RESUMO A queima das folhas do inhame, causada por Curvularia eragrostides pode comprometer a produção do inhame (Dioscorea spp.), uma cultura de extrema importância para a região Nordeste do Brasil. O uso de produtos naturais vem sendo utilizados com eficiência no controle de doenças de plantas por sua ação fungitóxica ou pela presença de compostos eliciadores, reduzindo o uso de defensivos agrícolas. O objetivo deste trabalho foi avaliar o efeito de diferentes formas de controle da queima das folhas do inhame causado por C. eragrostides. Foi analisada a esporulação de C. eragrostides em placas de Petri contendo meio BDA com os seguintes produtos: manipueira (40 %); óleo de hortelã (100 μL/mL), extratos de alho, e de melão de São Caetano, folha de inhame (20 %), Ecolife® (2 %); Trichoderma sp. (2,0 g. L-1); acibenzolar-S-metil (0,15 g. L-1 e 0,3 g. L-1), separadamente. A testemunha foi placa com meio BDA sem adição de produto. A avaliação do efeito preventivo dos produtos in vivo, sobre o desenvolvimento da doença foi realizada com tratamentos selecionados no ensaio in vitro. Nesse ensaio in vivo as plantas de inhame cv. Da Costa, com 45 dias de idade foram pulverizadas 48 e 72 h antes da inoculação do patógeno com óleo de hortelã (100 μL), extratos de alho e de folhas de inhame (20%); Trichoderma sp. (2,0 g.L-1); ASM (0,15 e 0,3 g L-1) e água (testemunha). Os produtos naturais inibiram a esporulação de C. eragrostides. O fungo Trichoderma sp., óleo de hortelã e ASM 0,3 foram eficientes em reduzirem a severidade da queima das folhas de inhame. O tempo de inoculação influenciou a severidade da doença, de acordo com o produto testado. Trichoderma sp. foi o tratamento que mais reduziu a severidade da doença (51%), sendo este, o tratamento podendo ser recomendado para integrar as táticas de manejo da queima das folhas do inhame.

ABSTRACTUMO Leaf blight, caused by Curvularia eragrostides, can compromise the production of yam (Dioscorea spp.), a crop of extreme importance for the northeast region of Brazil. Natural products have been efficient in controlling plant diseases due to their fungitoxic action or to the presence of eliciting compounds, reducing the use of pesticides. The objective of this study was to evaluate the effect of different methods to control yam leaf blight caused by C. eragrostides. Sporulation of the fungus was analyzed in Petri plates containing PDA medium plus the following products: “manipueira” (40%); mint oil (100 μL/mL); garlic, ‘São Caetano’ melon and yam leaf extracts (20%); Ecolife® (2%); Trichoderma sp. (2.0 g.L-1), and acibenzolar-S-methyl (0.15 g.L-1 and 0.3 g.L-1), separately. Control treatment consisted of a plate containing PDA medium without addition of products. The preventive effect of products on the disease development in vivo was assessed with treatments selected in the in vitro test. In the in vivo experiment, yam plants cv. ‘Da Costa’, 45 days old, were sprayed with the products selected in the in vitro test at 48h and 72h before inoculation of the pathogen and mint oil (100 μL); garlic and yam leaf extracts (20%); Trichoderma sp. (2.0 g.L-1); acibenzolar-S-methyl (0.15 and 0.3 g L-1), and water (control). All natural products inhibited C. eragrostides sporulation. The best severity reduction was provided by Trichoderma sp. applied at 72h before inoculation of the pathogen (51%), followed by mint oil at 48h (41.4%) and 72h (31.5%), Trichoderma sp. at 48h (31.5%) and acibenzolar-S-methyl 0.3 g.L-1 at 72h (25.5%). The inoculation time influenced the disease severity according to the tested product. Trichoderma sp. was the treatment that most reduced the disease severity (51%) and can be recommended to integrate yam leaf blight management strategies.

RESUMO Os badnavírus (família Caulimoviridae) possuem genoma de dsDNA semicircular encapsidado em partículas baciliformes. Dioscorea bacilliform virus (DBV) é um importante Badnavirus infectando várias espécies de Dioscorea (inhame) no hemisfério sul. No presente estudo, 235 amostras foliares de inhame foram coletadas em diferentes bancos de germoplasma mantidos pela Escola Superior de Agricultura Luiz (ESALQ/USP), Universidade Federal de Viçosa (UFV) e Universidade Federal do Recôncavo Baiano (UFRB). A incidência média de badnavírus nas amostras avaliadas foi de 75,3%. As análises moleculares e filogenéticas, realizadas a partir de 42 sequências nucleotídicas, da região da RT/RNaseH de badnavírus, revelaram a ocorrência do Dioscorea bacilliform AL virus (DBALV) e Dioscorea bacilliform SN virus (DBSNV) em diferentes espécies de inhame. Até o momento, somente o DBALV havia sido relatado em inhame no Brasil, estando restrito à região Nordeste. Portanto, este é o primeiro relato do DBSNV infectando a cultura do inhame, no país. Além das espécies espissomais, também foram detectadas sequências endógenas, com porcentagem de identidade de nucleotídeos variando entre 89% e 100% com endogenous Dioscorea bacilliform virus (eDBV), pertencentes aos grupos 9 e 12. Os resultados obtidos corroboram com a hipótese de que o DBALV e o DBSNV estão presentes em materiais propagativos de inhame e amplamente disseminados no Brasil.

ABSTRACT Badnaviruses (family Caulimoviridae) have semicircular dsDNA genomes encapsidated into bacilliform particles. Dioscorea bacilliform virus (DBV) is an important Badnavirus infecting several species of Dioscorea (yam) in the southern hemisphere. In this study, 235 yam leaf samples were collected from different germplasm banks maintained by “Luiz de Queiroz” College of Agriculture (ESALQ/USP), Federal University of Viçosa (UFV) and Federal University of Recôncavo da Bahia (UFRB). The average incidence of badnavirus in the evaluated samples was 75.3%. Molecular and phylogenetic analyses, carried out from 42 nucleotide sequences, of the badnavirus RT/RNaseH region, revealed the occurrence of Dioscorea bacilliform AL virus (DBALV) and Dioscorea bacilliform SN virus (DBSNV) in different yam species. So far, only DBALV has been reported in yams in Brazil, being restricted to the Northeast region. Therefore, this is the first report of DBSNV infecting yam crop in the country. In addition to the episomal species, endogenous sequences were detected, and the percentage of nucleotide identity varied between 89% and 100% with endogenous Dioscorea bacilliform virus (eDBV), belonging to groups 9 and 12. The obtained results corroborate the hypothesis that DBALV and DBSNV are present in yam propagating materials and are widely disseminated in Brazil.

RESUMO A presença de Lasiodiplodia theobromae (Podridão seca) em videiras implantadas em áreas irrigadas da região semiárida brasileira, tem mostrado elevada agressividade ao infectar várias partes das plantas. Os óleos essenciais têm apresentado eficiência no controle de doenças, tanto pela ação fungitóxica, inibindo o crescimento micelial e a germinação de esporos, além da presença de compostos eliciadores. Com isso, o presente trabalho teve por objetivo obter o controle de L. theobromae pela utilização do óleo de Citronela (Cymbopogon nardus L) em diferentes dosagens. O crescimento micelial de L. theobromae foi analisado em placas de Petri contendo o meio batata-dextrose-ágar suplementado com o óleo essencial nas seguintes concentrações: 0; 0,25; 0,5; 1,0; 1,5 e 2,0%. Para avaliação do efeito direto do óleo essencial sobre o desenvolvimento da doença, os cachos de uva foram inoculados com o patógeno e, após 4 horas, aspergidos com soluções contendo as mesmas concentrações descritas anteriormente; posteriormente, com o objetivo de avaliar o potencial do óleo essencial no tratamento preventivo, os mesmos tratamentos foram aplicados em cachos de uva 4 horas antes da inoculação. O óleo de Citronela em todas as concentrações testadas foi capaz de inibir o crescimento micelial de L. theobromae em 100%. No tratamento curativo, com relação a incidência, Citronela 0,25% reduziu 38% na infecção de bagas de cachos de uva inoculadas com L. theobromae, sendo capaz de reduzir a severidade da doença em 56%. No tratamento preventivo, o óleo de Citronela a 0,25% reduziu em 52% a infecção das bagas de uva cv. Itália, reduzindo a severidade da doença em 66,3%. Sendo possível concluir que a concentração 0,25% foi a melhor dosagem testada para o controle da doença com o óleo essencial de Citronela.

ABSTRACT Presence of Lasiodiplodia theobromae (dry rot) in vines established in irrigated areas of the Brazilian semiarid region has shown to be highly aggressive when infecting several parts of the plants. Essential oils have shown efficiency in controlling diseases, both by the fungitoxic action, inhibiting mycelial growth and spore germination, and by the presence of eliciting compounds. Thus, the objective of this study was to obtain control of L. theobromae by using citronella oil (Cymbopogon nardus L) at different levels. Mycelial growth of L. theobromae was analyzed in Petri dishes containing potato-dextrose-agar medium supplemented with essential oil at the following concentrations: 0; 0.25; 0.5; 1.0; 1.5 and 2.0%. To evaluate the direct effect of the essential oil on the development of the disease, grape bunches were inoculated with the pathogen and, after 4 h, sprayed with solutions containing the same concentrations described above; subsequently, in order to evaluate the potential of the essential oil in the preventive treatment, the same treatments were applied in grape bunches 4 hours before inoculation. Citronella oil at all tested concentrations was capable of inhibiting the mycelial growth of L. theobromae by 100%. For the healing treatment, considering incidence, 0.25% citronella reduced by 38% the infection in berries of grape bunches inoculated with L. theobromae and was capable of reducing the disease severity by 56%. As regards the preventive treatment, 0.25% citronella oil reduced by 52% the infection in berries of grape bunches cv. ‘Itália’, reducing the disease severity by 66.3%. These results lead to the conclusion that 0.25% was the best concentration tested for disease control with citronella essential oil.

RESUMO Com a realização deste trabalho objetivou-se verificar o controle de L. theobromae através da utilização de extratos vegetais. Inicialmente, foi avaliada a inibição do crescimento micelial de L. theobromae em placas de Petri contendo o meio BDA suplementado com os extratos de Boldo Baiano, Jurema Preta, Pata de Vaca e Pau-ferro, nas concentrações 10, 20 e 30%, para todos os extratos. Posteriormente, foram realizados dois experimentos in vivo com o objetivo de avaliar o efeito direto e protetor dos extratos vegetais testados in vitro sobre a intensidade da podridão seca em cachos da videira cv. Itália, utilizando-se a concentração de 30%, sendo a água destilada esterilizada (ADE) utilizada como testemunha. Os resultados mostraram que extratos de Pau-ferro, Pata de vaca, Jurema preta e Boldo baiano com concentrações de 30% inibiram o crescimento micelial em 85,6; 83,3; 85,6 e 88,9%, respectivamente, contudo, os extratos testados não foram eficientes para inibir a incidência e a severidade da doença.

ABSTRACT This study aimed to verify L. theobromae control by using plant extracts. Initially, L. theobromae mycelial growth inhibition was evaluated on Petri dishes containing PDA medium supplemented with extracts of “boldo baiano”, “jurema preta”, “pata-de-vaca” and “pau-ferro” at the concentrations of 10, 20 and 30% for all extracts. Afterwards, two in vivo experiments were carried out to evaluate the direct and protective effect of the plant extracts tested in vitro on dry rot intensity in bunches of grape cv. Itália, using the concentration of 30%, while sterile distilled water (ADE) was adopted as control. Results showed that extracts of “pau-ferro”, “pata-de-vaca”, “jurema preta” and “boldo baiano” at concentrations of 30% inhibited mycelial growth by 85.6, 83.3, 85.6 and 88.9%, respectively; however, the tested extracts were not efficient in inhibiting the disease incidence and severity.

RESUMO O objetivo deste estudo foi avaliar o efeito dos extratos vegetais sobre L. theobromae pela utilização de produtos como método alternativo. Foi analisado o crescimento micelial de L. theobromae em placas de Petri contendo o meio BDA suplementado com os extratos de Aroeira, Angico, Umburana e Quebra-faca nas concentrações de 10, 20 e 30%. Para avaliação do efeito direto dos extratos sobre o desenvolvimento de L. theobromae, cachos da cv Itália foram inoculados com o patógeno e, após 4 h, pulverizados com os diferentes extratos: Angico e Umburana (30%) e Quebra-faca (10%). Água destilada esterilizada (ADE) foi utilizada como testemunha. Posteriormente, para avaliar o potencial dos extratos como indutores de resistência, cachos da cv Itália foram submetidas aos mesmos tratamentos e, inoculados após 4 horas com L. theobromae. Os resultados mostraram que os extratos de Angico e Umburana nas concentrações de 30%, inibiram o crescimento micelial in vitro em 61,66 e 60% respectivamente, contudo os extratos testados não foram eficientes em inibir a incidência e a severidade da doença.

ABSTRACT The aim of this study was to evaluate the effect of plant extracts on L. theobromae by using products as an alternative method. Mycelial growth of L. theobromae was analyzed on Petri dishes containing PDA medium supplemented with extracts of Brazilian peppertree, angico, amburana and “quebra-faca” at the concentrations of 10, 20 and 30%. To evaluate the direct effect of extracts on the development of L. theobromae, bunches of grape cv Itália were inoculated with the pathogen and, after 4 h, sprayed with the different extracts: angico and amburana (30%) and “quebra-faca” (10%). Sterile distilled water (ADE) was used as control. Subsequently, to evaluate the potential of extracts as resistance inducers, bunches of grape cv Itália were subjected to the same treatments and inoculated, after 4 hours, with L. theobromae. Results showed that extracts of angico and amburana at the concentration of 30% inhibited in vitro mycelial growth by 61.66 and 60%, respectively; however, the tested extracts were not efficient in inhibiting the disease incidence and severity.

O crescimento micelial de L. theobromae foi analisado em placas de Petri contendo o meio batata-dextrose-ágar suplementado com o óleo essencial nas seguintes concentrações: 0; 0,25; 0,5; 1,0; 1,5; 2,0% de óleo de Menta Para avaliação do efeito direto do óleo essencial sobre o desenvolvimento da doença, os cachos de uva foram inoculados com o patógeno e, após 4 h, aspergidos com soluções contendo as mesmas concentrações descritas anteriormente; posteriormente, com o objetivo de avaliar o potencial do óleo essencial no tratamento preventivo, os mesmos tratamentos foram aplicados em cachos de uva 4 horas antes da inoculação. O óleo de Menta, em todas as concentrações testadas foi capaz de inibir o crescimento micelial de L. theobromae em 100%. No tratamento curativo, com relação à incidência, Menta 0,25% diferiu estatisticamente dos demais tratamentos, apresentando 88% de bagas infectadas, e em todas as concentrações não foi capaz de reduzir a severidade da podridão seca em cachos de uva. Enquanto no tratamento preventivo, com relação a incidência, o óleo de Menta 0,5% diferiu estatisticamente da testemunha, apresentando 86% e 94% bagas infectadas respectivamente, e apresentou melhor redução da severidade da podridão seca, apresentando índice de doença de 73,7% em relação a testemunha, com 98% de índice de doença.

ABSTRACT The presence of Lasiodiplodia theobromae (dry rot) on the vine, in irrigated areas of the Brazilian semi-arid region, has been very aggressive, infecting several plant parts. Essential oils show efficient disease control, both for their fungitoxic action, which inhibits mycelial growth and spore germination, and for the presence of elicitor compounds. Thus, this study aimed to obtain L. theobromae control by using the oil from mint (Mentha arvensis). The mycelial growth of L. theobromae was analyzed on Petri plates containing potato-dextrose-agar medium supplemented with the essential oil at the following concentrations: 0; 0.25; 0.5; 1.0; 1.5; 2.0% of mint oil. To evaluate the direct effect of the essential oil on the disease development, the grape bunches were inoculated with the pathogen and, after 4 h, sprayed with solutions containing the same previously described concentrations; then, to evaluate the potential of essential oil in the preventive treatment, the same treatments were applied on grape bunches at 4 hours before inoculation. The mint oil, at all tested concentrations, was capable of inhibiting the mycelial growth of L. theobromae by 100%. In the curative treatment, considering incidence, 0.25% mint was statistically different from the remaining treatments, showing 88% infected berries, while dry rot severity of grape bunches could not be reduced by any of the concentrations. For the preventive treatment, considering incidence, 0.5% mint oil statistically differed from control, showing 86% and 94% infected berries, respectively, and there was greater reduction in dry rot severity, since the disease index was 73.7%, compared to control, which had disease index of 98%.

RESUMO Entre as infecções fúngicas que afetam a pós-colheita da videira se destacam a podridão seca ocasionada por Lasiodiplodia theobromae que provoca perdas superiores as perdas no campo, pelo fato de serem adicionados aos custos de colheita, transporte e armazenamento. Produtos naturais vêm sendo utilizados no controle de doenças de plantas, apresentando eficiência na redução do uso de defensivos agrícolas. Desta forma, o presente trabalho teve por objetivo obter o controle da podridão seca pela utilização de produtos alternativos. Foi analisado o crescimento micelial de L. theobromae em placas de Petri contendo o meio BDA suplementado com os seguintes produtos: óleo de citronela, cravo e menta, nas concentrações de 1,0% e 1,5% e quitosana (1,5%). A testemunha foi placa com meio BDA sem adição de produto. Para avaliação do efeito curativo dos óleos sobre o desenvolvimento da podridão seca, cachos da cv Itália foram inoculados com o patógeno e, após 4 h, aspergidos com concentrações 1,0% e 1,5% de solução de óleos de cravo, citronela e menta e de Quitosana a 1,5% e água (testemunha). Posteriormente, para avaliar o efeito protetor dos cachos da cv Itália foram submetidos aos mesmos tratamentos e, inoculados após 4 horas com L. theobromae. Na avaliação do tempo de inoculação, cachos da cv. Itália foram aspergidos com solução de óleos essenciais: cravo (1,5%) e citronela (1,0% e 1,5%), e água (testemunha) e, inoculados após 24, 48, 72 ou 96 h com o fungo L. theobromae. Os óleos de Citronela e Menta, em todas as concentrações foram capazes de inibir o crescimento micelial de L. theobromae. Os óleos essenciais e Quitosana foram eficientes em reduzirem a incidência da podridão seca em cachos da videira cv Itália. No entanto, em relação à severidade apenas os óleos de citronela e menta, na concentração de 1,5% mantiveram a capacidade de reduzir a doença em 30 e 29,2%, respectivamente. O tempo de inoculação influenciou a incidência e a severidade da doença. Quanto mais tempo o material esperou para ser inoculado, maior foi à incidência e a severidade da doença.

ABSTRACT Among fungal infections that affect the grapevine post-harvest is dry rot caused by Lasiodiplodia theobromae, which leads to losses greater than those in the field since they are added to the costs of harvest, transport and storage. Natural products have been used to control plant diseases, showing efficiency in reducing the use of pesticides. Thus, this study aimed to obtain control of dry rot by using alternative products. Mycelial growth of L. theobromae was analyzed on Petri plates containing PDA medium supplemented with the following products: citronella, clove and mint oil at the concentrations of 1.0% and 1.5%, and chitosan (1.5%). Control was a plate with PDA medium with no product addition. To evaluate the healing effect of oils on dry rot development, bunches of grapevine cv Itália were inoculated with the pathogen and, after 4 h, sprayed with the concentrations of 1.0% and 1.5% clove, citronella and mint oil solution; 1.5% chitosan solution, and water (control). Subsequently, to evaluate the protective potential, bunches of grapevine cv. Itália were subjected to the same treatments and inoculated, after 4 hours, with L. theobromae. For the assessment of inoculation time, bunches of grapevine cv. Itália were sprayed with solution of the essential oils: clove (1.5%) and citronella (1.0% and 1.5%), and water (control), and inoculated after 24, 48, 72 or 96 h with the fungus L. theobromae. Mint and citronella oils, at all concentrations, were capable of inhibiting the mycelial growth of L. theobromae. Essential oils and chitosan were effective in reducing the incidence of dry rot in bunches of grapevine cv. Itália. However, regarding the severity alone, citronella and mint oils, at the concentration of 1.5%, retained the ability to reduce the disease by 30 and 29.2%, respectively. The inoculation time influenced the disease incidence and severity. The longer the time for the material to be inoculated, the higher the incidence and the severity of the disease.
