorial analyses of this beverage were done by QDA (Quantitative Descriptive Analysis). Results: Nineteen compounds were identified, showing acid, buttery/caramel, smoke/roasted, and fruity odour notes, which are common to those of roasted coffee. Ketones, aldehydes, aliphatic acids, furanic alcohols and pyrazines were identified as relevant for this coffee substitute beverage. The major volatile compounds were furfural, 5-methyl furfural, furfuryl alcohol, and 3-hydroxy-2-butanone, some of them related to the sugar breakdown by heating. Odour attributes characterized by QDA, were in agreement with those detected by GC-O (Gas Chromatography coupled to Olfactometry), but some off-taste notes were detected. Conclusions: The results of molecular sensory approach confirmed that this product resembles coffee; however, QDA analyses showed the presence of undesirable taste. More studies are needed in order to improve the taste quality of this coffee substitute.

Los frutos de guanábana (Annona muricata L. cv. Elita) en diferentes estados de madurez se evaluaron para determinar las características fisico-químicas: sólidos solubles totales, porcentaje de acidez, pH e índice de madurez. Los cambios en la composición de los compuestos volátiles se determinaron mediante la técnica de Espacio de Cabeza-Microextracción en Fase Sólida y el posterior análisis por Cromatografía de gases acoplada a espectrometría de masas. Los compuestos volátiles se identificaron por comparación de sus propiedades cromatográficas y espectrales con sustancias de referencia. Los factores de calidad sensorial mostraron valores que variaron con el estado de madurez. Además, se estableció que la cantidad de ésteres, especialmente los provenientes de los ácidos C6, C4 y C8, saturados e insaturados, se incrementaron con la maduración. Los compuestos alifáticos C6, como el (Z)-3-hexenol y el (Z)-3-hexenal, fueron los volátiles mayoritarios en la fruta verde; en tanto que el hexanoato de metilo y el (E)-2-hexenoato de metilo, fueron los más abundantes en las frutas maduras, sobremaduras y de madurez intermedia. En la sobremaduración aumentó la concentración de acetato de etilo, butanoato de metilo y butanoato de etilo; y aparecieron los ácidos alifáticos butanoico y hexanoico, los cuales pueden ser indicadores del inicio de la etapa fermentativa y de la pérdida de la calidad sensorial de la fruta.

Fruits of soursop (Annona muricata L. cv. Elita) were evaluated at different ripening stages to determine the changes in their physicochemical characteristics: total soluble solid content, percentage of acidity, pH, and ripeness index. The change in volatile compounds were determined through Headspace-Solid Phase Microextraction and subsequent Gas Chromatography-Mass Spectrometry analyses. The volatile compounds were identified through the comparison of their chromatographic and spectral properties against the ones exhibited by reference substances. The sensorial quality factors showed values that changed according to the ripening stage. It was established that the ester levels, particularly those from C6, C4, and C8 saturated and unsaturated aliphatic acids, increased with ripening. C6 aliphatic compounds, such as (Z)-3-hexenol and (Z)-3-hexenal, were observed to be the major volatile constituents in the green stage. In contrast, methyl hexanoate and methyl (E)-2-hexenoate were the main volatile in ripe, overripe and half ripe fruits. The overripe stage was characterized by an increase in the ethyl acetate, methyl butanoate and ethyl butanoate levels, and the appearance of butanoic and hexanoic aliphatic acids. These facts could be used as an indicator of the beginning of the fermentation stage and the loss of fruit sensory quality.

La composición de la fracción hexánica de la corteza de Croton stipuliformis se estudió por cromatografía de gases acoplada a espectrometría de masas; se caracterizaron los sesquiterpenos y esteroles como los principales constituyentes. Adicionalmente a estos compuestos, tres diterpenoides labdanos y seco-labdanos, el ácido 8(17),12E,14-labdatrien-18-oico [3], el ácido 12E-3,4-seco-labda-4(18),8(17),12,14-tetraen-3- oico [2] y su ester metílico [1] se aislaron de esta fracción por HPLC preparativa y sus estructuras se elucidaron con base en los análisis por HRFABMS, espectroscopía de RMN mono y bidimensional. Adicionalmente, se determinó la actividad citotóxica de estos tres compuestos frente a las líneas celulares de tumores humanos HEp-2, HT-29, MKN-45, MCF-7 y HeLa. Todos los compuestos mostraron una citotoxidad moderada y no específica frente a las líneas celulares mencionadas anteriormente.

The composition of the cytotoxic hexane fraction of the Croton stipuliformis bark was studied by means of a gas chromatography coupled to a mass spectrometry; sesquiterpenes and sterols were characterized as the main constituents. Along with these compounds, three labdane and seco-labdane diterpenoids, 8(17),12E,14-labdatrien-18-oic acid [3], 12E-3,4-seco-labda-4(18),8(17),12,14-tetraen-3-oic acid [2], and its methyl ester [1] were isolated from this fraction through preparative HPLC, and their structures were elucidated by HR-FABMS, 1D and 2D NMR analyses. Additionally, the cytotoxic activity of these three compounds against human tumor cell lines HEp-2, HT-29, MKN-45, MCF-7, and HeLa was assessed. The three compounds showed a non-specific and moderate cytotoxicity against the abovementioned cell lines.

Two food products (powders) were obtained by hot-air drying or lyophilisation methods on the whole guava fruits. The powders were characterised by sensory and thermal analyses (TGA-DSC), infrared spectroscopy (IR), X-ray diffraction (XRD) and scanning electron microscopy (SEM). Thermal, morphological and structural characterisations showed a similar behaviour for the two solids. TGA-DSC and IR showed the presence of pectin as the main constituent of solids. A semi-crystalline profile was evidenced by XRD, and lamellar/spherical morphologies were observed by SEM. Sensory analyses revealed an aroma highly related to guava. These value-added food products are an alternative to process guava and avoid loss during postharvest handling.

Durante el estudio de los compuestos activos olfativamente en el aroma de la uchuva (Physalis peruviana L.) se identificaron los compuestos volátiles, obtenidos por 3 técnicas, extracción líquido-líquido con pentano-diclorometano (1:1) (LL), Solvent-Assisted Flavor Evaporation (SAFE) y í/eadspace-Microextracción en fase sólida (HS-MEFS), y se encontraron diferencias en la calidad del aroma de los extractos mediante el análisis olfato-métrico. La técnica SAFE permitió obtener un extracto con un aroma muy semejante al de la fruta fresca. Los compuestos que presentaron los valores más altos de factor de dilución de aroma (FD) fueron: hexanal, 3-hidroxi-2-butanona, 2-metil-propanol, 2-hidroxibutanoato de etilo, octanoato de etilo y 3-hidroxibutanoato de butilo. En el extracto LL se detectaron algunos compuestos sin actividad olfativa en la fruta; en contraste, mediante la técnica de HS-MEFS no fue posible detectar todos los compuestos activos olfativamente, principalmente los de mayor polaridad. Estos resultados muestran la utilidad del nuevo enfoque de análisis en química de aromas, el cual le da más relevancia al análisis olfativo en estos estudios.

During the study of odor-active active compounds in the aroma of cape gooseberry (Physalis peruviana L.), volatile compounds obtained by three techniques-liquid liquid extraction with penta-ne-dichloromethane (1:1) (LL), Solvent-Assisted Flavor Evaporation (SAFE) and Headspace-Solid Phase Microextraction (HS-SPME) were identified. Differences in the aroma quality of the extracts were found by olfactometric analyses. The SAFE technique allowed obtain in ganextract with similar aroma to the freshfruit. Thecompounds with highest Flavor Dilution factor (FD) value were hexanal, 3-hydroxy-2-butanone, 2-methylpropanol, ethyl 2-hydroxybutanoate, ethyl octanoate, and butyl 3-hydroxybutanoate. Some compounds without olfactive activity in the fruit aroma were detected in the LL extract; in contrast, it was not possible todetectalloftheodor-activevolatilesby using HS-SPME technique, especially those of the most polarity. These results showtheutilityinusingthenewapproach of flavor chemistry analysis, which gives more relevance to the odor analysis during these studies.

Durante o estudo de compostos olfativos ativos no aroma de uchuva (Physalis peruviana L.), foram identificados os compostos voláteis obtidos por três técnicas: extração líquido-líquido com pentano-diclorometano (1:1) (LL), Solvent-Assisted Flavor Evaporation (SAFE) e Heads-pace-Microextração em Fase Sólida (HS-MEFS). Também se acharam diferenças na qualidade do aroma dos extratos por meio de análise de olfatometria. A técnica SAFE permitiu a obtenção de um extrato com um aroma semelhante à fruta fresca. Os compostos que apresentaram os maiores valores de fator de diluição de aroma (FD) foram: hexanal, 3-hidroxi-2-butanona, 2 metilpropanol, octanoato de etilo e 3-hidroxibutanoato de butilo. No extracto LL foram detectados alguns compostos sem atividade olfativa no fruto; em contrapartida, pela técnica de HS-MEFS não foi possível detectar todos os compostos ativos olfativamente, principalmente os de maior polaridade. Estes resultados mostram a utilidade do uso de uma nova abordagem para a análise de química do aroma, que dá mais importância á análise olfativa em estes estudos.
