A cardiomiopatia hipertrófica (CMH) é uma forma de doença do músculo cardíaco de causa genética, caracterizada pela hipertrofia das paredes ventriculares. O diagnóstico requer detecção por métodos de imagem (Ecocardiograma ou Ressonância Magnética Cardíaca) de qualquer segmento da parede do ventrículo esquerdo com espessura > 15 mm, sem outra causa provável. A análise genética permite identificar mutações de genes codificantes de diferentes estruturas do sarcômero responsáveis pelo desenvolvimento da CMH em cerca de 60% dos casos, permitindo o rastreio de familiares e aconselhamento genético, como parte importante do manejo dos pacientes e familiares. Vários conceitos sobre a CMH foram recentemente revistos, incluindo sua prevalência de 1 em 250 indivíduos, não sendo, portanto, uma doença rara, mas subdiagnosticada. A vasta maioria dos pacientes é assintomática. Naqueles sintomáticos, a obstrução do trato de saída do ventrículo esquerdo (OTSVE) é o principal distúrbio responsável pelos sintomas, devendo-se investigar a sua presença em todos os casos. Naqueles em que o ecocardiograma em repouso ou com Manobra de Valsalva não detecta gradiente intraventricular significativo (> 30 mmHg), devem ser submetidos à ecocardiografia com esforço físico para detecção da OTSVE. Pacientes com sintomas limitantes e grave OTSVE, refratários ao uso de betabloqueadores e verapamil, devem receber terapias de redução septal ou uso de novas drogas inibidoras da miosina cardíaca. Por fim, os pacientes adequadamente identificados com risco aumentado de morta súbita podem receber medida profilática com implante de cardiodesfibrilador implantável (CDI).
La miocardiopatía hipertrófica (MCH) es una forma de enfermedad cardíaca de origen genético, caracterizada por el engrosamiento de las paredes ventriculares. El diagnóstico requiere la detección mediante métodos de imagen (Ecocardiograma o Resonancia Magnética Cardíaca) que muestren algún segmento de la pared ventricular izquierda con un grosor > 15 mm, sin otra causa probable. El análisis genético permite identificar mutaciones en genes que codifican diferentes estructuras del sarcómero responsables del desarrollo de la MCH en aproximadamente el 60% de los casos, lo que permite el tamizaje de familiares y el asesoramiento genético, como parte importante del manejo de pacientes y familiares. Varios conceptos sobre la MCH han sido revisados recientemente, incluida su prevalencia de 1 entre 250 individuos, por lo tanto, no es una enfermedad rara, sino subdiagnosticada. La gran mayoría de los pacientes son asintomáticos. En los casos sintomáticos, la obstrucción del tracto de salida ventricular izquierdo (TSVI) es el trastorno principal responsable de los síntomas, y su presencia debe investigarse en todos los casos. En aquellos en los que el ecocardiograma en reposo o la maniobra de Valsalva no detecta un gradiente intraventricular significativo (> 30 mmHg), deben someterse a ecocardiografía de esfuerzo para detectar la obstrucción del TSVI. Los pacientes con síntomas limitantes y obstrucción grave del TSVI, refractarios al uso de betabloqueantes y verapamilo, deben recibir terapias de reducción septal o usar nuevos medicamentos inhibidores de la miosina cardíaca. Finalmente, los pacientes adecuadamente identificados con un riesgo aumentado de muerte súbita pueden recibir medidas profilácticas con el implante de un cardioversor-desfibrilador implantable (CDI).
Hypertrophic cardiomyopathy (HCM) is a form of genetically caused heart muscle disease, characterized by the thickening of the ventricular walls. Diagnosis requires detection through imaging methods (Echocardiogram or Cardiac Magnetic Resonance) showing any segment of the left ventricular wall with a thickness > 15 mm, without any other probable cause. Genetic analysis allows the identification of mutations in genes encoding different structures of the sarcomere responsible for the development of HCM in about 60% of cases, enabling screening of family members and genetic counseling, as an important part of patient and family management. Several concepts about HCM have recently been reviewed, including its prevalence of 1 in 250 individuals, hence not a rare but rather underdiagnosed disease. The vast majority of patients are asymptomatic. In symptomatic cases, obstruction of the left ventricular outflow tract (LVOT) is the primary disorder responsible for symptoms, and its presence should be investigated in all cases. In those where resting echocardiogram or Valsalva maneuver does not detect significant intraventricular gradient (> 30 mmHg), they should undergo stress echocardiography to detect LVOT obstruction. Patients with limiting symptoms and severe LVOT obstruction, refractory to beta-blockers and verapamil, should receive septal reduction therapies or use new drugs inhibiting cardiac myosin. Finally, appropriately identified patients at increased risk of sudden death may receive prophylactic measure with implantable cardioverter-defibrillator (ICD) implantation.

ABSTRACT This work investigated the combination of agitation and aeration conditions in a bench-bioreactor to identify the optimal biosurfactant production from substrate based on beet peel and glycerol from a biodiesel process. Thus, a central composite rotatable design (CCRD) and responses were evaluated by response surface methodology (RSM) modeling. The optimal operation values determined were 200 rpm (agitation) and 0.5 vvm (aeration), reaching values of 1931.2 mg/L of crude biosurfactant concentration and 28.37 mN/m of surface tension. For the development of a mathematical model based on an artificial neural network (ANN), the experimental data from each run (CCRD) of the bioreactor were used. The results indicated a topology of 6-6-1 neurons with an excellent predictive capacity of biosurfactant concentration: dispersion plot with R2 of 0.995, and error criteria SSE of 0.31, MSE of 7.29×10-4 and RSME of 2.7×10-2. A soft sensor was then designed in an electronic spreadsheet, computing the biosurfactant production from secondary measurements. Furthermore, the produced biosurfactant showed the ability to remediate oil spreading, evaluated through the appearance of clear zones on the surface of water covered with oil, and also from high emulsification indexes obtained on most of the solvents tested, such as toluene (~65%).

ABSTRACT At the Brazilian Amazon forest, studies were carried out to estimate the community of arbuscular mycorrhizal fungi (AMF), from this it was used a bioassay of dilutions of samples collected from preserved and regenerated areas after bauxite extraction. To regenerate areas, tree species were introduced and samples were taken after 2, 6, 12, and 16 years, the spores obtained were compared to those obtained by direct extraction and the number of species recovered from the bioassay was significantly higher. Therefore, the species founded after different periods regeneration was similar to the ones from the native forest. Since the early years of revegetation, the number of rare species was high with strong dominance of G. macrocarpum. Among older communities this high dominance decreased while at the same time, there was an increase in the number of individuals from other AMF species, concluding that the number of species did not change with the age of the revegetation.

The effectiveness of basal shoot mechanical leaf removal at the onset of bloom to control crop on cv. Sangiovese (V vinifera L.): report on a three-year trial

Vine basal shoot leaves are known to be the primary nutritional source for fruit set at the onset of bloom. The effectiveness of mechanical removal of this foliage at that date was tested from 2012 to 2014 to control the cropping of high-yielding cv. Sangiovese in a "Toscana rosso" TGI (Typical Geographic Indication) district in Tuscany, where the yield threshold is 16 t/ha. A tractor-mounted leaf remover featuring side-by-side rotary suction and feed rollers was employed along a 50 cm basal area of cropping shoots at the beginning of bloom; control was the usual manual thinning of clusters carried out at véraison in the same vineyard. Mechanical leaf removal eliminated about 30% of leaf area and some shoots and inflorescences, thus reducing cropping potential and even resulting in a physiological effect, as the resulting clusters were composed of lower numbers of berries and were less compact and less susceptible to mould than the control. By harvest, the defoliated vines showed higher leaf area, most likely because leaf removal at the onset of bloom may have triggered compensatory new growth, and their grapes had a higher content of soluble solids than the thinned control. While both treatments kept the yield below the 16 t/ha threshold - leaf removal at an estimated 15.1 t/ha and manual cluster thinning at 15.6 t/ha - mechanical defoliation notably reduced yearly labour input: an estimated 4 h/ha against the 38 h/ha from thinning.

The determinations of cellulose and lignin are important in wood analysis from both perspectives: chemistry composition, commercial and industrial application of woods. In this context, Raman image spectroscopy was applied in the determination of cellulose and lignin distribution in wood surfaces of Swietenia macrophylla King (Mahogany) and Eucalyptus hybrid (E. urophylla × E. camaldulensis). The relative concentration maps were obtained by applying a multivariate curve resolution procedure. The estimated Raman spectra for cellulose and lignin agrees with the ones available in the literature. The cellulose concentration maps showed similar variations for both studied species with an average value of nearly 85% as obtained by the reference method. The lignin concentration maps were somewhat different. The Eucalyptus hybrid presents a broader distribution (20 to 45%) while for Mahogany it is nearly constant, around 25%, with localized regions presenting contents of 45 to 65% of lignin. The reference method showed that the mean lignin content of both species is 27%. The cellulose and lignin concentration maps agree with the biological functions of the anatomic structures observed in the images.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Reconhecer com precisão indivíduos com maior risco imediato de morte súbita cardíaca (MSC) ainda é uma questão em aberto. A natureza fortuita dos eventos cardiovasculares agudos não parece se adequar ao conhecido modelo de indução de taquicardia/fibrilação ventricular por um gatilho em sincronia a um substrato arritmogênico estático. Quanto ao mecanismo da MSC, uma instabilidade elétrica dinâmica explicaria melhor a raridade da associação simultânea de um gatilho certo a um substrato cardíaco apropriado. Diversos estudos tentaram medir essa instabilidade elétrica cardíaca (ou um equivalente válido) em uma sequência de batimentos cardíacos no ECG. Dentre os mecanismos possíveis podemos citar o prolongamento do QT, dispersão do QT, potenciais tardios, alternância de onda T ou T-wave alternans (TWA), e turbulência da frequência cardíaca. Este artigo se atém em particular ao papel da TWA no panorama atual da estratificação de risco cardíaco. Os achados sobre TWA ainda são heterogêneos, variando de um desempenho prognóstico muito bom até um quase nulo, dependendo da população clínica observada e protocolo clínico usado. Para preencher as atuais lacunas no conhecimento sobre TWA, profissionais médicos e pesquisadores devem explorar melhor as características técnicas das diversas tecnologias disponíveis para a avaliação de TWA e atentar ao fato de que os valores de TWA respondem a diversos outros fatores, além de medicamentos. Informações sobre mecanismos celulares e subcelulares da TWA estão fora do escopo deste artigo, mas são referenciados alguns dos principais trabalhos sobre este tópico, com o intuito de auxiliar no entendimento dos conceitos e fatos cobertos neste artigo.
Accurate recognition of individuals at higher immediate risk of sudden cardiac death (SCD) is still an open question. The fortuitous nature of acute cardiovascular events just does not seem to fit the well-known model of ventricular tachycardia/fibrillation induction in a static arrhythmogenic substrate by a synchronous trigger. On the mechanism of SCD, a dynamical electrical instability would better explain the rarity of the simultaneous association of a correct trigger and an appropriate cardiac substrate. Several studies have been conducted trying to measure this cardiac electrical instability (or any valid surrogate) in an ECG beat stream. Among the current possible candidates we can number QT prolongation, QT dispersion, late potentials, T-wave alternans (TWA), and heart rate turbulence. This article reviews the particular role of TWA in the current cardiac risk stratification scenario. TWA findings are still heterogeneous, ranging from very good to nearly null prognostic performance depending on the clinical population observed and clinical protocol in use. To fill the current gaps in the TWA base of knowledge, practitioners, and researchers should better explore the technical features of the several technologies available for TWA evaluation and pay greater attention to the fact that TWA values are responsive to several factors other than medications. Information about the cellular and subcellular mechanisms of TWA is outside the scope of this article, but the reader is referred to some of the good papers available on this topic whenever this extra information could help the understanding of the concepts and facts covered herein.

En cuanto al mecanismo de la MSC, una inestabilidad eléctrica dinámica explicaría mejor la rareza de la asociación simultánea de un gatillo correcto a un substrato cardíaco apropiado. Diversos estudios trataron de medir esa inestabilidad eléctrica cardíaca (o un equivalente válido) en una secuencia de latidos cardíacos en el ECG. Entre los mecanismos posibles podemos citar la prolongación del QT, dispersión del QT, potenciales tardíos, alternancia de onda T o T-wave alternans (TWA), y turbulencia de la frecuencia cardíaca. Este artículo se atiene en particular al papel de la TWA en el panorama actual de la estratificación de riesgo cardíaco. Los hallazgos sobre TWA aun son heterogéneos, variando de un desempeño pronóstico muy bueno hasta uno casi nulo, dependiendo de la población clínica observada y protocolo clínico usado. Para llenar las actuales lagunas en el conocimiento sobre TWA, profesionales médicos e investigadores deben explotar mejor las características técnicas de las diversas tecnologías disponibles para la evaluación de TWA y estar atento al hecho de que los valores de TWA responden a otros diversos factores, además de medicamentos. Informaciones sobre mecanismos celulares y subcelulares de la TWA están fuera del objetivo de este artículo, pero son referenciados algunos de los principales trabajos sobre este tópico, con el propósito de auxiliar en la comprensión de los conceptos y hechos cubiertos en este artículo.

Este trabalho descreve os métodos de produção e os usos potenciais das peneiras moleculares de carbono. A principal rota sintética para estes materiais é a replicação, onde uma peneira molecular de sílica ou aluminossilicato é usada como gabarito (template) para o crescimento de uma fase carbonácea nos seus canais e cavidades. Estes materiais podem ter aplicações tão variadas como separação, adsorção e armazenamento de gases, como eletrodos em baterias e como suportes de catalisadores, todas elas extremamente dependentes da porosidade da peneira molecular de carbono.
This paper describes the production methods and the prospective uses of carbon molecular sieves. The main route to these materials is replication synthesis, where a silica or aluminosilicate molecular sieve is used as template to grow the carbonaceous phase in the voids. These materials may have applications as varied as in separation, adsorption and storage of gases, as electrodes in batteries, and as catalyst supports, all of them highly dependent on the molecular sieve porosity.

Production of lipase from Geotrichum sp and adsorption studies on affinity resin

There is a growing interest in microbial lipase production due to its great potential for industrial applications such as food additives, industrial reagents and stain removers, as well as for medical applications. Specially for medical applications a high degree of purity is required, which is accomplished with high resolution chromatographic techniques. Affinity chromatography is considered a very high resolution chromatographic technique. In this work the adsorption isotherms and kinetics of the adsorption of lipase from Geotrichum sp on biospecific resin were determined. The resin was prepared using EAH sepharose 4B gel (Pharmacia), made to react with oleic acid as the specific ligand.The lipase was produced in a five-liter fermenter, with both complex and synthetic media. Fermentation conditions were a temperature of 30°C, an aeration of 1VVM and an agitation of 400 rpm. Maximum lipase activity was around 28 U/ml after 10 hours of fermentation for the complex medium. The kinetic model and parameters were determined by dynamic fitting to experimental results using the fourth-order Runge-Kutta method.
